Explore the life and legacy of graphic design pioneer Jacqueline Casey, whose bold, modernist posters defined MIT's visual identity for decades — with insights from designer Michael Bierut, we uncover how Casey infused Swiss design principles with American ingenuity, led campus-wide design efforts, and created work that remains timeless and influential._______Support this podcast with a small donation: Buy Me A CoffeeThis show is powered by Nice PeopleJoin this podcast and the Patreon community: patreon.com/womendesignersyoushouldknowHave a 1:1 mentor call with Amber Asay: intro.co/amberasay_______Sources:Article: Eye Magazine Feature "Jacqueline Casey. Science and design"Article on Thérèse MollAbout Jacqueline (Jackie)Jacqueline Casey was a pioneering graphic designer whose work defined the visual identity of MIT for over three decades. Born in 1927 in Quincy, Massachusetts, she studied fashion design and illustration at MassArt before finding her way into graphic design. In 1955, her lifelong friend Muriel Cooper recruited her to the MIT Office of Publications, where Casey absorbed the principles of Swiss modernism and gave them her own twist—infusing wit, visual metaphors, and bold typography into her work.As Director of MIT's Design Services Office, she led campus-wide design efforts, creating posters and materials for scientific conferences, exhibitions, and cultural events. Her ability to simplify complex ideas with elegance and playfulness made her work timeless and celebrated globally. Today, her posters are held in the collections of MoMA, the Cooper Hewitt, and the MIT Museum, solidifying her as a quiet yet powerful force in modernist design history. About MichaelMichael Bierut is one of the most influential graphic designers of our time. A partner at Pentagram since 1990, his work spans iconic logos, brand identities, and campaigns for clients like The New York Times, Mastercard, and Saks Fifth Avenue. Bierut has been a longtime educator at Yale, co-founded Design Observer, and authored celebrated books, including How to Use Graphic Design to Sell Things. His work is held in the permanent collections of MoMA and the Cooper Hewitt, and his thought leadership continues to shape the design world.Follow Michael:Instagram: @mbierutWebsite: Pentragram.comThreads: @mbierut ____View all the visually rich 1-min reels of each woman on IG below:Instagram: Amber AsayInstagram: Women Designers Pod

We sat down with Colin and Antoine Massart from Massart Biketrails as well as our former guest and fan favourite Greg Jolliffe.Earlier in 2024, the boys were building the “Dreamline” for a Kriss Kyle edit in Glassie Bike Park and that video got released later in the summer to much fanfare. The trail itself is part of the bike park now and open to the public for everyone to enjoy! In our chat, we get to hear some more insights into the build of the line, about working with Kriss and the many challenges of digging trail in the Scottish countryside. But the conversation turns into much more than that as we dive into the “heart in the art” of trail building, the inspiration and influences coming from the community, competing builders, multi-faceted athletes like Kriss, different disciplines or other sports altogether, progression in the sport and what it means to actually put these very diverse “dreams” of what a trail should be into the dirt.MentionsAll of our "trail building" episodes Greg Jolliffe: Instagram, Jolliffe Builds, Ep17 Massart Biketrails: Instagram, Website, Colin, Antoine Dreamline videos: Edit, Build series Ep1, Ep2, Ep3 and Ep4 Kriss Kyle: Instagram, Youtube Glassie Bike Park: Instagram, Trailforks, Ray Will

From her first memories of receiving the pure magic of a Lion King bike at the age of 5, through her most recent bike-packing adventure across New England, Jordan has a very wonderful and happy connection to bikes. We hear fun stories of learning about the “turnaround point” while riding with her mom as a kid, to being a pedicab operator while attending college at MassArt, and how she's found “her people” and a great home base living in Maine. Jordan is also an amazing visual artist, surfer, adventurer, tinkerer and advocate for making riding bikes accessible and approachable for everyone. She even brought along a surprise guest — Gordie the dog — to keep Louie the dog company as they played in the background. But she landed a Summer internship at Pixar and decided to drop out of art school to continue to work in industry and study independently online and though professional work. In October 2003 Maggie landed a job at ILM. She started in render support working in the trailer and had an interesting run in with a family of raccoons working the graveyard shift. She later worked as a technical director on films including; Star Wars: Episode III, Evan Almighty, Chicken Little 3D, Lions for Lambs, Lemony Snicket, Pirates of the Caribbean, and many others. She also worked developing the HDR pipeline. In 2008 Maggie left to work at the California Academy of Sciences to work on their first planetarium show. She then moved over to doing freelancing for various visual effects companies including Polygon, Cafe FX, Spy Post, and then Image Movers Digital. Then Maggie went to 343 Industries to work on Halo 4 & 5 followed by time at Microsoft helping develop the HoloLens. From there she spent a year at NVIDIA and then returned to ILM for a few years working with the ILMxLAB. Today Maggie is working at Google alongside Dan Goldman on Project Starline. Maggie is brilliant. Her career journey is a who's who of high-end technology. Her love of working in the space where art meets engineering has guided her choices. It was great fun to chat and hear about her story.

Learn about Lisa's passion for curating inspiration, her artistic process and what inspires her to create.    Inspire, create, do. We are all artists. Don't let anyone tell you otherwise. Including yourself. Lisa L. Sears is a fine artist creating paintings and drawings in water-based media on paper and canvas. She earned her BFA in studio art at the School of Visual Arts, University of North Texas, with additional painting and photography study at University of Texas and mixed media painting at Art New England through MassArt. Her work has been described as symbolic, mythic, emotional, colorful, dream-like and nightmarish, which mixed all together might equal the moniker “whimsical depression.”  Dedicated to making art available to the community, Sears has been involved in Malden public arts projects including several years with Windows Art Malden, the Switchbox Project, “Pictures at an Exhibition Reframed” (a free piano concert and art exhibit organized by Malden Creates), and a recipient of a Malden Cultural Council grant for “Lines Connecting Lines”— a collaboration between area poets and artists—which she created and curated. Additionally, she was involved in the workings of Malden Pops Up, and participated in its later restructuring as The Gallery Inc., serving as Exhibit Coordinator and Curator for six-months. She has also enjoyed exhibiting her work locally at Malden Public Library and UMA Gallery. Sears is once again involved with Malden arts through The Gallery@57 on Pleasant Street in Malden. One would be hard-pressed to think of any other artist working with glass whose work reflects as many varied and compelling styles as Dan Dailey's. From vessel forms to his Individuals to lamps, sconces and chandeliers, these beautiful, sometimes humorous pieces dazzle through a combination of colored glass and intricate metal work. No matter the format, Dailey's work expresses humanity, historical reference, and reverence for the natural world.  Dailey credits his successful career to his education in the arts. Born in Philadelphia in 1947, he attended Philadelphia College of Art, where he encountered glass through ceramic teacher, Roland Jahn, and discovered a mentor in William P. Daley, who taught basic design and color to his freshman class. Dailey, who completed graduate studies at Rhode Island School of Design (RISD) as Dale Chihuly's first graduate student, says: “Under Chihuly's influence, I focused totally on glass. That was a breakthrough for me. It was a lucky time for me to be there.'' Following graduate school, with the support of a Fulbright fellowship, Dailey moved to Italy and worked in Murano's famed Venini Factory during 1972 and 1973 as an independent artist/designer. He later worked with other established glass companies such as Critsallerie Daum in Nancy, France, and Steuben Glass Works, in Corning, New York. In 1973, Dailey returned to the US and established the glass program at the Massachusetts College of Art and Design in Boston, which he headed until 1985. Now Professor Emeritus, he transitioned into a new relationship with MassArt, creating a lecture series titled Materialism, in collaboration with Joe Rapone, a professor of design at the University of the Arts in Philadelphia. Dailey continues his role as independent designer at both Venini and Daum, and serves on the National Advisory Board for The University of the Arts.  Among his many awards, Dailey received a Fulbright Hayes Fellowship, Venice, Italy, 1972-1973 and a Fellowship at the MIT Center for Advanced Visual Studies, Cambridge, Massachusetts, 1975-1983. He was elected a Fellow of the American Craft Council in 1998, honored in 2000 with the Libensky Award, and in 2001 with the Masters of the Medium Award by the James Renwick Alliance. Shown in over 300 exhibitions, including a retrospective at the Renwick Gallery, his work is included in more than 50 museum and public collections internationally, and currently represented by Schantz Galleries, Stockbridge, Massachusetts; Hawk Galleries, Columbus, Ohio; Habatat Galleries, Royal Oak, Michigan; and Sandra Ainsley Gallery, Toronto, Canada.  Dailey's process for transforming glass into compelling and unexpected forms is almost as interesting on paper as it is in three dimensions. Drawings and watercolors are used to refine ideas, but also to direct his team, which can include glassblowers in Seattle; acid polishing in West Virginia; waterjet cutters in local machine shops; and cutting, grinding, metal working, and assembling assistants at his New Hampshire studio. Working from his titles forward, the artist keeps a list of thoughts and key phrases, illustrating words with the objects he makes.  He states: “I emphasized drawing as a teacher for many years, because it would help me to help somebody realize their own ideas. It doesn't have to be a beautiful drawing. It just needs to include information. However, in my own work, I make accurate drawings that really represent the piece.” Focusing part of his time on producing sculptural lighting and large installations for residences and public buildings, Dailey says being diversified has kept him continuously busy, though he notes, not everyone makes a connection between all of his work. “Someone interviewed me at an exhibition in Chicago and did not realize that I made all of the work on exhibit. She thought it was three different artists. It was the first time I considered that perhaps my work wasn't clearly all mine, even though to me it all looked like it belonged. If you look through my sketchbooks and see the black-and-white ink on paper drawings, you can see that as different as the finished work can be, it is all connected by my stylistic approach.” Emerging from the Studio Glass movement initiated by Harvey Littleton, Dailey's work goes beyond its historical glass roots to combine with metal in a variety of formats, all of which communicate a subjective, narrative message. A vast array of forms has always been required to express the multitudes of ideas generated by Dailey's mind, and style is the common thread that binds them.

Jacqueline Casey was born in Quincy, Massachusetts in 1927 and  grew up with a passion for art. While her parents encouraged her to study bookkeeping, she pursued art against their wishes by entering the Massachusetts College of Art. It was at MassArt that Casey met and befriended Muriel Cooper. In 1949, Casey graduated from MassArt with a degree in fashion design and illustration. She entered the workforce which was very gradually becoming more accepting of women and worked in fashion design, advertising, and interior design. Except none of them felt quite right. Frustrated, Casey took three months to travel across Europe and clear her head. In 1955, Muriel Cooper hired Casey to work with her at MIT's Office of Publications. Casey would soon become responsible for posters, catalogs, and other promotional materials. In 1958, Casey worked with Thérèse Moll, an assistant to seminal Swiss designer Karl Gerstner. Moll introduced Casey to the International Typographic Style. Casey was later appointed director of the Office of Publications, and MIT's public image became recognizably influenced by the International Typographic Style. Under Casey's directorship, the MIT Office of Publications adapted ITS and went on to play a critical role in popularizing Swiss modernism across America.TIMELINE1927 –  b Jacqueline Casey in Quincy, Massachusetts.1949 – Casey graduates from Massachusetts College of Art with a Bachelor's degree in Fine Arts and a focus in design/illustration.1955 – Casey is hired by friend and fellow MassArt alumna Muriel Cooper to work at MIT's Office of Publications.1957 – Cooper leaves MIT and Jacqueline Casey takes on a larger, more active role within MIT's Office of Publications.1958 – Casey that summer works with Thérèse Moll, friend and assistant to famous Swiss designer Karl Gerstner. Marketing for Humans: Bringing the Human Back into Marketing. Hello. I’m Christina Frei, founder of Innate Marketing Genius, where how you love to help people informs how you get business. Do you know your deepest why in serving others?  I have identified 5 Innate Marketing Genius types, and knowing your type answers this question. The types  include Nurturers, Adventure Guides, Door Openers, Steady Presences, and Celebrators.  Discover your genius type here.Season 5: Marketing HotseatsI decided that I needed to show the magic of people’s deepest why in action.  In Season 5 of Marketing for Humans, I get to do this. I put myself in a Marketing Hotseat,  invite dynamic entrepreneurs as my guests, and act as my guest’s marketing consultant for 45 minutes or so.  If you want some calmer, more authentic marketing ideas, these are your episodes. Sometimes, you’ll even get the input of my colleague, Jenn Wentworth, a digital marketing expert based in New Hampshire. Today we talk to Stephanie Audette Connor, a graphic designer based in Worcester County, Massachusetts.  Stephanie does beautiful work with clients, a great person to hire for a re-design, and she started with a BFA in graphic design from MassArt.  Then after working at a few agencies, Stephanie stuck out on her own and hasn’t looked back since.  In my Pepperlane community, I hear good things about Stephanie consistently, and today we’re going to look at her marketing through the lens of a Nurturer. That means Stephanie loves to make it safe for others to thrive. This factors into her work as a graphic designer and her marketing decisions, which you will see in a moment.  Please welcome, Stephanie Audette Connor.Meet with ChristinaStephanie Audette Connor

**Silent Bob is Silent no more! Director Kevin Smith brings his Mooby's Pop-up to Boston at House of Blues for a limited engagement. We got a chance to sit down with him and talk all the nerdy tidbits about Stan Lee, his upcoming Masters of the Universe: Revelation, helping restaurant workers in the pandemic, and his shot-from home sitcom, Son in Lockdown featuring his entire family and introducing Austin Zajur.** Snootch to the Bootch! Do617: So good to see you. you don't mind being on video, do you?. Kevin Smith: Not at all. That's why I wore the funny hat! By the way, I'm still happy to see you. Trying to stay alive, man. The restaurant business doesn't make it easy though. Because, suddenly, you get access to free food where you're like, "I'll take three orders of Hater Tots"! "I'm the man" Why not? It's my place. Free food! How are you feeling? Feeling good? I'm feeling fantastic. It's been, just past the three-year mark since the heart attack knocks wood you know, so far so good. Thanks for coming to Boston. Lovely to be here, man. Boston has always been a big part of everything. I was just telling my wife today. I was like, the first time I came to the Commonwealth was with Clerks. Like even though I lived in New Jersey, I'd never come up to Massachusetts. There was no call for it and stuff. Yeah. It was from Philly to New York. They're close. Boston's like a three-hour drive. So it wasn't until Clerks that I came to the city with the Boston Film Festival and then I kept going back and like, Chasing Amy and Dogma and stuff like that. And the boys are from here. And the boys being from here! You know, me and Scott (Moser) were involved with Good Will Hunting as well. So there's, always a wonderful tie to the city, to the community. Also, this is a big college town and at one point in my career, I was "The College Guy". Yeah. You're my "College Guy", I suppose. I went to MassArt right down the street from here. In the Nineties, we were looking up to you, man. You've been inspiring people forever. You were so close with Stan Lee, I got to interview Stan it was an honor. an honor. You've always said good things and, and you were taking care of him. And I just want to say, I appreciate that you did that. What a great life. So the thing about the loss that breaks the heart is he was a vital 90-year-old and he didn't have that the last year or two years of his life, the way. He was kind of tossed around and literally suffered from elder abuse. That shaved years off his life. He was so vital, but there came a certain point where he's not only was he has physically beaten down but just the thought of like, "this is what it's come to". How could you do that to everyone's Grampa? So, you know what I'm saying, who gave so much joy in his lifetime, but at least, you know, he had that entire ten-year period of Marvel cameos, where at the end of his life, he got celebrated by the entire world. Everybody knows his name, even people that never heard of him before were, "Oh, that's Spider-Man's baby daddy". You know, maybe it was your idea to put him in the films? I remember seeing them in a movie years ago. Yeah. That was, for us, it was a no-brainer. Cause you got a movie about a guy that loves comic books. You got to Stan... Perfect marriage. And when we did it, I remember the movie came out in '95, there was just like, "Oh, Who? Stan Lee? That's quaint". You know, years later it became. A stamp of quality. I love seeing him in that movie. He's younger, obviously. I remember when he came to work with us, they were telling us "you gotta be careful, man, because this man is 73 years and he can go at any point". And he lasted another 23 years! *God bless him, man. Well, another thing, you've come here to Boston, to the restaurant at the House of Blues, which is a great venue. Lots of people, we've been out of work, but there are ways that people can keep it going. I appreciate that you came here to help us out. * I first heard of the Mooby's Pop-Up in the show that you did, Son in Lockdown and, that was just such a great thing to have your wife at that and Harley and Austin. Was it fun to just shoot if you're own your house? It was so adorable, especially considering it was in the middle of the quarantine and nobody was doing anything. So this TBS show Celebrity Show-Off presented itself as an opportunity. And they were like, "you can do anything you want, we're gonna give you a camera, shoot, whatever you want". And I was like, "can I shoot a sitcom"? They're like, "absolutely go ahead". So the entire town was shut down and you know, it's dopey to say, but we were literally the only sitcom in town that was shooting. It was a weird moment and it was fun to do it. It's the best job in the world, you don't even have to get dressed and drive to the studio. You wake up and you weren't in the studio. So it was such a good time! But it was a Kevin Smith joint, man! It was really, really funny Austin is such a rockstar! He's great. I put him in Clerks III... Do you have any updates, about Masters of the Universe: Revelation? I think the first time folks are going to see any imagery is coming up in May. We're in April right now. So next month. Do you like animation? I love it. It's phenomenal. The boys at Powerhouse Animation did a wonderful, wonderful job. They do_ Castlevania_ on Netflix, Seis Manos, Blood of Zeus. So all the characters look exactly the way they're supposed to. They just look like they were drawn now as opposed to the '80s. And so hey, if you want to talk about people who elevate it, man. I love the scripts that we came up with, but Powerhouse absolutely knocked it out of the park. They elevated our scripts and made far better cartoons than we provided them the "word power" to do so. Watching the whole thing come together, man. Because so many people work on it from your writing staff to then, your recording, and the vocal tracks. Then it all gets handed over to the animators and they do. iI's like movie making and it's like comic books at the same time. What's really wonderful about it is when you're working on a movie or something or a TV show, you know, you're seeing results on a regular basis because you're doing it. You got this window of time to do it when you're just the writer or showrunner on this animated series. My main job of just "let's get this thing written" was done early for us, it was the first job done. So I go about my life and then every few days there's a delivery of an art pack and you're like, "Holy shit". They drew all the characters and then as time goes on, it's like, "here's the first animatic". "Here's the second animatic". "This animatic is locked. Here's the automatics for episode two". Episode three, four. Oh my God. The finished animation for episode one is coming in. So it's this, this waterfall of productivity that you have nothing to do, with, other than getting to appreciate it. You wrote and your part is done. I mean, you can't go and make a script change. We can, as long as "lip flap" is off-camera. That continued and as much as periodically, we need to pick up these lines, we need to add new lines. Netflix thinks this needs clarity. So then you come up with that stuff and make that work and stuff. So you might need to even make new scenes, etc... Generally, it's not too deep in the process and going like, "Oh, I'm gonna write a whole new scene". if you do that, it's a big deal because the process is eight to ten months in. And once it starts going, it starts going, but being at the receiving end of all, the joy, the here's what Battle Cat looks like, you know, Oh my God, this is what Evil-Lyn looks like. This is what Evil-Lyn looks like with the helmet off. Every design before actually beginning to see the animatics and the animatics is the barest form. It's a digital storyboard or a moving storyboard. And even those were so pretty. I was like, "we could air this, fuck finishing, let's air this"! Let's just say thankfully Powerhouse push to finish it because the finished product is gorgeous. You can't just put out the pencils, you gotta ink this thing! I would! I come from comics, so I like when they do. So I love when they do Batman Black and White and just letters inked and pencils version. So I could have dealt with it, but people will be delighted to see how colorful the whole thing is. *Not for nothing. I love your Green Hornet. It was so great, I didn't expect to like the character, I mean, I liked what you did in comics, but I was just like, "Oh, this is a really great thing". It was drawn really awesome. * Jonathan Lau, man. He made that him and Phil. Phil Hester took my stuff and then he broke it down. Um, but wow. They turned it into a beautiful book. What was so satisfying about that for me was at the end of when I read all the books together, all the comics together... I was like, "It would have worked. We could have pulled that off as a movie!" Somebody could have, not me! *Absolutely. And coming up with comics and you are reading Batman, you are reading Spider-Man. You don't think of Green Hornet for comics, but you can do something cool with it. I could talk to you all day! * I tried to come up with the best "Cantina" question for you, one that I would ask on your podcast Fatman Beyond and not be embarrassed. But before I get to that, I watched the scene last night with the Ball Lickers, on the "Mooby's Internet" internet. So, you know, people get a chance to come down here and they can... ...and actually relive it. You are the ones who are the Ball Lickers! They can just do that from their phones! That was just so funny. But my, my "Ultimate Cantina Question" is about the Snyder Cut! Let's say Warner or whoever gives you $30 million dollars to redo a Kevin Smith movie, or to redo the CGI. What Kevin Smith movie would you give the #SnyderCut treatment to? Is it Tusk? I would go Red State. I would just shoot the ending. We cheaped out at the ending because we don't have any money. So, the ending was supposed to be the apocalypse. Angels showing up, slaying people and shit like that, these giant angels. So at a certain point, we were like "Alright, do you want to shoot the ending of that movie and that ending?" "Do you want to wait to shoot that movie in that ending for however long it takes to find the money for it?" "...or do you want to shoot a version of the movie now where you've let go of the supernatural element that was in the last scene?" So then I changed it to him being over the loudspeaker and "it was a hoax and blah, blah, blah". So if I had like #SnyderCut money, I'd go back and I would shoot the angels show up, ending like the apocalyptic world ending. I'd have a lot of money left over! But I think that would be the one that I would love to take in the direction of the original vision. And I love Red State what it became and stuff, but that ending would have been so fucking weird. Do617: Well, you know, when people after seeing this interview, they might demand the release of the #SnyderCut of Red State. KS: Release the #RedderCut! Yes! Well, I'm just so happy to talk to you, man. I'm really excited to be here at Mooby's. Thanks for helping out Boston. Thanks for helping out everybody here at the House of Blues Restaurant and, everybody makes a couple of bucks and people can have some fun doing it because you came here. I think this is a perfect time. just about to open up. This is like a preview. This is a preview of the good time for the summer that's going to come. They were asking me when I rented the car today, when I was leaving Logan, the lady was like "You're from, California, how is it about there? I was like "the exact same thing as it is here. You guys are about to open". That's been the fun part of taking Mooby's around the country is that it's always at a restaurant that's been closed for a while. So it's good for the business. It's also good for the staff morale. You can forget when you're in the restaurant business, your friends are your coworkers, you see them every day. A lot of folks haven't seen their friends for a year and stuff. "Zoom Cocktail Hour" just doesn't cut it. It really, really doesn't at the end of the day. At the end of the day, being in a room with people, human interaction, I'm not saying you got to lick each other's faces and shit..being in a room with people and talking and nothing will replace it. Well, thanks so much, Kevin. It's an honor, man! Damn pleasure man! ** BUY TICKETS AT THE LINK ABOVE AND LEARN MORE: http://moobyspopup.com (http://moobyspopup.com)** Special Guest: Kevin Smith.

Gene-corrected Parkinson's disease neurons show the A30P alpha-synuclein point mutation leads to reduced neuronal branching and function

M., Pavelka, L., Kwon, Y.-J., Krueger, R. A. Abstract: Parkinson's disease is characterised by the degeneration of A9 dopaminergic neurons and the pathological accumulation of alpha-synuclein. In a patient-derived stem cell model, we have generated dopaminergic neurons from an individual harbouring the p.A30P SNCA mutation and compared those neurons against gene-corrected isogenic control cell lines. We have used confocal microscopy to assess the neuronal network, specifically segmenting dopaminergic neurons and have identified image-based phenotypes showing axonal impairment and reduced neurite branching. We show using multi-electrode array (MEA) technology that the neurons carrying the endogenous p.A30P alpha-synuclein mutation are functionally impaired and identified mitochondrial dysfunction as a pathogenic cellular phenotype. We report that against gene-corrected isogenic control cell lines the neurons carrying the p.A30P SNCA mutation have a deficit and are susceptible to the mitochondrial toxin and environmental pesticide Rotenone. Our data supports the use of isogenic cell lines in identifying image-based pathological phenotypes that can serve as an entry point for future disease modifying compound screenings and drug discovery strategies. F., Massart, F., Grossmann, D., Shiga, T., Ishikawa, K.-i., Akamatsu, W., Finkbeiner, S., Hattori, N., Krueger, R. Abstract: Parkinson's disease (PD) is characterized by the loss of A9 midbrain dopaminergic neurons and the accumulation of alpha-synuclein aggregates in remaining neurons. Many studies of the molecular and cellular basis of neurodegeneration in PD have made use of iPSC-derived neurons from patients with familial PD mutations. However, approximately half of the cells in the brain are glia, and their role facilitating neurodegeneration is unclear. We developed a novel serum-free protocol to generate midbrain astrocytes from patient-derived iPSCs harbouring the pathogenic p.A30P, p.A53T mutations in SNCA, as well as duplication and triplication of the SNCA locus. In our cellular model, aggregates of alpha-synuclein occurred only within the GFAP+ astrocytes carrying the pathogenic SNCA mutations. Assessment of spontaneous cytosolic calcium (Ca2+) release using Fluo4 revealed that SNCA mutant astrocytes released excess Ca2+ compared to controls. Unbiased evaluation of 3D mitochondrial morphometric parameters showed that these SNCA mutant astrocytes had increased mitochondrial fragmentation and decreased mitochondrial connectivity compared to controls, and reduced mitochondrial bioenergetic function. This comprehensive assessment of different pathogenic SNCA mutations derived from PD patients using the same cellular model enabled assessment of the mutation effect, showing that p.A53T and triplication astrocytes were the most severely affected. Together, our results indicate that astrocytes harbouring the familial PD mutations in SNCA are dysfunctional, suggesting a contributory role for dysfunctional astrocytes in the disease mechanism and pathogenesis of PD. Today Clyde and I welcome a very special guest: Julie Toole. During our conversation, we look at what is white privilege and how we can practice DEI+J in the TAB studio,   Julie has been a teacher for 30 years, 20+ in Chicago Public schools and is currently teaching art at a private progressive school, pre-k-8th grade. She has been a TAB teacher and TAB mentor for the last 10 years and co-lead at the elementary level group at the TAB Summer Institute at MassArt and part of the Executive Board for TAB, INC. As a transracially adoptive mother of two children of color, She has been on a personal journey on understanding her whiteness and privilege in order to be actively anti-racist in her work and life. This has led her to participate in SEED (Seeking Educational Equity and Diversity) cohorts and then to do the training to become a SEED facilitator at her school. She has co-led groups for parents and for staff. She is also part of the EQaT (equate) team at her school (Equity in Action), the parent EqAT group, and teaches an elective course for her middle schoolers called Artist as Activist. Five years ago she started a GSA (Gender Sexuality Alliance) club at her school and together they do school-wide events such as Day of Silence and Ally Week.  Show Notes: White Privilege  Unpacking the Invisible Knapsack Windows and Mirrors of Your Child's Bookshelf - Ted Talk William Pinar - What is Curriculum Theory? What started as Design Museum Boston, now includes Design Museum Portland in Oregon, and is growing across the country. Their mission is to educate the world about the importance of design in our lives. Design is everywhere, so the museum has no permanent address — instead, Sam and his team turn the museum inside out and turn entire cities into locally-focused, nomadic design museums. Sam earned a BFA in Industrial Design from Rochester Institute of Technology and went on to design consumer electronics at Bose Corporation, as well as to teach design at Wentworth Institute of Technology, MassArt, and Babson College — where he recently earned his MBA. With a passion for design, creativity, and learning, Sam creates a long-term vision for the museum. He leads a dedicated team while managing key programs and strategic operations. In 2013 Sam and his team were named one of BostInno’s 50 on Fire. Sam routinely speaks at conferences, keynotes, and workshops about design, entrepreneurship, innovation, branding, placemaking, and business strategy. Recent speaking engagements include IBM SmartCamp, Future M, TEDxBoston, TEDxFenway, Portland Monthly’s Bright Lights, MIT Hacking Arts, and Harvard iLab. What is Design Museum Boston? We’re a new kind of museum focused on educating the world about design. We’re based in Boston, but we don’t have our own gallery space. We’re a distributed museum with exhibits & events all over town. What is a nomadic museum? We turn the museum inside out, turning the entire city into the museum. We put exhibitions and events in places where people already go: retail space, public space—even outside. What will I learn at Design Museum Boston? From architecture to video game design, furniture to fashion, products to graphics, design is more than an industry: it’s a process and way of thinking that solves complex problems. Do I need to be a designer to visit the museum? Absolutely not! Design is for everyone! We offer the most accessible, inclusive way to engage with design and designers. Our exhibitions and events are open to all. Nick Sherman is a typographer and typographic consultant based in New York City, co-founder of Fonts In Use and columnist at A List Apart, serves on the board of directors for the Type Directors Club, the Adobe Typography Customer Advisory Board, as well as the artistic board for the Hamilton Wood Type & Printing Museumh He is both a graduate of and consultant for the typeface design program at Cooper Union. He graduated with honors from the Graphic Design program at MassArt in Boston, where he also taught undergraduate typography and letterpress printing. Nick has worked at Font Bureau, Webtype, and MyFonts, directing web design and promotional material for typefaces in print and digital media. In this interview we talk about Nick’s rituals involving pizza. We wonder if there’re any similarities between pizza and type. Nick explains how he got interested in graphic arts at an early age through music. And how he later on started focussing more on typography. We also talk about the project he did for the Hamilton Wood Type Museum and his work for MyFonts, Font Bureau, Webtype and Fonts in Use. We wonder if Nick can see how type trends go, where it’s getting influenced by, and if he thinks that we’re reaching some sort of a saturation point? Recorded at the Robothon 2015 Conference in The Hague, the Netherlands. The idea of selecting a model via penalizing a log-likelihood type criterion goes back to the early seventies with the pioneering works of Mallows and Akaike. One can find many consistency results in the literature for such criteria. These results are asymptotic in the sense that one deals with a given number of models and the number of observations tends to infinity. A non asymptotic theory for these type of criteria has been developed these last years that allows the size as well as the number of models to depend on the sample size. For practical relevance of these methods, it is desirable to get a precise expression of the penalty terms involved in the penalized criteria on which they are based. We will discuss some heuristics to design data-driven penalties, review some new results and discuss some open problems.
