Abstract: Altered brain network activity and the resulting hypersynchrony are important for the pathogenesis of cognitive decline in Alzheimer's disease (AD) mouse models. Treatments that reduce epileptiform discharges (EDs) or network hyperactivity improve cognition in AD models and humans. We first show that ketogenic diet, but not fasting, rapidly and persistently reduced EDs in the hAPPJ20 Alzheimer's mouse model over timescales of hours to months. Then, to identify the specific mechanism of the pleiotropic ketogenic diet, we developed small molecule ketone esters to deliver ketone bodies pharmacologically. Two ketone esters recapitulate ED suppression without other dietary manipulation, over time scales of minutes to one week. This small molecule rescue was associated with reduced low-frequency oscillatory activity similar to the recently reported mechanism of an NMDA receptor modulator molecule in this model. Long-term KD resulted in cognitive improvement and in a sex-stratified analysis also improved survival in the more severely affected hAPPJ20 males. Agents that deliver ketone bodies via small molecules or act on downstream targets may hold therapeutic promise in AD through the mechanism of improved network function and reduced epileptiform activity. Let's dive into the "Sports Protocol": SUGGESTED SERVING SIZE Each "serving" is technically 60ml of KE4 (30g of the Ketone Ester), but do not start with a full bottle. More is NOT always better. More may lower blood glucose too much, lower top speed, and reduce performance substantially. It is based on weight, so if you weight 2x the average person, you may need more. And if you are lightweight, you will need much less. Numbers below are based on 70 kilos. Start in practice, NOT in a race. (Updated March 2021) Micro dosing KE4: (Micro dosing is a fancy word for taking smaller amounts of KE4) If using KE1 (compare) and adjust. 1 bottle of KE1 is = to about 2 capfuls of KE4) Pre-Workout: Start here. (All Athletes, regardless of diet) Regardless of your diet.A) Full Keto & fat adapted B) New to keto or intermittent fasting, or C) Full carb/western diet. Step 1: Empty stomach (at least 2-3 hours, if not overnight). This really is key. Without this, you may have to take more or do the dual fuel protocol with carbs and it still not be better. Why? (see this video) and more details on Fasted Workouts here Step 2: MOST COMMON: 1 to 3 capfuls of KE4 (same as 1/2 to 1.5 bottles of KE1) about 15 min before warmup or workout. * 1-2 capful for females or much lighter males, * 2-3 capfuls for NON keto males. * 1-4 capfuls for keto males, (Keto athletes find benefit at lower amounts, BUT some are able to take more without the blood sugar dropping effect that non keto people get.) Warning, if you take it too early like 30-60 min and don't use the fuel, you may get tired (i.e. blood sugar crash). Some sprinters have taken it 45 min before without issue. Just test in practice the EXACT protocols you plan to use. Note that glucose is not necessary (and could be worse) with this small amount since blood glucose does not drop too much with micro dosing. Glucose also causes inflammation, it is a dirty fuel with lactic acid as a byproduct (ketones create zero lactic acid). The theory here is micro dosing may trigger endogenous ketone production (where your body makes ketones), whereas more may hault that production or it can also drop blood sugar too much. Some may stick with this protocol and not move up in quantity. Play around with different amounts Then try 15ml up to 25ml of KE4 (again, until you reach 30ml, you may not need carbs.) If you do notice a difference with 10-15ml, some may drop to 5ml to see how that works. I wouldn't start with 5ml because it may be too subtle, but you can if you feel the energy is still available. Tips: Don't go out faster expecting a better final result. Go out as normal as possible and then later in the practice you can increase your speed if you want. Brain aging is inevitable, but not uniform; every brain ages differently. Are there lifestyle and nutritional interventions we can follow to reduce the rate of decline? A recent study found that those who followed a ketogenic and low-carb diet did indeed see positive results that we can attribute to preventing (perhaps even reversing) cognitive decline as they age. Fascinatingly, even those who followed a standard American diet found that simply ingesting an exogenous ketone ester (H.V.M.N. Ketone Ester) resulted in similar, positive neurological effects. This study, led by Lilianne R. Mujica-Parodi, PhD, and published in PNAS, is titled "Diet modulates brain network stability, a biomarker for brain aging, in young adults". When interviewed about her findings, she is quoted: "...as people get older, their brains start to lose the ability to metabolize glucose efficiently, causing neurons to slowly starve, and brain networks to destabilize. Thus, we tested whether giving the brain a more efficient fuel source, in the form of ketones, either by following a low-carb diet or drinking ketone supplements, could provide the brain with greater energy. And with that has come a bunch of ways to "hack" your way into ketosis so you can (in theory) have your cake and eat it too! One of such hacks are exogenous ketones. So, the whole point of following a ketogenic diet is to use your body's fat reserves as energy. Basically, your liver converts your fat into energy which your cells can utilize instead of glucose. This process generates ketone bodies (simply referred to as ketones) which fuel your cells. The problem is that most of us become carb-dependent very early on in our lives. So when we decide to transition and switch from carbs to fat for fuel, it is not a smooth transition. It usually takes time and a very strict low carb diet for us to become fat adapted. And during this period of time we might suffer of the "keto flu". The keto flu is simply a collection of flu-like symptoms that may occur to people who are transitioning to a fat adapted metabolism. It usually resolves with a few modifications to our diets such as adding salt and drinking more water. Unsurprisingly, many people give up the keto diet at this point and never get to experience true ketosis. Enter exogenous ketones. By supplementing your body with the ketones it needs while it reaches the point of ketosis by diet alone can be a very effective way of getting you fat-adapted. The problem came when some companies decided to start marketing their exogenous ketones for weight loss and as a way for you to be able to eat all the carbs you wanted while staying in ketosis. This is not possible. Or healthy. And to add to the mix, it turns out that not all the ketones are created equal. There's ketone salts and there's ketone esters. Complete Show Notes

One of the greatest challenges of training is successfully attaining "functional overreaching". Pushing your maximal capacity results in stress that leads to beneficial adaptations...making your body stronger, faster, & better. What happens when you do it wrong? Aptly named, "non-functional overreaching" occurs. Nothing will help besides rest and recovery. In the case of "overtraining", the more serious form of overreaching, it might take months for even the most elite athletes to bounce back. A recent study found promising signs that ketone esters can alleviate overreaching symptoms. The researchers didn’t necessarily look at how ketones acutely influence exercise performance, but rather how consistent supplementation could affect recovery and training capacity. The lead researcher, Professor Peter Hespel, described the potential effect of ketones in recovery as “unseen” and “many times greater” than what he’s seen with other nutritional supplements. And many athletes have resorted to the morning supplement cocktail believing it will make them better cyclists. But there’s a dark side. Those cocktails can actually hurt performance, certainly affect health, and lead to even darker, ethically-challenged places. Today, we’re going to talk about supplements and our concerns with them, and then cover a few foods that actually do work. We’ll discuss: - We thought about bashing all the supplements that don'’t work, but then realized we only have an hour. So instead, Trevor will read a description of every supplement that does work. If you have questions about the Keto diet...go no further. Our guest today is pretty much the Keto science guy. Dr Dominic D'Agostino is a professor in molecular pharmacology and physiology at the University of South Florida Morsani College of Medicine as well as a Visiting Research Scientist at the Florida Institute for Human and Machine Cognition. Dr D'Agostino tells us how he found his way into the filed of ketosis. He combined interests in nutritional and neuro sciences. From there, Dom developed a deep expertise in underwater medicine and oxygen toxicity seizures where he made the connection with ketones and the brain. What is Nutrtional Ketosis Ketones - more specifically ketone bodies- are chemical produced by our livers. Ketone bodies are produced by avoiding most carbohydrates in the diet (eg sugars, breads, fruits, and certain vegetables). A much higher percentage of healthy fats are eaten during a ketogenic diet. When someone follows this diet the body begins to make and utilize ketones more so than on it's typical energy source - glucose. Nutritional ketosis is the sustained elevation of ketones in the blood. To be clear, as long as someone isn't completely lacking insulin (eg Type 1 diabetic), this low-level of ketosis is NOT the dangerous state of ketoacidosis which develops from having a lack of insulin. What does it feel like once you are in Ketosis and what are the Benefits of Ketosis Many people feel more cognitively alert and more even brain energy. Lower hemoglobin A1c Dom explains the synergy between intermittent fasting (caloric restriction) and low carbohydrate/modified ketogenic diet. We discuss the evidence or more specifically the lack of evidence for the Ketogenic diet in Alzheimer's and cognitive decline and where the research is headed. Late in 2017, a San Francisco startup company brought one of the commercial ketone esters to market. Today’s episode features an interview with a scientist and world-class athlete who has spent the past year helping develop and rollout HVMN Ketone, an FDA-approved drink that promises increased athletic ability as well as heightened focus and energy. Dr. Brianna Stubbs earned her PhD in biochemical physiology from Oxford University in 2016 where she researched the effects of ketone drinks on elite athletes. During Brianna’s collegiate athletic career, she won two gold medals while representing Great Britain at the World Rowing Championships. She first made international news when as a 12-year-old she became the youngest person ever to row across the British Channel. Brianna graduated from Oxford’s Pembroke College with a BA in preclinical sciences with the idea of becoming an MD. But after spending a year working as a research assistant helping to investigate the effect of exogenous ketones on human performance, she decided instead to pursue her doctorate in biochemical physiology and investigate how ketone compounds might be applied in a sporting and healthcare setting in the future. While at Oxford, she worked alongside Dr. Kieran Clarke to develop a novel ketone monoester that has been shown to improve exercise performance in endurance athletes. She also was a member of the Great Britain Rowing Team and in 2016 become the World Champion in the lightweight guadruple sculls. Brianna’s time at Oxford gave her a unique opportunity to combine her scientific interest in sports physiology and metabolism while also competing at an international level. Dr. Veech's solution was something called "ketone esters", which are extremely expensive, but, according to him, a far more natural, safe and healthy way to quickly get the body into ketosis - for anything from managing medical conditions, to improving cognition, to increasing endurance and beyond. In the video below, I actually experimented with these myself and, as you can see, the results were astounding... After shooting this video, I took another bottle of ketone esters to a Tough Mudder event and recorded a crazy video on Snapchat during which I elevated both blood glucose by drinking 75 grams of pure glucose and elevated blood ketones by drinking a bottle of the ketone salts, and the results were equally as astounding, resulting in one of the fastest races of my life. But the question remains: are these fancy, expensive ketone esters actually far better than ketone salts? Are ketone salts, which is what most people use, actually going to result in long term health issues? In today's podcast, I bring on ketosis expert Dr. Dominic D’Agostino to get his opinion.
