Abstract: Pain often persists in patients with inflammatory diseases, even when the inflammation has subsided. The molecular mechanisms leading to this failure in resolution of inflammatory pain and the transition to chronic pain are poorly understood. Mitochondrial dysfunction in sensory neurons has been linked to chronic pain, but its role in resolution of inflammatory pain is unclear. Transient inflammation causes neuronal plasticity, called hyperalgesic priming, which impairs resolution of hyperalgesia induced by a subsequent inflammatory stimulus. We identified that hyperalgesic priming in mice caused disturbances in mitochondrial respiration, oxidative stress, and redox balance in dorsal root ganglia (DRG) neurons. Preventing these priming-induced disturbances restored resolution of inflammatory hyperalgesia. Concurrent with these mitochondrial and metabolic changes, the expression of ATPSc-KMT, a mitochondrial methyltransferase, was increased in DRG neurons in primed mice. ATPSc-KMT overexpression in DRG neurons of naive mice induced similar mitochondrial and metabolic changes as observed after priming, leading to failure in pain resolution. The boys have on guests Matt Mahalak and Josh Malecki. Matt was drafted by the Carolina Hurricanes in the 6th round and was the starting goalie for the Plymouth Whalers and the Kingston Frontenacs in the OHL. He went on to continue his career at the University of Prince Edward Island. Josh led the U16AAA Compuware team to a National Title and was one of the best goalies to come through that association. He went on to play for the Windsor Spitfires and the Barrie Colts in the OHL.
John Malecki is a woodworker and epoxier who started his YouTube channel in January, 2016 and had a big breakthrough just two years later when he posted a pair of live edge epoxy table videos within 5 days of each other. The second of these, Live Edge River Table Woodworking How To is still his most viewed video with 5.1 million views. Ever since then, the channel is about going big! Big epoxy, big slabs, and most recently a huge workshop. Before getting into YouTube, John was a football player who actually got to play a game in the NFL. Pittsburgh native, John Malecki grew up wanting one thing; to play in the NFL for the Steelers. After a star high school, and college football career, John reached his goal and played for the Steelers on the active roster for about 2 years in 2012. With his NFL dreams fulfilled, Malecki soon realized he wasn't prepared for the "real word" and life after professional football. He found a passion for wood working while doing some DIY house projects, and ran with that. Since 2016 John has had his own YouTube channel sharing his love for building and learning new skills. Fast forward to present, a now new Father, he and his wife Sara are raising their daughter and navigating with no play book for parenting. Allerdings gehört doch noch viel mehr dazu. Unter anderem ein feines Gespür für die Luft. Es gewinnt häufig nicht unbedingt der Pilot mit den –salopp gesagt – „dicksten Eiern“, sondern dem sensibelsten Hintern. Der hilft dabei, die Aufwinde und die tragenden Linien zwischen den Bärten zu erfühlen. Gehaltene Höhe ist beim Wettbewerbsfliegen ein Pfund, mit dem man als Pilot am meisten wuchern kann. Einer, der das meisterlich beherrscht ist Andreas Malecki – in der Szene allgemein als Pepe bekannt. Der heute 60-jährige fliegt seit 20 Jahren in der Weltelite der Gleitschirmflieger. Er ist mehrfacher Deutscher Meister, zuletzt 2020. In 2012 stand er auf Platz 1 der Weltrangliste. 2015 wurde er Weltmeister im Team mit der Deutschen Nationalmannschaft. FNMA, Freddie Mac and HUD have recently announced upcoming sales of non-performing and re-performing loan sales. Fannie Mae has three larger NPL pools totaling about $613 million in unpaid principal balance, the Community Impact Pools is $34 million in UPB and the re-performing loan pool totals $3billion in UPB. HUD announced a vacant NPL sale totaling $170 million in UPB and Freddie Mac announced a previously modified loan sale totaling $292 million in UPB. The NPL market continues to be driven by the government which feeds the secondary and tertiary markets. Positioning your firm and raising capital is critical to be able to participate. John Malecki - We got John's feedback on the antics from Vontaze Burfict and the Cincinnati Bengals again last Sunday vs. the Steelers. He talked about how the Bengals implode leads directly to Steeler victories. He also shared stories of some moments that he retaliated when he played in college at Pitt. Tony Collins - We went through our 5 Star Picks of the Week for Week 16 in the NFL season with Tony. Victor Green - We got his thoughts on the Jets situation including whether they should focus on developing Bryce Petty or Christian Hackenberg or if they should bring in a free agent QB over the off season. Victor shared his memory of his first pick 6 which came off Drew Bledsoe and the 90 yards pick 6 he had on his first game facing the Jets after they cut him and he went to New England. Paul Alexander - We talked with Paul about it being Ravens week in Pittsburgh and what that means and the Steelers need to develop another offensive threat besides Antonio Brown and Le'Veon Bell. I also got Paul's thoughts on why Mike Tomlins struggles with math and anything having to do with numbers. Gene Lang - We got Gene's insights into the frustrations that led to a postgame meltdown between the Broncos defense and offense last week. We also got his thoughts on his alma mater LSU and Offensive Coordinator Ed Orgeron taking over as Head Coach plus his thoughts on star players now deciding to sit out Bowl games.

John Malecki - John was recently featured in a Titleist advertisement during the US Open at Oakmont. We'll hear about how that came about plus his memories from from being in training camp with the Steelers. He played for the following teams in the NFL: Tennessee Titans, Cleveland Browns, Tampa Bay Bucs, Pittsburgh Steelers, and Washington Redskins.
