Den díkuvzdání je za námi a přinesl nám spoustu fotbalové zábavy. Neuvěřitelných osm televizních přenosů, studio až do noci a samozřejmě parádní bitvy! V druhé polovině základní části už se definitivně začíná oddělovat zrno od plev. Někteří favorité drží svoje pozice, ale někteří naopak selhali. Utkání v Mnichově nám definitivně odzvonilo celou Evropskou šňůru NFL. Teď už se musíme spokojit pouze ze zápasy ve starých dobrých Spojených státech. Favorité jednotlivých konferencí se nám pomalu ale jistě sice profilují, ale pořád je před námi spousta a spousta kvalitního fotbalu. Zápas ve Wembley nám ukončil trojdílnou londýnskou sérii. Nebyla to sice žádná velká přehlídka skvělého fotbalu, ale i tak jsme si to všechno parádně užili. O to zajímavější byl televizní zápas mezi Vikings a Lions, ve kterém tedy poprvé v letošním ročníku tým z Minnesoty našel přemožitele. Posledním mohykánem bez negativního zářezu na pažbě zůstává Kansas City Chiefs. Vydrží mu tahle série? Rozjel se někdo k velkému trháku? Poseł PiS Maciej Wąsik mówi w Poranku Wnet, że w budżecie nie ma pieniędzy na kosztujący 10 mld zł program Tarcza Wschód, który zapowiada premier Donald Tusk. Na razie to deklaracja wyborcza – dodaje. Kandydujący do Parlamentu Europejskiego polityk deklaruje poparcie dla wszystkich działań rządu, zmierzających do realnego wzmocnienia bezpieczeństwa Polski. Jednak, jak zauważa, póki co są to tylko zapowiedzi. „Tak wiarygodne, jak benzyna po 5,19 zł czy kwota wolna 60 tys. zł” – tłumaczy Wąsik. Premier Donald Tusk zapowiedział w sobotę w Krakowie realizację programu „Tarcza Wschód”, który miałby na celu rozbudowę infrastruktury na granicy wschodniej, która pozwoliłaby zatrzymać ewentualną agresję rosyjską i białoruską. „Te 10 mld zł uruchomiliśmy już w tej chwili” – mówił Tusk. Maciej Wąsik komentując słowa premiera zwraca uwagę, że pieniędzy na nowy program nie przewidziano w budżecie państwa. Budżet trzeba by znowelizować, na razie to są deklaracje wyborcze. Dotychczas jedyny konkret z tym związany to „bannerki” Platformy Obywatelskiej – stwierdza polityk Prawa i Sprawiedliwości. Wąsik podkreśla hipokryzję konkurencyjnej partii. Wylicza sprzyjające Rosji posunięcia PO z przeszłości. Brak sprzeciwu rządu PO wobec budowy gazociągu NS2, wypowiedź Sikorskiego o Rosji w NATO, otworzenie małego ruchu granicznego z Kaliningradem, reset, „nasz człowiek w Warszawie” – mówi. Z drugiej strony, jak podnosi Wąsik, „to rząd PiS pierwszy wsparł poważnymi środkami Ukrainę w walce Rosją, to my odparliśmy atak hybrydowy z Białorusi”. Pytany o twierdzenie gen. Simon is a prominent former England rugby union player, current Head of Sustainable Technology Partnerships for NS2, and President of charity, Love of the Game.Simon had an idyllic childhood in Nairobi until his family had to flee from a volatile political election, his family suddenly moved to Gainsborough for a short while before relocating to Madrid. These sudden changes in his youth prepared Simon for an equally storied career. At age 15, Simon started his career in rugby and he maintained a successful career in professional rugby, during which he played for the biggest teams in rugby: Bristol, London Wasps and Toulon. He was part of the winning World Cup team in 2003, winning 71 caps for England between 1996 and 2011 and 2 for the British & Irish Lions. With Nina Müller, Ethical Commerce Alliance Director and host of the Ethical Allies podcast. __ This was a pretty active season in terms of regulatory updates and decisions or guidelines coming out of supervisory bodies: Spain's AEPD issued a decision on the use of Google Analytics by the Royal Academy of Spanish Language (“RAE”), becoming the first EU Data Protection Agency to see the glass half full in the use of the widespread digital data collection service (having been considered high-risk in Denmark, Italy, France, the Netherlands and Austria). It must however be noted that the RAE was only using the most basic version of the tool, without any AdTech integrations or individual user profiling - and in this regard aligned with the CNIL's long-standing guidelines for the valid use of the tool. At EU level, the Artificial Intelligence Act (which we have covered this quarter in a couple of Masters of Privacy interviews) made fast progress with the Council adopting its final position. At the same time, new common rules on cybersecurity became a reality with the approval of the NS2 Directive (or v2 of the Network and Information Security Directive) on November 28th. The updated framework covers incident response, supply chain security and encryption among other things, leaving less wiggle room for Member States to get creative when it comes to “essential sectors” (such as energy, banking, health, or digital infrastructure). Across the Channel, the UK's Data Protection Agency (ICO) issued brand new guidelines on international data transfers, providing a practical tool for businesses to properly carry out Transfer Risk Assessments and making it clear that either such tool or the guidelines provided by the European Data Protection Board will be considered valid. Already into the new year, the European Data Protection Board (EDPB) issued two important reports, on valid consent in the context of cookie banners (in the hope to agree on a common approach in the face of multiple NOYB complaints across the EU) and the use of cloud-based services by the public sector. The former concluded that the vast majority of DPAs (Supervisory Authorities) did not accept hiding the “Reject All” button in a second layer - which most notably leaves Spain's AEPD as the odd one out. They did all agree on the non-conformity of: a) pre-ticked consent checkboxes on second layer; b) a reliance on legitimate interest; c) the use of dark patterns in link design or deceptive button colors/contrast; and d) the inaccurate classification of essential cookies. The latter concluded that public bodies across the EU may find it hard to provide supplementary measures when sending personal data to a US-based cloud (as per Schrems II requirements) in the context of some Software as a Service (SaaS) implementations, suggesting that switching to an EEA-sovereign Cloud Service Provider (CSP) would solve the problem and getting many to wonder whether it also refers to US-owned CSPs, which would leave few options on the table and none able to compete at many levels in terms of features or scale. All of which can easily lead us to the latest update on the EU-US Data Privacy Framework: The EDPB released its non-binding opinion on the status of the EU-US Data Privacy Framework (voicing concerns about proportionality, the data protection review court and bulk data collection by national security agencies). The EU Commission will now proceed to ask EU Member States to approve it with the hope of issuing an adequacy decision by July 2023. This would do away with all the headaches derived from the Schrems II ECJ decision (including growing pressure to store personal data in EU-based data centers), were it not for the general impression that a Schrems III challenge looms in the horizon. In the United States, long-awaited new privacy rules in California (CPRA) and Virginia (CDPA) entered into force on January 1st. Although both provide a set of rights in terms of ensuring individual control over personal data being collected across the Internet (opt-out, access, deletion, correction, portability…), California's creates a private right of action that could pave the way for a new avalanche of privacy-related lawsuits.In any case, only companies meeting a minimum threshold in terms of revenue or the amount of consumers affected by their data collection practices (both of them varying across the two states) will have to comply with the new rules. Lastly, Privacy by Design will become ISO standard 31700 on February 8th, finally introducing an auditable process to conform to the seven principles originally laid out by Anne Cavoukian as Ontario(Canada)'s former Data Protection Commissioner. Enforcement updates It's been interesting to see how continental Data Protection Agencies (“DPAs”) keep milking the cow of the ePrivacy Directive's lack of a one-stop-shop for US or China-based Big Tech giants. The long-awaited ePrivacy Regulation never arrived to keep this framework in sync with the GDPR (which does have a one-stop-shop), and this leaves an opening for any DPA to avoid referring large enforcement cases involving such players to the Irish Data Protection Commissioner (“DPC”) whenever cookie consent is involved. This criterion has been further strengthened by the recent conclusions of EPDB cookie banner task force. Microsoft was the last major victim of this particular gap (following Meta and Google), receiving a 60-million euro fine from France's DPA (CNIL), which shortly after honored TikTok with a 5m euro fine (once again, due to the absence of a “Reject All” button on its first layer - or “not being as easy to reject cookies as it is to accept them”) and, not having had enough, went on to give Apple an 8m euro fine for collecting unique device identifiers of visitors to its App Store without prior consent or notice, in order to serve its own ads (which is akin to a cookie or local storage system when it comes to article 5.3 of the ePrivacy Directive). The CNIL ePrivacy-related enforcement spree did not stop short at Big Tech. Voodoo, a leader in hyper-casual mobile games, was also a target, receiving a 3 million euro fine for lack of proper consent when serving an IDFV (unique identifier “for vendors”, which Apples does allow app publishers to set when IDFA or cross-app identifiers have been declined via the App Tracking Transparency prompt). Putting the ePrivacy Directive aside, and well into pure GDPR domain, Discord received a 800k euro fine (again, at the hands of CNIL) on the basis of: a) a failure to properly determine and enforce a concrete data retention period; b) a failure to consider Privacy by Design requirements in the development of its products; c) accepting very low security levels for user-created passwords; and d) failing to carry out a Data Protection Impact Assessment (given the volume of data it processed and the fact that the tool has become popular among minors). And yet, one particular piece of news outshined mostly everything else in this category: Ireland's DPC imposed a 390 euro fine on Meta following considerable pressure from the EDPB for relying on the contractual legal basis in order to serve personalized advertising - itself the core business model of both social networks. We had a debate on the matter with Tim Walters (English) and Alonso Hurtado (Spanish) on Masters of Privacy, and published an opinion piece on our blog. This last affair is a good segue into Twitter's latest troubles. Its new owner, Elon Musk, not content with having fired key senior executives in charge of EU privacy compliance (including its Chief Privacy Officer and DPO), has suggested that he will oblige its non-paying users to consent to personalized advertising. The Irish DPC (once again, in charge of its supervision under the one-stop-shop rule) asked Twitter for a meeting in the hope to draw a few red lines. Meanwhile, the Spanish AEPD, still breaking all records in terms of monthly fines, sanctioned UPS (70,000 euros) for handing out a MediaMarkt (consumer electronics) delivery to a neighbor, thus breaching confidentiality duties. This will have a serious impact on the regular practices of courier services in the country. Back in the United States, Epic Games and the FTC agreed to a $520m fine for directly targeting children under the age of 13 with its Fortnite game (a default setting that allows them to engage in voice and text communications with strangers has made it worse), as well for using for “dark patterns” in in-game purchases. Separately, in what we believe it is a first case of its kind, even in the EU (with the ECJ FashionID case possibly being the closest we have been to it). Betterhelp has received an FTC $7,8m fine for using the Facebook Lookalike Audiences feature (and alternative offerings in the programmatic advertising space, including those of Criteo, Snapchat or Pinterest) to find potential customers on the basis of their similarity with the online mental health service's current user base. This involved sensitive data and follows repetitive disclaimers by Betterhelp that data would in no case be shared with third parties. On the private lawsuits front (especially important in the US), Meta agreed to pay $725m after a class action was brought in California against Facebook on the back of the ever-present Cambridge Analytica scandal. Also, the Illinois Biometric Information Privacy Act (BIPA) kept putting money into the pockets of claimants and class action lawyers, in this case forcing Whole Foods (an upscale organic food supermarket chain owned by Amazon) to settle for $300.000 - we have previously previous cases against TikTok, Facebook or Snapchat, albeit it was the monitoring, via “voiceprints”, of its own employees (rather than its customers) that triggered this particular lawsuit. Legitimate Interest strikes back To finish with this section, very recent developments justify turning our eyes back to the UK and the EU as there is growing momentum for the acceptance of the legitimate interest as a legal basis for purely commercial or direct marketing purposes: While the CJEU decides on a question posed by a Dutch court in January, in which the DPA issued a fine to a tennis association for relying on legitimate interest to share member details with its sponsors (who then sent commercial offers to them), a UK court (First-Tier Tribunal) has ruled against the ICO (UK DPA) and in favor of Experian (a well-known data broker) for collecting data about 5.3m people from publicly available sources, including the electorate register, to build customer profiles and subsequently selling them to advertisers. Experian has relied on legitimate interest and found it too burdensome to properly inform every single individual (this being the ICO's main point of contention). The decision does appear to indicate that using legitimate interest would not be possible if the original data collection had been based on consent, but even this is not entirely clear. So, just to make it even more clear and simple, the UK Government presented a new draft of a new UK Data Protection Bill on March 8th that includes a pre-built shortcut to using legitimate interest without need for the so-called three part test (purpose, necessity, balancing). Data controllers can now go ahead with this legal basis if they find their purpose in a non-exhaustive list provided - which includes direct marketing. Competition and Digital Markets Google was sued by the Department of Justice for anti-competitive behavior in its dominance of the AdTech stack across the open market (or the ads that are shown across the web and beyond its own “walled gardens”), using its dominance of the publisher ad server market (supply side) to further strengthen its stranglehold of the demand side (advertisers, many of them already glued to its Google Ads or DV360 platforms in order to invest in search keywords or YouTube inventory) and, worse, artificially manipulating its own ad exchange to favor publishers at the expense of advertisers - thereby reinforcing the flywheel, as digital media publishers found themselves with even less incentives to work with competing ad servers. Zero-Party Data and Future of Media (The piece of news below obliges us to combine both categories this season) The BBC has rolled out its own version of SOLID pods to allow its own customers to leverage their own data (exported from Netflix, Spotify, and the BBC) in order to obtain relevant recommendations while staying in full control of such data. Perhaps a little step towards individual agency, but a giant one for a digital media ecosystem mostly butchered by the untenable notice-and-consent approach derived from the current legal framework - which takes us back full circle to Elizabeth Renieris' new book.

According to Don Lemon of CNN hurricane Ian is a new phenomenon. He said this as he scolded Jamie Rhome, acting director of NOAA's National Hurricane Center, for telling him he was wrong about Ian's strength having no relation to climate change. According to the New York Times Ian is the first major hurricane to hit Tampa in over a century, but this is so provably wrong it makes the story totally fictitious. Hurricane wind speed, sustained wind speed, pressure, storm surge, and economic damage, all are weighed, and then the worst condition is selected to create a narrative about the storm. Ian is not hitting Tampa head on, in terms of eye-wall. Joe Biden then warned the public that the best was to prepare for these major storms was to get a ‘shot'. Likewise, when the Nord Stream pipelines were blown up earlier this week the white House told us it was a great opportunity to go ‘green' and the same was said about rising gas prices. But there is more to the story. Just as technology exists to control, intensify, and denitrify hurricanes officially, the pipeline explosions were likely not accidental. Joe Biden and Victoria Nuland both warned that if Russia entered Ukraine the Nord Stream pipelines, particularly NS2, would be dealt with. A few months after Russia ‘invades' we find the US Navy and NATO conducting an unmanned underwater exercise in the Baltic sea near Nord Stream! Another US naval exercise was taking place in the Baltic Sea at the beginning of September. Then the CIA informed Berlin of a coming attack just days before the pipes depressurized from explosions, according to seismographs, and the very day Norway and Poland opened a new gas pipeline to replace Nord Stream 1 and 2. Meanwhile, the largest produce market in the world, Rungis International Market in Paris, has gone up in flames alongside of the dozens of food facilities over the last year that have also burned, exploded, or been hit by planes. Energy and food are under sustained attack. If United States did blow up the pipelines, it is an act of terrorism and war on Russia and Germany, which had control of the project.

According to Don Lemon of CNN hurricane Ian is a new phenomenon. He said this as he scolded Jamie Rhome, acting director of NOAA's National Hurricane Center, for telling him he was wrong about Ian's strength having no relation to climate change. According to the New York Times Ian is the first major hurricane to hit Tampa in over a century, but this is so provably wrong it makes the story totally fictitious. Hurricane wind speed, sustained wind speed, pressure, storm surge, and economic damage, all are weighed, and then the worst condition is selected to create a narrative about the storm. Ian is not hitting Tampa head on, in terms of eye-wall. Joe Biden then warned the public that the best was to prepare for these major storms was to get a ‘shot'. Likewise, when the Nord Stream pipelines were blown up earlier this week the white House told us it was a great opportunity to go ‘green' and the same was said about rising gas prices. But there is more to the story. Just as technology exists to control, intensify, and denitrify hurricanes officially, the pipeline explosions were likely not accidental. Joe Biden and Victoria Nulled both warned that if Russia entered Ukraine the Nord Stream pipelines, particularly NS2, would be dealt with. A few months after Russia ‘invades' we find the US Navy and NATO conducting an unmanned underwater exercise in the Baltic sea near Nord Stream! Then the CIA informed Berlin of a coming attack just days before the pipes depressurized from explosions, according to seismographs, and the very day Norway and Poland opened a new gas pipeline to replace Nord Stream 1 and 2. Meanwhile, the largest produce market in the world, Rungis International Market in Paris, has gone up in flames alongside of the dozens of food facilities over the last year that have also burned, exploded, or been hit by planes. Energy and food are under sustained attack.
Nord Stream Pipeline HEADLINE: Germany and EU Have Been Handed Over a Declaration of War by Pepe Escobar The pipes on both NS and NS2 can be repaired, of course, but hardly before the arrival of General Winter. The question is whether Gazprom – already focused on several hefty Eurasian customers – would bother, especially considering that Gazprom vessels could be exposed to a possible NATO naval attack in the Baltic. As if all of that was not serious enough, Germany is contractually obligated to purchase at least 40 billion cubic meters of Russian gas a year until 2030. QUESTION: Remember when Donald Trump told them NOT to do this? Just say no? They can't: Gazprom is legally entitled to get paid even without shipping gas. That's the spirit of a long-term contract. And it's already happening: because of sanctions, Berlin does not get all the gas it needs but still needs to pay. Mr. Saunders served in the Bush Administration from 2003 to 2005 as Senior Advisor to the Under Secretary of State for Global Affairs. In that capacity, he worked on a broad range of transnational issues, in particular with respect to Russia, Ukraine, and the former Soviet Union, as well as Iraq, China and India. Earlier, Mr. Saunders served as Director of the Center from 1997 to 2003, and was Assistant Director of the Center from its founding in 1994 until 1997. In 2000, he was a Senior Policy Advisor to the Speaker's Advisory Group on Russia, established by the Republican Policy Committee of the U.S. House of Representatives. He has written extensively for major newspapers and journals, and is a frequent commentator in national media, including CNN, Fox, and MSNBC. Abstract: To better understand the genetic diversity, host association and evolution of coronaviruses (CoVs) in China we analyzed a total of 696 rodents encompassing 16 different species sampled from Zhejiang and Yunnan provinces. Based on the reverse transcriptase PCR-based CoV screening CoVs of fecal samples and subsequent sequence analysis of the RdRp gene, we identified CoVs in diverse rodent species, comprising Apodemus agrarius, Apodemus latronum, Bandicota indica, Eothenomys miletus, E. eleusis, Rattus andamanesis, Rattus norvegicus, and R. tanezumi. Apodemus chevrieri was a particularly rich host, harboring 25 rodent CoVs. Genetic and phylogenetic analysis revealed the presence of three groups of CoVs carried by a range of rodents that were closely related to the Lucheng Rn rat coronavirus (LRNV), China Rattus coronavirus HKU24 (ChRCoV_HKU24) and Longquan Rl rat coronavirus (LRLV) identified previously. One newly identified A. chevrieri-associated virus closely related to LRNV lacked an NS2 gene. This virus had a similar genetic organization to AcCoV-JC34, recently discovered in the same rodent species in Yunnan, suggesting that it represents a new viral subtype. Notably, additional variants of LRNV were identified that contained putative nonstructural NS2b genes located downstream of the NS2 gene that were likely derived from the host genome. Recombination events were also identified in the ORF1a gene of Lijiang-71. In sum, these data reveal the substantial genetic diversity and genomic complexity of rodent-borne CoVs, and greatly extend our knowledge of these major wildlife virus reservoirs. Respiratory syncytial virus (RSV) is recognised as the most frequent cause of severe lung infections in infants and cattle worldwide. Currently, no effective treatments are available and the development of a successful vaccine has been hampered by the fact that natural infection does not provide complete and durable protection. RSV nonstructural proteins, NS1 and NS2, are strong inhibitors of IFN α/β-production by specifically preventing interferon regulatory factor (IRF)-3 phosphorylation. However, the exact mechanisms leading to NS protein-mediated inhibition of IRF3 remain to be unravelled. One of the objectives of this study was to identify amino acid domains in the human respiratory syncytial virus (HRSV) nonstructural proteins (NS) responsible for their ability to ablate the IFN-β signalling pathway. Furthermore, I wanted to find out at which level of this signalling pathway the NS proteins exert their suppressive activity and which are their major cellular targets. HRSV strains A2 and Long differ in their ability to block interferon type I synthesis. Sequence analysis of their NS proteins revealed the presence of an amino acid residue in the NS2 protein with a potential role for RSV IFN-inhibitory functions. Two recombinant bovine respiratory syncytial (BRSV) viruses harbouring HRSV NS1 and NS2 genes were generated and tested in their ability to restrict IFN-β synthesis. These recombinant viruses differed only in the identity of the residue at position 26 of the HRSV NS2 protein: rBRSVh1/2 has a Threonine as in the Long strain, while in rBRS h1/2*T26I this amino acid was mutated into an Isoleucin similarly to A2 virus. Sets of in vitro tests revealed that IFN-β induction was impaired by rBRSVh1/2*T26I when compared to rBRSV h1/2. Analysis of the transcriptional factors (AP-1, NF-kB and IRF3) involved in the activation of IFN-β synthesis provided evidence that the inhibitory ability of rBRSVh1/2*T26I was correlated to a selective block of IRF3. The mutation (T26I) in the NS2 protein did neither effect the NF-kB activation pathway nor perturbed the IFN-resistance characteristics of the chimeric viruses. IRF3 is activated upon phosphorylation mediated by IKK-related kinases (TBK1 and IKK-ε). TBK1 was therefore cloned from a human lung cDNA library and its biological activities regarding the induction of IFN-β were compared in mock-infected and infected cells. rBRSVh1/2*T26I and HRSV A2 precluded virus-induced IRF3 activation by interfering with TBK1 functions. No direct interaction between TBK1 and NS2 protein was demonstrated indicating that the kinase TBK1 may not be the sole target involved in RSV mechanisms of evasion of the innate immune response. Recombinant IFN-inducible rabies viruses expressing HRSV Long-derived (rGFP-Ph2/1) or HRSV A2-like (rGFP-Ph2*T26I/1) NS proteins were also generated. The HRSV NS2 protein expressing an Isoleucin (NS2*T26I) at position 26 was not able to suppress IFN-β induction and to rescue the growth of the recombinant eGFP-Ph2*T26I/1 in interferon-competent cells. A low expression of the mutated NS2*T26I protein was probably the reason of this failure. In summary, these results show that the HRSV NS2 protein possesses an intrinsic IFN-β inhibitory activity, which is achieved throughout a selective inhibition of the IRF3 activation pathway. The block appears to be exerted at the level of IRF3-kinase TBK1. Interferonantagonist functions of the HRSV NS2 protein are linked to a particular amino acid motif in the N-terminus of the protein. Identification of this amino acid domain and of TBK1 as the cellular target provide a better insight of how the HRSV NS2 protein prevents the establishment of the antiviral innate immune response and therefore it might contribute to the development of an effective vaccine.

HRSV ist eine häufige und weltweit verbreitete Ursache von Infektionen des Respirationstraktes. Es führt zu einer entzündlichen Erkrankung der respiratorischen Schleimhäute mit Mukosaödem, Hypersekretion und Bronchospasmus. Die Übertragung des viralen Erregers erfolgt durch Tröpfcheninfektion oder Kontakt mit kontaminierten Gegenständen. HRSV-Infektionen zeigen die höchste Inzidenz bei Säuglingen, vor allem in den ersten zwei bis sechs Lebensmonaten. Bei 25% bis 40% dieser Säuglinge nimmt die Erkrankung einen schweren Verlauf mit Befall des unteren Respirationstraktes in Form einer HRSV-Bronchiolitis oder -Pneumonie. Bei 0,5% bis 2,0% ist eine stationäre Behandlung im Krankenhaus erforderlich. Die Inzidenz nimmt wegen des zunehmend effektiveren Immunsystems mit dem Alter ab. Erwachsene und ältere Kinder zeigen meist keine Symptome bzw. Symptome einer leichten Erkältung. Reinfektionen im Laufe des Lebens sind häufig. Eine effektive kausale Therapie bei HRSV-Infektionen steht derzeit nicht zur Verfügung. Bei Patienten mit leichtem Krankheitsverlauf ist keine spezielle Behandlung erforderlich, therapiert wird symptomatisch. Aktuell ist keine spezifische Prävention in Form einer aktiven Impfung oder als effektive antivirale Therapie etabliert. Angesichts der hohen Inzidenz von HRSV-Infektionen und -Reinfektionen sowie der enormen gesundheitlichen und wirtschaftlichen Auswirkungen ist ein effektiver Impfstoff gegen HRSV als Forschungsziel vorrangig. Das Genom von HRSV, das zur Ordnung der Mononegavirales gehört, besteht aus einem negativ-orientierten RNA-Einzelstrang mit einer Länge von 15 222 Nukleotiden (beim A2-Stamm) und kodiert für zehn subgenomische mRNAs in der Reihenfolge 3’-leader, NS1, NS2, N, P, M, SH, G, F, M2(1+2), L, trailer-5’, die zur Expression von elf viralen Proteinen führen: fünf RNP-assoziierte Proteine, das sind das Nukleoprotein N, das Phosphoprotein P, die große katalytische Untereinheit L der RNA-Polymerase und der Transkriptionselongationsfaktor M2-1 sowie das nicht essentielle M2-2-Protein; vier Hüllproteine, dazu zählen das nicht-glykosylierte Matrixprotein M und drei Oberflächenproteine, im Einzelnen das Fusionsprotein F, das Anheftungsprotein G und das kleine hydrophobe Protein SH; zwei Nicht-Strukturproteine NS1 und NS2. NS1 und NS2 zeichnen die Pneumoviren vor allen anderen Viren der Ordnung der Mononegavirales aus. Beide NS-Proteine sind im Virion nur in Spuren nachweisbar, während sie in infizierten Zellen akkumulieren. Die beiden für die Proteine NS1 und NS2 kodierenden, nichtüberlappenden Gene liegen am 3‘-Ende des Genoms direkt im Anschluss an die leader-Region. NS1 und NS2 stimmen in den vier carboxyterminalen Aminosäuren überein, ansonsten weisen sie keine Sequenzähnlichkeiten auf. Das NS1-Gen hat eine Länge von 552 nt und kodiert für ein leicht saures Protein von 139 AS und 15,7 kD. Das NS2-Gen ist 503 nt lang und kodiert für ein basisches Protein von 124 AS und 14,7 kD. Die für die Ordnung der Mononegavirales charakteristische progressive Attenuation der Transkription sowie die Genlokalisation von NS1 und NS2 am 3‘-Ende lassen auf die höchste Transkriptionsrate für NS1- und NS2-mRNA unter den zehn HSRV-mRNA schließen, was auf eine bedeutende Rolle der NS1- und NS2-Proteine in infizierten Zellen hindeutet. NS1 und NS2 antagonisieren im Zusammenwirken die durch alpha-IFN und beta-IFN induzierte antivirale Antwort des Wirtsorganismus. Hierfür ist eine Koexpression beider NS-Proteine unbedingt erforderlich, ein NS-Protein allein zeigt keine derartige Aktivität. Der Mechanismus, mit dem HRSV die IFN-Antwort des Wirtsorganismus umgeht, ist unklar. In dieser Arbeit wurde die Funktion der NS-Proteine von klinischen HRSV-Isolaten aus fünf bis fünfzehn Monate alten Kindern untersucht. Durch die Anzucht der klinischen HRSV-Isolate in HEp-2-Zellkultur unter identischen Bedingungen wurden zunächst patientenabhängige Faktoren ausgeschaltet und damit die Grundlage für die Vergleichbarkeit der Wachstumseigenschaften der Isolate geschaffen. In den daraufhin erstellten Wachstumskurven konnten deutlich voneinander abweichende Wachstumverhalten der Isolate aufgezeigt werden. Der Befund, dass 3/4 der Bronchiolitis hervorrufenden HRS-Viren hohe infektiöse Titer (>106 infektiöse Viruspartikel/ ml an Tag 3) erreichten, während dies nur bei 1/3 der Bronchitis verursachenden Viren zu beobachten war, könnte auf eine Korrelation zwischen Wachstum in vitro und Pathogenität in vivo hindeuten. Um dies zu belegen, müsste eine größere Zahl von klinischen Isolaten analysiert werden. Die beiden Nicht-Strukturproteine versetzen HRSV in die Lage, die antivirale IFN-Antwort der Wirtszelle zu umgehen. Durch Behandlung von Virus-infizierten Zellkulturen mit IFN ließ sich nachweisen, dass alle klinischen HRSV-Isolate die Eigenschaft der IFN-Resistenz gleichermaßen besitzen und erst durch unphysiologisch hohe IFN Dosen eine wesentliche Inhibierung der Virusreplikation erreicht werden kann. Die in gleicher Weise ausgeprägte α-IFN-Resistenz bei den in Virulenz und Wachstumsgeschwindigkeit unterschiedlichen Viren deutete bereits darauf hin, dass diese Resistenz essentiell für alle klinischen RSV-Isolate ist, und dass zusätzliche Faktoren für das Maß der Aggressivität der Erreger verantwortlich sind. Mittels Nukleotid- und Aminosäuresequenzanalysen von NS1 und NS2 konnte dies weitgehend bestätigt werden. Anhand von RNA aus den HRSV-Isolaten wurde mit Hilfe des Enzyms Reverse Transkriptase cDNA von NS1 und NS2 synthetisiert, die nach dem Prinzip der PCR in vitro amplifiziert wurde. In anschließenden Klonierungsarbeiten wurden aus dem Vektor pBluescript II SK (–) und NS1-DNA bzw. NS1+NS2-DNA als Insert Plasmide konstruiert, in denen die Gensequenzen von NS1 und NS2 ermittelt und rechnergestützt in die entsprechenden Aminosäuresequenzen translatiert wurden. Die Analyse der NS-Sequenzen zeigte eine überraschend hohe Konservierung. Die Isolate waren einschließlich des Long-Stamms diesbezüglich untereinander sehr ähnlich. Diese Beobachtung stimmt mit der IFN-Resistenz überein und zeigt die Bedeutung der NS-Proteine. Die Ergebnisse dieser Arbeit deuten darauf hin, dass Abweichungen in den Sequenzen der übrigen Gene sowie patientenbezogene Faktoren wie Abwehrlage und anatomische Beschaffenheit des Respirationstraktes als Grund für die Unterschiede im Schweregrad der HRSV-Infektion eine Rolle spielen. Angesichts der stabilen Koexpression beider Nicht-Strukturproteine und des dadurch bedingten effektiven IFN-Escape sichern die Gene NS1 und NS2 die Überlebensfähigkeit von HRSV in vivo und stellen ebenso geeignete wie interessante Angriffspunkte in der Entwicklung eines attenuierten Lebendimpfstoffs dar.

Among the Paramyxoviridae, only members of the subfamily Pneumovirinae like Respiratory Syncytial Virus (RSV) encode two nonstructural proteins, NS1 and NS2. These two proteins cooperatively mediate type I interferon resistance and prevent induction of interferon in infected cells. Interactions of NS1 and NS2 proteins in every combination were shown by using the yeast-two-hybrid system. Therfore, NS1 and NS2 are able to form homo- and hetero(oligo)mers in infected cells. Although RSV replicates exclusively in the cytoplasm, NS-Proteins are localized in the cytoplasm as well as in the nucleus. Expression of an enlarged NS1 fusion protein, EGFP-NS1-BRSV N, resulted in the same nuclear and cytosolic localisation indicating that nuclear localisation is not due to diffusion but rather to an active transport. Thus, NS-Proteins should have particular functions in the nucleus of infected cells. Furthermore, yeast-two-hybrid screening of a lung cDNA expression library using NS1 of bovine RSV (BRSV) as a bait, identified cDNA clones encoding several nuclear proteins and one cytosolic protein. BRSV NS2 protein and NS-Proteins from other pneumoviruses (HRSV, PVM) were also able to interact with the identified cellular proteins in yeast. The isolated cDNAs encode the nuclear proteins CDK4BP (p34SEI-1 or TRIP-Br1), RanBP16, MM-1, DEAD Box Helicase p68 and the cytosolic β-COPI. Specific interactions were determined by mutational analysis of BRSV NS1 in yeast. Co-immunoprecipitation from lysates of eukaryotic cells confirmed the interaction of both BRSV NS-Proteins with the cellular proteins. The interaction of MM-1 and p68 with both NS-Proteins was also shown in GST pull down assay in vitro. Engineered BRSV encoding a truncated p68 showed accelerated replication in MDBK and Vero cells, whereas growth of NS1/NS2 deletion mutants expressing the truncated p68 was unaffected. This indicates that the presence of NS-Proteins is a prerequisite for the acceleration of BRSV growth by truncated p68. Furthermore, replication of BRSV was attenuated on HeLa cells in which expression of p68 was knocked down by specific siRNA, whereas replication of the unrelated Rabies virus was not. Thus, p68 is a nuclear target protein for the NS-Proteins and supports BRSV replication in vitro. Growth and division of host cells is necessary for optimal BRSV replication and like p68, most of the identified nuclear protein interactors are related to regulation of the cell cycle and cell division, respectively. Therefore, NS-Proteins appear to influence the cell cycle for optimal replication of BRSV by targeting such proteins. Hence, with the yeast-two-hybrid system, the first cellular interaction partners were identified indicating new functions of NS-Proteins in the viral replication cycle.

Bovine respiratory syncytial virus (BRSV), family Paramyxoviridae, subfamily Pneumovirinae, is a major etiological agent of respiratory tract disease in calves. A peculiarity of pneumoviruses is the presence of two unique nonstructural protein genes, NS1 and NS2. We show that both BRSV NS proteins are necessary and sufficient to mediate IFN resistance. This function can also be transferred to another, unrelated virus, Rabies virus. The mechanism of IFN resistance is conserved among the Pneumovirinae since the NS proteins of human RSV (HRSV) and of Pneumoniavirus of Mice (PVM) are also able to mediate IFN resistance in a similar manner. Furthermore, both BRSV NS proteins are able to block the induction of IFN by inhibiting the activation of interferon regulatory factor 3 (IRF-3). As a result, BRSV is able to block the establishment of a cellular antiviral state. The characterization of interactions between virus (NS proteins) and the host cell (IFN system) will have important implications in the development of HRSV/BRSV vaccines
