T., Auluck, P. K., Gibbs, J. R., Marenco, S., Cookson, M. R. Abstract: Age is a major common risk factor underlying neurodegenerative diseases, including Alzheimer's disease, Parkinson's disease, and amyotrophic lateral sclerosis. Previous studies reported that chronological age correlates with differential gene expression across different brain regions. However, prior datasets have not disambiguated whether expression associations with age are due to changes in cell numbers and/or gene expression per cell. In this study, we leveraged single nucleus RNA-sequencing (snRNAseq) to examine changes in cell proportions and transcriptomes in four different brain regions, each from 12 donors aged 20-30 years (young) or 60-85 years (old). We sampled 155,192 nuclei from two cortical regions (entorhinal cortex and middle temporal gyrus) and two subcortical regions (putamen and subventricular zone) relevant to neurodegenerative diseases or the proliferative niche. We found no changes in cellular composition of different brain regions with healthy aging. Surprisingly, we did find that each brain region has a distinct aging signature, with only minor overlap in differentially associated genes across regions. Moreover, each cell type shows distinct age-associated expression changes, including loss of protein synthesis genes in cortical inhibitory neurons, axonogenesis genes in excitatory neurons and oligodendrocyte precursor cells, enhanced gliosis markers in astrocytes and disease-associated markers in microglia, and genes critical for neuron-glia communication. Importantly, we find cell type-specific enrichments of age associations with genes nominated by Alzheimer's disease and Parkinson's disease genome-wide association studies (GWAS), such as apolipoprotein E (APOE), and leucine-rich repeat kinase 2 (LRRK2) in microglia that are independent of overall expression levels across cell types. We present this data as a new resource which highlights, first, region- and cell type-specific transcriptomic changes in healthy aging that may contribute to selective vulnerability and, second, provide context for testing GWAS-nominated disease risk genes in relevant subtypes and developing more targeted therapeutic strategies.

Our history dates back to 1925s and grandfather Michele's dream of making wine from his vineyard in the heart of the Bagnario Valley, Strevi, an area especially suitable to viticulture. His son Giuseppe then began the Company's evolution, gradually acquiring the best land of the Monferrato and choosing the most typical grape varieties to plant. In 1956, he built his winery in the center of the village of Strevi, to be near the train station so that he could send his wines directly to his customers.A man with deep roots, he was able to convey his passion to the entire family.Today his daughters , Michela, with her husband Giovanni Costa, Patrizia, and Doretta carry on “the family dream ” with his same enthusiasm and dedication. They personally supervise and control every step of the production process, all the way from grape to glass.Andrea Costa, Michela and Giovanni's first son, joined the company in 2014. Topics: Tom’s background growing in a small town in Northern Mississippi and the story of how he got a late start as a young musician. Some of the people who influenced his career and the wisdom he learned from those educators. The band programs at Meridien High School and the University of Southern Mississippi. Regionalism in school band programs and traditions around the country and the success of the major universities in the Southeast. Tom’s work with wind bands in Italy and the interviews in the ADA archives at the University of Maryland. Links: Thomas Fraschillo American Bandmasters Recordings Zaninelli: Roma Sacra Biography: Thomas V. Fraschillo, DMA, has served as a catalyst and mentor for members of the teaching profession. His high standards of performance have had a sustained influence on ensembles at every level, and his performances serve as models in both the professional or academic arena. Through his recordings, The Music of Luigi Zaninelli and The University of Southern Mississippi Wind Ensemble LIVE IN ITALY (recorded in Italy), and his publishing, conducting, and lecturing in the United States, Europe, Asia, and Australia, he is considered an international musician/scholar. His publications, translations from the original Italian of Alessandro Vessella’s Studi di strumentazione (Instrumentation Studies), and La Tecnica dell’orchestra contemporanea (The Technique of Contemporary Orchestration) by Alfredo Casella and Vittorio Mortari, both published by BMG Ricordi, Milan, have put his name in libraries of the entire English speaking world. As a writer/scholar Dr. Fraschillo is a contributing editor to the American Grove Dictionary, 2nd Edition and serves as a frequent conductor and lecturer in Italy. His most recent appearances in Italy have been with La Banda dell’esercito/The Italian Army Band in Rome. One of his most significant engagements with them signaled a very important milestone for the Italian Army in that Dr. Fraschillo was the first American-born conductor to have been invited to appear in a public performance by what is considered Italy’s most prestigious military concert band. The concert with Dr. Fraschillo conducting was the opening concert of the International Festival in Spoleto, “The Festival of Two Worlds, Festival dei due mondi.” His appearance was enormously significant for conductors of bands in that the opening performance featured such international artists as Gian Carlo Menotti, the renowned composer, the Orchestra and Giuseppe Verdi Chorus of Milan, and the Italian actress, Claudia Cardinale. Finally, in Italy Dr. Fraschillo often serves as a member of the judging panel for many international festivals, most notably the Concorso Internazionale di Composizione “R. Marenco” di Novi Ligure, and the Concorso Internazionale di Composizione Originale per Banda di Corciano. Aside from Dr. Fraschillo’s work in Europe, he is often engaged throughout the continents of Asia, and Australia. The Melbourne, Australia, Summer Youth Music Program has invited Dr. Fraschillo to be their guest conductor for their summer session for five years. In addition the Australian Band and Orchestra Directors Association has invited him to lecture and to adjudicate at their large ensemble festivals. Dr. Fraschillo’s other work in the Pacific Rim includes having served as clinician and guest conductor of the Central Armed Forces Band in Singapore and as conductor of the Singapore All-College Band sponsored by the Wind Band Association of Singapore. In 2009 and 2010 Dr. Fraschillo served as Artistic Director for the weeklong Winter Band Festival at Disney, Hong Kong. Dr. Fraschillo has recently been recognized by election to the National Band Association’s Hall of Fame of Distinguished Conductors, a distinct honor that recognizes his lengthy career as a public school, university, and professional conductor. Dr. Fraschillo devoted a significant amount of his career to the education of young people in Mississippi. For example his ten-year tenure at Meridian High School was highlighted by a performance at the Midwest Clinic in Chicago, the nation’s oldest and most prestigious music event for wind and string educators. The invitation was only the second to have been given to a band from Mississippi. Dr. Fraschillo’s Meridian students obtained successes not before reached, for he taught and helped place the first African-American students in the Mississippi All-State Band. Not only were they among the first minority members, but they were also the first African-Americans to attain the very highest positions in the group. Dr. Fraschillo has attained a significant level in the realm of international leadership in that he serves as a past president of the prestigious American Bandmasters Association, past president of the world’s largest organization for band directors, the National Band Association, and President of the CBDNA Southern Division.

This month’s Grape Gab is all about Albarossa. We were fortunate to have a conversation earlier this month with Andrea Costa from the Marenco family winery in Piemonte, Italy. He was one of the first handful of producers to experiment with it, and he generously shares some of his experiences with us. We also tried a traditional May Wine – in July, because that’s how we roll – and have shoutouts and announcements to wrap up this week’s fun. Fiona Joy poured her heart and soul into Signature – Synchronicity, a New Age album submitted to the 59th Grammy Awards for consideration. You can listen on: http://www.thebcompany.com/fionajoy-fyc.html. Or a Soundcloud link https://soundcloud.com/user-260052746 From Fiona I'm very proud of this album about fairy-tales told from an adult perspective - it has taken me a long time to be able to find a little 'naïve' in some of these heart-felt issues. I’m a full-time composer, recording and touring musician and just returned from my third tour of China. Signature - Synchronicity is my 11th album with Little Hartley Music and I’m also an audiophile who records in hi-res formats with Blue Coast Records (content partners with Sony). I have been an ARIA Finalist (Australia), 8 x New Age Music Award Winner, a 5x IMA Finalist, and I’ve been Runner Up or Finalist in the International Acoustic Music Awards (IAMAs) over the last three years. On this album, I believe I’ve worked with some of the finest session musicians in the world including Tony Levin, Jeff Haynes, Eugene Friesen and legendary Producer and guitarist Will Ackerman (Founder, Windham Hill Records), who says: “One of the brightest lights in contemporary instrumental music, Fiona Joy is poised to move into stardom.” - Will Ackerman, Founder of Windham Hill Records Australian Pianist, Producer & Singer Fiona Joy is enjoying the success of her recent release Signature Synchronicity after returning from her third major tour of China, recording a new album in the US, and featuring in several Australian concerts with her Blue Dream Ensemble at the Joan Sutherland Performing Arts Centre and the Sydney Women's International Jazz Festival. In China Fiona Joy is nick-named The Piano Angel, not just for her romantic, melodic songs and lush arrangements, but also for her gracious and mesmerizingly ballet-like performance style in concert. A prolific composer, she has always been interested in creating music that evokes images, emotions and stories. The album so far has won two catetories in the Independent Music Awards, a Finalist in the International Acoustic Music Awards, Hollywood Music in Media Awards and the USA Songwriting Competition. Her recording schedule for 2017 includes Blue Coast Records solo piano release (SACD/CD/DSD) Into the Mist and FLOW - the debut album by Fiona Joy, Lawrence Blatt, Jeff Oster & Will Ackerman. FLOW will be a groundbreaking release by a New Age group fof successful artists inspired by the iconic music of Windham Hill. It follows that her upbringing was filled with music. She recalls that her father embraced Andrew Lloyd Webber's music, playing Jesus in the Tamworth Musical Society production of Jesus Christ Superstar. Her classical piano training is evident in her thematic and melodic compositions. Ravel's “Bolero” emerged as an early influence, she says, adding, "I loved it, I played it over and over and deconstructed every part. George Winston, Prokofiev and Mendelssohn also intrigued me.” In 2015 Fiona contributed to a GRAMMY-winning album and traversed three continents, performing solo on a grand tour of China, entertaining in the U.S. at the ZMR Awards and at the audiophile Mecca, T.H.E. Show, closing out the year in Australia at the Sydney Women's International Jazz Festival with her renowned Blue Dream Ensemble. Once home, she attracted the national TV show ‘Lateline’, which ran a feature lifestyle piece on her life and music. (https://youtu.be/6RCTtmkwdSI) In 2014, a version of Fiona’s song “Grace” was chosen for a GRAMMY®-winning album that also reached #1 on the Billboard charts, and in 2013, her evocative piano and ethereal vocals earned her the global broadcaster’s choice and honor as the Best Instrumental Piano Album for600 Years in a Moment at the ZMR Awards, whose airplay charts she also graced at #1 for several months with Signature Solo. Fiona Joy also placed as Runner Up for Best Instrumental Song in the International Acoustic Music Awards, nominated alongside Pete Seeger for Best LIVE Performance Album in the Independent Music Awards, and she was an ARIA Finalist in Australia in 2008. As a forward-looking artist, Fiona is interested in pioneering new musical concepts and recording techniques to better serve her highly-interactive fan base. She frequently appears at audiophile conferences, and is signed to Blue Coast Records for high-fidelity recordings; owner/producer Cookie Marenco is considered and leader in the field, partnering with Sony Music to deliver hi-resolution music to discerning audiophile consumers. Marenco says, "Fiona's melodic solo piano and masterful performances are the perfect music to debut the kind of clarity double DSD audio brings to the home listener. Her music is a cross of George Winston's simple melodies and Ludovico Einaudi's musical sophistication." Fiona's hard work and conceptual vision for the Signature Series is now coming to fruition in Signature Synchronicity, produced in collaboration with Will Ackerman's team in full orchestral form. Ackerman, founder of both Windham Hill Records and Imaginary Road Studios, embraced and encouraged Fiona’s music years ago, stating, “Fiona’s Blue Dream album was the most ambitious project of my entire career and resulted in one of the most remarkable collaborations this genre has ever known. Blue Dream is unique and I’m as proud of it as anything I’ve ever worked on in my 35-year career of GRAMMY® Awards and Gold/Platinum records.” As a collaborator, Fiona has worked with Will Ackerman, Lawrence Blatt, T-Bone Wolk (Hall & Oates), Tony Levin (Peter Gabriel, Dire Straits), Jeff Haynes (Pat Metheny), Eugene Friesen (Paul Winter Consort), Heather Rankin, Charlie Bisharat (Shadowfax), Rebecca Daniel (ACO), Jeff Oster and many others. In 2014, she and Ackerman co-produced the successful debut album of Jennifer DeFrayne, who won Best New Artist in the 2015 ZMR Awards for her album By A Wire. Music writer Michael Diamond, recently reviewed one of Fiona’s live concerts in the U.S., writing “What I appreciated most was being able to listen and watch Fiona at the piano. The passion and elegance in her performance was breathtaking, as her flawless technique combined with pure emotional expression.” When not recording or touring, Fiona is busy directing the entrepreneurial side of her music business, an effort which is crucial to the success of every independent artist. She manages her growing record label, Little Hartley Music, and promotes Fiona Joy Jewellery, her new line offering the upscale Swarovsky crystal-adorned bracelets and foot jewellery that she wears while performing barefooted in concert. When not on the road, Fiona escapes to her peaceful home in a small rural village in Australia. Part two of my chat with bay area Producer Cookie Marenco. I've learned to just hit RECORD as soon as I start talking to someone, which I have to admit was a very good idea in this case. With this tour, we get even more insight into Cookie's approach to music, analog recording, and her engineering intern program.
In "Songs and Stories" #82, we talked to Houston Jones about their latest release and why they love working with bay area producer Cookie Marenco. In these next two episodes, we talk with Cookie herself. She describes how she was an early advocate of many digital recording advancement, and why these days she records exclusively on two-inch analog tape.
