I talk with Claire Breedlove, a licensed clinical social worker who has a lot of experience treating OCD, especially when it comes to religious themes. We discuss what scrupulosity is, how it can impact missionaries and young adults, and tips for managing anxiety. Claire also shares about effective treatment options like exposure therapy. Abstract: Identified across multiple psychiatric disorders, the dopamine (DA) transporter (DAT) Ala559Val substitution triggers non-vesicular, anomalous DA efflux (ADE), perturbing DA neurotransmission and behavior. We have shown that DAT Val559 mice display a waiting impulsivity and changes in cognitive performance associated with enhanced reward motivation. Here, utilizing a within-subject, lever-pressing paradigm designed to bias the formation of goal-directed or habitual behavior, we demonstrate that DAT Val559 mice modulate their nose-poke behavior appropriately to match context, but demonstrate a perseverative checking behavior. Although DAT Val559 mice display no issues with the cognitive flexibility required to acquire and re-learn a visual pair-wise discrimination task, devaluation of reward evoked habitual reward seeking in DAT Val559 mutants in operant tasks regardless of reinforcement schedule. The direct DA agonist apomorphine also elicits locomotor stereotypies in DAT Val559, but not WT mice. Our observation that dendritic spine density is increased in the dorsal medial striatum (DMS) of DAT Val559 mice speaks to an imbalance in striatal circuitry that might underlie the propensity of DAT Val559 mutants to exhibit compulsive behaviors when reward is devalued. Thus, DAT Val559 mice represent a model for dissection of how altered DA signaling perturbs circuits that normally balance habitual and goal-directed behaviors. Francine Glasser is a Clinical Social Worker and Therapist who recently published a memoir, "Shards of Glass: A Kaleidoscopic Life's Memoir," about her experiences as a child and how she took a path to healing from her difficult experiences.A bit about her book: Because her family constantly had to flee the repercussions of her father's gambling addiction, Francine Glasser was consigned to a childhood devoid of connection and marked by loss after loss of everything she cherished. Friends, relatives, pets, and possessions were left behind each time her family ran from debt collectors, sometimes in the middle of the night, and left town for yet another hopeless new start. For Francine, painful memories of those rootless years were like the fragments of color, light, and darkness in a kaleidoscope. But years later, clinging to her love for the father responsible for her lost childhood, those shards of glass would reorganize into patterns of forgiveness, stability, beauty, and peace.This book was conceived long ago, because Francine had always wanted to tell the story of her search for a forever home. Years later, as a practicing psychotherapist and social worker her life was invested in hearing the stories and traumas of those whom she counseled and helped make peace with their past. Abstract: Obsessive-Compulsive Disorder (OCD), characterized by intrusive thoughts (obsessions) and repetitive behaviors (compulsions), is associated with dysfunction in fronto-striatal circuits. There are currently no fast-acting pharmacological treatments for OCD. However, recent clinical studies demonstrated that an intravenous infusion of ketamine rapidly reduces OCD symptoms. To probe mechanisms underlying ketamine's therapeutic effect on OCD-like behaviors, we used the SAPAP3 knockout (KO) mouse model of compulsive grooming. Here we recapitulate the fast-acting therapeutic effect of ketamine on compulsive behavior, and show that ketamine increases activity of dorsomedial prefrontal neurons projecting to the dorsomedial striatum in KO mice. Optogenetically mimicking this increase in fronto-striatal activity rescued compulsive grooming behavior in KO mice. Conversely, inhibiting this circuit in wild-type mice increased grooming. These studies demonstrate that ketamine increases activity in a fronto-striatal circuit that causally controls compulsive grooming behavior, suggesting this circuit may be important for ketamine's therapeutic effects in OCD.   We spend a lot of time and money on the treatment of chemical addictions but often not enough on the subtler process addictions; things like compulsive gambling, spending, hoarding, or eating, and least of all on compulsive sexual behavior.  Process addictions don't come to our attention often until they are well beyond crisis because they are so neatly masked by the notion of normalcy.  But like it's cousin alcoholism, sexual addiction fills it's sufferers with shame, ruins lives and destroys relationships.  Join us tonight to hear a story of hope and recovery from sexual addiction.
