David Barreto es de origen venezolano pero reside en España. Tiene más de 15 años realizando clases, talleres y retiros de yoga. He went to his car and brought his check book, and gave him advance as much as he got entire rumenuration as writer for Nuvve Kavali, he bought a bike with that money.  At that point Nuvve Kavali still hadn't released and he had confidence in him to do it. So basically everybody who worked for Nuvve Kavali worked again for Nuvve Nuvve as a formality basically as goodwill. Went to Mumbai to meet Shriya to narrate the story  I directed Nuvve Nuvve film after the release of Nuvvu Naaku Nachav. I couldn't do that film the way I wanted. I was not completely satisfied with my work. I think the dialogue writer in me dominated the director in that film. However, it was a commercially successful project for the producer and was a hit at the box office.  Writing/Direction:  So actually the main point is that a father's love for his daughter can never be beaten. The entire movie is actually a flashback because Prakash raj narrates entire story to Tanikella Bharati of what happened.  The best part of the movie is that its very realistic but also the whole world created for the characters. Basically all of them face the same problem which is everybody is torn between father and lover all 3 main characters.  Also with Rishi, Anjali, and Viswanath, you get to travel with them, learn their worlds, learn their ideologies, and learn the way they think and go through this wile journey with them which is rare for a film.  But much like the movie idiot, this is one of those point of view movies, and if you ask me the only correct POV is Prakash raj. But yourself in his shoes and whatever doubts he has they are valid. Praksh raj isn't even the villain, hes just a man trying to understanding veeyrhting and fears that someone other than him will love his daughter kore than him.  Vishwananth seems like a man with such high morals and everything but theres a crucial scene with Rajeev Kanakala where he calls out Viswanath's hypocrisy and he starts to think about it.  Also Rishi and Anjali's actions start to affect their families as well and its like everything that's going on is basically like a chain reaction, IN that aspect this is a brilliantly written and directed film.  He eventually loses and gives Anjali to Rishi.  That being said two points seemed very over the top: First one was them just casually going to Mumbai and back, idk how that works but it seemed quire unrealistic and illogical and then they run out of petrol and just keep pushing the motorcycle towards Annavaram. What he did there was actually stalking even though they like each other.  Also the movie pace was really slow and especially 2nd half it just completely slowed down. The copy iw watched was 2 hrs 48 mins. Also movie is so dialogues heavy that I get exhausted. Its one thing to have heavy dialogues and they work, like theyre really good, the opposite would be Aagadu so I hope it makes sense.  These drama movies aren't my thing no matter how good the drama might be, I need some other commercial elements in the movie. Very little Trivikram mark comedy as well.  Direction by Trivikram is good for the characterizations, writing, and dialogues. But huge pacing issues as the movie is very slow and feels like a lot of lag in between giving boredom.  In the end, Amma avakayi Anjali bore kottindi actually.

Abstract: ApoE4 isoform contributes to increased risk for Alzheimers Disease (AD) over the life course of individuals. Much remains unknown about the biological pathways that connect APOE4 genotype with the development of pathology that eventually leads to AD, nor do we know how early in life these cellular alterations begin. To answer these questions, we derived neural precursor cells (NPCs) from induced pluripotent stem cells (IPSCs) that were CRISPR-edited at the APOE locus. We intended to characterize the protein expression landscape in the NPCs subsequent to targeted deletion of E4 from a parent IPSC line of APOE3/4 genotype. Differentially expressed proteins (DEPs) following mass spectrometric analysis were determined from the protein abundance fold change values obtained for each protein. Proteins which showed greater than 1.5-fold difference with FDR adjusted P-value less than 0.05 were considered differentially expressed. DEPs were mapped to the STRING database (v11.5) for retrieval of interacting proteins and functional enrichment. CRISPR-editing of E4 from the parent line revealed 98 differential expressed proteins. Of these, 54 were upregulated, and 44 were downregulated. Further analysis of the DEPs via STRING database showed that these changes primarily affect pathways linked to RNA processing, plasma membrane repair, and cytoskeleton organization. Indeed, we find the effects of E4 extend beyond proteins considered central to AD pathology. Knowing more about the protein interactions regulated by ApoE, in an isoform-specific manner, can reveal new mechanistic insights into development of AD. Copy rights belong to original authors. Editors - Craig Hayes, Varun Viswanath ACE, Chris Chandler and Trevor Penna The Blindspotting editing team of Craig Hayes, Varun Viswanath ACE, Chris Chandler and Trevor Penna collectively crafted a show unique in both its structure as well as its visual style.  On the audio side, the show is underscored by a soundtrack that pays homage to the Bay Area roots of Blindspotting creators, Rafael Casal and Daveed Diggs.  Hayes and Viswanath took on the task of cutting Season One, while Chandler and Penna split editorial duties for the sophomore effort. Taking place six months after the events of the film Blindspotting, the tv series begins with Miles, Ashley's partner of 12 years and father of their son, suddenly being incarcerated.  The situation leaves Ashley to navigate a chaotic and humorous existential crisis when she and her son Sean are forced to move in with Miles' mother and half-sister. CRAIG HAYES Craig Hayes has cemented a career as a spirited and versatile editor. He studied film at Ithaca College in New York, which inspired Craig to hone his craft and move forward in his career.  From there, Craig worked as associate editor on Gus Van Sant's To Die For and M. Night Shyamalan's Lady in the Water, and as assistant editor for Steven Spielberg's War of the Worlds and Quentin Tarantino's Django Unchained.  Craig was also first assistant editor for the award-winning HBO series John Adams.  Craig has recently worked on the critically acclaimed The Hate U Give, directed by George Tillman Jr. as well as being nominated for a best editing award for HBO's Righteous Gemstones. VARUN VISWANATH ACE Varun is a Film & TV Editor based in Los Angeles, CA. He is a member of the American Cinema Editors and the Motion Pictures Editors Guild, and has an MFA in Film Editing from The American Film Institute Conservatory. Abstract: Adults with bipolar disorder or epileptic seizures are commonly prescribed sodium valproate. In utero exposure to this drug is linked to a multitude of defects in normal brain development, from neural tube defects to autism spectrum disorders. During the course of brain development, neural precursor cells (NPCs) give rise to neurons and glia, and therefore to understand the valproate-induced defects, it is crucial to understand its effect on NPCs. Two NPC lines, both derived from healthy individuals, were used for all experiments. Cells were treated with 0.7mM valproate for one week. Fresh media (+/- drug) was replenished every alternate day. RNA was extracted on day 7 of drug treatment, and transcriptomics performed. All experiments were performed in biological replicates. Genes that showed greater than 1-fold difference (with FDR adjusted q-value less than or equal to 0.05) were considered differentially expressed. We further investigated the interacting partners of the differentially expressed genes using PINOT, as well as cellular pathways using DAVID. Our primary endpoint of analysis were genes that were differentially expressed (DEGs) with valproate treatment in both the NPC lines used. We found 21 such genes that were common in the two lines. PINOT revealed 504 interacting partners of the DEGs. Functional annotation analysis showed significant enrichment of four signaling pathways - Wnt, Notch, Rho-GTPase and PI3K-AKT. While the role of Rho-GTPase is a novel finding, we have replicated previously reported findings on Wnt, Notch and PI3K-Akt pathways, which further strengthens their role in mediating neurodevelopmental anomalies. It may be recalled that Dr Swamy already filed case before SC against places of worship act, 1991 and notice was issued to Govt of India. It's strange that current dispensation which came to power on lofty promises refuses even to do basic thing. Instead, sales automation is designed to simply make sellers more productive at doing the same work. If there are repetitive steps that are done, then this is where sales automation can shine. But, the important thing to consider is that you must first understand your prospecting process and then look for ways to get more done, faster.  That's where tools like FlyMSG can be really useful in scaling how you communicate with prospects to book more meetings, with less time and energy spent. Listen into the conversation and hear how exactly sales leaders should use sales automation to enhance their outreach cadences. What Are the Drawbacks of Sales Automation? Over the past two years of our prospecting research, we've found that: 1-2 word subject lines including your prospect's name perform best Incorporating a specific personal event or interest in the copy of your sales messages improves response rates by 61% Using an omnichannel approach to prospecting gets better results than sticking to only one prospecting method Inserting a short, 30-45 second welcome video in your first sales outreach message can dramatically increase responses The power of personalization is that you can never do ‘too much' of it.  If your sales teams are not already investing time in looking on LinkedIn at prospect and buyer profiles before reaching out, then you could be sabotaging your results. Recently, Yiddish became the fortieth language to join Duolingo, an achievement that followed hot on the heels of Harry Potter being translated into Yiddish. And would you believe that a brother and sister were responsible for these separate accomplishments? Well, when we found out about it we were struck by how many of the same kinds of conflicts and considerations that faced Irish came up again in thre context of Yiddish. So we had to find out more.On today’s episode, Darach chats with siblings Meena and Arun Vishwaneth about their family’s relationship with Yiddish. They tell him how the phenomenon of Yiddish loanwords in slang has a downside when promoting the language, about the sense of post-assimilation loss that defines the relationship some American Jews have with the language. G., Sali, A., Davis, T. N., Agard, D. A. Abstract: Microtubule (MT) nucleation is regulated by the {gamma}-tubulin ring complex ({gamma}TuRC), conserved from yeast to humans. In Saccharomyces cerevisiae, {gamma}TuRC is composed of seven identical {gamma}-tubulin small complex ({gamma}TuSC) sub-assemblies which associate helically to template microtubule growth. {gamma}TuRC assembly provides a key point of regulation for the MT cytoskeleton. Here we combine cross-linking mass spectrometry (XL-MS), X-ray crystallography and cryo-EM structures of monomeric and dimeric {gamma}TuSC and open and closed helical {gamma}TuRC assemblies in complex with Spc110p to elucidate the mechanisms of {gamma}TuRC assembly. {gamma}TuRC assembly is substantially aided by the evolutionarily conserved CM1 motif in Spc110p spanning a pair of adjacent {gamma}TuSCs. By providing the highest resolution and most complete views of any {gamma}TuSC assembly, our structures allow phosphorylation sites to be mapped, suggesting their role in regulating spindle pole body attachment and ring assembly. We further identify a structurally analogous CM1 binding site in the human {gamma}TuRC structure at the interface between GCP2 and GCP6, which allows for the interpretation of significant structural changes arising from CM1 helix binding to metazoan {gamma}TuRC. The Great Flood of 1924 is often considered a landmark moment in Kerala's history, such that people who experienced it anchor their memories in relation to this event. Smaller floods occurred in 61, 84, 94. Then 2018 brought with it a once-in-a-century flood, with three times more rain than the Great Flood. With the rains came destruction - loss of property, culture, livelihoods, and life. Kerala's identity seems to have been irrevocably changed by these 2018 floods. The romance about the rain has been replaced by a wariness, but also a drive for real change to build resilience and the proof of what can be achieved through the community surge of action. In this episode we look into Kerala's changing relationship with its water, land, and geography. This week's Yiddish Voice presents an interview, in Yiddish, with the creator of the recently published Yiddish translation of the first of theHarry Potter book series, Arun ("Arele") Viswanath (אַרעלע ווישוואַנאַט). Harry Potter and the Philosopher's Stone is the wildly popular first novel in the Harry Potter series, written by British author J. K. Rowling. The book was first published in 1997. The Yiddish edition, with the title הערי פּאָטער און דער פֿילאָסאָפֿישער שטיין (Harry Potter un der filosofisher shteyn), was published by Olniansky Tekst in 2020 and immediately sold out its first print run. A second printing has since sold out as well. A third printing is planned but delayed due to COVID-19. The publisher web site for information and future purchases is here: https://harrypotter.olniansky.com Arun “Arele” Schaechter Viswanath is the grandson of the prominent Yiddish linguist and professor Dr. Mordkhe Schaechter. He grew up in Teaneck, New Jersey, in a Yiddish- and Tamil-speaking home. He has stated that his desire to translate Harry Potter grew from a strong feeling that a translation of such a popular children's novel into Yiddish would be a boon and a resource for Yiddish students and teachers, Yiddish-speaking children and parents, and the Yiddish world more broadly. Arun lived for a time in the Boston area as a Harvard undergrad majoring in linguistics, and he now resides in New York and works in high tech as an operations strategist and data analyst. Viswanath was interviewed by regular Yiddish Voice contributor Lillian ("Leah") Shporer-Leavitt, who prepared by reading the entire Yiddish translation, having already read every Harry Potter book in the series with her children. Leavitt, a daughter of Shoah survivors, grew up speaking Yiddish and graduated from the Boston Workmen's Circle Yiddish Shule. A former computer business analyst and software trainer, she is an experienced Yiddish teacher and translator, having taught at the Workmen's Circle, Gann Academy of Greater Boston, Boston University Hillel, and Brandeis University. In addition, she has subtitled Yiddish films for the National Center for Jewish Film and translated Yizkor (memorial) books for the Jewish Genealogical Society. Excerpts of Harry Potter in Yiddish! (and English) In addition to short readings by Arele Viswanath during the interview, the show presented excerpts of his longer reading of the entire first chapter of this book. The so called “Pariah Problem” emerged in public consciousness in the 1890s in India as state officials, missionaries and “upper”caste landlords, among others, struggled to understood the situation of Dalits (those subordinated populations once called untouchables). In The Pariah Problem: Caste, Religion, and the Social in Modern India (Columbia University Press, 2014) Rupa Viswanath unpacks the creation and application of this so called “problem.”The interview explores the ways in which land, labour and ritual combined in producing the Pariah and the affect Protestant missionaries had in reshaping Pariah-ness, as well as the role of the colonial state and changes in house site ownership among other issues. Amazingly rich in detail and theoretically dynamic throughout, the book is relevant to numerous discussions in present day India and beyond.
