Against the backdrop of debates around 'success' and 'failure' in anti-corruption, they talk to Professor Robert Barrington about approaches to Monitoring, Evaluation and Learning (MEL). The discussion covers the basics of what MEL is and the role it has to play in building knowledge on anti-corruption effectiveness; the current state of practice for MEL; what can be learnt from other sectors; and what changes might be needed to shift towards more adaptable, learning-focused approaches. Marius Cruceru este soț, tată și bunic, roluri cu care îi place să se mândrească. Pe lângă asta, este prof. univ. dr. și predă în cadrul Universității Emanuel din Oradea. De aproape trei ani, împreună cu soția sa, Natalia, au inițiat proiectul Casa Păcii și Grădina împăcării. Acest proiect este alcătuit, de fapt, din două case în care cuplurile aflate în impas au posibilitatea de a se izola și de a beneficia de ajutorul specialiștilor, pentru a-și restaura căsnicia. Acesta este un proiect anti divorț. Știu că multă lume se luptă cu gândul divorțului, chiar dacă nu verbalizează și nu acționează în consecință. Adeseori, luptele zilnice sunt puternice, ne simțim neînțeleși, lucrurile nu se desfășoară așa cum ne-am imaginat, sacrificiile par prea multe și prea dese doar de partea noastră, strigăm către Dumnezeu și parcă nimic nu trece de tavan, ceea ce ne duce aproape de prăpastia disperării. Prăpastie care naște în noi dorința separării în speranța că asta va aduce pacea, calmul, liniștea. Acest episod este o verbalizare a frământărilor pe care poate nu ai avut curaj să le spui nimănui. Pentru mine și pentru tine am pus cărțile pe față, iar domnul Marius Cruceru răspunde extrem de clar și direct la multe din întrebările care se ascund de atâta vreme sub preș. Ce fac în caz de adulter, infidelitate? Trebuie să stau, pot să rămân? Și dacă a plecat, să mă recăsătoresc? Ce fac cu copiii? Aleg separarea, îi mai dau o șansă? Dar, dacă mă abuzează fizic și nu știu cum să îmi mai ascund vânătăile? Stau să sufăr ca și cum aș face-o pentru Hristos sau pot să plec, să-mi salvez literalmente viața? Unde sunt pastorii/consilierii cu adevărat pregătiți pentru acest gen de provocări și de ce biserica nu intervine, nu vorbește, nu strigă, ci tace? Acest episod este pentru oricine căruia îi pasă de familie și care își dorește ca această instituție să reziste peste ani și ani, chiar și în fața vremurile neprielnice în care trăim. Acest episod este pentru tine și soțul/soția ta. Ascultați-l împreună și relegați mâinile, împreunându-le din nou în legământul căsniciei, dacă pe pragul ușii voastre scrie de ceva vreme DIVORȚ! Sie fand im Club Celeste, im Schikaneder und im Spektakel statt.LIFE BALLDer Ballorganisator Gery Keszler hat im Jahr 1993 den Life Ball ins Leben gerufen und sich zum Ziel gesetzt, mit dieser Benefiz-Veranstaltung Bewusstsein für die Krankheit Aids zu schaffen. Bis 2019 fand diese queere Ballnacht im Wiener Rathaus jedes Jahr mit unzähligen nationalen und internationalen Gäst:innen statt.HEAVENDonnerstags traf man sich von Herbst 1989 bis 1992 im Wiener Nachtclub U4, um die queere Szene am ‚Heaven‘ zu feiern. Ab nun gehts weiter mit den Meilensteinen des Babys. In Folge 64 haben wir schon darüber gesprochen, was denn so alles in den ersten beiden Lebensmonaten passiert. In dieser Folge stehen die Lebensmonate 3 und 4 im Fokus. Dabei gilt für jede dieser Folgen: Jedes Baby ist einzigartig! Auch innerhalb einer Familie. Daher sind Vergleiche mit anderen Babys (im ähnlichen Lebensmona)t oder zum Erstgeborenen nicht ratsam. Somit gelten auch unsere geschilderten Entwicklungen in diesen Folgen nur als Orientierung, weil jedes Baby sein eigenes Tempo hat. Es kann also sein, dass sich ein 3-Monate altes Baby schon drehen und das Köpfchen halten oder auch nicht. Auf jeden Fall nimmt es seine Umgebung wieder etwas aufmerksamer wahr. Es reagiert auf Umgebungsgeräusche und kann auch schon besser räumlich sehen. Die Neugier Dinge zu ertasten nimmt zu. Das lässt sich gut an der Hand-Mund-Koordination beobachten. Dabei ist ganz wichtig, dass Kleinteile nicht in der Nähe vom Baby liegen! Zudem kann zu verstärkten Kopfgneis kommen und evtl. zeigt sich auch der erste Zahn bzw. das Baby fängt an zu zahnen. Auch in diesen beiden Lebensmonaten ist Körperkontakt mit Massagen und Kuscheleinheiten wichtig, um Nähe und Geborgenheit zu fühlen und das Vertrauen mit Bezugspersonen zu stärken. In dieser Zeit steht auch die U4 beim Kinderarzt mit ggf. Impfung an. Mehr dazu dann in dieser Folge, wenn Sunny Andrea fragt: Was passiert denn im 3. und 4. Lebensmonat?

Bei der zweiten Begegnung fällt Rüdiger jedoch auf, dass der vielleicht gerade mal 16-Jährige alles andere als gesund aussieht. Ein paar Tage später erfährt er in einem an ihn gerichteten Brief auch warum. Doch die entscheidende Frage, die sich auch der Verfasser des Briefes stellt, bleibt offen. In der zweiten Geschichte geht es um den Steinmetz und Bestatter William Markland. Eines Tages wird er von einer Kundin mit einem für ihn etwas ungewöhnlichen Auftrag betreut, denn der Grabstein, den William fertigen soll, soll möglichst billig und unscheinbar sein. William ist verwundert darüber, dass jemand an der letzten Ruhestätte am falschen Ende sparen möchte, noch viel mehr wundern ihn jedoch die Änderungswünsche des unbekannten Besuchers am Tag darauf.

Early nomads in the Eurasian steppes since the beginning of the 1st millennium BC played a key role in the formation of the cultural and genetic landscape of populations of a significant part of Eurasia, from Eastern Europe to Eastern Central Asia. Numerous archaeological cultures associated with early nomads have been discovered throughout the Eurasian steppe belt. The Tagar archaeological culture existed in the Minusinsk basin (Sayan Mountains, Southern Siberia, Russia) in the northeastern periphery of the Eurasian steppe belt from the 8th to 1st century BC during the pre-Scythian, Scythian, and Early Xiongnu-Sarmatian periods. In this study, we evaluated mtDNA diversity in the Tagar population based on representative series (N = 79) belonging to all chronological stages of the culture. The Tagar population had a mixed mtDNA pool dominated by Western Eurasian haplogroups and subgroups (H, HV6, HV*, I, K, T, U2e, U4, U5a, and U*) and, to a lesser degree, Eastern Eurasian haplogroups (A*, A8, C*, C5, D, G2a, and F1b). The Tagar population showed a similar mtDNA pool structure to those of other Iron Age populations representing the “Scythian World.” We observed particularly high similarity between the Tagar and Classic Scythians from the North Pontic region. Our results support the assumption that genetic components introduced by Bronze Age migrants from Western Eurasia contributed to the formation of the genetic composition of Scythian period populations in Southern Siberia. Another important component of the Tagar mtDNA pool was autochthonous East Eurasian lineages, some of which (A8 and C4a2a) are potential markers of the westward genetic influence of the eastern populations of the Scythian period. Our results suggest a genetic continuity (at least partial) between the Early, Middle, and Late Tagar populations. In dieser Episode sprechen wir darüber, wie dein Buchcover diesen Job erfüllen kann — und was du tun solltest, wenn du merkt, dass dein Cover nicht funktioniert. A., Barbosa-Morais, N. L., Garinis, G. A. Abstract: RNA splicing, transcription and the DNA damage response are intriguingly linked in mammals but the underlying mechanisms remain poorly understood. Using an in vivo biotinylation tagging approach in mice, we show that the splicing factor XAB2 interacts with the core spliceosome and that it binds to spliceosomal U4 and U6 snRNAs and pre-mRNAs in developing livers. XAB2 depletion leads to aberrant intron retention, R-loop formation and DNA damage in cells. Studies in illudin S-treated cells and Csbm/m developing livers reveal that transcription-blocking DNA lesions trigger the release of XAB2 from all RNA targets tested. Immunoprecipitation studies reveal that XAB2 interacts with ERCC1-XPF and XPG endonucleases outside nucleotide excision repair and that the trimeric protein complex binds RNA:DNA hybrids under conditions that favor the formation of R-loops. Thus, XAB2 functionally links the spliceosomal response to DNA damage with R-loop processing with important ramifications for mammalian development and transcription-coupled DNA repair disorders. Ein kleiner Bericht über den Tag der Großbaustellen in Hamburg. Vor allem geht es um die neue U4 und den Bau der Elbphilharmonie auf historischem Grund. Weiterhin gibt es einen kleinen Ausblick auf das große Event im Mai - den Hamburger Hafengeburtstag.

Ribonukleoproteinpartikel (RNPs) sind Komplexe aus RNA und Proteinen, die entscheidende Funktionen bei Prozessen wie Translation, Telomer-Synthese, Protein-Import in das endoplasmatische Retikulum oder RNA-Prozessierung übernehmen. Obwohl stets neue Beispiele die Bedeutung von RNPs untermauern, sind grundlegende Aspekte ihrer Funktion noch unklar. So stellte sich zu Beginn dieser Arbeit die Frage, wie sich die Komponenten von RNPs zu funktionellen Gebilden zusammenlagern. In frühen in-vitro-Studien war beobachtet worden, dass sich RNPs spontan ausbilden und dieser Vorgang keine weiteren Faktoren benötigt. Daraus war die Hypothese abgeleitet worden, dass dies möglicherweise auch der in vivo Situation entsprechen könnte. Unerwartete Einblicke in die Biogenese von RNPs lieferten schliesslich Studien zum "survival motor neurons"-Protein (SMN), dem Krankheitsgenprodukt der spinalen Muskelatrophie. Antikörper gegen SMN und seinem Bindungspartner Gemin2 inhibierten in Xenopus laevis Oocyten die Ausformung von RNP-Untereinheiten des Spleißosoms - den U snRNPs und nährten den Verdacht, dass diese Proteine Hilfsfaktoren der U snRNP-Biogenese sein könnten. Das Ziel der vorliegenden Arbeit war daher, mechanistische Details über die Zusammenlagerung von U snRNPs in vivo zu ermitteln und die Rolle von SMN und Gemin2 zu untersuchen. Die wesentlichen Schritte der Biogenese von U snRNPs können experimentell in X. laevis Oocyten verfolgt werden. Nach dem Export der U snRNAs U1, U2, U4 und U5 in das Cytosol lagern sich dort jeweils sieben sogenannte Sm-Protein an ein gemeinsames Motiv der U snRNAs an und formen so die Grundstruktur jedes U snRNPs, die Sm-Core-Domäne. Hierauf folgen die Hypermethylierung der U snRNA-Kappe und der Import der Sm-Core-Domäne in den Zellkern, wo sich U snRNP-spezifische Proteine anlagern, ehe die reifen snRNPs am Spleißprozess teilnehmen. In der vorliegenden Arbeit wurde zunächst ein zellfreies System entwickelt, durch das die Zusammenlagerung von U snRNPs in der Komplexität des Cytosols untersucht werden konnte. Unter Verwendung von Extrakten aus Xenopus laevis-Eiern oder HeLa-Zellen konnte gezeigt werden, dass die Ausbildung der Sm-Core-Domäne, entgegen bisheriger Vermutungen, nicht spontan erfolgt, sondern Energie in Form von ATP benötigt. Aus Depletionsversuchen wurde deutlich, dass SMN unter diesen zellähnlichen Bedingungen für die snRNP-Biogenese unbedingt erforderlich ist. SMN, dies zeigten immunbiochemische Reinigungen, ist in der Zelle mit 17 verschiedenen Proteinen assoziiert, die hier erstmals vollständig identifiziert wurden. Dieser SMN-Komplex enthält bereits alle Sm-Proteine, jedoch keine U snRNAs. Anhand direkten Sm-Protein-Transferstudien wurde klar, dass der SMN-Komplex allein nicht nur notwendig sondern auch hinreichend für die Ausbildung der Sm-Core-Domäne, ist. Dennoch konnte mit dem pICln-Komplex ein Proteinkomplex entdeckt werden, der mit dem SMN-Komplex interagiert und dessen Aktivität erheblich steigert. Der pICln-Komplex enthält eine neuartige Methyltransferase, die Arginylreste in den Sm-Proteinen B/B’, D1 und D3 zu symmetrischen Dimethylargininen modifiziert. Es ist bekannt, dass hierdurch die Bindung von Sm-Proteinen an SMN verstärkt wird. Die vorliegenden Daten weisen darauf hin, dass SMN- und pICln-Komplexe eine funktionelle Einheit bilden, in der Modifikation und Transfer der Sm-Proteine koordiniert ablaufen. Erste Erkenntnisse aus Versuchen mit HeLa-Zellen und Patientenzelllinien deuten an, dass reduzierte Menge des SMN-Komplexes mit einer reduzierten U snRNP-Zusammenlagerungsaktivität einhergehen, und dass dies einen biochemischen Defekt in Spinaler Muskelatrophie darstellen könnte. In einem weiteren Projekt wurde mit Hilfe von Datenbankanalysen und biochemischen Strategien das SMN-homologe Protein SMNrp identifiziert und charakterisiert. Biochemische Studien zeigten, dass SMNrp eine Komponente des U2 snRNPs ist und eine essentielle Rolle beim Spleißen ausführt. Kernextrakte die kein SMNrp enthalten wiesen einen Defekt der Spleißosomen-Zusammenlagerung auf der Stufe des „prä-Spleißosoms“ auf. SMNrp ist demnach ein Zusammenlagerungsfaktor des Spleißosoms und bezüglich dieser Funktion dem U snRNP-Zusammenlagerungsfaktor SMN ähnlich.
