Left unchecked, these harmful attitudes and behaviours will see rates of male violence against women continue to rise. In this panel discussion, three experts consider where these attitudes begin, and the importance of positive male role modelling in addressing them. The problem is evident – now it's time to identify the steps that will lead to systemic, meaningful change. The hype around cryptocurrencies and blockchain art was almost as annoying as the glee with which crypto sceptics welcomed the sudden onset of the crypto winter. But for all the popularity of Bored Apes and Ponzi scheme stories, there seems to have been little serious engagement with the philosophical, political, and aesthetic implications of the blockchain. Abstract: Embryos have a remarkable ability to repair wounds rapidly, with no inflammation or scarring. Embryonic wound healing is driven by the collective movement of the surrounding cells to seal the lesion. During embryonic wound closure, the cells adjacent to the wound polarize the cytoskeletal protein actin and the molecular motor non-muscle myosin II, which accumulate at the wound edge forming a supracellular cable around the wound. Adherens junction proteins including E-cadherin are internalized from the interface with the lesion and localize to former tricellular junctions at the wound margin, in a process necessary for cytoskeletal polarity. Using quantitative live microscopy, we found that the cells adjacent to wounds in the Drosophila epidermis also polarized Talin, a core component of cell-extracellular matrix (ECM) adhesions that links integrins to the actin cytoskeleton. Integrin knock-down and inhibition of integrin binding delayed wound closure and were associated with a reduction in actin levels around the wound. Additionally, disrupting integrins caused a defect in E-cadherin reinforcement at tricellular junctions along the wound edge, suggesting crosstalk between integrin-based and cadherin-based adhesions. Together, our results show that cell-ECM adhesion contributes to embryonic wound repair and reveal an interplay between cell-cell and cell-ECM adhesion in the collective cell movements that drive rapid wound healing. Abstract: The carious lesion is a bacteria caused destruction of tooth mineralized matrices marked by concurrent tissue reparative and immune responses in the dental pulp. While major molecular players in tooth pulp decay have been uncovered, a detailed map of the molecular and cellular landscape of the diseased pulp is still missing. Here we used single-cell RNA sequencing analysis, to generate a comprehensive single-cell atlas of the carious human dental pulp tissue. Our data demonstrated modifications in various cell clusters of the carious pulp, such as immune cells, mesenchymal stem cells (MSC) and fibroblasts, when compared to the healthy dental pulp. These changes include upregulation of genes encoding extracellular matrix (ECM) components and the enrichment of the fibroblast cluster with myofibroblasts. Assessment of the Fibronectin fibres' mechanical strain showed a significant tension reduction in the carious human pulp, compared to the healthy one. Collectively, the present data demonstrate molecular, cellular and biomechanical alterations in the carious pulp tissue, indicative of extensive ECM remodelling and reminiscent of fibrosis observed in other organs.   A woman's weight and BMI during and after pregnancy is often discussed and debated. This is especially true in the case of high-weight women, who, as research shows, have a  higher risk of complications like cesarean birth.  Today's episode is to discuss high weight women in pregnancy and when we need to concerned and when we don't  PLUS, we also discuss some very questionable myths and treatments followed for high-weight women that you need to be aware of. But these in no way imply that people with very low body weight have uncomplicated pregnancies. To prove that, we compare pregnancy risks, body weight gain, and recovery in low-weight, normal weight and high-weight pregnant women and see clearly why a lower than ideal body weight could also pose significant birth complications. PS. Excessive weight is sometimes associated with ovulation issues. That, in turn, could be caused due to PCOS (PolyCystic Ovarian Syndrome), which is associated with a hormonal imbalance that messes up your ability to metabolize. Here are a few known complications faced by high-weight women during pregnancy: >> Preeclampsia -- Condition associated with high blood pressure and other complications, including retaining fluid and end-organ dysfunction. In extreme cases, it can lead to eclampsia or seizures, a severe form of preeclampsia. >> Diabetes (pregestational and gestational) -- Diabetes that's detected during the first trimester or the beginning of the second trimester is called pregestational diabetes. These can lead to abnormalities of development and more chances of miscarriages. Gestational diabetes is a form of diabetes that's developed in women during pregnancy. Women who have this condition wouldn't have had diabetes previously. >> Preterm delivery -- A condition that might cause pregnant women to go into labor early. >> Macrosomia -- High weight of the baby >> Higher weight loss -- High-weight women tend to lose more weight during pregnancy, which is unhealthy. It's healthy to end a pregnancy with a greater weight than what they started with. >> High risk of C section -- High-weight women are at a 70 % risk of a C-section -- even after complications like hypertension and diabetes have been neutralized. >> Less sensitivity to oxytocin -- Hence an increased need for oxytocin augmentation. >> The need to use IPCs (Intrauterine Pressure Catheter) -- a device placed inside the pregnant woman's uterus to monitor uterine contractions during labor. >> Pelvic outlet is narrower -- This is alleviated by making positional changes. >> Uterine tilt if a woman previously had high weight and lost it abruptly -- This is alleviated by making positional changes. >> Remodelling of bones -- As the bones become more narrow, changes are caused in getting the baby. >> Shoulder dystocia and postpartum hemorrhage -- Massage from postpartum hemorrhage is also harder. >> Difficult labor -- Caused due to positional issues of internal organs and the baby A common pregnancy complication seen (especially) in low-weight women: >> Hyphemeresis -- A condition that causes severe metabolic disturbances, including abnormal amounts of nausea and vomiting. A self-described computer nerd, Dave Cooper was always fascinated by the way technology could push fundamental science research forward.    After bone density scans became commonplace in the 1980s, Cooper watched imaging technology and forensic science begin to advance.   Today, his laboratory uses the Canadian Light Source to take detailed snapshots of bones evolving and changing in real time, honing treatments and improving outcomes for patients with osteoporosis.    “People are living longer, and part of living longer and healthier lives will be protecting the skeleton,” said Cooper, who says unraveling the mysteries inside bones and their evolution over time can have ramifications across health disciplines, even for space travel.   “Now, we can watch it as it's happening rather than using a blunter instrument after the fact," he said.

Abstract: Corpus callosum dysgenesis (CCD) is a congenital disorder that incorporates either partial or complete absence of the largest cerebral commissure. Remodelling of the interhemispheric fissure (IHF) provides a substrate for callosal axons to cross between hemispheres, and its failure is the main cause of complete CCD. However, it is unclear whether defects in this process could give rise to the heterogeneity of expressivity and phenotypes seen in human cases of CCD. We identify incomplete IHF remodelling as the key structural correlate for the range of callosal abnormalities in inbred and outcrossed BTBR mouse strains, as well as in humans with partial CCD. We identify an eight base pair deletion in Draxin and misregulated astroglial and leptomeningeal proliferation as genetic and cellular factors for variable IHF remodelling and CCD in BTBR acallosal strains. Eukaryotic genomes are condensed into a multilevel structure called chromatin which serves to organize and package the DNA, but at the same time needs to be flexible to permit regulated access to the stored information. ATP-dependent chromatin remodelling factors largely contribute to this dynamic nature of chromatin by catalysing processes such as the disruption of histone-DNA contacts, nucleosome repositioning and histone exchange. ATP-dependent remodelling has been well documented on a mononucleosomal level, but little is known about its regulation in a more physiological chromatin environment, where neighbouring nucleosomes and linker histones might interfere with the remodelling reaction. If and to what extent remodelling can work on chromatin bound by linker histones remains controversial, in spite of their high abundance and their strong influence on chromatin folding. We therefore investigated chromatin remodelling in the presence of linker histones H1 or H5 using regularly spaced, oligonucleosomal substrates reconstituted from purified components. Surprisingly, we found that both the remodelling complex ACF – consisting of the ATPase ISWI and the regulatory subunit ACF1 – and ISWI alone were able to catalyse the repositioning of entire chromatosomes (nucleosomes + H1). Linker histones inhibited their remodelling activity by only about 50%. In contrast, the related ATPase CHD1 remodelled chromatin only in the absence of linker histones, suggesting that linker histones allow remodelling by selected factors only. In addition, our data indicate that repositioning in the presence of H1 might be unidirectional. ACF1 is abundant during early Drosophila development, when H1 gradually replaces its early placeholder HMG-D. HMG-D binds to chromatin less tightly than H1 and unlike the latter, did not affect the remodelling activity of ACF in our assay. H1 was able to displace HMG-D from and bind to our reconstituted arrays without the help of cofactors. Strikingly, both H1 and HMG-D are more abundant in embryonic nuclei of acf1 null flies compared to the wild-type, raising the possibility that an ACF1-containing complex controls linker histone incorporation.

Die koronare Herzkrankheit (KHK) stellt momentan die häufigste Krankheits- und Todesursache in Europa dar. Häufig kommt es als Folge einer KHK zum akuten Myokardinfarkt mit den gefürchteten Folgen, wie kardiogenem Schock und plötzlichem Herztod. Während sich die etablierten konservativen Therapiestrategien bislang auf eine Verminderung des pathologischen Remodellings beschränken, gewinnt die Forschung an alternativen Therapiemöglichkeiten zur längerfristigen Regeneration des geschädigten Myokards zunehmend an Bedeutung. Ein neuer bisher noch nicht zur Therapie des Herzinfarkts eingesetzter Kandidat könnte das Parathormon (PTH) sein. Dessen Fragment PTH (1-34) befindet sich bereits seit Jahren im klinischen Einsatz zur Bekämpfung schwerer Osteoporosen. Im Tiermodell verbesserte PTH durch Steigerung des myokardialen Blutflusses die Herzfunktion und verhinderte dadurch die Ausbildung eines kardiogenen Schocks. Kürzlich konnte darüber hinaus im Mausmodell gezeigt werden, dass PTH die Stammzellnische im Knochenmark reguliert. So führte die PTH-Gabe zu einem Anstieg verschiedener Stammzellpopulationen im Knochenmark und verminderte bei bestrahlten Mäusen nach Knochenmarktransplantation signifikant deren Mortalität. Ziel dieser Arbeit war es, den Einfluss von PTH (1-34) auf die Mobilisation von Knochenmarkstammzellen ins periphere Blut sowie mögliche Effekte auf Pumpfunktion und Remodelling nach akutem Myokardinfarkt im Mausmodell zu untersuchen. Die Behandlung mit PTH (1-34) führte zu einer Mobilisation verschiedener Stammzellpopulationen aus dem Knochenmark ins periphere Blut. Dabei kam es nach PTH-Gabe im Gegensatz zu Granulocyte-colony stimulating factor (G-CSF) nicht zum Abfall von CD45+/CD34+ Stammzellen im Knochenmark. Nach chirurgisch induziertem Myokardinfarkt führte die Gabe von PTH zu einer signifikanten Abnahme der Mortalität im Vergleich zu den Kontrolltieren. Dies war bei den PTH-Tieren assoziiert mit einer signifikanten Verbesserung der globalen Herzfunktion. So waren das Herzzeitvolumen und die Auswurffraktion nach PTH-Gabe deutlich gesteigert. Wir konnten anhand einer erniedrigten arteriellen Nachlast in den hämodynamischen Untersuchungen zeigen, dass PTH am arteriellen Gefäßbett zu einer Vasodilatation führt. Über diesen bekannten Einfluss von PTH auf den arteriellen Gefäßwiderstand kommt es zu einem gesteigerten myokardialen Blutfluss. Dadurch verbessert PTH in der Akutphase nach akutem Myokardinfarkt die Herzfunktion und schützt vor einem akuten Herzversagen. Auf histologischer Ebene fanden sich nach PTH-Behandlung kleinere Infarktgrössen und eine verminderte Abnahme der linksventrikulären Vorderwand im Vergleich zu den Kontrolltieren. Diese Veränderungen im Remodelling nach PTH-Behandlung waren durch eine Zunahme von CD31+ Kapillaren in der Grenzzone um den Infarkt (Borderzone) erklärbar. Die Gefäßneubildungen waren assoziiert mit einer gesteigerten Expression von Vascular Endothelial Growth Factor (VEGF) sowie des Insulin like Growth Factor-1 (IGF-1)-Rezeptors in der Borderzone. PTH bewirkt somit entweder direkt oder indirekt über die Mobilisation von Knochenmarkstammzellen eine vermehrte Sekretion von vaskulären Wachstumstumsfaktoren wie VEGF und IGF-1. So kommt es nach akutem Myokardinfarkt im Mausmodell zu einem abgeschwächten „Remodelling“ mit konsekutiver Verbesserung der myokardialen Pumpfunktion. Neben weiterer Untersuchungen bezüglich der Mechanismen, über die PTH zu den gezeigten Veränderungen im kardialen Remodelling führt, müßte in einem nächsten Schritt geklärt werden, ob PTH (1-34) beim Menschen in der zur Osteoporosebehandlung üblichen Tagesdosis von 20-40 µg oder in konsekutiv höheren Dosierungen zur Freisetzung verschiedener Populationen von Knochenmarkstammzellen ins periphere Blut führt (Phase I Studie).

Eine neue Methode zur Dampfsterilisierung von Knochenmaterial unter Erhalt der mechanischen Steifigkeit und Osteokonduktivität

Knochenaufbau mittels Biomaterialien und Transplantaten gehört zur Standardmethodologie in vielen chirurgischen Fachgebieten. Dampfsterilisiertes Knochenmaterial zeigt, trotz Attraktivität der Entkeimungsmethode überwiegend ein Verknöcherungsversagen bei stark verminderter Steifigkeit. Es wurde in dieser dreigeteilten Studie ein neues Knochentransplantatmaterial untersucht, Powerpore® duro, welches seine Steifigkeit bei einer Dampfsterilisierung nicht verliert,. Das Material zeigte eine primäre Osseointegration und eine lamelläre Knochenneubildung. In einer geschlossenen Tierversuchsstudie an 17 Kanninchen wurde am Patellagleitlagermodell das Einwachsverhalten des neuen Materials untersucht. Nicht entkalkte Knochenhistologien zeigen nach 30 (n=5) und 100 (n=12) Tagen eine regelrechte Osseointegration in allen untersuchten Proben. Hierbei waren die allogenen Powerpore® duro Transplantate im Vergleich zur allogenen, unbehandelten Kontrollgruppe (frischer Knochen) adäquat im Hinblick auf Einwuchstiefe, Resorption und Remodelling. Die Technologie ist eine Möglichkeit, Knochenmaterial einfach und kostengünstig zu sterilisieren, und bietet eine alternative Möglichkeit zum Führen einer Knochenbank.
