Are you interested in how multifunctional urban spaces can contribute to increasing the quality of urban life in dense cities? Summary of the article titled Multifunctional Urban Spaces – a solution to increase the quality of urban life in dense cities from 2020 by Atieh Ghafouri and Christiane Weber, published the Manzar journal. Since we are investigating the future of cities, I thought it would be interesting to see how urban spaces can be designed and managed to promote social inclusion, community engagement, and sustainable development in the face of rapid urbanization and increasing density. The article presents the potential of these spaces to provide a range of social, cultural, and recreational amenities that meet the diverse needs of urban residents. As the most important things, I would like to highlight 3 aspects: The main goal is to improve the quality of life in densely populated cities and this can be achieved through efficiency but we must be able to create spaces for new functions – we must create multifunctional spaces. A multifunctional space could be identified with these two characteristics: allow different functions, actors and different users to use it and fulfill the different roles a space might have in line with the environmental, economical and social sustainability. When multifunctional cells are put together with a comprehensive vision, the benefits are much more significant than the sum of the advantages each cell could achieve individually. Abstract: As urban population increases, cities face a dilemma on one hand, the horizontal growth causes devastating effects on the environment, and on the other hand, vertical growth leads to a decrease in the quality of life of citizens and causes a variety of mental illnesses. This dilemma has left city managers with a complex puzzle: a city that must be able to accommodate more people with minimal interference ensuring their optimum quality of life. This research presents the idea of multifunctional urban space design as a solution to the mentioned problem. The definition of multifunctional spaces in this research has been investigated using its definition in agriculture and environmental studies. The hypothesis of this research is based on the fact that the comprehensive definition can take a new perspective on urban design, improving the functional performance of existing urban spaces and thus reducing the need to build new facilities while maintaining the quality of life in dense cities. The research method is qualitative and based on bibliographic studies. First, the notion of functional performance and multifunctional space definition are examined in disciplines that produce a more specific interpretation of the subject, and then the definition of concepts is extended to urban design and urban space, highlighted and discussed. Is the future of product development within the supplement industry heading towards more condition-specific or multifunctional formulation approaches? This juxtaposition in supplement industry product development approaches has been on mind for a while now...and there are so many considerations at play here coming from both the brand-level and supply side of the supplement industry, but also with how it all interacts to consumer behavior. We know that ingredient advances are enabling formulators to create products that can address a broader cluster of health conditions. At the same time, supplement brands are finding opportunities in smarter, more targeted products that appeal to specific subsets of consumers. While condition-specific is not a new approach in the supplement industry, it has gotten more prominent in the last few years though because of the overwhelming product proliferation within the supplement industry. Alternatively, we would be ignorant if we didn't believe a subset of consumers appreciate convenient multifunctional products that can meet several wellness goals. But there's also some other attributes at play in this condition-specific versus multifunctional debate. Though there's no central “best” approach to attack the supplement market, I'll explore this debate by looking at purchase criteria like price, personalization, marketing, and sales channel strategy. To further add depth to this content, I also included snippets of a recent conversation I had with Richard Wang, the CEO of ingredient company NuLiv Science, at the SupplySide West tradeshow. However, there is a dearth of tools for simultaneous recording and localized receptor modulation in large animal models. We address this limitation by translating multifunctional fiber-based neurotechnology previously only available for rodent studies to enable cortical and subcortical neural modulation in macaques. We record single unit and local field potential activity before, during, and after intracranial GABA infusions in the premotor cortex and putamen. We apply state-space models to characterize changes in neural activity and investigate how neural activity evoked by a working memory task varies in the presence of local inhibition. The recordings provide detailed insight into the electrophysiological effect of neurotransmitter receptor modulation in both cortical and subcortical structures in an awake, behaving macaque. By the year 2050, it’s estimated that there will be as much plastic in the ocean as there are fish. The industrial fishing industry actually generates a staggering 10% of this waste in the form of abandoned fish nets and fishing gear, approximately 640,000 tons each year. It’s estimated that 46% of the plastic in the Great Pacific Garbage Patch comes from fishing gear alone. Luckily, I’m talking to someone today who is tackling the problem of ocean plastic in a really unique way. Hannah Tomita is the founder and designer of Kaira Active. Kaira Active makes all of their products out of recycled fishnets and fishing gear! Hannah grew up in Hawaii and moved to California for college. After stumbling into her passion for fashion while a senior, she decided to sew her own swimwear to avoid spending hundreds of dollars on a swimsuit from a store. She taught herself how to make her own swimsuits by watching YouTube videos and reading articles online. After building a portfolio of her work, Hannah landed a job as an assistant designer at a swimwear consultancy where she worked with several high-profile international brands. I love hearing people who use their unique passions to solve problems in really unique ways, and I can’t wait for you to hear how Hannah is making a difference for the environment with her business. 5:20 - The Hannah 101 Growing up in Hawaii, Hannah visited the ocean at least a couple times a week. Experiences in nature were very accessible, and she spent her childhood exploring the island of Oahu. Hannah studied industrial design in college at Arizona State University. Hannah started sewing swimsuits and learned how to make designs from patterns and online tutorials. It was sharing her designs online that ultimately led Hannah to her first job as an assistant fashion designer at a consultancy. She worked with big clients like Urban Outfitters, Tommy Bahama, True Religion and more. Hannah immersed herself in fashion and while she loved it, she quickly realized that the industry was riddled with fast fashion and little to no concern for sustainability. No one was diving deep into thinking through designs and materials used. A few years after working in the fashion industry, Hannah realized the need for companies that thought through how their practices would affect the world. That’s when she started Kaira Active. When she was a student, Hannah loved designer swimwear, but couldn’t afford designer swimsuits on a college student budget. Missing the beach while studying in Arizona and not being able to afford the swimsuits she liked led her to start playing around with sewing her own swimwear. 8:40 – Kaira Active What began as a curiosity and hobby turned into a unique fashion brand that recycles industrial fishing nets and turns them into functional, beautiful active wear. Sadly, there are a lot of ways these nets end up polluting the ocean. Sometimes fisherman will illegally dump their nets to avoid getting caught. In other situations, nets are lost due to bad weather or getting snagged. There’s currently no good system in place to regulate the situation. When Hannah first started working at the swimwear consultancy, a vendor came in from Italy selling fabric made out of recycled fish nets. At the time, sustainability wasn’t being paid attention to very much, but seeing the fabric made Hannah excited about merging her love of nature and her very active lifestyle. Hannah was also looking for a way to get the most out of her activewear. Her fitness and outdoor hobbies are eclectic, and she wanted a way to use one outfit for both the yoga studio and surfing. 16:26 – Fishing Nets to Fabrics to Clothing Kaira Active partners with a nonprofit organization called Healthy Seas that sends divers to pull nets out of the ocean. Since industrial fishing nets are made of Nylon, the nets are taken to a facility and chemically regenerated back into their pure form of Nylon and made into ECONYL. ECONYL is the technology that forms the Nylon into a thread which is then sewn in with Spandex to make fabric for Kaira Active clothing. Kaira Active is also committed to reducing their waste, energy, emission, and water consumption. They partner only with manufactures that focus on sustainability, their electricity is derived from renewable sources, and they have adopted a water system aimed at reducing polluting agents before it is sent to the municipal water treatment plant. Actin is one of the most abundant proteins in eukaryotic cells and regulation of the microfilament system is crucial for a wide range of cellular functions including cell shape, cell motility, cell division and membrane dynamics. The aim of this thesis was (1) to gain a better understanding of the function of distinct actin binding domains in the regulation of the actin cytoskeleton and (2) to elucidate the role of actin variants. WH2 domains (WH2, Wiskott-Aldrich syndrome protein homology 2) are ubiquitous multifunctional regulators of actin dynamics. The protein Spire contains four central WH2 domains A-B-C-D with about 20 amino acids each and the cyclase-associated protein CAP2 contains only one WH2 domain. Under certain conditions, they can (1) nucleate actin polymerization, (2) disintegrate actin filaments and (3) sequester actin monomers. Here, the influence of selected Drosophila melanogaster Spire-WH2 and Mus musculus CAP2-WH2 domain constructs on actin dynamics was tested in vitro. To act as a filament nucleator, at least two WH2 domains are required, and nucleation of actin polymerization was only observed at substoichiometric concentrations of WH2 domains over actin. At higher concentrations, the sequestering activity of WH2 domains takes over. Preformed and purified SpireWH2-actin complexes act as extremely efficient nuclei for actin polymerization, even at superstoichiometric WH2 concentrations, under which free WH2 domains would sequester actin. All analyzed constructs, including these with only a single WH2 domain, sequester actin as well as they can disrupt filaments. This latter and most peculiar behavior of WH2 domains was observed in fluorometric, viscometric and TIRF assays. The WH2 domains seem to have such a high affinity for actin that they can forcefully sequester monomers even from filaments and filament bundles, thus breaking the whole structures. Taken together, the data clearly show that SpireWH2-actin complexes are the intermediates that account for the observed nucleating activity, whereas free WH2 domains can disrupt filaments and filament bundles within seconds, again underlining the intrinsic versatility of this regulator of actin dynamics. These data have been confirmed by crystallography in collaboration with the groups of Prof. Dr. Tad Holak and Prof. Dr. Robert Huber (Martinsried, Germany). Besides the well-studied conventional actins many organisms harbor actin variants with unknown function. The model organism Dictyostelium discoideum comprises an actinome of a total of 41 actins, actin isoforms and actin-related proteins. Among them is filactin, a highly conserved actin with an elongated N-terminus. The 105 kDa protein has a distinct domain organization and homologs of this protein are present in other Dictyosteliidae and in some pathogenic Entamoebae. Here, the functions of filactin were studied in vivo and in vitro. Immunofluorescence studies in D. discoideum localize endogenous and GFP-filactin in the cytoplasm at vesicle-like structures and in cortical regions of the cell. A most peculiar behavior is the stress-induced appearance of full length filactin in nuclear actin rods. To perform in vitro analyses recombinant filactin was expressed in Sf9 cells. Fluorescence studies with the filactin actin domain suggest that it interferes with actin polymerization by sequestering G-actin or even capping filaments. Gel filtration assays propose a tetrameric structure of full length filactin. Protein interaction studies suggest that filactin is involved in the ESCRT (endosomal sorting complexes required for transport) pathway which is responsible for multivesicular body formation. The data on filactin suggest that only the conventional actins are the backbone for the microfilamentous system whereas less related actin isoforms have highly specific and perhaps cytoskeleton-independent subcellular functions.

Inhibitors of coagulation factors from blood-feeding animals display a wide variety of structural motifs and inhibition mechanisms. We have isolated a novel inhibitor from the cattle tick Boophilus microplus, one of the most widespread parasites of farm animals. The inhibitor, which we have termed boophilin, has been cloned and overexpressed in Escherichia coli. Mature boophilin is composed of two canonical Kunitz-type domains, and inhibits not only the major procoagulant enzyme, thrombin, but in addition, and by contrast to all other previously characterised natural thrombin inhibitors, significantly interferes with the proteolytic activity of other serine proteinases such as trypsin and plasmin. The crystal structure of the bovine alpha-thrombin.boophilin complex, refined at 2.35 A resolution reveals a non-canonical binding mode to the proteinase. The N-terminal region of the mature inhibitor, Q16-R17-N18, binds in a parallel manner across the active site of the proteinase, with the guanidinium group of R17 anchored in the S(1) pocket, while the C-terminal Kunitz domain is negatively charged and docks into the basic exosite I of thrombin. This binding mode resembles the previously characterised thrombin inhibitor, ornithodorin which, unlike boophilin, is composed of two distorted Kunitz modules. Unexpectedly, both boophilin domains adopt markedly different orientations when compared to those of ornithodorin, in its complex with thrombin. The N-terminal boophilin domain rotates 9 degrees and is displaced by 6 A, while the C-terminal domain rotates almost 6 degrees accompanied by a 3 A displacement. The reactive-site loop of the N-terminal Kunitz domain of boophilin with its P(1) residue, K31, is fully solvent exposed and could thus bind a second trypsin-like proteinase without sterical restraints. This finding explains the formation of a ternary thrombin.boophilin.trypsin complex, and suggests a mechanism for prothrombinase inhibition in vivo.
