“A couple of things I think are really important when you look at this class of drug: It developed by a concerted effort in cancer drug development to look at new agents that would be effective based on the mechanism. Highlights From This Episode “With the nitrosoureas, there's something really interesting because there's another mechanism that has been identified. And that is that when you put these nitrosoureas in the body, they break down into intermediates, and one of them is an isocyanate. … These isocyanates, what they do is they inhibit DNA repair, therefore have an impact on cells that are damaged. You can think of it as the second mechanism, and people that work in the neuro-oncology space think of this when they think of drugs like lomustine in brain cancer, how that drug decreases the DNA repair protein O6-methylguanine-DNA methyltransferase.” TS 4:11 “These drugs are very lipophilic, meaning they cross the blood-brain barrier. That's why we use them in brain tumors, so that's one of the key things. That's also one of the toxicities we see when drugs cross blood-brain barrier; we see neurotoxicity. So that's one to at least always consider but also the benefit of it crossing over and being able to treat cancers within the CNS.” TS 8:19 “As a group, these drugs are alkylating agents, so definitely the safe handling is essential. And with DNA-damaging agents, that means anybody who is going to come in contact with these drugs. So, carmustine is given intravenously. Lomustine or CCNU, those are capsules. So handling is different depending on the agents.” TS 12:45 “The thing with the lomustine or the CCNU capsules, the thing that's really important here is that the dosing is really different than how we normally give oral medications. And so, it's really important that patients are aware of exactly how much they take and not that they don't repeat the dose every day. So I think just like with other oral regimens that are not daily, we really have to make sure patients are aware of the specifics of how they take the drug.” TS 14:25
BUFFALO, NY- March 20, 2024 – A new #research paper was #published in Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 16, Issue 5, entitled, “FoxO6-mediated ApoC3 upregulation promotes hepatic steatosis and hyperlipidemia in aged rats fed a high-fat diet.” FoxO6, an identified factor, induces hyperlipidemia and hepatic steatosis during aging by activating hepatic lipoprotein secretion and lipogenesis leading to increased ApoC3 concentrations in the bloodstream. However, the intricate mechanisms underlying hepatic steatosis induced by elevated FoxO6 under hyperglycemic conditions remain intricate and require further elucidation. In this new study, researchers Dae Hyun Kim, Seulah Lee, Sang Gyun Noh, Jaewon Lee, and Hae Young Chung from Pusan National University aimed to delineate the regulatory pathway involving ApoC3 controlled by FoxO6 and its resultant functional impacts. “[...] we employed a spectrum of models including liver cell cultures, aged rats subjected to HFD, transgenic mice overexpressing FoxO6 (FoxO6-Tg), and FoxO6 knockout mice (FoxO6-KO).” Their findings indicate that FoxO6 triggered ApoC3-driven lipid accumulation in the livers of aged rats on an HFD and in FoxO6-Tg, consequently leading to hepatic steatosis and hyperglycemia. Conversely, the absence of FoxO6 attenuated the expression of genes involved in lipogenesis, resulting in diminished hepatic lipid accumulation and mitigated hyperlipidemia in murine models. Additionally, the upregulation of FoxO6 due to elevated glucose levels led to increased ApoC3 expression, consequently instigating cellular triglyceride mediated lipid accumulation. The transcriptional activation of FoxO6 induced by both the HFD and high glucose levels resulted in hepatic steatosis by upregulating ApoC3 and genes associated with gluconeogenesis in aged rats and liver cell cultures. “Our conclusions indicate that the upregulation of ApoC3 by FoxO6 promotes the development of hyperlipidemia, hyperglycemia, and hepatic steatosis in vivo, and in vitro. On this episode of the Lessons Learned for Vets podcast, we conclude our 2-part series with Ken Davenport. Author of The Stoic Transition, a Guide to How Veterans can Thrive in their Next Adventure, Ken brings a fresh, civilian perspective to the military transition process. Ken encourages transitioning service members to start the process at least 18 months out. While this is common advice, Ken further explains why. There is a massive amount of information that is passed to service members as they exit the military. By starting early, a person has a runway in which to process and think through the information. It's important to allow yourself the time to decompress, assess and review what it is that you want after the military. Set realistic expectations. In some cases, a person may have to take a couple of steps back to move forward. An O5 or O6 may have expectations of running a private business. Although they are equipped to lead, working in the private sector requires learning a new language, a new system and a new environment. Learning to recognize what is within or out of one's control is helpful in the transition process. It's impossible for a person to completely control how they will fit into an opportunity given to them. Know that what you bring to the right environment will be utilized and rewarded. If it's not the right environment, it will be made clear. In such a situation, it will be necessary to pivot to something new. The culture of an organization is paramount to having a successful next career. There are certain indicators on the surface such as how people dress and present themselves, but there is a foundation of intangibles that can make or break a culture. For instance, does the company reward risk or value individual opinions? How do you begin to research culture? The best way to understand culture is to network. Find other veterans on LinkedIn who work at the company and ask questions. Be true to yourself and know that there is a culture for everyone. Ken talks through the idea of backward planning. In this process, a person plans for different eventualities and makes course corrections based on the feedback they're getting as they move toward their goal. In general, civilians tend to be reactive. Instead of thinking about how they want to reach their goal, they react to what's right in front of them and make plans for only the next step. Backward planning is important for achieving the lifestyle that you are looking for while you are still active and productive. Fear is prevalent when facing the unknown. If you are facing fear, try getting out of your head and freeing yourself from overanalyzing the situation. Take a break and a step back. Reach out to a mentor to gain new perspective. Ken encourages people to create their own board of advisors. Find people who know you and support you. Find people who will give you unfiltered advice in the area that you are seeking. Find a person who may be able to open doors for you in your chosen industry. Ken talks about the concept of memento mori. Don't put off for tomorrow what you could do today. Distance and age of the massive stellar cluster Westerlund 1 II The eclipsing binary W36 by Danilo F. Rocha et al. on Tuesday 11 October Westerlund 1 (Wd 1) is one of the most relevant star clusters in the Milky Way to study massive star formation, although it is still poorly known. Here, we used photometric and spectroscopic data to model the eclipsing binary W36, showing that its spectral type is O6.5 III + O9.5 IV, hotter and more luminous than thought before. Its distance $d_{rm W36}$ $=$ 4.03$pm$0.25 kpc agrees, within the errors, with three recent Gaia-EDR3-based distances reported in Paper I, Beasor & Davies, and by Negueruela's group. However, they follow different approaches to fix the zero-points for red sources such as those in Wd 1 and to select the best approach, we used an accurate modelling of W36. The weighted mean distance of our parallax (Paper I) and binary distances results in $d_{rm wd1}$ = 4.05 $pm$0.20 kpc, with an unprecedented accuracy of 5%. We adopted isochrones based on the Geneva code with supersolar abundances to infer the age of W36B as 6.4 $pm$ 0.7 Myr. This object seems to be part of the prolific star formation burst represented by OB giants and supergiants that occurred at 7.1 $pm$ 0.5 Myr ago, which coincides with the recently published PMS isochrone with age 7.2 Myr. Other BA-type luminous evolved stars and Yellow Hypergiants spread in the age range of 8--11 Myr. The four Red Supergiants discussed in paper I represent the oldest population of the cluster with an age of 10.7 $pm$ 1 Myr. Sledge(2012-03-12): Books - AmazonToday, I have Major General Dale Alford, USMC on the podcast. He is currently the commanding general of Training Command in Quantico Virginia, which is not to be confused with training and education command or T com. Training Command touches the entire marine corps both officer and enlisted which includes close to 90, regionally aligned formal learning centers aligned under 17 unique O6 level commands with 15 commanded by marine colonels and two commanded by Navy captains. These learning centers are spread across the continental United states with additional detachments in Hawaii and Okinawa. At any given time there can be more than 28,000 marines and sailors under the command of Major General Alford. This includes both the instructor staff and students. All of these personnel are engaged in training associated with 242 different military occupational specialties. While I certainly can't list all of the formal learning centers, some of the more recognizable ones are officer candidate school, the basic school, the weapons and training battalions, school of infantry both east and West coast, three marine aviation training support groups, the different Marine Corps intelligence schools, Marine corps communication-electronics school, the engineer school, combat service support schools to include the all the different training at Ft Leonard Wood for motor transport and MPs, artillery training at Fort Sill, and field medical training battalions which train our corpsman on both the East and West Coast, and the assault amphibian school.

May is a fun month, all month long to honor MotHER's day, we are bringing you 5 weeks of mother/daughter duos to the show. You get to hear about the family dynamics, honor and how the military really is a family business for these special mother-daughter teams. This week is week 3 and we have Kathy Donovan and Emily Cirocco. Both women are Navy, one retired after 30 years of faithful service and the other still serving and getting ready to be a recruiter! I was a captain in the Army an O3 but a Captian in the Navy is an O6, which is the rank right below generals/admirals. Really senior ranking and the competition is steep. Kathy Donovan achieved that rank in her 30-year career and is known as a legend to those who served under her. Her daughter Emily Cirocco is a Petty Officer 3rd Class and looking forward to continuing to serve and make a name for herself in the Navy. Emily is a new mom and we get to hear both women gush over the baby, hear about the fun they had served together, special memories and how they became a family. Our first blended family in the series with a very special bond and story. In our first podcast I asked Joel to provide any interventional studies showing harm from red meat, or any unprocessed meat. He was unable to provide these studies at that time, or in the 2 weeks that followed before I released our podcast debate. P., Fujii, S. Abstract: Temozolomide, a DNA methylating agent, is the primary chemotherapeutic drug used in glioblastoma treatment. TMZ induces mostly N-alkylation adducts (N7-methylguanine and N3-methyladenine) and some O6-methylguanine (O6mG). Current models propose that during DNA replication, thymine is incorporated across from O6mG, promoting a futile cycle of mismatch repair (MMR) that leads to DNA double strand breaks (DSBs). To revisit the mechanism of O6mG processing, we reacted plasmid DNA with N-Methyl-N-nitrosourea (MNU), a temozolomide mimic, and incubated it in Xenopus egg extracts. We show that in this system, mismatch repair (MMR) proteins are enriched on MNU-treated DNA and we observe robust, MMR-dependent, repair synthesis. Our evidence also suggests that MMR, initiated at O6mG:C sites, is strongly stimulated in cis by repair processing of other lesions, such as N-alkylation adducts. Importantly, MNU-treated plasmids display DSBs in extracts, the frequency of which increased linearly with the square of alkylation dose. We suggest that DSBs result from two independent repair processes, one involving MMR at O6mG:C sites and the other involving BER acting at a nearby N-alkylation adducts. We propose a new, replication-independent mechanism of action of TMZ, that operates in addition to the well-established cell cycle dependent mode of action. Copy rights belong to original authors. For the career minded Naval professional, to have a chance for the greatest advancement and promotion, you have to push and push hard. The reputation you build in your first 10 years sets the tone for the rest. Except for very rare exceptions, there are no second chances. There are no pauses - one iffy set of orders - one poorly timed FITREP, and you are on an off-ramp. You must work harder, you must sacrifice, and if you are to have a family young, you need a very strong support structure. For men - there is always the RADM Sestak, USN (Ret) option; wait until post O6, then start. For women though, there are some hard biological facts.The average American woman gets married at age 26. For college-educated women the average age at first birth is ~30. If you want to have more than 2 kids, you need to start earlier. You need to time it right - and Mother Nature has her own schedule that doesn't often match yours. With women making up more of the military than ever, what are the challenges out there biological, cultural, psychological, and relationship wise to "making it happen?" You can't have it all - but how do you get the best mix you can? For the career minded Naval professional, to have a chance for the greatest advancement and promotion, you have to push and push hard. The reputation you build in your first 10 years sets the tone for the rest.Except for very rare exceptions, there are no second chances. There are no pauses, one iffy set of orders, one poorly timed FITREP, and you are on an off-ramp. You must work harder, you must sacrifice, and if you are to have a family young, you need a very strong support structure.For men - there is always the RADM Sestak, USN (Ret) option; wait until post O6, then start the family your peers did 20-yrs ago. Glioblastoma (GBM) is the most common and aggressive brain tumor in adults. Despite multimodal treatments including surgery, chemotherapy and radiotherapy the prognosis remains poor and relapse occurs regularly. The alkylating agent temozolomide (TMZ) has been shown to improve the overall survival in patients with malignant gliomas, especially in tumors with methylated promoter of the O6-methylguanine-DNA-methyltransferase (MGMT) gene. However, intrinsic and acquired resistance towards TMZ makes it crucial to find new therapeutic strategies aimed at improving the prognosis of patients suffering from malignant gliomas. Cold atmospheric plasma is a new auspicious candidate in cancer treatment. In the present study we demonstrate the anti-cancer properties of different dosages of cold atmospheric plasma (CAP) both in TMZ-sensitive and TMZ-resistant cells by proliferation assay, immunoblotting, cell cycle analysis, and clonogenicity assay. Importantly, CAP treatment restored the responsiveness of resistant glioma cells towards TMZ therapy. Concomitant treatment with CAP and TMZ led to inhibition of cell growth and cell cycle arrest, thus CAP might be a promising candidate for combination therapy especially for patients suffering from GBMs showing an unfavorable MGMT status and TMZ resistance.
