In unserem Gespräch gehen wir auf die 6 (momentan testbaren) Varianten ein- was charakterisiert die einzelnen Varianten, was sind ihre Besonderheiten nicht nur symptomatisch sondern eben auch Fütterungs- und Trainingstechnisch. Hier fließt natürlich sehr viel eigene Erfahrung mit ein und natürlich kann man (gerade bei Mischformen) niemals verallgemeinern. Inga Wessling hat darauf, gelinde gesagt, keinen Bock mehr. Im Jahr 2022 sollte die Periode der Frau keine peinliche, unangenehme Angelegenheit mehr sein, die die Natur irgendwie im weiblichen Körper verbaut hat. Aus diesem Grund geht die TV-Autorin und Moderatorin das Thema frontal an und spricht Klartext. Und zwar in ihrem Audible-Podcast „Periodensystem“ - und auch in der heutigen Episode von „Das Ziel ist im Weg“. Frappierend offen und direkt befreit Inga das Thema von Stigmata, Vorurteilen und Fehlinformationen und klärt dabei in bester, blutigster Weise auf. Abstract: Immunopeptidomics refers to the science of investigating the composition and dynamics of peptides presented by major histocompatibility complex (MHC) class I and class II molecules using mass spectrometry (MS). Here, we aim to provide a technical report to any non-expert in the field wishing to establish and/or optimize an immunopeptidomic workflow with relatively limited computational knowledge and resources. To this end, we thoroughly describe step-by-step instructions to isolate MHC class I and II-associated peptides from various biological sources, including mouse and human biospecimens. Most notably, we created MhcVizPipe (MVP) (https://github.com/CaronLab/MhcVizPipe), a new and easy-to-use open-source software tool to rapidly assess the quality and the specific enrichment of immunopeptidomic datasets upon the establishment of new workflows. In fact, MVP enables intuitive visualization of multiple immunopeptidomic datasets upon testing sample preparation protocols and new antibodies for the isolation of MHC class I and II peptides. In addition, MVP enables the identification of unexpected binding motifs and facilitates the analysis of non-canonical MHC peptides. We anticipate that the experimental and bioinformatic resources provided herein will represent a great starting point for any non-expert and will therefore foster the accessibility and expansion of the field to ultimately boost its maturity and impact. The quarantine life has me making old school phone calls. One of those calls was to an old radio friend, John Wessling. He and I played a significant role in bringing a Rock station to Victoria, TX in the mid 90s. This might not sound like an epic tale in today's time of streaming music, but in 1993 the internet was in its infancy. The radio options in Victoria were country, adult contemporary and religious programming. We helped change that with a coup d'etat of KLUB 106.9 which is now known as ‘The Rock of Victoria'. After a dramatic exit from KLUB John went on to become a stand up comedian and podcast host. This is a fun episode and I hope you enjoy it!
