Abstract: The endoplasmic reticulums (ER) structure is directly linked to the many functions of the ER but its formation is not fully understood. We investigate how the ER-membrane curving protein reticulon 4 (Rtn4) localizes to and organizes in the membrane and how that affects local ER structure. We show a strong correlation between the local Rtn4 density and the local ER membrane curvature. Our data further reveal that the typical ER tubule possesses an elliptical cross-section with Rtn4 enriched at either end of the major axis. Rtn4 oligomers are linear-shaped, contain about five copies of the protein, and preferentially orient parallel to the tubule axis. Our observations support a mechanism in which oligomerization leads to an increase of the local Rtn4 concentration with each molecule increasing membrane curvature through a hairpin wedging mechanism. This quantitative analysis of Rtn4 and its effects on the ER membrane result in a new model of tubule shape as it relates to Rtn4. Copy rights belong to original authors. Ron & Adam catch up with Alex Baddeley, a friend of Ron's, who joined the duo on Day 183 in Invercargill. Alex has had the opportunity to cycle across his country and has acknowledged that it really is one of the most incredible ways to see New Zealand. He was able to adapt to the cycling routine quickly and has proven to be a great support with all the pronunciation of places in New Zealand. Ron & Adam talk through their route from Invercargill, one of the most southern parts of the world that the trio have been to and started making their way towards Auckland, where they will hand over the official Rugby World Cup whistle to referee, Maggie Cogger-Orr. The weather has been temperamental but towns that they have camped in and visited have been beautiful. On their way to Auckland, they were able to visit Wellington Girls College - which is where the first ever female game of rugby was played - which was a special stop on our way to the Women's Rugby World Cup a week later. They also were able to visit the New Zealand Rugby Museum, which brought back a few great memories for Ron. There is a lot of other interesting history, especially seeing the old official whistle which Ron & Adam were able to compare theirs to! This whole journey is to raise awareness and funds for ChildFund Rugby, an amazing charity that promotes community growth and development through sport. And as Cameron likes to say, "Only around 2% of the golf population can make all the technical stuff work for them. Everyone else needs a better way". For the last 25 years Cameron has been writing and thinking deeply about the golf learning process. He has been on a mission to make golf learning easier and more fun for the other 98%. We discuss how he learned to play the game. A unique journey of self discovery Going from shooting in the high hundreds to sub par rounds without any kind of formal instruction. Then after this stellar progress he took some lessons and began to really struggle to implement the positional theories into his game. He got really bogged down with the theories and began to fall out of love with the game. Then the discovery of the ‘Inner Game of Golf' by Timothy Gallwey and how the ideas in the book transformed his approach to the game and led to a lifelong fascination with the learning process in golf. Cameron details the work he did with Aaron Baddeley on his putting. Sue Gathercole is a cognitive psychologist with interests in memory, learning and language. Until 2018 she was the Director of the MRC Cognition and Brain Sciences Unit (CBU) in Cambridge and is now based in Developmental Psychiatry at the University of Cambridge. In this podcast Sue talks about a classic paper in which she examined how memory for words was affected in children with language disorders and whether this might provide an explanation for those language problems. On 28th September 1985 Lee Lawrence’s mother Cherry Groce was shot by police during an armed raid on her Brixton home. Lee Lawrence talks to Samira Ahmed about his Costa Biography award winning memoir The Louder I will Sing in which he recounts the devastating impact the shooting had on the family’s life and his courageous fight for justice. As British musicians warn that costly post-Brexit bureaucracy could decimate European touring, we discuss the potential impact of the recent Brexit Trade Deal on the music industry. With Deborah Annetts from the Incorporated Society of Musicians, Mark Pemberton from the Association of British Orchestras and conductor Paul McCreesh, founder of the Grammy award-winning baroque ensemble, the Gabrieli Consort. Actor Robert Baddeley, a member of David Garrick’s company at Theatre Royal Drury Lane, created a tradition when he died in 1794. In his will, he left £100 to be invested and each year, the money from that sum be spent on “the purchase of a twelfth Cake or Cakes and Wine and Punch or both of them which it is my request the Ladies and Gentlemen performers of Theatre Royal Drury Lane will do me the favour to accept on twelfth night in every year in the Green Room”. Ever since the company playing has enjoyed Baddeley's largesse on January 6th. Abstract: Localization-based super-resolution microscopy techniques such as PALM, STORM, and PAINT are an increasingly critical tool for biological discovery. These methods generate lists of single fluorophore positions that capture nanoscale structural details of subcellular organisation, but to develop biological insight, we must post-process and visualize this data in a meaningful way. A large number of algorithms have been developed for localization post-processing, transforming point data into representations which approximate traditional microscopy images, and performing specific quantitative analysis directly on points. Implementations of these algorithms typically stand in isolation, necessitating complex workflows involving multiple different software packages. Here we present PYMEVisualize, an open-source tool for the interactive exploration and analysis of 3D, multicolor, single-molecule localization data. PYMEVisualize brings together a broad range of the most commonly used post-processing, density mapping, and direct quantification tools in an easy-to-use and extensible package. What does behavioral economics and finance have to do with understanding teens’ decision-making? More than you’d think. Michelle Baddeley’s research in Copycats and Contrarians: Why We Follow Others...and When We Don’t, helps explain why people copy each other and why they rebel: a common focal point in parent/teen conversations. With the best of intentions, parents often struggle to help their teen develop a critical perspective when navigating mature decisions. But by considering the various internal and external forces at play in their teens’ experience, parents can better guide their teens to making healthier decisions. Much of what we know about decision making pivots on social learning, or the theory that new behaviors are acquired through observation and imitation. Anyone in a new situation is susceptible to this: the less we know about a topic, or the less social cues we have in a given setting, the more apt we are to copy those around us. And the most vulnerable group to this influence is first-year high school/university students. When I asked what wisdom Dr. Baddeley could offer parents who want to help their teens critically combat this influence, Dr. Baddeley focused on the importance of shared trust, communication, and anecdotes. By illuminating the consequences of a misguided decision more tangibly, Dr. Baddely noted, parents can help their teen better consider the outcomes without eroding trust. Such illustrations are reinforced in economics: when the implications are imagined more immediately, we can make stronger decisions. Another important topic covered was fruits of contrarians’ labor, and how dissent is actually quite essential to functional families. By striking the balance between unity and rebellion whilst acknowledging their teens’ voice, parents will have an easier time navigating conflict. This too is supported in her research. While contrarians may simply enjoy the risk of being different, they do push the copycats to reconsider their position. The same can be said about rebellious teens. Most of the time, they may be wrong. But sometimes, they’re right. And by squashing their perspective immediately by pulling rank, parents can do more harm than good. Ultimately, though, sometimes the decision is entirely in the teens’ hands. Once they leave the house to go to college, they will have the final say. And the best way Dr Baddely believes parents can support their teens without eroding their trust is to engage them in system 2 levels of thinking. Simply put, we have two main systems of thought: system 1 being the instinctive side and system 2 being more rational and controlled. Dr. Baddeley explains that while system 1 was more useful for early humans in hunter-gather settings, system 2 is more fruitful for today’s circumstances. And teens, often quick to engage system 1 in their decision-making, can be redirected into system 2 through parental encouragement. This is best exemplified when parents ask their teen to consider making the decision over a longer period of time. Having the teen make a comprehensive case for the decision also works in accomplishing this task. Alan Baddeley is a professor of psychology at the University of York. He is best known for his model of working memory, which essentially won out in the history of cognitive psychology as the model of working memory. He's been in the game for many years, and a couple years back published a career retrospective called Working Memories, which details his experiences, where his ideas came from, and what he observed around him during the Cognitive Revolution. What is perhaps less known about Professor Baddeley is that he began his PhD doing memory working at Cambridge because his studentship was sponsored to help develop memorable postal codes for the U.K.'s postal service. Otherwise, he might not have become a memory researcher at all. In this episode we talk about how Bertrand Russell influenced him to become a psychologist, the difference between the U.S. and the U.K. during the Cognitive Revolution, and how to develop an academic career by proceeding through a series of logical steps.

Today on the podcast I talk with Jessica Baddeley and she is telling me all about how to fly Space-A. Space-A means space available and is a service the military provides for members and families in certain situations. If there is space available on a military flight that is already going somewhere you could possibility get on that flight for free! However, it’s not that easy.Jessica tells us exactly where to start in the processes by going to SpaceA.net and then finding your terminal's Facebook page to get information on flights that might be going out near you. She then explains what documents you need in order to fly Space A this also depends on what category you are in, which she explains as well. Jessica then lays out what you should be doing 2 weeks before you want to fly, 48 hours, and the day of! She makes is clear that you are giving your time instead of your money. It takes her on average about 18 hours to get from Hawaii to California and the flight is only 5 hours. That is a lot of waiting time. But hey, if you are saving $1500 it’s worth it!We talk about the worst times to fly Space-A because it is so busy: summer, holidays, and school holidays! So even if you don’t have school age children check the calendar of your local area and the area you are going to and make sure it won’t be busy because school is out. Good communication is one of the foundations of the Agile mindset and one of the core values in Extreme Programming. As programmers, we’re used to identifying anti-patterns in our code, and yet at the same time we fail to recognize communication anti-patterns that exist on the teams that we work on, and how they’re impacting our ability to produce software that generates value. In this talk we'll learn to detect communication anti-patterns that can exist on teams, and how they can impact the ability to translate business requirements into implemented features. We'll also explore some strategies and approaches for mitigating and managing these anti-patterns to improve the software development process.

Human beings are social animals, notes economist Michelle Baddeley, and as such the instinct to herd is hardwired into us. And so while this has changed from (in most cases) physically clumping into groups, it does translate into behavior linked to financial markets, news consumption, restaurant-picking and Brooklyn facial hair decisions. In this latest Social Science Bites podcast, Baddeley – a professor in economics and finance of the built environment at University College London -- tells interviewer David Edmond how modern herding often follows from an information imbalance, real or perceived, in which a person follows the wisdom of crowds. The decision to join in, she explains, is often based an astute reading of risk; as she quotes John Maynard Keynes, “It’s better to be conventionally wrong than unconventionally right.” As a real world example of that, she points to the plight of the junior researcher, whose career is best advanced by serving up their innovative insights along conventional lines. Apart from reputational damage control, there are pluses and minuses to human herding, Baddeley notes there are advantages to finding safety in numbers: “It’s a good way to find a hotel.” But there are pernicious outcomes, too, like groupthink. In that vein, the economist says she finds partisan herding “more prevalent in a ‘post truth age,’” as individuals join thought groups that reinforce their existing world-view. And it doesn’t help, her research finds, that people are more likely to herd the less well-informed they are. This has also had dire consequences in financial markets (Baddeley was principal investigator on a Leverhulme Trust project focused on neuroeconomic examination of herding in finance), where pushing against the grain makes for a short career for anyone other than the luckiest professional stockpicker. Baddeley’s early education was in Australia and her first professional work was as an economist with the Australian Commonwealth Treasury. She then completed masters and doctorate work at Cambridge. Her most recent book is 2013’s Behavioural Economics and Finance  and other works include Running Regressions - A Practical Guide to Quantitative Research (2007) and Investment: Theories and Analysis (2003).

In geostatistics, a common problem is to predict a spatial exceedance and its exceedance region. This is scientifically important since unusual events tend to strongly impact the environment. Here, we use classes of loss functions based on image metrics (e.g., Baddeley's loss function) to predict the spatial-exceedance region. We then propose a joint loss to predict a spatial quantile and its exceedance region. The optimal predictor is obtained by minimizing the posterior expected loss given the process parameters, which we achieve by simulated annealing. Various predictors are compared through simulation. This methodology is applied to a spatial dataset of temperature change over the Americas. This research is joint with Jian Zhang and Peter Craigmile. Noel Cressie. Das Konzept exekutiver Funktionen hat eine lange Geschichte.(vgl. Benton, 1991, Tranel et al, 1994), welche im Laufe der Zeit zu den bekannten kognitiven Modellen geführt hat (Baddeley & Hitch, 1974, Norman & Shallice, 1980, Grafman, 1989, 1995, Damasio et al., 1991). Aufgrund der methodischen Probleme, eine zentral exekutive Instanz zu bestätigen, gehen einige Autoren von einem „multikomponenten“ exekutiven System aus (McCarthy & Warrington, 1990, Milner & Petrides, 1984), welches eigene Zuständigkeitsbereiche für bestimmte Anforderungen beinhaltet. Baddeley (1996) hingegen geht in seinen Überlegungen von einem einzelnen koordinierten System aus, welches verschiedene Funktionen bereitstellt. Es kann mit Sicherheit behauptet werden, dass die zentrale Exekutive keine monolithische Einheit darstellt (vgl. Baddeley et al., 1997, Burgess et al., 1997). Es hat sich herausgestellt, dass es sinnvoll ist, die kognitiven Anforderungen von Aufgaben systematisch zu untersuchen und diese anschließend für die Konstruktion eines kohärenten Modells der zentralen Exekutive zu verwenden (Baddeley, 1996, 1997). Eine Doppelaufgabe ist ein geeignetes kognitives Modell, um die zentrale Exekutive zu untersuchen, weil im Sozialverhalten und in der Selbstkontrolle eine Dual-Task Anforderung beinhaltet ist. Aus diesem Grund wurde die Anforderung einer Dual-Task Aufgabe als Brückenmaß zwischen einer kognitiven Dysfunktion und einem gemessenen beeinträchtigten Verhalten herangezogen. Beeinträchtigtes Sozialverhalten ist deshalb von Bedeutung, weil es mit einem Leben in sozialer und finanzieller Unabhängigkeit interferiert und weil es die Rehabilitation anderer kognitiver Defizite reduzieren kann. Der Zusammenhang dieser zwei Verfahren untermauert demnach Baddeley`s Theorie einer zentralen Exekutive, die für kognitive Leistungen als auch für Verhaltensaspekte zuständig ist. Die Ergebnisse der hier vorliegenden Arbeit unterstützen die Theorie einer zentralen Exekutive (vgl. Baddeley, 1997, 1996, Baddeley et al, 1997) und sprechen somit gegen ein Multikomponentensystem (McCarthy & Warrington, 1990, Milner & Petrides, 1984), welches autonome Kontrollprozesse ausführt, da die erfasste verbale Arbeitsgedächtnisleistung als neuropsychologisches Verfahren mit einer Verhaltensproblematik einhergeht. Die gefundenen Ergebnisse im visuo-spatialen Arbeitsgedächtnisverfahren können in der hier vorliegenden Studie weder für eine Bestätigung noch Widerlegung der angenommenen Hypothese herangezogen werden, da aufgrund des festgestellten Bodeneffektes keine tendenziellen Aussagen getroffen werden können. Jedoch konnte gezeigt werden, dass neben dem statistischen Zusammenhang des DEX mit einem verbalen Dual-Task Verfahren, die Leistungen in einer verbalen Flüssigkeitsprüfung wiederum eng mit der verbalen Dual-Task Leistung verknüpft sind (vgl. Baddeley, 1996, Daneman & Carpenter, 1980), und dass die Anforderungen im ToL Parallelen zu den Anforderungen beim Generieren von Wörtern zeigen.
