Though they are not well read by most Christians today, they remain authoritative for pastors, elders, and deacons. Today we'll cover 1-2 Timothy, Titus, and Philemon. Read his bio here —— Notes —— The Pastoral Epistles 1 Timothy, 2 Timothy, Titus, Philemon Philemon is not always included as a pastoral Epistle since it only deals with a specific issue regarding a single person. Household Paul employs the metaphor of a household in the pastorals to illustrate the order he'd like to see in the churches (1 Tim 3:14-15). ZIBBC: “In Xenophon's essay, the husband instructs his fourteen-year-old bride (the average age for marriage among Greek and Roman girls was fourteen to sixteen) on her vital role in managing the household resources, including the care and supervision of household slaves, while he supervised the gathering of produce from the farms. Because of the presence of slaves and freedmen in an average ancient household, management of sometimes large households could be demanding.”[1] Lynn Cohick: “The family in the Greco-Roman world valued the community over the individual and promoted corporate honor and fortune. Those living in the domus (“home”) included parents and children, and perhaps extended family, such as adult siblings, cousins, and grandparents, as well as slaves, freedmen, and freedwomen. Each individual had a specific status within the home, and each family member deemed the social status of the family, including its wealth and social prestige, as of equal or greater value than their personal happiness.”[2] Paul is concerned for how outsiders will perceive the churches (1 Tim 3:7). Everyone has a place and a role. Men's role (1 Tim 2:8), older men (Tit 2:2), younger men (Tit 2:6-8). Women's role (1 Tim 2:9-15), older women (Tit 2:3), younger (Tit 2:4-5) Respect elders (1 Tim 5:1-2, 17-19). Young widows should marry (1 Tim 5:11-15). Take care of your own family (1 Tim 5:7-8, 16). Support real widows (1 Tim 5:3-6, 9-10). Slaves obey & submit (1 Tim 6:2; T 2:9-10) Rich be humble/generous (1 Tim 6:18-19) Godliness (Eusebia) 1 Tim 4:7-8; Tit 2:11-12 (See also 1 Tim 2:1-2; 5:4.) Translated “godliness” but has nothing to do with being like God or imitating God. Showing expected reverence Piety (Latin = pietas) Pious, reverent, dutiful Shown not just to God (or gods) but also to the city and to one's household Church Leadership Overseer's qualifications (1 Tim 3:2-7; T 1:7-9) Elders' qualifications (Tit 1:6) Deacon's qualifications (1 Tim 3:8-13) Standards are high for service in the church. Overseers must manage their own households well (1 Tim 3:4-5). Warnings Against False Teachers Some strange Jewish teachings (1 Tim 1:4, 6-7; Tit 1:10-11, 14) Asceticism (1 Tim 4:1, 3) Proto-gnostics (1 Tim 6:20) Unlabeled false teachings (2 Tim 2:14, 16, 23, 25; Tit 3:9) Warn those who cause divisions (twice), then have nothing more to do with them (Tit 3:10-11) Names individuals: Hymenaeus & Alexander (1 Tim 1:20), Phygelus & Hermogenes (2 Tim 1:15), Hymenaeus & Philetus (2 Tim 2:17-18), Alexander the coppersmith (2 Tim 4:14) Sound Teaching (Healthy Doctrine) Sin is when you live contrary to healthy doctrine (ὑγιαινούση διδασκαλία) (1 Tim 1:10). Sound teaching nourishes you (1 Tim 4:6). Encouraged to hold the standard of sound teaching (2 Tim 1:13) “People will not put up with sound teaching” (2 Tim 4:3). “Rebuke them sharply, so that they become sound in the faith” (Tit 1:14). “Teach what is consistent with sound instruction” (Tit 2:1). Reading the Pastoral Epistles Mostly focused on church leadership Church leaders NEED to read them. The pastorals tell us who is qualified and who is disqualified for church leadership. It's good for everyone to know what they say so you can hold your leaders accountable. Philemon Philemon was a wealthy Christian leader who had a house church. His slave, Onesimus, ran away and later became a Christian. Paul sent Onesimus back to Philemon, asking Philemon to treat him as a brother. This Epistle is a masterful piece of rhetoric, Paul at his finest. Ben Witherington III calls Philemon the emancipation proclamation of the NT. Review Paul wrote the pastoral Epistles toward the end of his life. The metaphor of a household illustrates how the churches should function. Everyone should play their role according to their duty, whether men or women or elderly or young. Godliness is a major concept in the pastoral Epistles. It refers to performing your duty with proper honor. It's important that Christians live respectfully and with proper dignity before a watching and suspicious world. The pastoral Epistles include clear qualifications for church leaders, including that they manage their own households well. The churches in Ephesus and Crete were beset with false teachers from both Jewish and Greco-Roman sources. Paul encourages Timothy and Titus to adhere to sound teaching (healthy doctrine), so that people will escape the dangers of false teachers. Philemon was a wealthy church leader who managed a household that included slaves. Paul sent Philemon his runaway slave, instructing him to accept Onesimus as a brother. Paul told Philemon to charge any debt Onesimus owed him to his account. Such a request radically reconfigured social norms in a way that undermined the system of slavery. [1]Baugh, S. The 2023-24 Marquette University men's basketball season will be the first for Chris Howell, as a program assistant.Prior to arriving at Marquette, Howell served as an assistant coach at Hill Community College in Hillsboro, Texas (2022-23) and University of Mary (2021-2022). Howell spent one season at Jackson State as a graduate transfer during 2018-2019, he averaged 12.2 points, 7.4 rebounds, and 2.3 assists per game and helped the program finish third in the Southwestern Athletic Conference.A two-year letterwinner with the South Dakota State men's basketball team from 2016-2018, Howell helped the Jackrabbits win 46 games in two years. He led the program to two Summit League Conference Tournament championships and two NCAA Tournament appearances. Playing one season at Butler Community College during 2014-2015, Howell helped the Grizzlies to a 29-4 record and guided the program to a Jayhawk West Conference championship. He earned All-Jayhawk West Conference honorable mention and All-Region VI honors. Further, the action space is a keyboard and a mouse, which has to be operated only given the game's video input. OpenAI tackles this challenge using Video PreTraining, leveraging a small set of contractor data in order to pseudo-label a giant corpus of scraped footage of gameplay. The pre-trained model is highly capable in basic game mechanics and can be fine-tuned much better than a blank slate model. This is the first Minecraft agent that achieves the elusive goal of crafting a diamond pickaxe all by itself. Abstract: Pretraining on noisy, internet-scale datasets has been heavily studied as a technique for training models with broad, general capabilities for text, images, and other modalities. However, for many sequential decision domains such as robotics, video games, and computer use, publicly available data does not contain the labels required to train behavioral priors in the same way. We extend the internet-scale pretraining paradigm to sequential decision domains through semi-supervised imitation learning wherein agents learn to act by watching online unlabeled videos. Specifically, we show that with a small amount of labeled data we can train an inverse dynamics model accurate enough to label a huge unlabeled source of online data -- here, online videos of people playing Minecraft -- from which we can then train a general behavioral prior. Despite using the native human interface (mouse and keyboard at 20Hz), we show that this behavioral prior has nontrivial zero-shot capabilities and that it can be fine-tuned, with both imitation learning and reinforcement learning, to hard-exploration tasks that are impossible to learn from scratch via reinforcement learning. For many tasks our models exhibit human-level performance, and we are the first to report computer agents that can craft diamond tools, which can take proficient humans upwards of 20 minutes (24,000 environment actions) of gameplay to accomplish. Abstract: Machine learning can infer how protein sequence maps to function without requiring a detailed understanding of the underlying physical or biological mechanisms. It's challenging to apply existing supervised learning frameworks to large-scale experimental data generated by deep mutational scanning (DMS) and related methods. DMS data often contain high dimensional and correlated sequence variables, experimental sampling error and bias, and the presence of missing data. Importantly, most DMS data do not contain examples of negative sequences, making it challenging to directly estimate how sequence affects function. Here, we develop a positive-unlabeled (PU) learning framework to infer sequence-function relationships from large-scale DMS data. Our PU learning method displays excellent predictive performance across ten large-scale sequence-function data sets, representing proteins of different folds, functions, and library types. The estimated parameters pinpoint key residues that dictate protein structure and function. Finally, we apply our statistical sequence-function model to design highly stabilized enzymes. Our special guest today is Dr. Cara Tucker from rural Victoria, Australia. When you have any part of your family tree online on any of the “” websites (Ancestry, MyHeritage, Findmypast and FamilySearch) they do a lot of the hunting for you. They deliver hints that have a good chance of matching up with your ancestors. Your job is to carefully review them and determine if they are your ancestor's records. Whether we are helping our parents downsize to a smaller house, or we need to downsize our own belongings to carve out a spare bedroom or just make room in a closet. it's never really an easy task. And I think it's safe to say it's even more difficult for the family historian, because we collect a lot of paper, photos and other things that are often near and dear to our hearts. Devon Noel Lee and her husband Andrew Lee of the Family History Fanatics YouTube channel have taken on this challenge themselves and they've written a new book called Downsizing with Family History in Mind. Here to help you make the tough choices and clear the clutter is Devon Noel Lee. There are many reasons for downsizing: To move to a smaller place Absorbing inherited genealogy Divorce To free up space in your own home Downsizing the sentimental items is the hardest part of downsizing. Question: A lot of us just think, well it's a Saturday morning, I think I'll just do some decluttering. But you say in the book that decluttering doesn't work. Why is that? Devon's Answer: “There are three things that experts teach us that are absolutely wrong:” We don't give ourselves enough time for nostalgia. We're really bad at evaluating what's going to last for the long term We use the wrong boxes when decluttering – all the experts say to use Keep, Sell and Donate. Devon recommends the following boxes: Keep Giveaway (combining sell and donate) – to family, societies, archive, university special collections, libraries, etc. Trash (or recycle) Process How to “process”: Digitize Process the information in your binders and get rid of the binders if no one wants them. Use it: Sad to say, most people don't want your family china. Give yourself permission to use it and enjoy it now. Make memories with it! Let your children play with things. Four Basic Downsizing Principles in the book: Reduce: Divide things into the boxes. Preserve: This is when you're going to digitize the things in your process box. Photograph objects. Transfer your genealogy into software and online trees. Reclaim: Take everything out of the process box after processing, and divide into Giveaway, Trash and Keep. Don't put things into storage! Showcase: Put on display what you found worth keeping so it can be enjoyed. Transform what you have into something that is easier to pass on like videos, podcasts, scrapbooks. Focus on story-based items. From Lisa: It puts us back in control as to what happens to it. Making sure the right people get it. I'm a big fan of displays. If we haven't taken a moment to get something on the wall - to put a display together - how can we expect our family to appreciate it and embrace our family history values? Question: Many downsizing projects are much more than a single day. When you're faced with a really big job, where do you recommend that people start, and where should they put their primary focus? The book includes action plans for folks who have: just an hour Weekend 3-6 months 6-12 months Capture what is right now: Photograph the outside of the home. Photograph what's inside. Then focus on photographing the collections in their context. Mentioned by Lisa: Genealogy Gems Podcast includes a Gem called Thanks for the Memories. In it, I share an example of mentally walking through my Grandma's house and capturing all of my memories on paper. Get a piece of paper or pull up a word document. Close your eyes for a moment and visualize a favorite memory from your childhood. In my case I started with a favorite place, my maternal grandma's house. But perhaps yours is the back alley where you and your friends played baseball, or your great uncle's garage where he showed you how to work on cars. Whatever is meaningful to you. Now, open your eyes, and write your thoughts one at a time. Just free flow it. They don't have to be complete sentences. Later you can try your hand at writing more of your actual experiences or memories of a person. Again, it doesn't have to be a novel or sound really professional. It's just the memories from you heart. Family Photos: Question: If we have piles and piles of family photos, particularly ones we've inherited, how to do we decide which to keep and which to toss? Or do you ever toss? Devon's answer: Get rid of the duplicates! Keep 1 of the biggest and best and throw the rest away. Don't bog yourself down with hours spent trying to track down someone else to give them to. Get rid of blurry, overexposed, underexposed, and meaningless photos. Unlabeled photos: There will be some circumstances where you will not be able to keep them. You can't go into debt for unlabeled photos. You want to separate them from the labeled so that other family members don't throw them all out together. If you have time, try to identify them by asking relatives, and posting them to DeadFred.com. If you can, donate the remaining unlabeled photos to orphaned photo collectors, or toss. You did the best you can. Don't feel guilty because your ancestors didn't label their photos. Question: What advice do you give your readers who are faced with what to do with their genealogy when they don't have descendants or when no one in the family wants it? What encouragement can you offer when there is no one who descends from you, or there is no one who wants them. Devon's answer: If you think you don't have anyone in your family who is interested, you're wrong. Downsizing and organizing will increase the chances of someone willing to take it later. If you don't have anyone in your immediate family who wants your stuff, start looking for distant cousins actively working on a surname. They won't want everything. You will have to divide the material. They want it organized. Do it while you're living – don't leave it to someone else. Digitize it and get it online where it can be shared. It’s a new month and with it a brand new theme for our upcoming Build episodes! When designing products we often think about usability: how easy to use a product is. But we often overlook another aspect of product design: accessibility. So all this month we’re going to dive into accessibility. One reason accessibility gets overlooked is because we think it’s a challenge to prioritize it given a company’s size and resources. We may think accessibility makes sense for a big company, but a startup that is getting off the ground just doesn’t have the resources to incorporate it. Well, actually that’s not true... In fact, accessibility maybe just the differentiator you need when it comes to product design that is going to give your product a competitive advantage and increase adoption! And in today’s episode, we’re going to explore what accessibility is, why it’s important for any size company to incorporate, and show you how to do an accessibility audit for your product. In each episode of *Build*, I host innovators and together we debunk a number of myths and misconceptions related to building products, companies, and your career in tech. Now, one often overlooked aspect of building products is accessibility. In today's *Build* episode, we're going to talk about what accessibility is, why it's important, and how you can do an accessibility audit for your product. To help us out, I've invited Laura Allen, who is a accessibility program manager at Google for Chrome and the Chrome operating system. Thanks for joining us today, Laura. Laura Allen: Absolutely. Thank you so much for having me. Poornima Vijayashanker: Sure. I know that a lot of times people think about usability when they're building products but they don't often think about accessibility. Let's talk about what is accessibility and how is it different from usability. Laura Allen: Accessibility is the design of products, services, devices, and environments for people with disabilities. I always like to take that one step further and think about accessibility as really empowering users with disabilities to be productive, to be socially engaged, and to be independent. This is super closely aligned with the concept of usability and also even just universal design and inclusive design. You think about universal design being this idea of building products that are going to be usable by the widest range of people and the widest range of situations. It's so closely aligned with this, that absolutely includes designing for people with disabilities. This whole concept of usability, yes it's critical to be thinking about all the time, of course, but we can make products functionally accessible, we can go through checklists, we can incorporate design principles and what not to make things technically work, but if you don't think about how is this actually going to be used, what is the experience for someone with assistive technology—like a screen reader, for example—if you don't think about that experience and usability of that experience it might not be productive or efficient at all. All these things are really closely linked together and they all help to move towards building an inclusive product. Poornima Vijayashanker: Why is this even important? I think a lot of people would say, "Oh, we have a really niche customers customer base, we don't think anybody has accessibility concerns so why even bother?" Laura Allen: Accessibility is something that impacts everyone at some phase or at some point in their life. Fifteen percent of the global population has some form of disability—that's a huge number, that's over a billion people. We tend to think about a few different distinct groups when we're thinking about design. We might be thinking about people who are low vision or blind, people who are deaf or hard of hearing, people who have motor or dexterity challenges. Then people who are, what we consider to be, neurologically diverse that can be anything ranging from dyslexia, to perhaps being on the autism spectrum, to any forms of intellectual disabilities. When you think about these different groups of people, people might be developing disabilities at different phases of their life, different severity levels, different combinations of disabilities, and then you start to think about, what about temporary impairments? Like what if you break your arm and all of a sudden you can't type on your computer for a few months? Situational impairments, like what if you're at a loud restaurant or a loud bar and there’s something on the TV that you want to be listening to, it's too loud to hear and you have to actually rely on those captions that were there specifically for the deaf population but they're helpful to everyone. Then, you take it one step further, and you think about this growing aging population, which thanks to increasing life expectancy, which is great, the aging population of people over 60 is growing, and growing, and growing, and the World Health Organization estimates that by 2050 it'll be over 2 billion people that are over the age of 60. Poornima Vijayashanker: Wow, so it's like doubling. Hopefully not, but yeah. Laura Allen: As we all age, at any point in our lives, we may experience some slight deteriorations in vision, or of hearing, or of dexterity, so these concepts are really, really critical to be building in, in general. Poornima Vijayashanker: That makes sense. Now, some would say that this makes sense for really big companies with hundreds of millions of users, but does it really make sense for our tiny little startup that's just getting started? Laura Allen: I would honestly say, accessibility is something that is critical for all companies, at all stages, all phases. To be totally honest with you, it's actually easier to build this in four startup-sized companies, smaller teams, smaller processes. Of course, it's completely doable at large companies as well that have established processes, but at a startup, you're building from the ground up, you're defining what you want your product processes to look like, and it's so much better just to be able to integrate accessibility in at that level, get people understanding what these concepts are, make this just a core part of inclusive design from the very beginning, and it'll be that much easier as you grow, and grow, and grow. Another thing to think about here is accessibility because it impacts such a large number of people this presents, honestly, a growth opportunity in many cases. It just opens doors for a lot more business, a lot of growth potentially. One thing that I like to think about, especially for startups and just hiring in general, if companies are focused on actually making their own products accessible then it opens the doors as well for being able to hire a more diverse and inclusive workforce. You can hire assistive technology users and have them come in and be able to use your products and that opens the door. A lot of us, obviously, at the companies we're thinking about how do we further diversify? How do we get people in the room who have a diverse set of perspectives? This whole idea of diversity a lot of times we are thinking about race, and ethnicity, and gender, sexual orientation, but disability is a huge part of this. It is a very, very big part of this group and we need a voice. Poornima Vijayashanker: Making it into your process, your priorities, your core values can really open doors for you in terms of your customer base and make things, hopefully, easier as you grow. Laura Allen: Absolutely. I will say, too, for a lot of people, like I mentioned before, accessibility will touch everybody at some point and in many cases it'll make the experience better, and more usable for many, many users. For someone like me, I happen to be low vision myself— Poornima Vijayashanker: What does that mean, “low vision?” Laura Allen: Really good question, because it can mean a lot of different things. For me, I basically have a central vision disorder, so if you can imagine all in my peripheral vision is still intact, it's still clear, but anything I'm looking directly at is this blend of flashing lights, and distortion, and blurring, and whatnot. This all happened for me when I was about 14, happened really quickly, really rare condition. I basically went from having typical 20/20 vision to being what's considered legally blind within about a week when I was 14. At that point, it was like I'm getting ready for high school, and all of a sudden I'm going to be moving to a bigger school, and then what happens? I couldn't read a book at that point. I couldn't see a blackboard. I couldn't recognize faces in the hallway. It was a huge period of transition for me, and for my family. For a few years there, it was one of those things where if materials weren't actually accessible in formats that I could listen to, for example, instead of visually read, I was stuck. I had to literally come home from school and my parents and my brother would read to me. That, to me, was the definition of dependence and I really, really hated it. I was so fortunate to have a family that was able to help me that way. It was just unbelievable the amount of effort they went through to get me through to the point where then I was able to regain my independence through discovering assistive technology like text-to-speech software, or magnification, or a larger mouse cursor, things like that. It was that period of my life that really propelled me into this world of accessibility and usability, because I saw the huge potential of what technology can do for someone's life and I just want to help to make that better for the rest of the world. Poornima Vijayashanker: It's great to hear you have a personal stake and it inspires everybody out there, but it also inspires you to realize and relate to people who might also be having these recognitions so that's wonderful to hear. For people in our audience out there who are building products, how can they get started? How can they prioritize this and gain the benefits? Laura Allen: That's a great question. There are a lot of different things to be considering. One thing that I would recommend is doing an audit, understanding where is your product right now, what's the level? This may vary. If you haven't really been thinking about accessibility yet, that's OK. It's a good opportunity to look at the holistic picture and see what's going on, and what bugs you may have. I would recommend just going through and leveraging a lot of the different resources that are out there and using those to create your own audit, however that works for you. For example, there is a great resource out there from the web content accessibility guidelines and we abbreviate that to WCAG. This is a W3C standard guidelines for accessibility. They've been really widely adopted by a lot of designers, engineers, companies, and they're wonderful. They outline different steps and different things to be considering. For example, they break it down into four different categories: perceivable, operable, understandable, and robust. Each of these things has a lot of different checkpoints, but just as a brief example, when we think about “perceivable,” what assumptions are you making about your users basically? What are we assuming that they can perceive? Are we assuming they have perfect sight or sight at all? Are we assuming that they can hear? Thinking about how they're perceiving the product and then different design guidelines that go hand-in-hand with that. “Operable,” similarly, is what are we assuming about the users, how they're actually operating with the product? Are we assuming they have really fine motor skills? That they can use a mouse, that they can use a keyboard? Are we assuming that they are able to use really quick reaction times, things like that? “Understandable,” what is the general understandability of the product? Are you assuming really high language skills to be able to navigate? Or the ability to just remember really complex sequences, all kinds of things like that? Then, “robust” is a little bit different in that it talks about how is your product working with assistive technology? Like a screen reader, for example, which would be leveraged by someone who's blind to be able to listen to the product, listen to the phone, or the computer, whatever it may be, and get that audio output instead of the visual. Poornima Vijayashanker: Nice. Laura Allen: The WCAG is a great resource. I tend to think when I'm thinking about checklists and working with designers and whatnot, I break down into a few key groups as well. The first around keyboard and focus, just really taking a quick poll of—let's say you've got a site, how does it work with just the keyboard, no mouse whatsoever? It's a great thing for engineers and designers to be able to try that out themselves as well. Just try using the keyboard only and as you're navigating through, can you get to everything that you need to? Can you also see visual focus indication? If you don't see that and you're just tabbing through, you don't know what you can actually take action on. Have you thought about that in the design process, basically? Then, I start to think about semantics. How do we actually make it more clear for screen reader users what the page is actually all about or what the app's all about? For example, do we have labels in place for buttons so that as you navigate with a screen reader, you don't just hear, "Button," or, "Unlabeled button," which is not helpful at all. Thinking about how do we just convey that experience and make sure that it's clear for a screen reader. Then a third bucket, which I like to call think about in my audits, is just this idea of flexible interface. That can be anything from color contrast—so WCAG actually says we should have a minimum color contrast ratio of 4.5:1 for text against its background color. That's super helpful because for anyone, like me, with a low vision or just anyone who doesn't have the perfect 20/20 vision, it can be really hard to actually distinguish those colors, or a low contrast text, so that's a really helpful usability improvement for a lot of people. In this same group of flexible UI, you think about things like how does this interface look with magnification at a 200% zoom level, for example? Or are we using just color, or just sound to convey information? Just color, one example there, is if you have an input field and you type an error and all of a sudden maybe just the text will appear red. In that case, people who can't distinguish color will miss that information, screen reader users, or braille readers will completely miss that information as well. Thinking about how do you go one step further and convey that and make sure there's also error messaging. You can still use the color red and all, that's fine but it can't be the only way that you're identifying that information. I like to think through questions like that using the WCAG guidelines and other things that help there—like I know Vox Media has a really great checklist—and just get a sense of where's the current level? From that point, you may have a lot of different bugs, you may have different things that you want to be able to address, and the next step is naturally to work on, “How do we triage this? How do you prioritize?” I think one really helpful thing to do there is just to think about each of these bugs, what is the typical user impact? How critical is this? Would this bug stop somebody from being able to actually interact or take action on your site and your core purpose of your site or your app? I like to think about that, and help to prioritize, and just go from there. Poornima Vijayashanker: Wonderful. We'll be also sure to include the resources you mentioned below in the show notes. You've mentioned a number of things that happen during the audit. What happens after the audit? Laura Allen: I think the next natural step, of course, is going through that triage and prioritization process. Then as you're solving these problems, as you're fixing bugs, continuing to go through and help to honestly integrate accessibility into each step of the process. I think that's the really critical step. One holistic audit is not going to take you all the way. We have to start bringing this into our development process and building it from the ground up. Then, honestly, getting out there and working with users, understanding what the feedback is. I think that's a really critical component to understanding how to improve. Hannes Leitgeb (MCMP/LMU) gives a talk at the Workshop on Mathematics: Objectivity by Representation (11 November, 2014) titled "On Mathematical Structuralism. A Theory of Unlabeled Graphs as Ante Rem Structures". Abstract: There are different versions of structuralism in present-day philosophy of mathematics which all take as their starting point the structural turn that mathematics took in the last two centuries. In this talk, I will make one variant of structuralism—ante rem structuralism—precise in terms of an axiomatic theory of unlabeled graphs as ante rem structures. I will then use that axiomatic theory in order to address some of the standard objections to ante rem structuralism that one can find in the literature. Along the way, I will discuss also other versions of mathematical structuralism, and I will say something on how the emerging theory of ante rem structures relates to modern set theory.
StatLearn 2012 - Workshop on "Challenging problems in Statistical Learning"
We consider a classification problem: the goal is to assign class labels to an unlabeled test data set, given several labeled training data sets drawn from different but similar distributions. In essence, the goal is to predict labels from (an estimate of) the marginal distribution (of the unlabeled data) by learning the trends present in related classification tasks that are already known. In this sense, this problem belongs to the category of so-called "transfer learning" in machine learning. The probabilistic model used is that the different training and test distributions are themselves i.i.d. realizations from a distribution on distributions. Conceptually, this setting can be related to traditional random effects models in statistics, although here the approach is nonparametric and distribution-free. This problem arises in several applications where data distributions fluctuate because of biological, technical, or other sources of variation. We develop a distribution-free, kernel-based approach to the problem. This approach involves identifying an appropriate reproducing kernel Hilbert space and optimizing a regularized empirical risk over the space. We present generalization error analysis, describe universal kernels, and establish universal consistency of the proposed methodology. Experimental results on flow cytometry data are presented.

In this work, gold and semiconductor nanoparticles are used as building blocks for nanostructures, in which energy transfer is investigated. Nanoparticles have size-dependent controllable optical properties. Therefore, they are interesting objects to study different aspects and applications of energy transfer. Fluorescence quenching by gold nanoparticles is investigated and used to develop novel immunoassays for medically relevant molecules. The range of fluorescence quenching by gold nanoparticles is effective over longer distances than for dye molecules. The reason for this is the large absorption cross-section of gold nanoparticles and the radiative rate suppression of dyes caused by gold nanoparticles. The influence of gold nanoparticles on radiative and non-radiative rates of Cy3 and Cy3B dyes is studied here. A competitive, homogeneous immunoassay for digoxigenin and digoxin, a drug used to cure heart diseases, is developed. Dye-labeled digoxigenin is bound to the gold nanoparticles functionalized with anti-digoxigenin antibodies, quenching the dye fluorescence. Unlabeled digoxigenin partially replaces the dye-labeled digoxigenin leading to an increase of fluorescence. The assay has a limit of detection of 0.5 nM in buffer and 50 nM in serum. Time resolved spectroscopy reveals that the quenching is due to energy transfer with an efficiency of 70%. A homogeneous sandwich immunoassay for cardiac troponin T, an indicator of damage to the heart muscle, is developed. Gold nanoparticles and fluorophores are functionalized with anti-troponin T antibodies. In the presence of troponin T the nanoparticles and fluorophores form a sandwich structure, in which the dye fluorescence is quenched by a gold nanoparticle. The limit of detection of the immunoassay in buffer is 0.02 nM and 0.11 nM in serum. Energy transfer, with up to 95% efficiency, is responsible for the fluorescence quenching, as found through time resolved spectroscopy. Energy transfer is demonstrated in clusters of CdTe nanocrystals assembled using three methods. In the first method, clusters of differently-sized water soluble CdTe nanocrystals capped by negatively charged mercaptoacid stabilizers are produced through electrostatic interactions with positively charged Ca(II) cations. The two other methods employ covalent binding through dithiols and thiolated DNA as linkers between nanocrystals. Energy transfer from smaller nanocrystals to larger nanocrystals in aggregates is demonstrated by means of steady-state and time-resolved photoluminescence spectroscopy, paving the way for nanocrystal-based light harvesting structures in solution. Multi-shell onion-like CdSe/ZnS/CdSe/ZnS nanocrystals are presented. In these structures the CdSe core and the CdSe shell produce two emission peaks upon UV light excitation. When the emission peaks are well matched, the resulting emission appears as pure white light. The shade of the white light can be controlled by annealing the particles. Evidence for intra-nanocrystal energy transfer is presented.
