F., Buitelaar, J. K., the EU-AIMS LEAP group Abstract: Background: Voxel-based Morphometry (VBM) studies in Autism Spectrum Disorder (autism) have yielded diverging results. This might partly be attributed to structural alterations being associating with the combined influence of several regions rather than with a single region. Further, these structural covariation differences may relate to continuous measures of autism rather than with categorical case-control contrasts. The current study aimed to identify structural covariation alterations in autism, and assessed canonical correlations between brain covariation patterns and core autism symptoms. Methods: We studied 347 individuals with autism and 252 typically developing individuals, aged between 6 and 30 years, who have been deeply phenotyped in the Longitudinal European Autism Project (LEAP). All participants' VBM maps were decomposed into spatially independent components using Independent Component Analysis. A Generalized Linear Model (GLM) was used to examine case-control differences. Next, Canonical Correlation Analysis (CCA) was performed to separately explore the integrated effects between all the brain sources of gray matter variation and two sets of core autism symptoms. Results: GLM analyses showed significant case-control differences for two independent components. The first component was primarily associated with decreased density of bilateral insula, inferior frontal gyrus, orbitofrontal cortex, and increased density of caudate nucleus in the autism group relative to typically developing individuals. The second component was related to decreased densities of the bilateral amygdala, hippocampus, and parahippocampal gyrus in the autism group relative to typically developing individuals. The CCA results showed significant correlations between components that involved variation of thalamus, putamen, precentral gyrus, frontal, parietal, and occipital lobes, and the cerebellum, and repetitive, rigid and stereotyped behaviors and abnormal sensory behaviors in autism individuals. Conclusions: Covaried areas associated with autism diagnosis and/or symptoms are scattered across the whole brain and include the limbic system, basal ganglia, thalamus, cerebellum, precentral gyrus, and parts of the frontal, parietal, and occipital lobes. Some of these areas potentially subserve social-communicative behavior whereas others may underpin sensory processing and integration, and motor behavior. Copy rights belong to original authors. Efforts to unravel sex differences in the brain organization of autism have, however, been challenged by the limited availability of female data. Methods: We addressed this gap by using the largest sample of male and female autistic and neurotypical (NT) control individuals, to date (ABIDE; Autism: 362 males, 82 females; NT: 410 males, 166 females; 7-18 years). Discovery analyses examined main effects of diagnosis, sex and their interaction across five resting-state fMRI (R-fMRI) metrics. Secondary analyses assessed the robustness of primary results to different pre-processing approaches and their replicability in two independent samples, the EU-AIMS Longitudinal European Autism Project (LEAP) and the Gender Explorations of Neurogenetics and Development to Advance Autism Research (GENDAAR). Results: Discovery analyses in ABIDE revealed significant main effects across the intrinsic functional connectivity (iFC) of the posterior cingulate cortex, regional homogeneity and voxel-mirrored homotopic connectivity (VMHC) in several cortical and subcortical regions. Sex-by-diagnosis interactions were confined to VMHC in dorsolateral occipital cortex with reduced VMHC in females with autism. Findings were highly robust to different pre-processing steps and replicable in another sample. Specifically, the sex-by-diagnosis interaction replicated in the larger of the two replication samples - EU-AIMS LEAP. Conclusions: Results emphasize that atypical cross-hemispheric interactions are neurobiologically relevant to autism. Systematic assessments of the factors contributing to their replicability are needed and necessitate coordinated large-scale data collection across studies. Copy rights belong to original authors. SC and RRB severity can markedly differ within and between individuals and is underpinned by different neural circuitry and genetic mechanisms. Modeling SC-RRB balance could help identify how neural circuitry and genetic mechanisms map onto such phenotypic heterogeneity. Here we developed a phenotypic stratification model that makes highly accurate (96-98%) out-of-sample SC=RRB, SC>RRB, and RRB>SC subtype predictions. Applying this model to resting state fMRI data from the EU-AIMS LEAP dataset (n=509), we find replicable somatomotor-perisylvian hypoconnectivity in the SC>RRB subtype versus a typically-developing (TD) comparison group. In contrast, replicable motor-anterior salience hyperconnectivity is apparent in the SC=RRB subtype versus TD. Autism-associated genes affecting astrocytes, excitatory, and inhibitory neurons are highly expressed specifically within SC>RRB hypoconnected networks, but not SC=RRB hyperconnected networks. On December 4, 1956, a legendary jam session was held at rock and roll pioneer Sam Phillips’ Sun Records studio in Memphis, Tennessee. Johnny Cash, Jerry Lee Lewis, Carl Perkins, and Elvis Presley were labeled the “Million Dollar Quartet” by a local journalist and the moniker stuck to the recordings of the session released decades later. In 2006, Colin Escott and Floyd Matrux unleashed a highly fictionalized and time-compressed theatrical version of the event also titled Million Dollar Quartet. After several successful bay area productions, Santa Rosa’s 6th Street Playhouse gives the North Bay a chance to check out this popular jukebox musical now running through March 24. Jukebox musicals are usually comprised of a couple of dozen well-known songs connected by expositionary material and Million Dollar Quartet is no different. Sam Phillips (Benjamin Stowe) narrates the tale of the event, filling in the backstory and presenting the dramatic conflict (Will Johnny Cash sign a contract extension or fly the coop?) around which the music swirls. At a recording session for Carl Perkins (Jake Turner) with Jerry Lee Lewis (Nick Kenrick, also music director) on piano, who should happen to drop by but Elvis Presley and his girlfriend (played by Daniel Durston and Samantha Arden) and Johnny Cash (played by Steve Lasiter)! In no time, there’ll be a whole lot of shakin goin’ on as we’re treated to “Blue Suede Shoes”, “Folsom Prison Blues”, “That’s All Right”, “Great Balls of Fire” and 20 other classics. Director Michael Ray Wisely – who has played Phillips and directed this piece before - had the benefit of 6th Street expending significant coin on this production beginning with an impressive set (Conor Woods adapting Kelly James Tighe’s original scenic design) and imported talent. It’s not an easy show to cast as each performer must be a “triple threat” - an actor, a singer, and a musician. Kenrick reprises his TBA Award-winning performance as Jerry Lee Lewis and steals the show with his kinetic piano playing and entertaining characterization. Local performer Jake Turner manages to hold his own against Kenrick as Carl Perkins, and Durston and Lasiter do fine in capturing the essence of their characters while avoiding simple caricatures. They receive good musical support by locals Nick Ambrosino on drums and bassist Shovanny Delgado Carillo. Ignore the shaky musical history and often-pedestrian exposition that’s presented and you’ll find yourself enjoying a well-performed staged concert of some of rock and roll’s greatest hits. 'Million Dollar Quartet' runs Thursdays through Sundays through March 24 at 6th Street Playhouse in Santa Rosa. K., Oranje, B., Durston, S., the TACTICS consortium Abstract: Repetitive behavior is a core symptom of Autism Spectrum Disorder (ASD) and Obsessive-Compulsive Disorder (OCD), and has been associated with impairments in cognitive control. However, it is unclear how cognitive control and associated neural circuitry relate to the development of repetitive behavior in children with these disorders. In a multicenter, longitudinal study (TACTICS; Translational Adolescent and Childhood Therapeutic Interventions in Compulsive Syndromes), the development of cognitive control was assessed during late childhood using a longitudinal fMRI design with a modified stop-signal task in children with ASD or OCD, and typically developing (TD) children (baseline: N=122 (8-12y), follow-up: N=72 (10-14y), average interval: 1.2y). Stop-signal reaction time (SSRT) decreased over development, regardless of diagnosis. Repetitive behavior in children with ASD and OCD was not associated with performance on the stop-signal task. There were no whole-brain between-group differences in brain activity, but ROI-analyses showed increases in activity in right precentral gyrus over development for children with OCD. In sum, even though subtle differences were observed in the development of brain activity in children with OCD, the findings overall suggest that the development of cognitive control, as assessed by the stop signal task, is similar in children with and without ASD or OCD.
