Like an outdated operating system running in the background, these limiting beliefs quietly sabotage our potential. What is possible when we uncover them and align our goals with what we really want? Cindy walks us through discovering and changing the beliefs holding us back. The first step is identifying all those secret rules we've internalized that have been silently dictating what we do. Cindy explains that the limiting beliefs come from well-meaning advice, societal expectations, and passing comments we internalize. She delves into four of the most common: 1) “I don't deserve better”, 2) “I'm not qualified for anything else”, 3) “I'm lucky to have this job”, and 4) “My work doesn't count unless I'm miserable”. So how do we shake these limiting beliefs and move past them? Cindy's advice starts with four questions designed to uncover the ones we hold about ourselves. She then explains how to replace them with new, empowering beliefs by breaking down why the old beliefs are limiting. Replacing the limiting beliefs includes choosing a path that is both challenging and believable, paying attention to intuition, and taking a leap past ingrained fear. Kennedy Institute for the United States Senate, a non-profit organization in Boston, Massachusetts that educates the public about the U.S. system of government and works to inspire new generations of citizens and leaders. She served as an advocate for expanded medical research and access to health care for all Americans and actively promoted the passage of the Affordable Care Act of 2010. On the underestimation of dust mass in protoplanetary disks: Effects of disk structure and dust properties by Yao Liu et al. on Sunday 16 October The total amount of dust grains in protoplanetary disks is one of the key properties that characterize the potential for planet formation. With (sub-)millimeter flux measurements, literature studies usually derive the dust mass using an analytic form under the assumption of optically thin emission, which may lead to substantial underestimation. In this work, we conduct a parameter study with the goal of investigating the effects of disk structure and dust properties on the underestimation through self-consistent radiative transfer models. Different dust models, scattering modes and approaches for dust settling are considered and compared. The influences of disk substructures, such as rings and crescents, on the mass derivation are investigated as well. The results indicate that the traditional analytic method can underestimate the mass by a factor of a few to hundreds, depending on the optical depth along the line of sight set mainly by the true dust mass, disk size and inclination. As an application, we perform a detailed radiative transfer modeling of the spectral energy distribution of DoAr 33, one of the observed DSHARP disks. When the DSHARP dust opacities are adopted, the most probable dust mass returned from the Bayesian analysis is roughly 7 times higher than the value given by the analytic calculation. This week I would like to welcome Stacy, a thirty-four year old mother of a two year old little girl; Stacy and her husband are expecting a second daughter in August. Living on the central coast of Oregon, Stacy and her family have a dairy farm as well as a footwear manufacturing company. Originally from Kansas, born and raised, she moved away shortly after college and eventually she and her husband made their way to Oregon. Stacy learned that she was a donor conceived person (DCP) when she was around fourteen or fifteen years old. After their original disclosure, it wasn't something that her parents ever discussed with her again. Stacy is the only child of her parents aside from her father's adopted son from a previous marriage; he is fifteen years her elder. Her biological father is an anonymous donor who she was able to identify about a year ago through DNA testing and family research. After confirming his relation to her and divulging his medical history, he requested they have no further contact. She is still searching for answers to understand him..... and to understand herself. Atrial Fibrillation and Underestimation of Aortic Stenosis Severity Guest: Vuyisile T. Nkomo, M.D., M.P.H. Host: Malcolm R. Bell, M.D. Aortic valve stenosis is common in the general population. Echocardiography is the primary tool used for assessing the severity of the disease. In patients with atrial fibrillation, however, Doppler echocardiography may underestimate the severity of aortic valve stenosis, resulting in the potential for grave consequences. Abstract: The prediction of acid dissociation constants (pKa) is a prerequisite for predicting many other properties of a small molecule, such as its protein-ligand binding affinity, distribution coefficient (logD), membrane permeability, and solubility. The prediction of each of these properties requires knowledge of the relevant protonation states and solution free energy penalties of each state. The SAMPL6 pKa Challenge was the first time that a separate challenge was conducted for evaluating pKa predictions as part of the Statistical Assessment of Modeling of Proteins and Ligands (SAMPL) exercises. This challenge was motivated by significant inaccuracies observed in prior physical property prediction challenges, such as the SAMPL5 logD Challenge, caused by protonation state and pKa prediction issues. The goal of the pKa challenge was to assess the performance of contemporary pKa prediction methods for drug-like molecules. The challenge set was composed of 24 small molecules that resembled fragments of kinase inhibitors, a number of which were multiprotic. Eleven research groups contributed blind predictions for a total of 37 pKa distinct prediction methods. In addition to blinded submissions, four widely used pKa prediction methods were included in the analysis as reference methods. Collecting both microscopic and macroscopic pKa predictions allowed in-depth evaluation of pKa prediction performance. This article highlights deficiencies of typical pKa prediction evaluation approaches when the distinction between microscopic and macroscopic pKas is ignored; in particular, we suggest more stringent evaluation criteria for microscopic and macroscopic pKa predictions guided by the available experimental data. Top-performing submissions for macroscopic pKa predictions achieved RMSE of 0.7-1.0 pKa units and included both quantum chemical and empirical approaches, where the total number of extra or missing macroscopic pKas predicted by these submissions were fewer than 8 for 24 molecules. A large number of submissions had RMSE spanning 1-3 pKa units. Molecules with sulfur-containing heterocycles or iodo and bromo groups were less accurately predicted on average considering all methods evaluated. For a subset of molecules, we utilized experimentally-determined microstates based on NMR to evaluate the dominant tautomer predictions for each macroscopic state. Prediction of dominant tautomers was a major source of error for microscopic pKa predictions, especially errors in charged tautomers. The degree of inaccuracy in pKa predictions observed in this challenge is detrimental to the protein-ligand binding affinity predictions due to errors in dominant protonation state predictions and the calculation of free energy corrections for multiple protonation states. Underestimation of ligand pKa by 1 unit can lead to errors in binding free energy errors up to 1.2 kcal/mol. The SAMPL6 pKa Challenge demonstrated the need for improving pKa prediction methods for drug-like molecules, especially for challenging moieties and multiprotic molecules. A few weeks ago, Kowalski & I decided to run the 72-mile loop around Lake Tahoe... We have ridden our bikes around the lake countless times but figured why not go ahead and give it a run this time. Several sections of the loop have long paths that stretch many miles at a time, but there are also a lot of sections that have no path and very little to no shoulder... Well aware of this challenge, we figured if we made the voyage on foot from Chicago to NYC, how bad could it be navigating the roads around the lake? BAD... Very BAD. In the entire Chicago to NYC journey we never encountered anything like Hwy 50 on the South and East Shore of the lake... ZERO bike lanes coupled with speeding cars going 80 plus mph literally had us running for our lives. We expected a long day of running and a little traffic dodging in a few areas, but we never expected to find ourselves in the midst of a real life game of Frogger. Oftentimes in life we underestimate a scenario and then find ourselves trying to figure out how it happened... Well, let me enlighten us... Underestimation is caused by one main thing… LACK of preparation. Being the Tahoe local that has navigated the lake many times before, I take full responsibility for not comprehending just how HAIRY running the same route would be... I took for granted the fact that I thought I knew exactly what the route would be like but in reality, I had never gone counterclockwise around the lake and I surely had never run it. Had I done more navigational preparation, I would have paid attention to all of the areas with blind turns and no shoulders and simply realized it wasn't worth the risk... That said, I didn't, so Kowalski & I got to experience one helluva adventure. We here at the Daily Hustle will always encourage and support hard charging lifestyles and doing EPIC shit is pretty much a GO HARD prerequisite... Yet, if we can learn one thing from our Mistake By The Lake, don't ever underestimate the unknown and always be sure to diligently prepare for any and all scenarios… -EB P.S. Today’s show addresses and debunks the current jobs statistics just released today by the US Labor Dept. The official announced 14.7% unemployment rate and 23m jobs lost (20m in April) are dissected to reveal the true scope of those without work. Why the 14.7% only represents full time workers’ job loss and excludes millions more part time, temp, contract, gig jobs now 60m out of a 164m labor force. Rasmus explains even Govt stats show 22.4% unemployment rate when part time, ‘missing labor force and other categories are included. Rasmus further explains why the 22.4% is under-reporting as well, when the 8m who have dropped out of the labor force are included. How we get 33.5m having filed for unemployment benefits but report only 23m jobless. Why govt stats exclude many sheltering at home, without jobs, as not unemployed. Why the economy’s ordered ‘reopening’ will lower the official 14.7% unemployed, but only temporarily and why jobless numbers will rise again as a second infection wave hits the economy, and economic conditions developing, converge by year end. Rasmus explains it will take 5-7 yrs to just recover lost jobs, under best of conditions. Why the economic recovery will be ‘W Shape’, not V-Shape, and follow Rasmus’ past predictions of the trajectory of an ‘Epic’ Recession.
