Abstract: Fragile X syndrome (FXS), a neurodevelopmental disorder caused by the inactivation of the FMR1 gene, is one of the most common causes of autism spectrum disorder (ASD). People with FXS and ASD often have difficulty regulating their emotions and behaving in a socially appropriate way. The specific neuronal circuits and synaptic dysfunctions involved in social deficits in FXS are not yet fully understood. The basolateral amygdala to accumbens pathway is central to normal social behavior, and we investigated the functionality of this pathway in a novel mouse model of FXS (Fmr1/y::Drd1a-tdTomato). We used optogenetics to distinguish synapses between BLA neurons and spiny projection neurons in the NAcC. We found that the BLA-NAcC connection in FXS mice displayed heightened synaptic strength along with specific alterations in synaptic plasticity. In FXS, enhanced AMPA and NMDA postsynaptic currents elevated BLA-D2 SPN synaptic strength, while BLA-D1 SPNs increased firing probability occurred without altered transmission. This results in an overall increase in postsynaptic integration in both D1 and D2 SPN BLA-NAcC pathways compared to WT littermates. Further, the absence of FMRP leads to a specific deficit in long-term depression for BLA-D1 SPNs connections. The neurobiological changes identified in this study may underlie the susceptibility to stressors, socio-communicative difficulties, and altered reward-related processes that are characteristic of FXS pathology. Abstract: Fragile X syndrome (FXS) is a highly-prevalent genetic cause of intellectual disability, associated with disrupted cognition and sleep abnormalities. Sleep loss itself negatively impacts cognitive function, yet the contribution of sleep loss to impaired cognition in FXS is vastly understudied. One untested possibility is that disrupted cognition in FXS is exacerbated by abnormal sleep. We hypothesized that restoration of sleep-dependent mechanisms could improve functions such as memory consolidation in FXS. We examined whether administration of ML297, a hypnotic drug acting on G-protein-activated inward-rectifying potassium channels, could restore sleep phenotypes and improve disrupted memory consolidation in Fmr1-/y mice. Using 24-h polysomnographic recordings, we found that Fmr1-/y mice exhibit reduced non-rapid eye movement (NREM) sleep and fragmented NREM sleep architecture, alterations in NREM EEG spectral power (including reductions in sleep spindles), and reduced EEG coherence between cortical areas. These alterations were reversed in the hours following ML297 administration. Hypnotic treatment following contextual fear or spatial learning also ameliorated disrupted memory consolidation in Fmr1-/y mice. Hippocampal activation patterns during memory recall was altered in Fmr1-/y mice, reflecting an altered balance of activity among principal neurons vs. parvalbumin-expressing (PV+) interneurons. This phenotype was partially reversed by post-learning ML297 administration. These studies suggest that sleep disruption could have a major impact on neurophysiological and behavioral phenotypes in FXS, and that hypnotic therapy may significantly improve disrupted cognition in this disorder. Each month, our Carrier Connections program features a different X-linked condition with the goal to increase awareness and education of X-linked conditions and how they impact females. This month, we are featuring Fragile X syndrome. Fragile X syndrome is a heritable X-linked disorder caused by an abnormality in the FMR1 gene. This gene makes the protein, FMRP, which plays a critical role in the development of nerve cells and the production of other cellular proteins. An issue in this FMR1 gene causes an absence or severe deficiency of the FMRP protein, resulting in a range of developmental problems and distinctive physical features. It typically results from a specific mutation, or alteration of DNA, that causes a CGG base repeat to occur over 200 times in the damaged FMR1 gene. Molecular genetic testing can be used to diagnose this condition by identifying the 200 CGG repeats in the FMR1 gene. Women with a fragile X premutation, or 55 to 200 repeats of the FMR1 gene, have an increased risk of having a child with fragile X syndrome.  Additionally, female fragile X premutation carriers may experience menstrual and menopausal abnormalities, infertility, diminished ovarian reserve, and chronic hypoestrogenism. Such chronic hypoestrogenism has been associated with impaired bone health and an increased risk of cardiovascular disease. Furthermore, female carriers of a fragile X premutation have been found to have neuropsychiatric issues including neuropathy, increased anxiety and depression, and tremor/ataxia syndrome. Some studies have also associated thyroid abnormalities and hypertension to fragile X syndrome in women.  Today, we are joined by Rebecca Kronk, PhD, MSN, CRNP, ANEF, FAAN, CNE. Dr. Rebecca Kronk joined Duquesne University School of Nursing as an Assistant Professor in August 2010, earned tenure in 2016 and was promoted to Full Professor in 2022. She is a board certified pediatric nurse practitioner. Dr. Kronk earned her MSN from the University of Pittsburgh School of Nursing and a PhD in Applied Development Psychology at the University of Pittsburgh, School of Education. Dr. Kronk was a fellow in the Maternal and Child Health Training program providing leadership and education in neurodevelopmental disabilities and autism (LEND) at the University of Pittsburgh. Dr. Kronk has been in the field of pediatric nursing for over 40 years and she is experienced in helping children and families with a range of developmental issues. She has enjoyed combining clinical work with teaching and research. The theoretical framework of her research has been based on the International Classification of Functioning, Disability, and Health published by the World Health Organization. Dr. Kronk has conducted several research studies on the sleep patterns of children with Fragile X syndrome and developmental functioning of children in the FXS gray zone alleles. Her ongoing research has focused on undergraduate educational interventions to promote learning in genetics and caring for people with disabilities. However, the role of a major pathway controlling protein synthesis, the integrated stress response (ISR), in ASD remains poorly understood. Here, we demonstrate that the main arm of the ISR, eIF2 phosphorylation (p-eIF2), is suppressed in excitatory but not inhibitory neurons in a mouse model of fragile X syndrome (FXS; Fmr1-/y). We further show that the decrease in p-eIF2 is mediated via activation of the mTORC1. Genetic reduction of p-eIF2 only in excitatory neurons is sufficient to increase general protein synthesis and cause autism-like behavior. In Fmr1-/y mice, genetic restoration of p-eIF2 solely in excitatory neurons reverses elevated protein synthesis and rescues autism-related phenotypes. Thus, we reveal a previously unknown causal relationship between excitatory neuron-specific translational control via the ISR pathway, general protein synthesis and core phenotypes reminiscent of autism in a mouse model of FXS. Abstract: Fragile X syndrome (FXS) is the single most common monogenetic cause of autism spectrum disorders in humans. FXS is caused by loss of expression of the Fragile X mental retardation protein (FMRP), an mRNA-binding protein encoded on the X chromosome involved in suppressing protein translation. Sensory processing deficits have been a major focus of studies of FXS in both humans and rodent models of FXS, but olfactory deficits remain poorly understood. Here we conducted experiments in wild-type and Fmr1 KO (Fmr1-/y) mice (males) that lack expression of the gene encoding FMRP to assess olfactory circuit and behavioral abnormalities. In patch-clamp recordings conducted in slices of the olfactory bulb, output mitral cells (MCs) in Fmr1 KO mice displayed greatly enhanced excitation, as evidenced by a much higher rate of occurrence of spontaneous network-level events known as long-lasting depolarizations (LLDs). The higher probability of LLDs did not appear to reflect changes in inhibitory connections onto MCs but rather enhanced spontaneous excitation of external tufted cells (eTCs) that provide feedforward excitation onto MCs within glomeruli. In addition, in a go/no-go operant discrimination paradigm, we found that Fmr1 KO mice displayed impaired discrimination of odors in difficult tasks that involved odor mixtures but not altered discrimination of monomolecular odors. We suggest that the higher excitability of MCs in Fmr1 KO mice may impair fine odor discrimination by broadening odor tuning curves of MCs and/or altering synchronized oscillations through changes in transient inhibition. Abstract: The Fragile X Syndrome (FXS) is the most common form of inherited intellectual disability, and the first monogenic cause of Autism Spectrum Disorder. FXS is caused by the absence of the RNA-binding protein FMRP (Fragile X Mental Retardation Protein). Neuronal migration is an essential step of brain development allowing displacement of neurons from their germinal niches to their final integration site. The role of FMRP in tangential neuronal migration remains unexplored. We studied the consequences of FMRP absence on tangential migration, using the postnatal Rostral Migratory Stream (RMS) as an experimental system. In Fmr1-null RMS, neurons exhibit a slowed-down migration and an impaired trajectory, associated with defects of their centrosomal movement. Through RNA-interference-induced knock-down of Fmr1, we show that these migratory defects are cell-autonomous. Mutated neurons also display altered morphology as well as defects in their microtubule tyrosination distribution. Finally, we show that the FMRP mRNA target involved in these defects is MAP1B (Microtubule-Associated Protein 1B), whose knock-down rescues most migratory defects. Our results thus unveil a new neurodevelopmental role of FMRP, as a crucial actor of postnatal tangential migration, potentially important for the understanding of FXS pathophysiology. ​Regulating Together: An Evidence Based Group for Children with Autism That Utilizes the Parents As Coaches, Creating Lasting Change

In this episode, I speak with Rebecca, who discusses her career working with children, which led her to focusing on treatment and research of children on the autism spectrum. She discussed being influenced by her training in Philadelphia, which had a strong family systems component, and how working with the parents and children is a foundation for her Regulating Together work. She explains that the children are in a group where they learn affect regulation skills, while the parents are in another group, also learning affect regulation skills, how to coach the kids at home, and prevention and behavioral management skills. The skills the children learn are relaxation skills, identifying triggers and physical reactions, rating emotions, problem solving skills, mindfulness, radical acceptance and cognitive flexibility. She discussed how the caregiver training has a lot of focus on preventing the emotional dysregulation, as well as techniques for managing the dysregulation and behavior problems when they do occur. Additionally, the caregivers are encouraged to use the skills in order to regulate themselves, and how this helps with coregulation with their child. Rebecca discussed using CBT with a child with autism and modifications you might make since many autistic children can struggle with rigidity. She also remarked on how the group leaders have the ability to work with the children in vivo, at the end of the group where the kids earn time to play games. The group facilitators help the children implement the skills they learned if they become triggered during that time socializing. We discuss the research and how they found that the biggest gains were realized between five to ten weeks after the regulating together series was over, which highlights that the benefits of affect regulation and that a shift in behavior may take time to appear. We discussed other applications for the model and future potential research directions and a trial starting using a canine assisted version of the model. Rebecca explains that her and her team will be publishing their manual and are currently training clinicians in the use of this model. Rebecca C. Shaffer, PsyD, is a clinical psychologist and currently serves as an Associate Professor of Pediatrics at Cincinnati Children's Hospital with an affiliated appointment at the University of Cincinnati. Rebecca is the director of Psychological Services for the Cincinnati Fragile X Center, where she oversees the assessment and treatment of individuals with fragile X syndrome (FXS). Rebecca and her team have created an emotion dysregulation treatment program for children with ASD called Regulating Together. Regulating Together treats emotion dysregulation, especially with reactivity and irritability, in a group setting with concurrent caregiver training. She currently leads several research studies, as well as publications, focused on the development and efficacy of this program. She also serves as the primary investigator of the Simons Foundation Powering Autism Research (SPARK) study at Cincinnati Children's and other ASD-specific studies. Rebecca has had numerous publications and trains clinicians in Regulating Together throughout the country. When we yearn for the past, we want to re-capture the magic, the old emotion we once felt. Full Show: HERE   Seg 2 Rich Singer has a 20 year career in advertising, working for New York firms. He started in print ads, then radio, billboards, smaller TV ads and worked his way up to doing major TV ads and Super Bowl ads. When you first meet with a client to create an ad campaign what do you discuss? Try to determine what the client wants to say. Boil down the client message to one sentence. The ad needs to be clear. What the client wants to say, needs to be in one sentence. TV is expensive. There is a tendency for clients to want to jam in too much into an ad. They want to have multiple messages, when you need one message that can be laid out in one sentence. What is the objective of the ad? Is it a brand story, building the company brand. Is it an offer story, like a special sales package being run in all the stores. There are different levels to a sales funnel and your advertising has to match what level you believe your customers are on. Customers who are new to a brand, would need to see a different ad from a company that has a well established brand where customers already know what they're getting. The analogy Rich uses when talking with clients, is like a dog with a tennis ball. If you throw one tennis ball at a dog he catches that. If you throw two tennis balls at a dog he just stands there, because he doesn't know what to do. This is what customers feel like if you try to throw too many messages at them in one ad. After the initial meeting with the client the creative team has typically 2 weeks to create a concept. Then you will check in with the client to discuss the overview of the concept, see if it is to the client's liking. Then typically you have 2 more weeks to work it out more. Meeting number 2, creative team would show the client a first draft of a script. Rich is a copywriter and creative who works with his partner who is an art Director. They run a team of six people total, broken into three teams of 2, each also consisting of a copywriter and an art Director.   Seg 3 Super Bowl ads ship to Network about 2 weeks before the big game. The process to create the Super Bowl ads started months before. Networks will approve the ad script, with lawyers also working on that process. Networks have the right to reject an ad from an agency. The last thing you want, and it's rare, is to send an ad for the Super Bowl to a Network and they rejected it. Agencies start working on Super Bowl ads in August of the previous year, and it is about a 6 month process. A client is spending $7 million with the net work to buy a 30 second spot during the Super Bowl. After the initial meeting, the creative team spends 3 to 4 weeks writing the first draft of the spot. In September Rich and his team would present the script or Scripts, could be three to the client. In the presentation will be Rich, his partner, his boss, meeting with the client team which could be 3 to 4 people. Rich and his team has created six Super Bowl ads. Ads are initially filmed in November, or about 2 1/2 months before the Super Bowl. Sometimes, though an ad could be filmed as late as New Year's. Many people plus lawyers review Scripts and there are multiple revisions throughout the process. The final script is put together before filming. Lawyers and clients are looped in throughout the process and approving the script. Once you have approved Scripts you go out and you hire a Director and a production team to make the commercial. The Director would then be responsible to create a treatment and may be a storyboard for the commercial. The ad agency has a producer who works with the production company and Director to get the process going. There's also a line producer who is on site during a commercial shoot keeping the process moving. At an actual commercial shoot there might be a production team that could be upwards of 100 people plus actors and extras who are in the ad. The agency will send 6 to 10 people, as well as the client probably has 3 to 7 people also at the shoot.   Seg 4 After a commercial is filmed goes into post production. This involves editing the commercial, possible re-shoots, and post edits. This is also where computer graphics and digital effects are added to an ad as well as music and sound. Special effects and computer graphics are very advanced where you can change backgrounds within an ad. Amount of special FXs all depends on the budget of the commercial. Rich has been involved in commercials filmed at multiple NFL stadiums where there could be as many as 100 extras to sit in a crowd. Usually when you create a Super Bowl, the agency and client are in agreement on what they're trying to accomplish. The most likely goal is brand buzz. You know you have a successful Super Bowl ad when it's a crowd pleaser, people are entertained, and they are talking about it the next day. The goal of any great Super Bowl ad is to be memorable and entertaining. Again clients are spending a lot of money, $10 million for this process. $7 million with the NFL and Network to air the ad, and a $3 million production cost. You're hiring actors, production teams, possibly celebrities and athletes. There is a lot involved in a 30 second ad. Rich said the script needs to be extremely tight. Abstract: Attention deficit is one of the most prominent and disabling symptoms in Fragile X Syndrome (FXS). Hypersensitivity to sensory stimuli contributes to attention difficulties by overwhelming and/or distracting affected individuals, which disrupts activities of daily living at home and learning at school. We find that auditory or visual distractors selectively impair visual discrimination performance in both humans and mice with FXS, but not their typically developing controls. Vasoactive intestinal polypeptide (VIP) neurons were significantly modulated by incorrect responses in the post-stimulus period during early distractor trials in WT mice, consistent with their known role as error signals. Strikingly, however, VIP cells from Fmr1-/- mice showed little modulation in error trials, and this correlated with their poor performance on the distractor task. Thus, VIP interneurons and their reduced modulatory influence on pyramidal cells, could be a potential therapeutic target for attentional difficulties in FXS. Copy rights belong to original authors. Abstract: Individuals with fragile X syndrome (FXS) are frequently diagnosed with autism spectrum disorder (ASD), including increased risk for restricted and repetitive behaviours (RRBs). Consistent with observations in humans, FXS model mice display distinct RRBs and hyperactivity that are consistent with dysfunctional cortico-striatal circuits, an area relatively unexplored in FXS. Using a multidisciplinary approach, we dissected the contribution of two populations of striatal medium spiny neurons (SPNs) in the expression of RRBs in FXS model mice. We found that dysregulated protein synthesis at cortico-striatal synapses is a molecular culprit of the synaptic and ASD-associated motor phenotypes displayed by FXS model mice. Cell-type-specific translational profiling of the FXS mouse striatum revealed differentially translated mRNAs, providing critical information concerning potential therapeutic targets. Our findings represent the first evidence of a cell-type specific impact of the loss of FMRP on translation and the sequence of neuronal events in the striatum that drive RRBs in FXS. Abstract: Fragile X Mental Retardation Protein (FMRP) regulates activity-dependent RNA localization and local translation to modulate synaptic plasticity throughout the CNS. Mutations in the FMR1 gene that hinder or ablate FMRP function cause Fragile X Syndrome (FXS), a disorder associated with sensory processing dysfunction. FXS pre-mutations are associated with increased FMRP expression and neurological impairments including sex dimorphic presentations of chronic pain. In mice, FMRP ablation causes dysregulated DRG neuron excitability and synaptic vesicle exocytosis, spinal circuit activity, and decreased translation-dependent nociceptive sensitization. Activity-dependent, local translation is a key mechanism for enhancing primary nociceptor excitability which promotes pain in animals and humans. These works indicate that FMRP likely regulates nociception and pain at the level of the primary nociceptor or spinal cord. Therefore, we sought to better understand FMRP expression in the human dorsal root ganglion (DRG) and spinal cord using immunostaining in organ donor tissues. We find that FMRP is highly expressed in DRG and spinal neuron subsets with substantia gelatinosa exhibiting the most abundant immunoreactivity in spinal synaptic fields. Here, it is expressed in nociceptor axons. FMRP puncta colocalized with Nav1.7 and TRPV1 receptor signals suggesting a pool of axoplasmic FMRP localizes to plasma membrane-associated loci in these branches. Interestingly, FMRP puncta exhibited notable colocalization with calcitonin gene-related peptide (CGRP) immunoreactivity selectively in female spinal cord. Our results support a regulatory role for FMRP in human nociceptor axons of the dorsal horn and implicate it in the sex dimorphic actions of CGRP signaling in nociceptive sensitization and chronic pain. No matter what I predict (or anyone else), you want your portfolio set up so that if the market goes up you make a lot of money but if it goes down you are in a position to buy stuff cheap. Recommendations: - Cash 20-30%: Most people think that's insane, but we aren't most people! This is liquid money that won't go down. Alternatively, you can look into Gemini exchange (GUSD) and earn 8% interest. - Stablecoins: DAI 50%, USDC 25%, RAI 10%, Frax 10% - Ethereum 10% Layer 2's and new layer 1's are higher risk and I'm not interested in them right now. I recommend being very careful if you want to get into them. But maybe they will be attractive to me someday. - Other risk tokens 10%: Looking for things with good fundamentals. In this episode of the Ulysses Neuro Podcast, we celebrate Fragile X Awareness Day, which falls on the 22nd July in the US and other countries. We hear from Holly Roos, Community Services Director for the FRAXA Research Foundation, and her youngest child A. Holly is a mother of two children diagnosed with Fragile X Syndrome (FXS) and she shares her experience both as a mother and a carrier, discussing the challenges she has had to face from the very first day, sharing personal life stories and what kind of medical and educational support she had and how important it is to celebrate every single inchstone and milestone. A is 18 and lives in Illinois with Holly. A has just finished high school and is preparing to go to college soon. Holly and A share their experience of living with FXS, discussing the journey they have been on with Parker over the past two decades, and the challenges they have faced and overcome. FXS is a single gene disorder affecting 1 in 4,000 males and 1 in 8,000 females. It is the leading inherited cause of intellectual disability and it is caused by mutation of the FMR1 gene, which in turn reduces the production of the FMRP protein. FMRP is crucial for typical brain development as it controls expression of a number of other neurodevelopmental genes. Two main mutations have been identified: The full mutation giving rise to FXS, and a pre-mutation form in carriers whose children are of greater risk of having FXS. Carriers are also at risk themselves of a neurodegenerative disorder called Fragile X-associated Tremor/Ataxia Syndrome (FXTAS). Currently, despite the considerable progress in research in FXS, there is no cure or approved medication to treat the underlying causes of FXS and related conditions. This is in addition to the challenges faced by individuals with FXS and their families when it comes to diagnosis, support, and education.

Her son who is diagnosed with autism and Fragile X Syndrome had a very different level of support and resources than when her neurotypical daughter as they sought justice against the perpetrator. Abstract: Fragile X syndrome (FXS) is a neurodevelopmental disorder, characterized by intellectual disability and sensory deficits, caused by epigenetic silencing of the FMR1 gene and subsequent loss of its protein product, fragile X mental retardation protein (FMRP). Delays in synaptic and neuronal development in the cortex have been reported in FXS mouse models, however, the main goal of translating lab research into pharmacological treatments in clinical trials has been so far largely unsuccessful, leaving FXS a still incurable disease. Here, we generated 2D and 3D in vitro human FXS model systems based on isogenic FMR1 knock-out mutant and wild-type human induced pluripotent stem cell (hiPSC) lines. Phenotypical and functional characterization of cortical neurons derived from FMRP-deficient hiPSCs display altered gene expression and impaired differentiation when compared with the healthy counterpart. FXS cortical cultures show increased proliferation of GFAP positive cells, likely astrocytes, increased spontaneous network activity and depolarizing GABAergic transmission. Cortical brain organoid models show increased proliferation of glial cells, and bigger organoid size. Our findings demonstrate that FMRP is required to correctly support neuronal and glial cell proliferation, and to set the correct excitation/inhibition ratio in human brain development. Abstract: Drug repositioning is emerging as an increasingly relevant option for rare disease therapy and management. Various methods for identifying suitable drug candidates have been tried and range from clinical symptomatic repurposing to data driven strategies which are based on the disease-specific gene or protein expression, modification, signalling and physiological perturbation profiles. The use of Artificial Intelligence (AI) and machine learning algorithms (ML) allows one to combine diverse data sets, and extract disease-specific data profiles which may not be intuitive or apparent from a subset of data. In this case study with Fragile X syndrome and autism, we have used multiple computational methodologies to extract profiles, which are then combined to arrive at a comprehensive signature (disease DEG). This DEG was then used to interrogate the large collection of drug-induced perturbation profiles present in public databases, to find appropriate small molecules to reverse or mimic the disease-profiles. We have labelled this pipeline Drug Repurposing using AI/ML tools - for Rare Diseases (DREAM-RD). We have shortlisted over 100 FDA approved drugs using the aforementioned pipeline, which may potentially be useful to ameliorate autistic phenotypes associated with FXS. Abstract: Fragile X syndrome (FXS), the leading cause of familial intellectual disability, is an uncured disease caused by the absence or loss of function of the FMRP protein. FMRP is an RNA binding protein that controls the translation of specific proteins in neurons. A main target of FMRP in neurons is diacylglycerol kinase kappa (DGKk) and the loss of FMRP leads to a loss of DGK activity causing a diacylglycerol excess in the brain. Excessive diacylglycerol signaling could be a significant contributor to the pathomechanism of FXS. Here we tested the contribution of DAG-signaling in Fmr1-KO mouse model of FXS and we show that pioglitazone, a widely prescribed drug for type 2 diabetes, has ability to correct excessive DAG signaling in the brain and rescue behavioral alterations of the Fmr1-KO mouse. This study highlights the role of lipid signaling homeostasis in FXS and provides arguments to support the testing of pioglitazone for treatment of FXS. Copy rights belong to original authors. Abstract: Synaptic vesicle (SV) recycling is essential for the maintenance of neurotransmission, with a number of neurodevelopmental disorders linked to defects in this process. Fragile X syndrome (FXS) results from a loss of fragile X mental retardation protein (FMRP) encoded by the FMR1 gene. FMRP is an established translation repressor, however it also has translation-independent presynaptic roles, including regulation of the trafficking and function of specific ion channels. Since defects in SV recycling are exacerbated during intense neuronal activity, we investigated whether these events were disproportionately affected by the absence of FMRP. We revealed that primary neuronal cultures from a Fmr1 knockout rat model display a specific defect in activity-dependent bulk endocytosis (ADBE). ADBE is dominant during intense neuronal activity, and this defect resulted in an inability of Fmr1 knockout neurons to sustain SV recycling during trains of high frequency stimulation. Using a molecular replacement strategy, we revealed that a human FMRP interaction mutant failed to correct ADBE dysfunction in knockout neurons. Abstract: Fragile X syndrome (FXS) is caused by mutations in the FMR1 (fragile X mental retardation 1) gene. It is a significant form of heritable intellectual disability with comorbidity of other symptoms such as autism. Due to the lack of efficacious medication, repurposing the existing FDA-approved drugs may offer an opportunity to advance clinical intervention for FXS. Analysis of the whole-genome transcription signatures predicts new therapeutic action of vorinostat to correct pathological alterations associated with FXS. We further find that the administration of vorinostat restores object location memory and passive avoidance memory in the Fmr1 knockout (KO) mice. For the non-cognitive behavioral symptoms, vorinostat corrects the autism-associated alterations, including repetitive behavior and social interaction deficits. In the open field test, vorinostat dampens hyperactivity in the center area of the arena. Surprisingly, vorinostat does not affect the abnormally elevated protein synthesis in Fmr1 KO neurons, suggesting different outcomes from correcting behavioral symptoms and specific aspects of cellular pathology. Our data reveal the therapeutic effects of the FDA-approved drug vorinostat in a mouse model of FXS and advocate efficacy testing with human patients. Copy rights belong to original authors. Abstract: Axo-somatic K+ channels control action potential output in part by acting in concert with voltage-gated Na+ channels to set action potential threshold. Slowly inactivating, D-type K+ channels are enriched at the axo-somatic region of cortical pyramidal neurons of the prefrontal cortex where they regulate action potential firing. We previously demonstrated that D-type K+ channels are down regulated in extratelencephalic-projecting L5 neurons (ET) in the prefrontal cortex of the fmr1 knockout mouse model of Fragile X syndrome (FX mice), resulting in a hyperpolarized action potential threshold. To test whether K+ channel alterations are regulated in a cell autonomous manner in FXS, we used a viral-mediated approach to restore expression of Fragile X Mental Retardation Protein (FMRP) in a small population of prefrontal neurons in male FX mice. Outside-out voltage clamp recordings revealed a higher D-type K+ conductance in FMRP-positive ET neurons compared to nearby FMRP-negative ET neurons. FMRP did not affect either rapidly inactivating A-type or non-inactivating K+ conductance. ET neuron patches recorded with FMRP1-298, a truncated form of FMRP which lacks mRNA binding domains, included in the pipette solution had larger D-type K+ conductance compared to heat-inactivated controls. Viral expression of FMRP in FX mice depolarized action potential threshold to near wild type levels in ET neurons. These results suggest that FMRP influences the excitability of ET neurons in the mPFC by regulating somatic D-type K+ channels in a cell autonomous, protein-protein dependent manner. Abstract: Loss of the fragile X mental retardation protein (FMRP) causes fragile X syndrome (FXS). FMRP is widely thought to repress protein synthesis, but its translational targets and modes of control remain in dispute. We previously showed that genetic removal of p70 S6 kinase 1 (S6K1) corrects altered protein synthesis as well as synaptic and behavioral phenotypes in FXS mice. In this study, we examined the gene-specificity of altered mRNA translation in FXS model mice and the mechanism of rescue with genetic reduction of S6K1 by carrying out ribosome profiling and RNA-Seq on cortical lysates from wild-type, FXS, S6K1 knockout, and double knockout mice. We observed reduced ribosome footprint abundance in the majority of differentially translated genes in the cortices of FXS mice. We used molecular assays to discover evidence that the reduction in ribosome footprint abundance reflects an increased rate of ribosome translocation, which is captured as a decrease in the number of translating ribosomes at steady state, and is normalized by inhibition of S6K1. We also found that genetic removal of S6K1 prevented a positive-to-negative gradation of alterations in translation efficiencies (RF/mRNA) with coding sequence length across mRNAs in FXS mouse cortices. Our findings reveal the identities of dysregulated mRNAs and a molecular mechanism by which reduction of S6K1 prevents altered translation in FXS. Copy rights belong to original authors. When Missy Zolecki learned that she had a family history of Fragile X syndrome (FXS), she sought out carrier testing, thinking that she and her fiancé would not have children if she learned she was at risk for having a child with FXS. Fast forward 21 years later, Missy and her husband have been married 21 years and have three children, the oldest of whom is 18 years old and has Fragile X syndrome. Missy discusses how her religious faith influenced her decision making process around the option of IVF and Preimplantation Genetic Diagnosis (PGD), available only under a research protocol in 2000. She shares how she moved from anger at medical errors to appreciating the people and the opportunities that her son’s diagnosis of FXS has brought into her life. Missy explains how an accurate diagnosis of Fragile X—the leading genetic cause of autism—can be incredibly helpful in providing optimal care and education for children with Fragile X. She also shares her passion for women being offered preconceptual carrier testing for Fragile X for women, giving them choices and allowing them to make informed decisions. This talk revealed how the development of neuronal communication at synapses is altered in Fragile X Syndrome (FXS). It also examines how basic research into the role of genetics has led to the development of new medicines for treating FXS and hopefully will result in treatment possibilities for some forms of autism. Wild City #090 comes from Karachi based electronic producer TMPST – a genre bending artist who’s been at the forefront of the Pakistani scene for some time. With the third edition of the (Pakistan focused) Border Movement Lounge in India taking place tonight at the Max Mueller Bhavan, we thought it’s about time that we featured another exclusive, carefully curated mixtape from our contemporaries in the west. Asfandyar Khan is the man behind the 2-step, garage inspired beats and synths of TMPST as well as a range of downtempo, often hauntingly composed ambient music he records under his own name. He’s been making music since 2010 and equipped with nothing other than a guitar, released his first album, titled ‘Snow Makes Things Perfect’. The musician’s affinity for bass and beat only became apparent in 2012 with the birth of his ‘post-dubstep’ and garage inspired outfit TMPST. Despite TMPST’s foray into the world of bass however, he never quite manages to let go of the ambient sounds that began his journey and that peculiar seepage is what makes his sound to interesting. His last EP ‘Serpentine’ sees garage influenced choppy female vocals combined with undulating, atmospheric basslines and deep, dub wubs cleverly juxtaposed against drawling, emotive synths. TMPST’s exclusive Wild City Mix begins with an original ambient track by Asfandyar Khan. Apart from some excellent tunes by the likes of Lapalux and DEFT, this mix also features a bunch of music from the region. That includes some unreleased sounds by Asfandyar’s FXS peers, including a particularly impressive, unnamed mix closer from its founder Dynoman. Mix #090 starts on a placid note and ends on a high, occupies a comfortable space between your headphones and the dancefloor. It’s made of versatile stuff. Abstract: Fragile X Syndrome (FXS) is a neurodevelopmental disorder characterized by intellectual disability, autism spectrum disorders (ASDs), and anxiety disorders. The disruption in the function of the FMR1 gene results in a range of alterations in cellular and synaptic function. Previous studies have identified dynamic alterations in inhibitory neurotransmission in early postnatal development in the amygdala of the mouse model of FXS. Yet little is known how these changes alter microcircuit development and plasticity in the lateral amygdala (LA). Using whole-cell patch clamp electrophysiology, we demonstrate that principal neurons (PNs) in the LA exhibit hyperexcitability with a concomitant increase in the synaptic strength of excitatory synapses in the BLA. Further, reduced feed-forward inhibition appears to enhance synaptic plasticity in the FXS amygdala. These results demonstrate that plasticity is enhanced in the amygdala of the juvenile Fmr1 KO mouse and that E/I imbalance may underpin anxiety disorders commonly seen in FXS and ASDs.
