Unlocking the Secrets of the Six-Base GenomeEpigenetics was somewhat vague when I was an undergraduate ( a long time ago). So I was curious to get an update on how we can investigate it more closely and what we are learning. I talked to Tom Charlesworth, Director of Market Strategy and Corporate Development at biomodal, a sequencing technology company focused on epigenetics. Tom explained how modifications beyond the traditional four DNA bases impact gene expression, development, and disease. What is the Six-Base Genome?Tom introduced biomodal as a sequencing technology company spun out of the University of Cambridge, focused on the interface between genetics and epigenetics. Their technology goes beyond the traditional four-base genome (A, T, G, and C) by adding two epigenetically modified bases: methylcytosine (5-MC) and hydroxymethylcytosine (5-HMC). This “six-base” approach captures critical modifications that play distinct roles in gene regulation.5-MC is associated with repressing gene expression, often keeping certain genes “turned off,” while 5-HMC is linked to opening chromatin and activating gene expression. Understanding these modifications provides a more dynamic picture of how our genes are regulated—not just by the sequence of DNA but also by chemical marks that change over time.Bridging the Gap Between Genetics and FunctionThe traditional four-base genome gives us an invaluable map of our genetic code, but it falls short of explaining how the same genetic sequence could lead to such diverse outcomes—from development to disease. Epigenetic modifications, like 5-MC and 5-HMC, offer another layer of regulation that's essential for gene expression. Tom highlighted research that illustrates the value of this additional information. He mentioned the work of developmental biologist Emily Hodges, who uses the six-base data to study chromatin accessibility during neuronal stem cell differentiation. Emily found that early changes in 5-HMC could predict chromatin opening, an insight that would be invisible if one only looked at 5-MC. The “rebaptizers” or Anabaptists in Germany. Almost a curse word.  We fought the wrath of both the catholics and the Lutherans as they held to a very traditional understanding that the sprinkling of water made you a “Christian”. The act of faith was not needed or involved in the act of salvation. Gal.5:6 says circumcision is nothing, only faith expressing itself through love!A bold start: The extent of John the Baptist's ministry: at least from Egypt to Turkey.  The preparation for Jesus to come was significant. But it did need to be followed up with the introduction of Jesus into the lives of those who who had repented.  Just acknowledging the truth that we are sinners is not enough. Paul's approach to Jews, Gentiles, and now disciples of John.The event with these disciples of John – who believed in the One God! – shows us several important aspects of what receiving Jesus into our lives and being empowered by the Holy Spirit looks like in the lives of believers:The two events of baptism in water (into the death and resurrection of Jesus) and being baptized with the Spirit, can be separated by time, though not necessarily essential.  We do not know when the disciples were baptized in water, but we know when Paul was baptized in the Spirit and then in water in Acts 9:17. Cornelius was baptized in the Spirit and then in Water.  The Eunuch was baptized in water and we know nothing after that. Paul only baptized a few in Corinth but it is not recorded in Acts, but they obviously were actively using the gifts of the Holy Spirit. Luke sketchily records the events, not the teaching about why, function, usage, etc. for that we turn to the letters of the NT.  We do know that Jesus empowers his disciples to use the gifts he gives them.Who has the power to lay on hands? Disciples, Ananias, Timothy, and Hebrews is a foundational teaching not just for special people.All gifts from God are valuable and serve a functionThe baptism in the Spirit is accompanied by the Gifts, Fruit, Presence, and Activity of the Holy Spirit. He never changes, he is God! Those who say that the gifts stopped with the Bible completion have no biblical evidence for that.  How can they think that and then not say the same for the fruit of the Spirit? Or the functions of the Spirit listed in Romans 12:6-9 If you want to learn about the variety of gifts, then I am happy to do that on Wednesday nights where we can discuss and ask questions and share experiences. Acts only shows what happens.There is a difference between phenomena and repeated and taught matters in Scripture.  For example, the tongues of fire is phenomena and not repeated.  God can do it, but that would not be something one can expect to happen. The gifts taught in scripture can be expected to be a part of normal Christian life and used for the benefit of others, not to puff oneself up with pride. Focusing on building a firm foundationSeparating the unbelievers from believers to invest in: for understanding the Scriptures, for training in righteousness, for correction, 2 Tim 3:16Modeling and mentoring and releasing into ministryFulfilling the task, especially if you think Jesus is coming soon!  That is the attitude we all must haveThe fruitfulness of God's methodThe disciples were able to spread the Gospel through out their region!The investment was significant and life changingThe true power of the Gospel is evident in the fact that God is magnified by real growthWho do you invest your Christian life into?

Leaky Gut: Facts and FadsThe latest fad of supplement makers is to talk about "gut health." The latest boogyman for the gut is the "leaky gut." The other name for leaky gut is intestinal permeability.A Surgeon's ViewAs a surgeon, a "leaky gut" is a devastating surgical emergency. Surgeons emergently operate to remove the gut that no longer has a barrier function.The dark gut is dead and needs to be surgically removed. This gut has no barrier function and will allow bacteria from the inside of the small bowel to easily enter the patient's bloodstream. Leading to septic shock and death.The Non-Surgical Emergency or Chronic Leaky GutBut a leaky gut doesn't have to be so dramatic. Not leading to septic shock or needing to be removed. In this model, the intestinal barrier is "leaky," not to the point of causing sepsis. But the leaky gut enables small molecules to leak out of the gut into the bloodstream. Thus producing chronic inflammation.Chronic inflammation may cause obesity. Emphasis on "may."Emulsifiers and ObesityRodent studies have provided fascinating insights. Certain emulsifiers, when added to the diet of mice or rats, lead to obesity. In one experiment, rodents were fed identical amounts of kibble. But one group, emulsifiers, was added to their water. These emulsifiers, like polysorbate 80, not only produced obesity but also disrupted the microbiome of the gut (ref). But mice are not men.When dietary emulsifiers are examined in humans, high levels cause disruption of the gut. However, there is little evidence that small amounts found in the food supply cause issues. (ref 4)Non-Humans and Leaky GutWhile animal models are interesting, human studies provide useful information. That a rat or mouse has a leaky gut based on some intervention is not an equivalence in humans.Functional Medicine and Leaky GutIf you searched "leaky gut," you will find a host of "functional medicine" doctors providing supplements, bone broth, probiotics, and other scams. On the shelves of drug stores and "health food stores," you will find abundant supplements to "support gut health."Disease States and Leaky GutFunctional medicine physicians associate many disease states with leaky gut. Celiac disease and Crohn's disease have been studied for years (ref 2, ref 3). She is the bestselling author of her memoir My Stroke Of Insight which recounts her experience and recovery after a severe stroke, which left her unable to walk, read, write, or recall any of her life.
