The major traditional exchanges have adapted well to the migration of transactional activity to other trading platforms, notably by developing their data and post-trade revenues, so it is surprising that they remain indifferent to tokenisation. With some notable exceptions, traditional stock exchanges have shown little interest in the revolutionary potential of issuing, trading and servicing tokens. Explanations for this indifference vary. Some say the existing markets (especially equity trading) are already so efficient that tokenisation is unnecessary. The fact that exchanges which have invested in tokenisation have yet to earn a return, and are now making economies, is not encouraging. If tokenisation ever does take off, add some larger exchanges, they could simply acquire the successful platforms. The long lists of intermediaries that stand between issuers and investors – global brokers, local brokers, clearing houses, securities depositories, payments banks, custodian banks and so on – also have limited incentives to encourage the emergence of peer-to-peer exchanges. So even the leading traditional stock exchange groups are focusing on a narrow range of token opportunities, such as cryptocurrencies and privately managed assets, and leaving potential lavish operational cost savings and new product earnings untouched. Dominic Hobson, co-founder of Future of Finance, moderated a discussion of about traditional stock exchanges and tokenisation with Marco Kessler, business head of digital securities at six Digital Exchange SDX, Hirander Misra, chairman and CEO of GMEX Group, Ricardo Correia, a partner in the financial services practice at Bain and Company, Benedikt Schuppli, co-founder and CBDO at Obligate and Miryusup Abdullaev, managing director of the Deutsche Börse Digital Exchange (DBDX). OutlineChapter 14- Hypovolemic States- Etiology - True volume depletion occurs when fluid is lost from from the extracellular fluid at a rate exceeding intake - Can come the GI tract - Lungs - Urine - Sequestration in the body in a “third space” that is not in equilibrium with the extracellular fluid. - When losses occur two responses ameliorate them - Our intake of Na and fluid is way above basal needs - This is not the case with anorexia or vomiting - The kidney responds by minimizing further urinary losses - This adaptive response is why diuretics do not cause progressive volume depletion - Initial volume loss stimulates RAAS, and possibly other compensatory mechanisms, resulting increased proximal and collecting tubule Na reabsorption. - This balances the diuretic effect resulting in a new steady state in 1-2weeks - New steady state means Na in = Na out - GI Losses - Stomach, pancreas, GB, and intestines secretes 3-6 liters a day. - Almost all is reabsorbed with only loss of 100-200 ml in stool a day - Volume depletion can result from surgical drainage or failure of reabsorption - Acid base disturbances with GI losses - Stomach losses cause metabolic alkalosis - Intestinal, pancreatic and biliary secretions are alkalotic so losing them causes metabolic acidosis - Fistulas, laxative abuse, diarrhea, ostomies, tube drainage - High content of potassium so associated with hypokalemia - [This is a mistake for stomach losses] - Bleeding from the GI tract can also cause volume depletion - No electrolyte disorders from this unless lactic acidosis - Renal losses - 130-180 liters filtered every day - 98-99% reabsorbed - Urine output of 1-2 liters - A small 1-2% decrease in reabsorption can lead to 2-4 liter increase in Na and Water excretion - 4 liters of urine output is the goal of therapeutic diuresis which means a reduction of fluid reabsorption of only 2% - Diuretics - Osmotic diuretics - Severe hyperglycemia can contribute to a fluid deficit of 8-10 Iiters - CKD with GFR < 25 are poor Na conservers - Obligate sodium losses of 10 to 40 mEq/day - Normal people can reduce obligate Na losses down to 5 mEq/day - Usually not a problem because most people eat way more than 10-40 mEq of Na a day. - Salt wasting nephropathies - Water losses of 2 liters a day - 100 mEq of Na a day - Tubular and interstitial diseases - Medullary cystic kidney - Mechanism - Increased urea can be an osmotic diuretic - Damage to tubular epithelium can make it aldo resistant - Inability to shut off natriuretic hormone (ANP?) - The decreased nephro number means they need to be able to decrease sodium reabsorption per nephron. This may not be able to be shut down acutely. - Experiment, salt wasters can stay in balance if sodium intake is slowly decreased. (Think weeks) - Talks about post obstruction diuresis - Says it is usually appropriate rather than inappropriate physiology. - Usually catch up solute and water clearance after releasing obstruction - Recommends 50-75/hr of half normal saline - Talks briefly about DI - Skin and respiratory losses - 700-1000 ml of water lost daily by evaporation, insensible losses (not sweat) - Can rise to 1-2 liters per hour in dry hot climate - 30-50 mEq/L Na - Thirst is primary compensation for this - Sweat sodium losses can result in hypovolemia - Burns and exudative skin losses changes the nature of fluid losses resulting in fluid losses more similar to plasma with a variable amount of protein - Bronchorrhea - Sequestration into a third space - Volume Deficiency produced by the loss of interstitial and intravascular fluid into a third space that is not in equilibrium with the extracellular fluid. - Hip fracture 1500-2000 into tissues adjacent to fxr - Intestinal obstruction, severe pancreatitis, crush injury, bleeding, peritonitis, obstruction of a major venous system - Difference between 3rd space and cirrhosis ascities - Rate of accumulation, if the rate is slow enough there is time for renal sodium and water compensation to maintain balance. - So cirrhotics get edema from salt retension and do not act as hypovolemia - Hemodynamic response to volume depletion - Initial volume deficit reduced venous return to heart - Detected by cardiopulmonary receptors in atria and pulmonary veins leading to sympathetic vasoconstriction in skin and skeletal muscle. - More marked depletion will result in decreased cardiac output and decrease in BP - This drop in BP is now detected by carotid and aortic arch baroreceptors resulting in splanchnic and renal circulation vasoconstriction - This maintains cardiac and cerebral circulation - Returns BP toward normal - Increase in BP due to increased venous return - Increased cardiac contractility and heart rate - Increased vascular resistance - Sympathetic tone - Renin leading to Ang2 - These can compensate for 500 ml of blood loss (10%) - Unless there is autonomic dysfunction - With 16-25% loss this will not compensate for BP when patient upright - Postural dizziness - Symptoms - Three sets of symptoms can occur in hypovolemic patients - Those related to the manner in which the fluid loss occurs - Vomiting - Diarrhea - Polyuria - Those due to volume depletion - Those due to the electrode and acid base disorders that can accompany volume depletion - The symptoms of volume depletion are primarily related to the decrease in tissue perfusion - Early symptoms - Lassitude - Fatiguability - Thirst - Muscle cramps - Postural dizziness - As it gets more severe - Abdominal pain - Chest pain - Lethargy - Confusion - Symptomatic hypovolemia is most common with isosmotic Na and water depletion - In contrast pure water loss, causes hypernatremia, which results in movement of water from the intracellular compartment to the extracellular compartment, so that 2/3s of volume loss comes from the intracellular compartment, which minimizes the decrease in perfusion - Electrolyte disorders and symptoms - Muscle weakness from hypokalemia - Polyuria/poly dips is from hyperglycemia and hypokalemia - Lethargy, confusion, Seizures, coma from hyponatremia, hypernatremia, hyperglycemia - Extreme salt craving is unique to adrenal insufficiency - Eating salt off hands ref 18 - Evaluation of the hypovolemic patient - Know that if the losses are insensible then the sodium should rise - Volume depletion refers to extracellular volume depletion of any cause, while dehydration refers to the presence of hypernatremia due to pure water loss. Such patients are also hypovolemic. - Physical exam is insensitive and nonspecific - Finding most sensitive and specific finding for bleeding is postural changes in blood pressure - I don't find this very specific at all! - Recommends laboratory confirmation regardless of physical exam - Skin and mucous membranes - Should return too shape quickly - Elastic property is called Turgur - Not reliable is patients older than 55 to 60 - Dry axilla - Dry mucus membranes - Dark skin in Addison's disease Frim increased ACTH - Arterial BP - As volume goes down so does arterial BP - Marked fluid loss leads to quiet korotkoff signs - Interpret BP in terms of the patients “normal BP” - Venous pressure - Best done by looking at the JVP - Right atrial and left atrial pressure - LV EDP is RAP + 5 mmHg - Be careful if valvular disease, right heart failure, cor pulmonare, - Figure 14-2 - Shock - 30% blood loss - Lab Data - Urine Na concentration - Should be less than 25 mmol/L, can go as low as 1 mmol/L - Metabolic alkalosis can throw this off - Look to the urine chloride - Figure 14-3 - Renal artery stenosis can throw this off - FENa - Mentions that it doesn't work so well at high GFR - Urine osmolality - Indicates ADH - Volume depletion often associated with urine osm > 450 - Impaired by - Renal disease - Osmotic diuretic - Diuretics - DI - Mentions that severe volume depletion and hypokalemia impairs urea retension in renal medulla - Points out that isotonic urine does not rule out hypovolemia - Mentions specific gravity - BUN and Cr concentration - Normal ratio is 10:1 - Volume depletion this goes to 20:1 - Serum Na - Talks about diarrhea - Difference between secretory diarrhea which is isotonic and just causes hypovolemia - And osmotic which results in a lower electrolyte content and development of hypernatremia - Talks about hyperglycemia - Also can cause the sodium to rise from the low electrolyte content of the urine - But the pseudohyponatraemia can protect against this - Plasma potassium - Treatment - Both oral and IV treatment can be used for volume replacement - The goal of therapy are to restore normovolemia - And to correct associated acid-base and electrolyte disorders - Oral Therapy - Usually can be accomplished with increased water and dietary sodium - May use salt tablets - Glucose often added to resuscitation fluids - Provides calories - Promotes intestinal Na reabsorption since there is coupled Na and Glucose similar to that seen in the proximal tubule - Rice based solutions provide more calories and amino acids which also promote sodium reabsorption - 80g/L of glucose with rice vs 20 g/L with glucose alone - IV therapy - Dextrose solutions - Physiologically equivalent to water - For correcting hypernatremia - For covering insensible losses - Watch for hyperglycemia - Footnote warns against giving sterile water - Saline solutions - Most hypovolemic patients have a water and a sodium deficit - Isotonic saline has a Na concentration of 154, similar to that of plasma see page 000 - Half-isotonic saline is equivalent to 550 ml of isotonic saline and 500 of free water. Is that a typo? - 3% is a liter of hypertonic saline and 359 extra mEq of Na - Dextrose in saline solutions - Give a small amount of calories, otherwise useless - Alkalinizing solutions - 7.5% NaHCO3 in 50 ml ampules 44 mEq of Na and 44 mEq of HCO3 - Treat metabolic acidosis or hyperkalemia - Why 44 mEq and not 50? - Do not give with calcium will form insoluble CaCO3 - Polyionic solutions - Ringers contains physiologic K and Ca - Lactated Ringers adds 28 mEq of lactate - Spreads myth of LR in lactic acidosis - Potassium chloride - Available as 2 mEq/mL - Do not give as a bolus as it can cause fatal hyperkalemia - Plasma volume expanders - Albumin, polygelastins, hetastarch are restricted to vascular space - 25% albumin can pull fluid into the vascular space - 25% albumin is an albumin concentration of 25 g/dL compare to physiologic 4 g/dL - Says it pulls in several times its own volume - 5% albumin is like giving plasma - Blood - Which fluid? - Look at osmolality, give hypotonic fluids to people with high osmolality - Must include all electrolytes - Example of adding 77 mEw of K to 0.45 NS and making it isotonic - DI can be replaced with dextrose solutions, pure water deficit - Case 14-3 - Diarrhea with metabolic acidosis - He chooses 0.25 NS with 44 mEq of NaCl and 44 NaHCO3 - Talks about blood and trauma - Some studies advocate delaying saline until penetrating trauma is corrected APR about to. Keep BP low to prevent bleeding. Worry about diluting coagulation factors - Only do this if the OR is quickly available - Volume deficit - Provides formula for water deficit and sodium deficit - Do not work for isotonic losses - Provides a table to adjust fluid loss based on changes in Hgb or HCTZ - Says difficult to estimate it from lab findings and calculations - Follow serial exams - Serial urine Na - Rate of replacement - Goal is not to give fluid but to induce a positive balance - Suggests 50-100 ml/hr over what is coming out of the body - Urine - Insensibles 30-50 - Diarrhea - Tubes - Hypovolemic shock - Due to bleeding - Sequesting in third space - Why shock? - Progressive volume depletion leads to - Increased sympathetic NS - Increased Ang 2 - Initially this maintains BP, cerebral and coronary circulation - But this can decrease splanchnic, renal and mucocutaneous perfusion - This leads to lactic acicosis - This can result in intracellular contents moving into circulation or translocation of gut bacteria - Early therapy to prevent irreversible shock - In dogs need to treat with in 2 hours - In humans may need more than 4 hours - Irreversible shock associated with pooling of blood in capillaries - Vasomotor paralysis - Hyperpolarization of vascular smooth muscle as depletion of ATP allows K to flowing out from K channels opening. Ca flows out too leading to vasodilation - Glyburide is an K-ATP channel inhibitor (?) caused increased vasoconstriction and BP - Pluggin of capillaries by neutrophils - Cerebral ischemia - Increased NO generation - Which Fluids? - Think of what is lost and replace that. - Bleeding think blood - Raise the hct but not above 35 - Acellular blood substitutes, looked bad at the time of this writing - Di aspirin cross linked hemoglobin had increased 2 and 28 day mortality vs saline - Colloids sound great but they fail in RCTs - SAFE - FEAST - Points out that saline replaces the interstitial losses why do we think those losses are unimportant - Pulmonary circulation issue - Pulmonary circulation is more leaky so oncotic pressure less effective there - Talks about the lungs be naturally protected from pulmonary edema - Rate of fluid - 1-2 liters in first hour - Suggests CVP or capillary wedge pressure during resuscitation - No refs in the rate of fluid administration section - Lactic acidosis - Points out that HCO can impair lactate utilization - Also states that arterial pH does not point out what is happening at the tissue level. Welcome to The Nonlinear Library, where we use Text-to-Speech software to convert the best writing from the Rationalist and EA communities into audio. This is: Even an obligate omnivore can try to eat less meat, published by Joe Rogero on July 8, 2023 on The Effective Altruism Forum. I've seen some good posts lately about veganism and health. Here's my two cents. I'm convinced by the arguments that many animals might have conscious experiences. I'd prefer not to torture the ones that do. Unfortunately, if I adopted a fully vegan lifestyle I might well be hospitalized for malnutrition within the year. In this post I'll give an overview of how I balance nutrition, ethics, and allergy constraints, and solicit ideas for improvement. My problem I am demonstrably allergic to a startling variety of foods, including, but not limited to: Peas Celery Uncooked egg yolks Chickpeas (and derivatives like hummus) Beans (but not peanuts, which are also legumes) Not deathly allergic, but definitely my-afternoon-is-ruined allergic. A tablespoon of hummus can give me 2-4 hours of intensely distracting pain. I have also experienced an identical, though sometimes inconsistent, reaction to: Fish Soylent Shellfish Eggplant Sweet potato Cranberry sauce Butternut squash Every protein shake mix I have ever tried Several mixtures containing none of the above but some hard-to-isolate combination of ingredients You will notice that several of the items on the above lists are widely considered key sources of vital nutrients in a vegetarian or vegan diet. Coupled with my strong dislike of spicy food, a typical restaurant menu typically contains at most 1-5 entrees I can safely eat. Catered meals often hedge me out entirely. At one (non-EA) event, after listing my allergies in the RSVP, I was served a dinner plate consisting of fish, lima beans, and rice pilaf with peas and celery. Even with these honestly ridiculous constraints, I've found some cheap ways to reduce my dependence on harmfully-produced meat. Reducing harm in an omnivorous diet It's easier to cut the first half of my meat intake than the second half. Diminishing returns apply. By the same logic, two people eating half as much meat is just as good as one person eating none. Here are some things I've learned while trying to minimize the suffering my diet imposes: Tofu is a fine supplement in many dishes, and rice is a cheap and flexible staple. Green vegetables like spinach and broccoli make good additions to any diet. Nuts and many dried fruits are nutrient-rich. Also, peanuts and cashews add a nice crunchy texture to homemade rice dishes. I can also eat cooked eggs and yogurt, and I've heard some convincing arguments in favor of eggs being less harmful than chicken and dairy less harmful than beef. I'm distrustful of "cruelty-free" branding because so many standards for that kind of thing are false or misleading, but with further research I expect I could find more harm-minimizing options there too. Being allergic to fish really hurts, because I think fish probably suffer less than birds or mammals, if at all. If I could eat more fish instead of meat, I would. I could probably bring myself to eat insect-based protein if it were a) actually available where I live and b) not recognizably still a whole bug at the time. Still working on that angle. I'm tentatively excited about lab-grown meat. My wife and I did some math and determined we aren't getting enough protein in our diets, so we're stepping up meat intake overall, but it's still a significant improvement over my past eating habits. I used to cook lots of chicken. My wife can eat beans and likes them fine. We don't have to eat the same things, especially when leftovers are available. We do a lot of home cooking, buy foods that keep, and try not to waste anything. If you have other ideas for reducing diet-induced harm, please share! Conclusion Even if you can't or don't want to fully expunge meat from your diet, it's possible to signific...

He studied law at the University of Zürich and always had a knack for the way financial markets work. After law school, he worked at a private bank, and in 2016 (right around the time when Benedikt took his bar exam) Switzerland started to become a hub for FinTech, which allowed Benedikt to see a completely new side of the Swiss financial market. He then began getting into blockchain from a legal perspective and joined Lykke Corp, his first job in the blockchain crypto space. Benedikt comes from an entrepreneurial family: both his father and brothers built their own businesses because there was a strong urge in all of them to challenge the status quo and do things their way. After working as a lawyer for a few years, Benedikt himself felt compelled to get more involved in business decisions and launch his own company. In 2019, he founded FQX. Along with his co-founders, he shared a vision to build a platform and infrastructure for tokenized debt assets that are recognized as legal assets. FQX is a blockchain startup that uses eNotes to enable companies to issue debt securities via the blockchain directly to investors, changing the way companies secure funding. In this episode, he talks more about his experience launching a blockchain company in Switzerland. He also goes into more detail about the e-Note concept and discusses the difficulties he encountered while attempting to obtain funding for his blockchain start-up. Memorable Quotes: “I think the financial market is one of the most complex things humans have created.” “Throwing a bunch of very smart people in a room doesn't guarantee you're going to build a great product.” If you would like to listen to another founder in the blockchain market, check out our conversation with Robert Lauko. On page 290 of the Artscroll biography of Rav Pam, it says that the Gemara (Yoma 35b) tells us that after 120 years we are going to be judged. People will have all kinds of excuses for why they couldn't do this or that, but the Gemara says that there are three great Rabbis that are going to obligate others. If someone says that he was too wealthy so he had no time to learn, we have Rabbi Eliezer ben Harsun, who was fabulously wealthy, and he obligates the rich. If someone says that he was very handsome and surrounded by beautiful people so he could not control his desires, Yosef HaSaddik, who resisted the enticement of Potiphar's wife, will obligate those people. If someone says that they were poor, and because of their poverty they could not learn, Hillel, who suffered extreme poverty and climbed on top of the roof on a snowy day and almost froze to death in order to learn Torah will obligate the poor. Rav Pam added to that Gemara, quoting the Elder of Novardok that there is a moment in every person's life, when he achieves something great, and that moment obligates him to maintain that level of achievement for the rest of his life. That means that you will obligate yourself. You might say that you can't be nice to people because that is not your style- but what about the one time that you were nice? That will obligate you, because you do know how to do it. Of course, this doesn't mean that we should avoid doing great things in order to avoid the obligation. God will see through that as well. What it means is that we have to do our best, and once we show ourselves what we are capable of, we have to live up to that. Rav Pam related that in his mind, he knew the moment in his life that obligated him for the rest of his life. It was when an elderly member of the Torah Vadaat minyan took ill and was hospitalized. Rav Pam was a Kohen, and so he could not visit the man, but he took the time to write him a letter. Although halachically, writing a letter does not fulfilled the mitzva of Bikur Holim, he wanted a man to know that he had been thinking of him. When man passed away shortly thereafter, Rav Pam was told that the man had kept the letter under his pillow, and whenever a visitor would enter his room, he would take out the letter and show them the letter that Rav Pam, the Rosh Yeshiva , had written to him. Rav Pam pointed out that a simple letter, which took only moments to write, had lifted the spirits of that sick man in a way that he had never imagined. This incident served as a lesson to him, of how seemingly insignificant acts, that take only a little bit of time, can result in great achievements. This obligated him to utilize such opportunities in the future. How Beautiful! Let us look back at our lives and see all the great moments, and B'Ezrat Hashem, keep it up. Have a wonderful day.

Obligate hibernators– can sleep regardless of access to food and temperatures. Ex. European Hedgehogs, mouse lemurs, various ground squirrels & butterflies Facultative hibernators– hibernate when it's too cold and the food supply decreased Ex. Black-tailed prairie dogs (Bears, squirrels, groundhogs, marmots, skunks, raccoons, opossums, dormice and bats are examples of mammals that hibernate.) Diapause – (Observed in all life stages of arthropods, like insects, common during egg and pupa stages. Marker by lower metabolic activity.) Aestivation – (desert plants & animals- low rainfall and high temps i.e. “dry heat”) Brumation- Dormancy in reptiles (only wake up to drink water and then go back to sleep) Can last 8 months. Triggered by decrease in daylight hours. (e.g. Snakes) Just like us, plants are deeply affected by temperature and sunlight. Plant growth slows as the day length slows, thus commencing the dormancy process. In today's show we discuss the idea of slowing down in the winter.  Instead of fighting these feelings of being a little more tired than usual, we invite you to get into the flow with Nature and slow down a bit…go dormant, as much as possible until spring when there is more sunlight and the days start to grow longer.  And… use this dormant time to look back at past failures and successes and plan for future outings and hikes and camping trips, and other adventures!

Plus Hari's still incorporeal, Gaal indulges in some casual vandalism, Salvor and Phara go on a trip, and Ethel the Aardvark goes quantity surveying. As usual, we talk about it. Most of us understand what it means to be a carnivore, as a human or an animal. Being an obligate carnivore, however, requires something completely different. If a cat does not get all its nutritional requirements met, it will become ill and die. Almost all cat food is processed, and that initiates a prolonged and gradual nutritional deficiency for cats.Understanding the protein requirements of cats has made Dr. Karl become a more serious carnivore for the sake of his health.A lesson from cats that can be applied universallyAlthough Dr. Karl always knew that cats are obligate carnivores, he did not truly understand what that meant until recently. One of his cats would only eat ground-up chicken meat, and she was not doing well. Now Dr. Karl understands that having only one kind of protein is wrong. It can get you into nutritional deficiency.Different kinds of proteinThere is a huge difference between muscle meat and organ meat. There is also a big difference between muscle meat from fish, muscle meat from beef, and muscle meat from chicken, pork, or lamb because all of those animals eat different things. A nutrient deficiencyIf you eat only muscle meat, you will have some kind of deficiency. So, if you are a carnivore, you are obligated to eat organ meats, like liver, too, because organ meat contains a vastly different nutrient profile to muscle meat. Obligate carnivoreAn obligate carnivore is an animal that has to eat other animals. A cat is an obligate carnivore, so there is no value in feeding cats vegetables.Amino acidsHumans need eight essential amino acids because our bodies are unable to produce them. Our bodies can make the other amino acids that we need, however. Obligate carnivores need eleven amino acids. Their bodies are unable to make amino acids in the same way that we, as humans, can.Feeding a cat a raw food dietIf you feed a cat a raw food diet it has to contain a variety of different kinds of meats to cover all the nutritional needs of the cat and prevent any deficiencies.Secondary carbohydratesIn the wild, obligate carnivores get secondary carbohydrates from eating the viscera of their prey which contains undigested carbohydrates.Cats cannot store fuelCats in the wild cannot store fuel as humans can, so they need to hunt daily. GluconeogenesisHumans can make glucose from the amino acids in protein. That is called gluconeogenesis. Cats need glucoseCats also need glucose. They produce it via gluconeogenesis from the protein they eat.Protein is the perfect foodIf you have nothing other than meat to eat, it will give you enough appropriate glucose to sustain your glucose activity for your red blood cells and nervous system.DiversifyDiversify your protein sources, or else you could self-induce nutritional deficiencies.Links and resources:PSMF 30-day courseCheck out the Keto Naturopath website, join our Facebook Group, and follow us on Pinterest!Subscribe to our YouTube Channel!Download our Free Keto Foods ListVisit our partnersBuy Keto Friendly Dry Farm WinesGet your KetoMoJo Here and test your ketones.

Pastor Randy Ruiz is a traveling evangelist who teaches on healing and prayer. This weekend, he joined us at River of Life Church to speak on the biblical truth of who God is and what He can do through the power of His Holy Spirit. We serve a God of patterns. In each of the scriptures below, we see that God PASSES BY."And she said to her husband, “Look now, I know that this is a holy man of God, who passes by us regularly." -2 Kings 4:9 (NKJ)"They told him, “Jesus of Nazareth is passing by.”- Luke 18:37 (ESV)"Jesus entered Jericho and was passing through." -Luke 19:1 (NIV)"He saw the disciples straining at the oars, because the wind was against them. Shortly before dawn he went out to them, walking on the lake. He was about to pass by them," -Mark 6:48 (NIV)"On the last and greatest day of the festival, Jesus stood and said in a loud voice, “Let anyone who is thirsty come to me and drink.” -John 7:37 (NIV)There is a Biblical Truth we're experiencing today - the obvious need does not obligate God to do a miracle. What makes the Holy Spirit stand still?1.) Our desperation2.) Our honesty with God3.) Our faithWhen you are desperate for Jesus, you are willing to do whatever it takes to catch His attention.Luke 19:1 - 9 (NIV) Story of Zacchaeus"Jesus entered Jericho and was passing through. A man was there by the name of Zacchaeus; he was a chief tax collector and was wealthy. He wanted to see who Jesus was, but because he was short he could not see over the crowd. So he ran ahead and climbed a sycamore-fig tree to see him, since Jesus was coming that way. When Jesus reached the spot, he looked up and said to him, “Zacchaeus, come down immediately. I must stay at your house today.” So he came down at once and welcomed him gladly. All the people saw this and began to mutter, “He has gone to be the guest of a sinner.” But Zacchaeus stood up and said to the Lord, “Look, Lord! Here and now I give half of my possessions to the poor, and if I have cheated anybody out of anything, I will pay back four times the amount.” Jesus said to him, “Today salvation has come to this house, because this man, too, is a son of Abraham. For the Son of Man came to seek and to save the lost.”Zacchaeus was DESPERATE for a move of the Holy Spirit, so he did something out of the ordinary to get the attention of Jesus. Luke 18: 35 - 43 (NIV) Story of Bartimaeus "As Jesus approached Jericho, a blind man was sitting by the roadside begging. When he heard the crowd going by, he asked what was happening. They told him, “Jesus of Nazareth is passing by.” He called out, “Jesus, Son of David, have mercy on me!” Those who led the way rebuked him and told him to be quiet, but he shouted all the more, “Son of David, have mercy on me!” Jesus stopped and ordered the man to be brought to him. When he came near, Jesus asked him, “What do you want me to do for you?” “Lord, I want to see,” he replied. Jesus said to him, “Receive your sight; your faith has healed you.” Immediately he received his sight and followed Jesus, praising God. When all the people saw it, they also praised God."Bartimaeus called out to God because he was desperate. He did something out of the ordinary to get the attention of Jesus. How desperate are you for the presence of the Holy Spirit to pour out into your life?

Gene Editing Reveals Obligate and Modulatory Components of the CO2 Receptor Complex in the Malaria Vector Mosquito, Anopheles coluzzii

Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2020.05.13.094995v1?rss=1 Authors: Liu, F., Ye, Z., Baker, A. P., Sun, H., Zwiebel, L. Abstract: The sensitivity to volatile carbon dioxide (CO2) produced by humans and other animals is a critical component in the host preference behaviors of the malaria vector mosquito Anopheles coluzzii. The molecular receptors responsible for the ability to sense CO2 are encoded by three putative gustatory receptor (Gr) genes (Gr22,23,24) which are expressed in a distinctive array of sensory neurons housed in maxillary palp capitate peg sensilla of An. coluzzii. Despite the identification of these components and subsequent studies, there is a paucity of understanding regarding the respective roles of these three GRs in the mosquito CO2 transduction process. To address this, we have used CRISPR/Cas9-based gene editing techniques combined with in vivo electrophysiological recordings to directly examine the role of Gr22,23,24 in detecting CO2 in An. coluzzii. These studies reveal that both Gr23 and Gr24 are absolutely required to maintain in vivo CO2 sensitivity while, in contrast, Gr22 knock out mutants are still able to respond to CO2 stimuli albeit with significantly weaker sensitivity. Our data supports a model in which Gr22 plays a modulatory role to enhance the functionality of Gr23/24 complexes that are responsible for CO2 sensitivity of mosquitoes. This title makes no sense. This was a super exciting epsiode, and Sun is finally boosted from and I've been saying for weeks that they need to do something about Sun. And it finally happens.Sun sucks up all the energy of this episode, and it's glorious. Of all the sensates, she's the one who has never really needed much assistance from anyone else, so it's nice to see everyone pitching in to get her out of a jam.Wolfgang has something vaguely interesting going on- he manages to lock eyes with another sensate, so I'm back on board with his storyline.Lito gets a new apartment, this time with Dani as a permanent roommate. I love this. There's some very beautiful Jonas time. But of course the guy who ruins it is named Todd. And we have another suicide mission from a sensate- seems you can't keep Mr. Whispers down.Super tense, and I can't wait for the next one!
