Abstract: Imaging of living synapses has relied for over two decades on the overexpression of synaptic proteins fused to fluorescent reporters. This strategy changes the stoichiometry of synaptic components and ultimately affects synapse physiology. To overcome these limitations, here we introduce a nanobody that binds the calcium sensor synaptotagmin-1 (NbSyt1). This nanobody functions in living neurons as an intrabody (iNbSyt1) and is minimally invasive, leaving synaptic transmission almost unaffected, as demonstrated by the crystal structure of the NbSyt1 bound to synaptotagmin-1 and by our physiological data. Its single-domain nature enables the generation of protein-based fluorescent reporters, as we showcase here by measuring spatially-localized presynaptic Ca2+ with an NbSyt1- jGCaMP8 chimera. Moreover, its small size makes the NbSyt1 ideal for various super-resolution imaging methods. Overall, NbSyt1 is a versatile binder that will enable imaging in cellular and molecular neuroscience at a higher precision than possible in the past, over multiple spatiotemporal scales. How to start running and reach the national top at an age at which other professional runners start retiring? In this episode, I talk to Ingrid Prigge, who has been 17 times Dutch Masters Champion over various distances from 10k to marathon. At age 35, shortly after giving birth to her second daughter, Ingrid wanted to get back in shape and started running. Coming from a background that was not too enthusiastic about running, she discovered her talent and passion for the sport. Talking about the past 20 years, Ingrid explains, how and why running became one of her big passions. We talk about her fast, and at times naive and bumpy way to running to the national top. On January 10th, 1883, the famed Newhall House hotel in Milwaukee, Wisconsin, caught fire. Made worse by a series of blunders by hotel staff and a poorly trained and equipped fire department, seventy five people would ultimately lose their lives in the horrific, raging inferno. My guest, historian Matthew J. Prigge, is author of “Damn the Old Tinderbox! Milwaukee’s Palace of the West and the Fire That Defined An Era”. Angela has lived in Hawaii her whole life, and we talk about her love for her home and her deep family roots there. We talk about how Angela’s grandparents moved to Hawaii from the Philippines, how her father and grandfather worked and lived on sugar plantations. We talk about Angela’s memories of the bakery her mom worked at in Kauai, and we talk about how her parents instilled in her, from a young age, an appreciation for the necessity and power of unions. We also discuss what it was like to witness firsthand the major changes that have happened to Hawaii’s economy, as farming and small businesses gave way to a tourism and hospitality industry that has come to dominate life on the islands. We talk about the ongoing struggles for workers who can hardly afford to live in Hawaii, which has the highest cost of living out of any state in the U.S. And we talk about how Angela, who is on the executive board of her union, UNITE HERE Local 5, and her coworkers are banding together to fight not only for better wages and fair treatment, but for a more dignified and enjoyable life—one that doesn’t completely revolve around work and bills. The names of the French New Wave have become staples for both French Cinema and beyond.
