Given the widespread legalization of cannabis for medical and recreational uses, you'd think we'd have a better understanding of how it works. But ask a neuroscientist exactly how cannabinoid compounds like THC and CBD alter our perceptions or lead to potential medical benefits, and you'll soon learn just how little we know.We know that these molecules hijack an ancient signaling system in the brain called the "endocannabinoid" system (translation: the "cannabinoids within"). Jonathan Long is a biochemist who studies the chemicals produced during exercise. In Long's world, “you always start with molecules,” which offer “clean handles” to understanding complex processes. His lab has identified a chemical produced in the digestive tract during exercise that can make a person stop eating. Dr. Raphael Mechoulam was an organic chemist at the Hebrew University of Jerusalem, and the father of cannabis research. His team was the first to elucidate the structure of cannabidiol, or CBD. Abstract: Animal models indicate that the endocannabinoid system (ECS) plays a modulatory role in stress and reward processing, both crucially impaired in addictive disorders. Preclinical findings showed endocannabinoid-modulated synaptic plasticity in reward brain networks linked to the metabotropic-glutamate-5 receptor (mGluR5), contributing to drug-reinforcing effects and drug-seeking behavior. Although animal models postulate a link between ECS and cocaine addiction, human translational studies are lacking. Here, we tested previous preclinical findings by investigating plasma endocannabinoids (eCBs) anandamide (AEA), 2-arachidonoylglycerol (2-AG), and the related N-acylethanolamines (NAEs) palmitoylethanolamide (PEA) and oleoylethanolamide (OEA), including their interaction with cerebral mGluR5, in chronic cocaine users (CU). We compared basal plasma concentrations between chronic CU (N=103; 69 recreational CU and 34 dependent CU) and stimulant-naive healthy controls (N=92). Follow-up basal eCB/NAE plasma levels after 12 months were used for reliability and stability check (CU: N=33; controls: N=43). In an additional analysis using 11C-ABP688 positron emission tomography (PET) in a male subsample (CU: N=18; controls: N=16), we investigated the relationships between eCBs/NAEs and mGluR5 density in the brain. We found higher 2-AG plasma levels in dependent CU compared to controls and recreational CU. 2-AG levels were stable over time across all groups. In the PET-subsample, a positive association between 2-AG and mGluR5 brain density only in CU was found. Our results corroborate animal findings suggesting an alteration of the ECS in cocaine dependence and an association between peripheral 2-AG levels and cerebral mGluR5 in humans. The incredibly balancing endocannabinoid system's benefits are being overshadowed by the stigma of marijuana and getting high. Unfortunately, most educators and medical providers are not fully educated or have written off medical marijuana as an ineffective medicine due to it's stigma. Sadly, what's being overlooked is that research has found life's stress depletes the endocannabinoids that provide calm and balancing within your body. There are many plants, not just just the cannabis plant utilizing this same pathway to help restore balance. Many are finding that using cannabis in combination with other plant medicine or on it's own is providing the sedative effects they desire. The Endocannabinoid System (or ECS for short) is a complex cell-signalling system in the body that is responsible for maintaining homeostasis (balance) in virtually every system in the body. The ECS was identified in 1992 by Professor Raphael Mechoulam and his researchers who were exploring exactly how the Cannabinoid, THC, works in the body. The Endocannabinoid System plays a role in your body's functions such as sleep, mood, appetite, memory, reproduction & fertility. There are three main parts to our Endocannabinoid System (ECS): Endocannabinoid molecules Cannabinoid receptors Enzymes Endocannabinoids are molecules made by your body. The 2 known Endocannabinoids are 2-AG & Anandamide. These help keep the body's internal functions running smoothly. Your body produces them as needed so it's hard to keep track if there's a deficiency. The Cannabinoid receptors around our body are called CB1 & CB2. CB1 receptors are mostly found in the central nervous system. CB2 receptors are mostly found in your peripheral nervous system, especially immune cells. Endocannabinoids can bind to either receptor. The effects that result depend on where the receptor is located and which endocannabinoid it binds to. There are two main enzymes: fatty acid amide hydrolase monoacylglycerol acid lipase These are responsible for creating endocannabinoids, transporting them to our receptors and breaking down endocannabinoids once they have performed their function. Research has linked the ECS to the following processes: Appetite and digestion Metabolism Chronic pain Inflammation Mood Learning and memory Motor control Sleep Cardiovascular system function Muscle formation Bone remodelling and growth Liver function Stress Reproductive system function Skin and nerve function Ways to give your Endocannabinoid System a boost. CBD & THC are the 2 most common Cannabinoids that people know about. THC being a psychoactive Cannabinoid and CBD being a non psychoactive Cannabinoid. Along with CBD & THC there are 481 other identifiable chemical components known to exist in the cannabis plant, including terpenes. Terpenes are aromatic compounds found in many plants, though many people commonly associate them with cannabis because cannabis plants contain high concentrations of them. For CBD to be legal in the UK it must be derived from hemp, contain less than 0.2% THC and any products must clearly state the amount of CBD in mg on the packaging. If a product doesn't show this, DON'T spend your money on it. There are 3 different types of CBD that you may see on the market. Isolate - This is CBD isolated from the other compounds found in the cannabis plant. Full Spectrum - This is CBD along with the other cannabinoids found in the cannabis plant. Broad Spectrum - This is CBD with the other compounds but with THC removed. You may see these types of CBD in all kinds of different products such as... Pure oil Tinctures Creams Balms Bath Bombs Soap Haircare Skincare E-liquids Tea & Coffee Chocolate Lollipops Soft drinks and many, many more. The list is endless. In order to make these products, CBD & other cannabinoids must be separated from the plant matter. This can be done in a few different ways with one of the most common being the C02 extraction method. CO2 extraction is a process that uses pressurized carbon dioxide to pull the desired phytochemicals from a plant. In the world of cannabis, the process pulls all of the essential cannabinoids, terpene oils and waxes out of the plant material. How does CBD Work? CBD works alongside our endocannabinoid system to aid better health and wellness. It's long been accepted that the edible experience and “the munchies” go hand in hand. But one edible is aiming to change that. In this episode, Arthur Jaffe joins Jonathan to discuss a new type of gummy hitting the market that actually curbs your hunger. Arthur is the CEO of ECS Brands, a company that's in the business of providing unique whole-plant extracts to brands, manufacturers, and processors around the world. On the pod, Arthur talks about developing the Wana Optimals Fit Gummy in partnership with Wana Brands, as well as the positive results people have been seeing with the product. He also gets into some of the ingredients in the edible, notably THCV, and emphasizes the need to focus on the endocannabinoid system in the edible creation process.
Health care has been a constant presence in Matt Parks's life for much longer than most people. His mother is an ER nurse. His older brother was born with a rare condition and was often hospitalized as a child. And as a college baseball player, Matt started taking nonsteroidal anti-inflammatory drugs (NSAIDs) to cope with a shoulder injury. He became dependent on the medications and started to experience migraines. He also developed depression when his parents divorced, which he still struggles with. By 2019, having been Vice President and Sales Manager at Williams Medical Supply Inc. for over a decade, Matt was a veteran of the medical supply industry. Conversations with people in his field who had looked into cannabidiol — better known as CBD — and customers who had used it, convinced him that the majority of CBD products were ineffective. This inspired him to see if he could come up with something better. He had three main areas of improvement. He wanted to create a hemp-derived topical CBD cream that he could verify did not contain tetrahydrocannabinol (THC), the main psychoactive compound in cannabis. He also wanted to build a company that was directly accessible to customers who had questions, and a brand that didn't traffic in the usual hype. Nick talks to Saoirse O'Sullivan, PhD, a physiologist specializing in the physiological effects of cannabinoids. Dr. O'Sullivan ran a lab at the University of Nottingham for many years before becoming a consultant and Vice President of Translational Research at Artello Biosciences. Nick and Saoirse discussed some of the physiological effects of cannabinoids in the body, including endogenous cannabinoids that our own bodies produce, and plant cannabinoids associated with the Cannabis plant (e.g. THC, CBD, THCV, and CBG). They cover a number areas of physiology and systems in the body, including inflammation, brain injury, the blood-brain barrier, gut health, and the cardiovascular system. Dr. Greg Gerdeman, PhD, is a pioneer in researching the endocannabinoid system's physiology. In this episode, he describes cannabinoids as the 'first responders' of herbal medicine. Greg researched the ECS for over 20 years, and talks about cannabis as a gateway to understanding the roots of our biological evolution and how we can learn to change our health's narrative with this knowledge. He clears up over-simplifications of the ECS, which he sees as a complex conductor of a ‘symphony of biological interaction.' Common misinformation is spread unwittingly through popular demand for sound-byte explanations. As a Ph.D. student, he discovered that endocannabinoids act as "retrograde messengers" to regulate synaptic plasticity in the brain, findings that have become foundational to a modern understanding of the ECS and the neuroprotective actions of cannabinoids. He later co-directed research on exercise-mediated endocannabinoid signaling, revealing new insights into the "runner's high" and its evolutionary roles in exercise's neurological benefits. As a college professor, his lab focused on the ancient evolutionary origins of cannabinoid receptors. Upon co-founding a medical cannabis company in Florida, he directed scientific activities at every level of the supply chain, ranging from integrative pest management to product development to physician education. He is Chief Scientific Officer of UCANN and a voluntary Assistant Professor at the USF College of Pharmacy. The cannabis that most people have access to isn't what it could be. A big part of that is a result of the way the plant has been bred to have high THC levels for the recreational market over the years."For the most part, around the world, we're still dealing with cannabis that is mostly high THC and high myrcene, which is gonna be very sedating, producing what we call colloquially couch lock, where the person feels immobilized," Dr. Ethan Russo explains on The Cannabis Enigma podcast. "So that might be fine for the person that's trying to get to sleep, but it's not at all good for the person that might need to work, or study, and function well in the process, and particularly for chronic pain conditions."Science has made great advances in identifying what chemical properties of a given cannabis chemovar would be beneficial for various diseases and treatments, "but that's a far cry from saying that they would be able to access a chemical variety of cannabis that would be appropriate for their treatment," Russo explains."We really haven't seen the capabilities of cannabis properly harnessed at this point," he adds.Dr. Russo, one of the premier cannabis researchers who is responsible for the theory of clinical endocannabinoid deficiency and has done a great deal of work on proving and harnessing the entourage effect, also discussed different approaches to dosing, and why developing a tolerance can actually be a good thing."The beauty of cannabis is, even though one gets accustomed to the psychoactive effects, the benefits on whatever you're treating remain. In other words, if we have a chronic pain patient and they get benefit from using cannabis, as long as that condition is stable — it's not getting worse — we don't see dose escalation over time, and in fact, there are many people, who have taken cannabis therapeutically for decades that are using the same dose."Dr. Russo will be speaking at the Whole Plant Expo.In the second part of the episode, Dustin McDonald of Americans for Safe Access interviews Sue Lewtin, a medical cannabis patient treating lyme disease, about her journey with the plant.
