BUB1 Regulates EGFR Signaling by Reducing EGFR Internalization

A new research paper was published in Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 15, Issue 13, entitled, “Budding uninhibited by benzimidazoles-1 (BUB1) regulates EGFR signaling by reducing EGFR internalization.” EGFR signaling initiates upon ligand binding which leads to activation and internalization of the receptor-ligand complex. In this new study, researchers Shyam Nyati, Grant Young, Corey Speers, Mukesh K. Nyati, and Alnawaz Rehemtulla from the University of Michigan, Henry Ford Health System and Case Western Reserve University evaluated if BUB1 impacted EGFR signaling by regulating EGFR receptor internalization and activation. “We postulate that BUB1 helps in the formation and stabilization of EGFR dimers at the membrane and may regulate endocytosis of activated EGFR into either clathrin dependent (EEA1 coated) or independent (caveolin coated) vesicles thus impacting receptor recycling or degradation and subsequently signaling amplitude and duration [38, 39].” BUB1 was ablated genomically (siRNA) or biochemically (2OH-BNPP1) in cells. EGF ligand was used to initiate EGFR signaling while disuccinimidyl suberate (DSS) was used for cross linking cellular proteins. EGFR signaling was measured by western immunoblotting and receptor internalization was evaluated by fluorescent microscopy (pEGFR (pY1068) colocalization with early endosome marker EEA1). siRNA mediated BUB1 depletion led to an overall increase in total EGFR levels and more phospho-EGFR (Y845, Y1092, and Y1173) dimers while the amount of total EGFR (non-phospho) dimers remained unchanged. BUB1 inhibitor (BUB1i) decreased EGF mediated EGFR signaling including pEGFR Y845, pAKT S473 and pERK1/2 in a time dependent manner. Additionally, BUB1i also reduced EGF mediated pEGFR (Y845) dimers (asymmetric dimers) without affecting total EGFR dimers (symmetric dimers) indicating that dimerization of inactive EGFR is not affected by BUB1. Furthermore, BUB1i blocked EGF mediated EGFR degradation (increase in EGFR half-life) without impacting half-lives of HER2 or c-MET. Abstract: Objective: High cholesterol levels in pancreatic {beta}-cells cause oxidative stress and decrease insulin secretion. {beta}-cells can internalize apolipoprotein (apo) A-I, which increases insulin secretion. This study asks whether internalization of apoA-I improves {beta}-cell insulin secretion by reducing oxidative stress. Approach: Ins-1E cells were cholesterol-loaded by incubation with cholesterol-methyl-{beta}-cyclodextrin. Insulin secretion in the presence of 2.8 or 25 mM glucose was quantified by radioimmunoassay. Internalization of fluorescently labelled apoA-I by {beta}-cells was monitored by flow cytometry. The effects of apoA-I internalization on {beta}-cell gene expression was evaluated by RNA sequencing. ApoA-I binding partners on the {beta}-cell surface were identified by mass spectrometry. Mitochondrial oxidative stress was quantified in {beta}-cells and isolated islets with MitoSOX and confocal microscopy. Results: An F1-ATPase {beta}-subunit on the {beta}-cell surface was identified as the main apoA-I binding partner. {beta}-cell internalization of apoA-I was time-, concentration-, temperature-, cholesterol- and F1-ATPase {beta}-subunit-dependent. {beta}-cells with internalized apoA-I (apoA-I+ cells) had higher cholesterol and cell surface F1-ATPase {beta}-subunit levels than {beta}-cells without internalized apoA-I (apoA-I- cells). The internalized apoA-I co-localized with mitochondria and was associated with reduced oxidative stress and increased insulin secretion. The ATPase inhibitory factor 1, IF1, attenuated apoA-I internalization and increased oxidative stress in Ins-1E {beta}-cells and isolated mouse islets. Differentially expressed genes in apoA-I+ and apoA-I- Ins-1E cells were related to protein synthesis, the unfolded protein response, insulin secretion and mitochondrial function. Conclusions: These results establish that {beta}-cells are functionally heterogeneous and apoA-I restores insulin secretion in ?-cells with elevated cholesterol levels by improving mitochondrial redox balance. The systemic nature of this defense mechanism is one of its more fascinating characteristics and the recognition and transport of double-stranded RNA (dsRNA) of viral origin is required for the systemic activity of the siRNA pathway. Indeed, cellular internalization of dsRNA from the environment is a widespread phenomenon among insects. Here we aimed to identify cell surface proteins that bind to extracellular dsRNA and mediate its internalization. To this end, we developed a novel co-immunoprecipitation protocol that we followed with proteomics analysis. Among the hits from our screens was Hsc70-4, a constitutively expressed member of the heat shock protein family that has been implicated in clathrin-mediated endocytosis. We found that silencing Hsc70-4 impaired dsRNA internalization. Surprisingly, despite lacking a predicted transmembrane domain, Hsc70-4 localizes to the cell membrane and this localization was preserved when Hsc70-4 was expressed in mammalian cells, suggesting a conserved role at the cell surface. Furthermore, Hsc70-4 shows a previously undescribed dsRNA-specific binding capacity. Our results show that Hsc70-4 is a key element of the dsRNA internalization process and its detailed study may facilitate the development of RNA interference (RNAi)-based technologies for pest and vector borne disease control. ImportanceTo protect plants from pathogens or pests, the technology of "Host-induced gene silencing" has emerged as a powerful alternative to chemical treatments. This is an RNAi-based technology where small RNAs made in the plant silence the genes of the pests or pathogens that attack the plant. The small RNAs are generally derived from dsRNA expressed in transgenic plants. Alternatively, dsRNA can be sprayed onto the plant surface, where it can be taken up into the plant or ingested by pests. We have identified a cell surface protein that mediates the early steps of extracellular dsRNA internalization in insect cells. So I've asked one of the few experts in the world on this subject to join me and walk us through what internalization is, how it works, and more. Her name is Katie Frost, and she was actually my teacher a long time ago for a workshop on oral drafting that I attended in Dallas. Katie is an Ethnoarts consultant with SIL, and a professor at Dallas International University. Her experience in both ethnoarts and linguistics/translation led to co-founding the Psalms: Layer by Layer project as part of pursuing how these disciplines intersect in biblical Hebrew poetry. She's done fieldwork in the Netherlands and the Middle East, and is married to Josh who also serves in Bible translation. And they have a baby girl. Abstract: Growth hormone (GH) and many other hormones or growth factors have been shown to be rapidly internalized and translocated into the nucleus. The first event of peptide action is binding to its receptor which initiates both signal transduction pathways and internalization. The latter process involves the nucleus and probably gene transcription. In order to analyze the consequences of internalization of GH on gene expression, we used different populations of CHO cells, transfected with either full length GH receptor, or with defective receptor unable to trigger either signal transduction (deletion of box1) or the internalization of GH (Phe346 mutation in Ala). In addition, functional isolated nuclei were incubated 10 and 30 min with 50 nM GH in order to analyze the direct effect of GH on gene expression without surrounding cytoplasmic structures. The genes involved in signal transduction pathways were not revealed if GH internalization is the only functional activity in the whole cell as well as with isolated nuclei. In intact cell, internalization increased expression of 297 genes and decreased fewer than 10% of those known to be influenced by GH. Variations of expression in purified nuclei showed large variations with time. If cell signaling was not modified, cellular growth and proliferation, nucleic acid metabolism, cellular development, cell cycle and gene expression showed many variations with time. GH internalization shows direct effects on gene expression, different from those stimulated by signal transduction. Copy rights belong to original authors. This is a term that we're hearing more and more, but I'm not sure it's something that everybody fully understands just yet. There are a lot of misconceptions around exactly what it means and how it works, but ultimately the impact on a victims mental health, and the people around them, is huge. And so it's an issue that you've been asking us to dive into on Blended. Today, our guests will be exploring exactly what gaslighting means; sharing their personal experiences; discussing whether the rise in popularity of the term is a good thing or a bad thing; giving us tips on how to spot gaslighting at work and in our homes; and sharing advice on how we can help victims. IN THIS EPISODE WE DISCUSS:   [02.10] Introductions to our Blended panelists. Self-doubt Narcissism Manipulation Male-female dynamics in the workplace Whitewashing of history Why empathy is in direct opposition of gaslighting [17.59] The panel shares their personal experiences; the impact those experiences have had on their work and personal lives; and how they dealt with them. “I'm sorry you feel that way” vs “I'm sorry I made you feel that way” Zero tolerance policies and following through in the workplace Personal responsibility Conscious vs unconscious gaslighting Love bombing Broken promises Control “A prime example, from my experience, was being told “you blew it out of proportion, I don't know why you had to report that, I was just trying to help you, I think so highly of you… ” Jolene [39.17] The group discusses the many and varied damaging impacts that gaslighting can have on victims. The gaslighting phenomenon can have many and varied adverse impacts. It can cause victims to doubt their own reality, question their sanity, and withdraw from social interactions. Gaslighting can also lead to decreased work performance, relationship difficulties, and mental health problems. Self-harm and suicide Internalization, introversion and isolation Further abusive relationships Triggering PTSD Trauma Anxiety and depression “It's like we attract it… like there's something we haven't dealt with, within ourselves, that causes it to repeat from other people.” Jenn [49.23] The panel reflects on the growing popularity of the term ‘gaslighting' and whether or not that popularity is a good thing; how we can identify gaslighting; and what we can do about it. Has it become a buzzword? Preference 3. Identity 4. Internalization 5. Fanaticism Level 5 - Fanaticism: At this level... "My knowledge controls my every action." Fanaticism: a rigid attachment to knowledge with an obsessive intolerance of opposing views. It is driven by the need to believe in something 100% even though that something's meaning is dependent on the agreement of others. Anything that contradicts or puts into question the sustainability of the belief is a direct threat, and the fanatic will believe the threat at any cost… Prejudice, intolerance, and violence are the instruments by which the beliefs are imposed onto the dream of the planet. The driving force behind fanaticism is not hate or anger, but rather an extreme form of conditional love for self and others. This is how any beautiful belief in the world can become lost in corruption, as knowledge controls a persons will for the sake of its own existence. Fanaticism is the complete loss of respect for another living being. When we no longer see an individual as a living being, and instead only an idea or a number. The image of oneself can become so distorted that one's own perception of their own humanity becomes lost. There is no awareness that a line has been crossed, only an illusion remains.

Preference 3. Identity 4. Internalization 5. Fanaticism Level 4 - Internalization: At this stage... "my identity in the form of my knowledge gives me the rules and guidelines by which I live my life." Internalization describes a degree of attachment to knowledge where our identity becomes the model in which we accept ourselves. This is domestication through attachment. At this level our narrators have begun to set the conditions by which we domesticate our identity. They measure our acceptance and rejection of ourselves and others based on the beliefs we use to construct the mask of our identity. We will distort the information we receive to reinforce the conditions of which we expect from life. The narrators also serve our need to validate who we are in our personal dream, as well as the face we present to the world . Our knowledge is corrupted. Our sense of self is the personification of our beliefs and our will is subjugated by the need to fit in with the dream.. Thus our mask may not necessarily be in the form of our passion, but we will wear whatever mask we think we need to be accepted. At this point we've lost our sense of respect for self and others, and conditional love is all we know.

But this identity is a mask that blurs our awareness of the authentic self… An attachment at this level occurs when we identify ourselves with our knowledge.

Currently settled in Portland, Oregon, Danny has been on a recovery journey after coming to terms with the sexual abuse he suffered as a child at the hands of his own brother. With his other siblings threatened into silence, Danny suffered for years before finally realizing that what he was experiencing was not normal. Once he opened up to someone about what he had lived through, he went down a path of healing. Transformed from his experience, Danny opens up about being a male sexual assault survivor, his struggles with masculinity, isolation, and balancing strength with vulnerability. He shares with us his trauma, what his thoughts are the term "survivor", why authenticity led him to share his story, and how he has sought wisdom throughout his recovery journey. All this week we've been looking at helpful insights for Christian parents from Dr. Christian Smith's research on how parents pass their faith on to the next generation. In addition to reminding Christian parents of the role that walking their talk plays in the faith formation of their kids, Smith has found a number of traits that are present in families where the Christian faith is passed on to kids. One of Smith's best and most helpful suggestions is that parents engage in channeling for internalization. What does this mean? Channeling for internalization is a term describing how parents purposefully arrange for their kids to have involvements and relationships that reinforce rather than replace their more direct parental influence on their kids. In other words, we want to get them involved with Christian adults from our church, with a youth group, with a youth pastor, and others who will have a positive spiritual input on your kids. You have some people that have internalized their dating history and see my content and are like “omg, I love your content, please help me” and they are just extremely disappointed with themselves to the point of not feeling worthy of dating and therefore being stagnant while dating. On the flip side, you have the women that are also internalizing and feel regret for their dating experiences, so they response in a negative way to me as a way to deflect from the shame they feel in themselves. Let me tell both types of women. Stop. Stop internalizing and start powerfully acknowledging. So that’s what we’re going to talk about today.

C., Hyman, B. T., Strickland, D. Abstract: ABSTRACT The identification of the apoE receptor, LRP1, as an endocytic receptor for tau raises several questions about LRP1s role in tauopathies. Is internalized tau, like other LRP1 ligands, delivered to lysosomes for degradation? Does LRP1 internalize pathological tau leading to cytosolic seeding? Do other, related receptors participate in these processes? We confirm that LRP1 rapidly internalizes tau, leading to efficient lysosomal degradation. Employing brain homogenates from human Alzheimer brain, we find that LRP1 also mediates cytosolic tau seeding. We additionally found that another apoE receptor, SORL1, a gene implicated in AD risk, also mediates tau endocytosis, degradation, and release into the cytoplasm of seed competent species. These data suggest a role for these apoE receptors in tau uptake, as well as the competing processes of degradation and release to the cytoplasm. He was a CFL (Command Fitness Leader) which is basically a personal trainer in the Navy... People who failed the Navy's body fat requirements would end up on a mandatory program designed to help them lose body fat and it was ran by the CFL. He had been trying to help the people assigned under his program but was not receiving very good results... He remembers one guy specifically that showed up for the final weigh in to either fail the Navy's requirements which could mean he would get kicked out of the Navy... His face was sucked in around his eye sockets... He looked pail... But he still had a huge stomach... He looked so week that if I pushed him lightly he would fall over... He was very nervous as he stepped on the scale... He then had his waist and neck measured.... He had failed again... The poor guy had starved himself 2 weeks leading up to this day and hadn't drank any water for 1 week... He spent the last 4 hours sitting in the sauna... All was for nothing... The CFL later realized that he wanted to figure out a way to help people better under him program... He looked into the nutrition options available for people in the Navy and found the 8 week Ship Shape program... He called the hospital and enrolled... It turned out to be an amazing course on weight loss and even though he wasn't trying to lose weight himself, he learned a ton about it.... For the next 4 years he went through many different courses on weight loss and body building... He became hooked on learning more and more.... Until finally one day he knew it so well it was like breathing... See how I lost 35 pounds in 2 months before I knew about the Internalization method... At this point I was at 195 pounds which was pretty heave for me because I normally sat around 160... My clothes started to fit really tight and I start to develop this ugly looking 'S' shape... Meaning my stomach was sticking out one way and my but was sticking far out the other way... This was actually causing me to have back pain when I sleep I think because my but was getting so big that it was putting pressure on my back... The worst part of all was when I developed the heart burn.. It hurt so bad that I ended up having to go to the doctor to get prescribed medicine to treat it... Not to mention people kept making comments of how fat I was looking... I got to this point because I wanted to bulk up and build muscle but the opposite happened... I ended up building fat instead... I really was trying to look like Brad Pitt from Troy but instead I looked like the marshmallow man... I ended up taking a 20% deficit in calories to cut the fat down because I was tired of living like this... In order to do this I had to take in 1,000 less calories then I was currently taking in on the surplus I was doing... I was at 3,500 calories and now I was doing 2,400 calories... The hunger pains immediately hit me and they felt like a knife stabbing me in the stomach... That definitely brought on the cravings for things like pizza, cookie dough ice cream, cookies, brownies, etc... The worst part was the weeks where I didn't lose any weight at all... I hate wasting time and when you go through all this hard work and don't see any results that is like the worst feeling in the world... I eventually got back down in body fat but the twist was I ended right back up where I was before... Same body fat percentage and same weight... I didn't gain any muscle after 6 months of work... I did eventually learn why though... I learned that I wasn't tearing my muscles correctly in the gym... Eventually I acquired what I call the Internalization Method where you lose weight without thinking about it... It's almost like breathing... I was in the gym working out with my buddy like we had been doing for months prior... We looked about average with no amazing results... The gym was really an open room style.... It had a wall of nothing but windows in the back so it was really well lit up... It was a huge open floor plan with really new equipment everywhere... Then I remember seeing this guy walk in with one of those low cut tank tops on... And he looked good! Do you control knowledge or does knowledge control you? Don Miguel Ruiz, Jr. is a Toltec Master of Transformation and the son of Don Miguel Ruiz, author of The Four Agreements discusses how we have unwittingly made agreements about how we choose to live our lives and that these agreements are actually attachments -- limiting filters on who we think we are. According to Ruiz, "When we believe in something so strongly that we lose awareness of our authentic self within the stories and comments of our internal narrators, we are allowing our preconceptions to make decisions for us. Therefore, it is important to be aware of where we are on the scale of attachment with any particular belief. With awareness we can regain the power to make our own decisions." He explains that our attachments can be divided into five levels -- The Authentic Self, Preference, Identity, Internalization, and Fanaticism and discusses how we can re-evaluate these potentially harmful attachments.
