On this episode, host Sima Vasa welcomes Basel Fakhoury, CEO and Co-Founder of User Interviews, for an enlightening conversation about Basel's work in improving data acquisition specificity. During their conversation, Basel and Sima discuss: - Using data to uncover needs in the market to better cater products and services to a particular niche. - The lessons Basel and his team learned from their first failed startup, and how that experience helped build User Interviews. - How finding the right survey respondents, rather than using a random sample, can provide more substantive and relevant data. - Why AI is an aid to finding human respondents rather than a threat to human-centric data. - The potential for growth in the data research industry. Basel and his team's journey is an excellent case study of both the power of data in the present as well as the industry's future trajectory. J., Robishaw, J. D. Abstract: GPR37L1 is an orphan receptor that couples through heterotrimeric G-proteins to regulate physiological functions. Since its role in humans is not fully defined, we used an unbiased computational approach to assess the clinical significance of rare GPR37L1 genetic variants found among 51,289 whole-exome sequences from the DiscovEHR cohort. Briefly, rare GPR37L1 coding variants were binned according to predicted pathogenicity, and then analyzed by Sequence Kernel Association testing to reveal significant associations with disease diagnostic codes for generalized epilepsy and migraine, among others. Since associations do not prove causality, rare GPR37L1 variants were then functionally analyzed in SK-N-MC cells to evaluate potential signaling differences and pathogenicity. Notably, receptor variants exhibited varying abilities to reduce cAMP levels, activate MAPK signaling, and/or upregulate receptor expression in response to the agonist prosaptide (TX14(A)), as compared to wild-type receptor. In addition to signaling changes, knockout of GPR37L1 or expression of certain rare variants altered cellular cholesterol levels, which were also acutely regulated by administration of the agonist TX14(A) via activation of the MAPK pathway. Finally, to simulate the impact of rare nonsense variants found in the large patient cohort, a knockout (KO) mouse line lacking Gpr37L1 was generated, revealing loss of this receptor produced sex-specific changes implicated in migraine-related disorders. Collectively, these observations define the existence of rare GPR37L1 variants in the human population that are associated with neuropsychiatric conditions and identify the underlying signaling changes that are implicated in the in vivo actions of this receptor in pathological processes leading to anxiety and migraine. Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2023.01.05.522910v1?rss=1 Authors: Koshko, L., Scofield, S., Debarba, L., Stilgenbauer, L., Sacla, M., Fakhoury, P., Jayarathne, H., Perez-Mojica, J. E., Griggs, E., Lempradl, A., Sadagurski, M. Abstract: The hypothalamus is essential in the regulation of metabolism, notably during critical windows of development. An abnormal hormonal and inflammatory milieu during development can trigger persistent changes in the function of hypothalamic circuits, leading to long-lasting effects on the body's energy homeostasis and metabolism. We recently demonstrated that gestational exposure to benzene at smoking levels induces severe metabolic dysregulation in the offspring. Given the central role of the hypothalamus in metabolic control, we hypothesized that prenatal exposure to benzene impacts hypothalamic development, contributing to the adverse metabolic effects in the offspring. C57BL/6JB dams were exposed to benzene in the inhalation chambers exclusively during pregnancy (from E0.5 to E19). The transcriptome analysis of the offspring hypothalamus at postnatal day 21 (P21) revealed changes in genes related to metabolic regulation, inflammation, and neurodevelopment exclusively in benzene-exposed male offspring. Moreover, the hypothalamus of prenatally benzene-exposed male offspring displayed alterations in orexigenic and anorexigenic projections, impairments in leptin signaling, and increased microgliosis. Additional exposure to benzene during lactation did not promote further microgliosis or astrogliosis in the offspring, while the high-fat diet (HFD) challenge in adulthood exacerbated glucose metabolism and hypothalamic inflammation in benzene-exposed offspring of both sexes. These findings reveal the persistent impact of prenatal benzene exposure on hypothalamic circuits and neuroinflammation, predisposing the offspring to long-lasting metabolic health conditions.
See omnystudio.com/listener for privacy information.

Ever thought about what you eat, struggled with your weight or wanted to know more about nutrition? In this episode we had the opportunity to sit down with Fatimah Fakhoury a Registered Dietitian providing virtual nutrition coaching services to people living with paralysis and spinal cord injury. She was inspired by her husband, who was injured in a car accident many years ago, to focus on providing nutrition services specifically to people living with paralysis, who are often left out of popular wellness culture. Fatimah empowers people living with paralysis to manage their weight and improve their bowel health by providing support to make healthier choices, without restrictive dieting. Welcome back to The BeREAL Podcast! This week, Diana and Ednesha are joined by physical therapist Farris Fakhoury to discuss physical therapy, his new book, and his practice during the COVID-19 pandemic. Farris is a physical therapist in a neurological rehab setting. He is also the author of “The Happiness Perspective” being published in December 2020. The premise of his book is happiness and the common threads his patients exhibit to help them unlock their happiness in recovery. Farris begins the episode with a little backstory into how he got into physical therapy. He explains that it began back when he was a kid playing sports, and he had to seek injury recovery through physical therapy. From here, he developed a passion for the practice and pursued a psychology degree in college. Now he works with patients who have suffered neural damage through strokes and spinal cord injuries, but he primarily works with amputee patients. Farris discusses the topic of happiness a lot throughout the episode. He explains how he sees happiness as a muscle that needs to be worked and can grow and shrink. He doesn't view happiness as a final destination, but rather a journey. Diana chimes in with her view of happiness and how she doesn't see it as a constant state either. Instead, they both acknowledge how happiness ebbs and flows. From here, Farris discusses the mental health of his patients. He gives an overview of how he witnesses mental health change as his patients progress through recovery and gain more confidence. Lastly, Farris discusses how his life has been since the COVID-19 pandemic has hit. He goes over how it has given him the time to collect his journal entries from over the years and adapt them into a book that is to be published in December 2020. He also explains how his practice has changed. He definitely ran into some roadblocks doing physical therapy virtually, but he has been adjusting his techniques to give the best experience possible to his patients. His in person visits have also been different, as patients cannot bring support with them. He discusses how this is difficult both mentally and physically on the patients. Thank you for tuning into BeREAL this week! Be on the lookout next Tuesday for a brand new episode. So, to lose weight, if they're trying to do it on their own, they can't necessarily calorie count on those applications because our foods aren't there. So, it's a pure guesstimate like a roti or chapati ends up becoming a tortilla.” - Sarika Shah Guest Resources: Fatimah's Instagram: @paralysis.nutrition Fatimah's Website:: https://paralysisnutrition.com/ Sarika's Instagram: @indian.nutritionist Sarika's Website: http://www.thenourishedrd.com Free Resources from Libby Are you ready to start your journey? “What is it that you love to do?” Dr. Manal Fakhoury is President and CEO of Fakhoury Leadership International; she also serves as a beacon of inspiration for women stepping into doing what they love to do. Quoting Warren Buffet, “Investing in yourself if the best thing you can do”, Dr. Fakhoury walks her talk. She is President of Toastmasters and Rotary, both organizations dedicated to personal growth and achievement, is a graduate of Leadership Florida and invests in many seminars and women’s conferences, and facilitates many others. A key statement in her interview was “You don’t have to have certification to provide value. Most women don’t understand that and we use lack of education and certification as an excuse to play small”. Isn't that powerful to think about? One of Manal Fakhoury’s achievements was hiking Mount Kilimanjaro. The story about how that came about is both inspiring and funny! Her climb became inspired by Climb of Hope, and her climbing group helped two young amputees make the ascent up Mount Kilimanjaro. Dr. Fakhoury may be reached at myfli.com or Info@myfli.com
