Today's guest is my good friend and past client, Adam Johnson, CPA, CA, CBV, CFA—who, at the time, was my youngest client and didn't have as many letters behind his name. Adam's not your average CPA. He's also a Chartered Business Valuator CBV and Chartered Financial Analyst CFA, making him one of less than 100 people in Canada with all three designations. He even ranked #1 in the country on his CBV exam. But what really sets him apart is his hands-on experience with one of Canada's biggest investment banks, where he analyzed stocks and worked directly with equity traders which prepared him nicely for a long-time career in business valuations. Now, Adam's expanded his investing to the U.S. after Ontario deals no longer made sense, and he's the founder of Synthesis Valuations, providing top-tier valuation reports for everything from mergers to shareholder disputes. Note we recorded this episode before the most recent hurricanes Helen and Milton devastated Florida the deadliest since Katrina in 2005, each with a combined $100 billion is losses, $50 billion each. For context, that major storm Toronto had in the summer caused $1 billion in damage, these hurricanes were 100 times worse. My thoughts and well wishes are with Florida.
DairyBeef 500 Programme Co-ordinator, Alan Dillon, joins Stuart Childs on this week's Dairy Edge to discuss late breeding season bull choice. Alan explains that late born calves are as much a hassle for the beef farmer as they are for the dairy farmer as they don't fit into any system, so a defined breeding season is important to eliminate May/June calves as they are of no value to dairy beef farms nor to dairy farmers. Alan then outlines the criteria for bull selection at this stage of the season with high carcass weight >10kg for traditional breeds such as Angus and Hereford and a minimum of 20kg for the continental breeds. The focus must be to deliver a quality calf that has potential to deliver a return. Gestation length and calving ease are the other two important criteria for dairy farmers particularly at this stage of the season where the objective must be to get the cow calved as early as possible at this late stage. Alan says there are buyers for most calves throughout the season with a variation similar to that of calving start date to when calves are bought across the country. Dairy farmers should try to use the best beef genetics either through AI or the stock bull they buy to generate calves of quality to move into dairy beef systems. In the future CBV will have a significant role to play in deciding the value of the calf but all calves can leave a return as long as the purchase price is right. Alan advocates developing a relationship with a local buyer to ensure calves move off the dairy farm for a reasonable price and in an orderly fashion rather than being at the mercy of the market each week. This is very useful for the calf buyer as they have animals that have a health history behind them so dairy farmers should try to develop these relationships ahead of the calving season when time will be hard to come by. Using a good beef animal is as easy as using a bad one and is to the benefit of the industry as a whole. Finally, Alan invites all farmers to attend the BEEf2024 Open Day in Grange, Co. Meath on June 26th, to see all things dairy beef with something for everyone. Sean Coughlan, CEO of ICBF, joins Stuart Childs on this week's Dairy Edge podcast to discuss the origins of ICBF, what it has done since its creation back in 1998 and what the future looks like for dairy breeding. Sean first gives a brief description of how ICBF came into being under the stewardship of Dr. Brian Wickham as the first CEO in 1998 when it was formed. He then explains the creases that had to be ironed out when ICBF started highlighting the variation in calving dates that existed initially due to the multiple layers at the time and how over time with the cooperation of all stakeholders, this has become a thing of the past with much more streamlined data flows ensuring accurate recording with no duplication. This has allowed ICBF to create the database that is the engine of dairy and beef breeding in Ireland. The links with the genetics and modelling departments of Teagasc have allowed ICBF to develop EBI initially, DBI and CBV more recently as well as suckler beef indexes all of which are created to drive genetic gain and ultimately profitability for Irish farmers. Sean says that the power of genetics is enormous and that while the improvement is often perceived to be slow, it works and that the potential of new technology such as the genotyping scheme to further improve genetic gain is very real and a huge opportunity for the Irish dairy and beef breeding industry. Abstract: Directed motion of cerebrospinal fluid (CSF flux) is instrumental for removing waste products from the brain. Fluctuations in global cerebral blood volume (CBV), induced by concerted changes of brain-wide neuronal activity, are suggested to drive CSF-flux. To test this hypothesis, we performed two related experiments in healthy humans, namely electro-encephalography and blood oxygenation level-dependent functional-MRI during burst-suppression anesthesia, and arterial spin labeling and functional-MRI during transient hypercapnia challenges. Changes in total CBV, induced by changes in instantaneous global neuronal activity during burst-suppression or brain blood flow during hyper-normocapnia transitions, cause opposite functional-MRI signal changes in the basal cisternae, marking CSF flux from and into the brain. This direct coupling of global neuronal activity, brain blood volume and CSF flux indicates a mechanism to enhance brain waste clearance. Copy rights belong to original authors. Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2023.03.15.532735v1?rss=1 Authors: Dresbach, S., Huber, R., Gulban, O. F., Goebel, R. W. Abstract: Layers and columns are the dominant processing units in the human (neo)cortex at the mesoscopic scale. While the blood oxygenation dependent (BOLD) signal has a high detection sensitivity, it is biased towards unwanted signals from large draining veins at the cortical surface. The additional fMRI contrast of vascular space occupancy (VASO) has the potential to augment the neuroscientific interpretability of layer-fMRI results by means of capturing complementary information of locally specific changes in cerebral blood volume (CBV). Specifically, VASO is not subject to unwanted sensitivity amplifications of large draining veins. Because of constrained sampling efficiency, it has been mainly applied in combination with efficient block task designs and long trial durations. However, to study cognitive processes in neuroscientific contexts, or probe vascular reactivity, short stimulation periods are often necessary. Here, we developed a VASO acquisition procedure with a short acquisition period (895 ms volume acquisition) and sub-millimetre resolution. During visual event-related stimulation, we show reliable responses in visual cortices within a reasonable number of trials ([~]20). Furthermore, the short TR and high spatial specificity of our VASO implementation enabled us to show differences in laminar reactivity and onset times. Finally, we explore the generalizability to a different stimulus modality (somatosensation). With this, we showed that CBV-sensitive VASO provides the means to capture layer-specific haemodynamic responses with high spatio-temporal resolution and is able to be used with event-related paradigms. It takes a strong leader to be vulnerable and ask their teams, "can we do it this way?" and if it doesn't work, admit that it's okay. It takes an even stronger leader to speak with all new hires as they come on board, empowering and encouraging them to be reckless. Imagine starting a new job and having you leader tell you to use your first 90 days to try and do things your way, and to break and change things as you see opportunity, without fear of repercussion. Chris Daly from ICBF joins Catherine Egan on this week's episode of the Beef Edge podcast to explain the new Commercial Beef Value (CBV) which will be launched at the Teagasc Virtual Beef Conference next week on 6th December at 8pm. Chris explains how it is a new tool being made available for farmers buying in cattle and bringing them on to finish to know what the genetic merit is. The euro value will be available for commercial suckler males and uncalved females, dairy males and dairy cross males and uncalved females being brought to finish. It is comprised of the following traits: carcass weight, carcass conformation, carcass fat , feed intake and docility. Chris highlights that while there will be a range across the breed, the within breed ranges will be very interesting to farmers. Farmers will be able to access their animals' CBV under their Herdplus account in the coming weeks. Chris says the plan is that it will be rolled out in marts in early 2022 to be available for farmers purchasing cattle to see the CBV on mart boards. The Teagasc Virtual Beef Conference takes place on Monday 6th & Wednesday 8th December at 8pm. It is free to tune in and you can register in advance at: https://www.teagasc.ie/beefcon21/ For more episodes and information covered on the Beef Edge, visit the show page at: https://www.teagasc.ie/thebeefedge Produced on behalf of Teagasc by LastCastMedia.com

Abstract: Current approaches to high-field fMRI provide two means to map hemodynamics at the level of single vessels in the brain. One is through changes in deoxyhemoglobin in venules, i.e., blood oxygenation level-dependent (BOLD) fMRI, while the second is through changes in arteriole diameter, i.e., cerebral blood volume (CBV) fMRI. Here we introduce cerebral blood flow (CBF)-fMRI, which uses high-resolution phase-contrast MRI to form velocity measurements of flow and demonstrate CBF-fMRI in single penetrating microvessels across rat parietal cortex. In contrast to the venule-dominated BOLD and arteriole-dominated CBV fMRI signal, the phase-contrast -based CBF signal changes are highly comparable from both arterioles and venules. Thus, we have developed a single-vessel fMRI platform to map the BOLD, CBV, and CBF from penetrating microvessels throughout the cortex. This high-resolution single-vessel fMRI mapping scheme not only enables the vessel-specific hemodynamic mapping in diseased animal models but also presents a translational potential to map vascular dementia in diseased or injured human brains with ultra-high field fMRI. Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2020.08.21.255802v1?rss=1 Authors: Poublanc, J., Sobczyk, O., Shafi, R., Uludag, K., Wood, J., Vu, C., Dharmakumar, R., Fisher, J. A., Mikulis, D. J. Abstract: BACKGROUND: The paramagnetic properties of deoxyhemoglobin shorten T2* as do gadolinium based contrast agents. Induction of abrupt changes in arterial deoxyhemoglobin concentration ([dOHb]) can mimic the action of intra-vascular boluses of gadolinium based contrast agents (GBCAs) used for perfusion imaging. AIM: To demonstrate the feasibility of rapidly changing pulmonary venous hemoglobin saturation for generating boluses of blood with altered T2* properties for measuring flow metrics in the systemic circulation. METHODS: A gas blender with a sequential gas delivery breathing circuit and software enabling prospective arterial blood gas targeting was used to implement rapid lung changes in the partial pressure of blood oxygen (PaO2) while maintaining isocapnea. Lung PaO2 was initially lowered to induce a low baseline deoxyhemoglobin concentration [dOHb]. PaO2 was then rapidly raised to normal for 10 seconds and then rapidly lowered to the initial low baseline creating a oxyhemoglobin (OHb) bolus. R2* changes were measured using blood oxygenation dependent (BOLD) MRI signal changes in large arteries and veins as well as in the microcirculation. This enabled generation of the following maps: bolus arrival time delay (TD) cerebral blood volume (CBV), mean transit time (MTT) and cerebral blood flow (CBF). RESULTS: BOLD signal in the middle cerebral artery showed a sharp increase during the OHb bolus transit indicating minimal dispersion confirming effective rapid modulation of pulmonary venous PO2 with reasonable cortical contrast-to-noise ratio of 3. Signals sorted by amplitude of signal changes and arrival times enabled the visualization of major arteries and veins. Contrast to noise ratio was adequate for a single gas challenge to provide most of the contrast, little improved by averaging over the remaining set of challenges. Values of the flow metrics derived from the perfusion maps were within normal ranges from published literature values. CONCLUSION: Non-invasive induction of abrupt changes in OHb saturation can function as a novel non-invasive vascular contrast agent for use in perfusion imaging. Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2020.07.24.219378v1?rss=1 Authors: Huber, L., Poser, B., Kaas, A. L., Fear, E. J., Dresbach, S., Berwick, J., Goebel, R. W., Turner, R., Kennerley, A. J. Abstract: Cerebral blood volume (CBV) has been shown to be a robust and important physiological parameter for quantitative interpretation of functional (f)MRI, capable of delivering highly localized mapping of neural activity. Indeed, with recent advances in ultra-high-field (>=7T) MRI hardware and associated sequence libraries, it has become possible to capture non-invasive CBV weighted fMRI signals across cortical layers. One of the most widely used approaches to achieve this (in humans) is through vascular-space-occupancy (VASO) fMRI. Unfortunately, the exact contrast mechanisms of layer-dependent VASO fMRI have not been validated and thus interpretation of such data is confounded. Here we cross-validate layer-dependent VASO fMRI contrast in a preclinical rat model using well established (but invasive) imaging methods in response to neuronal activation (somatosensory cortex) and respiratory challenge (hypercapnia). In particular VASO derived CBV measures are directly compared to concurrent measures of total haemoglobin changes from high resolution intrinsic optical imaging spectroscopy (OIS). Through direct comparison of response magnitude, across time, negligible changes in hematocrit ratio during activation (neuronal or vascular) are inferred. Quantified cortical layer profiling is demonstrated and in agreement between both VASO and contrast enhanced fMRI (using monocrystalline iron oxide nanoparticles, MION). Responses show high spatial localisation to layers of cortical excitatory and inhibitory processing independent of confounding large draining veins which hamper BOLD fMRI studies. While we find increased VASO based CBV reactivity (3.1 +/- 1.2 fold increase) in humans compared to rats it is demonstrated that this reflects differences in stimulus design rather than confounds of the VASO signal source. Together, our findings confirm that the VASO contrast is indeed a reliable estimate of layer-specific CBV changes. This validation study increases the neuronal interpretability of human layer-dependent fMRI results and should supersede BOLD fMRI as the method of choice in neuroscience application studies. Other questions focused on Daniel's Seventy Weeks, the first century rapture, and how the NT manuscripts survived and remained uncorrupted in spite of the rapture removal of all the true Christians at the Parousia. Several more questions and comments dealt with the two resurrection views within Preterism: CBV versus IBV. Advocates for the Collective Body View (CBV) of the resurrection claim that the Present Passive form of the Greek verb EGEIRO ("are raised") can ONLY be legitimately translated as "are BEING raised" with the sense of an ongoing process of resurrection of the collective body. On the basis of that claim, they label other preterists (IBV) as being futurists. We show that their collective body interpretation of 1 Cor 15 simply does not hold up under grammatical, historical, and contextual scrutiny, and that the Individual Body interpretation of this text is the correct one. Advocates of the Collective Body resurrection view have accused the Individual Body View of being futurist because we teach that we get a new immortal body and go to heaven at physical death. However, we reverse those charges, and show that the Collective Body View has produced some very rotten fruit by teaching that we are in "heaven now," and have our "immortal bodies now," and have "perfection now," and that "sin no longer exists now," or that "temptation and sin will continue to plague us even in the afterlife." If you are confused by all those strange unbiblical ideas that the Collective Body View has produced, this podcast is for you. Introduction: Stroke is the third leading cause of death, and is the leading cause of disabilities worldwide. Although stroke may result from localized cerebral ischemia, intracerebral hemorrhage, subarachnoid hemorrhage or venous sinus thrombosis, ischemic stroke is the most frequently cause of the total cases. In ischemic stroke, occlusion of the MCA or its branches accounts for more than 3/4 of infarcts and two thirds of all first strokes. The main mechanisms causing ischemic strokes are embolism and arterial thromboembolism. No matter what the mechanism an ischemic stroke is, they eventually lead to a focal reduction of perfusion in the brain. In the hyperacute stage the recognition of the ischemia using both clinical assessment and routine neuroimaging technique implies some uncertainties, which in turn makes it difficult to predict the outcome, either to improve or to reverse spontaneously, to persist or worsen. The concept of diffusion/perfusion mismatch attracted great attention since it may represent the tissue at risk or at least an index of penumbra. Our interest was to investigate whether the hemodynamic parameters had correlation with clinical severity and if they were useful for prediction of outcome in the mismatch region. Since diffusion/perfusion mismatch was recognized as a simple and feasible means to identify the ischemic penumbra, we evaluated the hemodynamic parameters in acute stroke patients and compared these parameter to the stroke scale NIHSS and to the outcome score MRS to investigate our hypothesis. Materials and Methods: 35 acute stroke patients (male:female=20:15, age: 61.3±15.2 years) who met the study inclusion and exclusion criteria were selected. Significant cerebrovascular risk factors were recorded in 27 patients. The NIHSS assessment was immediately performed at the patients’ admission by a neurologist. Functional outcome was measured on the day of hospital discharge following MRS. Routine MRI sequences and DWI and PWI (dynamic susceptibility contrast-enhanced [DSC] imaging) were employed in our patients study. The perfusion maps were processed with MEDx® and the parameters were obtained by identifying ROIs on both ischemic core and mismatch region, and the normal mirror region. Relative values of the hemodynamic perfusion parameters were used in the evaluation. Statistic treatment was used to test the significance of the result. Results: The NIHSS score ranged from 0 to 19 (10.2±4.4) and the outcome MRS scale ranged from 0 to 6 (mean: 3.23). Between the good outcome group (MRS 0 to 3) and the poor outcome group (MRS 4 to 6), time to scan, type of treatment, DW/PW volume ratio, and age and female/male ratio did not show significant differences. In ischemic core: rCBF showed a remarkable decrease in all patients on average by 59.3±33.7% (range: 23.2 - 97.4%). rCBV decreased in 29 patients by 41.7±23.7% (range 19.6 - 55.6%), while 6 patients showed an increase of rCBV by 60.4±57.1% (range 0.7 -139%). The mean rCBV change of the entire group was 26.3±52.5%. MTT, TTP and T0 prolonged for 4.7 (SD=15.1), 2.8 (SD=12.9) and 0.5 (SD=10.4) seconds, respectively. In mismatch region: rCBF decreased in 15 patients by 26.2±19.9% (range: 5.3-58.4%) and increased in 20 patients by 35±23.2% (range: 6.8–74.4%). The change of the rCBF of the whole patients group was 5.8±38.4%. rCBV decreased in 7 patients by 14.7±16.5% (range: 0.8-44.5%) and increased in 28 patients by 39.5±36% (range: 2.2-91.1%). The mean change of the rCBV of the whole group was 19.9±31.2%. The mean value of MTT, TTP and T0 prolonged for 2.7 (SD=8.5), 3.2 (SD=5.2) and 1.3 (SD=4.2) seconds respectively. In both core and mismatch region, rCBF showed statistically significant regression to MRS. The more the rCBF decreased the higher the MRS (poor outcome) was. Also, the MTT delay in the core region was significantly related to MRS. TTP delay, in both core and mismatch region, was related to both NIHSS and MRS significantly. No statistic significance was found comparing CBV and T0 in relation with NIHSS or MRS. Conclusion: The hemodynamic parameters derived from perfusion MR imaging may be helpful adjunct to predict the outcome and severity in acute stroke patients. In mismatch region, the rCBF and TTP are predictive for the stroke outcome.
