一、思想問題:當世人在哀嘆生命的短暫有限,念天地之悠悠,獨愴然而涕下時;我們短暫的生命有什麼盼望呢? 二、鑰節: 102:27 惟有你永不改變;你的年數沒有窮盡。102:28 你僕人的子孫要長存;他們的後裔要堅立在你面前。 這世界一切都會改變,天地要如外衣漸漸舊了,而惟有神是永遠不變,祂是我們的標竿與盼望,認識主、敬畏事奉主的人必然長存,生命、家庭要永遠被主堅立;這是無懼的人生,這是得勝的人生。 三、經文結構: A1-11 哀嘆人的年日短少。 →B12-17 神的永恆,與憐憫。 →→C18-22 後代要永遠讚美神。 A23-24 哀嘆人的日子短少。 →B24-27 神的永恆,24世世無窮 。 →→C28 後代讚美。 四、結構亮光: 本篇詩篇提到詩人正面對極大的苦難,可能是身體疾病,也可能是國破家亡的苦境,不斷地強調人生命的短暫與痛苦;但是當轉眼看見神,就看見那超越短暫苦難的永恆主在掌管一切,因此發出大讚美。 本篇結構採ABCABC平行結構,A是哀嘆人年日短少,B是看見神的永恆,C是後代要讚美神。 A的部分: vv.1-11與vv.23-24出現了七次「日子」:v.2急難之日、呼求之日、v.3消滅之日、v.8辱罵之日、v.11將亡之日、v.23短少之日、v.24中年去世之日,表達了人生命年歲的短少,而在這短少的日子中,又充滿了許多的急難、羞辱、死亡等威脅。 在這樣的苦情中,詩人向神禱告呼求,v2求主不要掩面、不要塞住耳朵。 其中用了許多生動筆法來形容人的苦情:v.3骨頭如火把燒著,v.4心好像草一般的枯乾,v.11日子好像草一般要消失,v.6又好像不潔淨的鵜鶘、鶚鳥等被人排斥,v.7好像最孤單的麻雀,被人撇棄,v.9我吃爐灰、喝眼淚;再再表達自己無法訴說的苦情。 B的部分: 而v.12,24b是轉捩點,從自己的短暫轉眼看見神的永恆。v.12「惟你─耶和華」、v.24「你的年數」,都提到了「萬代(1755)」,v.12神的名存到萬代,v.18後代人要記下並且讚美,因v.24神的年數世世無窮。人看自己是短暫痛苦的,但是看見神的永恆,生命開始有了新的座標、新的眼光、新的價值與智慧。 v.12神是永恆的,存到萬代,並且在永恆的權柄中,要重新憐憫、祝福錫安(v.13,16),v.14可憐聖殿的石頭。很可能此時已經國破家亡了,而詩人正為著國家哀慟時,卻轉眼看見神要重新地憐恤、可憐,並重新地建造,使神的榮耀回到錫安,這是詩人真正的盼望。 而vv.24b-27是更廣闊的眼光,不只是解決一個國家的問題,更是解決所有人類、受造物的問題:為什麼我們今天會在這麼多的眼淚、嘆息中?出路在哪裡? v.24神的年數世世無窮,v.25從創造的起點,神立的大地根基,神所創造的天,v.26而直接跳到世界的終了,有一天天地要滅沒,外衣漸漸舊了(這與熱力學第二定律相同,也與宇宙膨脹論相同),但是神沒有改變,並且神要重新再做一次創造的工作。 v.27這世界萬物都會改變,但是神永遠不改變,這一段偉大的時間論、永恆論,被新約希伯來書1:10-12引用,以此證明聖子的神性,成為希伯來書13:8的背景:來13:8 耶穌基督昨日、今日,一直到永遠,是一樣的。 C的部分: 認識神永恆的人,看人生有什麼不同的眼光呢?vv.18-22,28回答了這個問題。提到了後代、將來受造的民、子孫(v.28)、後裔,不只是我這一代有指望,而是世世代代、子子孫孫都有一個極大的盼望:v.18我們可以讚美神,v.21傳揚神、讚美神的話,v.22萬國要聚集事奉神,v.28長存堅立在神的面前。 自古至今,人在尋找永恆、尋找長生不老之術,從秦始皇派徐福去尋找蓬萊仙人;到春秋魯國大夫孫叔豹提到三不朽,追求精神上的永恆;到尼采、佛教涅槃的永恆觀等,沒有一種藥物與思想可以給人充滿動力的永恆觀,惟有認識永恆的主,我們的生命才能得著極大的喜樂與盼望,我們當好好把握現在,並過著盼望永恆的每一天。 五、反省問題: 我是否體會人生苦短?我的盼望在哪裡?神的永恆與我的盼望有什麼關係? 認識永恆的人,與眼中只有今天的人,在面對人生、後代有什麼不同的看法?什麼是永恆的盼望? ---------------- 講員: 貴格會合一堂 徐坤靖牧師 聖經之鑰-各卷書播放清單: https://thfc.pse.is/3epsdf 【聖經之鑰 相關資源】YouTube: https://thfc.pse.is/3cfams電子書: https://thfc.pse.is/3ccluu Powered by Firstory Hosting

Mark Ceilley of St. Paul has picture books on the mind, as a debut author himself. On a recent trip through Minneapolis-St. Paul International Airport, Ceilley discovered Picture Book Parade. The series features one picture book by a Minnesota author every two months through next August. Individual pages are blown up on panels so that visitors may feel they are stepping into the story. The book for this month and next is “In Search of the Gingerbread Man” by Timi Bliss, followed in 2023 by Peter Pearson's “How to Eat an Airplane,” Molly Beth Griffen's “Ten Beautiful Things,” Cheryl Minnema's book “Johnny's Pheasant” and Bao Phi's “A Different Pond.”   The exhibit is located between gates C18 and C19 at Terminal 1. Wendy Knox of Frank Theatre stopped at Prove Collective in Duluth to see Carolyn Sue Olson's work. Olson created a series of brightly colored oil pastel portraits of essential workers, beginning with the stay-at-home order in March 2020 and continuing through July 2021. The portraits have been on display in various locations around the state, but this is the first time all 100 have been exhibited together. Olson has put the complete series in a book, which is available through her website. The show runs until Nov. 18. Knox found encountering the entire collection a powerful experience. “You forget certain things that have happened” over the course of the pandemic, Knox said, “it is a little stroll down a not-pleasant memory lane, but it is really informative to see” what we've been through. Breanne Tepler, lead singer/songwriter of Breanne Marie and the Front Porch Sinners, loves the art and music scene in Duluth. This Thursday evening, she's heading to Wussow's Concert Cafe, which she calls “the heart and soul of the Duluth music scene,” for a concert and artist's opening reception. The artist is Annmarie Geniusz, a “wickedly talented” Duluth-based artist who creates joyful, whimsical creative works, from illustrations and murals to stained glass creations. Geniusz will be the November featured artist at Wussow's, and the artist's reception will feature local ska band, Woodblind, from 6 p.m. to 8 p.m. Tepler also says to check out the mural Geniusz painted next door Wussow's, at Zenith Bookstore.

The Whole View, Episode 461: Oats-M-G, Are These Actually Healthy? Welcome back to episode 461! (0:28) Stacy has mentioned in the past how great she feels when she includes some oats in her diet. She clarified that it's not something she loads up on daily. Finding high-quality, gluten-free, and non-glyphosate was a game-changer for her as well. For years, Stacy and Sarah have talked about how grains were problematic for digestion, proper absorption of nutrients, and gut health. However, it's important to revisit how we look at food with a scientific approach as often as possible. Both Stacy and Sarah's health journeys took a path through the Paleo Diet to a point where they are now very open to experimenting with foods they were not maligned to for a long time. For Sarah, that means revisiting the science of these foods to get away from the idea of grouping foods together and instead look at the pros and cons of individual foods and how they can fit into an overall healthy diet to empower others to experiment. Listener Question Sarah and Stacy have received many questions on this topic. This question in particular resonated the most: First off, I ADORE you both. I feel like you are my best friends and just don't know it yet :) I want to eat raw heads of lettuce (I've done this for years!) and get angry about poorly conducted research with Sarah, and go lift heavy weights while discussing clean beauty with Stacy. I can't get enough of you guys! So, my question is related to oats. Because my diet has been restrictive for so long while I've tried to heal my digestive system after being diagnosed with celiac three years ago, I have now developed anaphylactic responses to a lot of paleo foods. I was really happy this month when I was successfully able to reintroduce rice and oats, finding them to actually feel calming on my stomach. Because my list of paleo foods that I am reacting to keeps growing longer the more I rely on them, I am finding myself turning to these other foods like legumes, rice, and oats, on a daily basis in order to simply get enough food in. While I seem to tolerate it, it makes me nervous that I may be doing long term damage. I know you have discussed briefly how rice and some beans are actually not bad for the gut, but that was still always with the caveat of them being eaten "occasionally." And I don't think I've heard you discuss oats much at all.  Could you perhaps address the effect of GF oats on the microbiome, as well as the overarching effects of a diet that includes a lot of these "non paleo" foods, but in the context of an otherwise veggie and fruit rich diet with only healthy fats, high quality meat and organ meats, and no refined sugars?  Thank you! I love you both and am so thankful for all you do! - Anna Dogma of Paleo and Other Rigid Diets Stacy shares how much this question resonates with her. She first started on this health journey from guilt and feeding into the dogma of paleo that glorifies some foods and demonizes others. (8:00) In fact, what qualifies as a "paleo" food will vary from person to person. It's okay to lean into foods that make you feel good even if they don't fit a diet "framework" of what good and bad for you. For example, it was easy for Sarah to lump oats in with wheat and other grains and not critically evaluate it independently. Paleo has a very rigid set of rules, and it's important to Sarah to give listeners the tools and confidence needed to find what works best for them as individuals. Even if you stray from the framework of a particular diet but still identify with the term Paleo, there's nothing wrong with that. What works today might not work two years from now. It's okay to change to something better suited for what we need in a moment. Stacy stresses that they are not here to tell you what good, bad, and qualifies under a certain label.   Health Benefits of Oats Oats have been very well studied for at least 30 years and have some well-established health benefits. (16:27) Whole grain oat products have shone to positively impact coronary heart disease, diabetes, satiety/weight management, low glycemic index, and blood pressure.  These characteristics are mainly attributed to the high content of oat-specific beta-glucans, which are soluble food fibers. However, some people don't do well with oats. This is likely due to gluten cross-contamination. Gluten is a prolactin that is inflammatory that increases intestinal permeability, and is linked to adiposity signals. But avenin, prolamin type lectin, may not be as bad. Oats are often grouped in with wheat and barley as problematic, but not because oats themselves are a problem. Avenins are present at a lower concentration (10%–15% of total protein content) in oat as compared to gluten in wheat (80%–85%). The avenins in the genus Avena are free of the known CD immunogenic epitopes from wheat, barley, and rye. T cells that recognize avenin-specific epitopes have been found very rarely in CD patients. In fact, avenins are highly digestible and don't come out half-digested like gluten often does. Unlike gliadin peptides, oat peptides are also sensitive to digestion by pepsin, trypsin, and chymotrypsin in the gastrointestinal tract. They are unlikely to have any clinical relevance. Some cultivars of oats are gluten cross-reactors, meaning that if your body forms antibodies against gluten. But this appears to be a fairly low-frequency cross-reactivity and even not a problem for most people with celiac.   High Likelihood of Cross Contamination However, oats are often processed on the same wheat, making the potential contamination pretty high. For example, the U.S. specification for No. 1 oats allows the presence of up to 2% foreign material, which could be all wheat and barley! So yes, the biggest con of oats is that it's frequently contaminated with gluten. As we discussed on a recent show, upwards of 55% of people, have at least one gluten sensitivity gene.  Stacy references Oat O's and Thrive Market (not sponsored on this show!) for reduced risk of cross-contamination and what she personally leans on. For more information, Sarah recommends: frontiersin.org/articles/10.3389/fped.2019.00384/full tandfonline.com/doi/full/10.3402/fnr.v60.30324 journals.plos.org/plosmedicine/article?id=10.1371/journal.pmed.0010001 Oats Nutrient-Density Although oats fare better than other grains, they are nutritionally underwhelming per calorie. (26:50) A 100-gram (2/3 cup) serving of oats has a moderate glycemic load (37) but serves a fairly high (389) in calories. That serving of oats does provide an impressive 246% DV for manganese and 40-50% DV for vitamin B1, magnesium, and phosphorous. It also provides 20-30% DV of iron, copper, zinc.   Not too shabby... but not crazy impressive either. Sarah has been calculating Nutrivore Scores, and oats are the third-highest grain after corn and buckwheat. She adds that this is neither a pro nor a con for oats but shouldn't really be considered a "foundational" part of your diet, but rather fit in for added benefits.  Revisit 29:30 to figure out what Stacy is talking about with more context.   Oats and the Microbiome That same 100-gram serving contains a whopping 10.6 grams of fiber, including being particularly rich in a very special soluble fiber: mixed linkage beta-glucan. (34:31) This is a highly fermentable fiber that feeds anaerobic microbes in the gut and significantly increases the production of the short-chain fatty acids butyric and propionic acids. Oats contain 3-7% beta-glucan! Studies show over and over again that beta-glucans from oats significantly increase the growth of very important probiotic strains of Bifidobacterium as well as the Lactobacillus-Enterococcus group.   And oat beta-glucans seem to be better than beta-glucans from other grains. One study showed that an oat-based diet resulted in higher Bifidobacteria levels than either a barley-based or wheat-based diet supplemented with equivalent amounts of beta-glucan. In addition, these high levels of beta-glucan appear to be the "magic ingredient" behind oat's most famous properties, including their cholesterol-lowering effects.  This occurs because beta-glucan increases the viscosity of the chyme in the upper GI tract, leading to increased binding and secretion of bile acids.  In turn, plasma cholesterol becomes a substrate for newly formed bile, causing blood levels to decrease. Beta-glucans contribute to the maintenance of normal blood cholesterol levels. Consumption of beta-glucans from oats and barley as part of a meal (4 g/30 g carbohydrates) contributes to reducing the blood glucose rise after that meal (EU 432/2012). Oat grain fiber contributes to an increase in fecal bulk (EU 432/2012). Reducing saturated fat consumption contributes to the maintenance of normal blood cholesterol levels (EU 432/2012). Oat beta-glucan (3 g/day) has been shown to actively lower/reduce blood cholesterol. High cholesterol is a risk factor in the development of coronary heart disease (EU 1160/2011). In addition, oat fibers increase the fecal bulk, which contributes to a normal stool and positively impacts the functioning of the microbiome. Oats in particular also have high bioactive potential and relatively high lipid content relative to other grains. Their unique composition contributes some special benefits to gut health compared to other whole grains.   Surprise! Oats Are Super Digestible Protein content in the oat groat is relatively high (15%–20% by weight), as is the digestibility of oat protein (90%), and the majority (85%–90%) of oat protein consists of globulins. (43:15) This is in contrast to wheat, in which the vast majority consists of gluten.  Starch (about 60% of the total dry weight) is a major component of the oat grain. The amylopectin: amylose ratio is about 3, and the digestibility of oat starch is about 100%.  Oat starch digests slowly, partly due to high amounts of fiber and the high oil content in whole grain oat, which retards stomach emptying and improves digestion.  This results in a gradual supply of glucose to the intestine, which maintains a long feeling of satiety. As a result, whole grain oat foods have a low glycemic index (GI), which is advantageous in cases of diabetes and obesity. Compared to other cereals, oat grains have a relatively high oil content of on average 7% oil, but some varieties can have up to 18%: The three most abundant fatty acids are palmitic (C16:0; 20%), oleic (C18:1; 35%), and linoleic (C18:2; 35%) acids, which account for about 90%–95% of the total fatty acids.  The largest part is thus unsaturated, but there is more omega-6 than omega-3.  Alpha-linolenic acid (18:3, omega-3) is notably present in the oat germ.  The high lipid content can also hurt the sensory quality of oat products because of lipid oxidation, which produces fatty acid hydroperoxides and volatile aldehydes, causing rancidity.  Therefore, before further processing, oats are kilned—a high-temperature treatment to inactivate especially lipases.   So, Which Oats Are Best? Sprouted is best! Sprouting happens the seed is trying to turn itself into a plant. However, they are tough to sprout at home unless you buy unkilned oat groats. (48:35) The nutritional and bioactive properties and key enzymatic activities were studied in sprouted oat powder and compared with those of oat grain powder (control).  Sprouted oat powder was an excellent source of protein (10.7%), β-glucan (2.1%), thiamine (687.1 μg/100 g), riboflavin (218.4 μg/100 g), and minerals (P, K, Mg, and Ca).  This presented better amino acid and fatty acid compositions and levels of γ-aminobutyric acid (54.9 mg/100 g), free phenolics (507.4 mg GA/100 g), and antioxidant capacity (1744.3 mg TE/100 g) than control.  Enhanced protease and α-amylase and reduced lipase activities were observed in sprouted oat powder. Promising features to improve its nutritional, sensorial, and health-promoting properties! Since 2009 (EC 41/2009) and 2013 (FDA), oat products may be sold as gluten-free in several countries provided a gluten contamination level below 20 ppm. Note: 20ppm may still be too high for celiac or gluten allergy! Types of Oats Interestingly, when it comes to oats, the size of the flakes appears to make a difference in how the gut microbiota are affected.  One study found that thick oat flakes, but not thin oat flakes, increased Bifidobacterium levels during fermentation while also producing 2.5 times more butyrate than the thin oat flakes. The authors of this study suggested the resistant starch content of the thick oat flakes may explain these differences. Steel-cut oats have a little more resistant starch, slightly higher beta-glucans, and higher total fiber than old-fashioned rolled oats. Remember, old-fashioned rolled oats are better than quick oats.   One small study showed slightly higher butyrate production with steel-cut oats than old-fashioned, but it wasn't statistically significant.   Sarah recommends One Degree brands for the best-sprouted oats (not sponsored! Just a Sarah-approved favorite). Personally, Stacy and Sarah have both Old Fashion and Steel Cut oats currently in their pantries. Sarah recommends using an instant pot as well! Sarah's ideal recipe is 2 cups steel cut oats, 5-6 Cups of water (depending on how thick you like your porridge), and 1/2 a teaspoon of salt (scale up or down as desires). In Instapot, cook for 4 minutes on high pressure and then let the pressure naturally decline. Add flavoring as needed, such as maple syrup to slightly sweeten. Stacy also recommends trying this pumpkin oatmeal recipe, which has always been a crowd-pleaser.   How to Experiment  Sarah recommends starting by utilizing a food journal to see if you're reaching your nutrient targets for the day. If you are, first cut out the food from your diet for at least two weeks, even including trace amounts. This ensures you're testing your actual reaction and not the reaction you're currently having while already consuming the food. This helps you isolate and identify. Next, pick a day to challenge when you're feeling good, slept well, not stressed, and not eating anything else out of the ordinary. If you're worried about an allergy response, make sure you go slow since they can get really big very fast. If you check for sensitivity, like mood, stomach, or skin issues, it's okay to go more all-in. If you're not sure if something was a reaction (mild reactions can be tough), give yourself a few days to reset, and then try again to see if you have the same experience. If you're thinking about experimenting with oats, Sarah recommends buying sprouted oats (which reduces anti-nutrient content) that are certified gluten-free.  Be sure to follow a methodical reintroduction protocol!  Also, oats are way less likely to be tolerated by people with celiac disease (and potentially other autoimmune diseases) or anyone who has tested positive for wheat/gluten or oats on a food allergy/intolerance screen in the past.    Final Thoughts Sarah views oats as a "sometimes food" due to the importance of quality. (1:01:30) Stacy's philosophy is all about variety and introducing foods to develop a palate. And the more science we look at, the more we can emphasize foods we know are beneficial to our gut and health overall. It's important not to emphasize "yes" and "no" in terms of food, but instead look at each food as an individual in terms of a healthy diet overall. Identifying your triggers is very important, and knowing that not every food will work for every person. As interest in oligonucleotide therapeutics increases, there is an ever-growing need for analytical techniques that can properly analyse and quantitate these complex biomolecules. Mass spectrometry (MS) is required to analyse impurities, degradants, and other biological/chemical modifications and is the method of choice for the analysis of synthetic oligonucleotides. Sixfold Bioscience, in collaboration with the National Measurement Laboratory (NML) have been using the Agilent AdvancedBio 6545XT LC/Q-TOF to develop a method to analyse complex oligonucleotides of various lengths with volatile ion pair reagents. This method is used for quality control of oligonucleotides that are subsequently assembled into Mergos™ for use in in vivo applications. In addition to the analytical capabilities currently being utilised by Sixfold, the Agilent 1290 Infinity II Preparative system is also being used for oligonucleotide purification. Both reverse-phase (C18) and anion-exchange (IEX) chromatography is regularly carried out to assist in Sixfold's purification workflow. As a result of this work, and as an extension to the work currently being carried out by the Tate group at Imperial College, a Metabolomics group is in the process of being formed at the Molecular Sciences Research Hub. This group will aim to solve challenges related to the analysis of biomolecules and will take advantage of the combined know-how and expertise of scientists at Imperial College, Sixfold Bioscience and Agilent. Abstract: Lipid-based carriers of the hydrophobic drug paclitaxel (PTX) are used in clinical trials as next-generation agents for cancer chemotherapy. Improving the loading capacity of these carriers requires enhanced PTX solubilization. We compared the solubility of PTX in cationic liposomes (CLs) with lipid tails containing one (oleoyl; C18:1 {Delta}9 DOTAP/DOPC) or two (linoleoyl; C18:2 {Delta}9 DLinTAP/DLinPC) cis double bonds with newly synthesized cationic DLinTAP (2,3-dilinoleoyloxy-propyl-trimethyl-ammonium methylsufate). We used differential-interference-contrast microscopy to directly observe PTX crystal formation and generate kinetic phase diagrams representing the time-dependence of PTX solubility as a function of PTX content in the membrane. Replacing tails bearing one cis double bond (DO lipids) with those bearing two (DLin lipids) significantly increased PTX membrane solubility in CLs. Remarkably, 8 mol% PTX in DLinTAP/DLinPC CLs remained soluble for approximately as long as 3 mol% PTX (the membrane solubility limit which has been the focus of most previous fundamental studies and clinical trials) in DOTAP/DOPC CLs. The large increase in solubility is likely caused by enhanced molecular affinity between lipid tails and PTX upon replacement of oleoyl by linoleoyl tails, rather than by the transition in membrane structure from lipid bilayers to inverse cylindrical micelles observed in small-angle X-ray scattering. Importantly, the efficacy of PTX-loaded CLs against human prostate cancer (PC3) cells from measurements of the IC50 of PTX cytotoxicity was unaffected by changing the lipid tails, and toxicity of the CL carrier alone was negligible. Moreover, efficacy was approximately doubled against human melanoma (M21) cells for PTX-loaded DLinTAP/DLinPC over DOTAP/DOPC CLs. The findings demonstrate the potential of chemical modifications of the lipid tails to increase the PTX membrane loading well over the typically used 3 mol% while maintaining (and in some cases even increasing) the efficacy of CLs. The increased PTX solubility will aid the development of liposomal PTX carriers that require significantly less lipid to deliver a given amount of PTX, reducing side effects and costs. Robinson, aka "The Olive Oil Hunter ®" who is one of the world's most respected authorities on all matters olive oil.   Known for his “platinum palate,” he is one of the few Americans invited to serve as a judge in prestigious Italian olive oil tasting competitions.   These days he is dedicated to importing rare fresh-pressed olive oil via , the most flavorful and healthiest extra virgin olive oil on the planet, until now virtually impossible to obtain year-round in the U.S. All his oils are independently lab tested and certified for 100% purity. New Generation Thinker Alun Withey on what made 18th-century men shave off centuries of manly growth. Recorded before an audience at the York Festival of Ideas. You can hear audience questions from the event as an episode of the BBC Arts&Ideas podcast. To be clean-shaven was the mark of a C18 gentleman, beard-wearing marked out the rough rustic. For the first time, men were beginning to shave themselves instead of visiting the barber, and a whole new market emerged to cater for rising demand in all sorts of shaving products - soaps, pastes and powders. But the way these were promoted suggests there was confusion over exactly what the ideal man should be. On the one hand, razor makers appealed to masculine characteristics like hardness, control and temper in their advertisements whilst perfumers and other manufacturers of shaving soaps, stressed softness, ease and luxury. So enter the world of Georgian personal grooming to discover the 18th century's inner man. Alun Withey lectures in the Centre for Medical History at the University of Exeter and is a Wellcome Research Fellow and a BBC/AHRC New Generation Thinker. He has edited an essay collection on the history of facial hair (Palgrave), curated a photographic exhibition of Victorian beards in the Florence Nightingale Museum in London and has written for BBC History Magazine and History Today. As life expectancy has risen steadily over the last years and diseasefree aging is more and more challenging, understanding the complexity of age and aging is of great importance. Metabolomics is one of the novel approaches in systems biology with high potential to deliver answers to these questions. However, only a few metabolic studies based on large samples are available so far. In this thesis, I present results from two population-based studies, the German KORA Follow-Up 4 (KORA F4) study as a discovery cohort with a sample of 1,038 female and 1,124 male healthy participants (32–81 years) and 724 healthy females from UK Adult Twin Registry (TwinsUK) as replication. Targeted metabolomics of fasting serum samples with flow injection analysis coupled with tandem mass spectrometry (FIA-MS/MS) positively quantified 131 metabolites after stringent quality control. Among these, 71 and 34 metabolites were significantly associated with age in females and males, respectively, after adjustment for body mass index (BMI), which is highly correlated (r=0.9) with age. These results indicate that metabolic profiles are age dependent and sex specific. Then, a set of the 12 most age-discriminative, independent metabolites was identified in women with an approach based on random forest and stepwise variable selection. This set showed highly significant differences between subjects aged 32–51 years and 52–77 (p-values range 1.3E-09 – 1.9E-46, significance threshold p=0.004). Ten out of these 12 metabolites replicated in unrelated females from the TwinsUK study, including five metabolites the concentrations of which increased with age (C12:1, C18:1, sphingomyelin (SM) C16:1, SM C18:1 and phosphatidylcholine (PC) aa C28:1), while histidine decreased gradually. Three glycerophospholipids (PC ae C42:4, PC ae C42:5, PC ae C44:4) showed declines around the age of 51 years. Meta-analysis of both studies gave virtually the same results as KORA alone. These observations might reflect many different processes of aging such as incomplete mitochondrial fatty acid oxidation, counteracting oxidative stress, and changes in vascular functions. The identification of these ten age-related metabolites should help better understand aging pathways and networks and with —more discoveries in the future— eventually help enhance healthy aging and longevity.

Patients with phenylketonuria (PKU) have to follow a lifelong phenylalanine restricted diet. This type of diet markedly reduces the intake of saturated and unsaturated fatty acids especially long chain polyunsaturated fatty acids (LC-PUFA). Long-chain saturated fatty acids are substrates of mitochondrial fatty acid oxidation for acetyl-CoA production. LC-PUFA are discussed to affect inflammatory and haemostaseological processes in health and disease. The influence of the long term PKU diet on fatty acid metabolism with a special focus on platelet eicosanoid metabolism has been investigated in the study presented here. 12 children with PKU under good metabolic control and 8 healthy controls were included. Activated fatty acids (acylcarnitines C6-C18) in dried blood and the cholesterol metabolism in serum were analyzed by liquid chromatographic tandem mass spectrometry (LC-MS/MS). Fatty acid composition of plasma glycerophospholipids was determined by gas chromatography. LC-PUFA metabolites were analyzed in supernatants by LC-MS/MS before and after platelet activation and aggregation using a standardized protocol. Patients with PKU had significantly lower free carnitine and lower activated fatty acids in dried blood compared to controls. Phytosterols as marker of cholesterol (re-) absorption were not influenced by the dietary fatty acid restriction. Fatty acid composition in glycerophospholipids was comparable to that of healthy controls. However, patients with PKU showed significantly increased concentrations of y-linolenic acid (C18:3n-6) a precursor of arachidonic acid. In the PKU patients significantly higher platelet counts were observed. After activation with collagen platelet aggregation and thromboxane B(2) and thromboxane B(3) release did not differ from that of healthy controls. Long-term dietary fatty acid restriction influenced the intermediates of mitochondrial beta-oxidation. No functional influence on unsaturated fatty acid metabolism and platelet aggregation in patients with PKU was detected.

Ziel der vorliegenden Studie war die quantitative und qualitative Bestimmung der Gesamtfettsäuren in bovinen Serumproben sowie die Darstellung der Konzentrationsverläufe der einzelnen Fettsäuren während des peripartalen Zeitraums. Des Weiteren sollte festgestellt werden, ob zwischen Kühen, die postpartal an einer Gebärmutterentzündung erkranken, und gesunden Tieren Unterschiede in den Konzentrationen einzelner Fettsäuren bzw. Fettsäuregruppen vorliegen. Die in dieser Studie verwendeten Serumproben stammen von 50 Kühen eines Milcherzeugerbetriebes in Brandenburg. Von den ausgewählten Tieren zeigten 25 Tiere während des Untersuchungszeitraums Symptome einer klinischen Gebärmutterentzündung (puerperale Metritis, klinische Metritis, klinische Endometritis oder Pyometra) und wurden demnach als uteruskrank eingestuft. Die anderen 25 Tiere wiesen keine gesundheitlichen Störungen auf und wurden als uterusgesund eingestuft. Von jedem Tier wurden während des Zeitraums 17 Tage vor bis 28 Tage nach der Kalbung sechs bzw. sieben Serumproben gewonnen. Für die quantitative und qualitative Analyse der Gesamtfettsäuren in den Serumproben wurden die Fettsäuren zu Fettsäuremethylestern (FAME) derivatisiert. Die Probenanalyse wurde mit einem Gaschromatographie/Massenspektrometrie-System (GC/MS-System) durchgeführt. Außerdem wurden die Konzentrationen der freien Fettsäuren (NEFA) in den Serumproben mittels Enzym-Test bestimmt. Sowohl vor wie auch nach der Kalbung konnten bei verschiedenen Fettsäuren Konzentrationsunterschiede zwischen uterusgesunden und uteruskranken Tieren festgestellt werden. Die Konzentrationen der gesättigten Fettsäuren Octadecansäure (C18:0) und Tetradecansäure (C14:0) zeigten in beiden Tiergruppen nach der Kalbung einen leichten Anstieg, wobei die Werte der gesunden Kühe ab dem elften Tag p.p. signifikant höher lagen als bei erkrankten Kühen (P

The aim of this doctoral thesis was to investigate the factors relevant in plant interaction of two plant growth promoting rhizobacteria (PGPR). For this, the strain Acidovorax sp. N35 isolated from surface sterilized wheat roots and the two strains F4 and F7 of Rhizobium radiobacter, a bacterium associated with the plant growth promoting fungus Piriformospora indica, were chosen. First of all, the isolate N35 was characterized using phylogenetic and taxonomic methods. The 16S rRNA gene sequence analysis showed that strain N35 has the closest sequence similarities (98.2, 98.5 and 99.0 %) to the environmental Acidovorax species A. delafieldii, A. facilis and A. defluvii. The DNA-DNA hybridization values clearly separated the isolate from these three species. Additionally, phenotypic properties, such as substrate metabolization profiles as determined by a Biolog GN2 assay and cell wall fatty acid profiles concerning the fatty acids C16:0, C16:1ω7cis/trans, C17:0cyclo and C18:0cyclo and C19:0cyclo, facilitated the differentiation of the newly isolated strain N35 from its closest relatives. Thus, the strain N35 was classified as representative of a new species within the genus Acidovorax, and the name Acidovorax radicis sp. nov. is suggested. “Cand. A. radicis” N35 undergoes an irreversible phenotypic variation, resulting in different colony shapes on agar plate. In soil system, both phenotypes showed a plant growth promoting effect both on barley roots and shoots. The wild type N35 (rough colony type) had a better plant growth promoting effect on barley in comparison with phenotype variant N35v (smooth colony type). Wild type and phenotype variant cells of “cand. A. radicis” N35 were labeled with GFP and/or YFP and their separate and co-colonization behavior was investigated in a monoxenic system and a soil system using a CLSM for detection. Both types of N35 could endophytically colonize barley roots after 12 weeks inoculation in the soil system. Competitive root colonization behavior was observed after co-inoculation with differentially labeled wild type N35 and phenotype variant N35v bacteria, where the wild type showed dominant colonization of barley roots compared to the phenotype variant. Moreover, the variant N35v lost its motility due to missing flagella and swarming ability. The differences of both types at genetic level were investigated using whole genome sequence data obtained from 454 pyrosequencing (Roche) using the GS FLX Titanium chemistry. As only difference in the genome sequence, a 16 nucleotides deletion was identified in the mutL gene, which encodes for the mismatch repair protein MutL. In phenotype N35v, the frame shift caused by this deletion leads to the formation of a stop codon in the coding gene, resulting in a truncated MutL protein with a missing functional MutL C-terminal domain. This mutation occurred in exactly the same way in all investigated phenotype variants. These results suggest that MutL might be directly or indirectly responsible for the phenotypic variation in “cand. A. radicis” N35. Quorum sensing signaling molecules produced by “cand. A. radicis” N35 were identified using biosensors as well as Fourier transform ion cyclotron resonance - mass spectrometry (FT-ICR-MS) and ultra performance liquid chromatography (UPLC). Both types of “cand. A. radicis” N35 possess the same AraI/AraR quorum sensing system, which belongs to the LuxI/LuxR type. The two N35 phenotypes produced nearly the same amount of 3-OH-C10-HSL in the exponential growth phase. A co-inoculation experiment of AHL producing wild type N35 and a constructed AHL negative mutant N35 ΔaraI showed that wild type N35 had a dominant colonization behavior compared to the AHL negative mutant on barley roots in a monoxenic system. These data indicate that quorum sensing is involved in regulation of root colonization by “cand. A. radicis” N35. The second examined PGPR, R. radiobacter, which occurs naturally as endofungal bacterium in the plant growth promoting fungus P. indica, was demonstrated to colonize the surface of barley roots with fluorescence in situ hybridization (FISH) in a monoxenic system. The interaction of P. indica harboring R. radiobacter with other rhizobacteria was investigated using plate confrontation assays. Antibiotics and lipopeptides produced and excreted by the plant growth enhancing rhizobacterium Bacillus amyloliquefaciens FZB42 and the biocontrol rhizobacterium Pseudomonas fluorescens SS101 were shown to be responsible for the observed inhibition of P. indica by these bacteria. R. radiobacter F4 and F7 were able to synthesize a variety of oxo- and hydroxyl-C8- to C12-HSL compounds. In addition, both strains also produced coumaroyl-HSL when coumaric acid was supplied in the medium. The lactonase expressing transformants F4 NM13 and F7 NM13, which are the AHL negative phenotypes, abolished the lipase and siderophore activity. Considering this, quorum sensing influences the production of metabolites including lipase and siderophores in R. radiobacter F4 and F7. Further work should be directed to the question whether quorum sensing also plays a role in the interaction of the bacterium with fungus and/or plant.

Background/Aim: Previous studies in vitro and in animals in vivo found that alpha-linolenic acid (C18:3 omega 3) may enhance oxidative damage of essential amino acids. We investigated whether the addition of the long-chain polyunsaturated fatty acids (LCPUFA) arachidonate (C20:4 omega-6; AA) and docosahexaenoate (C22:6-omega 3; DHA) in the form of egg phospholipids to infant formula affects plasma amino acid concentrations and indices of protein metabolism in term infants. Methods: In a double-blind, randomized clinical trial, healthy infants were fed from day 5 of life formula with or without preformed LCPUFA (n = 10 and 12, respectively). At the age of 5 days and 1, 2, 3 and 4 months, blood samples were obtained and analyzed for plasma amino acids by high-performance liquid chromatography and for plasma phospholipid fatty acid composition by gas chromatography. Results: At the age of 3 months, plasma threonine concentrations were significantly lower in infants receiving dietary LCPUFA than in controls (124 +/- 16 vs. 216 +/- 28 mu mol/l, p < 0.05). Values of other plasma essential amino acids, total protein, albumin, creatinine and urea nitrogen did not differ between the two feeding groups throughout the study. At the age of 5 days, plasma phospholipid AA and DHA concentrations were inversely correlated with histidine concentrations (AA: r = -0.60, p = 0.01; DHA: r = -0.53, p < 0.05). At the age of 3 months, DHA concentrations were inversely related to plasma histidine, methionine and threonine concentrations (r = -0.66, -0.62, and -0.64, respectively, p < 0.05). Conclusions: The dietary LCPUFA supplementation of infant formula used in this study has no adverse effects on infant plasma amino acid concentrations and indicators of protein metabolism. Nonetheless, the apparent interaction of LCPUFA with some amino acids in formula-fed infants warrants further investigation.

Patients with glucocorticoid-remediable aldosteronism (GRA) possess a chimeric gene resulting from fusion of the genes encoding steroid aldosterone synthase and 11 beta-hydroxylase. In the adrenal zona fasciculata, this may lead to ectopic expression under ACTH control of aldosterone synthase activity and increased formation of cortisol C18 oxidation products. We assessed mineralocorticoid and glucocorticoid pathways in three patients with GRA. Baseline plasma progesterone, 17 alpha-hydroxyprogesterone, corticosterone, and cortisol were normal in all patients, whereas 11-deoxycorticosterone, aldosterone, and 11-deoxycortisol were above normal. The ratios of both corticosterone/11-deoxycorticosterone and cortisol/11-deoxycortisol were abnormally low, and decreased further 60 min after administration of ACTH-(1-24) (250 micrograms) as an i.v. bolus. A low corticosterone/11-deoxycorticosterone ratio is consistent with an increased aldosterone synthase activity forming aldosterone by corticosterone. Similarly, a decreased cortisol/11-deoxycortisol ratio could reflect enhanced cortisol C18 oxidation. Our findings are in agreement with a hyperfunction of the 11 beta-hydroxylase/aldosterone synthase complex in the adrenal zona fasciculata of GRA induced by the new chimeric gene.
