ob Zukis is a man on a mission to improve the ability of corporate America to succeed in a complex digital world, even when under constant cyber attack. Bob is the CEO and founder of the Digital Directors Network, the global pioneer in helping corporate directors advance their understanding of systemic risk. We consider Bob to be the world's leading advocate for improving cybersecurity governance. His many articles published in major business journals and impactful books on the topic make this case well. Bob has worked with, studied, and been on corporate boards for years and now teaches corporate governance as an Adjunct Professor of Management at the USC Marshall School of Business. He is co-author of the book The Great Reboot. We examine the book and Bob's approach to helping corporate directors mitigate cyber risk in this OODAcast. Topics covered include: How the 1200 strong members of the Digital Directors Network collaborate together to seek to reduce systemic risk. The creation of the Qualified Technical Expert (QTE) program and how the need for QTEs on boards is analogous to the need to have a Qualified Financial Expert (QFE) on boards when Sarbanes-Oxley drove that requirement. The new SEC regulations on cybersecurity that will require corporate boards to designate cybersecurity experts. How the new US Cybersecurity Strategy is helping create positive momentum in corporate America (Bob says "the White House has declared war on systemic risk with this strategy"). Actions directors can take to ensure corporate management is appropriately engaging to mitigate not just cyber attacks against the company, but broader systemic risks. Bob explains that "It's not just enough for board members to ask questions on cyber risk, as the questions are meaningless if corporate directors don't understand the answers." Very well put! Board members should continuously seek to improve their ability to understand. And then on top of that should ask the right questions. What is Bob's view of a powerful question boards should be asking? " What's the value of what we are trying to protect, and how safe is it for what we're spending?" Bob provides information on an event that brings together the Digital Directors Network called Domino (16-17 May 2023 in Chicago). This is a gathering of 200 of DDN's corporate director, CIO and CISO members for a unique executive learning experience. This year's event will feature keynotes from experts like SEC Commissioner Jaime Lizarraga explaining the new cyber rules being rolled out by the SEC. AI applications have large data volumes with lots of clients and conventional storage systems aren’t a good fit. In this episode, James Coomer from DDN talks about the lessons they have learned building storage systems to support AI applications. Inferencing requires terabytes or petabytes of data, often large files and streaming data. For example, autonomous driving applications generate hundreds of terabytes of data per vehicle drive, resulting in petabytes of data to ingest and process. DDN’s parallel filesystem goes a step further than NFS with an intelligent client that directs I/O to leverage all network links and storage endpoints available. Deep learning loves data, and a smart client can make the whole application faster. Because data is the biggest AI challenge today, an advanced storage solution can really help deliver AI solutions in the enterprise. Although most companies realize that finding expertise (data scientists, etc) is a major challenge, building infrastructure to support them is just as critical. Da la impresión de que las reflexiones que tienen sobre el mundo en el que viven son automáticamente inválidas por su condición etaria. ¿Qué se hace entonces? ¿Mejor que jueguen y hagan sus cosas en paz, sin contaminarse con la “política” y los adoctrinamientos de los adultos? Pero resulta que tienen mucho que decir sobre el país en el que viven, y dentro de las muchas cosas que develó el estallido social del 18 de octubre, es que desean y exigen ser escuchados en el espacio público. Quizás sea hora de cumplir de una vez por todas el mandato de la Convención de los Derechos del Niño y escucharlos, pero de verdad. En noviembre del 2019, la Facso, a través de su programa de Estudios Interdisciplinarios de la Infancia, realizó un cabildo orientado solo a niños, niñas y adolescentes desde los 4 hasta los 17 años. Según Camilo Morales, el psicólogo y coordinador de dicho estudio, el propósito fue “saber cómo ha sido para ellos la experiencia del conflicto social, cómo eso les afecta su vida cotidiana y qué les gustaría proponer para tener un país mejor” (Biblioteca del Congreso Nacional de Chile, 2019). De esta forma, y a través de papelógrafos, post-it, lápices y juegos, los niños, niñas y adolescentes tuvieron el espacio para expresar sus emociones y reflexiones. En los diversos grupos se iban repitiendo las mismas palabras, tales como “respeto, escucharse, justicia, derechos y derechos humanos” (BCN, 2019). Si nos damos cuenta, no distan mucho de lo que los adultos dicen también. Un estudio realizado por la Defensoría de la Niñez (DDN) sobre el efecto que tiene la crisis social en niños, niñas y adolescentes (2020), dilucida mejor algunas opiniones recogidas en este rango etario. Por ejemplo, dice una niña de 9 años que está “un poco enojada porque el semáforo, la banca y el basurero no les hicieron nada a los manifestantes”. En otro caso, una adolescente de 16 años de Arica, está muy feliz porque la gente por fin está dando la cara y son “los mismos jóvenes los que se ponen de acuerdo”. También está el caso de un niño migrante de 11 años que opina que los Carabineros están muy mal, porque le pegan a la gente. Una adolescente de 13 años dice que “los carabineros están matando a una persona que está protestando” e incluso un niño de 8 años se siente enojado, triste y con miedo, ya que a su papá “le cayó una bomba lacrimógena en los pies”. Nunca debemos olvidar que los niños, niñas y adolescentes se ven afectados por lo que sucede, y así como internalizan y reproducen la cultura, tienen un rol activo en la producción de significados (DDN, 2020). Todas y todos tienen una perspectiva única, en un contexto único, que habla mucho de ellas y ellos, pero también de todas y todos nosotros. No por nada las y los estudiantes saltaron los torniquetes del metro por sus padres, madres, tías, tíos, vecinas, vecinos, profesores, profesoras y sus compañeros y compañeras. Desestimar eso sería una enorme falta de respeto. Ellas y ellos quieren hablar, y es hora de escuchar mirando a la altura de sus ojos y a la altura de sus expectativas.

As we mentioned on our last episode, recently, at the invitation of Dave and Andi from Double Date Nation, we joined a group of podcasters for a meet and greet event called Podcaster Shinanigans at Menages in Nashville. It was a real treat to have so many varied-style podcasters together in one place so, to memorialize the occasion, we all sat around with our microphones just before the club opened and attempted to record a huge joint show, but sadly, it was an epic fail. The next day it was discovered that the whole audio file was distorted and completely unusable. The conversation was fun and interesting with fantastic observations and opinions from Euphoria Chronicles, Our Naughty Escapades and Spiritual Swingers. It was disappointing to have lost it. We were so thankful for the hospitality of DDN and Menages, and so sad to have lost the insights that we had all recorded, that last week we made a return trip to Nashville and attempted to recreate just a little bit of the lost information and ideas. In this casual, unscripted recording, we and the founders of DDN took up the same prompt that was ill-fatedly discussed by our podcaster's group, "What's so great about lifestyle clubs?" In a format very different than our typical show, the four of us got comfortable on Dave and Andi's couch, poured some drinks, pushed record, and enjoyed a great evening of relaxed conversation. Welcome to the History of Computing Podcast, where we explore the history of information technology. Because by understanding the past, we're able to be prepared for the innovations of the future! Todays episode is on the history of the Domain Name System, or DNS for short.  You know when you go to www.google.com. Imagine if you had to go to 172.217.4.196, or the IP address, instead. DNS is the service that resolves that name to that IP address. Let's start this story back in 1966. The Beatles released Yellow Submarine. The Rolling Stones were all over the radio with Paint It Black. Indira Ghandi was elected the Prime Minister of India. US Planes were bombing Hanoi smack dab in the middle of the Vietnam War. The US and USSR agreed not to fill space with nukes. The Beach Boys had just released Good Vibrations. I certainly feel the good vibrations when I think that quietly, when no one was watching, the US created ARPANET, or the Advanced Research Projects Agency Network.  ARPANET would evolve into the Internet as we know it today. As with many great innovations in technology, it took awhile to catch on. Late into the 1980s there were just over 300 computers on the Internet, most doing research. Sure, there were 254 to the 4th addresses that were just waiting to be used, but the idea of keeping the address of all 300 computers you wanted to talk to seemed cumbersome and it was slow to take hold. To get an address in the 70s you needed to contact Jon Postel at USC to get put on what was called the Assigned Numbers List. You could call or mail them.  Stanford Research Institute (now called SRI) had a file they hosted called hosts.txt. This file mapped the name of one of these hosts on the network to a IP address, making a table of computer names and then IP addresses those matched with, or a table of hosts. Many computers still maintain this file. Elizabeth Feinler maintained this directory of systems. She would go on to lead and operate the Network Information Center, or NIC for short, for ARPANET and see the evolution to the Defense Data Network, or DDN for short and later the Internet. She wrote what was then called the Resource Handbook.  By 1982, Ken Harrenstien and Vic White on Feinler's group at Stanford created a service called Whois, defined in RFC 812, which was an online directory. You can still use the whois command on Windows, Mac and Linux computers today. But by 1982 it was clear that the host table was getter's slower and harder to maintain as more systems were coming online. This meant more people to do that maintenance. But Postel from USC then started reviewing proposals for maintaining this thing, a task he handed off to Paul Mockapetris. That's when Mockapetris did something that he wasn't asked to do and created DNS.  Mockapetris had been working on some ideas for filesystems at the time and jumped at the chance to apply those ideas to something different. So Jon Postel and Zaw-Sing Su helped him complete his thoughts which were published by the Internet Engineering Task Force, or IETF, in in RFC 882 for the concepts and facilities and RFC 883 for the implementation and specification in November 1983. You can google those and read them today. And most of it is still used.  Here, he introduced the concept that a NAME of a TYPE points to an address, or RDATA and lives for a specified amount of time, or TTL short for Time To Live. He also mapped IP addresses to names in the specifications, creating PTR records. All names had a TLD or Top Level Domain name of ARPANET.  Designing a protocol isn't the same thing as implementing a protocol. In 1984, four students from the University of California Berkeley wrote the first version of BIND, short for Berkeley Internet Name Domain, for BSD 4.3. Douglas Terry, Mark Painter, David Riggle, and Songnian Zhou using funds from a DARPA grant. In 1988 Paul Vixie from Digital Equipment Corporation then gave it a little update and maintained it until he founded the Internet Systems Consortium to take it over.  BIND is still the primary distribution of DNS, although there are other distributions now. For example, Microsoft added DNS in 1995 with the release of NT 3.51.  But back to the 80s real quick. In 1985, came the introduction of .mil, .gov, .edu, .org, .com TLDs. Remember John Postel from USC? He and Joyce K Reynolds started an organization called IANA to assign numbers for use on the Internet. DNS Servers are hierarchical, and so there's a set of root DNS servers, with a root zone controlled by the US Dept of Commerce. 10 of the 13 original servers were operated in the US and 3 outside, each assigned a letter of A through M. You can still ping a.root-servers.net. These host the root zone database from IANA and handle the hierarchy of the TLD they're authoritative for with additional servers hosted for .gov, .com, etc. There are now over 1,000 TLDs! And remember how USC was handling the addressing (which became IANA) and Stanford was handling the names? Well Feinler's group turned over naming to Network Solutions in 1991 and they handled it until 1998 when Postel died and ICANN was formed. ICANN or the Internet Corporation for Assigned Names and Numbers, merged the responsibilities under one umbrella. Each region of the world is allowed to manage their own IP addresses, and so ARIN was formed in 1998 to manage the distribution of IP addresses in America.  The collaboration between Feinler and Postel fostered the innovations that would follow. They also didn't try to take everything on. Postel instigated TCP/IP and DNS. Postel co-wrote many of the RFCs that define the Internet and DNS to this day. And Feinler's showed great leadership in administering how much of that was implemented. One can only aspire to find such a collaboration in life and to do so with results like the Internet, worth tens of trillions of dollars, but more importantly has reshaped the world, disrupted practically every industry and touched the lives of nearly every human on earth.  Thank you for joining us for this episode of the History Of Computing Podcast. So I don’t want to steal your flame cause you already liked did your speech and so you can just tell everybody those slides again if you want. But I’m going to do it from start to beginning. Sure. Um, so tell, tell us real quick about your studio, what you guys are doing and all that stuff. [01:09] Sure. So our studio was in Ashland, Virginia, just outside of Richmond. We photograph about 200 seniors a year in addition to some families and babies and some corporate work that we do. I started out of my home originally or I started on location really and then to my home. And then we opened our retail space in 2005. Um, our first studio was about 1500 square feet and we lived across the street because the street that you want to be on in our small town. And um, now we have a 3000 square foot studio and that’s been since 2011. I have a fabulous team that works with me. I’m very fortunate to have, um, Karen and Robin support me and help me so that I’m able to really do the thing that I love, which is photography. Yeah. [01:46] Yeah. Kendra I was going on today good morning good morning how you doing I am doing well how are you doing this more well than what I am on the day number four of the fasting mimicking diet how about you I am also on day four the fasting mimicking diet made by prolonged made by prolonged so I want do a shout out to Dr. Joseph onto the CEO prolonged and Dr. Walter Longo who wrote the longevity diet they like we brought in a professor of stem cells may ease size pretty amazing how he actually played football at Oklahoma State went to med school I perform indices with him several years ago whenever he was still heavily just as he is today doing orthopedics he's a fantastic surgeon all of this is led to somebody's actually can remind me you he never wants to stop learning and so it's it's led to areas where he is today and he is I would say quite the expert with stem cells and where the future is going with them so this is so exciting that we have to time to fast for this episode because you know people throw around the word stem cell a whole lot and much like the CBD industry people throw around there's lots of misinformation there's lots of quality differences the people that are actually giving stem cells there's lots of differences with that so we could clarify all of it I really feel like there's some parallels you and I have brought in several CBD experts right and there's some parallels here and so you know this is a super exciting for that anything going on with the family anything going on socially well this last week whenever Morreale moved into preparing to do this fast I would say that the boys have the have enjoyed teasing us and other than that they've they thought back into all season basketball quite busy and is kind of the every day as usual at the round Renner household self typical tennis weekend both the Lucas and Karla were playing tennis and they both did really well it was kind of a little curb although I'm really proud of my team because as there are part of that institution it doesn't allow us to do certain things will go out of the country to get that kind help is such a small world and its fate in whatever it is I think a lot of things that have happened in both our lives have been opportunities that we take advantage of and I love you and I were working one day and I was like dude did you see that Joe Rogan we had Mel Gibson on and some other guys some PhD knew it yeah I know those guys and I was like laughing because no Gibson said the same thing a lot of people think is like you think about stem cells you have a mouth going up the side of your face and now not at all as it turns out he took his dad down to Panama and his nonmaterial dad and he got better and that's what I was like whoa and you started saying to me so the first question is your on day four of the prolonged fast why the world even doing that well it has been shown that in cellular metabolism in July 2015 summarize whatever you and will fast it promotes stress resistance so basically when you're put on a fasting mimicking diet or I should back up we've always known that the caloric restriction diet has been shown in yeast and other animals to prolong life Walter Longo in his book figure out a way that you can eat a little bit and trick your body to believe that it is completely fast that's the fasting mimicking diet is so this study in 2015 looked at putting mice on the fasting limiting diet and they demonstrated that these mice decrease the size of multiple organs improved glucose control decreased visceral fat lower blood pressure improved bone mineral density rejuvenated the immune system and reduce cancer risk always too good to be true and Academy just five days three times a year and can accomplish this but wait there's more we got more here they also showed after they re-fed the mice solicit what this is the coolest thing about this when you listen to vaulter give lectures he said it's not so much the fast it's the recovery from the fast because when they re-fed him they showed that in older mice areas of the brain like the hippocampus showed neurogenesis and improved cognitive performance while yeah so it's pretty wild with the re-feeding that super important which makes it fun because on Monday for I'm really looking to some refuted yeah can't wait for some re-creating but any that's all part of the process I will say going through at my second fast that this is becomes easier it's it's not as hard as the first run of some that's with prolonged but it is the expectation is they are know what to expect I know struggle for so what would be contributing to better neural thinking that is you a question so the question is what's going on there so then we dug up an article in the Journal of stem cell research in 2016 what they showed is that fasting protects against immune system damage and induces regeneration by waking up stem cells or by catalyzing dormant stencils so all those what they realize is what this article describing is what vulture was figuring out right there what he had figured out that all comes down to stem cells at the refuting stage basically not with her not stenciled several times which is to find what is real quick stem cells are the body's raw material their pre-sells for all other cells stem cells are the only cells that can generate new cell types and they can divide into form what are called daughter cells which become specialized cells that eventually come specific organs that's all me to say about it because wage may come in here and blow basically after you fast and then you re-feed you flip a regenerative switch which promotes stem cell regeneration in the blood making organs so that the important thing so when you go into starvation mode the body will save energy and one ways to recycle immune cells and that causes autophagy so old and sick and dying cells are programmed to hate go away right and then the autophagy gets rid of the old site sells them when you re-feed the stem cells wake up and they go around there's a bunch of fallen soldiers they don't bother them but they go around and say we need new people to replace this over simplistic way to lead into a much cooler explanation of that and then with each cycle you re-feed you get rid of sick and dying cells and replace them is like a janitor it's exactly like a janitor so five day fast three times a year you just cannot quit you just doing some serious housecleaning I member Saturn panda when he talks about that that's the godfather of circadian rhythm fasting or intermittent fasting right he's got mouse models he still does a prolonged fast because he describes it as you brush your teeth every day and then you a couple times a year you going to get the deep cleaning from the dentist I like that analogy you're always doing maintenance which is you keep in the nine date of of what you're eating but every once while you get to do that deeply definitely and then trying to fast for the first time you you can look at you like why would I not want to eat but if you look at the history of time where people were in the movement nowadays to return to health where people are trying to get healthier and you look at things like Paleolithic's for instance they talk about new diets and ways to eat in a pale lifestyle some of that also includes fasting and the reason is the primal man also went a long time without having food and for a few days at a time they would have intermittent fasting org or a few days fasting themselves yeah and so basically you're just returning to what it is that we've all been programmed to do for a long time that it just so happens the last several generations we've had ubiquitous amount of food here in the US and so now we it'll thicker bigger let's okay so this is no doubt about it feet eating is awesome right but eating is an inflammatory process so when you eat your you to become inflamed a little bit and then you incorporate the nutrients and so will the way that were doing and how will we eat so much it's probably not the healthiest way so right now where the fast let's talk about was actually a lot going on with our bodies before talk about Howard to tie all this together two stencils. So day one this is basically the five defenses what's happened to you and I so day one just upon your body day to start doing some fat burning day three start doing some cellular recycling basically you're going to clean up start realizing were on day three now for backing cavemen times they one day to that's normal day three your body starts going up oh we better get ready for something because were you have to go out and get some food soon and that's when you start doing the cleanup and this is when a lot of people will reach ketosis day for you and I right now are in this this is the cell regeneration this is where autophagy started yesterday in autophagy is when those old sick and dying cells are programmed to go away much like we talked about with the polyphenols when they get in there in your list and causes my top a G5 in the foods we eat will do that then this starts artists are stem cell-based regeneration is starting to ramp up so when people talk about how I fascinated 24 hours 09 and identity a fast we've come this far when were this far into it tomorrow's what all the real magic happens all the magic day five regeneration continues now we've turned on our stem for you to go and eat and I had in them and an abundant oriented interview time for you to get up have the energy to go kill the woolly mammoth so that you can eat and you can feel it a mere year manic and it was time and you saw me run when I quit the fast I mean I think of the time I my blood sugar was 54 and and and I felt fine with that and at key tones I think for 4.8 and the moment that we drew ride today the labs and that in the blood that one time it was it's time to eat yeah so both of us have ever similar expenses this is the third day that we done this so now let a geek out here for the last four minutes because this is where I think it comes in really cool and so we found an article that has a really long title and I just like I like saying it just because I realize that environment possible so there's different mediums to do this this study looked at taking stem cells out in vitro meeting in a dish and they bathed them in CBD and more range in MO R which is a program coming in today really appreciate absolutely so see what I do know him yeah you know whatever no word of her Rhonda Patrick goes on Joe Rogan's economy gets better paper out you'll be taking some notes I favor Gerardi, Patrick never feels more like absolutely just a caveman. Dr. she's a lot of knowledge of sausage yellow management so you since you do already went through a couple of small things in the last half-hour you played football look on the state you been orthopedic surgeon for several years but that's not really what you here to talk about today the cool part is I actually can you hear we good we are little my problem were going to get something fixed here real quick right area is going the cool part for me they spent is kind of been allowed to reinvent myself as a traditional surgeon when the science, caught up to what we do and figure out that a lot of what we would think of as traditional medical approaches were less than optimal from patients and point so exited a fellowship in trauma and post dramatic reconstruction after an orthopedic surgery residency and after a general surgery internship so in during a general surgery internship when I thought I was going to do transplant surgery because that was the coolest guys at the hospitalists on which Trina here in Dallas Fort Worth multiple hospitals with DFW medical service rendered him orthopedic residency I did my fellowship and trauma to the general I was Roy Sanders 2000 but hospital where as to trauma fellows panorama the program we had 10 residents in for helicopters and no sleep with a whole new episode on what lack of sleep will do to your stem cells well it lack of sleep and not healthiest UW not like lack of sleep induced by fasting where you feel like Superman lack of sleep induced of note falling asleep in the boat in the lounge chair waiting for the nurse to tapping the short essays time to go from zero to hero so but during the trauma fellowship we became very adept because we got stuck with a lot of the fractures that other people treated heel so the posttraumatic part of that is acutely we were stabilizing multiple extremity injuries but we also do those people over multiple extremity injuries and we would get referred a lot of the trauma patients the gentleman traumas what we called it which comes to visit you in clinic does it come to you in the middle night by helicopter gentlemen trauma that watching your clinic is like I get this knife stuck out as well hey I've had five surgeries on my femur fracture I still can't walk and still no bonus I have is soon healed and we had to find a way to not only promote tried to trick the body and the human something that already showed didn't want to heal but in the least invasive way possible to turn the table can help the patient generate new bone best way to do that early on was Boomer us were concentrate the very Baston first uses of bone roster concentrated in traditional surgery was in the treatment of nonunion when someone has a fracture that doesn't heal there is delayed union which is means takes forever that but there's nonunion wishes means it doesn't heal there's no bone if you have a leg doesn't have a hip fracture can't walk can't but wait on it given upper extremity with an unhealed fracture pretty much flail you can you have a non-nonfunctional extremity bone rest will concentrate in the treatment protocol this allowed us to be much less invasive instead of just it doesn't make a lot of sense to just take out all the other plates and stripped the blood supply room muscle replay fracture further destroying the blood supply to the fracture that already the numbness blood supply to heal so let's go ahead and revisit that really quick because as a surgeon it's interesting to dear somebody because that's typically that's a knee-jerk reaction all that last surged and worked when he operated as exactly so say one more time with re-operating guys will here's the problem with re-operating if you didn't heal the first time it's because of the formation of scar tissue not healed tissue so the healing gets stopped the fibrotic tissue begins scar tissue hurts scar tissue does not much blood supply and scar tissue is very functional it's fibrotic it it can take up some space for the most part the difference between the interface between healthy tissue and scar tissue continues to be painful forever every time you move something severe big fibrotic knot of scar with an attendant and you have some healthy tissue that generate the connects to it the mismatch in pliability that mismatch of you would never use metals that have different hardness when you put together an engine it is the soft metal in the hard metal caused threading and corrosion in significant problems and and metal mismatch soft tissue mismatches just as big a problem we create scar tissue and people hurts generates pain generates an inflammatory response or chronic inflammatory response from cytokines that without decent blood supply to scar tissue you want to get rid of so you end up with long-term and continued muscle that and a lot of our surgery approaches and a lot of surgery where you just strip off the blood supply to the bone that it needs to help heal don't work very well because were not focused on how the body needs to really heal this fracture were focused on making x-ray and I did just make you look nice so your I think it's fascinating because essentially I've done the same I've done the same thing in my practice rub to move from traditional gastroenterology become almost a functional orthopedist yeah I would like to think that that I I just come to the table with some of the extra tools that I need to kinda set the table for the patient to help them heal I the body has unique ability to overcome a lot of things Systematic analysis of a novel human renal glomerulus-enriched gene expression dataset.

Glomerular diseases account for the majority of cases with chronic renal failure. Several genes have been identified with key relevance for glomerular function. Quite a few of these genes show a specific or preferential mRNA expression in the renal glomerulus. To identify additional candidate genes involved in glomerular function in humans we generated a human renal glomerulus-enriched gene expression dataset (REGGED) by comparing gene expression profiles from human glomeruli and tubulointerstitium obtained from six transplant living donors using Affymetrix HG-U133A arrays. This analysis resulted in 677 genes with prominent overrepresentation in the glomerulus. Genes with 'a priori' known prominent glomerular expression served for validation and were all found in the novel dataset (e.g. CDKN1, DAG1, DDN, EHD3, MYH9, NES, NPHS1, NPHS2, PDPN, PLA2R1, PLCE1, PODXL, PTPRO, SYNPO, TCF21, TJP1, WT1). The mRNA expression of several novel glomerulus-enriched genes in REGGED was validated by qRT-PCR. Gene ontology and pathway analysis identified biological processes previously not reported to be of relevance in glomeruli of healthy human adult kidneys including among others axon guidance. This finding was further validated by assessing the expression of the axon guidance molecules neuritin (NRN1) and roundabout receptor ROBO1 and -2. In diabetic nephropathy, a prevalent glomerulopathy, differential regulation of glomerular ROBO2 mRNA was found.In summary, novel transcripts with predominant expression in the human glomerulus could be identified using a comparative strategy on microdissected nephrons. A systematic analysis of this glomerulus-specific gene expression dataset allows the detection of target molecules and biological processes involved in glomerular biology and renal disease.
