This episode carries content warnings for mental manipulation, body horror, and death. Hours after defeating Artifice, and hours before Motion's flagship breaches the far end of the battlefield, an uncomfortable buzz runs through the air. Even with the reinforcements from Millennium Break and the Apostolosians loyal to Cas'alear Rizah, the defenders of Perennial are outnumbered. Worse: Great heroics on the battlefield might fell a Divine or rout a batallion, but how do you stop a machine god who exists across the whole galaxy, dormant in every particle of the Perennial Wave. Such a feat would require more than battlefield brilliance... This week on PALISADE: Above the Earth Pt. 5 Damn, you look like you had a bad time / You look like you don't even know why Dossier People Audacious Opportunity "A.O." Rooke (he/him): Once an Orion mercenary, then the commander of the Rapid Evening's “toughs” squad, and now a unit leader for Millennium Break. K.O Rooke's son. Kueen Overture "K.O" Rooke (she/her):  A long time dispatcher and operator of the Scrivener's Guild, KO has taken the role as a major commander in Millennium Break. Mother of A.O. Rooke. Passiflora (she/her): Elect of the divine Artiface, naive but devoted to the vision promised to her by The Pact of Free States. Xiphion, aka Zig'ell Barbos, aka The Red Roc (zig/zig's, he/him): Elect of the Divine Valour and icon of the Apostolosian military. Zig'ell kept his Apostolosian name in personal use even after being chosen by Valour as its elect. Famed for zig's talent as a pilot and commander, zig spent years serving as Cas'alear Rizah's confidant and lieutenant, before being promoted and transferred into service directly under the Divine Motion. In her absence, his profile only grew as he secured wins against the Bilateral Intercession. Now that Motion has revived, Zig'ell has returned to her service with optimism and loyalty--quieting any doubts through a commitment to the larger project of the Pact. Organizations The Pact of Free States: One of the two factions vying for leadership of the Principality, a joint operation by Stels Columnar and Apostolos. Before they were killed by Kalvin Brnine, they were led in name by the Glorious Princept, Dahlia (they/them), but were controlled in day-to-day matters by members of the original Pact of Necessary Venture, including Rye (he/him) and Gallica (she/her).  With all three dead, the Divine Motion has rushed to fill a vacuum many feel but can hardly give word to. Under her control, the long-held "reformist" goals of the Stel have been set aside for simple, metaphysical domination.  Places Earth: Ancient home of humanity. Moved into orbit around the center of the galaxy by the Old Earth Cult, under guidance by a manipulative and influential divine called Independence. Divines The Divine Integrity (it/its): Sometimes appearing as an articulated staff or a metallic spine, Integrity integrates itself into its chosen user. Once the two are connected, Integrity becomes a powerful exoskeleton, and supports its user in matters of military and morale. Until being assassinated by Brnine, Dahlia, the Glorious Princept, was the elect of Integrity. Now, it has found a new home in Thisbe. The Divine Motion (she/her): One of the founding members of the Pact, the necromantic Apostolosian divine once led its infamous retinue, the Black Century, on Partizan. They were defeated and dispersed, turned into part of Kalmeria, during Operation Shackled Sun. Now, she has returned to take control of the Pact of Free States, and has set her sights on Perennial at the heart of the galaxy. The Divine Present (it/its): On first blush, Present might be confused for a small, if luxurious frigate. And sometimes, it is just that: Its oval structure houses living quarters, a humble armory, enough cargo space to hold a single hollow, and even a fully stocked bar. But at a snap of its elect's fingers, it transforms into a humanoid war machine that serves its elect loyally, outclasses any Altar currently in operation, and is capable of holding its own against many other Divines in combat. The Divine Commitment (it/its): A malleable divine made of inky black metal that has become one with the body of the Ekpyrosis, flagship of the Pact's fleet. Has historically required two pilots (both in a condition of ego death) in order to be operated, but currently operates alone, tethered by something like loyalty to the Divine Motion. How such obedience was garnered is unclear.   The Divine Valour: A machine the size of a large Hallow, Valour can transform between a vast, roc-like bird form and a more traditional humanoid model. Soldiers claim that serving under Valour gives them a noticeable boost to their morale, but it is unclear if this is a feature of their proximity to the Divine or to its elect. Objects The Ekpyrosis: Flagship of the Pact's fleet, a transforming, beautiful monstrosity designed to casually destroy worlds while simultaneously supporting life in a vacuum. In the time since Commitment's takeover, though, it has changed dramatically.  Columnar Altars: Though Exenceaster March took his research with him when he defected from Columnar and the Zenith Fund he once led, those engineers who remained loyal to their Stel had learned enough to develop their own rival machine, powered not only by Kalmeria, but also by upscaled (and stolen) Equiaxed designs. The result: The ZFC-Wizard.  Central to this design is the modularity and iconic visual design that has been core to Columnar technology for generations. The stock Wizard is rarely seen as-is. Instead, it is used as a frame for a number of other variants. The ZFC-TOME is the workhorse model of the Wizard, able to hold its ground defensively with a massive claymore and an arcane "breath" weapon. The ZFC-STAVE is a designated marksman unit, while the CLOAK was designed to operate behind enemy lines without being caught. Two commander class units were created as well. The ORB houses the Wizard frame inside of a sphere, which projects a directed energy field to its allies, serving as a shield. The TALISMAN, with its swarm of hard-to-control drones, was designed especially for ace pilots who lead from the front with near-impossible acumen.  Apostolosian Altars: In the age of Kalmeria, Apostolosian culture became fascinated with mastery over (and blending with) the natural and elemental. As such, it is no surprise to see that their Altars (designed in collaboration with Columnar's Zenith Fund), reflect those new interests.  The ZFA-FANG is a new version of an old classic: A Hypercat model with mounted freeze cannons and the ability to skate on ice it creates. The ZFA-TALON offers air supremacy, firing powerful, flaming beams and dropping dense explosive salvos. Both operate with greater efficiency when deployed directly alongside their allies. Operating as an on field commander, the ZFA-TUSK is a defensive powerhouse that can chase down enemies with incredible, wind-driven speeds.   Mysteries Perennial (she/her):  The Principality's so-called 'adversary,' who lives at the center of the galaxy and whose chaotic whims spread through her "Perennial Wave," an ever-present nanoparticle that has bonded with elements of Autonomy Itself and the Divine Motion to create Kalmeria. Arbor: The result of Thisbe's year of research of Divines, Axioms, and plant life--all three of which have homes in her own body. A growing organism with unknown powers. At the Irmo Middle School, maypops, Passiflora incarnata, found their way to the pollinator garden without being planted. Probably they started when a songbird, many of which love maypop seeds, flipped its tail and deposited that seed when it landed in a shrub growing there.

A seaweed and a vine-flower, how are they alike? We started out this episode feeling like these two herbs were completely different from one another. By the time we got to the end, though, we found a unifying quality or two.Dulse, whose Latin name is Palmaria palmata, is our favorite choice for those who are new to seaweeds. It has a mild flavor, isn't too ‘fishy' or too ‘slimy'. It may not exactly be “bacon of the sea”, but it sure does add a nice salty & umami flavor to dishes! Dulse is also a great provider of minerals (but not too much iodine, so don't worry). Its nourishing qualities support us in a very grounded way, at the mineral levels of bone, muscle, and nerve health. In archetypal terms, it is an “earth of water” herb.Passiflora incarnata is the botanical name for passionflower. This is an astonishingly beautiful flower – make a web search to check out some photos, you'll see what we mean. (Oh, and don't neglect ‘passionflower UV light' as a search term: see what it looks like to bees!) It's an excellent plant to sit with for meditation – and it can help you move into a more meditative state of mind, too. Famously helpful in sleep formulae, passionflower helps rein in the spinning, anxous mind, and brings it inward and downward. If you're comfortable with elemental language, it might make sense to look at this herb as embodying the “earth of air.”If you're having trouble sleeping, if anxiety is keeping you up at night, we've got a plan for that. Our short course, Holistic Help for Better Sleep, teaches you key strategies for lengthening and deepening your rest each night. You'll also meet our favorite herbs to help with sleep disturbances and insomnia, and how to choose the right herbs for your own personal sleep needs. What's more, you receive everything that comes with enrollment in our courses, including: lifetime access to current & future course material, twice-weekly live Q&A sessions with us, open discussion threads integrated in each lesson, an active student community, study guides, quizzes & capstone assignments, and more!If you have a moment, it would help us a lot if you could subscribe, rate, & review our podcast wherever you listen. From rising temperatures, to endless wildfire smoke, it's hard to not feel its effects on our mental health as well. In this episode, Grizel talks with her good friend Ben Bishop, who is an agroforestry researcher at the University of Missouri studying agroforestry systems and tree crops. Through his research, he hopes to expand the utilization of trees and woody shrubs as both profitable crops and as a strategy to off-set greenhouse gas emissions. They discuss ways to stay hopeful, how to navigate the connection between mental health and the climate crisis, and about the different ways we can all make a change. Resources from this episode: Underneath It All Instagram: @underneathitallpodJoin the Underneath It All Facebook GroupFollow Grizel on Instagram: @_grizel_Follow Ben on Instagram: @the.forest.gardener and TikTok: theforestgardenerCheck out his podcast The Forest Garden on all platforms and on Instagram @forestgardenpodcastReverse in Climate Change PodcastSunrise MovementProject DrawdownA word about this season's sponsor: Since 2021, Salomon has increased its representation from 14% to 20%, including representation from race, gender, sexual orientation, size, and ability. Does that involve others? Does that mean being by yourself? What keeps you from pursuing hobbies? What keeps you from experiencing happiness? Are you living a lifestyle you want? If you change one thing about your life, what would it be?What societal roles are you fulfilling that you don't want to fill anymore? What societal rules did you grow up believing? What keeps you from sharing your mental health with others? What parts of yourself are difficult to change? What parts of yourself are easy to victimize and not move on from? What is the goal for outdoor adventures and when can it be changed from getting to that peak, to checking in and listening to your body, mind and emotional needs?Who is one person you trust to talk about REAL life with? Gute Laune garantiert. So viel steht fest. NatureNotes with Rudy Mancke

The Gulf fritillary or passion butterfly (Agraulis vanillae) is a bright orange butterfly in the subfamily Heliconiinae of the family Nymphalidae. The caterpillar food plants for gulf fritillaries are members of the genus Passiflora. The adult butterflies use nectar from many flowers, including Lantana plants. The Passiflora host plants are frequently called passion vines; in some Texan counties where this butterfly's population is high, gulf fritillaries will feed on specific species such as Passiflora lutea and Passiflora affinis.

Today’s Patreon-fueled shout-out is for Abundant Life Ministries, “working hard to create a better future for the Charlottesville community.”*As the week begins, the statewide seven-day average for positive COVID test results is 5.7 percent, down from 7.2 percent last Monday. Over the weekend, the Virginia Department of Health added 2,436 new cases with 953 on Saturday, 856 on Sunday and 627 this morning. There have been 3,021 deaths, with many of the ones reported last week actually having happened earlier in the pandemic and not reported until now. There were 137 cases of COVID-19 added over the weekend in the Thomas Jefferson Health District, with 64 on Saturday, 54 on Sunday and 19 reported today. Two more deaths have been reported since Friday for a total of 67. The University of Virginia’s COVID tracker has not been updated since Friday, despite reports in the Cavalier Daily that day that as many as four residence halls have had their populations tested due to positive tests and early detection in wastewater. *With the end of September approaching, Albemarle and Charlottesville are coming closer to the first nine-week period of the public school year. Last week, news came out that Charlottesville City Schools have put together a committee to help “plan a safe return to in-person learning.” Slides distributed after a meeting last week show that one option is to return pre-K through second grade as early as October 13 for four days a week. Older students would begin in-person instruction soon after. (reopening slides)“A survey will be sent to families next week regarding their intent to remain on a virtual schedule or plan for their children to return to school with the proposed plan,” reads one of the slides.Krissy Vick, the school’s community relations liaison, sent an email to parents that said the slides were a starting point for the committee’s discussion. “Returning to in-person instruction will take time, information, and multiple revisions to a plan,” Vick said “The School Board will vote on the final plan. Keep in mind that we will continue to offer families the choice for all-online learning -- and to the greatest degree possible, we will allow staff this choice, also.”The next scheduled School Board meeting is on October 1. *Habitat for Humanity of Greater Charlottesville has broken ground for the redevelopment of the Southwood Mobile Home Park off of Old Lynchburg Road in Albemarle County. Habitat purchased the site in 2007. Dan Rosensweig is president and CEO of Habitat.  “See, thirteen years ago, this community was facing a deeply uncertain future,” Rosensweig said. “Like a lot of trailer parks in the nation, it’s underground infrastructure had basically passed its useful life.” Rosensweig said there was also development pressure on the property and residents asked the owner at the time to sell to Habitat. The organization was working on redevelopment of the Sunrise Trailer Court in Charlottesville. Now Habitat has been working with Albemarle County on a multiphase effort to redevelop the site without displacing residents. “We come up with a plan of development that within the next ten years or so is going to turn this wonderful community of people into a wonderful community of people who are here, new people who are joining the community, new homes, and a little downtown core,” Rosensweig said. Albemarle Supervisors voted on a rezoning last year that paves the way for the first phase, which will take place along Old Lynchburg Road and does not involve any of the existing trailer homes. Scottsville District Supervisor Donna Price was not on the board when the rezoning happened, but she is supportive of the funding Albemarle has committed to the project. “When I think of Southwood, I think of the core values of non-displacement and sustainability,” Price said. “When I think I see Southwood, I see a representation of an essential public-private partnership that will produce a neighborhood consistent with Albemarle County’s Comprehensive Plan.”*In a time of restaurant closings, one space on the Downtown Mall is set to reopen tomorrow. The Champion Hospitality Group will take over the Commonwealth Restaurant with a new space called Passiflora. The new restaurant will offer “a Baja-Mediterranean-inspired menu of small plates.” According to a release, the restaurant will open for dinner with limited indoor and outdoor seating, following CDC guidelines for safe social distancing. Last night, Bashir’s restaurant in the same block had a farewell dinner to mark its impending closure. *Today in meetings, the Albemarle Architectural Review Board meets at 1 p.m. (meeting info)Sheetz is building a convenience store in the northwest quadrant of the U.S. 29 and Airport Road intersection and needs design approval. The ARB did not grant an approval at its June 1 meeting. Should the current members of Charlottesville city council wait until after the November 3, 2021 Charlottesville city council election to appoint a new Charlottesville city manager? Remember, both Mayor Nikuyah Walker and Councilor Heather Hill are up for reelection on November 3, 2021. If a city manager is appointed before this November 3, 2021 election, will this new hire be a lameduck city manager with two of five council spots possibly changing less than one year after his/her appointment? 2. Steve Sakala is the co-founder and CEO of Mana Artisan Botanics. He has been a voice for sustainability, regenerative agriculture and medical cannabis for more than two decades. He was educated about the numerous benefits of the cannabis plant at a young age and quickly became passionate about its thousands of uses.  As part of a growing movement of companies that want to create a better world, Mana Artisan Botanics is dedicated to crafting pure, chemical and pesticidefree, whole-plant botanicals. They are committed to empowering consumers with evidence-based data on the potential health benefits of phytocannabinoids and leading as a force for good in the rapidly growing cannabis industry. Background Steve’s interest in cannabis stemmed from his interest in the environment and becoming an environmentalist. Shortly after high school, he felt that is what he wanted to do. He looked at all the environmental challenges we faced and felt the solutions to these challenges were rather simple to implement. Steve considered hemp an important part of these solutions. He was also interested in hemp because of its psychoactive properties. He worked on the first medical marijuana initiative way back in 1992. In the process of learning about hemp and its industrial uses as well as cannabis and its medicinal uses, he became devoted to getting the word out about the potential of cannabis on so many different levels. His major education has been on natural resources. His focus has always been on sustainable resources, renewable energy, green building materials etc. Can you tell us about how health plays a role in your mission? The fact that cannabis has a great potential to deal with health issues and major diseases is something that keeps Steve motivated. He has been in the industry for over 20 years and has worked with numerous medical professionals. Cannabis can help us in so many different ways. Cannabis can help cholesterol patients and blood pressure patients. It is extremely affective against inflammation and is also used to treat sleep disorders. Various studies have been conducted and results have shown that cannabis can also be used to treat diseases like Parkinson’s, Cancer and Alzheimer’s. Steve aims to educate people about the use of cannabis other than its usage as a medicine. He does a lot of education around nutrition. Can you tell us a little about our endocannabinoid system? Endocannabinoid system also known as ECS relates to the receptors in our body. ECS has more receptors in the body than any other receptors. Our body produces its own natural cannabinoids called endocannabinoids or endogenous cannabinoids. Our lifestyle has drastically changed in recent years. Due to the modern day toxins, the food we eat, the environment in which we breath and due to our day to day stress, we are not producing enough of our endogenous cannabinoids. Therefore, our indoor cannabinoid system is not playing its role to keep our health in balance. This is called chronic endocannabinoid deficiency. This is resulting in autoimmune and degenerative diseases among a lot of people. Through plants, mainly cannabis, we can try to fulfill the role that endogenous cannabinoids would have played. Tell us a little about your garden. After college, Steve left for Africa and spent four years in West Africa focusing on sustainable agriculture. Sustainable agriculture is the foundation of how we can build a sustainable society. Since college, Steve has been interested in growing food and growing medicine. It has become his life’s work. Steve moved to Hawaii about twelve years ago for some work and ended up buying a farm. He was able to grow some very diverse plants and raise animals on the farm as well. His agriculture philosophy is based on diversity. His farm consists of numerous fruit trees; he grows many medicinal plants as well. Steve also has a medicinal cannabis garden. He has been able to grow about 15 to 20 different types of cannabis in the garden. How much time do you put into nurturing your garden? Steve spends at least 5-10 hours a week on his cannabis garden. For his farm, he makes sure he spends about 30-40 hours a week. Can you share someone’s experience of using your product? There have been numerous people who have been able to undergo health transformation after using Steve’s products. These stories inspire Steve to carry on with his work. Most of these stories relate to sleep disorders of how people were able to treat their irregular sleep patterns through these products. Recently, Steve received feedback from a person who recently had a shoulder surgery and was in extreme pain. He used the cannabis products and was extremely surprised with the results. His pain went away in no time and was amazed with the results. Can you tell us more about inflammation and how it affects the body? There are so many health challenges that stem from chronic inflammation. Being able to reduce the amount of internal inflammation is key to being more aligned with how we want to be in our lives. It can help us reduce stress, irritation, pain and anxiety. Steve has extensively worked on his products to make sure they help in reducing inflammation. What is passiflora? It is the flower that grows on the passion vines and passion vines are known as passiflora. Passiflora has been used for hundreds of years for various different purposes. It is known to contain a compound that is very helpful for sleep. It can also help deal with anxiety. Steve has also built a product on top of passion flower. Which one of your own product would you recommend for someone dealing with day to day anxiety or stress? He recommends his most popular product called the ‘Hawaiian Turmeric Hemp Oil’. The reason it is so popular is because it does not taste like cannabis. Many of his clients do not want to feel like they are consuming cannabis. What is the best advice you have ever been given? The best advice that he has been given is to take care of himself. Steve says a lot of people have said this to him in his life. He believes that being a farmer and an entrepreneur, he tends to be work driven. Since he makes products that help people improve their health, Steve realizes that a lot of people are depending on these products. This gives him a greater sense of responsibility and at times he puts himself in a space where he is not able to take good care of himself. How can we connect with you? You can find more about Steve’s work and his products through his website. I was lucky enough to visit Cristina in person to record this episode and we had great fun enjoying a cup of Passion flower tea while we chatted. Passion flower is most commonly used to help with sleep and you'll find it in a lot of over the counter sleep tea mixes. But it's got so much more going on than its sedating properties. We talk about where the name comes from, its association with Jesus and how it has a kinda trippy vibe.

Investigation of interspecific genome-plastome incompatibility in Oenothera and Passiflora

Interspecific genome-plastome incompatibility is a widely observed phenomenon but its primary causes are still unknown. It reflects genome-plastome interactions that play a direct role in speciation processes, such interspecific combinations of nuclear genomes and plastomes that fail to develop fully autotrophic plants which then are usually eliminated by natural selection. We have investigated two plant models displaying genome-plastome incompatibility, Oenothera and Passiflora, using strategies of molecular biology in order to contribute to an analysis of primary causes of interspecific genome-plastome incompatibility. 1. Expressed sequence tags in Oenothera: In this study we present the first analyzed EST data set for Oenothera. 3,532 cDNA sequences derived from 9-week-old Oenothera plantlets were the analysed and assembled into 1,621 nonredundant clusters, including 1,133 singletons and 488 multi-member unigenes which contain a total of 875,940 nonredundand nucleotides. EST sequences were analysed by Sputnik algorithm. They were also used in the development of gene-specific PCR-based codominant markers (SNPs, CAPS, micro-satellites). The cDNA library could be directly used for macroarray applications including gene expression studies and for physical mapping. 2. Genotyping analyses in Oenothera using AFLP technology: The comparison of AFLPs from Oenothera with AFLPs from Arabidopsis was used to obtain an approximation of the genome size. The genotyping data provide evidence that genome of Oenothera is only six times larger than that of Arabidopsis corresponding to a size of about 750 Mb. The AFLP markers were also successfully applied to construct first genetic maps using F2 mapping population of interspecific hybrids between Oenothera elata ssp. hookeri, line johansen, AA-III, x Oenothera grandiflora ssp. tuscaloosa, BB-III. The linkage maps contain 88 AFLP markers covering a total map length of 154.4 cM for dominant markers in johansen, AA-III and 104 AFLP markers and a total size of 155.3 cM for dominant markers in grandiflora, BB-III. In addition, it was possible to assign genome-plastome incompatibility locus to the margin of coupling group 2B with 13 cM distance to the next AFLP marker. SUMMARY 91 The EST project followed by genotyping analysis increases knowledge and requirements in discovering primary causes of genome-plastome incompatibility. Oenothera with genome-plastome incompatibility, chromosomal translocations and many chromosomal arrangements provides an elegant tool in the study of genomeplastome interactions, speciation processes and species evolution. 3. Investigation of genome-plastome incompatibility in Passiflora: We present the first evidence of hybrid bleaching in this genus. The hybrid between Passiflora menispermifolia x Passiflora oerstedii showed bleaching regions during plant development. Reciprocal crosses have also shown hybrid bleaching but as well significant differences in leaf shape. Molecular analyses of cpDNA showed that Passiflora plastids are inherited bi-parentally and that the P. menispermifolia plastome is incompatible in F1 hybrids with P. oerstedii. This is the first evidence of genome-plastome incompatibility in Passiflora, which differ from Oenothera incompatibilities. The analysis of plastid ultrastructure showed that green tissues in the F1 generation have fully developed chloroplasts with thylakoids and grana; the incompatible material in F1 hybrids lacks differentiated plastids and contains plastids with only rudimentary membranes. An unexpected plastid ultrastructure was found in P. menispermifolia. The leaf from plant growing at greenhouse conditions contains plastids in different development stages including etioplasts, which normally develop from proplastids in darkness. Electron micrographs also indicated retardation of grana formation in P. menispermifolia which shows that vesicles could deliver parts of thylakoid components and that they may directly participate in the formation grana stacks. Northern and Western analyses demonstrated that genome-plastome incompatibility affects both transcription and translation, but with differences for nuclear and plastome encoded genes.
