Sibylle Mittnacht, PhD discusses the current landscape of PARP inhibitor use in cancers and osteosarcoma and discusses preclinical evidence and translation of biomarker-driven use of therapeutics in osteosarcoma. Dr. Mittnacht heads the laboratory of Cancer Cell Signalling at the University College London (UCL) Cancer Institute, with a long-standing interest in concepts that drive tumour development and therapy response in cancer. She acts as the cancer expert for the Royal College of Radiologists and is a founding member of the FOSTER (Fight Osteosarcoma Through European Research) initiative, where she co-leads the cell model and drug response focus groups and is a participant in a newly established initiative, icure, which aims to establish a living biobank and therapeutics assessment pipeline for rare and ultrarare cancers.Work in the Mittnacht team centers on understanding how cell cycle checkpoint aberrations affect the behaviour of cancer cells and the identification of therapeutic opportunity that arises therefrom. The team uses functional genomics and chemical biology to discover new treatment paradigms with a current focus on unmet needs in sarcoma and breast cancer, and is involved in establishing patient-derived and tissue-mimetic preclinical cancer models for these cancers, in order to accelerate therapeutic discovery.

Abstract: Huntington disease (HD) is an autosomal dominant genetic neurodegenerative disease caused by a CAG expansion in the Huntingtin (HTT) gene, translating to an expanded polyglutamine tract in the huntingtin (HTT) protein. Age at disease onset is correlated to CAG repeat length, but varies by decades between individuals with identical repeat lengths. Genome-wide association studies link HD modification to DNA repair and mitochondrial health pathways. Recent clinical studies show elevated DNA damage in HD, even at the premanifest stage of disease. One of the major DNA repair nodes influencing neurodegenerative disease is the PARP pathway. Accumulation of poly ADP-ribose (PAR), produced by PARP1 and PARP2, has been implicated in the pathology of Alzheimers and Parkinsons diseases, as well as autosomal recessive cerebellar ataxia. We report that HD mutation carriers have lower cerebrospinal fluid PAR levels than healthy controls, starting at the premanifest stage. Patient-derived fibroblasts have reduced PARP1/2 activity and elevated DNA damage, while elevated PAR levels are only revealed upon inhibition of PAR degradation. These phenotypes are rescued by moderate huntingtin level reduction via the huntingtin-lowering splice modulator drug, LMI070 (Branaplam). As a direct mechanism, we have defined a PAR-binding motif in huntingtin, detected huntingtin complexed with PARylated proteins in human cells during stress, and localized huntingtin to mitotic chromosomes upon inhibition of PAR degradation. Direct huntingtin PAR binding was measured by fluorescence polarization and visualized by atomic force microscopy. These results provide insight into a very early molecular mechanism of HD, suggesting possible targets in HD to design early preventive therapies. Copy rights belong to original authors. BUFFALO, NY- September 20, 2022 – A new research paper was published in Oncotarget's Volume 13 on September 14, 2022, entitled, “Targeting Krebs-cycle-deficient renal cell carcinoma with Poly ADP-ribose polymerase inhibitors and low-dose alkylating chemotherapy.” Loss-of-function mutations in genes encoding the Krebs cycle enzymes Fumarate Hydratase (FH) and Succinate Dehydrogenase (SDH) induce accumulation of fumarate and succinate, respectively, and predispose patients to hereditary cancer syndromes including the development of aggressive renal cell carcinoma (RCC). Fumarate and succinate competitively inhibit αKG-dependent dioxygenases, including Lysine-specific demethylase 4A/B (KDM4A/B), leading to suppression of the homologous recombination (HR) DNA repair pathway. Researchers Daiki Ueno, Juan C. Vasquez, Amrita Sule, Jiayu Liang, Jinny van Doorn, Ranjini Sundaram, Sam Friedman, Randy Caliliw, Shinji Ohtake, Xun Bao, Jing Li, Huihui Ye, Karla Boyd, Rong Rong Huang, Jack Dodson, Paul Boutros, Ranjit S. Bindra, and Brian Shuch from Yale University School of Medicine, West China Hospital/School of Medicine, Wayne State University, and University of California, Los Angeles have developed new syngeneic Fh1- and Sdhb-deficient murine models of RCC, which demonstrate the expected accumulation of fumarate and succinate, alterations in the transcriptomic and methylation profile, and an increase in unresolved DNA double-strand breaks (DSBs). “In this study, we sought to examine the activity of combined TMZ and PARPi in Krebs-cycle-deficient renal cancer models. Fh1 and Sdhb are the murine counter part of human FH and SDHB, respectively.” The efficacy of poly ADP-ribose polymerase inhibitors (PARPis) and temozolomide (TMZ), alone and in combination, was evaluated both in vitro and in vivo. Combination treatment with PARPi and TMZ results in marked in vitro cytotoxicity in Fh1- and Sdhb-deficient cells. In vivo, treatment with standard dosing of the PARP inhibitor BGB-290 and low-dose TMZ significantly inhibits tumor growth without a significant increase in toxicity. These findings provide the basis for a novel therapeutic strategy exploiting HR deficiency in FH and SDH-deficient RCC with combined PARP inhibition and low-dose alkylating chemotherapy. “Using newly developed Fh1 and Sdhb deficient syngeneic mouse models, we demonstrate that oncometabolite-induced HR defects can be leveraged with PARPi treatment to enhance sensitivity to low-dose TMZ in Krebs-cycle-deficient renal cancer.” DOI: https://doi.org/10.18632/oncotarget.28273 Correspondence to: Ranjit S. Rare diseases. The only thing really rare about them is their name! While it is true that individually, each rare disease only affects a handful of people, together, they represent 300 million individuals worldwide. That is 4% of the human population.  These 300 million people suffer from over 7.000 different diseases that are catalogued under the generic name ‘rare diseases', simply because they have a common feature: they affect fewer than 5 in 10.000 people. But that is not the only thing these diseases have in common. Most of them are genetic, appear during childhood and are incredibly challenging to diagnose.  Did you know that it takes an average of 7 to 8 years before a diagnosis is reached? Abstract: Ribose-Map is a user-friendly, standardized bioinformatics toolkit for the comprehensive analysis of ribonucleotide sequencing experiments. It allows researchers to map the locations of ribonucleotides in DNA to single-nucleotide resolution and identify biological signatures of ribonucleotide incorporation. In addition, it can be applied to data generated using any currently available high-throughput ribonucleotide sequencing technique, thus standardizing the analysis of ribonucleotide sequencing experiments and allowing direct comparisons of results. This protocol describes in detail how to use Ribose-Map to analyze raw ribonucleotide sequencing data, including preparing the reads for analysis, locating the genomic coordinates of ribonucleotides, exploring the genome-wide distribution of ribonucleotides, determining the nucleotide sequence context of ribonucleotides, and identifying hotspots of ribonucleotide incorporation. Ribose-Map does not require background knowledge of ribonucleotide sequencing analysis and assumes only basic command-line skills. [Schau dir den TED Talk von Amy Cuddy an] Muskelspannung hat Einfluss auf unseren Hormonhaushalt: Während Muskeltraining werden Myokine ausgeschüttet und folglich ein Nervenwachstumsfaktor (BDNF). BDNF wirkt im Gehirn auf Serotoninausschüttung, deshalb wirkt Training auf Emotionen und macht glücklich. Blutzuckerkontrolle bzw. Insulinsensitivität ist eine wichtige Voraussetzung für Gesundheit und Leistungsfähigkeit. Insulin ist ein Energiesensor im Gehirn Blutzuckerkontrolle lässt sich am einfachsten verbessern durch Bewegung und „nicht ständig essen“ bzw. intermittierendes Fasten. Weniger Essen ist nicht unbedingt zielführend gegen Insulinresistenz. Sportler, die sich außerhalb ihres Trainings nicht ausreichend bewegen, können trotzdem Insulinresistent werden. Wie schreit das Gehirn nach Energie? Über Emotionen: Hunger, Durst und Aktivierung der Stressachsen. Hunger sorgt für Dopaminausschüttung (Motivation) und macht Lust auf Bewegung. Negative Emotionen bei Hunger sind ein Signal für ein Problem bei der Energiebereitstellung. Energiegewinnung hat die höchste Priorität im Körper – auf Kosten anderer Funktionen: Wenn natürliche Energiegewinnungswege nicht genutzt werden können, werden andere Organe zur Energiegewinnung genutzt und vernachlässigen ihre anderen Funktionen. Kann die Leber zur Energiegewinnung genutzt werden, aber vernachlässigt dann ihre Entgiftungsfunktion. Für große Änderungen im Lebensstil fehlt oft die Energie, deshalb können „Quick Wins“ ein guter Einstieg sein für mehr Energie, um Veränderungen durchzuführen. This is a major contributor to the pathogenesis of Parkinsons disease, one of the most common neurological diseases in older individuals. Ribose-D : Ribose-D is a five-carbon sugar found in every cell in our bodies that combines with oxygen and ATP (adenosine triphoshate) to give energy to each cell. Ribose is also present in RNA (ribonucleic acid), which is one of the main information carriers of living organisms. Oxidative stress, as measured by free radical damage to cells, leads to systematic inflammation. Fortunately, our bodies handle this problem daily. Learn Why L-Carnitine May Be A Valuable Therapy In Helping Manage Heart Failure Dr. Mohammad Reza Movahed received his MD from Medizinische Hochschule Hannover, Germany. He completed his medical residency at the University of Rochester, New York and his cardiology training at the University of South Carolina, Columbia. His interventional cardiology training was completed at Yale University Hospital, New Haven, Connecticut in 2000. He has been an Assistant Clinical Professor at the University of California, Irvine and in 2007 joined the Southern Arizona VA Health Care System, University of Arizona College of Medicine for his clinical work and as the medical director of heart transplantation. Since July 2010, Dr Movahed has been a tenured Professor of Medicine at the University of Arizona College of Medicine. He has contributed to over150 publications in major peer reviewed medical journals. He recently was a co-author on a paper with lead investigator Dr. Ali Reza Serati MD and colleagues from Modarres Hospital, Tehran, Iran entitled: "L-Carnitine Treatment in Patients with Mild Diastolic Heart Failure is Associated with Improvement in Diastolic Function and Symptoms" in the jounral Cardiology 2010.
