Monica Coenraads, the mother of a daughter with Rett Syndrome, has played a critical role in catalyzing development of therapies to treat the rare, neurological disease. She co-founded and served as director of research for the Rett Syndrome Research Foundation. She later founded and today serves as CEO of the Rett Syndrome Research Trust. Her work shows how patient organizations can bridge the translational divide and de-risk rare disease drug development for biopharmaceutical companies. Karen Thomas of Naturally Recovering Autism interviews Zen Honeycutt from Moms Across America on Aluminum, Arsenic, Cadmium, Lead, and Mercury Found in Baby Formula. Catalyzed by the FDA's failure to regulate heavy metals in baby formula, a coalition of consumer advocacy groups commissions testing of 20 different brands. Groups urge the FDA and legislators to protect infants from toxic exposure. Severe health risks posed by these toxic metals include developmental delays, cancer, kidney damage, and neurocognitive difficulties. A coalition of consumer advocacy groups, including Moms Across America, GMOScience, and The New MDS, has uncovered alarming levels of toxic metals in baby formula. Testing of 20 popular brands revealed that 100% of samples contained aluminum and lead, with over half also testing positive for arsenic, mercury, and cadmium. Disturbingly, six formulas contained all five metals in both samples. Notably, aluminum levels in one goat's milk formula were up to 40,000 times higher than other metals tested, far exceeding FDA safety limits. Mercury and cadmium levels in several samples also surpassed allowable limits in drinking water. Despite the FDA's “Closer to Zero” initiative launched in 2021, no substantial regulatory limits have been set, leaving babies vulnerable. Recent efforts, including the Baby Food Safety Act of 2024, which was announced by Congress on May 9, one week after the heads of Moms Across America and GMOScience met with lawmakers and the FDA in D.C. to deliver the results of the baby formula testing, aim to give the FDA authority to regulate and enforce limits on levels of heavy metals found in baby food and potentially formula. Talk points in this podcast: Alarming rates of toxic metals when testing 20 different baby formula brands. 100% of the 40 samples tested contained aluminum and lead. 57% of the samples tested positive for arsenic, 55% for mercury, and 35% for cadmium. Six of the 20 formulas were positive for all five toxic metals in both samples. At 41,000 ppb, aluminum levels in a goat's milk baby formula were 4000 – 40,000x higher than other metals in the formulas tested and exceed limits set by the FDA for maximum safety level of aluminum for a preemie. Levels of mercury in four samples measured above the limit allowed by the FDA in drinking water. Levels of cadmium in both samples of one formula were nearly twice the level allowed in drinking water. Amidst a broad-based rally in risky assets, commodities stand out as under-valued, under-owned, and backed by compelling fundamentals and technicals—a good entry point for a catch-up tactical allocation. The BCOM index gained 5% in July, but there is likely more where this came from. Should recessionary fears not materialize over the next six months, idiosyncratic risks leave the entire complex exposed to the upside, with the rally in BCOM extending. In order of preference, we favor natural gas, ags, oil and gold, and maintain neutral stance on base metals. The paths to monetization are seemingly endless but the most obvious converge on making humans more productive or supercharging existing business models like search advertising or subscription licenses. Much of AI adoption in enterprise IT is hidden. Our research shows a very high overlap (around 40-60%) between AI adoption in enterprise tech and embedded AI inside software from the likes of Salesforce, ServiceNow, Workday, SAP, Oracle and other major players. But the rapid advancements of tools from AI leaders and an emerging group of independent firms is causing customers to think differently. Catalyzed by the OpenAI Microsoft partnership, organizations are rapidly trying to figure out how to apply these tools to create competitive advantage. Every firm on the planet wants to ride the AI wave. Lee spent ten years in prison after that arrest, and very possibly might have spent his whole life there if it were not for the dogged advocacy of Asian American activists who saw in his case a grave injustice born of racism and indifference. By the time of Lee's death in 2014, this movement had largely been forgotten, but a new documentary film is reviving the memories. The production of hydrinos results in direct electricity. 3. BlackLight process has four principle applications to motivate power: ciht, charging batteries, electrolysis to charge batteries, heat converted to electricity to charge batteries, and hybrid electric system. 4. Water has 55 moles/liter. Hydrogen to hydrino releases 50 MJ/Mole H2. One liter of water would produce 2.75 billion joules of energy. 5. A Prius consumes 9 x 10^5 J/Mile. The prius could travel 3000 miles, but most fuel cells are 50% efficient, reducing the range to 1500 miles. The onboard electrolysis unit providing the hydrogen would use 1% of the electrical output. 6. The UNDP says an analysis of 159 developing countries globally indicates that price spikes in key commodities are already having immediate and devastating impacts on the poorest households, with clear hotspots in the Balkans, countries in the Caspian Sea region, and Sub-Saharan Africa.

As such, she has always had an interest in access and equity in both the government and society. Courtney's studies in Political Science, Religion, and Philosophy at Middlebury College were generalist and explored both the philosophical and observed aspects of governance on a national scale. After college, however, she found myself drawn towards the kind of community-building and empathetic advocacy that is necessary to address radicalization after experiencing the polarization and radicalization of her own father. Catalyzed to take action, Courtney decided to pursue her master's to explore the nexus between policy, society, and innovative counter extremism measures. The following is a conversation between Sean Doherty, the Chairman of the JDRF Type 1 Diabetes Fund, and Denver Frederick, the Host of The Business of Giving. In 2015, a group of JDRF volunteers, formerly known as the Juvenile Diabetes Research Foundation, observed that there was virtually no private investment in type 1 diabetes, which was causing a gap between scientific advancements and the delivery of solutions to the people who need them. Their solution: launch the JDRF T1D Fund, a venture philanthropy fund to create a market in T1D by using their capital and expertise to convince venture capital, biotech, and pharma to make the investments needed to cure type 1 diabetes. Almost 41% of those looking to buy a car are considering buying an electric vehicle (EV). The EV sales boom is a part of a wider resurgence in car buying. What happens when the vision crystalizes and we realize that what we remember...what has helped us make decisions that our future now depends on...is not the whole story, much less the whole truth? My dear friend recently helped me access the most profound, shiver-worthy message that sent me back into the past so that I could re-experience it, so that I could see what I couldn't see the first time - so that I could scoop the new energy, and recapitulate the timeline between then and now... When, in 2017, I battled a whole array of conflicting emotions that I did not quite understand, a particular scene of Moana pierced my heart! It has accurately articulated what I had been experiencing; it solidified into a masterfully visual form the homesickness, the unresolved issues in my relationships, the feeling of having outgrown our space and the roles expected to play. 

It was the ancestral calling. And, it was the grieving of my old self as the process of the emergence of the new-me was just about to begin. When, in 2018, my daughters and I embarked on our 4-month Slovenian adventure, the only awareness I had, was to slow down. To heal. To reconnect. And when we returned, I experienced another dark night of the soul; I felt I no longer belonged to our home, into our union. I felt like a wild cheetah now caged and confined to a small patch of artificial grass - ready to break out. By the time 2020 pressed pause on our lives and the world seemingly stopped spinning, and the collective was filled with anger and sadness and resentment and haterage... I saw a glimpse of hope. I felt at peace. And, I felt courage. Courage to let the new me emerge: Catalyzed by awareness. Powerful in convictions. Rooted in deep knowing. In ownership of divine gifts. 

And ready to begin working on Legacy book. As words kept pouring out and on the paper, as new memories kept emerging, and pages kept being written, time has come to change the narrative. To intentionally deepen it until it feels light again - until it feel true. It was time to revisit the past... In today's journey, I also walk you through the process of making some of the most difficult decisions as a woman, wife, mom; the experience of having the most uncomfortable conversations with loved ones; the tunnel vision when advocating for oneself with no tangible or measurable proof other than the gut feeling.

Abstract: P-glycoprotein (P-gp) is a critical membrane transporter in the blood brain barrier (BBB) and is implicated in Alzheimer's disease (AD). However, previous studies on the ability of P-gp to directly transport the Alzheimer's associated amyloid-{beta} (A{beta}) protein have produced contradictory results. Here we use molecular dynamics (MD) simulations, transport substrate accumulation studies in cell culture, and biochemical activity assays to show that P-gp actively transports A{beta}. We observed transport of A{beta}40 and A{beta}42 monomers by P-gp in explicit MD simulations of a putative catalytic cycle. In in vitro assays with P-gp overexpressing cells, we observed enhanced accumulation of fluorescently labeled A{beta}42 in the presence of Tariquidar, a potent P-gp inhibitor. We also showed that A{beta}42 stimulated the ATP hydrolysis activity of isolated P-gp in nanodiscs. The second of a fascinating two-part discussion, this episode delves deeper into Ming Peiffer’s Usual Girls. The play premiered in 2018 at Roundabout Underground for an extended, sold-out run. Catalyzed by the allegations against American Apparel’s Dov Charney, playwright Peiffer began to investigate the stories of women and the milestones in the sexual maturation of girls in America that can lead to a fraught and vulnerable relationship to one’s own sexuality. Peiffer put the patriarchy, rape culture, sexism, misogyny, and racism on trial in her professional debut work. What does healthy sexual development look like? How can femmes claim (or reclaim) their own sexuality? Is it possible to shed the culturally imposed shame and guilt and adopt an outlook of pleasure? Abstract: Discovering the physical principles directing organelle assembly is a fundamental pursuit in biology. Centrioles are evolutionarily conserved organelles with a 9-fold rotational symmetry of chiral microtubules imparted onto the cilia they template. Centriole assemble from likewise symmetrical ring polymers of SAS-6 proteins, orthogonal to a toroidal surface surrounding the resident centriole. How surface properties ensure ring assembly with proper symmetry and orthogonal arrangement is not known. Here, we deployed photothermally-actuated off-resonance tapping high-speed atomic force microscopy (PORT-HS-AFM) to decipher physical principles of surface-guided SAS-6 self-assembly. Using machine learning to quantify the polymerization reaction and developing a coagulation-fragmentation model, we discovered that the surface shifts the reaction equilibrium by ~104 compared to the solution situation, explaining orthogonal organelle emergence. Moreover, molecular dynamics and PORT-HS-AFM revealed that the surface converts helical SAS-6 polymers into 9-fold ring polymers with residual asymmetry, which may impart chiral features to centrioles and cilia. Abstract: Amyloid seeds are nanometre-sized protein particles that accelerate amyloid assembly, as well as propagate and transmit the amyloid protein conformation associated with a wide range of protein misfolding diseases. However, seeded amyloid growth through templated elongation at fibril ends cannot explain the full range of molecular behaviours observed during cross-seeded formation of amyloid by heterologous seeds. Here, we demonstrate that amyloid seeds can accelerate amyloid formation via a surface catalysis mechanism without propagating the specific amyloid conformation associated with the seeds. This type of seeding mechanism is demonstrated through quantitative characterisation of the cross-seeded assembly reactions involving two non-homologous and unrelated proteins: the human A{beta}42 peptide and the yeast prion-forming protein Sup35NM. Our results suggest experimental approaches to differentiate seeding by templated elongation from non-templated amyloid seeding, and rationalise the molecular mechanism of the cross-seeding phenomenon as a manifestation of the aberrant surface activities presented by amyloid seeds as nanoparticles. Oxidation reactions of sugars to their corresponding lactones and the selective oxidation of a secondary alcohol of sugar substrates to keto-sugars using transition metal catalysis were investigated. Research results demonstrate that Shvo’s hydrogen-transfer catalyst, [(η4-C4Ph4CO)(CO)2Ru]2, selectively oxidized unprotected sugars to δ-lactones under very mild conditions. This is the first method to synthesize δ-D-galactonolactone, the first oxidation product of the 3rd most abundant sugar. δ-D-Galactonolactone was fully characterized by conformational analysis through NMR experiments and computational methods. We propose that in aprotic, anhydrous solvents the δ→γ lactone rearrangement mechanism proceeds via a bicyclic transition state. The oxidation of alicyclic vicinal diols to α-hydroxy ketones was investigated as a model system for the selective oxidation of a secondary hydroxyl group in sugar substrates to keto-sugars. The challenge was to develop a method that allows selective oxidation, yet prevents over-oxidation to the dione or dicarboxylic acids products. Based on a mathematical model of consecutive bimolecular reactions, the rate constants for the initial oxidation step k1(diol→α-OH ketone) and second oxidation step k2(α-OH ketone→dione) must be k1 > 10 k2 in order to obtain a synthetically useful method for α-hydroxy ketone synthesis. The metal-ligand bifunctional hydrogen transfer reactions of Shvo’s catalyst and Noyori’s η6-arene N-tosyl-1,2-diaminoethane ruthenium(II) complexes were investigated with alcohol model systems and it was concluded that hydrogen transfer reactions are insufficient in the oxidation of vicinal diols due to a unfavourable position of the equilibrium. Under oxidizing conditions, the 16-electron ruthenium complexes of the Noyori systems compete with a β-elimination process and thus new degradation resistant ligands were synthesized. The apparent slower oxidation of Noyori’s η6-arene N-tosyl-1,2-diaminoethane ruthenium(II) complexes under oxidizing conditions, i.e. in acetone or cyclohexanone solvent, was investigated through NMR and IR experiments finding no evidence of a kinetic inhibition by the solvent. We propose that the slower reactions depend on a relatively high energy barrier for the reaction of the 16-electron complex with a hydrogen donor and that a kinetic model must account for two effectively different oxidation and reduction catalysts. In an alcohol/ketone equal-concentration experiment with Noyori-type ruthenium(II) complexes a linear relationship was found between the initial rate of alcohol consumption/production and ∆G°. Reactions involving peroxides as the oxidant were investigated in order to avoid equilibrium processes. The oxidation of alcohol model systems with several Mo and W catalysts and peroxide sources indicated a deactivation of the Mo and W catalysts by formation of water or hydroxide complexes. The product distribution of the oxidation of trans-1,2-cyclohexanediol with the MoO2(acac)2/Na2CO3 x 1.5 H2O2 method was in agreement with the theoretical model, yet only had a k1 = 1.5 k2 rate ratio resulting in a maximum α-hydroxy cyclohexanone content of 45 %. The NiBr2 mediated oxidation of alcohols and benzoylperoxide was investigated. The selective oxidation of vicinal diols was unsuccessful with this method. However, the oxidation reaction of mono-alcohol substrates was greatly improved using water in the reaction. The reaction mechanism was investigated and we propose that the actual oxidant in the NiBr2 mediated benzoylperoxide method is [Br+], which is generated by the hydrolysis product of benzoylhypobromite, hypobromic acid (HOBr). Ishii’s stoichiometric NaHSO3/NaBrO3 reagent selectively oxidized vicinal diols to α-OH ketone without overoxidation. This oxidation reaction was investigated with regards to substrate, concentration and pH. The reactivity and selectivity were studied and it was found that the oxidation mechanism is based on a multitude of comproportionation and disproportionation equilibria at low pH. A small but steadily replenished HOBr concentration is the source of the actual oxidant and effectively acts as a redox buffer. While the NaHSO3/NaBrO3 reagent demonstrated excellent selectivities with the alcohol model systems, the oxidation of sugar substrates failed due to side-reactions occurring at the required low pH.

Gradual depletion of the ozone layer and consequently, increased ultraviolet (UV) radiation on the Earth's surface induces DNA-lesions inside the genome, thereby causing mutations. Three kinds of photoproducts are mainly formed, namely: cyclobutane pyrimidine dimers (CPD), pyrimidine-(6-4)-pyrimidone dimer [(6-4)PP] and the Dewar valence isomer of (6-4)PP lesion. The formation of these photolesions is a major cause of cell death (aging) and fatal disease like skin cancer. A part of this research was performed to investigate the formation and characterization of DNA-lesions under UVC radiation. Small, fluorescent labeled oligonucleotide hairpins (DNA, RNA and mixed DNA-RNA) were employed to this purpose. The amount of damage was quantified using HPLC. Here, a new method was developed, using ion-exchange SAX-column which works at pH = 13, to measure the lesion formation in a direct way. In order to know which lesions are formed, the irradiated hairpins were enzymatically digested. The lesions were separated by HPLC followed by their characterization using MS/MS fragmentation analysis. Investigation was performed to understand the impact of the neighboring nucleobases on the photo-reactivity of a dTpdT-dinucleotide. For this, hairpins were prepared in various sequential contexts. Analysis of these irradiated hairpins revealed the surprising result that the reactivity is strongly reduced when a dTpdT dinucleotide is locked between two neighboring 2'- deoxyguanosines, strongly implying that genomic DNA will be inhomogeneously damaged and hence mutated under UVC radiation. In order to account for the effect of oligonucleotide conformations (A versus B) on the photolesions formation, DNA, RNA and mixed DNA-RNA hairpins were irradiated. The most surprising observation is that the oligonucleotide hairpins, possessing more A-like conformations were found to be very much resistant to UVC degradation. RNA hairpins containing UpU dinucleotides were found to be fully protected from being damaged. A short, dTpdT-containing DNA stretch, embedded in an A-like RNA environment, was also found to be highly stable under UVC light. In the second part of this study, investigation was performed to assay the activities of CPD-photolyase enzyme isolated from different organisms. A synthetic cis-syn thymidine dimer with an open backbone was incorporated into DNA. CPD-photolyases were found to recognize and split this CPDlesion, via a cycloreversion process, into two 2'-deoxythymidine monomers. In order to profile the activities of photolyases and also to study photolyase activity inside living cell, a sensitive DNA-probe, known as molecular beacon was synthesized. The hairpin probe, which features a loop and stem structure contains the model CPD-lesion in its loop region. The molecular beacon (MB 1) in its closed form, is non-fluorescent due to efficient energy transfer (FRET) from the 5'-FAM to 3'-Dabsyl. The MB 1 was designed to undergo strand break on reaction with photolyase in presence of light, thus separating the FAM and the Dabsyl and causing the fluorescence of FAM to be restored. The activities of the photolyase can be studied by monitoring the fluorescence change. Activities of CPDphotolyase isolated from A. nidulans and A. thaliana were studied using this MBprobe. The sensitivity of this probe was tested with wild-type cell-extract from A. thaliana. The fishing-out of photolyase activity from this wild-type extract was possible. Effort was made to investigate the repair process within a living cell, using laser scanning fluorescence microscopy. The insertion of this artificial, chemically modified DNA-substrate in the cell nucleus was achieved.

In the first part, the use of chirality transfer in acyclic allylic systems was assessed for an intra- and an intermolecular reaction. The thermal [2,3] sigmatropic rearrangement of acyclic allylic phosphinites proved to be highly enantioselective, even at 110 °C and led to enantiomerically pure allylic phosphine oxides. The substitution pattern of the substrate and the reaction conditions were fully optimized.The chirality transfer concept was also used intermolecularly in asymmetric allylic substitution reactions. This allowed to build up enantiomerically pure quaternary centers. The product were then reacted to obtain enantionerically pure tertiary alcohols, desymmetrized 1,3 diols and aldol compounds. In a second part, it was found that Heck reaction leading to neopentylic palladium species underwent, in the absence of nucleophile, a C-H activation reaction, leading to spiro cyclopropyl compounds.

In summary, reaction conditions have been found for a versatile nickel-catalyzed crosscoupling reaction in the presence of an electron deficient styrene promoter, allowing for the first time: · effective coupling of readily available functionalized alkylzinc halides and benzylic zinc bromides with functionalized alkyl iodides in the presence of TBAI · the application of alkyl bromides in a Csp3-Csp3 cross-coupling reaction. Furthermore, · the presence of sensitive functional groups such as an amino function with an unprotected NH group is tolerated · coupling of diastereomerically enriched zinc reagents proceed with high retention of configuration at the zincated carbon · scaling up can be achieved without significant loss of chemical yield. In summary, an effective and high yielding protection-deprotection protocol has been developed for the iodine-magnesium exchange reaction of 2-iodo-1,4-phenylenediamine. The corresponding 2-magnesiated-1,4-phenylenediamine derivatives can be transmetallated either to copper: · a subsequent allylation reaction furnishes the allylated products in good yield, or to the corresponding zinc reagents: · a subsequent smooth palladium-catalyzed cross-coupling reaction with a range aryl iodides furnishes the polyfunctionalized cross-coupling products in good yield, · this cross-coupling reaction is highly tolerant towards electronic as well as steric effects.

Lewis acid catalyzed addition reactions of alkyl halides 1 with unsaturated hydrocarbons 2 have been studied. 1:l addition products 3 are formed if the addends 1 dissociate faster than the corresponding products 3; otherwise, polymerization of 2 takes place. For reaction conditions under which 1 and 3 exist mainly undissociated, solvolysis constants of model compounds can be used to predict the outcome of any such addition reactions if systems with considerable steric hindrance are excluded.
