Aakash Nambiar is a barefoot runner from the beautiful state of Kerala. Aakash is super passionate about traveling and running. He combines the two wherever he goes. He carries his Insta 360 camera, and documents his experiences on instagram, where he can be found by the name ‘barefootmallu'. Since the pandemic, he has travelled to and worked from numerous places including Kashmir, Manali, Meghalaya, Nagaland, Bangalore, Spain, Portugal and a few more. And wherever he has been, he has done a few long runs. For examples, he ran from Abu dhabi to Dubai, Madrid to Toledo in Spain Peniche to Lisbon in Portugal And he has done most of the LLRs that I've done. For those who don't know, LLR stands for Last long run of the year. It's a tradition we've followed for many years. We end the year running from one beautiful city to another and celebrate the year at a beautiful destination but not before we've done two back to back ultras distance runs. More on LLRs in a future episode perhaps. This conversation though is a catchup with Aakash as he just came back from Kashmir, where he did a bunch of runs with the Indian army. Dr. Anand Padmanabhan is a pathologist, transfusion medicine physician, and Associate Professor of Laboratory Medicine and Pathology at the Mayo Clinic. Anand studies blood clotting, also known as “thrombosis”. He is working on a particular type of blood clotting that can occur when our bodies produce antibodies for proteins or other molecules inside or on the surface of blood platelets. Anand is interested both in finding better ways to diagnose and to treat patients with these potentially deadly diseases. When he's not working, Anand loves spending time with his wife, their 15-year-old son, and their two dogs. They enjoy watching movies and traveling together across the U.S. and around the world. He received his Bachelor of Medicine, Bachelor of Surgery (M.B.B.S.) degree from Thanjavur Medical College and completed a Clinical Internship in Medicine and Surgery at LLR and Associated Hospitals in India. He was awarded his PhD in biochemistry from Brown University. Afterwards, Anand completed his residency in clinical pathology at Columbia University and New York Presbyterian Hospital and transitioned into a Fellowship in transfusion medicine at the University of Pittsburgh. Anand worked at the Blood Center of Wisconsin and the Medical College of Wisconsin from 2010 - 2019, and he joined the Mayo Clinic in 2020. Anand has received various awards and honors over the course of his career, including the JN Tata endowment for the higher education of Indians, National Blood Foundation Award for Innovative Research, ASFA Lecturer award, and Choosing Wisely Champion Award from the American Society for Apheresis, and he was recently inducted into the National Blood Foundation Hall of Fame. Scientists said this week that they used a high-powered laser to change the direction of lightning strikes for the first time. The laser was aimed at the sky atop Mount Santis in northeastern Switzerland. Lightning is a powerful electrical discharge between a cloud and the ground, within a cloud or between clouds. It kills thousands of people and causes billions of dollars of damage to communication systems, power lines, and equipment each year. American inventor Benjamin Franklin first invented the lightning rod in the 1750s. The laser redirected all four lightning strikes that happened during the laser's operation. "We demonstrate for the first time that a laser can be used to guide natural lightning," said physicist Aurelien Houard of Ecole Polytechnique's Laboratory of Applied Optics in France. He is the coordinator of the Laser Lightning Rod project and lead writer of the research published in the journal Nature Photonics. Abstract: Voluntary movements are prepared before they are executed. Preparatory activity has been observed across the CNS and recently documented in first order neurons of the human PNS i.e., in muscle spindles. The changes seen in these sensory organs suggest that the independent modulation of stretch reflex gains may represent an important component of movement preparation. The aim of the current study was to further investigate the preparatory modulation of short- and long-latency stretch reflex responses ('SLR' and 'LLR') of the dominant upper limb. Specifically, we investigated how different target parameters (target distance and direction) affect the preparatory tuning of stretch reflex gains in the context of goal-directed reaching, and whether any such tuning depends on preparation duration and the direction of background loads. We found that target distance produced only small variations in reflex gains. In contrast, both SLR and LLR gains were strongly modulated as a function of target direction, in a manner that facilitated the upcoming voluntary movement. This goal-directed tuning of SLR and LLR gains was present or enhanced when the preparatory delay was sufficiently long ( greater than 250 ms) and the homonymous muscle was unloaded i.e., when a background load was first applied in the direction of homonymous muscle action (assistive loading). The results extend further support for a relatively slow-evolving process in reach preparation that functions to modulate reflexive muscle stiffness, likely via the independent control of fusimotor neurons. Such control can augment voluntary goal-directed movement and is triggered or enhanced when the homonymous muscle is unloaded. Copy rights belong to original authors. LLR's podcast hiatus during every September is due to their involvement with Hell's Half Mile Film & Music Festival. Stay tuned! ___ HHM weekend's peak of music events for our 16th annual festival took place on Saturday, September 24th at the Dunlop Building (located at 517 Washington Ave., Bay City, MI), and per usual brought 3 amazing bands! This year's lineup featured Party Nails (Los Angeles), Ratboys (Chicago), and Sugar Tradition (Detroit), who recently opened for Jack White on his Supply Chain Issues Tour. As a festival, we were thrilled with the opportunity to use the Dunlop Building and transform it into a HHM-style venue for one evening, and bring 3 very different bands together to make it's mark on "Midtown Bay City"! It's Hell's Half Mile's sixteenth birthday, and we celebrated the only way we know how. The Art Department of Bay City provided stunning custom neon and unique art installations in the bar/lounge (former showroom) area, while Lightnin' Licks Vinyl Preservation Society provided venue layout, logistics, design and decorations (service garage). Dunlop Building owner Mike Beattie allowed HHM volunteers to showcase the history of the building, utilizing many promotional materials from the business' glory days as a Pontiac dealership and service center. Dunlop Pontiac was regularly voted as the #1 auto dealership in the Greater Bay region, selling both new and used vehicles to the community. Most longtime residents of Bay City have a connection to the building due to the once sizable workforce at nearby GM Powertrain, allowing family and friends to purchase their cars from the lot with GM discounts. It was important to HHM and Lightnin' Licks Vinyl Preservation Society that the aesthetics of the venue reflected the history of the building, marrying the city's auto manufacturing past with it's cultural present, as any space where working people spent their time away from their families deserves to be treated with respect. The sounds of this soon-to-be-season are as satisfying as she gets, folks! In honor of MLB's Spring Training games kicking off (acceptable mixed-sports metaphor) this weekend, we're placing a special treat in our episode feed. In episode 6, Deon told the story of creating the original pilot episode of Lightnin' Licks Radio in May of 2019. He did so to test all of the music and recording equipment in the basement and also to convince Jay that their stories could ultimately be rendered as a sonically palatable product, therefore presenting podcasting as a worthwhile endeavor. Well...it worked! Jay agreed to partner with Deon thus Lightnin' Licks Radio podcast was born. This is that fateful pilot episode. Although it doesn't sound too similar to what the podcast would eventually become as far as format is concerned, the storytelling element was strong from day one, with vinyl records and other found-sound/field recordings/archival audio sources playing an important role in the story. If you haven't already heard this episode and you enjoy baseball or poetry or stories of childhood memories, give it a listen. We think it's pretty cool. When many Michiganders think spring, we think baseball. And many spring-loving, baseball-minded Mitten-dwellers have a special place in their hearts for Ernie Harwell, the voice of the Detroit Tigers for many generations. This episode was written and recorded a few days after news broke that Ernie's widow Lulu had passed away at the age of 99, not long before the 9th anniversary of her legendary broadcaster husband's death. Fabricio is a postdoc in the LLR laboratory at École Polytechnique of Université Paris-Saclay. Fabricio is an experimental particle physicist and a friend of mine for many years. Abstract: The centeral nervous system uses feedback processes that occur at multiple time scales to control interactions with the environment. Insight on the neuromechanical mechanisms subserving the faster feedback processes can be gained by applying rapid mechanical perturbations to the limb, and observing the ensuing muscle responses using electromyography (EMG). The long-latency response (LLR) is the fastest process that directly involve cortical areas, with a motorneuron response measurable 50 ms following an imposed limb displacement. Several behavioral factors concerning perturbation mechanics and the active role of muscles prior or during the perturbation can modulate the long-latency response amplitude (LLRa) in the upper limbs, but the interaction between many of these factors had not been systematically studied before. We conducted a behavioral study on thirteen healthy individuals to determine the effect and interaction of four behavioral factors -- background muscle torque, perturbation direction, perturbation velocity, and task instruction -- on the LLRa evoked from the flexor carpi radialis (FCR) and extensor carpi ulnaris (ECU) muscles following the application of wrist displacements. The effects of the four factors listed above were quantified using both a 0D statistical analysis on the average perturbation-evoked EMG signal in the period corresponding to an LLR, and using a timeseries analysis of EMG signals. All factors significantly modulated LLRa, and that their combination nonlinearly contributed to modulating the LLRa. Specifically, all the three-way interaction terms that could be computed without including the interaction between instruction and velocity significantly modulated the LLR. Analysis of the three-way interaction terms of the 0D model indicated that for the ECU muscle, the LLRa evoked when subjects are asked to maintain their muscle activation in response to the perturbations (DNI) was greater than the one observed when subjects yielded (Y) to the perturbations ({Delta}LLRa -- DNI vs. Y: 1.76{+/-}0.16 nu, p

Abstract: Variation in ploidy occurs naturally in select plant and animal species. Ploidy variation can also occur spontaneously or be induced during artificial propagation of fish and shellfish. Studying species and systems that have variable ploidy requires techniques to infer ploidy of individuals. Massively parallel sequencing of biallelic SNPs has been used to infer ploidy, but existing techniques have several drawbacks. These include being limited to only comparing a fixed number of ploidies (diploidy, triploidy, and tetraploidy) and requiring that heterozygous genotypes in an individual be identified prior to ploidy inference. We describe a method of inferring ploidy from sequencing of biallelic SNPs based on beta-binomial mixture models. This method is generalized to apply to any ploidy and does not require prior identification of heterozygous genotypes. We demonstrate efficacy of this method for comparing ancestral octoploidy, decaploidy, and dodecaploidy (tetraploidy, pentaploidy, and hexaploidy for the sequenced SNPs) in white sturgeon and diploidy and triploidy in Chinook salmon with amplicon sequencing (GT-seq) data. Results indicated that ploidy could be reliably estimated for individuals based on distinct distribution of log-likelihood ratios (LLR) for known ploidy samples of both species that were tested. Confidence in ploidy estimates increased with sequencing depth. We encourage users to explore the sequencing depths and LLR critical values that provide reliable estimates of ploidy for a given organism and set of SNPs. We expect that the R package provided will empower studies of genetic variation and inheritance in organisms that vary in ploidy naturally or as a result of artificial propagation practices. Abstract: Knowledge on the organization of motor function in the reticulospinal tract (RST) is limited by the lack of methods for measuring RST function in humans. Behavioral studies suggest the involvement of the RST in long latency responses (LLRs). LLRs, elicited by precisely controlled perturbations, can therefore act as a viable paradigm to measure motor-related RST activity using functional Magnetic Resonance Imaging (fMRI). Here we present StretchfMRI, a novel technique developed to study RST function associated with LLRs. StretchfMRI combines robotic perturbations with electromyography and fMRI to simultaneously quantify muscular and neural activity during stretch-evoked LLRs without loss of reliability. Using StretchfMRI, we established the muscle-specific organization of LLR activity in the brainstem. The observed organization is partially consistent with animal models, with activity primarily in the ipsilateral medulla for flexors and in the contralateral pons for extensors, but also include other areas, such as the midbrain and bilateral pontomedullary contributions. The main problem with localized discriminant techniques is the curse of dimensionality, which seems to restrict their use to the case of few variables. This restriction does not hold if localization is combined with a reduction of dimension. In particular it is shown that localization yields powerful classifiers even in higher dimensions if localization is combined with locally adaptive selection of predictors. A robust localized logistic regression (LLR) method is developed for which all tuning parameters are chosen data¡adaptively. In an extended simulation study we evaluate the potential of the proposed procedure for various types of data and compare it to other classification procedures. In addition we demonstrate that automatic choice of localization, predictor selection and penalty parameters based on cross validation is working well. Finally the method is applied to real data sets and its real world performance is compared to alternative procedures.
