In the latest episode of the Next ReGeneration podcast, hosts Anina and Elena welcome Ian Needleman, Professor of Periodontology and Healthcare Research at UCL Eastman Dental Institute in London, UK. A passionate runner himself, Needleman has devoted substantial research to exploring the oral health of athletes and its impact on their performance and overall well-being.   Needleman shares fascinating insights from his assessments of athletes' oral health during the London 2012 Olympic Games. His studies consistently reveal that elite athletes frequently suffer from poor oral health, a phenomenon observed across both developing and developed nations. Although the exact mechanisms linking oral health to sports performance are not yet fully understood, inflammation seems to be a key factor. Needleman's research underscores that enhancing oral health can significantly boost athletic performance.

Katherine Needleman is a wonderful oboe player, who has been the principal oboist of the Baltimore Symphony since 2003. She's also active as a solist and chamber musician, and is on faculty at the Curtis Institute of Music. She is a champion of music of our time and you'll find her recordings and many projects on her website. To many listeners in the Classical Music world, she's probably better known for her Substack articles and posts on social media that address misogyny, sexual misconduct and assault, and the lack of diversity and equity in the Classical music world. In this conversation you'll hear about some of her activism and advocacy work , and different aspects of Katherine's life as a musician, including her work as a composer and educator, and mentors including Jennifer Higdon. “In that early stage, you really have to leverage your network and those around you who have done it before. Because they're only advisors, they can only go so far. It's kind of wild in that first year of having nothing and creating everything. You have to trust your instincts”, says Lauri.She shares her journey from growing up in a family of entrepreneurs through to joining Pie Insurance at its launch. Lauri gives insight in to her discussions with Co-Founder John Swigart and the vision for building the team in those early days. “The idea of making something much more accessible affordable and easier to understand for small business owners was a mission that I could rally around. But I shared with John, ‘Why don't you call me in six months to a year?'He said no for two reasons. He said he wanted to surround himself with a team that he trusted. The second reason was he and Dax (Co-Founders) were committed to getting the culture and the hiring right from the outset and they were willing to make the investment to do so.”Year one was spent establishing the core HR functions of the insurance business and recruiting from their networks. With 2 employees on day 1 in 2017, the business now has over 400 pie-oneers and has expanded from Workers' Comp to offer Commercial Auto Insurance, General Liability and Business Owners Insurance. “We wanted to make sure the individuals we were bringing in, understood the business, understood what it was like to join an early stage start up because it's not for everyone.”Lauri highlights how being a remote-first business impacts culture. “It's definitely a dichotomy a lot of companies face. On the one hand, being purely remote gives you access to much of the talent across the US. But it also has the challenge of building a culture.”“You have to have a fantastic on-boarding program. How you set up your new team members for success on day one, those first six months is everything. But it's important to have some live collaboration. We don't believe it should be 100% remote all the time.”Lauri discusses the investment in tech to facilitate communication as well as the complexities of supporting team members based across different States. She gives her advice to candidates coming to interview and shares her thoughts on best practice for fellow people leaders across the insurance industry. “You have to have a very thorough and tight talent process. You have to execute it similarly across the business. We do competency-based interviewing. We prep all of our interview panels before we interview and then we debrief together as a group. That ensures that everyone is on the same page going into and leaving the process.”To drive their insurance careers forward, Lauri encourages people to be integrative thinkers. “Something that's helped me be successful in my career is being an integrative thinker. It's not looking at a problem as ‘I've got path A or B' as the solution. It's considering all of the perspectives and all of the needs and ‘how do I solve for that that greater needs set'.Lauri shares the importance of working with an Executive Search firm that understands the business culture and calibrates with their recruitment needs. She concludes by sharing the value of mentoring and leveraging your insurance network. “I don't have all the answers and I'm not supposed to. I have points of view and perspectives but I hold them loosely. I try to take in other perspectives that will refine my views on things. Mrs Erika Needelman - Life CoachShe knew she would be young when her father died. She knew because she saw how much younger all of her friend's parents were as compared to her own. The young Erika grew up, married and had children. Her father, although elderly, was still alive and a loving grandparent. And then, one day, her father-in-law, who had been perfectly healthy, died. Just like that.She had to make sense of her elderly sick father still being alive, while her relatively young, healthy father-in-law was not. She had to support a grieving husband while she herself was also grieving. It was a hard time.One lesson she took to heart was to enjoy whatever time she had with her father. During the last bit of his life, she made sure to have him over and to visit him. She made sure that her children knew their Zaidy.Now that he has passed on, she misses her father. But she also feels so sad that her children have no Zaidies.How she aches for her father to continue shepping nachas.And as the only frum family members, she and her husband know it is up to them and their children to continue giving their Zaidies nachas up in Shamayim.  With that in mind, Mrs. Needelman and her husband continue to work on themselves to grow in Yiddishkeit and in yiras Shamayim. Because after all, her children do have Zaidies. If life is a garden, the writer, editor, and craftsperson Deborah Needleman certainly knows how to dig and cultivate it. Early in her career, she followed a nonlinear path in the media industry that was, for the better part of a decade, slow and steady—and then, upon launching the home design bible Domino in 2004, meteoric. Over the next dozen years, Needleman rose to become one of the magazine world's most in-demand editors, serving as the editor-in-chief of both WSJ. Magazine and T: The New York Times Style Magazine. Across this work, her deep appreciation for beauty, craft, gardening and nature, and unfussy, richly layered interiors shined through. By the end of 2016, though, Needleman decided that she had had enough of the whirlwind hustle of the magazine business. She sought a way to work with her hands, not just her head. So she slowed down—way down—and turned to the meditative acts of gardening and craft. She headed to the John C. Campbell Folk School in North Carolina, where she took an introductory broom-making course. Soon, she began producing a limited-edition “Garden Tea” of herbs such as chamomile, lemon balm, and mint. Everything suits me that suits your designs, O my universe. Nothing is too early or too late for me that is in your own good time. All is fruit for me that your seasons bring, O nature. All proceeds from you, all subsists in you, and to you all things return. (Meditations 4.23) The Roman Emperor Marcus Aurelius was a deeply spiritual person, and that fact comes across clearly in his Meditations. The American philosopher and religious scholar Jacob Needleman suggests the combination of “metaphysical vision, poetic genius, and the worldly realism of a ruler” within the Meditations of Marcus Aurelius inspire us and give us “honorable and realistic hope in our embattled lives.”[1] As a result, he argues, [The Meditations] deserves its unique place among the writings of the world's great spiritual philosophers.[2] Needleman elaborates on the spiritual impact Marcus' Meditations has on many of its readers, Marcus is seeking to experience from within himself the higher attention of what he calls the logos, or Universal Reason, so too the sensitive reader begins to listen for that same finer life within his own psyche. That is to say, the reader— you and I— is not simply given great ideas which he then feeds into his already formed opinions and rules of logic. The action of many of these meditations is far more serious than that, and far more interesting and spiritually practical. In a word, in such cases, in many of these meditations, we are being guided—without even necessarily knowing what to call it—we are being guided through a brief moment of inner work. We are being given a taste of what it means to step back in ourselves and develop an intentional relationship to our own mind.[3] The practice of Stoicism for Marcus was a means to find his place in the cosmos. He sought congruity with Nature and learned to love what fate had in store for him because he trusted in a providential cosmos. As David Hicks asserts, The Stoicism in which Marcus believed is rooted in an all-encompassing nature. Everything in man and in the universe, everything that is or ought to be, everything fated and everything free, and the logos or rational principle that informs everything and ties everything together and is ultimately identified with the deity – all of this is found in nature, and there is nothing else.[4] Stoicism provided Marcus with more than an abstract, intellectual understanding of human and cosmic Nature. The religious nature of Stoic philosophy differentiated it from other philosophies as well as organized religions. I covered the religious nature of Stoicism previously, so I will not address it fully here. However, it is important to understand that Stoicism was more than an intellectual endeavor for Marcus. Stoicism provided a rational form of spirituality for Marcus, and it offers the same for moderns. Stoicism is an alternative for those who consider themselves spiritual but not religious. If you're uncomfortable with the dogmas of organized religion and the nihilism of atheism, Stoicism offers a middle ground. Stoicism provides a spiritual way of life guided by reason. Stoicism relies on our innate connection with the rationality permeating the cosmos to guide our human reason toward a relationship with the divine that inspires us to develop our moral character and thereby experience true well-being. As Mark Forstater wrote in his insightful book The Spiritual Teachings of Marcus Aurelius: Until the time of Neoplatonism, Stoicism was the most highly spiritualised form of philosophy in ancient Greece and Rome. It was so spiritualised that it is as accurate to call it a religion as a philosophy.[5] As Henry Sedgewick points out in his biography of Marcus Aurelius, the traditional religions did not provide what he was looking for, Marcus was seeking a religion, as I have said, but there was none at hand that he could accept. WSB is an irreverent, often profane community of folks sharing stock tips, self-styled due diligence analysis, and screenshots of their brokerage accounts.Users of WSB noticed that shares in GameStop – a brick-and-mortar video game retailer which found itself on the wrong end of several trends (digitization of game purchasing, decline of physical retail, pandemic stress, etc.) – were sold short at a ratio of 140% of the float.Users started buying up positions in GameStop, driving up the price over 8,100 percent over a 1-year period (based on the closing price of $325.00 on January 29, 2020), causing a partial short squeeze.Meanwhile, several online brokerages – most notably Robinhood – limited trading activity on GameStop and several other stocks which the WSB community have been buying. Some assert a conspiracy between Robinhood and its order clearinghouse/counterparties drove the limits (which included temporarily blocking buy orders while allowing sell orders to go through, before simply placing limits on the number of shares which can be purchased). Robinhood asserts that it was simply trying to met requirements set by its clearinghouse and financial regulators.The fallout so far:On February 1, Robinhood announced it had raised an additional $3.4B in capital led by late-stage finch firm Ribbit Capital, with participation from existing investors including ICONIQ Capital, Andreessen Horowitz, Sequoia, Index Ventures, and NEA. That figure includes $1B in equity financing announced on January 29th. Abstract: Mitochondria are central to metabolism and their dysfunctions are associated with many diseases. Metabolic flux, the rate of turnover of molecules through a metabolic pathway, is one of the most important quantities in metabolism, but it remains a challenge to measure spatiotemporal variations in mitochondrial metabolic fluxes in living cells. Fluorescence lifetime imaging microscopy (FLIM) of NADH is a label-free technique that is widely used to characterize the metabolic state of mitochondria in vivo. However, the utility of this technique has been limited by the inability to relate FLIM measurement to the underlying metabolic activities in mitochondria. Here we show that, if properly interpreted, FLIM of NADH can be used to quantitatively measure the flux through a major mitochondrial metabolic pathway, the electron transport chain (ETC), in vivo with subcellular resolution. This result is based on the use of a coarse-grained NADH redox model, which we test in mouse oocytes subject to a wide variety of perturbations by comparing predicted fluxes to direct biochemical measurements and by self-consistency criterion. Using this method, we discovered a subcellular spatial gradient of mitochondrial metabolic flux in mouse oocytes. We showed that this subcellular variation in mitochondrial flux correlates with a corresponding subcellular variation in mitochondrial membrane potential. The developed model, and the resulting procedure for analyzing FLIM of NADH, are valid under nearly all circumstances of biological interest. Thus, this approach is a general procedure to measure metabolic fluxes dynamically in living cells, with subcellular resolution. Copy rights belong to original authors. Visit the link for more info

Emma shared pieces of her journey of becoming a mother. *Disclaimer* In this episode we do talk about child loss which can be triggering. “After becoming a mother myself, I found that there was a huge discrepancy in how I thought I was "supposed" to parent and how I actually felt drawn to parent. I had a career as a midwife that I had worked hard for and that was my passion in life, but after having my first son, the immense amount of time spent away from him felt wrong in the core of my belly.” During my return to work as a midwife, I experienced two complicated pregnancies. One which ended in a ruptured ectopic pregnancy requiring emergency surgery and the other was my son, Jonah, who was stillborn at 26 weeks gestation. Following my son's death, I was thrown into a world of immeasurable grief but it also shone the light on what was working in my life and what was not. Needleman joined the Twofivesix: Gaming and Marketing Podcast to talk about 2019’s major moments in video games, her predictions for 2020, and trends in interactive entertainment. Here are highlights from the conversation, lightly edited for clarity. The nature of loot boxes will change “As far as loot boxes themselves go, I think we're going to see less of them or they won’t give you some sort of competitive advantage. It'll just be cosmetic. And then I do think that will die down. But the broader issue surrounding loot boxes also ties to this concept of game addiction. And I don't think that's going to go away.” Apex Legends owes its success to Fortnite “What [the surprise Apex Legends launch] showed to me is that the influence streamers have on consumers is significant. But let's also keep in mind is that Apex Legends is free-to-play, just like Fortnite. And what made Fortnite so special is the fact that it is free-to-play and very easy to learn in short sessions. So, we've learned a lot about gaming behavior from Fortnite.” Streaming games isn’t going to take off anytime soon “It's gonna take a while for [game] streaming to become, anywhere near mainstream. Unlike other forms of media, video games are interactive, and if you're playing competitively, even like the slightest delay is going to be a problem. And we know that online multiplayer gaming is very popular. So in the near term, I can't see people giving up dedicated land devices for gaming. Streaming will be more like a bonus option that you would consider for certain situations, like when you're out of storage or when you're traveling. Ian Needleman is a Professor of Restorative Dentistry at the Centre of Oral Health and Performance, University College London (UCL) Eastman Dental Institute. Prof Needleman is on a mission to combat poor oral health in athletes.
