Abstract: Evolution depends upon genetic variations that influence physiology. As defined in a genetic screen, phenotypic performance may be enhanced or degraded by such mutations. We set out to detect mutations that influence motor function, including motor learning. Thus, we tested the motor effects of 36,444 non-synonymous coding/splicing mutations induced in the germline of C57BL/6J mice with N-ethyl-N-nitrosourea by measuring changes in the performance of repetitive rotarod trials while blinded to genotype. Automated meiotic mapping was used to implicate individual mutations in causation. 32,726 mice bearing all the variant alleles were screened. This was complemented with the simultaneous testing of 1,408 normal mice for reference. 16.3% of autosomal genes were thus rendered detectably hypomorphic or nullified by mutations in homozygosity and motor tested in at least 3 mice. This approach allowed us to identify superperformance mutations in Rif1, Tk1, Fan1 and Mn1. These genes are primarily related, among other less well characterized functions, to nucleic acid biology. We also associated distinct motor learning patterns with groups of functionally related genes. These functional sets included preferentially histone H3 methyltransferase activity for mice that learnt at an accelerated rate relative to the rest of mutant mice. The results allow for an estimation of the fraction of mutations that can modify a behavior influential for evolution such as locomotion. They may also enable, once the loci are further validated and the mechanisms elucidated, the harnessing of the activity of the newly identified genes to enhance motor ability or to counterbalance disability or disease. This time last year I had just pressed GO LIVE on my podcast and sat with bated breath to see if anyone, anywhere would consider downloading it and listening to me. I spent hundreds, HUNDREDS of hours editing, ripping, recording and trying with all my might and then-skill-level (which has improved!) to produce a show I love. I hit refresh and saw a download. It was my husband in the next room and I could hear him giggling softly when he heard my breath draw in at the sight of that one single download that meant the world to me. A few more downloads trickled in and I started to check out Twitter a little bit, Libsyn, PodBean and eventually ITunes and started listening to other podcasts. I guested on PassersbyPodcast with Matt and his listeners started reaching out to me, I will always be grateful for this because many of these folks have become my podcast hub. I hit 100 downloads and cried (you will see a theme here of me crying when happy and I blame my parents for this weird trait). I met up with the Angry Ginger and Salty Language's Tony and his beer expert wife Jeannie and laughed until our sides hurt somewhere in the middle of Michigan. I hit 1000 downloads and felt dizzy. I was asked to join Shumway Industries with a group of people I consider to be some of the best independent podcasters and I cried at the idea I could be considered among them for even a moment. Bill from Wicked Theory, Angry Ginger and TK1 of 7DAG, Stef of Gotham Lights, Diamond Dave, and a funny little proxy-caster named Agent Palmer who would go on to become one of my podcast confidants for every major change I have made to improve my show. I went to Podcast Movement in Chicago and met the great and astoundingly talented Carrie Zylka, the gentle giant (and I mean GIANT) Dan Lizette of Podcast Digest, and a host of other talented podcasters I now consider friends. I guested on over 20 shows and brought in as many fellow podcasters as I could manage to my show and these were more fun than I can write accurately about. I hit 10,000 downloads and told my family I might have a shot at this for real. I was invited to Podtoberfest and cried, I got to meet my heroes and it only made them larger in my mind somehow. I spent 15 hours on the road with Dougie, the Shumway IT guy and that truck contained some of the best conversations I've had, ever. I spent 36 hours podcasting with this group of awesomeness and finally gave in to the idea I might indeed be a music geek. I hit 30,000 downloads and forgot what numbers meant and I'm a math-lover people. I podcasted at so many music festivals I almost lost count, but not actually because each one was so meaningful and important to me I have kept every media pass I've ever been handed. I'm not kidding, I have them all on my dresser so when I'm getting ready in the morning for my soulless 9-5 to remind myself there is more out there for me, this work is for something, my audience growth is happening and I will do this for a living. My show got picked up by a college radio station and is now being prepped for a weekly Indie Music spotlight show, hosted by little old me. A year. I've been a podcaster for a year and I still can't believe how well its going. The setbacks have been bumps, the hours have been long and beloved, the chances people have taken on me have been humbling and wonderful and so deeply appreciated. As I look forward to 2017 I am a stronger podcaster and one more focussed on taking this to the next level. I don't know how to get there, there is no manual for this, its still too niche, its still too new - but I'm trying. Every day I am trying to make this happen for my family and for me and the little music nerd inside me that's screaming to get to the next show and record the next story. Thank you for coming along for the ride, I'm so happy you are here. I'm so happy we are. The downloads are a steady heartbeat now, rhythmic and reassuring that the audience I've built are sticking around and telling their friends about the show, every single one of them matters to me. This week on BSDNow, we have all sorts of interesting news, including a Kernel Fuzzing audit done for OpenBSD, a much improved This episode was brought to you by Headlines Multiple Bugs in OpenBSD Kernel (http://marc.info/?l=oss-security&m=146853062403622&w=2) Its patch Wednesday! (OR last Thursday if you were watching the mailing lists) Jesse Hertz and Tim Newsham (part of the NCC Group calling themselves project Triforce) have been working with the OpenBSD team to fix some newly discovered bugs in the kernel using fuzzing. Specifically they were able to track down several potential methods to corrupt memory or panic the kernel: mmap_panic: Malicious calls to mmap() can trigger an allocation panic or trigger memory corruption. kevent_panic: Any user can panic the kernel with the kevent system call. thrsleeppanic: Any user can panic the kernel with the _thrsleep system Call. thrsigdivertpanic: Any user can panic the kernel with the _thrsigdivert system call. ufsgetdentspanic: Any user can panic the kernel with the getdents system call. mount_panic: Root users, or users on systems with kern.usermount set to true, can trigger a kernel panic when mounting a tmpfs filesystem. unmount_panic: Root users, or users on systems with kern.usermount set to true, can trigger a kernel panic when unmounting a filesystem. tmpfsmknodpanic: Root can panic kernel with mknod on a tmpfs filesystem. This was a great find, and we have a link to more of the results, if you would like to explore them in more detail. NCC Group OpenBSD Kernel fuzzing results (http://www.openwall.com/lists/oss-security/2016/07/14/5) Would like to see more work like this done in all of the BSDs *** Running CockroachDB in a FreeBSD Jail (https://www.cockroachlabs.com/blog/critters-in-a-jar-running-cockroachdb-in-a-freebsd-jail/) The developers behind CockroachDB have written up a nice walkthrough of getting their software to run inside FreeBSD jails. “Manually encapsulating CockroachDB using Linux cgroups is no easy task, which is why tools like Docker exist in the first place. By comparison, running server processes natively in FreeBSD jails is straightforward and robust.” The walkthrough begins with compiling CockroachDB straight from source (A port is pending), which is pretty easy relying upon bash / git / gmake and GO. With the compile finished, the next step will be mounting linprocfs, although that may be going away in the future: “(Note: Linux compatibility files / packages / libraries are not needed further. CockroachDB uses Linux's procfs to inspect system properties via gosigar. If/when gosigar evolves to read FreeBSD properties natively, CockroachDB will not need linprocfs any more.)” With the initial setup complete, the walkthrough then takes us through the process of creating the rc.d script (Which should be included with the port) and ultimately setting up ezjail and deploying CockroachDB within. With the word getting out about jails and their functionality, we hope to see more projects also provide walkthroughs and FreeBSD support natively. Kudos to the CockroachDB team! *** Usermount bugs (https://marc.info/?l=openbsd-announce&m=146854517406640&w=2) kern.usermount, (vfs.usermount on FreeBSD) is a sysctl that can be enabled to allow an unprivileged user to mount filesystems. It is very useful for allowing non-root users to mount a USB stick or other external media. It is not without its dangers though: > “kern.usermount=1 is unsafe for everyone, since it allows any non-pledged program to call the mount/umount system calls. There is no way any user can be expected to keep their system safe / reliable with this feature. Ignore setting to =1, and after release we'll delete the sysctl entirely.” In OpenBSD 6.0 and forward, the setting will no longer work, and root privileges will be required to mount a filesystem If there is a bug in the filesystem driver, the user could potentially exploit that and root the system > “In addition to the patched bugs, several panics were discovered by NCC that can be triggered by root or users with the usermount option set. These bugs are not getting patched because we believe they are only the tip of the iceberg. The mount system call exposes too much code to userland to be considered secure” This is a very pragmatic way of dealing with these issues, as it is not really possible to be sure that EVERY bug has been fixed, and that this feature is no longer an exploit vector usermount being removed from OpenBSD (http://undeadly.org/cgi?action=article&sid=20160715125022) I use this facility in FreeBSD extensively, combined with ZFS permission delegation, to allow non-root users to create and mount new ZFS datasets, and to do replication without requiring any root access There are some safety belts, for instance: the user must own the directory that the new filesystem will be mounted to, so they can't mount to /etc and replace the password file with their own *** Let's Encrypt client from BSD in C (https://kristaps.bsd.lv/letskencrypt/) File this one under the category of “It's about time!”, but Kristaps (Who we've interviewed in the past) has released some new software for interacting with letsencrypt. The header for the project site sums it up nicely: “Be up-front about security: OpenSSL is known to have issues, you can't trust what comes down the pipe, and your private key's integrity is a hard requirement. Not a situation where you can be careless. letskencrypt is a client for Let's Encrypt users, but one designed for security. No Python. No Ruby. No Bash.A straightforward, open source implementation in C that isolates each step of the sequence.” What specifically does it isolate you ask? Type of bootloader depends on the version of Linux4Tegra TK1 comes with. Mine had L4T R19, with some kind of “not u-boot” bootloader.” They tried using the provided tool, compiled on FreeBSD since it uses libusb, but it gave an error. Falling back to trying from Ubuntu, they got the same error. They then flashed the TK1 with newer firmware, and suddenly, uboot is available. The post then walks through pxe booting FreeBSD on the TK1 The guide then walks through replacing the UBoot with a version compatible with UBLDR, for more features We'll have to wait for another post to get FreeBSD burned onto the device, but at this point, you can reliably boot it without any user interaction I have one of these devices, so I am very interested in this work *** Why we use OpenBSD at VidiGaurd (https://blog.vidiguard.com/why-we-use-openbsd-at-vidiguard-4521f217b2b7#.9r86v742v) VidiGuard (Which makes autonomous drone solutions for security monitoring) has posted an interesting write-up on why they use OpenBSD. Specifically they start by mentioning while they are in business to provide physical security, they just as equally value their data security, especially their customer data. They name 4 specific features that matter to them, starting with Uncompromising Quality and Security: “Over the past 20 years, OpenBSD's focus on uncompromising quality and code correctness has yielded an operating system second-to-none. Code auditing and review is core to the project's development process. The team's focus on security includes integrated cryptography, new security mitigation techniques, and an optional-security-is-no-security stance, making it arguably the most secure operating system available today. This approach pays off in the form of only a few security updates for a given release, compared to other operating systems that might release a handful of updates every week.” High praise indeed! They also mention the sane-defaults, documentation and last but not least, the license as also winning factors in making OpenBSD their operating system of choice. Thanks to VidiGuard for publically detailing the use of BSD, and we hope to see other business follow suit! *** "You can (and should) slow down and learn how things work" – Interview with Dru Lavigne (https://bsdmag.org/dru_lavigne/) If you've been around the BSD community for any length of time, you no doubt have heard of Dru Lavigne (Or perhaps own one of her books!) She was recently interviewed by Luca Ferrari for BSD Magazine and you may find it a fascinating read. The 2nd question asked sounded a lot like our opener to an interview (How did you get into BSD) “ In the mid 90s, I went back to school to learn network and system administration. As graduation grew near and I started looking for a work, I noticed that all the interesting jobs wanted Unix skills. Wanting to increase my skills, and not having any money, I did an Internet search for “Free Unix”. The first hit was freebsd.org. I went to the website and started reading the Handbook and thought “I can do this”. Since I only had access to one computer and wanted to ramp up my skills quickly, I printed out the installation and networking chapters of the Handbook. I replaced the current operating system with FreeBSD and forced myself to learn how to do everything I needed to do on that computer in FreeBSD. It was a painful (and scary) few weeks as I figured out how to transition the family's workflow to FreeBSD, but it was also exhilarating to learn that “yes, I can do this!. Since then, I've had the opportunity to try out or administer the other BSDs, several Linux distros, SCO, and Solaris. I found that the layout, logic, and release engineering process of the BSDs makes the most sense to me and I'm happiest when on a BSD system.” When asked, Dru also had a good response to what challenges potential new UNIX or BSD users may face: “Students who haven't been exposed to open source before are used to thinking of technology in terms of a purchasable brand consisting of “black boxes” that are supposed to “just work”, without having to think about how they work. You can (and should) slow down and learn how things work. It can be a mind shift to learn that the freedom to use and change how something works does exist, and isn't considered stealing. And that learning how something works, while hard, can be fun. BSD culture, in particular, is well suited for those who have the time and temperament to dive into how things work. With over 40 years of freely available source and commit messages, you can dive as deep as you want into learning how things came to be, how they evolved over the years, how they work now, and how they can be improved. There is a diverse range of stuff to choose from: from user tools to networking to memory management to hardware drivers to security mechanisms and so on. There is also a culture of sharing and learning and encouragement for users who demonstrate that they have done their homework and have their own ideas to contribute.” The interview is quite long, and Dru provides fantastic insights into more aspects of BSD in general. McBlizzard needs some teeth pulled just to spill the beans on his trip to Tennesse and TK1 talks shop about cameras! We also get into a heavy debate about what the hell Marvel is or isn't up to with their movies. The whole Brian Singer using Quicksliver agianst Whedon is talked to death as is a "Shared World" could it happen? Is it possible? In this episode we #GeekOut about Instagram. Well, TK1 geeks why out about it! Follow him at Insta_Grants2 and tell him he's got a pretty house! We also talk a bit of phone tech stuff...then we move into the "Movie Shoot!" The first week of April I set out a challenge to anyone that thought they could write, shoot, edit and post a movie in 7 days to join me. The list started at 14 people. By the time the day came to start, the list was down to 6. Including myself. So everyone went out and assembled a crew of no more than 7 and they went out and made a movie! We discuss this and get into the meat of it all buy talking about actually shooting a movie. Did we make succeed or did we fail? Wouldn't that be funny if we failed at this 7 day challenge that I put forth? Ha! Ha! There is a possibility that TK1 was "Aces" during the shoot...and I became a tyrant on set. My inner Michael Bay was summoned from the darkness. We then move onto discuss a few Filmmakers and thier movies. First up is "Elysium." Neil Blomkamp's new flick starring Matt Damon. District 9 is discussed as well as his failed Halo movie. Next up is Louis Leterrier and the new trailer for "Now You See Me." This movie is filled with an all star cast. We break down the trailer. TK1 want's a "Spoilers" shoutout...even though we are months away from even seeing the actual movie.
In this episode, TK1 and I sit down together for the first time...since...the first time! Meaning we once hosted a podcast where it was only the two of us. So for roughtly 30 minutes, until Oz shows up...and show up he does....we talk about the movie we are going to make. I'm writing and directing while Grant is brave? enough to put on his Aces and do a little acting for us. The subject of getting caught having sex by your kids comes up...and away it goes with more perversion and hilarity than a convicted sex addict could stand! It's all in good fun people! Then OZ stops by for the "What have you been geeking out about" segment and boy do we all geek out! Wolverine, Batman, Robin, X-men, and animation...lots of good stuff for you to stick around and listen to!
In this episode we ATTEMPT to make a commentary track over the Tricks and Treats story that is in Episode 31: The Grindhouse Funhouse Radio Play. As you will see, it takes us a while to warm up to that part. We get wrapped up in the Kai The Homeless Hitchhiker Story, as well as talk a little Old Gregg! It takes a few minutes, but we actually start to play the first read through of the Radio Play. It's terrible. No point in playing the good one right. While listening to it, Grant (TK1, Ed Richter) The Seasoned "Actor", has a bit of trouble telling the recording apart from real life...in a bizarre twist of events, his past self even starts to talk to him! Here we are, smack in the middle of Epi49, just one 'sode away from our 50th show! Exciting! And we have a wonderful Special Guest this show — Hal Lublin! If you're a fan of The Thrilling Adventure Hour, then you've heard LOTS of Hal because he's all over that show as both the announcer and filling out a number of roles in the various sketches. When I interviewed TAH's co-creator Ben Blacker back in Epi37, he told me that I really should try to catch the show live. And I finally got the opportunity this past weekend when the entire cast and orchestra traveled north from Hollywood, California, America to appear at the 12th Annual San Francisco Sketchfest. I got to see Hal and the rest of the gang put on a very funny show (they performed twice, actually, back to back) and Mr. Lublin was kind enough to sit with me before making the trek back down south. Our podcaster friends can relate when I say that Hal, while he exercises his voice on behalf of TAH as well as cartoons and other voiceover work, still holds down a fulltime day job. AND is perfectly happy to continue doing so. He tells us why during the interview, as well as some "tricks of the trade" when it comes to taking care of those precious pipes. In this, the 65th episode of The Last Podcaster Standing, While not trying to beat around the bush so to speak, TK1 and The Sniper do just that as they have come up with a new method of pleasing The Angry Ginger's Wife during climax! Actually, to be completely honest, all they want him to do is holler something at that pinnacle moment! AG, with the help of the crew, decide to literally (yes literally) break down what exactly could be going on inside and out during that special "feel good moment". Also, the gender of Ginger's new baby is revealed and he almost shows why his first name is in fact "ANGRY"! After that special lifetime movie G.G. decides it would be a great idea to chew our food for us so we don't choke on it...or is it our words? Either way 2 dogs and a Granny get in the way and after a few words we decide to head out into the forest and do the only thing we know how to do to end an argument! GET HIGH ON CRACK! I'm not going to say who brought the 8 ball but once we cash in our earnings from the biggest lottery jackpot in history we decide that we'll keep this show around for you listeners...but only if we can kill some crackheads on our off hours! Through tears as big as horse turds for some of us, we devise a plan that is so genius that we immediately move on to talk about "The Angry Dragon" and how skillful the female must be to pull this method of pleasure off! In this, the 63rd episode of The Last Podcaster Standing, Back by popular demand! Ladies and Gentleman, whether you love him or hate him (we know you all enjoy him), I give you Mr. Nick TWIST! Hopefully everyone has paid their cellphone bills, because it's going to take some crazy state to state calling to find our friend TWIST! So crazy in fact, that it will take not one podcaster but two to help find TWIST who is lost in New York City! In this, the second "Bloopersode" of The Last Podcaster Standing", "Welcome to S#!TCAST!"... Once again we find The Kilted One sick from his POIS, so while he endures his hyposensitization therapy, the guys skip recording a new episode, and instead have Mr. KIA put together more previously unaired material. New listeners should forego this episode in favor of previous episodes as this is not our usual idiocy. This is our "behind the scenes, look what we cut that should never air" idiocy. Included in this little ditty, you will find the twigs an berries of "The Real Episode 6". "The never meant to be heard episode" This was recorded when The Kilted One and The Angry Ginger were spur of the moment absent. Filling in for them was Emmett TerBeek (who was supposed to be our first guest podcaster) and The Ninja (future brain slave and spouse of Mr. KIA). This episode was never posted for various reasons the least of which was Mr. KIA's ability to stand by his word that he could lead and direct an episode without TK1. Initially, the plan was to have Mr. KIA run and direct the program as The Kilted One would, but upon startup, instantaneously deferred to allowing guest podcaster Emmett to run it. 45 minutes later it was over and done with. Not bad...Just not good. What you hear of this dumped episode is what we could salvage. The best bit being "Gingers Have No Souls (Ginger Song Part 1)" played by Emmett on his guitar. Also included in this "bloopersode" from past episodes: The guys before they start recording, the guys after they finish a show, but The Kilted One kept recording anyway, and other bits cut for other various reasons. Fights. Mistakes. Douchebaggery. Sing-A-Longs. It's all here! Well... Not ALL of it. That would be a lot... So to our fans, followers, and listeners, who have made it through our first 13 episodes, which by the way is only the beginning of what we have planned, (insert evil laugh here) we say "Thank You" and "Welcome To S#!TCAST!" To our non fans or people hearing this as their first introduction to us... Leave. Come back here when you think we're funny. In this, the 11th episode of The Last Podcaster Standing, “Shooting The S#!t” this week, the guys discuss various aspects of marriage. From proposals to prenups and some shades of grey inbetween. Mr. KIA himself starts this weeks podcast off with the announcement of his impending marriage and the proposal that's gonna get him there. This leads the other 3 guys to recollect their own proposal stories. Set expectations low, however, as none of the stories are as exciting or funny as Mr. KIA's. Shortly after this romantic strip down memory lane, TK1 attempts to move into headlines, but Mr. KIA has other plans for him. Nefarious plans! And what pray tell are those plans you may ask? To teach TK1 a lesson. Yes, that's right, Mr. KIA sets out to set TK1 straight after TK1 offends and upsets him a week prior to the podcast. Although, as you will hear many times over (if you haven't fallen asleep, skipped ahead, or had “same difference” post traumatic stress flashbacks) TK1 CLEARLY states how he was wrong, apologizes for jumping the gun, interrupting, and making assumptions. Which was a clear attempt by TK1 to avoid the whole nasty business. But, alas, an apology, a concession and an admission of fault is no good to Mr. KIA unless he can embarrass you and make you feel stupid in general in front of LOTS of people rather than in private. Guess who those LOTS of people are? That's right you the lucky listeners! Woo hoo! You may be asking why not just edit this out of the podcast? Why bore and alienate we the listeners with this drivel? 'Cause this is real life b*tches! We fight, we argue, we don't get along sometimes, we offend, and we hurt each others “feelings”. For all the s*** Tom gets for being a Vulcan robot devoid of emotions, this was an obvious emotional outburst (we new he had them!). Mind you it was driven by revenge, spite, anger, frustration and malice, but hey, they're emotions to aren't they? So why not keep it on the record. Besides, by the end of it he had taught TK1 a very valuable lesson. And now TK1 knows... Don't assume, don't interrupt, don't blather to other people, but mostly, don't piss off Mr. KIA. And knowing is half the battle. High 5? In Headlines, a bank robber is foiled by magic words, an amputee proves he is not the sum of his parts, and a man gets mad because his beer goggles lied to him. And finally in the news, for one soon to be incarcerated man, “everything but the kitchen sink” takes on a whole new meaning. 30 IS the magic number! In our new segment “The Male Sack” (at least new for this week), the guys field these questions from emails submitted by listeners, fans, followers, and stalkers. “On Valentines Day my girlfriend was giving me a B.J.
