Künstliche Intelligenzen werden heute schon bei der Diagnose von malignen Melanomen eingesetzt. Sie helfen bei der Entscheidung, ob biopsiert werden soll oder nicht. Eine Entscheidung, die aber in letzter Instanz der Dermatologe/die Dermatologin trifft. Solche Entscheidungen erfolgen aber nicht gleichmäßig. Manche Ärzte sind vorsichtiger, andere weniger. Manche Patienten lehnen Biopsien eher ab, andere sind ohne Zögern dazu bereit. Wie kann man das diese subjektiven Faktoren ausschalten oder zumindest dämpfen? An der Abteilung für Dermatologie der Medizinischen Universität Wien hat ein Team um Prof. Dr. Harald Kittler ein KI so gestaltet, dass sie aus richtigen Biopsie-Entscheidungen lernt—und zwar durch Feedback des behandelnden Arztes. Abstract: Light Field Microscopy (LFM) is an imaging technique that offers the opportunity to study fast dynamics in biological systems due to its rapid 3D imaging rate. In particular, it is attractive to analyze neuronal activity in the brain. Unlike scanning-based imaging methods, LFM simultaneously encodes the spatial and angular information of light in a single snapshot. However, LFM is limited by a trade-off between spatial and angular resolution and is affected by scattering at deep layers in the brain tissue. In contrast, two-photon (2P) microscopy is a point-scanning 3D imaging technique that achieves higher spatial resolution, deeper tissue penetration, and reduced scattering effects. However, point-scanning acquisition limits the imaging speed in 2P microscopy and cannot be used to simultaneously monitor the activity of a large population of neurons. This work introduces a physics-driven deep neural network to image neuronal activity in scattering volume tissues using LFM. The architecture of the network is obtained by unfolding the ISTA algorithm and is based on the observation that the neurons in the tissue are sparse. The deep-network architecture is also based on a novel imaging system modeling that uses a linear convolutional neural network and fits the physics of the acquisition process. To achieve the high-quality reconstruction of neuronal activity in 3D brain tissues from temporal sequences of light field (LF) images, we train the network in a semi-supervised manner using generative adversarial networks (GANs). We use the TdTomato indicator to obtain static structural information of the tissue with the microscope operating in 2P scanning modality, representing the target reconstruction quality. We also use additional functional data in LF modality with GCaMP indicators to train the network. Our approach is tested under adverse conditions: limited training data, background noise, and scattering samples. We experimentally show that our method performs better than model-based reconstruction strategies and typical artificial neural networks for imaging neuronal activity in mammalian brain tissue, considering reconstruction quality, generalization to functional imaging, and reconstruction speed. De origem austríaca, ele vem de uma família de classe média alta de São Paulo, estudou em boas escolas, teve oportunidades que a maioria das pessoas nem sonha e mesmo assim, sua vida não seguiu o rumo dito tradicional. Foi velejador e tentou a carreira de tenista, mas foi mesmo o triathlon que o fez ir bem longe. Em busca de viver plenamente a experiência da modalidade ele foi estudar inglês em San Diego, meca do esporte nos anos 90 e uma cidade que acabou o conquistando. Entre um treino e outro, uma prova e outra, ele conseguiu um estágio na famosa revista Triathlete, o que viria a criar uma grande oportunidade profissional para ele. Curiosamente, não como ele esperava. Antenado nas novidades e oportunidades, ele conseguiu o sinal verde dentro da editora para criar o que viria a se tornar o primeiro site de divulgação e inscrições de provas de triathlon e corridas de rua dos EUA. De volta ao Brasil, adaptou essa idéia para o mercado nacional e criou o ativo.com, que por muito tempo dominou o mercado das inscrições online por aqui. Depois de 12 anos à frente do negócio, resolveu que era hora de partir para novas empreitadas. Abstract: The K+/Clco-transporter KCC2 (SLC12A5) allows mature neurons in the CNS to maintain low intracellular Cl- levels that are critical in mediating fast hyperpolarizing synaptic inhibition via type A -aminobutyric acid receptors GABAARs. In accordance with this, compromised KCC2 activity results in seizures but whether such deficits directly contribute to the subsequent changes in neuronal homeostasis and viability that lead to epileptogenesis, remains to be assessed. Here, we have investigated these issues using conditional ablation of SLC12A5 gene expression with AAVs. Decreasing KCC2 expression resulted in the rapid activation of the extrinsic apoptotic pathway, but not the intrinsic pathway, in mature hippocampal neurons, in vitro and in vivo. Intriguingly, direct pharmacological inhibition of KCC2 in mature hippocampal neurons was also sufficient to activate the apoptotic pathway within 10 minutes. Finally, ablating KCC2 expression during neuronal development had no discernable effects on neuronal development, viability and apoptosis. However, KCC2 was essential for the postnatal development of GABAergic inhibition. Collectively, our results demonstrate that correct KCC2 expression and activity play a critical role in neuronal survival. Perturbed KCC2 activity initiates the extrinsic pathway of apoptosis but has a minimal impact on regulating neuronal development. Thus, increasing KCC2 activity may be a useful strategy to limit neuronal death that underlies epiletogenesis. Copy rights belong to original authors. Visit the link for more info

Abstract: The spatiotemporal distribution of mitochondria is crucial for precise ATP provision and calcium buffering required to support neuronal signaling. Fast-spiking GABAergic interneurons expressing parvalbumin (PV) have a high mitochondrial content reflecting their large energy utilization. The importance for correct trafficking and precise mitochondrial positioning remains poorly elucidated in inhibitory neurons. Miro1 is a Ca2+-sensing adaptor protein that links mitochondria to the trafficking apparatus, for their microtubule-dependent transport along axons and dendrites, in order to meet the metabolic and Ca2+-buffering requirements of the cell. Here, we explore the role of Miro1 in PV+ interneurons and how changes in mitochondrial trafficking could alter brain network activity. By employing live and fixed imaging, we found that the impairments in Miro1-directed trafficking in PV+ interneurons altered their mitochondrial distribution and axonal arborization. These changes were accompanied by an increase in the ex vivo hippocampal {gamma}-oscillation (30 - 80 Hz) frequency and promoted anxiolysis. Our findings show that precise regulation of mitochondrial dynamics in PV+ interneurons is crucial for proper neuronal signaling and network synchronization. Copy rights belong to original authors. Abstract: Microglia use a highly complex and dynamic network of processes to sense and respond to their surroundings. Microglial dynamics differ throughout development and in neurological and neuropsychiatric disease, though mechanistic insight into these changes is lacking. Here we identify novel roles for regulators of the actin cytoskeleton in controlling microglial behaviour. We show that the actin branching complex Arp2/3 is critical for maintaining microglial morphology and required for surveillance but not chemotactic motility. Neuropsychiatric disease-associated Cyfip1, a core component of the WAVE regulatory complex that links Rac1 signalling to Arp2/3 activation, is highly expressed in microglia but has unknown function. We report that conditional deletion of Cyfip1 in mouse microglia impairs morphological complexity and surveillance of brain parenchyma, and increases activation state as defined by CD68 expression. Thus, altered actin-dependent microglial dynamics mediated by Cyfip1 and Arp2/3 may contribute to neuropsychiatric disease. Copy rights belong to original authors. Visit the link for more info

We are fortunate to have engaged the esteemed Professor Harald Kittler from the Medical University of Vienna in Austria and Associate Professor Caterina Longo from the University of Modena and Reggio Emilia in Italy to discuss their involvement with dermoscopy. ----more---- Both speakers offer their unique viewpoints and advice on dermoscopy, with Professor Kittler recounting his very early experiences with the technology during its development phase as a huge stereo microscope at the Medical University of Vienna in 1986. Associate Professor Longo came to dermoscopy more recently in 2012 by way of the large clinic where she worked in Italy. She was greatly inspired by the enthusiasm of her mentors there, which led to her focus on dermoscopy both academically and clinically.  This podcast episode reflects on the remarkable changes in dermoscopy over time and highlights our current rapid move into a phase of digitised dermoscopy.

Friedrich Kittlers „Grammophon. Film. Typewriter“ beginnt mit einer lakonisch-verdichteten Formel, welche das Programm einer im Veröffentlichungsjahr 1986 gerade zu sich kommenden Medienwissenschaft auf den Punkt bringt. Kittlers Satz lautet: “Medien bestimmen unsere Lage.” Der Vortrag wird den Hintergrund der von Kittler postulierten Bestimmtheit der Lage durch die Medien im literarisch-philosophischen Kontext der 1930er und 40er Jahre nachzeichnen. So wurde im Umkreis von Phänomenologie, philosophischer Anthropologie und Kulturanthropologie (Heidegger, Jaspers, Rothacker) die Diskussion der Differenz von Situation und Lage zu einem zentralen Ort, die existentielle Verfasstheit des Menschen als ein welterschließendes Wesen zu denken. Die paradigmatische Verdichtung zur zitierbaren Formel “Erkenne die Lage” geschieht dann im Werk Gottfried Benns, für den sich die militärische Lagebestimmung zum medientechnologisch vermittelten Existential des modernen Menschen verwandelt.

In February, Laurence Rickels stopped by Austin, Texas. Dr. Rickels, who is the Sigmund Freud Professor of Psychoanalysis at the European Graduate School as well as Professor of Art and Theory at the Academy of Fine Arts in Karlsruhe, Germany, was in town as part of the tour for his latest book: Germany: A Science Fiction. During his visit, he also swung by UT-Austin's Digital Writing and Research Lab and was generous enough to sit down for the following interview.In his new book, Rickels focuses on psychopathy as, quote, "the undeclared diagnosis implied in flunking the empathy test." He does so via an exploration of Germany's role in Cold War-era science fiction: from the Thomas Pynchon novel Gravity's Rainbow to B movies like 1962's The Day of the Triffids to the science fiction of Philip K. Dick. In addition to Germany, Dr. Rickels has written numerous works tracing connections between psychoanalysis, popular culture, critical theory, science fiction, and mourning. His books include The Case of California, The Vampire Lectures, a three-volume series entitled Nazi Psychoanalysis, and Spectre, in which Rickels turns his attention to Ian Fleming's James Bond. He's also the author of a recent article entitled "The Race to Fill in the Blanks: On (Animal) Testing in Science Fiction," which appeared in the 2014 issue of Philosophy & Rhetoric touched on in this podcast's premiere episode.In our conversation, I ask Dr. Rickels about his use of the term "psy-fi," the impetus behind his new book, the relationship between his work and that of the late media theorist Friedrich Kittler, as well as the puns and juxtapositions that punctuate his pages. Götter und Schriften rund ums Mittelmeer. Symposion in memoriam Friedrich Kittler | Symposium Fr, 19.10.2012 – Sa, 20.10.2012, ZKM_Medientheater Die Schrift ist das erste Medium. Das ist der Sinn der Behauptung: Am Anfang war das Wort. Mit der Schrift begann jene Kette von Medienrevolutionen, die uns von Runen und Piktogrammen über musikalische und mathematische Notationen bis zu den Symbolen auf den grafischen Benutzeroberflächen von Computermenüs führte. Die Schrift ist das Medium der Absenz und gemäß Freud (Das Unbehagen der Kultur, 1930) setzt die Technik die Arbeit der Schrift fort. Technik ist also ebenfalls ein Medium der Absenz. Deswegen lautet meine These: Alle Technologie ist Teletechnologie (Telefax, Telefon, Television etc.) und alle Teletechnologie ist Theotechnologie. Jede Medienrevolution ist daher nicht nur eine technische, sondern auch eine soziale, erkenntnistheoretische und sogar religiöse. Die Religion ist gewissermaßen ein Effekt der Medien. Die Schrift ist noch immer ein zentrales Medium und wegen ihrer Abhängigkeit von der Schrift, von der Bibel zum Koran, ist Religion selbst ein Medium (s. Boris Groys, Peter Weibel, Das Medium Religion, 2011). Die symbolische Ordnung aller Schrift- und Bildzeichen ist offensichtlich deswegen so wichtig, weil sie die soziale Ordnung nicht nur spiegelt, sondern auch mitkonstruiert. Daher war ich sofort fasziniert, als mir Kittler in Gesprächen andeutete, er hätte eine neue Theorie, wieso Jesus von Nazareth ermordet wurde. Als Ergebnis neuer Reflexionen zur Entstehung der Schriftkultur als Beginn aller Medientheorie hat Kittler mir seine Theorie über die Rolle Jesus’ von Nazareth als Medienrebell skizziert. Peter Weibel, 1944 in Odessa geboren, studierte Literatur, Medizin, Logik, Philosophie und Film in Paris und Wien. Durch seine vielfältigen Aktivitäten wurde er eine zentrale Figur in der europäischen Medienkunst. Seit 1984 ist er Professor an der Universität für Angewandte Kunst in Wien, 1984-1989 war er Professor für Video und Digitale Kunst am Center for Media Study an der State University of New York in Buffalo. 1989 gründete er das Institut für Neue Medien an der Städelschule in Frankfurt, das er bis 1995 leitete. Von 1986 bis 1995 war er künstlerischer Leiter der Ars Electronica in Linz und von 1993 bis 1999 Österreichs Kommissär der Biennale von Venedig. Von 1993 bis 2011 Chefkurator der Neuen Galerie in Graz, seit 1999 ist er Vorstand des ZKM | Karlsruhe. 2008 leitete er die Biennale von Sevilla (Biacs3). 2007 wurde ihm die Ehrendoktorwürde der University of Art and Design Helsinki verliehen, 2008 das französische Ehrenzeichen „Officier dans l'Ordre des Arts et des Lettres“. 2011 war er künstlerischer Direktor der 4. Moskau Biennale für zeitgenössische Kunst. In seinem Lebenswerk hat Medientheoretiker Friedrich Kittler (1943−2011), die Geschichte der Dichtung, der Philosophie, ja der Kultur als solche vom Kopf auf die Füße ihrer technischen und vortechnischen Medien gestellt. Was Aufschreibesysteme für die Literatur, was Befehlssätze für programmierbare Maschinen, das ist den Göttern das elementarste Medium im lateinischen Wortsinn von elementa: Buchstaben. Das Symposion »Götter und Schriften rund ums Mittelmeer«, noch zu Lebzeiten von dem deutschen Medientheoretiker Friedrich Kittler selbst vorbereitet, widmet sich dieser Hypothese. Wie bestimmen die Kontakte, Konkurrenzen, Innovationen der verschiedenen Schriften und Alphabete seit der frühesten Antike rund ums Mittelmeer die zukünftigen Geschicke des Abendlands? Seit dem Neolithikum gibt es im Mittelmeerraum Kulturen, deren Alphabete eng an Verwaltung und Handel, Befehlsflüsse und Gesetze gebunden sind, aber auch eine Kultur, die ihr Alphabet aus dem Schreiben von Musik und Gesangsvortrag, Vers und Götteranrufung schöpft. Einige Schriften des Mittelmeers − in Keilen, Bildschriftzeichen, Silbenschriften dargestellt − sind graphisch orientiert. Sie schreiben von den Worten der Sprache meist Konsonanten oder Konsonantengruppen. Andere, wie das griechische Alphabet, das als erstes der Welt auch Vokale schreibt, sind phonetisch orientiert und damit prinzipiell auf jede Sprache übertragbar. Mächtige Gesetzesgötter einerseits strafen und befehlen. Der Verkehr mit ihnen wird gesetzlich geregelt. Andererseits gibt es Götter, die an- und abwesend sind. Der Verkehr mit ihnen wird nicht verwaltet, sondern begangen. Wie sind diese verschiedenen Ausgestaltungen der Götterwelten in den Schriftsystemen widergespiegelt?

Nur Atavismen wie das Urheberrecht, das ja nicht umsonst aus der Goethezeit stammt, zwingen noch zur Namensnennung von Textern und Komponisten (als ob es dergleichen im Soundraum gäbe). Viel eher wären die Schaltpläne der Anlagen und […] die Typennummern der eingesetzten Synthesizer aufzuführen.“ (Friedrich A. Kittler, 1975) In den frühen 1980er Jahren baute Friedrich A. Kittler einen modularen Synthesizer in 5 Makro-Modulen/Gehäusen. Fünf Kuben aus gebürstetem Aluminium – heute golden nikotinbrüniert – in smartem, minimalistischem Post-Braun-Design sieht man auf einem Portraitfoto hinter Kittler in seinem Regalsystem von Dieter Rams stehen. Daneben findet man Bücher und die weißen Elementen der Atelier-Anlage von Braun (auch unter Mitwirken von Dieter Rams entworfen) sowie einen elektrostatischen Kopfhörer der japanischen Marke Stax. Seit Kittlers Tod am 18. Oktober 2011 verbleibt der Synthesizer zusammen mit seinen Aufzeichnungen und Datenträgern als Teil seines Nachlasses am Deutschen Literaturarchiv in Marbach. apparatus operandi ist ein konzeptuelles Kunstprojekt von Jan-Peter E.R. Sonntag aus Setzungen verschiedener Formate. Im Zentrum des ersten Teiles steht die Anatomie des Synthesizers Friedrich A. Kittlers. Das Projekt stellt die wahrnehmungsgenerative Potenz von Apparaten in Frage und argumentiert in allen Formaten archäologisch, historisch und sinnlich; Hardware nah am Prozess, am Werden orientiert, reformuliert es eine Absage an überkommene Substanzbegriffe. apparatus operandi1::anatomie// Der Synthesizer des Friedrich A. Kittler ist als Entwurf ein Vorwurf an veraltete Institutionen, die uns in einer Art von Trägheit immer noch dienen. Dem bei der Anatomie eröffneten Gefüge - Epistemologie von Schaltungen, Kittlerphilologie, Gedächtnispraxis - sinnt Sebastian Döring nach. Sebastian Döring, geboren 1977, konzeptioniert und veranstaltet seit 2005 fröhliche Wissenschaft im Medientheater, ergründet Wissenskonfigurationen und betreibt Medienarchäologie und -epistemologie; seit 2009 am Medienarchäologischen Fundus der Humboldt-Universität zu Berlin. Seine Forschungsschwerpunkte sind die Archäologie des neuzeitlichen Subjekts, die Mediengeschichte gesetzten Rechts  und die Analyse epistemischer Dinge. http://www.medientheater.org/ Jan-Peter E.R.
