I denne episoden snakker vi om retur til idrett etter korsbåndsskade. Korsbåndsskader er den hyppigste av de akutte kneskadene, og er en stor byrde for utøveren som rammes. Man vet at mange pasienter som pådrar seg en korsbåndsskade aldri returnerer til idretten de kom fra, og av de som gjør det så vil en andel av pasientene dessverre ryke korsbåndet på nytt. Korsbåndsskade, i likhet med andre traumatiske kneskader, medfører en betydelig økt sjanse for utvikling av symptomatisk artrose senere i livet. Desto større kneskaden er, desto større er risikoen for artrose. Følgelig er behovet for god forståelse rundt re-skade og retur til idrett stor hos våre lyttere og i denne episoden snakker vi om return to play-begrepet, hoppe- og styrketester, mentale aspekter ved retur til idrett inkl. self-effiacy, selvtillit og begrepet “psykologisk beredskap”. Hvor mange returnerer til idrett etter korsbåndsskade? Hvor mange pådrar seg en ny skade innenfor to år? Hvordan ser testbatteriene ut? Hvilken rolle har du som kliniker i møtet med disse pasientene? Abstract: The cellular complexity of the endochondral bone underlies its essential and pleiotropic roles during organismal life. While the adult bone has received significant attention, we still lack a deep understanding of the perinatal bone cellulome. Here, we have profiled the full composition of the murine endochondral bone at the single-cell level during the transition from fetal to newborn life and in comparison to the adult organ, with particular emphasis on the mesenchymal compartment. The perinatal bone contains different fibroblastic clusters with blastema-like characteristics in organizing and supporting skeletogenesis, angiogenesis, and hematopoiesis. Our data also points out to a dynamic inter- and intra-compartment interactions as well as a bone marrow milieu prone to anti-inflammation, which we hypothesize is necessary to ensure the proper program of lymphopoiesis and the establishment of central and peripheral tolerance in early life. Our study provides a integrative roadmap for the future design of genetic and cellular functional assays to validate cellular interactions and lineage relationships within the perinatal bone. Several industry heavyweights in Germany have recently made some interesting comments on switching to full electrification. Markus Heyn, the CEO of Bosch, warned that a switch to full electric would result in a shortage of batteries. Talking to a German news outlet Mr Heyn said the ramifications of a mass adoption of a single energy source are currently being played out in Europe – “we're seeing the consequences of the gas shortage because we haven't prepared enough alternatives.” His comments echoed by BMW's Chief Executive, Oliver Zipse, who said Europe isn't ready for a hard cut-off of petrol and diesel engines suggesting “it's wrong to switch off the combustion engine in Europe.” The comments echo those of Toyota and Ford who suggest motorists will still need a choice of what powers vehicles because not everybody is rolling out electric tech at the same speed. Yes, hydrogen is still in the running. The Fed indeed raised interest rates by another .75% yesterday marking the 5th rate hike this year and there is no sign of the increases slowing down. Fed Chair Jerome Powell announced the rate hike while adding that rates hikes will continue until inflation is reigned back in as they forecasted another 1.25% points in hikes throughout the end of the year noting that a ‘soft landing' for the economy was not likelyTo review the first hike of .25% was on March 17th of this year when the base rate was zero. From there we saw hikes in May, June, July, and yesterday bringing the rate to its current 3.25The rate was initially brought to zero in 2008 in an unprecedented move to stabilize the economy and stayed there for over 7 years before slight increases beganAn entire neighborhood of homes equipped with Tesla solar roofs and powerwalls has been steadily moving forward and is kicking into high gear. Her family ventured to several states during her youth, including: Nevada, Maryland, Arizona, and Colorado. The youngest of three kids, her flare for drama surfaced by age three when she told her parents she wanted to be an actress and refused to appear in public without her pink tutu.Growing up she was fascinated and inspired by classic films and their actors, especially MGM musicals. She was probably the only twelve year old who scouted the TV Guide every Sunday to see which movies from AMC she wanted to record and delight in throughout the week. She even asked for TCM for Christmas because she felt that they had a better choice of Judy Garland, Betty Grable, and Doris Day films. These movies motivated her to get serious about tap dancing and singing, and most of her after school hours were dedicated to lessons and practicing. Developing her talents turned her into a bonafide "Theatre Geek," and she graced her high school stage with pride and excitement!After high-school she left her family in Colorado and moved to Southern California to pursue her life-long love of acting. She attended University of CA, Irvine where she studied acting and musical theatre. It was there that she began doing improv and sketch comedy. Within 3 years she graduated and headed up to LA to start a TV/Film career.Once in LA, she began to perform at the Upright Citizens Brigade Theatre (UCB), mainly in their musical shows, specifically Matt Besser's show "The Freak Dance." In this episode of the Epigenetics Podcast, we caught up with Manel Esteller, Director of the Josep Carreras Leukemia Research Institute to talk about his work on Epigenetics and Epitranscriptomics in Cancer. The focus of Manel Esteller's research career and the focus of his current team is to characterize the epigenome and epitranscriptome of cancer cells in comparison to healthy cells, and their interplay. Ultimately, their goal is to use this knowledge to develop new therapies for cancer. Key achievements from the Esteller lab began with the discovery of the first miRNA that undergoes specific cancer-methylation associated silencing. The team further identified many more miRNAs that also play a role in cancer. It's the 4th since her first album, which she produced in 2016 when she was a drama student at Stellenbosch University. Emma grew up around music. Her father was an opera singer, and her grandparents were quite well-known musicians back in the day. She started playing the piano at the age of four and composed her first song at the age of 6. By 13, she was also writing the words to her music. Bosch y Microsoft han desarrollado una plataforma de software que conecta perfectamente los coches con la nube. El objetivo de esta colaboración es simplificar y acelerar el desarrollo y la implementación del software del vehículo durante toda la vida útil de un automóvil, de acuerdo con los estándares de calidad automotriz. La nueva plataforma, que estará basada en Microsoft Azure e incorporará módulos de software de Bosch, permitirá desarrollar y descargar software en las unidades de control y en los ordenadores del vehículo. Aufbauend auf bereits abgeschlossenen Ergebnissen verschiedener Forschungs- arbeiten des Lehrstuhls für Tierschutz, Verhaltenskunde, Tierhygiene und Tierhaltung der Tierärztlichen Fakultät der Ludwig-Maximilians-Universität München und dem Lehr-, Versuchs- und Fachzentrum für Geflügelhaltung Kitzingen (LVFZ) bezüglich dieser Thematik, war es Ziel dieser Studie den Einsatz der „modifizierten Rundtränke nach Heyn und Erhard“, die nun kommerziell unter dem Namen „AquaDuc T“ über die Firma Big Dutchman International GmbH (Vechta, Deutschland) vertrieben wird, in der Mastphase auf Praxistauglichkeit zu untersuchen. In drei Pekingentenmastbetrieben mit 7.140 bis 13.515 Mastplätzen, wurden von Februar 2008 bis Juli 2009, jeweils fünf bis acht sich abwechselnde Kontroll- (ausschließlich betriebsübliche Nippeltränken) und Versuchsdurchgänge (zusätzliches Angebot von Rundtränken) mit je einem Besuch zu Mastanfang (28.-32. LT) und Mastende (35.-39. LT) durchgeführt (Tierzahl gesamt n= 429.137 Enten). In allen Betrieben wurden Cherry Valley Pekingenten (Firma Wichmann Geflügelproduktionsgesellschaft mbH, Wachenroth, Deutschland) für die Dauer von 37 bis 47 Tagen in Bodenhaltung auf Stroheinstreu in Fensterställen gemästet. Jeweils auf der Gefälleseite der Mastställe wurde ein Rundtränkensystem installiert, zu dem die Enten in den Versuchsdurchgängen ab dem 25. Lebenstag zusätzlich zu dem Nippeltränkensystem für sechs Stunden täglich Zugang hatten. Die dabei eingesetzten Rundtränken „AquaDuc T“ der Firma Big Dutchman International GmbH (Vechta, Deutschland) hatten einen Durchmesser von 45,3 cm (Trogseitenlänge 142 cm). Eine in der Aufhängung integrierte Feder regulierte über das Gewicht der Tränke den Wasserzulauf und damit den Wasserstand auf 8 cm bis 10 cm. Das Verhalten der Enten im Tränkebereich wurde bei jedem Besuch 24-40 Stunden aufgezeichnet und über 6.300 Stunden Videomaterial mittels Scan Sampling und Instantaneous Sampling (MARTIN und BATESON, 1994) ausgewertet. Um wasserassoziierte Parameter der Tiergesundheit beurteilen zu können, wurden bei jedem Besuch 100 Tiere (n= 8.300 Enten) auf ihre Gefiederqualität, den Verschmutzungsgrad ihres Gefieders, Nasenlochverstopfungen, Durchgängigkeit der Nasenhöhlen und Augenentzündungen untersucht. Parallel zu dieser Arbeit lief ein Dissertationsvorhaben von Nicola Hirsch mit den Schwerpunkten Tierhygiene und -gesundheit (HIRSCH, 2011). Die Mastkennzahlen und wirtschaftlichen Aspekte wurden von den Mitarbeitern des Lehr-, Versuchs- und Fachzentrums für Geflügelhaltung Kitzingen (LVFZ) ermittelt. Alle gewonnenen Ergebnisse zeigen, dass Pekingenten die modifizierten Rundtränken „AquaDuc T“ gegenüber den Nippeltränken eindeutig bevorzugten. Sie ermöglichen den Tieren ein Eintauchen des Kopfes, arttypisches Trinken und Seihen, Gefiederpflege mit Wasser und ein Reinigen von Schnabel und Augen. In insgesamt fünf Versuchsdurchgängen, die alle in den Stallungen der Landesanstalt für Landwirtschaft in Kitzingen stattfanden, wurden Pekingenten in Wahlversuchen Duschen (ab dem 22. Lebenstag) oder modifizierte Rundtränken nach Heyn und Erhard (ab dem 24.–25. Lebenstag) für vier beziehungsweise sechs Stunden täglich zusätzlich zu den in Mastbetrieben üblichen Nippeltränken angeboten. Zielsetzung dieser Studie war es, eine zugleich tierfreundliche und unter wirtschaftlichen Gesichtspunkten realisierbare Alternative in der Wasserversorgung zu finden. Der Stall, in den die Enten als Eintagsküken eingestallt wurden, war in sechs Abteile von 32 m² Größe unterteilt. Jedes Abteil umfasste einen eingestreuten Bereich mit Futterschalen sowie auf der linken und rechten Stallseite je einen 4 m² großen, erhöhten Tränkebereich mit perforiertem Boden. In drei von fünf Versuchsdurchgängen erhielten die rund 200 Tiere pro Abteil Zugang zu einem 11,4 m² großen, überdachten Auslauf mit perforiertem Boden, in dem zeitweise Tränken installiert waren und der über eine Klappe auf der linken Stallseite zugänglich war. Um die Tierfreundlichkeit der verschiedenen Systeme zu überprüfen, wurde das Tierverhalten am Mastanfang (27.–31. Lebenstag) und -ende (41.–46. Lebenstag) anhand von 24-Stunden-Videoaufnahmen der Tränkebereiche und Ausläufe sowie einer 20-minütigen Direktbeobachtung, die das gesamte Abteil umfasste, mittels Scan Sampling und Instantaneous Sampling (MARTIN und BATESON, 1993) untersucht. Besonderes Augenmerk wurde dabei auf die mit dem Wasser assoziierten Verhaltensweisen „Trinken“, „Putzen mit Tränkewasser“ und „Badeverhalten“ sowie auf die „Badeersatzhandlungen“ gerichtet. Um die Tiergesundheit beurteilen zu können, wurden zu Mastanfang und -ende 15 Tiere aus jedem der sechs Abteile auf Gefiederverschmutzung, Verletzungen und Vorhandensein von ein- oder beidseitigen Nasenlochverstopfungen und Augenentzündungen untersucht sowie am Ende der Mast zusätzlich auf ihre Gefiederqualität. Von 15 Tieren jedes Abteils wurden außerdem zu beiden Terminen Blutproben gewonnen und auf ihren Hämatokrit-, Hämoglobin- und Corticosterongehalt untersucht. Die Wirtschaftsdaten zu Wasserverbrauch und Mastergebnissen ermittelten die Mitarbeiter in Kitzingen. Die Ergebnisse aus der Verhaltensbeobachtung lassen den Schluss zu, dass die Enten Rundtränken eindeutig gegenüber Nippeltränken bevorzugen. Während der vier- beziehungsweise sechsstündigen Zugangszeit zu den Rundtränken konnte an diesen stets eine deutlich höhere Tränkeaktivität („Trinken“ plus „Putzen mit Tränkewasser“) festgestellt werden als an den Nippeltränken in den gegenüberliegenden Tränkebereichen. Der prozentuale Anteil der Tiere im Tränkebereich, die an den Rundtränken mit Trinken oder Putzen mit Wasser beschäftigt waren, nahm zum Mastende hin zu, wohingegen die Tränkebereiche mit Nippeltränken vorwiegend als Ruheräume genutzt wurden. Die Enten suchten die Rundtränken gezielt auf, um zu trinken und ihr Gefieder mit Wasser zu reinigen. Anders verhielt es sich in den Tränkebereichen mit Dusche. Hier konnten während der Betriebszeit der Dusche zum einen insgesamt deutlich weniger Tiere beobachtet werden als in Tränkebereichen mit Rundtränken, und zum anderen lag die Tränkeaktivität an den Duschen, die stellenweise gegen Null ging, deutlich niedriger als an den Nippeltränken im Tränkebereich gegenüber der Dusche. Natürliches Badeverhalten konnte an keiner der drei Tränkeformen festgestellt werden. An den Rundtränken war den Tieren aber zumindest ein partielles Eintauchen des Kopfes möglich. Die Ausläufe wurden von den Enten sowohl mit als auch ohne dort installierte Tränken gern genutzt, wobei sie in letzterem Fall vorwiegend als Ruheräume dienten. Bei der Gefiederqualität schnitten die Enten aus Abteilen mit Rundtränken signifikant besser ab als solche aus Abteilen mit ausschließlich Nippeltränken und meist auch als solche aus Abteilen mit Dusche. Eine schlechte Gefiederqualität wurde allerdings bei keinem Tier festgestellt. Auch bei der Gefiederverschmutzung und der Verstopfung der Nasenlöcher erzielten die Tiere mit Rundtränken stets die besten Ergebnisse, gefolgt von jenen mit Dusche und Nippeltränken. Eine Reduktion der Tränkezahl von ursprünglich 3 Rundtränken für 4 Stunden auf 2 für 4 beziehungsweise 6 Stunden hatte keine nennenswerte Verschlechterung der Tiergesundheit zur Folge. Enten aus Abteilen mit 2 Rundtränken für 6 Stunden schnitten dabei genauso gut ab wie solche aus Abteilen mit 3 Rundtränken für 4 Stunden. Die Blutparameter Hämatokrit, Hämoglobin und Corticosteron sowie die Wirtschaftsparameter Schlachtgewicht, Futterverbrauch und -verwertung, Tierverluste und Europäischer Effizienzfaktor wurden von der angebotenen Tränkeform nicht signifikant beeinflusst. Der Wasserverbrauch in Abteilen mit Dusche lag deutlich über dem in Abteilen mit Rund- oder Nippeltränken und stand in keinem Verhältnis zur Nutzung durch die Enten. Demgegenüber waren die Unterschiede im Wasserverbrauch zwischen Abteilen mit ausschließlich Nippeltränken und solchen mit zeitlich begrenztem Zugang zu Rundtränken gering. Das Angebot der in dieser Studie eingesetzten Duschen kann aufgrund dieser Ergebnisse somit weder aus ethologischer noch aus wirtschaftlicher Sicht empfohlen werden. Durch den Einsatz modifizierter Rundtränken, die den Tieren arttypisches Trinken, Seihen, Schnabelwaschen und Putzen mit Tränkewasser ermöglichen, kann die Haltungsumwelt der Pekingenten dagegen attraktiver und tierfreundlicher gestaltet werden, insbesondere wenn diese in einem überdachten Auslauf angeboten werden. Für eine Tierzahl von rund 200 Tieren hat sich auch unter wirtschaftlichen Gesichtspunkten das Angebot von 2 Rundtränken mit einer Zugangszeit für 6 Stunden bewährt.

Um den möglichen Einsatz und die Vorteile von offenen Tränkeformen in der Entenmast besser beurteilen zu können, wurden im Zeitraum von August 2003 bis Dezember 2004 fünf Versuchs-Mastdurchgänge mit Pekingenten (Anas platyrhynchos f. domestica) in den Stallungen des Lehr- und Versuchsguts der Bayerischen Landesanstalt für Landwirtschaft (LfL) in Kitzingen (Bayern) durchgeführt. Dabei wurden in den beiden ersten Mastdurchgängen von jeweils 47 Tagen 900 bzw. 1044 Tiere in zwölf standardisierte Stallabteilen aufgeteilt. Die Abteile waren jeweils mit einer von vier möglichen Tränkevarianten (Spark-Cups, Rundtränke, Rinnentränke, Nippeltränke in je drei Wiederholungen) ausgestattet. In Versuchsdurchgang III wurden insgesamt 1152 Tiere, aufgeteilt in sechs Abteile mit je zwei Tränkeformen pro Abteil, zur Auswahl eingestallt. Die Tiere hatten die Wahlmöglichkeit zwischen einer Nippel- und einer Rinnentränke oder einer Nippeltränke und einer für Enten entwickelten, modifizierten Rundtränke (modifiziert nach Heyn und Erhard). Der Kontrollgruppe standen nur Nippeltränken zur Verfügung. Die Tränke-Kombinationen wiederholten sich bei sechs Abteilen je zweimal. In den Versuchsdurchgängen IV und V wurden, bei unverändertem Stall-Aufbau, zeitlich begrenzt zugängliche modifizierte Rundtränken (zwei, vier und acht Stunden täglich ) gegenüber Nippeltränken zur Auswahl gestellt. Jeweils zu Beginn (um Tag 21) und Ende (um Tag 47) der Mast wurden Untersuchungen zu Tierverhalten und Tiergesundheit durchgeführt mit dem Ziel, mögliche Einflüsse der verschiedenen Tränkeformen auf die erhobenen Parameter zu ermitteln. Dazu wurden 24-Stunden-Video-Beobachtungen der Tränkebereiche sowie Direktbeobachtungen der Gesamtabteile vorgenommen. Bei 240 Tieren pro Mastdurchgang (insgesamt 1200) wurden die Blutparameter Hämatokrit und Hämoglobin gemessen, verstopfte Nasenöffnungen erfasst, sowie die Gefiederqualität und –verschmutzung, unterteilt nach verschiedenen Körperregionen, evaluiert.
