    Aiden Deacon from the University of Minnesota-Twin Cities, Minneapolis, discusses a research paper he co-authored that was published in Volume 16 of Oncotarget, titled “Dissecting the functional differences and clinical features of R-spondin family members in metastatic prostate cancer.” DOI - https://doi.org/10.18632/oncotarget.28758 Correspondence to - Justin Hwang - jhwang@umn.edu Video interview - https://www.youtube.com/watch?v=OXKhWWU1gnY Abstract This study investigates the R-spondin family of genes (RSPO1/2/3/4), a group of secreted proteins that act as Wnt regulators, and their subsequent role in advanced prostate cancer (PC). When evaluating transcriptomic data from primary and metastatic PC patients, we found that alterations in RSPO2 were more prevalent than in other RSPO family members or Wnt-regulating genes APC and CTNNB1. Further, we found that RSPO2 alterations in PCs were significantly associated with worse disease-free survival. Through our in silico modeling, RSPO2 exhibited strong positive associations with genes regulating epithelial-mesenchymal transition (EMT) and double-negative prostate cancer (DNPC), but had negative correlations with androgen receptor (AR) and AR-associated genes. Furthermore, 3D modeling of RSPO2 revealed structural differences between itself and other RSPOs. In cell lines, RSPO2 overexpression caused up-regulation of EMT pathways, including EMT-regulatory transcription factors ZEB1, ZEB2, and TWIST1. Conversely, this was not observed when CTNNB1 was overexpressed in the same models. These findings highlight that, in PC, RSPO2 functions as a unique member of the R-spondin family by promoting genes and signaling pathways associated with aggressive PC, and RSPO2 amplifications are associated with poor outcomes in PC patients.     This Episodes Questions: Brians Questions: Hello gentleman, thank you again for one of the best woodworking podcasts available. I recently had the opportunity to travel to Lebanon IN for a few days of training at a tool manufacturer. I won't mention the company, but I got to use a Domino and a Rotex sander for the first time..., but I digress. While I was in the airport waiting for my flight home and enjoying a breakfast sandwich, I looked down at the table and noticed the "purposeful-design" logo branded on the top. This experience gave me two questions. One, what finish did Guy and Brian use on these tables to survive the abuse of so many travelers per day through an airport? Two, during the training we sanded a piece of cherry to 1500 grit, giving a very high sheen to the bare wood. What type of technique or finish would you use on something like that? Thank you for the great podcast, and I'm sorry I didn't have time to say hi while I was in Indianapolis. I waved from the plane, but you probably didn't notice. Josh Hello fellow wood-nerds. I would like some advice on finishing. I've been doing a bathroom remodel. Which means I've spent a very long time doing non-woodworking work and learning new skills and desperately missing my shop time. I'm near the end finally and actually get to do some woodwork to wrap it up. My new floor doesn't quite line up with the floor in the next room and the transition pieces available don't quite suit. So I'm making one custom out of walnut. My question is: what would be a good finish? I don't want it glossy or “plasticy” looking. But of course it will be stepped on and exposed to humidity from the shower so it does need to be durable. Thanks in advance Jason Guys Questions: This one may for Guy specifically but I'm interested in getting a 3D printer and would like your opinion on what I might want to look at. I have a total budget of around $1000 and want something that is very turnkey; I don't have a ton of time to learn how to use the machine and would like that will work for me out of the box. Ron Brewer Hi Guys, I am hoping to eventually upgrade my table saw from a jobsite saw.  I have heard a little bit of discussion about the advantages of an induction motor over a universal motor but I was wondering if you could provide some insight about voltage for a table saw.  Obviously higher power is available with table saws that operate on higher voltages but as a hobbyist I am wondering how much power I actually need.  I do make some cabinets and furniture as well as smaller projects. So here is an assortment of questions that are perhaps intertwined: Is the increase in power with a 240V saw worth it?  I would also have to get new electrical added to my garage.  Is the type of motor related to the voltage?  In your opinion are table saws that can be rewired from 120 to 240V a viable option? Would the power actually change with the rewiring?  Are there any other considerations that I am not thinking of? Thank you all for your insight. Brad Huys Questions: If you're installing a herringbone floor, do you need to think about chatoyance, in addition to colour and grain pattern, when you're laying out the pieces? Do you want all of the pieces to be oriented to have similar chatoyance? Lauris Hello Guys, I listen to your podcast while driving to and from work.  I really appreciate that you stay on track and offer several opinions regarding woodworking without the cackling and inside jokes, poor microphone control etc that other woodworking podcasts seem to have. I build a lot of Shaker style furniture with tapered legs and built my own taper jig, a sled which clamps the leg at the desired angle with the widest part of the cutoff leading into the blade.  This has worked very well except that occasionally, the wedge that is cutoff tips into the crack between the saw blade and the insert, jamming and sometimes tripping the saw's overload protection.  I have cut a brand new table saw insert with the intention of creating as close to a zero clearance slot as possible but it still is wide enough for the sliver edge of the taper cutoff to drop in.  How have you avoided this in the past?  Would a reverse orientation of the taper jig be safer such that the thinnest part of the cutoff is cut first and therefore at the back of the blade and the thickest part is the last part cutoff?  Thank you! The students were supervised as they document a Dutch Waterschip . This is a type of fishing vessel that was common in the Zuiderzee before it was reclaimed to form the province of Flevoland . As fish were caught they were kept alive in two chambers in the boat that were open to the sea¸ allowing the fishermen to deliver the freshest of fish to their market . As the water table in Flevoland is lowering, the shipwreck will soon rot away. So the team are out teaching students the craft of documenting the find and preserving what is unique. The legs of two people can be seen standing on the grass above two archaeologists who are in the wreck. From the grass level, there is a 30cm/1 Foot layer of sandy soil until there wreck begins. The body of the ship is exposed and we can see the interlocking planks that form the hull. There is a "stepping stone" path made from sand bags from the keel/center spine of the ship, to the surface. One archaeologist stands at the end of this path with a bucket and at his feet is a scoop that he is using to fill it. The lower part of the ship is filling with water that is seeping in from the surrounding ground. The bun/fish compartment In the body of the ship we see three wooden partitions, that form the two chambers that were used to keep the fish alive. Although not visible to the untrained eye, there are holes to the outside sea. The Mast hole in the keel A close up of the 2M/6Foot wooden sleeper that lies on a forklift pallet. It is about 15CM/6Inches thick. This has a about 30cm/1 Foot square hole in it, that is only about 5CM/2Inch deep. This was used to hold the bottom of the mast. The Timbers Views of the overlapping timbers with cut marks, and pins holding them together. The Back/Aft A view from the back from the back of the ship. We can see that there is a rip in the hull behind the last fish compartment and the back of the ship. Was this a collision or did it occur after sinking ? An archaeologist is fighting a loosing battle to scoop out water. The glamour of the dig. The back of the site tent showing the woodland in the background, a pile of soil that was removed, and a another pile of rotting wood from the ship. There is a makeshift pool of water used to store water pumped from the wreck. The front of the site tent showing the woodland in the background, some storage shipping containers, some steel plates used to spread the load of heavy machinery. A 2M/6Foot wooden sleeper lies on a forklift pallet. A earthen mound is in front of the woods. In front of that is a very wet and damp looking tarpaulin with parts of the ship. They are normally kept wet covered with another tarpaulin so they will not rot. Each has a label on it. Some are held vertical by sandbags. There is a view of the pile of rotten wood, the eacavated soiland the pump pool. There are some rusted iron as braces in the wood. There is a rust shaped axe. Information Displays A informational poster showing how they determined the date to be Dendroarchaeology. If two trees grew during the same period, the thickness of their rings during that period will be the same. Archaeologist were able to make a complete historical map of tree rings going back to prehistory. They did this by comparing the overlapping tree rings from in existing buildings, shipwrecks and artwork. In some cases there were receipts from wood choppers for the timbers. Further back can be used by comparing overlapping rings in different artifacts. A drawing of the ship cross-sections through the center, a top, front and back view. Then a drawing of the ship with triangle sail at the front, and a square sale at the back with rigging. The cross section shows where the bun/fish compartment was located in the middle behind the mast at the bottom of the ship. A informational poster tells us the shipwreck was discovered in 1972 when the land was drained. The ship is 16 meter long and 5.5 meter wide, which is a small version as they often got to 20 meter in length. The ship was complete to the top of the bun/fish compartment. It sank somewhere between 1520 and 1530. It continues to give a timeline of the discovery. The next poster shows the 3d scans from 2022 to 2025. 2022 Shows the front half of the ship with a rats nest of broken timbers. 2023 The entire top deck is exposed 2024 and 2025 show the wood is now cleared down to the frame of the ship. There are also 3D printed versions of the scans in cases. The 3d scanner used to record some of the models. A map showing the shipping routes through the Zuiderzee. Items that visitors are encouraged to touch are fresh moss used to seal the lats, a piece of timber, some iron nails, and braces to keep the planks together. Items that visitors are not allowed to touch include, barrel taps, rings, actual moss, fish bones, and a box of other finds.
