He first entered a lab at age 12 to study the chemical degradation of aspartame in diet sodas over extended storage periods at room temperature. At 14, he joined a research team in the Nutritional Sciences Department at Rutgers University to study adipocyte metabolism in adipocyte fatty acid binding protein (A-FABP) knockout mice (aP2-/-) after short-term high-fat feeding. His work contributed to the discovery of a functional compensation by the keratinocyte-FABP. From there, Chris went on to join a research team working on a NASA-funded project developing biologically based nano-robots at Rutgers in the Department of Mechanical and Aerospace Engineering. Rather than pursuing the conventional path after graduating from Cornell University with a BA in Economics, Chris blazed his own trail and pursued his passion for stand-up comedy on the New York circuit. His quick wit and even quicker ability to handle audiences made him a regular on the New York comedy scene. He has toured nationally at the Improv opening for some of the most popular and well-known acts. Chris has also headlined internationally and performed in front of large stadium crowds.

Abstract: Mutations of the adaptor protein-2 sigma subunit (AP2S1) gene which encodes AP2-sigma2, a component of the ubiquitous AP2 heterotetrameric complex involved in endosomal trafficking of the calcium-sensing receptor (CaSR), cause familial hypocalciuric hypercalcemia type 3 (FHH3). FHH3 patients have heterozygous AP2S1 missense Arg15 mutations (p.Arg15Cys, p.Arg15His or p.Arg15Leu) with marked hypercalcemia and occasional hypophosphatemia and osteomalacia. To further characterise the phenotypic spectrum and calcitropic pathophysiology of FHH3, we used CRISPR/Cas9 genome editing to generate mice harboring the AP2S1 p.Arg15Leu mutation, which causes the most severe FHH3 phenotype. Heterozygous (Ap2s1+/L15) mice were viable, and had marked hypercalcemia, hypermagnesemia, hypophosphatemia, and increased plasma concentrations of parathyroid hormone, fibroblast growth factor 23 and alkaline phosphatase activity, but normal pro-collagen type 1 N-terminal pro-peptide and 1,25 dihydroxyvitamin D. Homozygous (Ap2s1L15/L15) mice invariably died perinatally. The AP2S1 p.Arg15Leu mutation impaired protein-protein interactions between AP2-sigma2 and the other AP2 subunits, and the CaSR. Cinacalcet, a CaSR allosteric activator, ameliorated the hypercalcemia and elevated PTH concentrations, but not the diminished AP2-sigma2-CaSR interaction. Thus, our studies have established a mouse model with a germline loss-of-function AP2S1 mutation that is representative for FHH3 in humans, and demonstrated that cinacalcet corrects the abnormalities of plasma calcium and PTH. Abstract: alpha-Synuclein (a-Syn) is a protein implicated in the pathogenesis of Parkinson s disease (PD). It is an intrinsically disordered protein that binds acidic phospholipids. Growing evidence supports a role for a-Syn in membrane trafficking, including, mechanisms of endocytosis and exocytosis, although the exact role of a-Syn in these mechanisms is currently unclear. Here we have investigated the role of a-Syn in membrane trafficking through its association with acidic phosphoinositides (PIPs), such as phosphatidylinositol 4,5-bisphosphate (PI4,5P2) and phosphatidylinositol 3,4-bisphosphate (PI3,4P2). Our results show that a-Syn colocalizes with PIP2 and the phosphorylated active form of the clathrin adaptor AP2 at clathrin-coated pits. Using endocytosis of transferrin, an indicator of clathrin mediated endocytosis (CME), we find that a-Syn involvement in endocytosis is specifically mediated through PI4,5P2 levels. We further show that the rate of synaptic vesicle (SV) endocytosis is differentially affected by a-Syn mutations. In accord with their effects on PI4,5P2 levels at the plasma membrane, the PD associated E46K and A53T mutations further enhance SV endocytosis. However, neither A30P mutation, nor Lysine to Glutamic acid substitutions at the KTKEGV repeat domain of a-Syn, that interfere with phospholipid binding, affect SV endocytosis. Abstract: Recent genetic evidence revealed that endocytic pathway plays a major role in Parkinsons disease (PD) risk. However, the molecular mechanism of how endocytic defects contribute to dopaminergic neurodegeneration in PD is poorly understood. Here we report that LRRK2, the mutations of which are the most genetic causes of PD, binds to and phosphorylates AP2M1, the core component of endocytosis that has been recently implicated in PD risk. Our study revealed that abnormal AP2M1 phosphorylation cycle, regulated either by knockout or overexpression of LRRK2, cause endocytic defects. Our study also uncovered a novel tissue-specific regulation of AP2M1 phosphorylation by LRRK2. Further, we found that LRRK2 phosphorylation on AP2M1 mediates LRRK2-induced neuronal toxicity both in vitro in neuronal cultures and in vivo in Drosophila dopamine neurons. Importantly, AP2M1 phosphorylation levels are elevated in patient fibroblasts of both LRRK2-associated PD and sporadic PD, suggesting the clinical relevance of our finding in PD. Cytotoxic T cells use Flower power In order to efficiently kill multiple target cells, cytotoxic T lymphocytes must endocytose and recycle cytotoxic granule membrane components from the immunological synapse. Chang et al. reveal that a protein called Flower facilitates granule endocytosis in a calcium-dependent manner. This biosights episode presents the paper by Chang et al. from the February 5th, 2018, issue of the Journal of Cell Biology and includes an interview with one of the paper's senior authors, Jens Rettig (Saarland University, Saarbrücken, Germany). Är det dags att starta en snällhetsrevolution bland kvinnor i svenskt näringsliv? Det är ett av samtalsämnena i EY:s senaste podd med Eva Halvarsson, vd på Andra AP-fonden ("AP2"). Eva pratar om betydelsen av att vara snäll kvinnor emellan och inom näringslivet. "Vi skulle vinna på att vara lite snällare. Vara öppnare för att se varandras styrkor och mer förlåtande för svagheter. Ingen är perfekt", menar Eva. Eva har varit vd i 11 år på AP2, en av fem buffertfonder i det svenska pensionssystemet. Under den tiden har det hänt otroligt mycket på kapitalmarknaden, men Eva menar att synen på vd-posten inte hänger riktigt med. Hon träffar fortfarande personer som utgår från att någon av hennes manliga kollegor är vd istället. Eva uttrycker att hennes ledarskap utmärks av prestigelöshet och tydliga värderingar. I vardagen på AP2 är hon omgiven av experter inom olika områden, områden där hon själv inte har egen expertis. För att lyckas behöver man våga erkänna att man inte kan allt själv, vara överens om en gemensam kultur samtidigt som det är viktigt att inse att alla medarbetare är olika och därför behöver olika ledarskap.

I'm also only recently become a CSM player last week with WoM and Thousand Sons release (army already being built and getting some paint). However, I am a Horus Heresy nut and if any of you have listened to this podcast before (Combat Phase podcast) you'll know how I love me some heresy. If you are looking for new rules for Heresy models from FW or plastic GW HH products then I'm afraid to tell you that is not here. As I've said before, no new models accompany this or the Codex: Imperial Agents books this week or next, however, in 2017, so few weeks away…we should have quite a good first quarter (at the very least). That said, as a Heresy player I'm impressed with most of the flavoring given to these legions. Some like Alpha Legion or Death Guard (my 30K army) or even World Eaters are pretty sweet enemy opinion. But you decide for yourself. The episode of Combat Phase (www.combatphase.com) with this review is coming soon but in the meantime for all who are chomping at the bit, let's begin... Book contents Most of the book is data sheets and even more is creating the 9 legions to give you themey armies, special rules, artifacts, warlord traits, objective markers, expanded Nurgle and Slaanesh lore for DG & EC respectively.This means, like Tzeencth, now 6 + 1 primaries power. As with Wrath of Magnus, you can make any CSM army into a specific legion given certain modifications, then the army gains additional special rules and when an army is composed of at lest 1 core and 1 auxiliary formation you get a formation with extra benefits. If you warlord is from this legion detachment it becomes your primary detachment and all in the primary detachment gain the benefits. Just with recent supplements an example is given when building a legion force where a gamer has a collection of models and the example walks through how he/she would separate models into a primary detachment with the bulk and a HQ & 2+ troops at least to for a CAD which gains objective secured. The book opens with some fluff for the Long War and a page with killer artwork for each of the 9 legions presented here. We see the usual inclusion of high quality art and photography of models engaged in combat we're used to from GW. This is a supplement to codex Chaos Space Marines so any new datasheets here replace what is in CSM. New wargear is on pages 135-136. The new rules vehicle squadrons from Traitor's Hate are included where Chaos vindicators and predators can be taken as a squadron. I think especially the vindicator one is best, and would be quite fluffy in an Iron Warriors list. Very fluff choices, all around in my opinion. Legions hating their old Heresy enemies, how they score, relics and traits but especially primary detachment rules. Vindicators: 120 base/for 2nd & 3rd models (minimum cost: 360 pts) - gains Linebreaker Bombardment: Instead of firing all 3, select 1 vindicator to fire the demolisher cannon which now becomes 24”, S10, AP2, Ordinance but uses the 10” Apocalyptic Blast template and Ignores cover :) Predators: 75 base/ for 2nd & 3rd (minimum cost 225) if all 3 predators are on field they gain monster hunter and tank hunter. General layout formations reflect the fighting style of each legion, taking into account how steeped in Chaos they may be. So without further ado Legions- every legion has 6 exclusive warlord traits, artifacts, tactical objectives, rules for CSM in their legion army, and if its detachment is the primary detachment allows them to re-roll legion warlord trait. Amazing art, a stunning new line of models which I honestly did not expect. I can't say with any certainty on this, the day of pre-release in Nov., how the Traitor's Legion book (resale Dec 3) will address TS but I have faith in Tzeentch that it will at the worst mirror this and at the best, grant options for TS fans/players of 30K & 40K—but that's just my educated guess. ;) OK, so how does this work? Kickass models—stunning and a wider selection than I could have imagined—accompany new paints to build your TS collection. Demons do NOT have any new model kit releases at this time. However, blue horrors and brimstone horrors (they're so darn cute!) are available in Warhammer Quest: Silver Tower (WHQ) where you may want to look at your local GW or FLGS or online to buy, trade, or make dark pacts to acquire some. Though not confirmed, I am under the impression early in 2017 we'll see these models boxed to be used in 40K & AoS. The Tzaangor kit has a nice selection of bits; don't bother trying to mix WHQ Tzaangors into your 40K army, imo, just get the 40K kit and you can use those for AoS. Rubric marines make a gorgeous kit. Datasheets have units of 5 (4 + 1 aspiring sorcerer as infantry character) so the box can make the asp sorcerer + 9 marines, equipped with inferno boltguns or warp flamers. Soulreaper cannon and icon of flame…pretty. Oh, and a diverse CSM transfer sheet for other legions. Nice kit. That's all I have for models. Making an army These books are incredibly clear and perhaps that's future-proofing in addition to an evolution of the design studio. You need CSM codex for TS and Demon codex for the Tzeentch demon army here. The TS are vicious psykers and that comes through quite clear in this WoMagnus supplement! TS: This book contains new data sheets for Ahriman and Rubric Marines which replace their listings in the current CSM codex. The TS theme army is a Grand Coven (GC) which is made up of formations. Psykers get new and better powers and the CSM sorcerers can draw from 4 new disciplines: Sinistrum, Heretech, Ectomancy, Geomortis. We get 6 new objectives for TS focusing on psyker objectives mostly, or number 9. Also 3 cool fluffy altars of war. TS get 6 new relics, 6 new warlord traits (One gives eternal warrior!). CSM models can become TS with certain Tzeentchy condition to basically, ya know, make it work. Special rules: detachment from CSM can become TS meeting rules (this book), then all units in a TS detachment or formation get 3 special rules (hells yeah, imo!) 1. if unit w/vet of long war (all TS except Tzaangors) affected by a blessing then invuln save better by 1 until start of your next psyker phase. 2. Units as above reroll hits every round of close combat vs. Space Wolves but also SW get hatred vs. TS 3. Rubricae becomes troops instead of elites -Units from the formation can be in both a formation and detachment. You need your warlord in a GC as primary detachment to give the benefits to that whole primary detachment. Radial glial cells are a widespread non-neuronal cell type in the developing central nervous system (CNS) of all vertebrates. In the cortex, distinct subsets of radial glial cells coexist that are either multipotent or specified towards the generation of neurons or glial cells (Malatesta et al., 2000). Radial glial cells in the cerebral cortex are also the source of a second type of neurogenic progenitors, called basal progenitors. However, whether the generation of basal progenitors occurs in a stochastic manner or whether a specific lineage of radial glial cells is specified towards the generation of these progenitors has not been previously known. To identify functionally distinct lineages of cortical radial glial cells, I developed a new strategy using fluorescence-activated cell sorting (FACS) to isolate them and study their progeny. I isolated radial glial cells by FACS from a transgenic mouse line in which green fluorescent protein (GFP) expression is under the control of the human GFAP promoter. Strikingly, GFP intensity was correlated with cell fate. Selective enrichment of cells with a higher GFP intensity separated a largely non-neurogenic from a neurogenic (low GFP-intensity) subsets of radial glial cells. Notable differences on the progeny of these distinct sets of radial glia were found. The neurogenic radial glial cells subset generated neurons directly and those that are largely non-neurogenic also gave rise to a small proportion of Tbr2-positive basal progenitors that are then neurogenic. Thus, this last subset comprises an indirect neurogenic population of radial glial cells present in the developing cortex. Microarray analysis of these distinct sets of radial glial cells revealed profound differences in their gene expression. Genes related to gliogenesis, proliferation and cell-cycle regulation were expressed at higher levels in the largely non-neurogenic set of radial glia while genes related to neurogenesis, cell adhesion, neurotransmitter secretion and axon guidance were expressed mostly in the neurogenic subset. Moreover, the set of genes expressed at higher levels in the neurogenic radial glia was down-regulated at later stages (cortical radial glia at E18). Thus, this analysis reveals differences at the transcriptional level between direct neurogenic and largely non-neurogenic radial glial cells, supporting their intrinsic lineage differences. The functional analysis of a key fate determinant for neurogenesis from radial glia discovered in this transcriptome analysis will also be presented. This gene is the transcription factor AP2γ which was expressed at significantly higher levels in radial glial cells generating basal progenitors. AP2γ is restricted to the ventricular zone (VZ)/subventricular zone (SVZ) regions of the developing cerebral cortex in the entire nervous system and is also highly expressed in primate and human cortical progenitors. Its genetic deletion within the mouse cerebral cortex results in the molecular misspecification of basal progenitors with decreased levels of Tbr2 and Math3 expression, as well as their overproliferation associated with increased cell death specifically in the occipital cortex. This causes a reduction in upper layer neuron generation with intriguing functional defects in visual acuity. Gain-of-function studies also revealed the important role of AP2γ for the adequate specification and development of basal progenitors in the cerebral cortex, while apical progenitors were not affected by the loss- and gain-of-function of this transcription factor. Thus, I show for the first time the prospective isolation of distinct radial glia subtypes in the mouse developing cortex demonstrated at the molecular and functional level.
