Most importantly: They can cover themselves in a magical ash that will boost their might and give them a Fire resistance. Once they do this, they sort of become solid all-arounders, so it's best to knock them out before they can do that. Starter Tip: Hit them with Dark or Ice damage to bring them down to melee range! There's basically only one way through his conversation tree that doesn't draw you either into combat with him immediately (with reinforcements coming soon after) or up into the Barracks where his friends are. Personally, I prefer to just knock him out ASAP and then sneak up to the hidden passage a few rooms away. Starter Tip: If he manages to poison you, don't forget to use your IP for an Antitote. Poisoned doesn't do damage over times like in some games, instead it saps BOTH your Might and Willpower, so whether it hits a bruiser or a mage, you're in trouble!  Ushala, Conduit Traits: Mysterious, Possessed, Sacrificial Type: Demon Mage Level: 10 Rank: Soldier Stats: DEX 10, INS 8, MIG 10, WLP 6 Attacks: Heaven's Light, Heaven's Glory, Heaven's Call Special Abilities: Flying, Pressure Gauge Elemental Affinities: IMM: Light | RES: Physical, Poison | VUL: Dark | Pressure Points: Dagger, Sword In-Game Description: A member of the Creed who temporary gives their body up to be possessed by the spirit of the holy Oracles.  So, as far as I know, there's two versions of Ushala you can fight. The above is the "standard" one, which is what happens when the Gem Scene goes the way it almost always does. But I've heard that if you manage to get the gem away from the Draconic Elf safely, then Ushala can become possessed (by who? My theory is Lucena) and then she gets a second, much harder phase. Which is saying something, because even this version of her is NO JOKE. Starter Tip: If you can, hit her with Shaken and Poisoned (the status condition, NOT the damage type). All three of her attacks are based on WLP, so it helps to knock that down as far as possible. Starter Tip: Because she has a pressure gauge (from the Armidirge games), she doesn't take extra damage from the elements she's weak against. Instead, if you can hit her six times with something she's vulnerable to (including her pressure point weapon types), she'll get staggered and she loses all of her actions for the rest of the round. In this explainer episode, we've asked Ella Davyson, Genomics Data Scientist, to explain the meaning of the term genomics. [00:00:00] Florence: What is genomics? My name is Florence Cornish, and today I'm joined by Ella Davyson, who is a genomics data scientist here at Genomics England, and she is here to explain the topic in much more detail So, Ella, we obviously both work at Genomics England. This podcast is called Genomics 101, so I guess it's fitting that we have an episode dedicated to explaining the term 'genomics'. [00:00:26] But before we get into that, I think it would be good if you could first explain what we mean by the term 'genome'. [00:00:32] Ella: Thanks, Florence. The genome is, essentially you can think of it like a manual booklet, or instructions that the body uses in how to grow, survive, and function, and this is a manual that's in every single cell within our body, and it tells our cells exactly how to divide, how to survive. [00:00:54] For example, the genome in the pancreas, in pancreatic cells will tell those cells how to produce proteins such as insulin that we need to control our blood sugar. And also, the genome within our eye cells will tell the cells how to generate photoreceptors to enable us to see. So the genome is essentially like the ultimate guide that our body uses to tell it how to create everything that we need to survive going forwards. [00:01:25] Florence: So then, what do we mean by the term 'genomics'? [00:01:30] Ella: So, genomics is essentially the study of the entire human genome. So we study its structure and also how it functions, in terms of how is this instruction manual being read by the body, and how does that result in healthy human beings that we see today. [00:01:48] Florence: So when we're talking about studying DNA, lots of our listeners might have heard the term 'genetics', which kind of also refers to the study of DNA and genes, so it might be a little bit confusing. [00:01:58] So what's the difference between the two? What's the difference between genetics and genomics? [00:02:04] Ella: So genetics is specifically the study of genes in the genome, and genes are part of the instruction manual, that specifically tell the body to produce a certain thing. So, in our insulin example, there is an INS gene, so, which is the gene in the genome or the instruction manual that specifically tells the cells to make insulin and to produce this product. [00:02:30] There are many different genes in our genome, and genetics is the study of all of these. In contrast, genomics is the study of the entire instruction manual altogether, so that includes all of the genes in genetics and also everything else in the manual. So, genetics is limited to the study of these parts of the manual that clearly encode certain proteins or products such as insulin. Genomics is the study of everything all at once, everything under the bathroom sink. So yeah, the confusion I think can arise a lot because historically when we first started looking at DNA and researching genetics, we didn't have the technology to look at the whole genome all at once, and with older sequencing technologies we would focus on particular genes that we knew important for certain diseases. [00:03:19] So in diabetes, for example, they would instead specifically look at the insulin gene and see how does this influence diabetes, rather than looking at the entire instruction manual at once. Nowadays, we do have that technology, and that is what we do here at Genomics England, just use that to look at the entire genome rather than specific subsets of the genome, so specific genes. [00:03:45] We can look at everything in its entirety. So, you can kind of think of genomics as a much broader, more complete study of genetics. [00:03:56] Florence: So speaking of genomic testing, I don't know if you saw, but in the government's 10-year Health Plan that they published last year, they predicted that genomics could play a role in up to 50% of healthcare interactions. [00:04:08] Could you tell me a bit about why genomics is important in healthcare? [00:04:12] Ella: So that's a really exciting point, and I think one that we should be all striving towards. So, genomics can play a role in healthcare in so many different ways. I think before going into each of them, it's kind of maybe important just to illustrate that our genomes between two, two people are 99.9% the same. [00:04:38] So we're both humans. We are both the same species. There is 0.1% difference between two people's genomes, and those differences underlie all the uniqueness that makes a person a unique individual. [00:04:54] So personality, appearance and also risk to different health and disease outcomes. So that is where the role of genomics can come in, is to understand how the differences between people and their genetic makeup can influence maybe their risk for being more predisposed to developing a condition. Conditions such as Cystic Fibrosis or Huntington's disease that are specifically caused by genetic variants or mutations in genes that directly cause the condition. So it's a bit more maybe obvious, if you like, about how studying genetics in those, in those conditions can directly inform on how they arise, potential ways that we can better treat them. [00:05:52] So another way that genomics can be used in healthcare is through screening. So this is being piloted at the moment in the Generation Study by Genomics England which is applying whole genome sequencing to newborn babies to look for a range of conditions which are caused by genetic changes, all of which are treatable. [00:06:13] But importantly, screening will enable clinicians and families to know about these conditions much earlier and start life-changing treatment much, much sooner. So this is kind of already beginning to be, I think it will be showcased with this study in the next couple of years and the power of this in healthcare, I think can't really be overestimated. [00:06:40] Florence: And finally, just to finish off, is there anything coming up in the field of genomics that you're especially excited about? [00:06:48] Ella: There are loads of different things that I'm excited about in the field of genomics. I think probably maybe one that's most kind of relevant to clinical care is the possibility of doing more personalised medicine with treatments. [00:07:05] Often, at the moment, we majority have kind of one treatment for all when treating certain conditions, and sometimes these treatments aren't tolerated well by some people, and also some of these treatments just don't work well in some people as well. Sometimes there's a clear reason for these things, but more often than not, it's not entirely clear why some people might benefit more for some treatments or some people don't respond or don't react well to some treatments as well. [00:07:35] And understanding more, so there's a whole field about how genomics interacts with drugs and medicines, which is called pharmacogenomics, and its aim is to understand which medicine might be most effective or well-tolerated in certain people based on their genetics. And I think that will be kind of life-changing as well for some people, who are suffering from diseases where the medication is either not effective enough or is also affecting their quality of life. [00:08:10] Florence: Mm-hmm. [00:08:10] Ella: Because that is a whole other part of it as well is that sometimes these treatments for certain conditions are really hard to tolerate. [00:08:19] Other things that I'm excited about are just the technologies that are coming out at the moment mean that we can measure and understand a whole lot more about genomics than we used to be. So now we can say this gene is influencing this disease, but sometimes, you know, it's more complicated, and we now have the technology to measure all sorts of different things, so how our environment can influence our genes and how our genes react with each other. [00:08:57] So we're just getting, we're getting able to look at more and more, and I think we'll expand our understanding in a lot of conditions that unfortunately aren't very simple. Daniel came over from Norway for his first INS race and left with wins in both 2WD and 4WD Mod after a brutal weekend on one of the toughest carpet tracks we have seen. Where in the world am I? In San Diego, talking about Sao Paolo, Brazil Welcome back to the  Dr. Mary Travelbest Guide podcast. The FAQ is: Leslie asked, "After my injury/ operation, I'm concerned about travel. Can you tell me how hard it is to resume my former Step (1-5) for Travel? How should I set my expectations for future travel after I am healed? Answer:  The step for your next travel may be different than your last trip. For example, you may have been on an African safari, and that was a Step 5. Now you have had a broken bone, or you have had surgery, and you are wondering how to get back out there on your next adventure. You may need to re-group, drop down a step or two, and figure out your confidence again. You can always go on an overnight trip to a nearby city and start with Step 1 travel once again. There may be some things you missed or never saw that you've been meaning to visit, and now would be a good time to do so. Today's destination is São Paulo, Brazil.   São Paulo is the capital of Brazil and home to 22M people, a Step 5 destination.   São Paulo is worth seeing for women who like culture, neighborhoods, museums, and food, but it is not the easiest first-choice city in South America for a solo woman over 50. It can feel gray, crowded, and tiring. The traffic is real. The scale is real. The safety concerns are real. But if you stay in the right area, move with intention, and do not try to conquer the whole city, you can have a rich two-day visit that feels strong, smart, and independent.   I booked a cheap place on a travel website in São Paolo near the airport. Please listen to my mistakes at the end to understand why not to do that.   If you just arrived, you may want to take the metro into town. Get a 24-hour pass for about $.-5 Or just buy individual tickets. Metro here is free for people over 60, so just show your passport upon arrival at the station. I have a story at the end about that also.   If you are planning to go, here is a 2-day itinerary, along with a few of my comments. Day 1: Start with the easier parts of the city Morning: Avenida Paulista and MASP area Begin on Avenida Paulista because it is one of the city's best-known and most practical starting points for a solo visitor. It is central, busy, and lined with museums, cafes, shops, and hotels. MASP is one of the city's landmark museums and a strong anchor stop, rather than just wandering without a plan. São Paulo's official visitor materials also highlight Paulista as one of the city's defining areas. Midday: Long lunch instead of overpacking the day Do not try to "do São Paulo" in one sweep. Build in a proper lunch near Paulista or Jardins. This city can wear you out. Traffic, sidewalks, noise, and decision fatigue are real. Afternoon: Parque Ibirapuera Head to Ibirapuera Park for a calmer second half of the day. Official city materials list it among the major attractions, and it is a better late-afternoon choice than pushing deeper into more chaotic areas when your energy is lower. Evening: Dinner close to your hotel This is where I would be critical. São Paulo is not the city where I would tell a solo woman to "go out and see what happens" at night. Have dinner in a well-reviewed area near where you are staying, and use a car service back if needed.  Day 2:  Morning: Liberdade Liberdade is one of São Paulo's signature neighborhoods and gives you a different side of the city. Go in the morning, when you are fresh, and the area feels more manageable. It is photogenic and culturally distinct. It felt like an asian-like atmosphere for me. I felt the Japanese and other asian cultural influences, which made it different than other parts of the city. What to watch out for: Do not confuse "interesting" with "relaxing." Some parts can feel crowded, messy, and overstimulating. Keep valuables out of sight and do not stand around consulting your phone at the curb. Lunch: Stay put, then move intentionally Have lunch there or in a nearby planned stop. Avoid zigzagging across the city without a clear reason. In São Paulo, too much transit can waste time and cause more hassle. Afternoon option A: Municipal Market, if you like food stops The Mercado Municipal is iconic and worth considering for a focused visit, not an all-day outing. Go, sample, look around, then leave. Afternoon option B: Easier finish in Jardins or back to Paulista If you want a smoother second day, return to a more polished area such as Jardins or the Paulista zone. This is the better choice if you are tired, jet-lagged, or feeling cautious. Evening: End early I would lean toward Paulista/Jardins rather than picking a cheaper stay in a less convenient area. That is not because those neighborhoods are risk-free. They are not. It is because being in a more established, service-rich area usually makes solo travel simpler and lowers friction. São Paulo can be rewarding, but it is not a city to treat casually. Official advisories warn about crime, including street crime, and São Paulo's own Metro provides a dedicated safety reporting channel, which tells you something important: security is an active issue, not an afterthought. "São Paulo is not the city to improvise, but it is a city that rewards a solo woman who travels with judgment." This is not the kind of destination I would describe as easy. It is not relaxed. It is not as charming as some other cities in Brazil. And it is definitely not a place where I would suggest that a solo woman just show up and wander around without a plan. But here is the honest part. Interesting does not always mean comfortable. Some areas can feel crowded and overstimulating. So go early, stay alert, and do not stand on a sidewalk corner looking down at your phone like a lost tourist. São Paulo can be rewarding, but it is not casual travel. You need to pay attention. I would not wear flashy jewelry. I would not hold my phone out while walking. I would not arrive in a brand-new neighborhood after dark. And I would not assume that a place is fine just because it looks busy. Busy does not always mean safe. Another point for women over 50: choose your hotel area carefully. I would spend a bit more to stay in a better-located neighborhood rather than save money and make every outing harder. In a city like São Paulo, convenience is not a luxury. It is part of your safety strategy. São Paulo is better for the traveler who can say, "I do not need this city to entertain me every minute. I just want to experience it intelligently." If you go, keep your schedule realistic, stay alert, and let the city come to you in pieces. You do not need to conquer São Paulo. You just need to experience the right parts of it with confidence. I was able to store my luggage at the airport for the day for about $10. Worth it, so I did not have to carry it all day. The storage is in Terminal 2: Arrivals. Turn left. Luggage storage- "Guarda-Volumes", near parking garage entrance at far end of terminal.    There is an express train from Luz to the city every 2 hours. It's called. "GRU Airport train " Or CPTM. Take the free GRU SHUTTLE BUS. See show notes for many details. Beautiful building. Ornate. The Portuguese Language Center was located there, too, in the Parque de Luz. The police were guarding, but felt safe. Beautiful trees and a fresh smell after the rain. Walked to the Pina Art Museum. Lots of school groups. Modern. Plus some traditional: films, sculptures, paintings, 3d designs. Walked over a bridge to a long street still close to Luz. Walked about a mile. Stores and parking garages mostly. Optical, toys, and industrial products. Found Church of San Bento. Praying for them and others in this beautiful building. No video allowed. —————— My three Sao Paolo  missteps:    Spilling yogurt all over me early in the morning. Table, clothes, floor, everywhere. What a mess. Be more careful around yogurt containers.   In the San Bento metro station, the  Woman at the counter would not give me free subway access. She sent me to the ticket booth to buy a ticket. I showed my passport there, and she said,  Go back there. So I did. I told her my age. Finally, she smiled and let me pass. Maybe I looked too young for the free transport.   Booking a room near the airport that was not in a good neighborhood. Lots of reasons not to walk at night. In the western wing, three figures push forward into the passageways of this transcendent fortress—Uncle Nicky, criminal turned chef; the terrapine Jonathan, inventor and—now—airship captain; and Antistrophe Landrace, the Ruinbringer, though he may not know it yet.  Meanwhile, to the east, an octet walks Eschatonica's twisting paths: Famed adventurer, and Jonathan's sister, Maebela; Veile Lyndelle, destined devotee of the Ennead; Elena Millefiori, Tesserae chanter and eager traveler; the youthful and curious Waylon of Spillaway Peaks; Renegade Hexcloak Caiomhe Wake; and finally, Brontë Adelvys, dissipated sixth scion of the First Line, along with his attendants, Efta and Zolfta Above them, watching with all nine eyes, is Gnova, once the God of Distant Color and Light, now the steward of this Castle.
