In a devastating blow to a faithful Texas Catholic family with six children who have made their living running a private ranch, the Archbishop of San Antonio Texas has issued a public letter to all Catholics directing them not to do business with the ranch. The January 30th letter formally prohibits all clergy, schools, retreat centers, catholic groups “from contracting with or utilizing Sanctus Ranch for any Catholic-sponsored retreats, meetings, activities, or spiritual endeavors.” Even where contracts were already signed, the bishop directed them to discuss it with his legal team rather than fulfill their contractual obligations. Archbishop Gustavo Garcia-Siller instructs Catholics against Sanctus Ranch, telling them not “to participate in any of its activities.” Previous to the Archbishop's action against Sanctus Ranch, the family-run business had bookings nearly every weekend. Every Catholic booking has been canceled since they were by Catholic groups in the archdiocese. Creative projects have a habit of taking on their own lives and bringing the creator along for the ride. In this episode, we speak with science writer Melissa L. Sevigny about how her book Brave the Wild River: The untold story of two women who mapped the botany of the Grand Canyon surprised her and required her to tell a story different from the one she set out to write. She also shares how she managed to work a full-time job while researching and writing, how she created three-dimensional characters out of archival information and interviews, how the book let her know she was done, and what she learned from this project to apply to future projects. Melissa L. Sevigny grew up in Tucson, Arizona, where she fell in love with the Sonoran Desert's ecology and dark desert skies. She has worked as a science communicator in the fields of space exploration, water policy, and sustainable agriculture, and has a B.S. in environmental science from the University of Arizona and an M.F.A. in creative writing from Iowa State University.She is the science reporter at KNAU (Arizona Public Radio) in Flagstaff, Arizona and her stories have been awarded regional Edward R. In this episode, it was my pleasure to interview science journalist Melissa Sevigny about her new book, Brave the Wild River: The Untold Story of Two Women Who Mapped the Botany of the Colorado River. She is the science reporter for Arizona Public Radio as well as the author of two previous books, Mythical River: Chasing the Mirage of New Water in the American Southwest, and Under Desert Skies: How Tucson Mapped the Way to the Moon and Planets. Her work has also appeared in many places, including Orion, The Atavist Magazine, Science Friday, and more. I spoke with Melissa about writing a book while working a full time job as a reporter, how she created such detailed scenes, and the helpful metaphor she used to think about the book's narrative structure.

Aging is a complex and dynamic process which influences distinct cell types in a myriad of ways. The cellular architecture of the mammalian brain is heterogeneous and diverse, making it challenging to identify precise areas and cell types of the brain that are more susceptible to aging than others. Here, we present a high-resolution single-cell RNA sequencing dataset containing ~1.2 million high-quality single-cell transcriptomic profiles of brain cells from young adult and aged mice across both sexes, including areas spanning the forebrain, midbrain, and hindbrain. We find age-associated gene expression signatures across nearly all 130+ neuronal and non-neuronal cell subclasses we identified. We detect the greatest gene expression changes in non-neuronal cell types, suggesting that different cell types in the brain vary in their susceptibility to aging. We identify specific, age-enriched clusters within specific glial, vascular, and immune cell types from both cortical and subcortical regions of the brain, and specific gene expression changes associated with cell senescence, inflammation, decrease in new myelination, and decreased vasculature integrity. We also identify genes with expression changes across multiple cell subclasses, pointing to certain mechanisms of aging that may occur across wide regions or broad cell types of the brain. Finally, we discover the greatest gene expression changes in cell types localized to the third ventricle of the hypothalamus, including tanycytes, ependymal cells, and Tbx3+ neurons found in the arcuate nucleus that are part of the neuronal circuits regulating food intake and energy homeostasis. These findings suggest that the area surrounding the third ventricle in the hypothalamus may be a hub for aging in the mouse brain. Overall, we reveal a dynamic landscape of cell-type-specific transcriptomic changes in the brain associated with normal aging that will serve as a foundation for the investigation of functional changes in the aging process and the interaction of aging and diseases. Abstract: Gut stem cells are accessible by biopsy and propagate robustly in culture, offering an invaluable resource for autologous cell therapies. Insulin-producing cells can be induced in mouse gut, but it has not been possible to generate abundant and durable insulin-secreting cells from human gut tissues to evaluate their potential as a cell therapy for diabetes. Here we describe a protocol to differentiate cultured human gastric stem cells (hGSCs) into pancreatic islet-like organoids containing gastric insulin-secreting (GINS) cells that resemble {beta}-cells in molecular hallmarks and function. Sequential activation of the inducing factors NGN3 and PDX1-MAFA led hGSCs onto a novel differentiation path, including a SOX4High endocrine and GalaninHigh GINS precursor, before adopting {beta}-cell identity, at efficiencies close to 70%. GINS organoids acquired glucose-stimulated insulin secretion in 10 days and restored glucose homeostasis for over 100 days in diabetic mice after transplantation, providing proof of concept for a new approach to treat diabetes. Boys Don't Cry holds a 90% Rotten Tomatoes critics score and a pretty astonishing 86% Metacritic. And with scores like that, one might get the impression that was and is a universally admired classic. Billed as a dramatization of the events leading up to the 1993 rape and murder of 21-year-old trans man Brandon Teena, it was nominated for 57 major awards and won 37 of them. 14 of went to star Hilary Swank, 6 to costar Chloe Sevigny, and 8 to writer director Kimberley Peirce. And Swank of course won the Oscar for Best Actress, while Sevigny was nominated for Best Supporting Actress. But critics at the time almost always misgendered Brandon in their reviews. And the film effectively erases the trans identity of its protagonist, which some argue is the result of a conscious and unforgiveable narrative and creative decision made my Peirce, who cast a cisgendered woman to play Brandon and who shut out essentially all input and participation from anyone in the trans community. In the 23 years since its release, Boys Don't Cry has not aged well. But this week's guest, writer and trans horror historian Logan Ashley Kisner, argues that it's not all about aging - Boys Don't Cry is a fundamentally, irredeemably transphobic film. Greetings, Citizens! Dan Here. Welcome to the first of 2 episodes that you'll be getting this week. On Episode 119 Steve is out and I sit down with returning guests Joe Stark from Starkcast and Leftover Brian from Pop Culture Leftovers. This was actually going to be a bonus episode as we were only planning to talk about one particular movie but we ended up talking about 5. Before we get to the main event, Brian and I briefly talk about the documentary "Miss Snake Charmer" (Now on Amazon Prime Video), All 3 of us have seen "The Old Guard" starring Charlize Theron (Now on Netflix), Brian and I also talk about "Palm Springs" (Now on Hulu), as well as the 2014 movie that came from out of nowhere called "The One I Love" (also on Netflix and starring Elisabeth Moss and Mark Duplass). Now I want to take a moment to personally thank Brian and Joe for doing me a solid and entertaining my sick idea to review Harmony Korine's 1995 breakout film "Kids". If you're not familiar with the premise of "Kids" here it is: Kids is a 1995 American coming-of-age drama exploitation film directed by Larry Clark and written by Harmony Korine. It stars Leo Fitzpatrick, Justin Pierce, Chloë Sevigny, and Rosario Dawson, all in their film debuts. Primarily set during the course of one day, Fitzpatrick, Pierce, Sevigny, Dawson, and other newcomers portray a group of teenagers in New York City with hedonistic behavior toward sex and drugs during the mid-1990s. 2 Warnings: "Kids" is a very graphic movie that deals with some uncomfortable subjects and we definitely get into that. This is also a spoiler filled review so if you haven't seen it and plan to, you might want to consider coming back to this one after viewing. Lastly I want to publicly apologize to Rod Claerbout, cohost of Number One Comic Books, for being an absolute shit head and referring to him as "Dustin". I literally have no idea why I spaced out on your name and again, I apologize. Trust me that Brian and Joe let me have it when I said that. (This wasn't my best hosting job. I admit it) Brian, Joe, and Rebekah Dahling also host Number One Comic Books and I highly recommended that you listen to their podcast. Its super entertaining and informative. Sevigny was born in Miami and grew up in a home where he was encouraged to read, write and make art. His father was a painter and sculptor, and his mother, a feminist, photographer and activist. Sevigny himself has never been shy about expressing his opinions on the issues of the day, and has written in depth about everything from immigration and human trafficking to Reagan-era politics. He is best known for his uncompromising photographs, of Mexican prostitutes and portraits of Salvadoran gang members. Sevigny frequently speaks at universities across the Americas on structural violence, Abstract Expressionism, street photography, and frequently his own work. He taught photography for five years at la Universidad Centroamericana Jose Simeon Cañas (UCA) in El Salvador - a Roman Catholic school remembered for being the site of the killing of six Jesuit priests during that country's armed conflict. He has photographed for newspapers and magazines in the United States, Mexico, Guatemala, El Salvador and Brazil and been a guest artist at Benedictine University, Illinois and given guest lectures at the Pratt Institute and School of Visual Arts, both in New York; the Loyola University, Depaul University, and College of Dupage, all in Chicago; the University of Wisconsin at Oshkosh; and the University of Illinois, among many others. Dan Sevigny was sensitive kid who had learning difficulties and trouble connecting with his peers. He coped by cutting himself and started using alcohol and pills as a teenager. Which kicked off a 10+ year cycle where drug use, aggression, and petty crime would get him kicked out of school and then sent to rehab. The cycle became more destructive, the aggression turned more violent, the rehab leading to juvie and jail. Dan describes the feedback loops of addiction and how depression made him feel like he was "on fire, with no skin and with everything in his body hurting." Dan makes it extremely clear that there's no silver bullet to recovery, but help came in the form of a mentor, learning about Search Engine Marketing while in rehab, and numerous therapists, particularly around DBT (Dialectical Behavior Therapy). Mark Alan Sevigny graduated from the University of San Diego with a Juris Doctor in May 1977, after receiving a BA from San Diego State in Political Science with honors in 1973. He passed California State Bar Exam in August 1977 and went to work for the Orange County District Attorney's Office in February of 1978. In his thirty years as prosecutor, Mr. Sevigny tried over ninety jury trials, including twenty-five homicides, eleven felony sexual assault cases, and numerous major fraud cases. He worked in almost every unit of the office, including five years in the Major Fraud Unit in the 1980's and again between January 1999 and February 2008. His activity in gang homicide investigations and prosecutions between 1990 and 1994 helped to formulate the legal and evidentiary strategies by which gang-related homicide cases were vigorously pursued and tried in Orange County. From 1995 through 1997 he worked as one of the lead attorneys in the criminal investigation of the Orange County Bankruptcy case, including a nine-month presentation of evidence to the Orange County Grand Jury concerning the role of investment firms in the county's financial calamity. This investigation eventually led to a $30 million unfair business practices settlement with Merrill Lynch. After that he served as supervising deputy district attorney in charge of Law and Motion of the District Attorney's Office for over a year and a half before returning to the Economic Crimes Unit in 1999 to be part of the Major Fraud and High Tech Crimes division. During his tenure with the office Mr. Sevigny trained fellow prosecutors in the District Attorney's Office and through the California District Attorneys Association on documentary evidence, identity theft, complex case preparation, and major fraud issues. Mr. Sevigny officially retired from the district attorney's office in February 2008, but still works there as a prosecutor on a part-time basis. He is also available as a consultant to private industry and insurance companies on issues relating to fraud and computer-related crimes.
