Biomedical researchers have long sought ways to repair spinal cord damage with the holy grail of the pursuit being the reconstitution of lost function. In the mid 1990's with the successful culture of human embryonic stem cells, and about a decade later induced pluripotent stem cells (iPSCs), the field was energized with a potential new approach to replace the lost neurons and glia cells and restoring neural connections.  In the decades since that discovery some progress has been made, however many hurdles remain, including establishing a functional synaptic connection between the transplanted and host neurons which is crucial for motor function recovery. To boost therapeutic outcomes our guests tested an ex vivo gene therapy to promote synapse formation between the donor and host neurons by expressing the synthetic excitatory synapse organizer CPTX in hiPSCs-derived neural stem and progenitor cells. This episode focuses on social constructionism with one of the leading figures in the field, Kenneth J. Gergen Ph.D. We hear about how radical the ideas were at the time of its conceptiopn and the people who believed in its contribution to psychology and supported its evolution. Ken tells us his personal and professional stories of the history and development of his key ideas within social constructionism alongside some of the challenges when sharing with the wider field.  Ken discussed the key elements of its application in the therapeutic context and his futures hopes. Bio (taken from Taos Institute website)Kenneth J. Gergen, Ph.D., is a founding member, President of the Taos Institute and Chair of the Board, and the Mustin Professor of Psychology at Swarthmore College. Gergen also serves as an Honorary Professor at the University of Buenos Aires. Gergen received his BA from Yale University and his PhD from Duke University, and has taught at Harvard University and Heidelberg University. He has been the recipient of two Fulbright research fellowships, the Geraldine Mao fellowship in Hong Kong, along with Fellowships from the Guggenheim Foundation, the Japanese Society for the Promotion of Science, and the Alexander Humboldt Stiftung. Gergen has also been the recipient of research grants from the National Science Foundation, the Deutsche Forschungsgemeinschaft, and the Barra Foundation. He has received honorary degrees from Tilburg University and Saybrook Institute, and is a member of the World Academy of Art and Science. Gergen is a major figure in the development of social constructionist theory and its applications to practices of social change. He also lectures widely on contemporary issues in cultural life, including the self, technology, postmodernism, the civil society, organizational change, developments in psychotherapy, educational practices, aging, and political conflict. With Mary Gergen, he published an electronic newsletter, Positive Aging (www.positiveaging.net) distributed to 20,000 recipients. Gergen has served as the President of two divisions of the American Psychological Association, the Division on Theoretical and Philosophical Psychology, and on Psychology and the Arts. He has served on the editorial board of 35 journals, and as the Associate Editor of The American Psychologist and Theory and Psychology. His laboratory published the 2015 paper, Precise correction of the dystrophin gene in Duchenne muscular dystrophy patient induced pluripotent stem cells by TALEN and CRISPR-Cas9. Thomas Rando is the Director of the Eli and Edythe Broad Center of Regenerative Medicine and Stem Cell Biology at the University of California Los Angeles, where he is also Professor of Neurology and of Molecular, Cell and Developmental Biology. Dr Rando is a board-certified neurologist and a member of the National Academy of Medicine and a member of the American Academy of Arts and Sciences. He is also the 2023 recipient of the ISSCR Achievement Award, which recognizes the transformative body of work of an investigator that has had a major impact on the field of stem cell research or regenerative medicine. Dr. George Hajishengallis is a Professor in the Department of Microbiology at the University of Pennsylvania School of Dental Medicine. Through his research, George is trying to understand how the immune system interacts with oral bacteria. He is curious about why you see destruction of gums and the bone that supports the teeth in some people. They are also working on treatments to prevent gum disease called periodontists. Outside of his work life, George spends his time watching soccer, reading books, and hiking and swimming with his wife. He received his Doctor of Dental Surgery Degree from the University of Athens in Greece and his PhD in Cellular and Molecular Biology from the University of Alabama at Birmingham. Following postdoctoral studies in Immunology at the University of Alabama at Birmingham and the University at Buffalo, George served on the faculty at Louisiana State University Health Science Center and the University of Louisville before joining the Faculty at the University of Pennsylvania. George has been elected as a Fellow of the Japanese Society for the Promotion of Science and was a recipient of the Distinguished Scientist Award in Oral Biology from the International Association of Dental Research (IADR) in 2012 and the AADR/IADR William J. Gies Award in the Biological Research Category in 2014. In this interview, George shares more about his life and science.

This is the concept of denying trauma – even from awful life events – and recognising your have the choice on how to respond to it.  For instance, just because something bad happened to you it doesn’t mean that your life will be bad as a result of it. We often see this in our own lives, or those around us today, but by denying the idea of determinism, we realize that our past does not determine our future. In some cases, we behave in a certain way to achieve a goal or live according to other people’s view. One of the biggest points shared in this book is that most of us lack the courage to be happy – because it requires change. This is why some people choose to live a miserable or unhappy life because being happy requires challenging changes. BIG IDEA 2 (6:24) – Own your tasks. Each one of us has our own tasks and we should not interfere with other people’s tasks. These three tasks are work, friendship and love. One of the most important ideas here is not looking for or seeking recognition from others – just focusing on your own tasks and our contribution to others/society. If we need other people to interfere with us, recognize us or celebrate our achievement, we are worried about what other people think about us and not who we truly want to be. This idea comes down to freedom, or the courage to be disliked. This is about not needing other people’s recognition, reward or validation to feel like we have contributed. Focus on your own tasks – work, love and friendships. BIG IDEA 3 (7:33) – All problems are all interpersonal relationship problems. Most interpersonal relationship problems (think comparison, jealousy, imposter syndrome, regret etc) are solved by first accepting yourself as you are. When we do not accept ourselves, sometimes this is used as an excuse to dislike or make assumptions about what others think. The great idea here is that you are the only who is worried about you!  If we think about it, when we are only worried about ourselves, we can do more of what we really want to do. One great quote shared in the book is “We cannot alter objective facts but subjective interpretations can be altered as one likes.” Most of the things around us are purely subjective, so if someone does not like something, it’s their opinion and doesn’t matter. When it began services on 1 October 1964, in time for the Tōkyō Olympics, it heralded a new age in railway transport and since that time the shinkansen network has continued to grow with many changes since the break up and privatisation of the railways in 1987. Meanwhile the aviation market has also continued to evolve, with ANA becoming Japan's biggest airline in recent years, the construction and expansion of airports, and the introduction of Low Cost Carriers. But there is more to these trains and planes than merely means of transportation; they also reflect a range of aspects of Japanese society. This paper will look at a range of issues relating to their design and usage that will reveal ways in which they can help us to understand Japanese society.

This senior-junior culture, underlies practically all relationships in Japanese society such as schools, offices, clubs, organizations and almost any other setting where groups are involved. Who is the senpai and who is the kohai is determined by the time you enter an organization. It is more than just a rank where older members receive special treatment. Senpais are mentors and are responsible for their kohais, and should take this responsibility very seriously. If a kohai screws up, the senpai will also feel embarrassed and responsible.One of the main things that observers will notice is the way that the senpai and kohai communicate. The senpai usually uses casual Japanese whereas the kohai will always use polite Japanese (keigo, teinego, or sonkeigo).For foreigners, it can be difficult to fully appreciate the nuances of this relationship, so having Yuki on will hopefully shed some light on this fascinating topic. About the Authors Ichiro Kishimi was born in Kyoto, where he still lives, in 1956. He has aspired to become a philosopher since his days in high school. Since 1989, while specialising in Classical Western philosophy, with a special focus on Platonic philosophy, he has researched Adlerian psychology; he writes and lectures on the subject, and provides counselling for “youths” in psychiatric clinics as a certified counsellor and consultant for the Japanese Society of Adlerian Psychology.   Fumitake Koga, an award-winning professional writer and author, was born in 1973. This is why some people choose to live a miserable or unhappy life because being happy requires challenging changes.   BIG IDEA 2 (6:24) - Own your tasks. Each one of us has our own tasks and we should not interfere with other people’s tasks. These three tasks are work, friendship and love. One of the most important ideas here is not looking for or seeking recognition from others - just focusing on your own tasks and our contribution to others/society.   If we need other people to interfere with us, recognize us or celebrate our achievement, we are worried about what other people think about us and not who we truly want to be.   This idea comes down to freedom, or the courage to be disliked. This is about not needing other people’s recognition, reward or validation to feel like we have contributed. Focus on your own tasks - work, love and friendships.   BIG IDEA 3 (7:33) - All problems are all interpersonal relationship problems. Most interpersonal relationship problems (think comparison, jealousy, imposter syndrome, regret etc) are solved by first accepting yourself as you are. When we do not accept ourselves, sometimes this is used as an excuse to dislike or make assumptions about what others think.   The great idea here is that YOU are the only who is worried about you!  If we think about it, when we are only worried about ourselves, we can do more of what we really want to do. One great quote shared in the book is “We cannot alter objective facts but subjective interpretations can be altered as one likes.” Most of the things around us are purely subjective, so if someone does not like something, it’s their opinion and doesn’t matter.   The book is very clear that is not about doing things that would purposely hurt others, but that the subjective opinions of others aren’t as important and defining as we often make them out to be.   Welcome to another episode of Biotechnology focus radio! I am your host – Michelle Currie – here to give you the rundown on what is happening in the life sciences sector from coast to coast. Keep listening to find out more!  +++++  As regenerative medicine grows around the world, topping a whopping $36-billion annually and only expected to rise, it comes as no surprise that more and more international collaborations are happening – especially within Canada.  CCRM and the Japanese Society for Regenerative Medicine (JSRM) liaise to advance the field of regenerative medicine (RM) and cell and gene therapies in Canada and Japan, signing a Memorandum of Understanding (MOU) this week at the Annual Meeting of the Tissue Engineering and Regenerative Medicine International Society (TERMIS) in Kyoto, Japan.  Michael May, president and CEO, CCRM says that CCRM’s mission is to generate sustainable health and economic benefits through global collaboration in cell and gene therapy, and regenerative medicine. CCRM is catalyzing a global network of highly integrated commercialization centres working together to enable viable and cost-effective patient access to revolutionary new treatments. The Memorandum of Understanding with Japanese Society for Regenerative Medicine, through its vast research network and industry-enabling activities, is a positive step in that direction.”  The Memorandum of Understanding has been put in place to promote academic and industry partnership in Japan, Canada and internationally to advance the field of regenerative medicine and cell and gene therapies. This will include supporting knowledge translation about technologies, policies (e.g., regulatory and health economics), legal and ethical issues.  Prof. Sawa, president of Japanese Society for Regenerative Medicine.  Says that “There are many obstacles to establish a sustainable business model for regenerative medicine in Japan, as it requires a whole new value chain. Canada’s CCRM has been fostering and promoting a successful commercialization model since its launch. JSRM is proud to announce that we have entered into a partnership with CCRM to develop sound industrialization pathways, learning from CCRM’s excellent model to make regenerative medicine an available treatment worldwide.”  Regenerative medicine – that can be a bit of an umbrella term – includes cell and gene therapy, stem cells, biomaterials, molecules and genetic modification to repair, regenerate or replace diseased cells, tissues and organs. This approach is disrupting the traditional biotechnology and pharmaceutical industries with the promise of revolutionary new cures for devastating and costly conditions such as heart disease, diabetes and cancer.  This sector represents so many potential untapped possibilities. Forecasted to grow to US$49.41-billion by 2021, there were 977 clinical trials in cell, gene and tissue therapy underway worldwide at the close of the second quarter of 2018. The sector achieved the first global approvals and reimbursements for major cellular immunotherapies and gene therapies in 2017, that resulted in record-breaking investment and acquisitions in the sector. This field encapsulates the phrase “the world is truly their oyster”.  +++++  The Centre for Drug Research and Development, Canada’s national life sciences venture, announces the first cohort of the CDRD Academy’s Executive Institute.  Earlier this year, CDRD and Pfizer Canada announced the launch of the Executive Institute under the umbrella of The CDRD Academy. The Institute is a 10-month, focused executive development program open to a limited number of senior-level life sciences professionals annually. The first face-to-face session will happen in Vancouver September 12-13, 2018, with additional workshops to be held in Montreal and Toronto throughout the Winter and Spring 2019.  +++++  Concordia synthetic biology researchers develop a method to fight disease at a genetic level that may revolutionize patient care.  Steve Shih, an assistant professor of electrical and computer engineering in Concordia University’s Faculty of Engineering and Computer Science and with a cross-appointment in biology, is also the founder of the Shih Microfluidics Laboratory.  His team created a system that integrates the automation of complex biology experiments in order to find genes that are related to cancer and kill them before they develop into a potentially fatal disease.  The system is described in a paper published last July by the journal Lab on a Chip.  Shih says that “Finding genes related to cancer is already very difficult. It’s like finding a needle in a haystack, especially with current methods. But hopefully, with this new method, we can expedite the whole process and rapidly find the culprit genes.”  However, finding the genes is one thing. Preventing them from causing cancer is another.  To do that, Shih’s team uses CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) a genetic engineering technique that uses a Cas9 protein (essentially a pair of ‘programmed genetic scissors’) to find a cancer-causing gene and essentially snip it out of the DNA and replace it with a healthier one.  Shih says that “Once both ends of the gene are snipped, it degrades to the point where we won’t have it anymore. Now that gene won’t be able to go through pathways that cause cancer. To be able to do that on a typical platform is very difficult, because we’re dealing with very static, very manual techniques.  “By doing this in an automated way, and by also miniaturizing the scale which we’re working on, we’re able to expedite the whole process. Instead of looking at this process as a matter of weeks, we can look at it in a matter of days.”  The paper’s authors — Hugo Sinha, Angela Quach, Philippe Vo and Shih, all affiliated with Concordia’s Centre for Applied Synthetic Biology — created the first digital microfluidic method that automated arrayed gene editing in mammalian cells.  This involves using tiny amounts of fluid to culture lung cancer cells for up to six days, while at the same time automating gene transfection and knockout procedures.  The whole automation and miniaturization process that they have developed has not only saved them heaps of time, but it has also slightly augmented the efficiency of the knockout procedure itself.  The mission to eradicate cancer has been a personal aspect for Shih, and he believes his work will have direct material benefits for people diagnosed with cancer.  He hopes his project will contribute to the development of personalized platforms for treatment based on their genetic makeup. That platform would be easily transferable and can be set up in any kind of lab or hospital.  In fact, Sinha started a company called DropGenie that will create gene-editing platforms that can bring this idea to realisation.  Despite CRISPR being controversial, Shih believes that only now researchers are reaping its benefits.  He adds that “There still isn’t a killer application for microfluidics, but I think we’ve found it. I think we found that we can use these miniaturized platforms for something that can really save people’s lives down the line. That’s why I say it can be a killer app because hopefully, we’ll be able to eventually kill all cancer cells.”  +++++   Researchers from the Peter Munk Cardiac Centre (PMCC) at UHN suggest that the model used by the Canadian Institute for Health Information (CIHI) significantly underestimates mortality in specialized heart surgery centres. The study’s findings show that CIHI’s model does not encapsulate all the medical problems that patients have when at elevated risk for surgery.  The study, looked at the outcomes of 1,635 cardiac operations performed at the Peter Munk Cardiac Centre between 2013 and 2016. It compared the observed mortality within 30 days of surgery to the predicted mortality rates estimated with either the CIHI administrative data model or the clinical data model used by the Society of Thoracic Surgeons (STS).  Of the 1,635 patients that had heart surgery at the Peter Munk Cardiac Centre, 32 died within 30 days of surgery. These results were in line with the 1.96 per cent mortality predicted by Society of Thoracic Surgeons model – which uses comprehensive data to describe how sick patients are before surgery – for this group of patients. In contrast, the CIHI Cardiac Care Quality Indicator estimated that the mortality rate for these 1,635 patients would be 1.03 per cent.  Peter Munk Cardiac Centre researchers noted that the Society of Thoracic Surgeons model captures seven medical conditions that predict worse outcomes after heart surgery that are not captured in the CIHI model.  These risk factors include whether the patient had heart failure, an abnormal heart rhythm, a recent heart attack, very low blood pressure (shock), needed recent CPR or a mechanical heart pump to live, or had kidney failure. These serious conditions forecast a worse outcome after heart surgery, and are not included in the CIHI database.  Dr. Barry Rubin, medical director of the Peter Munk Cardiac Centre and one of the authors of the study  says that “The predicted mortality of patients undergoing heart surgery based on the Society of Thoracic Surgeons model is similar to what we actually observed,” says. “The failure to include these seven medical conditions causes the CIHI database to underestimate predicted mortality after heart surgery in high risk patients.”  According to Dr. Douglas Lee, senior scientist at the Peter Munk Cardiac Centre and lead author of the study, mortality prediction models – either based on clinical or administrative data – use risk adjustment to account for how sick patients are before surgery at different hospitals. This is necessary as outcomes may vary if leading academic institutions such as the Peter Munk Cardiac Centre operate on sicker patients.  Dr. Lee says that “The CIHI and Society of Thoracic Surgeons models aim to predict outcomes based on the medical complexity and acuity of the patient. In general, academic hospitals take on higher risk cases compared to community hospitals, and good risk adjustment models should factor that in when predicting mortality rates,” explains Dr. Lee.  Incorrect data may lend the impression that there is a higher level of mortality then there should be at the PMCC or other academic centres. This could have the inadvertent effect of causing the highest risk surgery patients to defer potentially-life saving heart surgeries.  CIHI has a legislative mandate to publicly release Cardiac Care Quality Indicator data and has done so since October 2017. Clinicians and researchers have been concerned that the CIHI model may underestimate how sick cardiovascular surgery patients are at Peter Munk Cardiac Centre.  Incorrect data may lend the impression that there is a higher level of mortality then there should be at the Peter Munk Cardiac Centre or other academic centres. This could have the inadvertent effect of causing the highest risk surgery patients to defer potentially-life saving heart surgeries.  The Society of Thoracic Surgeons model considers vital medical conditions to accurately predict how high-risk patients will do after surgery. The CIHI model does not accurately account for complexity of patients, which is recorded in databases specifically designed for the measurement of surgical quality, such as Society of Thoracic Surgeons.  Dr Rubin says that “Clinical data-based models like the Society of Thoracic Surgeons collect much more detailed patient information, but are also more costly to maintain. “Administrative models like CIHI’s continue to play a very important role in assessing quality of care across Canada. We will continue to work in collaboration with CIHI to improve the accuracy of quality report cards that can be used as valid evaluation tools for Canadian hospitals.”  The authors caution that there are limitations to the study, as it was completed in a single centre during a three-year period, observing 1,341 isolated coronary bypass grafts, 143 isolated aortic valve replacements and 151 combined procedures. PMCC researchers declared there were no conflict of interests but would suggest validation of these findings in other academic centres over a longer time interval that would include a larger subset of cardiac operations. Yoshiharu Nakagawa, PhD., is a professor of education at Doshisha University, in Kyoto, Japan. His current interests include holistic education, spirituality, transpersonal psychology, Eastern philosophy, Indian philosophy, and contemplation. He teaches holistic education and contemplative education at his school, and is active in the Japanese Society for Holistic Education & Care, and the Japanese Association of Transpersonal Psychology & Psychiatry. He is also one of the founding members of the Asia-Pacific Network of Holistic Education. He is the author of Education for Awakening: An Eastern Approach to Holistic Education (2000) and the co-editor of Nurturing Our Wholeness: Perspectives on Spirituality in Education (2002), in addition to contributing articles to many journals and anthologies.His translator:Sachiko Gomi, PhD. Dr. Rob Johns is a Research Scientist and Forest Insect Ecologist for the Natural Resources Canada Atlantic Forestry Centre. He is also an Adjunct Professor in Biology at the University of New Brunswick and St. Francis Xavier University. Rob received his undergraduate training in biology from St. Francis Xavier University and PhD in Biology from the University of New Brunswick. He was then was awarded a Postdoctoral Research Fellowship from the Japanese Society for the Promotion of Science before joining the Canadian Forest Service where he is today. Rob is here with us today to tell us all about his journey through life and science.

Dr. Park talks about the origins of MegaGen, the first revolution of implantology, and why a second one has already happened.   Dr. Kwang Bum Park graduated Kyung Pook National University, School of Dentistry of Korea in 1985. He finished his residency in periodontics in 1988. After three years of academic teaching and research at Kyung Pook National University, he has been practicing periodontic and implant surgery since 1993. He founded a private dental institute ‘Perio-Line,’ and trained hundreds private practitioners on the topic of clinical perodontics and implant dentistry. He was a visiting scholar at the UCLA School of Dentistry, Department of Periodontics in 2000. In 2001, as a founding member, he started a dental hospital network called ‘MIR Dental Network’ which now compises of 21 large multidisciplinary dental hospitals throughout the Korea. He is presently CEO and Chairman of an implant manufacturing company ‘MegaGen’ which he started in 2002. He is the director of MINEC Dental Education Center which provides continuing education to practicing clinicians on the topics of implant dentistry. Dr. George Hajishengallis is a Professor in the Department of Microbiology at the University of Pennsylvania School of Dental Medicine. He received his Doctor of Dental Surgery Degree from the University of Athens in Greece and his PhD in Cellular and Molecular Biology from the University of Alabama at Birmingham. Following postdoctoral studies in Immunology at the University of Alabama at Birmingham and the University at Buffalo, George served on the faculty at Louisiana State University Health Science Center and the University of Louisville before joining the Faculty at the University of Pennsylvania. George has been elected as a Fellow of the Japanese Society for the Promotion of Science and was a recipient of the Distinguished Scientist Award in Oral Biology from the International Association of Dental Research (IADR) in 2012 and the AADR/IADR William J. Gies Award in the Biological Research Category in 2014. George is here with us today to tell us all about his journey through life and science.

Project A-Ko This amazing movie from 1986 remains one of Eeeper's favourite anime of all time and set a benchmark for parodies of anime and manga to come. Captain Harlock, Maison Ikkoku and Fist of the North Star are some of the titles that get poked to death in this treat. Eeeper goes all over the world for sport in this review before actually getting to the PLOT. Try and stay with him on this one, it does pay off. Central Park Media released this movie along with the OVA sequels comprising of the DVD volumes Love and Robots and Uncivil Wars. Oni watched Uncivil Wars but Eeeper didn't so only the first film and part of the OVA's are covered. This film was one of three VHS available to rent from 24-Hour Video in Eeep's hometown (the other two being Giant Robo Part One of the OVA and StreetFighter II: The Animated Movie) so it got serious playtime in the Eeep household. The new Japanese history textbook by the revisionist Tsukuru-kai is at the center of a debate on neo-nationalism and the growing influence of right-wing intellectuals in Japan. This is part of an agenda that calls into question many assumptions on which post-war Japanese society has been built. As Hidetsugu Yagi, president of Tsukuru-kai until recently, pointed out, from the society's point of view "history textbooks are bad, but civic textbooks used in Japanese middle schools are worse" So far, the "New Civic Textbook" (Atarashii komin kyokasho) for middle schools has received less scholarly attention than the notorious history textbook.This paper will introduce the Civic textbook and its main assumptions about present and future Japanese society. In many ways the textbook tries to explain what makes "a good Japanese citizen" and therefore can serve as a key to understanding revisionist thinking on contemporary Japan. The analysis of texts and illustrations brings to light an image of Japanese society threatened from within and from outside. The paper focuses on how social change is dealt with in the "New Civic Textbook" and what solutions its narrative implies when addressing dramatic demographic challenges, changing family patterns and gender roles.Klaus Vollmer holds an M.A. and a PhD (1993) in Japanese Studies, Hamburg University. Post-doc research fellow Osaka City University, (prohibitions of killing and meat-eating in pre-modern Japan). Since 1998, chair of Japanese Studies, Japan Center Munich University; Numata-Fellow for studies in Japanese Buddhism in 2002. Fields of research and teaching include cultural and social history of Japan (both pre-modern and modern), focusing on representations and interpretations of Japanese culture. The topic of the presentation at the DIJ derives from a long-standing interest in issues of historical revisionism and historiography in Japan and Germany and its implications for images of and attitudes towards society and its norms.Since 2000, Klaus Vollmer is president of the German Association for Social Science Research on Japan (VSJF).
