Abstract: Dense core vesicles (DCVs) and synaptic vesicles (SVs) are specialised secretory vesicles (SSVs) in neurons/neuroendocrine cells harbouring cargo whose abnormal release is associated with pathophysiology. Endoplasmic Reticulum (ER) stress and inter-organellar communication are also associated with disease biology. In pursuit of investigating the cell physiological consequences arising from the crosstalk of a stressed ER and DCVs, ER stress was modelled in PC12 neuroendocrine cells using Thapsigargin (Tg). DCV exocytosis was severely compromised in ER-stressed PC12 cells, reversed by Docosahexaenoic acid (DHA). Experiments with Tunicamycin(Tm), an independent ER stressor, yielded similar results. Concurrently, ER stress caused impaired DCV exocytosis also in INS-1 cells. Molecular analysis revealed blunted SNAP25 expression, potentially attributed to augmented levels of ATF4 (a well-known CREB inhibitor) and its transcriptional regulator CREB (also known to regulate key granulogenic players Chromogranin A, Secretogranin II). Our studies revealed severe defects in DCV exocytosis in ER-stressed cells for the first time, mediated by reduced levels of key 'exocytotic' and 'granulogenic' switches regulated via the CREB/ATF4/eIF2 axis. Abstract: Axonal transport is key to neuronal function. Efficient transport requires specific motor-cargo association in the soma, yet the mechanisms regulating this early step remain poorly understood. We found that EBP-1, the C. elegans ortholog of the canonical microtubule end binding protein EB1, promotes the specific association between kinesin-3/KIF1A/UNC-104 and Dense Core Vesicles (DCVs) prior to their axonal delivery. Using single-neuron, in vivo labelling of endogenous cargo and EBs, we observed reduced axonal abundance and reduced secretion of DCV cargo, but not other KIF1A/UNC-104 cargo, in ebp-1 mutants. This reduction could be traced back to fewer exit events from the cell body, where EBP-1 colocalized with the DCV sorting machinery at the trans Golgi, suggesting that this is the site of EBP-1 function. In addition to its microtubule binding CH domain, mammalian EB1 interacted with mammalian KIF1A in an EBH domain dependent manner, and expression of mammalian EB1 or the EBH domain was sufficient to rescue DCV transport in ebp-1 mutants. Our results suggest a model in which kinesin-3 binding and microtubule binding by EBP-1 cooperate to transiently enrich the motor near sites of DCV biogenesis to promote motor-cargo association. In support of this model, tethering either EBP-1 or a kinesin-3 KIF1A/UNC-104 interacting domain from an unrelated protein to the Golgi restored the axonal abundance of DCV proteins in ebp-1 mutants. These results uncover an unexpected role for a microtubule associated protein and provide insight into how specific kinesin-3 cargo are delivered to the axon. Copy rights belong to original authors. Abstract: Neuronal dense-core vesicles (DCVs) contain neuropeptides and much larger proteins such as the 70 kDa protease TPA, which contributes to long-term potentiation and synaptic growth. Rather than using full collapse exocytosis that is common in endocrine cells, DCVs at a native intact synapse, the Drosophila neuromuscular junction, release their contents via fusion pores formed by kiss and run exocytosis. Here fluorogen activating protein (FAP) imaging reveals the limited permeability range of synaptic DCV fusion pores and then shows that this constraint is circumvented by cAMP-induced extra fusions with dilating pores that result in DCV emptying. These Ca2+ independent full fusions require PKA-R2, a PKA phosphorylation site on the fusion clamp protein complexin and the acute presynaptic function of Rugose/Neurobeachin, a PKA-R2 anchor implicated in learning and autism. Therefore, localized Ca2+-independent signaling triggers the opening of dilating fusion pores to release large DCV cargo proteins that cannot pass through fusion pores that normally dominate Ca2+-dependent synaptic protein release. B., Toonen, R. F., Verhage, M. Abstract: Tomosyn is a large, non-canonical SNARE protein proposed to act as a competitive inhibitor of SNARE complex formation in vesicle exocytosis. In the brain, tomosyn inhibits fusion of synaptic vesicles (SVs), whereas its role in the fusion of neuropeptide-containing dense core vesicles (DCVs) is unknown. Here, we addressed this question using a new mouse model allowing conditional deletion of tomosyn (Stxbp5) and its paralogue tomosyn-2 (Stxbp5l), and an assay that detects DCV exocytosis with single vesicle resolution in primary hippocampal neurons. Surprisingly, loss of both tomosyns did not affect DCV exocytosis but resulted in a strong reduction of intracellular levels of many DCV cargos, most prominently brain-derived neurotrophic factor (BDNF), granin VGF and prohormone convertase PCSK1. Reduced levels of DCV cargos were paralleled by decreased DCV size and impaired mRNA expression of the corresponding genes. We conclude that tomosyns regulate neuropeptide and neurotrophin secretion via control of DCV cargo production, and not at the step of cargo release. Our findings suggest a differential effect of tomosyn on the two main secretory pathways in mammalian neurons and argues against a conserved role of tomosyn as competitive inhibitor of SNARE complex formation. Bryan has a bit of an industry nerd out with Ross Trethewey from “This Old House” and TE2 Engineering at IBS 2022 (the International Builders' Show). Ross's education and career have focused on mechanical engineering, especially with sustainable solutions. In building science, the key mindset is to think of the building as a system. Using that school of thought, Ross has developed building science and HVAC solutions that also consider indoor air quality and ventilation, such as hybrid VRF systems. Many of Ross's solutions take the best aspects of air-source and ground-source heat pumps and apply those to hydronics. Some exciting applications for those types of systems could include simultaneous heating and cooling as well as the integration of domestic hot water. Demand control ventilation has been used for a long time in the commercial world, but its possible use in residential applications is another exciting thing to consider. With proper control devices, DCV would give us the opportunity to control temperature, humidity, VOCs, carbon monoxide, carbon dioxide, and radon. In residential applications, DCV has to be a delicate balancing act, as bringing in too much outdoor air would require us to condition that air. High latent loads also present challenges to some of the ventilation solutions in development. Serviceability is another challenge to DCV usage in residential applications; whenever an innovative system is brought to the market, very few people will know how to fix and maintain those systems. One of the possible solutions is to create instruction manuals and give education similar to what already exists for package units. 3D models and animations also help make complicated systems easier to understand. Ross's presence on “This Old House” marks the third generation of Tretheweys on the show. Ross is excited to talk about building science and HVAC innovations and concepts while on the show. Heat pumps are also getting better, especially due to inverter-driven compressors, enhanced vapor injection, advanced control systems, and ECMs. Heat pumps are safer than gas-fired equipment, and we have made them work well in subzero temperatures (because we're nowhere near absolute zero). This December will see a meaningful change in how CAs are allowed to conduct Domain Control Validation (DCV) using the method known as https token or file authentication or agreed up on change to web site. This method will be removed as an option for "domain spaces" including wildcards and subdomains. Join our hosts as they explain how DCV works and how the rules are changing and why. Auch durch seine Auftritte bei Stefan Raab - zuerst bei TV Total; inklusive dews Verschenkens seines Pullunders, später beim Bundesvision Song Contest- stieg seine Bekanntheit. Für mich persönlich (Rewow) ist dieses Album in seiner Einzigaritigkeit ikonisch und verfolgt sowohl einen ironischen Gesamtcharakter, als auch die auf Realtalk, Gesellschaftskritik und Seitenhiebe gegen die Szene basierenden Konzeptidee des Wolfes im Schafspelz. Hannes Norberg. Freier Künstler aus Düsseldorf. Hannes Norberg wurde 1969 in Worms geboren, lebt und arbeitet in Düsseldorf. Er studierte an der Kunstakademie Düsseldorf bei Christian Megert und Gerhard Merz. Seine Arbeiten sind eigens für die Kamera im Studio konstruierte architektonische Modelle und Landschaften. Er hatte zahlreiche Ausstellungen und Arbeitsstipendien für Paris, New York, Florenz, Xiamen, São Paulo und Seoul. 2002 erhielt er für seine Arbeit den Villa Romana-Preis. Vom Aussterben bedroht? - Männerchöre Wo Jan Schumacher ist, ist Energie im Raum. Es darf gelacht werden, viel geredet und gesungen werden. Ganz so wie man sich Fasnachtssitzungen rund um Limburg vorstellen kann. Kein Wunder, dass der kleine Jan dort seine ersten Auftritte hatte. Ob es ein Glück ist, dass er später zu den Limburger Domsingknaben stieß und dann doch nicht Musiklehrer wurde, können wir nur erahnen. Gelacht haben wir aber viel und Spaß hatten wir auch. Und Anne fühlt sich zumindest vorsichtig ermutigt, doch vielleicht mal als hohe Tenöse bei einem Männerchor vorstellig zu werden. Jan Schumacher (*1980) ist seit 2015 Universitätsmusikdirektor der Goethe Universität in Frankfurt/Main und unterrichtet seit 2019 an der Akademie für Tonkunst in Darmstadt. Von 2011 bis 2016 war er Professor für Chorleitung an der Hochschule für Kirchenmusik in Rottenburg. Nach dem Studium der Schulmusik und der Germanistik in Mainz studierte Jan Dirigieren bei Wolfgang Schäfer in Frankfurt. Schon als Kind und Jugendlicher (Musikalische Ausbildung bei den Limburger Domsingknaben und im Landesjugendchor Rheinland-Pfalz) prägte ihn die Faszination des gemeinsamen Musizierens, die bis heute Movens für seine vielfältigen Tätigkeiten ist. Neben der Frankfurter Universitätsmusik und den Ensembles der Akademie für Tonkunst in Darmstadt ist Jan Leiter des Chores der TU Darmstadt und der Camerata Musica Limburg. Mit seinen Ensembles erarbeitet er ein breites Repertoire von Gregorianik bis zur Uraufführung oder Jazz, von sinfonischem Orchester bis Big Band und vokaler oder elektronischer Improvisation. Regelmäßige Aufnahmen für HR, SWR, ORF, RTÉ, EBU und Deutschlandradio sowie zahlreiche CD-Produktionen dokumentieren seine Arbeit. Camerata Musica Limburg ist darüber hinaus 1. Preisträger renommierter nationaler (Deutsches Chorfest, Deutscher Chorwettbewerb) und internationaler (Tours, Maasmechelen) Wettbewerbe und Gast bedeutender Musikfestivals (z.B. Schubertiade Schwarzenberg). Jan ist gern gesehener Gast in Jurys und als Leiter von Seminaren für Chöre, Orchester und Dirigenten im In- und Ausland. Der deutschsprachigen Chorszene ist er durch die Leitung zahlreicher Workshops und Kurse bei AMJ, DCV (chor.com), Pueri Cantores, ACV u.a., als Jurymitglied beim Schweizerischen und Deutschen Chorwettbewerb sowie Austria Cantat und als Herausgeber von über zehn Chorbüchern bei den wichtigsten Chormusik-Verlagen (Helbing, Carus, Bärenreiter, Peters) bekannt. Abstract: Long range fast axonal transport of neuropeptide-containing dense core vesicles (DCVs), endolysosomal organelles and presynaptic components is critical for maintaining the functionality of neurons. How the transport of DCVs is orchestrated remains an important unresolved question. The small GTPase Rab2 has previously been shown to mediate DCV biogenesis and endosome-lysosome fusion. Here we use the Drosophila model system to demonstrate that Rab2 also plays a critical role in bidirectional axonal transport of DCVs, endosomes and lysosomal organelles, most likely by controlling molecular motors. We further show that the lysosomal motility factor Arl8 is required as well for axonal transport of DCVs, but unlike Rab2 is also critical for DCV exit from cell bodies into axons. Our results uncover the mechanisms responsible for axonal transport of DCVs and reveal surprising parallels between the regulation of DCVs and lysosomal motility. Ein Chorbuch, in der sich rund 500 Jahre Chormusik aus Mitteldeutschland, geistliche und weltliche Friedensmusik sowie klassische und moderne Volksliedarrangements auf rund 300 Seiten wiederfinden. Das Chorbuch ist in Kooperation des DCV mit den drei mitteldeutschen Verbänden (Sächsischer Chorverband, Chorverband Sachsen-Anhalt, Chorverband Thüringen). Prof. Andreas Göpfert, der die Werke zusammengestellt hat, spricht im Interview mit Holger Frank Heimsch über das Buch, seine Entstehung und warum es über das Chorfest hinaus ein Buch für die Chorlandschaft sein wird.
