Glycation Stress Promotes Arterial Stiffening and Is Reversed by a Natural Compound in Aging Mice

BUFFALO, NY — December 12, 2025 — A new #research paper was #published in Volume 17, Issue 11 of Aging-US on November 14, 2025, titled “Methylglyoxal-induced glycation stress promotes aortic stiffening: putative mechanistic roles of oxidative stress and cellular senescence.” The study was led by first authors Parminder Singh of the Buck Institute for Research on Aging and Ravinandan Venkatasubramanian of the University of Colorado Boulder, with senior contributions from corresponding authors Pankaj Kapahi (Buck Institute for Research on Aging) and Zachary S. Clayton (University of Colorado Boulder and University of Colorado Anschutz Medical Campus). The researchers investigated how methylglyoxal (MGO), a toxic byproduct that builds up in blood vessels with age or metabolic dysfunction like diabetes, contributes to artery stiffening. Their findings are especially important to aging and diabetes-related cardiovascular risk. Aortic stiffening, which reduces the flexibility of the body's largest artery, is a key predictor of cardiovascular disease in older adults. The research team used young and aged mice to study how MGO affects vascular health. In young mice, chronic exposure to MGO increased aortic stiffness by 21%. However, when treated with Gly-Low, a supplement containing natural compounds such as nicotinamide and alpha-lipoic acid, this stiffening was completely prevented. Gly-Low also reduced the buildup of MGO and its harmful byproducts, particularly MGH-1, in both blood and tissue. “Aortic stiffness was assessed in vivo via pulse wave velocity (PWV) and ex vivo through elastic modulus.” The research showed that MGO's damage goes beyond structural changes. It also caused the endothelial cells that line blood vessels to enter senescence, a state in which cells stop dividing and begin releasing inflammatory signals. This led to lower levels of nitric oxide, a molecule essential for blood vessel relaxation. In human vascular cells in lab culture, Gly-Low reversed these aging-like changes and restored nitric oxide production. In older mice, which naturally develop stiffer arteries, Gly-Low treatment during four months significantly reduced stiffness and lowered MGO and MGH-1 levels. This suggests that Gly-Low may help slow or even reverse vascular aging by reducing glycation stress. The study also identified the glyoxalase-1 pathway as a critical mechanism. This is a natural detox system that helps clear harmful molecules like MGO. Gly-Low appeared to boost this pathway. When the pathway was chemically blocked, Gly-Low's protective effects disappeared, confirming its role in the process. Overall, the findings highlight glycation stress as a modifiable contributor to vascular aging. The results suggest that natural compound-based therapies, like Gly-Low, may offer a potential strategy to protect arteries from age- and diabetes-related damage. How Growth Hormone Excess Accelerates Liver Aging via Glycation Stress

BUFFALO, NY — November 18, 2025 — A new #research paper was #published in Volume 17, Issue 10 of Aging-US on October 3, 2025, titled “Growth hormone excess drives liver aging via increased glycation stress.” In this study, led by first author Parminder Singh alongside with corresponding authors Pankaj Kapahi from the Buck Institute for Research on Aging and Andrzej Bartke from Southern Illinois University School of Medicine, researchers investigated how elevated growth hormone (GH) levels contribute to liver aging and dysfunction. They found that excess GH disrupts liver metabolism in ways that resemble aging-related liver damage. The study suggests that managing glycation stress may help prevent or treat liver diseases linked to abnormal hormone levels. Excess GH is known to cause different disorders, but its long-term impact on internal organs like the liver has remained unclear. To address this, researchers used a mouse model engineered to overproduce bovine GH and examined how chronic hormone exposure affects liver function over time. “Pathological conditions such as acromegaly or pituitary tumors result in elevated circulating GH levels, which have been implicated in a spectrum of metabolic disorders, potentially by regulating liver metabolism.” The team found that young mice with GH overexpression showed molecular and cellular patterns similar to those in naturally aged livers. In both groups, genes involved in metabolism were suppressed, while those linked to immune and inflammatory responses were activated. On one hand, the metabolic changes were associated with the buildup of advanced glycation end products, harmful compounds formed when sugars attach to proteins or fats without proper regulation. On the other hand, the immune and inflammatory changes reflected a process known as “inflammaging,” a form of chronic, low-grade inflammation commonly associated with aging. By revealing the overlap between hormone-driven and age-related liver dysfunction, the study provides new insight into how GH may accelerate aging processes. Importantly, the team showed that reducing glycation stress can reverse many of these negative effects. Mice treated with a compound that lowers glycation levels demonstrated improved liver health, reduced insulin resistance, and enhanced physical function. This intervention also corrected several abnormal genetic patterns caused by excess GH. The findings point to a potential therapeutic strategy for liver diseases associated with aging and hormonal imbalances. Overall, this research identifies glycation and its byproducts as key contributors to liver damage caused by excess GH. It suggests that targeting glycation could offer broad therapeutic benefits, not only for hormone-related conditions but also for supporting liver health during aging. All it takes is a small amount of oxidative stress to the eye's lens to develop a cataract. A healthy lens is composed of an organized protein called crystallin. When this tissue becomes damaged, it is not replaced. Glycation can damage the lens of the eye. It results from sugar connecting to a protein and then becoming stuck and unusable to the body. Steroids like Prednisone can also increase the risk of cataracts. Not all surgeries can eliminate cataracts, and they can come with complications. According to “The Cataract Cure” by Dr. Marios Kyriazis, there is a safe, effective remedy for cataracts. Carnosine is a unique compound that can penetrate the lens of the eye and help with cataracts. Carnosine is a potent antioxidant that can dissolve damaged proteins. It can also activate your own antioxidants. NAC (n-acetyl carnosine) eye drops are one of the best remedies for cataracts.In an experiment conducted at the Moscow Helmholtz Research Institute of Eye Diseases, scientists used NAC eyedrops at 1% concentration on rabbits. They found that NAC could reach the inner parts of the eye within 15 to 30 minutes. No side effects were reported during this experiment. After 6 months, researchers found that:•41% of people presented a significant reduction of clouding in the lens•90% showed a gradual improvement in vision•89% showed up to a 100% improvement in sensitivity to glare NAC for cataracts must be used for at least 3 to 5 months. This week I interview longevity queen, Dr Sandra Kaufmann the author of the Kaufmann Protocol and Aging Solutions. Dr Sandra is the head of department of Anesthesiology at  and just happens to be obsessed with longevity science. What started out as her own search to slow and reverse ageing turned into a comprehensive system that allows people to personalize their own regime to their particular set of problems or needs and goals. This ingenious system has revolutionized my approach and makes it so simple to construct a very personalized approach. There are innumerable things a person can read concerning aging, but at the most basic level, organisms age because their component cells age. We are all made of cells or cell products and as they falter, so do we. Because cell aging is multifactorial, the organization of these concepts is important. The Protocol organizes these various theories of cellular aging into seven categories or tenets. The information in this podcast discusses these concepts in the language of science. The following seven tenets encompass all of the presently known modes of cellular aging.  Based on this organizational scheme, Dr Kaufman then developed a strategy where aging is curtailed in every category.   The 7 Tenets are as follows: DNA alterations (telomere attrition, epigenetics) Mitochondrial failure Pathways (Sirtuins, AMP Kinase, mTOR) Quality control (DNA & Protein repair mechanisms and Autophagy) Immune and inflammatory systems Individual cell needs (Senescent, Stem cells) Waste management (Glycation, lipofuscin) Dr Kaufman explains her rating system for various longevity molecules and how to utilize these in your protocols. We also discuss many of the supplements and adjuvants that Dr Kaufmann recommends for each of these tenets and we look at other cutting edge regenerative modalities like Exosomes.  Stem cells are fantastic in terms of longevity, however there are limitations. Exogenous stem cells are currently not available in this country. Meanwhile, autologous stem cells are only as healthy as the person they which they come. Exosomes, on the other hand, constitute up to 80% of the benefits seen with stem cells and are available through Dr Kaufmann in the United States at least,  in addition to being less expensive. I highly recommend Dr Kaufman's books, these are major reference book that will serve those looking to optimise their health, performance and longevity. Dr. David P. Turner, a dedicated researcher at the Massey Cancer Center, is passionate about dogs and their well-being. Specializing in Cancer Prevention and Control research, Dr. Turner focuses on the adverse connections between advanced glycation end products (A.G.E.s) and human health. A.G.E.s, harmful oxidative compounds prevalent in processed and high-temperature-cooked foods, play a significant role in chronic diseases. In collaboration with his wife, Dr. Victoria Findlay, co-leader of the Cancer Prevention and Control research program, Dr. Turner co-founded the Anti-A.G.E. Foundation, a non-profit organization raising awareness about the health impact of processed foods. Driven by his extensive research on A.G.E.s' effects on human health, Dr. Turner shifted his focus to canine and feline companions. How gummed up with sugar are your blood cells? Hemoglobin A1C (hbA1c) measures your average blood sugar from the past 3 months. Your red blood cells carry oxygen to your tissues using a protein called Hemoglobin A. The higher your blood glucose, the more glucose sticks to the hemoglobin A1c protein. How glycated is your hemoglobin? Glycation is a spontaneous non-enzymatic reaction of free reducing sugars with free amino groups of proteins, DNA, and lipids that forms glycated residues. Sugar is sticky (think of cotton candy sticking to your fingers) and this sugar can gunk up your arteries and cells.  Our red blood cells keep a record of your behavior. The answer hides in your red blood cells – specifically, your hemoglobin. A1C measures average blood sugars by detecting the glucose stuck to the hemoglobin protein.  The higher A1C, the more glucose glycated your protein. Test your hemoglobin A1C to learn your average blood sugar. You can request this test from your doctor. There are also companies who have created A1C test kits to do at the comfort of your own home. A “normal” A1C level is below 5.7%, a level of 5.7% to 6.4% indicates prediabetes, and a level of 6.5% or more indicates diabetes. Within the 5.7% to 6.4% prediabetes range, the higher your A1C, the greater your risk is for developing type 2 diabetes. The functional medicine range of A1C is 5.2% or lower. C-Peptide. C-peptide is a substance that is created when the hormone insulin is produced and released into the body. The insulin C-peptide test measures the amount of this product in the blood. The pancreas makes insulin, but not directly. The beta cells in the pancreas first produce a protein called “proinsulin.” Each proinsulin breaks down to one molecule of insulin and one molecule of C–peptide. Both are released when blood sugar levels get high. The more your beta cells get stimulated for insulin production, the faster you gain weight and create inflammation. While moderate levels lower inflammation, even slightly higher levels have been linked to insulin resistance, metabolic syndrome, heart disease, and cancer. C-peptide can give you a good idea if you've been overworking your beta cells. The functional medicine range for C-peptide is 0.8 – 3.8 ng/mL. Levels below 0.6 ng/mL (0.2 nmol/L) are a sign of possible beta cell failure and type 1 diabetes. High C-peptide levels indicate that the body is producing too much insulin. Fasting Insulin. This is a great compliment to the C-peptide test. Testing your insulin can reveal hidden problems in the way your body controls your blood sugar, years before they escalate to more serious issues, such as diabetes. In many cases, your blood sugar levels might not change for 10+ years, meanwhile you are getting closer each day to developing diabetes. This is because insulin is working so hard to keep those sugar levels “normal”. After a period, insulin cannot keep up with the demand. At this point blood sugars start to rise, and a diagnosis of prediabetes or diabetes is made. A fasting insulin test allows you to see if you are insulin resistant or on the road to insulin resistance. The standard reference range for a fasting insulin is less than 25 uIU/mL. The functional medicine range for fasting insulin is 3 – 7 uIU/mL. 25-Hydroxy Vitamin D. Vitamin D is a fat-soluble vitamin (and hormone) the body needs to build and maintain strong bones. It helps absorb calcium in the gut, keeping calcium and phosphorus in balance to mineralize bones. Vitamin D also helps support immune balance. Most people are deficient in Vitamin D. Symptoms of vitamin D deficiency may include: -            Fatigue -            Not sleeping well -            Bone pain or achiness -            Depression or feelings of sadness -            Hair loss -            Muscle weakness -            Loss of appetite -            Getting sick more easily -            Pale skin  Is the solution to take Vitamin D supplements? Not really. I am not opposed to taking a fat-soluble complex supplement, but it isn't until you lower insulin that you'll be able to change your Vitamin D numbers for the better. High insulin levels block the production of Vitamin D! You can take all the vitamin D in the world, and get sunshine every day, but you will not optimize your vitamin D levels until insulin is optimized. The great news for you, this book will teach you how to do this. The standard reference range on your lab report is typically 30-80 ng/mL. The functional medicine range for Vitamin D is 60-80 ng/mL) Homocysteine. High homocysteine has been linked to many health problems including heart disease, dementia, and Alzheimer's. When homocysteine cannot be converted to other compounds, it builds up in the body and may cause damage. B vitamins play a very important role in this balance. The standard reference range for homocysteine is under 14.5 umol/L. The functional medicine range for homocysteine is 5 – 7 umol/L. High Sensitivity C-Reactive Protein. An important marker of systemic inflammation. While this protein helps you fight infections, it is also a marker of low-grade inflammation and a predictor of your heart disease risk. People with higher hs-CRP values have a more elevated risk of cardiovascular disease and those with lower values have less risk. Standard reference range for hs-CRP is 0 – 3.0 mg/L. Functional medicine range for hs-CRP is 0.5 mg/L or under. Let's talk about lipids! The most common questions I get asked on social media is about cholesterol. Here are the markers I suggest you get done to assess if you are risk for heart disease. You will not see total cholesterol on here because this marker is close to meaningless. High Density Lipoprotein Count (HDL-C). HDL (high-density lipoprotein) is often referred to as the “good” cholesterol because it helps to remove the “bad” cholesterol from the body and is required for many vital processes within the body. Having high levels of HDL cholesterol has been shown to decrease your risk for heart disease. Cholesterol transported by HDL is known as “good” cholesterol because it is being removed from artery walls, which helps prevent, reduce, and even reverse hardening of the arteries (atherosclerosis) and heart disease [2]. HDL also plays a role in reducing oxidative stress and inflammation, improving cell and immune function, enhancing insulin sensivity, and may protect against neurodegenerative disorders [5, 6, 7]. The standard reference range for HDL for men is 35 – 65 mg/dl, for women is 35 – 80 mg/dl. The functional reference range for HDL for men and women is 60-90 mg/dl. Triglycerides. High levels of triglycerides can increase your risk of obesity, heart disease, diabetes, and other chronic health conditions. Triglycerides can be stored in fat tissue, or can be used as energy. For example, the body can release stored fats into the bloodstream as fatty acids when you don't eat for a prolonged time. These fatty acids travel back to the liver, which transforms them into triglyceride-containing very low density lipoproteins (VLDL) that are returned into the blood. On the other hand, your body stores more triglycerides when you overeat or have an unhealthy diet, such as the good ol' standard American diet. The standard reference range for triglycerides is under 150 mg/dL. The functional medicine range for triglycerides is under 90 mg/dL. Triglycerides to HDL ratio (TG:HDL). Your Triglyceride/HDL ratio helps offer a lifelong metric for your health. TG:HDL is one of the criteria for the diagnosis of metabolic syndrome, which was briefly known as Reaven's syndrome, named after Gerald Reaven, MD, of Stanford University. He and his colleagues around the world pioneered much of the research on TG:HDL ratio that indicates cardiovascular risk. TG:HDL is a strong marker of lifestyle factors—Your TG:HDL ratio offers a way to see whether you have a metabolism that naturally has cardiac protection or whether you need to earn it. (18:05) Dr. David Perlmutter is the author of the bestseller "Grain Brain: The Surprising Truth about Wheat, Carbs, and Sugar--Your Brain's Silent Killers." It's all about gluten and your health. Gluten sensitivity may relate to 30-40% of all people walking the planet who have a significant inflammatory response when they consume this protein found in wheat, barley, and rye called gluten. (22:40) Gluten induces inflammation and inflammation is the cornerstone of everything you don't want to get, like Alzheimer's, heart disease, and even cancer. (23:30) Your risk of developing type 1 diabetes if you have celiac disease is increased by about 300%. (23:50) Gluten sensitivity is an issue that goes far beyond a gut and upset stomach; it can cause joint pain, skin issues, and brain-related problems like depression and dementia, ADHD, obsessive-compulsive activity, and even schizophrenia. (24:40) People are misled that once they go gluten-free they're going to lose a lot of weight, which isn't true. Gluten-free food still has carbohydrates. (26:35) Fruit should be limited as well; Dr. Perlmutter and Vinnie suggest what is reasonable, as fruit is a very concentrated form of sugar. (26:45)  SATURATED FATS AS SUPER FUEL We desperately need saturated fat; as a result, we have to revise the whole notion that fat is bad for you. (28:00 Humans have been on a high-fat, low-carb, gluten-free diet for about two million years and it served us very well until the government decided to tell us that we need to go on a low-fat diet. (28:20) There's this bizarre notion that you need to have your whole grains each day and it just isn't true. (29:40) In 1992 the US government said we should eat a low-fat or no-fat diet and within 10 years of that doctrine, the rate of diabetes rose by 300%. (30:50) A recent study shows that saturated fat has no relevance when it comes to heart disease whatsoever. (31:20) Egg substitutes are detrimental to health but whole eggs are extremely healthful. (34:00) When it comes to dementia it's all about the carbs, not cholesterol--the brain needs cholesterol to function. (36:00) When people go gluten-free, cut out the carbs and reintroduce fat, we see breathtaking things: gaining back brain function, reducing issues with ADHD, losing weight, sleeping better, better libido, and less depression. (40:30) Glycation is the combination of protein and sugar, which increases the production of free radicals and turns on inflammation, which is the cornerstone of brain degeneration. (41:10) Dr. Perlmutter explains A1C; there is a perfect correlation between A1C levels and the risk of brain shrinkage and dementia.
