Abstract: Background: The Microtubule-Associated Protein Tau (MAPT) is one of the proteins that are central to neurodegenerative diseases. The nature of intracellular tau aggregates is determined by the cell types whether neuronal or glial, the participating tau isoforms, and the structure of the amyloid filament. The transmembrane protein 106B (TMEM106B) has recently emerged as another significant player in neurodegeneration and aging. In the central nervous system, the composition of the gray and white matter differs considerably. The gray matter consists of nerve cell bodies, dendrites, unmyelinated axons, synaptic terminals, astrocytes, oligodendrocytes (satellite cells) and microglia. The white matter differs from the gray for the presence of axonal tracts as the only neuronal component and for the absence of nerve cell bodies, dendrites and synaptic terminals. Cryogenic electron microscopy (cryo-EM) studies have unveiled the structure of tau and TMEM106B, from the cerebral cortex, in several neurodegenerative diseases; however, whether tau and TMEM106B filaments from the gray and white matter share a common fold requires additional investigation. Methods: We isolated tau and TMEM106B from the cerebral cortex and white matter of the frontal lobes of two individuals affected by multiple system tauopathy with presenile dementia (MSTD), a disease caused by the MAPT intron 10 mutation +3. We used immunostaining, biochemical, genetics and cryo-EM methods to characterize tau and TMEM106B. Results: We determined that tau filaments in the gray and the white matter of MSTD individuals can induce tau aggregation and have identical AGD type 2 folds. TMEM106B amyloid filaments were also found in the gray and white matter of MSTD; the filament folds were identical in the two anatomical regions. Conclusions: Our findings show for the first time that in MSTD two types of amyloid filaments extracted from the gray matter have identical folds to those extracted from the white matter. Whether in this genetic disorder there is a relationship in the pathogenesis of the tau and TMEM106B filaments, remains to be determined. Furthermore, additional studies are needed for other proteins and other neurodegenerative diseases to establish whether filaments extracted from the gray and white matter would have identical folds. G., Dalkara, T., Karatas, H. Abstract: Migraine is a neurological disorder characterized by episodes of severe headache. Cortical spreading depression (CSD), the electrophysiological equivalent of migraine aura, results in the opening of pannexin-1 megachannels that release ATP and triggers parenchymal neuroinflammatory signaling cascade in the cortex. Migraine symptoms suggesting subcortical dysfunction bring subcortical spread of CSD under the light. Here, we investigated the role of purinergic P2X7 receptors on the subcortical spread of CSD and its consequent neuroinflammation using a potent and selective P2X7 antagonist, JNJ-47965567. P2X7 antagonism had no effect on the CSD threshold and characteristics but increased the latency to hypothalamic voltage deflection following CSD showing that ATP acts as a mediator in the subcortical spread. P2X7 antagonism also prevented hypothalamic neuronal activation following CSD, revealed by bilateral decrease in hypothalamic c-fos positive neuron count. P2X7 antagonism further stopped the CSD-induced neuroinflammation revealed by decreased nuclear translocation of NF-kappa B-p65 in astrocytes and decreased HMGB1 release. Following CSD we observed an increase in neuronal cytoplasmic P2X7R signal in the cortex and subcortical structures (thalamus, hypothalamus, striatum, hippocampus) concordant with the neuroinflammation which is also prevented by P2X7R antagonism. In conclusion, our data suggest that P2X7R plays an imperative role in CSD-induced neuroinflammation, the subcortical spread of CSD, and CSD-induced hypothalamic neuronal activation hence can be a potential target in migraine treatment. J., Karatas-Kursun, H., Dalkara, T., Ozkan, M. Y. Abstract: Ischemic stroke results in sudden blood flow cessation, thus, unmet energy requirements. Although the clotted artery can be recanalized and blood flow is restored, brain perfusion may not be fully attained due to microvascular constrictions. Under glucose-deprived and hypoxic conditions, glucose derived from the glycogen stored around peri-microvascular astrocyte end-feet may serve as an emergency fuel to meet the metabolic demand during the acute period of ischemic stroke. To elucidate the impact of glycogen utilization on brain microcirculation, we administered glycogen phosphorylase inhibitor 1,4-dideoxy-1,4-imino-d-arabinitol (DAB) intracerebroventricularly. Transgenic mice in which glycogen synthase-1 expression was selectively knocked out in central nervous system (GYS1Nestin-KO) were also used. Both approaches caused microvascular constrictions mediated by CD13-positive pericyte contractions. When mice with disrupted glycogen utilization were subjected to MCA ischemia, pericyte-mediated microvascular constrictions and the infarct volumes were further increased compared to untreated controls or wild-type littermates. Peri-microvascular glycogen depletions were highly correlated with microvascular constrictions as shown by Periodic acid Schiff (PAS) staining and immunolabeling with anti-glycogen antibodies. Imaging of regional cortical blood flow changes during ischemia disclosed severely compromised blood flow dynamics in mice with disrupted glycogen metabolism. In conclusion, disrupting glycogen utilization causes ischemic-like microvascular constrictions under non-ischemic circumstances and increases susceptibility to brain ischemia. Understanding the role of glycogen at neurogliovascular level in brain may provide novel insight to the pathophysiology of ischemic stroke and therapeutic opportunities. Hij vindt zichzelf pas de beste wanneer hij Petrosyan heeft verslagen. Buiten de ring geeft hij aan een heel rustig persoon te zijn: “Als ik bloed zie word ik al misselijk.” Maar in de ring komt hij in kill modus.  Zijn motivatie: “Ik kom van de bodem. Ik wil niet terug naar mijn verleden. Daar vecht ik tegen. “  Slapeloze nachten  De jeugd van Tayfun was niet makkelijk. “Ik begon onder de nul.” Als kind uit een gezin van negen personen, een oververmoeide moeder en een vader met meerdere verslavingen, groeide Tayfun op met een slechte basis. Er was zelden geld voor eten en geen geld voor sport. Hij schaamde zich wanneer hij als kind publiekelijk werd aangesproken dat de contributie niet was betaald. Hij vertelt dat zij 's nachts niet konden slapen, door de verslaving van hun vader. Er kwamen dan veel vreemde mensen over de vloer, er was lawaai en soms was iedereen ineens weg en stonden alle deuren open. Nog steeds heeft hij de gewoonte om rond 5:00 uur wakker te schieten en te controleren of alles op slot is. Inmiddels is zijn vader op eigen kracht na vijfentwintig jaar verslaving clean. Tayfun zegt: “Ik ben dankbaar dat ik dit heb meegemaakt, omdat ik mijn huidige leven nu extra kan waarderen.”  Ongekende liefde  Ondanks deze achtergrond ervoor heeft gezorgd dat hij moeilijk mensen toe laat, heeft hij het geluk in de liefde gevonden. Inmiddels is hij gelukkig getrouwd. Hij vertelt dat zij hem geeft wat hij nooit heeft gekend. Zij is de ontspanning wanneer hij thuiskomt. “Mijn moeder was uitgeput, dus liefde kenden wij niet. Mijn moeder had geen tijd om liefde te geven.”  Haatberichten en de beste lessen  Tayfun heeft van jongs af aan een sterke focus gehad voor kickboksen. Hij had altijd veel bewondering voor Andy Souwer en was dan ook verheugd om tegen hem te mogen vechten. Deze partij heeft Tayfun gewonnen. Hij vertelt: “Voor deze partij was ik het meest gemotiveerd. Ik was zo zenuwachtig dat ik de nacht ervoor niet kon slapen.” Ook vertelt hij over de partijen tegen Mohammed Jaraya en dat hij in deze tijd veel haat berichten kreeg door deze rivaliteit. Buiten de ring zijn deze twee kickboksers op goede voet met elkaar. Tayfun geeft zelfs aan dat deze partijen de beste lessen uit zijn leven zijn geweest. Volgens hem zit er zeker nog een partij aan te komen. “Dan ga ik hem weer kapot slaan!”  Voorlopig aan de top  Over de toekomst geeft hij aan dat hij niet bezig is met een carrière na kickboksen. Omdat kickboksen zijn passie is en hij zich hier extreem voor inzet, is hij nooit bezig geweest met een andere focus. The Oak Ridge National Laboratory is one of the 17 Department of Energy laboratories. It has the largest multi-program science and energy research lab in the U.S. On this latest episode of Sustainably Speaking, we discuss the new partnership between Oak Ridge and the American Chemistry Council. In this scope, Ozcan facilitates the development of composite recycling technologies and utilizes various composite techniques to repurpose them into useful applications. Ozcan and his team are developing manufacturing techniques and exploring new materials to improve energy efficiency during composite manufacturing, decrease material waste, lower costs, and improve material performance. This panel explored the interconnectedness of Turkey's domestic and foreign politics over the past two decades. How do geopolitical histories and imaginaries affect Turkey's foreign policy? What are the links between everyday culture and Turkey's foreign policy? To what extent have global and regional developments impacted on and informed domestic politics? In what ways has foreign policy been used as a technique of governance? Evren Balta is Professor of International Relations and chair of the International Relations Department at Özyeğin University. She is the author of 'The American Passport in Turkey: National Citizenship in the Age of Transnationalism' (with O Altan-Olcay, UPenn, 2020), 'Age of Uneasiness' (İletisim, 2019) and 'Global Security Complex' (İletisim, 2012). She is the editor of 'Neighbors with Suspicion: Dynamics of Turkish-Russian Relations' (with G. Ozcan and B. Besgul, İletisim, 2017); 'Introduction to Global Politics' (Iletisim, 2014) and 'Military, State and Politics in Turkey' (with I. Akca, Bilgi University Press, 2010). She served as a research fellow at the Institute for Human Sciences/Russia in Global Dialogue Program (Vienna, Austria 2017) and as a Fulbright visiting associate professor at New York University, Program in International Relations during the 2017-2018 academic year. Balta is a senior scholar at Istanbul Policy Center, a member of the Global Relations Forum and co-editor of International Relations Journal. She was appointed as the academic coordinator of TÜSİAD Global Politics Forum in 2021. Lisel Hintz is an Assistant Professor of International Relations at Johns Hopkins University's School of Advanced International Studies. She was a postdoctoral fellow at Cornell University and was visiting assistant professor at Barnard College, Columbia University. She studies the arenas in which struggles over various forms of identity – e.g., national, ethnic, religious, gender – take place. Her regional focus is on Turkey and its relations with the Middle East, Europe, and the US. Her 2018 book 'Identity Politics Inside Out: National Identity Contestation and Foreign Policy in Turkey' (Oxford University Press) examines how Turkey's Justice and Development Party (AKP) used foreign policy gambits to weaken its domestic obstacles and open up space for disseminating its own Ottoman Islamist understanding of Turkish national identity and, ultimately, the ways in which contestation over national identity spills over to shape and be shaped by foreign policy. Her current book project, under contract with Cambridge University Press, investigates Turkey's state-society struggles over identity in the pop culture sphere. Her work also appears in journals and news outlets contributing to discussions on Turkey's increasing authoritarianism, opposition dynamics, foreign policy shifts, and identity-related topics including Kurdish, Alevi, and gender issues. Spyros A. Sofos is a Researcher at the LSE Middle East Centre. Spyros has been a member of the Fragmentation of peacemaking and peacebuilding: Non-Western dynamics of peace and transition management project team, funded by the FCDO and the PeaceRep Consortium. His research explores the intersection of societal insecurity, identity, and collective action and, to date, it has focused on Turkish politics and society, nationalism, populism and Islamism in Europe and the Middle East, urban citizenship, and European Muslim identities and politics. His latest book Turkish Politics and ‘The People': Mass Mobilisation and Populism (Edinburgh University Press) – explores the emergence of populism in Turkey and its genealogy as a tradition of action and discourse. His other publications include 'Nation and Identity in Contemporary Europe' (Routledge), 'Tormented by History: Nationalism in Greece and Turkey' (Oxford University Press), 'Islam in Europe: Public Spaces and Civic Networks' (Palgrave).

Abstract from the paper we discuss with the author during the episode: Digital platforms have disrupted many sectors but have not yet visibly transformed highly regulated industries. This study of Big Tech entry in healthcare and education explores how platforms have begun to enter highly regulated industries systematically and effectively. We present a 4-stage process model of platform entry, which we term as “digital colonization”: (1) provision of data infrastructure services to regulated incumbents; (2) data capture in the highly regulated industry; (3) provision of data-driven insights; (4) design and commercialization of new products and services. We clarify platforms' sources of competitive advantage in highly regulated industries and conclude with managerial and policy recommendations.

Today, we’re joined by Aydogan Ozcan, Professor of Electrical and Computer Engineering at UCLA, where his research group focuses on photonics and its applications to nano- and biotechnology. In our conversation, we explore his group's research into the intersection of deep learning and optics, holography and computational imaging. We specifically look at a really interesting project to create all-optical neural networks which work based on diffraction, where the printed pixels of the network are analogous to neurons. We also explore some of the practical applications for their research and other areas of interest for their group. As a miner, chances are you're not too knowledgeable about law. But this ignorance, according to the latest guest on the Full Production podcast, is precisely what causes so many legal issues in our industry.   Müge Ozcan is a WA-based lawyer whose firm, Vitalis Legal, helps miners (and others) with employment issues, contract disagreements and other legal situations.
