Fisetin, a Natural Compound, Helps Prevent Artery Hardening from Aging and Kidney Disease

BUFFALO, NY — May 6, 2025 — A new #research paper was #published in Aging (Aging-US) Volume 17, Issue 4, on April 2, 2025, titled “Fisetin ameliorates vascular smooth muscle cell calcification via DUSP1-dependent p38 MAPK inhibition.” In this study, researchers at Johannes Kepler University Linz found that fisetin, a natural substance found in fruits and vegetables, helps protect blood vessels from hardening, which is a common problem in older adults and people with kidney disease. This discovery highlights fisetin's potential to prevent vascular calcification and reduce cardiovascular damage caused by aging and chronic kidney disease. The research, led by first author Mehdi Razazian and corresponding author Ioana Alesutan, focused on vascular calcification—a condition in which blood vessels stiffen due to calcium deposits. This process is common in aging and chronic kidney disease and increases the risk of heart attacks and strokes. Using human and mouse study models, the researchers tested fisetin's ability to prevent this calcification in vascular smooth muscle cells (VSMC), which play a key role in maintaining vessel health. Fisetin, known for its anti-inflammatory and antioxidant properties, significantly reduced calcium buildup and calcification markers under stress conditions that mimic disease. The team also discovered that fisetin suppresses activity in a signaling pathway called p38 MAPK, which is known to promote calcification. This effect depends on a protein called DUSP1. When DUSP1 was blocked, fisetin could no longer protect the cells, showing that this protein is essential for its anti-calcification activity. The researchers confirmed fisetin's protective effects in isolated mouse arteries and in living mice treated with high doses of vitamin D, which typically increases arterial calcification. “Mechanistically, fisetin requires the phosphatase DUSP1 to inhibit p38 MAPK in order to mediate its protective effect on VSMC calcification.” Importantly, the researchers tested fisetin under conditions similar to human disease. When VSMCs were exposed to blood serum from kidney dialysis patients—a condition known to trigger vascular calcification—fisetin again reduced calcium buildup and protected the cells. These findings suggest fisetin could be useful in countering the harmful vascular effects seen in chronic kidney disease. This study adds to growing evidence that fisetin may protect blood vessels from aging-related damage. While more research is needed before it can be used in clinical treatments, the study highlights fisetin as a promising candidate for slowing or preventing vascular calcification. The findings could have broad implications for aging populations and individuals with kidney disease, who are at greater risk for heart problems due to vascular stiffening. A Longitudinal Study on Dasatinib, Quercetin, and Fisetin Senolytic Interventions

BUFFALO, NY- March 6, 2024 – A new #research paper was #published in Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 16, Issue 4, entitled, “Exploring the effects of Dasatinib, Quercetin, and Fisetin on DNA methylation clocks: a longitudinal study on senolytic interventions.” Senolytics, small molecules targeting cellular senescence, have emerged as potential therapeutics to enhance health span. However, their impact on epigenetic age remains unstudied. In this new study, researchers Edwin Lee, Natàlia Carreras-Gallo, Leilani Lopez, Logan Turner, Aaron Lin, Tavis L. Mendez, Hannah Went, Alan Tomusiak, Eric Verdin, Michael Corley, Lishomwa Ndhlovu, Ryan Smith, and Varun B. Dwaraka from the Institute For Hormonal Balance, TruDiagnostic, Buck Institute for Research on Aging, and Cornell University aimed to assess the effects of senolytic treatments on DNA methylation (DNAm), epigenetic age, and immune cell subsets. “[...] this study aims to comprehensively assess the impact of senolytic drugs on epigenetic aging through two longitudinal studies to address our research objective. The initial investigation focuses on a combination treatment of Dasatinib and Quercetin, while the subsequent phase incorporates Fisetin into the treatment regimen.” In a Phase I pilot study, 19 participants received Dasatinib and Quercetin (DQ) for 6 months, with DNAm measured at baseline, 3 months, and 6 months. Significant increases in epigenetic age acceleration were observed in first-generation epigenetic clocks and mitotic clocks at 3 and 6 months, along with a notable decrease in telomere length. However, no significant differences were observed in second and third-generation clocks. Building upon these findings, a subsequent investigation evaluated the combination of DQ with Fisetin (DQF), a well-known antioxidant and antiaging senolytic molecule. After one year, 19 participants (including 10 from the initial study) received DQF for 6 months, with DNAm assessed at baseline and 6 months. Remarkably, the addition of Fisetin to the treatment resulted in non-significant increases in epigenetic age acceleration, suggesting a potential mitigating effect of Fisetin on the impact of DQ on epigenetic aging. “Furthermore, our analyses unveiled notable differences in immune cell proportions between the DQ and DQF treatment groups, providing a biological basis for the divergent patterns observed in the evolution of epigenetic clocks. Episode 57: In this episode, host Angie Gust talks about senescent cells and fisetin.  When normal cells get too damaged (as in accumulating too much DNA damage), they could turn into cancer cells. To prevent this, these damaged cells change to where they cannot divide anymore, so in that way, they cannot become cancer cells. Rather, they became senescent cells.  A bad thing is that. these cells secrete substances that damage the healthy surrounding cells, contributing to aging.  Fisetin has been shown to be good at clearing out this senescent cells.  Now turning to the environment. The 2023 UN Climate Conference, COP28, concluded on December 13th. While some think it was an improvement over other conferences, others thought it did not go far enough and left loop holes. The deal calls for transitioning away from (not phasing out) fossil fuels  in energy systems, in a just, orderly and equitable manner ... so as to achieve net zero by 2050. There was an agreement to triple renewables and double energy efficiency by 2030. S., Ravuri, S., Huard, J. Abstract: Mesenchymal stem cells (MSCs) have long been viewed as a promising therapeutic for musculoskeletal repair. However, regulatory concerns including tumorgenicity, inconsistencies in preparation techniques, donor-to-donor variability, and the accumulation of senescence during culture expansion have hindered the clinical application of MSCs. Senescence, in particular, is a driving mechanism for MSC dysfunction with advancing age. Often characterized by increased reactive oxygen species, senescence-associated heterochromatin foci, inflammatory cytokine and chemokine secretion, and reduced proliferative capacity, senescence directly inhibits MSCs efficacy as a therapeutic for musculoskeletal regeneration and repair. Furthermore, autologous delivery of senescent MSCs can further induce disease and aging progression through the secretion of the senescence-associated secretory phenotype (SASP) and mitigate the regenerative potentetial of MSCs. To combat these issues, the use of senolytic agents to selectively clear senescent cell populations has become popular. However, their benefits to human MSCs during the culture expansion process have not yet been elucidated. To address this, analyzed markers of senescence during culturing of human primary adipose-derived stem cells (ADSCs), a population of fat-resident MSCs commonly used in regenerative medicine applications. Next, we used the senolytic agent fisetin to determine if we can reduce these markers of senescence within our culture-expanded ADSC populations. Our results indicate that ADSCs acquire common markers of cellular senescence including increased reactive oxygen species, senescence-associated {beta}-galactosidase, and senescence-associated heterochromatin foci. Furthermore, we found that the senolytic agent fisetin works in a dose-dependent manner and selectively attenuates these markers of senescence while maintaining the differentiation potential of the expanded ADSCs. Regardless how frightening the possibility is, the chances of it happening to you aren't exactly slim, which means prevention should be at the forefront of your mind. A recent study from the Salk Institute for Biological Studies indicates prevention could be as simple as a natural foods diet—rich in fruits (such as strawberries) and vegetables containing something called fisetin. Fisetin is a flavonol found in strawberries, mangoes, cucumbers, and other vegetables and fruits. Researchers with the Salk Institute found this simple compound can actually reduce the risk of Alzheimer's in mice, and could be effective in humans as well. Maher and her team have documented that fisetin has both anti-inflammatory and antioxidant properties in the brain. It is also able to turn on a cellular pathway related to memory function. The team looked to a type of mouse with mutated genes making them vulnerable to Alzheimer's. At three months old, the researchers began feeding the mice a diet enriched with fisetin. Mice who hadn't received the fisetin began struggling in the mazes at nine months of age, but the fisetin mice performed as well as normal (non-predisposed) mice at both nine and twelve months of age. Avocados may hold the answer to beating leukemia University of Waterloo (Canada), June 16, 2022 Rich, creamy, nutritious and now cancer fighting. New research reveals that molecules derived from avocados could be effective in treating a form of cancer. Professor Paul Spagnuolo from the University of Waterloo has discovered a lipid in avocados that combats acute myeloid leukemia (AML) by targeting the root of the disease – leukemia stem cells. Worldwide, there are few drug treatments available to patients that target leukemia stem cells. “The stem cell is really the cell that drives the disease,” said Professor Spagnuolo, in Waterloo's School of Pharmacy. “The stem cell is largely responsible for the disease developing and it's the reason why so many patients with leukemia relapse. We've performed many rounds of testing to determine how this new drug works at a molecular level and confirmed that it targets stem cells selectively, leaving healthy cells unharmed.” Inability to stand on one leg for 10 seconds in mid to later life linked to near doubling in risk of death Exercise Medicine Clinic-CLINIMEX (Brazil) and University of Eastern Finland, June 21, 2022 The inability to stand on one leg for 10 seconds in mid- to later life is linked to a near doubling in the risk of death from any cause within the next 10 years, finds research published online in the British Journal of Sports Medicine This simple and safe balance test could be included in routine health checks for older adults, say the researchers. The researchers wanted to find out whether a balance test might be a reliable indicator of a person's risk of death from any cause within the next decade, and, as such, might therefore merit inclusion in routine health checks in later life. Participants were asked to stand on one leg for 10 seconds without any additional support.  To improve standardization of the test, participants were asked to place the front of the free foot on the back of the opposite lower leg, while keeping their arms by their sides and their gaze fixed straight ahead. Up to three attempts on either foot were permitted. In all, around 1 in 5 (20.5%; 348) participants failed to pass the test. The inability to do so rose in tandem with age, more or less doubling at subsequent 5 year intervals from the age of 51-55 onwards.  The proportions of those unable to stand on one leg for 10 seconds were: nearly 5% among 51-55 year-olds; 8% among 56-60 year-olds; just under 18% among 61-65 year-olds; and just under 37% among 66-70 year-olds.  More than half (around 54%) of those aged 71-75 were unable to complete the test. In other words, people in this age group were more than 11 times as likely to fail the test as those just 20 years younger. During an average monitoring period of 7 years, 123 (7%) people died: cancer (32%); cardiovascular disease (30%); respiratory disease (9%); and COVID-19 complications (7%). The proportion of deaths among those who failed the test was significantly higher: 17.5% vs. 4.5%, reflecting an absolute difference of just under 13%. Anxious Children have Bigger “Fear Centers” in the Brain Stanford University School of Medicine, June 16, 2022 The amygdala is a key “fear center” in the brain. Alterations in the development of the amygdala during childhood may have an important influence on the development of anxiety problems, reports a new study in the current issue of Biological Psychiatry.

 Researchers at the Stanford University School of Medicine recruited 76 children, 7 to 9 years of age, a period when anxiety-related traits and symptoms can first be reliably identified.  The researchers found that children with high levels of anxiety had enlarged amygdala volume and increased connectivity with other brain regions responsible for attention, emotion perception, and regulation, compared to children with low levels of anxiety. They also developed an equation that reliably predicted the children's anxiety level from the MRI measurements of amygdala volume and amygdala functional connectivity.

The most affected region was the basolateral portion of the amygdala, a subregion of the amygdala implicated in fear learning and the processing of emotion-related information.

 Our study represents an important step in characterizing altered brain systems and developing predictive biomarkers in the identification for young children at risk for anxiety disorders,” Qin said.  New research: Olive oil compound destroys cancer cells in 30 minutes Rutgers University & Hunter College, June 12, 2022  Oleocanthal, a polyphenolic, therapeutic compound found in olive oil is the subject of a new anti-cancer study performed by nutritional science and cancer biology researchers with The School of Environmental and Biological Sciences at Rutgers and Hunter's College in New York City. Programmed cell death, known as apoptosis takes approximately 16-24 hours. Dynamic new research just published in the Journal of Molecular and Cellular Oncology blew scientists away – when exposed to oleocanthal, a polyphenol compound found in olive oil, cancerous cells died within 30 minutes to an hour. While researchers previously understood that compounds in olive oil were capable of killing cancer cells, until now, such short apoptosis had not been observed. Even more fascinating was when the team looked closely to surmise why apoptosis was occurring under such swift circumstances – they discovered that cancer cells were being killed by their own enzymes. And, not only one isolated type of cancerous cell, but all of the cancerous cells they were examining. Unlike chemotherapeutic pharmaceuticals that devastate healthy cellular activity, the therapeutic polyphenolic compound found in olive oil kills cancer while maintaining vitality among healthy cells. As Paul Breslin, one of the study's authors at Rutgers noted, while cancerous cells died, healthy cells were not harmed, but rather the oleocanthal “put them to sleep.” The lifecycle of healthy cells was only temporarily affected in this way, without any negative observations and in approximately 24 hours, the healthy cells resumed their life cycle.  Sports, not screens: The key to happier, healthier children University of South Australia, June 21, 2022 Whether it's sports practice, music lessons or a casual catch up with friends, when children are involved in after-school activities, they're more likely to feel happier and healthier than their counterparts who are glued to a screen. In a new study conducted by the University of South Australia, researchers found that children's well-being is heightened when they participate in extra-curricular activities, yet lowered when they spent time on social media or playing video games. Published in BMC Pediatrics, the study analyzed data from 61,759 school students in years 4 to 9, assessing the average number of days per week children participated in after-school activities (3–6pm), and measure these against well-being factors—happiness, sadness, worry, engagement, perseverance, optimism, emotion regulation, and life satisfaction. It found that most students watched TV about four days of the school week and spent time on social media about three days of the week. Our study highlights how some out-of-school activities can boost children's well-being, while others—particularly screens—can chip away at their mental and physical health. “Screens are a massive distraction for children of all ages. And whether children are gaming, watching TV or on social media, there's something about all screens that's damaging to their well-being. Students in lower socio-economic backgrounds who frequently played sports were 15% more likely to be optimistic, 14% more likely to be happy and satisfied with their life, and 10% more likely to be able to regulate their emotions. Conversely, children who played video games and used social media almost always had lower levels of well-being: up to 9% less likely to be happy, up to 8% to be less optimism and 11% to be more likely to give up on things.

Episode Highlights [03:17] Travis' Background Travis has an undergraduate degree in biochemistry. He eventually took over the family business and got married.  He then finished his master's degree, focusing on cancer metabolism for his independent study. The prevailing teaching during that time was that somatic mutations cause cancer. Travis was inspired by the book, Cancer as a Metabolic Disease, to do a thesis on metabolic theory.  Otto Warburg proposed the metabolic theory many years ago, but the medical community dismissed it as simplistic. New evidence reignited interest in the approach. [08:10] The State of Healthcare Public and health institutions have long accepted the somatic approach to cancer, with billions of dollars backing it.  The United States spends the most per capita on health care expenditure among developed countries yet has the lowest life expectancy.  The issue with the healthcare system lies in variations of treatment, over-treatment, and drug development process.  The healthcare industry is focused more on treating diseases rather than preventing them.   Although technology has advanced to assist people in tracking their health, people's health is deteriorating. Even if people live longer, they often experience illness and disability in their final years. [12:48] Changing the Healthcare Industry  The healthcare incentive structure must change. Doctors get paid for every drug and procedure they prescribe, leading to overtreatment.  There are emerging practices where doctors get a big bonus per patient who doesn't need procedures, encouraging them to keep patients in good health. When blood sugar levels rise, doctors usually diagnose diabetes. However, we can test for fasting insulin regularly to intervene. This test can also help people prevent other diseases and become more health-conscious. Change takes time with systems and industries, but we can take simple lifestyle interventions now. [16:01] Why People Are Getting Sicker Maintaining our health can be challenging. We've been conditioned that taking a magic pill is preferable to putting in the effort. Remember that metabolic dysregulation causes aging, not the other way around. In the last 10,000 years, humanity has drastically shifted to consuming processed foods, carbohydrates, and oils, resulting in noticeable consequences. People have also become sedentary.  A hunter-gatherer society has no obesity, insulin resistance, or chronic systemic inflammation. However, we frequently see this in the Western world. [20:32] The Root of Diseases  Understanding cellular health and inflammatory processes are critical in keeping yourself healthy.  Periods of food scarcity have happened throughout humanity's evolution. Our bodies enter ketosis due to the caloric deficit but most never reach this state. Studies have shown that ketosis can help prevent diseases ranging from Alzheimer's to Parkinson's to cancer and diabetes. Constantly eating keeps our immune system active and prevents it from resting, which has implications for chronic inflammatory diseases. [24:53] What Happens During Ketosis?  Ketosis happens in a fasted state, and a ketogenic diet is nutritional maintenance. When we fast, our bodies will mobilise the fat we have in our bodies and begin a process of beta-oxidation to burn off fat.  A caloric deficit makes this process faster, converting fats into ketone bodies, which then take the place of glucose.  Ketones have more thermodynamic energy and ATP per carbon unit compared to glucose.  Ketosis also boosts antioxidant levels. Since diseases are oxidative, ketosis can help to combat illnesses. Listen to the episode to learn more!  [32:01] Growing Healthcare Trends  Research has shown that fasting before chemotherapy can reduce some radiation side effects Sugar is fuel for cancer cells, but doctors continue giving and allowing patients to consume it.  There's more awareness of the latest research. The annual Metabolic Therapeutic Conference has grown to around 1600 attendees.    When we approach doctors, we will receive information their institution has taught and allowed them to give us. Even with COVID-19, people have yet to explore treatments like monoclonal antibodies, hyperbaric, and Vitamin C.  [44:00] The Abscopal Effect on Cancer The abscopal effect is when you target a single tumour site, but it affects other tumour sites in the body.  Travis shares a case where someone with breast cancer on a checkpoint inhibitor received a much lower radiation dose.  In that case, radiation was sufficient to systematically stimulate the immune system to attack cancer, improving the patient's chances of survival. [47:41] Advancements in Immunotherapy Using peptides to manipulate the immune system is still a trial and error process. The immune system's job is to survey for cancer, get rid of it early, and create a checkpoint to avoid attacking itself.  [49:44] Emerging Cancer Treatment Research In detection, metastasis is an early stage event rather than a late-stage one. Cancer cells break off, dying when it reaches the bloodstream.  This selection pressure creates different genetic mutations in tumours.  The somatic mutation theory argues that a series of sequential mutations cause cancer, with each tumour site having a unique set of driving mutations. The epigenetic metabolic theory posits that these mutations are a side effect of the disease.  All cancers burn sugar and evade the immune system, but mutations vary too much. Treatments should focus on epigenetic expression rather than mutations. [55:16] Researching Treatment Options The medical field is filled with uncertainty. Cancer treatment options have not been compared.  After an in-depth look at clinical trials, we find that what seems to be the best treatment paradigm isn't always the right option. When conducting research, consider the data and evaluate it using a risk-reward framework. You can choose from low-risk alternatives. [59:59] The Soil Approach to Cancer  Cancer grows in the presence of a favourable microenvironment and molecular events. It takes a long time for a chronic irritant to develop cancer cells. Cancer cells take on characteristics of our earliest genes,  including replication and the appearance of early embryos. You can help your cells "behave" by providing oxygen and avoiding chronic irritants. Nurture the microenvironment through exercising and eating the right foods.  Instead of focusing on the seed or cancer, we should concentrate on the soil or microenvironment. [1:04:05] Health and Longevity While we don't know humankind's natural and optimal states, ketosis and fasting provide a glimpse. Cells stop metabolising as we age, causing inflammation. We may be able to extend life spans if we can reduce senescent cells. Travis shares a study in which mice received fisetin, found in strawberries, reducing senescent cells and prolonging their life.  There's a lot of new research on substances that kill senescent cells, like senolytics, quercetin, and other compounds. Every time you do that, it brings down those inflammatory processes, and so forth.' ‘We tend to think we're in this modern era, but with regard to cancer, we will look back 200 years from now, realise that we were just in the Dark Ages. We're still using radiation which was invented about 110 years ago; chemotherapy was invented around World War II. That's still the mainstay of cancer treatments.' ‘When you really look deep, the two problems really are variation in treatment, over-treatment. In this video Dr. Kirkland talks about the future of aging and how the biology of aging affects everyone and should not be thought of as only relevant to older people. He also discusses the path of bringing senolytics to wider use. Dr. James Kirkland is Director of the Robert and Arlene Kogod Center on Aging at Mayo Clinic and Noaber Foundation Professor of Aging Research. Dr. Kirkland's research focuses on cellular senescence, age-related adipose tissue and metabolic dysfunction, and development of agents and strategies for targeting fundamental aging mechanisms to treat age-related chronic diseases and disabilities and to extend healthspan. He published the first article about drugs that clear senescent cells, senolytic agents. A novel, mechanism-based, hypothesis-driven drug development paradigm was used to discover senolytic drugs. Based on the observation that senescent cells release factors that cause apoptosis of the cells around them, yet are themselves resistant to apoptosis, Dr. Kirkland hypothesized that senescent cells utilize senescent cell anti-apoptotic pathways (SCAPs) for protection from their own senescence-associated secretory phenotype (SASP). Using bioinformatics analyses of senescent vs. non-senescent cells and RNA interference, Dr. Kirkland identified these SCAPs and verified their importance for senescent cell survival. Dr. Kirkland used bioinformatics approaches to identify agents that target key nodes across the SCAP network and demonstrated these drugs are senolytic in rodent and human cultured cells and mice in vivo. These senolytic drugs include Dasatinib (D), Quercetin (Q), Fisetin, Navitoclax, and related compounds. Dr. Kirkland showed these agents delay, prevent, or alleviate multiple disorders in mouse models of human chronic diseases and aging phenotypes. Conditions alleviated in mouse models include frailty, diabetes, hepatic steatosis, cirrhosis, renal dysfunction, neuropsychiatric disorders, dementias, pulmonary fibrosis, osteoporosis, osteoarthritis, retinal degeneration, diastolic dysfunction, cardiac ischemia, vascular hyporeactivity, infertility, and skin disorders, among others. He demonstrated that intermittent, orally administered senolytics reduce senescent cell abundance in adipose tissue and blood markers of senescent cell burden in blood of patients with diabetic kidney disease. He and collaborators found that a brief course of senolytics enhances physical function and reduces frailty in patients with idiopathic pulmonary fibrosis, a fatal, cellular-senescence-driven disease for which available treatments have been unsatisfactory. Multiple clinical trials are currently underway of the senolytics that Dr. Kirkland discovered. He is a scientific advisory board member for several companies and academic organizations. In addition to being President-Elect of AFAR, he has been a member of the National Advisory Council on Aging of the National Institutes of Health, and past chair of the Biological Sciences Section of the Gerontological Society of America. He holds honorary appointments at Boston University and the University of Groningen in the Netherlands. He is a board-certified specialist in internal medicine, geriatrics, and endocrinology and metabolism. Among other risk factors, more and more studies point to diet as a major factor in the development of prostate cancer, as it is for cardiovascular disease, diabetes, and obesity. Using data from a study conducted in Montreal between 2005 and 2012, a research team led by Professor Marie-Elise Parent of Institut national de la recherche scientifique (INRS) has shown a link between diet and prostate cancer in the article “Dietary Patterns Are Associated with Risk of Prostate Cancer in a Population-Based Case-Control Study in Montreal, Canada”, published in Nutrients in June. Three main dietary profiles analyzed INRS PhD student Karine Trudeau, the lead author of the study, based her analysis on three main dietary profiles: healthy diet, salty Western diet including alcohol, and sugar-rich Western diet with beverages. The first profile leans heavily towards fruits, vegetables, and plant proteins like tofu and nuts. The salty Western diet with alcohol includes more meat and beverages such as beer and wine. The third profile is rich in pasta, pizza, desserts, and sugary carbonated drinks. The study took age, ethnicity, education, family history, and date of last prostate cancer screening into account. Marie-Elise Parent and Karine Trudeau found a link between a healthy diet and a lower risk of prostate cancer. Conversely, a Western diet with sweets and beverages was associated with a higher risk and seemed to be a factor in more aggressive forms of cancer. The study did not show any clear link between a Western diet with salt and alcohol and the risk of developing the disease. The newest heart-healthy vegetable could be a gigantic, record-setting radish. In a study appearing in ACS' Journal of Agricultural and Food Chemistry, scientists report that compounds found in the Sakurajima Daikon, or "monster," radish could help protect coronary blood vessels and potentially prevent heart disease and stroke. The finding could lead to the discovery of similar substances in other vegetables and perhaps lead to new drug treatments. Grown for centuries in Japan, the Sakurajima Daikon is one of the Earth's most massive vegetables. In 2003, the Guinness Book of World Records certified a Sakurajima weighing nearly 69 pounds as the world's heaviest radish. Radishes are good sources of antioxidants and reportedly can reduce high blood pressure and the threat of clots, a pair of risk factors for heart attack and stroke. But to date, no studies have directly compared the heart-health benefits of the Sakurajima Daikon to other radishes. To address this knowledge gap, Katsuko Kajiya and colleagues sought to find out what effects this radish would have on nitric oxide production, a key regulator of coronary blood vessel function, and to determine its underlying mechanisms. The researchers exposed human and pig vascular endothelial cells to extracts from Sakurajima Daikon and smaller radishes. Using fluorescence microscopy and other analytical techniques, the research team found the Sakurajima Daikon radish induced more nitric oxide production in these vascular cells than a smaller Japanese radish. They also identified trigonelline, a plant hormone, as the active component in Sakurajima Daikon that appears to promote a cascade of changes in coronary blood vessels resulting improved nitric oxide production.   Placebos prove powerful even when people know they're taking one Michigan State University, August 7, 2020   How much of a treatment is mind over matter? It is well documented that people often feel better after taking a treatment without active ingredients simply because they believe it's real -- known as the placebo effect. A team of researchers from Michigan State University, University of Michigan and Dartmouth College is the first to demonstrate that placebos reduce brain markers of emotional distress even when people know they are taking one. Now, evidence shows that even if people are aware that their treatment is not "real" -- known as nondeceptive placebos -- believing that it can heal can lead to changes in how the brain reacts to emotional information. "Just think: What if someone took a side-effect free sugar pill twice a day after going through a short convincing video on the power of placebos and experienced reduced stress as a result?", said Darwin Guevarra, MSU postdoctoral fellow and the study's lead author. "These results raise that possibility." The new findings, published in the most recent edition of the journal Nature Communications, tested how effective nondeceptive placebos -- or, when a person knows they are receiving a placebo -- are for reducing emotional brain activity.  "Placebos are all about 'mind over matter," said Jason Moser, co-author of the study and professor of psychology at MSU. "Nondeceptive placebos were born so that you could possibly use them in routine practice. So rather than prescribing a host of medications to help a patient, you could give them a placebo, tell them it can help them and chances are -- if they believe it can, then it will." To test nondeceptive placebos, the researchers showed two separate groups of people a series of emotional images across two experiments. The nondeceptive placebo group members read about placebo effects and were asked to inhale a saline solution nasal spray. They were told that the nasal spray was a placebo that contained no active ingredients but would help reduce their negative feelings if they believed it would. The comparison control group members also inhaled the same saline solution spray, but were told that the spray improved the clarity of the physiological readings the researchers were recording. The first experiment found that the nondeceptive placebos reduced participants' self-reported emotional distress. Importantly, the second study showed that nondeceptive placebos reduced electrical brain activity reflecting how much distress someone feels to emotional events, and the reduction in emotional brain activity occurred within just a couple of seconds. "These findings provide initial support that nondeceptive placebos are not merely a product of response bias - telling the experimenter what they want to hear -- but represent genuine psychobiological effects," said Ethan Kross, co-author of the study and a professor of psychology and management at the University of Michigan.     Greater coffee intake associated with decreased depressive symptoms among older Japanese women Nakamura Gakuen University (Japan), August 5, 2020   According to news reporting originating from Fukuoka, Japan, by NewsRx correspondents, research stated, “Depression in elderly people is a major global concern around the world. Coffee intake was associated with a lower prevalence of depressive symptoms, the ORs of which for the 4th versus the 1st quartiles of intake was 0.64 (95% CI, 0.46-0.88, P for trend = 0.01) in a fully adjusted model. Caffeine intake was marginally associated with depressive symptoms, but the association was not statistically significant (OR 0.75; 95% CL 0,55-1,02. P for trend = 0.058). The result suggests that the inverse association of coffee intake with depressive symptoms might be associated with not only caffeine intake but also some other substances in coffee or factors related to coffee intake.” According to the news editors, the research concluded: “Because of the cross-sectional design of the present study, longitudinal studies are required to confirm the present finding.”     Sugary drinks and disease: Chugging 2 sodas per day increases your risk of premature death University College Dublin (Ireland), August 8, 2020   On top of raising blood sugar and contributing to abdominal fat, European researchers found that soda can also lead to an earlier death. Published in JAMA Internal Medicine, the study demonstrates that daily consumption of two or more sodas – diet or not – and other sweetened drinks corresponds to a 17 percent increase in the risk of premature death from all causes. The “bitter truth” of soda consumption: premature death To examine the relationship between soda consumption and the risk of all-cause and cause-specific mortality, the researchers studied 451,743 individuals living in 10 European countries, including Denmark, France, Germany, Greece, Italy, the Netherlands, Norway, Spain, Sweden and the United Kingdom. The participants were from the European Prospective Investigation into Cancer and Nutrition (EIPC) study, one of the largest ongoing cohort studies on diet and its relation to cancer and other chronic diseases. None of the participants have any chronic conditions. The team studied their soda consumption for an average of 16 years. During that period, a total of 41,693 participants died from all causes, eleven percent of whom reported drinking at least two sodas daily, while nine percent reported drinking not more than one per month. The participants who drank two or more glasses of soda also had a higher risk of death from heart conditions. Meanwhile, those who reported consuming other beverages sweetened with either sugar or artificial sweeteners had a greater risk of death from digestive diseases. Participants who drank diet soda weren't off the hook either. The team reported that those who drank diet sodas also had a greater risk of earlier death from cardiovascular disease(CVD). Taken together, these findings indicate that the consumption of soda, diet soda and other sweetened beverages is linked to premature death from all causes, including CVD and digestive diseases. The researchers noted that their study supports public health campaigns aimed at limiting the consumption of sodas and other sugar-laden drinks.       Fisetin derivative shows promise against Alzheimer disease in mice Salk Institute, August 5 2020.    he September 2020 issue of Redox Biology published the finding of Salk Institute researchers of an ability for a compound derived from fisetin, a flavonoid occurring in many plants, to reverse memory loss in a mouse model of Alzheimer disease. The compound, known as CMS121, which was synthesized by Pamela Maher and colleagues, was recently demonstrated to slow brain cell aging. "This was a more rigorous test of how well this compound would work in a therapeutic setting than our previous studies on it," commented Dr Maher. "Based on the success of this study, we're now beginning to pursue clinical trials." In the current research, normal mice and mice that were genetically modified to develop Alzheimer disease were given CMS121 starting at nine months of age. Untreated Alzheimer mice and normal mice served as controls. At 12 months, memory and behavior tests revealed that treated Alzheimer mice performed as well as control mice and that Alzheimer mice that did not receive CMS121 performed worse.  An increase in lipid peroxidation was observed in brains cells of untreated Alzheimer mice in comparison with Alzheimer mice that received CMS121. "That not only confirmed that lipid peroxidation is altered in Alzheimer's, but that this drug is actually normalizing those changes," remarked first author Gamze Ates.  It was further determined that CMS121 lowered levels of the lipid-producing molecule fatty acid synthetase (FASN). Brain samples from human Alzheimer patients revealed that greater amounts of FASN were present in comparison with cognitively healthy patients, suggesting that FASN could be a drug target for Alzheimer disease.  "There has been a big struggle in the field right now to find targets to go after," Dr Maher stated. "So, identifying a new target in an unbiased way like this is really exciting and opens lots of doors."     REM sleep tunes eating behavior University of Bern (Germany),  August 7, 2020   Despite our broad understanding of the different brain regions activated during rapid-eye-movement sleep, little is known about what this activity serves for. Researchers at the University of Bern and the Inselspital have now discovered that the activation of neurons in the hypothalamus during REM sleep regulates eating behaviour: suppressing this activity in mice decreases appetite. While we are asleep, we transition between different phases of sleep each of which may contribute differently to us feeling rested. During (rapid eye movement) REM sleep, a peculiar sleep stage also called paradoxical sleep during which most dreaming occurs, specific brain circuits show very high electrical activity, yet the function of this sleep-specific activity remains unclear.  Among the brain regions that show strong activation during REM sleep are areas that regulate memory functions or emotion, for instance. The lateral hypothalamus, a tiny, evolutionarily well conserved brain structure in all mammals also shows high activity during REM sleep. In the awake animals, neurons from this brain region orchestrate appetite and the consumption of food and they are involved in the regulation of motivated behaviours and addiction. In a new study, researchers headed by Prof. Dr. Antoine Adamantidis at the University of Bern set out to investigate the function of the activity of hypothalamic neurons in mice during REM sleep. They aimed at better understanding how neural activation during REM sleep influences our day-to-day behaviour. They discovered that suppressing the activity of these neurons decreases the amount of food the mice consume. "This suggests that REM sleep is necessary to stabilize food intake", says Adamantidis. The results of this study have been published in the journal Proceedings of the National Academy of Sciences (PNAS). Long-lasting effect on neuronal activity and feeding behavior The researcher discovered that specific activity patterns of neurons in the lateral hypothalamus that usually signal eating in the awake mouse are also present when the animals were in the stage of REM sleep. To assess the importance of these activity patterns during REM sleep the research group used a technique called optogenetics, with which they used light pulses to precisely shut down the activity of hypothalamic neurons during REM sleep. As a result, the researchers found that the activity patterns for eating were modified and that the animals consumed less food. "We were surprised how strongly and persistently our intervention affected the neural activity in the lateral hypothalamus and the behaviour of the mice", says Lukas Oesch, the first author of the study. He adds: "The modification in the activity patterns was still measurable after four days of regular sleep." These findings suggest that electrical activity in hypothalamic circuits during REM sleep are highly plastic and essential to maintain a stable feeding behaviour in mammals.  It is a question of quality These findings point out that sleep quantity alone is not solely required for our well-being, but that sleep quality plays a major role in particular to maintain appropriate eating behaviour. "This is of particular relevance in our society where not only sleep quantity decreases but where sleep quality is dramatically affected by shift work, late night screen exposure or social jet-lag in adolescents", explains Adamantidis. The discovered link between the activity of the neurons during REM sleep and eating behaviour may help developing new therapeutical approaches to treat eating disorders. It might also be relevant for motivation and addiction.
