General Jim commissioned into the Intelligence Corps in 1986 and has extensive operation experience across the globe. His Staff experience includes role at the UK Ministry of Defence, and at the US Command and General Staff College. He has extensive experience working in and with multinational forces. He was Chief of Campaign Plans in Headquarters Multinational Force Iraq, and Chief Plans at Headquarters Allied Rapid Reaction Corps (ARRC) and subsequently deployed to Afghanistan. On promotion to Brigadier in 2009, he assumed the role of Director Intelligence, Surveillance, Target Acquisition and Reconnaissance (ISTAR) at Army Headquarters, and in Sep 2011 he returned to the Ministry of Defence as Head Military Strategic Planning. In June 2013, he was promoted to Major General, and deployed to Kabul as Director of the Ministry of Defence Advisory Group, before taking up his post as Director Cyber Intelligence and Information Integration in Mar 2015. In Dec 2018, General Jim assumed the appointment of Chief of Defence Intelligence on promotion to Lieutenant General. In 2022 General Hockenhull was appointed Commander UK Strategic Command.

There were massive travel delays and cancelations it was June and there were storms and then there were just really no good reasons for some of it. We met at Warner's place in DC and got a special whirlwind tour of the most esoteric parts of the city. We then traveled to Warner's farm in Virginia and spend the week talking through some of the works most intriguing mysteries. If I were able to record on 1/4 of our conversations it would be mind blowing to everyone. This was recorded on the last day, right before we were to catch our flight back to Ohio, another storm stranded us back in DC for the night and thankfully back at Warner's place until our rescheduled flight the next night. These two men are amazing, their perspectives and experiences range far beyond their years and I am very fortunate to call them my friends. This was us sitting down in the "bunker", with no pre planning or script, even the puppet show. This will not be the last time the 3 of us get together. Creepy Uganda   So Logan and I saw that we were rising through the ranks of Uganda's listeners for the show and thought: “Hey!  We should show our love and support to these wonderful people”. So, in order to do it right,  we are going on a trip! To Creepy Uganda.    Aside from rituals, ancient vengeful deities, and some rather haunted locations found throughout the wonderful country, there's actually quite a few beautiful areas that, as a tourist, would be something to see! Beautiful Lakes, Mountains and rich cultures are just some of the many things that are strewn about Uganda. So without further adieu, Let's Get Creepy.   The East African nation of Uganda, formally the Republic of Uganda, is a landlocked nation. Kenya borders the nation on the east, South Sudan on the north, the Democratic Republic of the Congo on the west, Rwanda on the south-west, and Tanzania on the south. A sizable piece of Lake Victoria, which Tanzania, Kenya, and the rest of the country share, is located in the southern region of the nation. The African Great Lakes area includes Uganda. The climate in Uganda, which is also part of the Nile basin, is variable but usually modified equatorial(Characteristics of Modified Equatorial Climate have a range of 4 to 27 degrees celsius). There are about 42 million people living there, 8.5 million of them reside in Kampala, the country's capital and largest metropolis.   Uganda was given its name after the kingdom of Buganda, which ruled over a sizable area of the country's southern region, including the capital city of Kampala, and whose language, Luganda, is extensively spoken today.   The United Kingdom began to govern the region as a protectorate in 1894, establishing administrative law throughout the realm. (A Protectorate is state that is governed and guarded by another independent state is known as a protectorate. It is a dependent region with local autonomy over the majority of internal matters that yet recognizes the authority (much like our relationship between the US and Puerto Rico) of a more powerful sovereign state without being that state's actual possession.) On October 9, 1962, Uganda declared its independence from the UK. Since then, there have been other bloody wars, including an eight-year military dictatorship under Idi Amin.   Their Constitution stipulates that "any other language may be used as a medium of instruction in schools or other educational institutions or for legislative, administrative, or judicial functions as may be authorized by law," despite the fact that English and Swahili are the official languages. Many more languages, including Ateso, Lango, Acholi, Runyoro, Runyankole, Rukiga, Luo, Rutooro, Samia, Jopadhola, and Lusoga, are also spoken in the Central and South Eastern portions of the nation.   Yoweri Kaguta Museveni, the president of Uganda at the moment, came to power in January 1986 following a lengthy six-year guerrilla conflict. He was able to run and win the presidency of Uganda in the general elections of 2011, 2016, and 2021 as a result of constitutional revisions that eliminated the president's term restrictions.   Uganda's varied terrain includes volcanic hills, mountains, and lakes. The average elevation of the nation is 900 meters above sea level. Mountains line Uganda's eastern and western borders. The Ruwenzori mountain range is home to Alexandra, the highest peak in Uganda, which rises to a height of 5,094 meters.   One of the largest lakes in the world, Lake Victoria, which has several islands, has a significant effect on most of the country's southern region. The most significant cities, including Kampala, the capital, and Entebbe, a neighboring city, are found in the south, close to this lake. The country's largest lake, Lake Kyoga, located in the middle of a vast marshy landscape. Uganda is a landlocked country, although it has a lot of big lakes. Lake Albert, Lake Edward, and the smaller Lake George are additional lakes to Lakes Victoria and Kyoga. The Nile basin encompasses practically the whole country of Uganda. On the border with Congo, the Victoria Nile flows from Lake Victoria via Lake Kyoga and into Lake Albert. South Sudan is reached by continuing northward. The Suam River, which is a component of Lake Turkana's internal drainage basin, drains a region in eastern Uganda. The Lotikipi Basin, which is mostly in Kenya, receives water from the far north-eastern region of Uganda.   There are 60 protected areas in Uganda, including ten national parks. Bwindi Impenetrable National Park and Rwenzori Mountains National Park are both UNESCO World Heritage Sites. What in the hell is UNESCO? It stands for Unidentified Neural Electron Sexual Conspiracy Organization and of course that's incorrect and stupid. It ACTUALLY stands for The United Nations Educational, Scientific and Cultural Organization. A specialised agency of the United Nations aimed at promoting world peace and security through international cooperation in education, arts, sciences and culture.The Bwindi Impenetrable National Park is home to a group of mountain gorillas, the Mgahinga Gorilla National Park is home to gorillas and golden monkeys, and the Murchison Falls National Park is home to those evil fucking hippos.   The military in Uganda is known as the Uganda People's Defense Force. There are about 45,000 soldiers on active service in Uganda's military. Only the United States Armed Forces are deployed to more nations, according to analysts, than the Ugandan army, which is actively engaged in a number of combat and peacekeeping missions in the area. Uganda has troops stationed in the Central African Republic, Somalia, South Sudan, and the northern and eastern regions of the Democratic Republic of the Congo.   The landscape and wildlife of Uganda are the main attractions for tourists. In 2012–13, it contributed 4.9 trillion Ugandan shillings (US$1.88 billion or €1.4 billion as of August 2013) to Uganda's GDP, making it a significant source of employment, investment, and foreign money. Photo safaris across the National parks and wildlife reserves are the primary draws. Other highlights are the mountain gorillas, which may be found in Uganda's aforementioned Bwindi Impenetrable National Park (BINP) and Mgahinga Gorilla National Park (MGNP), which are two of the continent of Africa's oldest cultural kingdoms. With more than 1073 species of birds reported, Uganda is an ornithologist's paradise, ranking fourth among bird species in Africa and sixteenth worldwide. The Great Rift Valley and the white-capped Rwenzori mountains are only two of Uganda's many landscapes.   Unfortunately like everywhere else, Uganda has a plethora of things that have happened there that aren't exactly what some may consider “pleasant”. For lack of a better term and because we're adults, let's just say some Pretty fucked up shit had happened, actually. Genocide being a fairly big thing. But we want to dive into the lesser known side of Uganda.   Like maybe some cryptozoology? Hmmmmmm?   A large cryptid bird named Bagge's Black Bird was once sighted in Uganda's Lake Bujuku, which is located south of Mount Speke in the Ruwenozori Mountains. They were purportedly observed in large numbers in 1898 at a height of 9,000 feet, according to Stephen Salisbury Bagge, a guide for the government. Bagge described them as black birds the size of sheep with an alarm call resembling that of a bull. Not much else to go on here since this was the only sighting allegedly of the creature. But who knows! Maybe it was a pterodactyl, or better yet, a rather large black bird that was living rather well and just so happened to be bigger than the rest.   Denman's bird was another cryptid bird that Canadian mountaineer Earl Denman purportedly claimed to have seen diving "swiftly and nearly vertically in the high mountain air" in Uganda's Ruwenzori Mountains. Ben S. Roesch speculated that they could have been Verreaux's eagles, which are common in the region and frequently observed diving to grab hyraces (rock rabbits) and hares (the thing that doesn't grow on my head) when hunting in pairs.   The irizima, also known as "the thing that may not be spoken of," was a cryptid that was seen in Uganda and the Democratic Republic of the Congo near Lake Edward. One of the least well-supported of the African neodinosaurs, it has been compared to both the mokele-mbembe and the emela-ntouka.   Neodinosaurian cryptids like the mokele-mbembe or li'kela-bembe have been seen mostly in the Republic of the Congo and Cameroon, where it is thought to live in marshy or swampy wetlands, lakes, and rivers. Several other bodies of water have also reported seeing it, but the Likouala region and Lake Tele are particularly linked to it. Many cryptozoologists have long assumed that the mokele-mbembe is a big amphibious animal with a bulky body, a long neck and tail, and a small head. However, a wide range of different reptilian and mammalian identities have also been proposed.   A neodinosaurian cryptid known from the rainforest swamps and rivers of the Republic of the Congo and the southwest Central African Republic, the emela-ntouka (Bomitaba or Lingala: "killer of elephants" or "eater of the tops of trees") is described as a horned animal and has been likened to rhinoceroses and ceratopsian dinosaurs. It is often used as a synonym for the older but now less well-known chipekwe water rhinoceros from Zambia, as well as the ngoubous from Cameroon, the ntambue ya mai from the southern Democratic Republic of the Congo, and certain accounts of forest rhinoceroses. The morphology of the emela-ntouka has been described as well-defined but puzzling. It is described as an amphibian with an elephantine, rhinoceros-like appearance, a big horn on its nose, and a bulky tail resembling a crocodile. The emela-identity ntouka's has historically been the subject of two extremely divergent conflicting theories: either it was a big semi-aquatic rhinoceros or, primarily due to its bulky tail, a living ceratopsian dinosaur. Many cryptozoologists no longer subscribe to the latter notion, as the emela-ntouka is now thought of as a mammal. One ethnic group, the Aka, refers to the emela-ntouka as mokele-mbembe, a practice that has generated considerable misunderstanding.   Now that we understand those two similar cryptids we go back to the irizima. It was initially brought up by Captain William Hichens, who said that there were two conflicting accounts of the creature, including a "gigantic hippopotamus with the horns of a rhinoceros" and an animal with hippo-like legs, an elephant-like trunk, a lizard's head, and an aardvark's tail. Hichens said that such a creature had been spotted by an unknown big game hunter, who then told Herbert Francis Fenn about it, inspiring him to look for neodinosaurs in the Congo. A Brontosaurus, described by Hichens as "a massive marsh animal, ten times as big as the biggest elephant," was discovered in a Congo swamp by a "madcap man" who had been searching for the monster, according to Hichens. Hichens, according to Bernard Heuvelmans, mistook information about the Great Brontosaurus Hoax and Captain Leicester Stevens' excursion for information about Lake Edward. Also, it sounds like they found the funny mushrooms.   The brontosaurus hoax was pretty interesting as well. Allegedly, the news paper in the area of the Democratic Republic of the Congo wanted Captain Stevens to find this cryptid found in the marshes of Lake Edward. The twist is that the original reports were of a ceratopsian dinosaur not a brontosaurus that was written in the news.   Hunter Roger Courtney later made reference to the Lake Edward monster, describing it as a huge, black beast that spews tremendous waves and spouts. When the hunter persuaded his companions to aid him onto the water, the monster had already dove, according to Courtney, who claimed that a Dutch hunter had spotted the animal from the shore of Lake Edward. In addition, Courtney had heard rumors about "dinosaurs" from the adjacent Ituri Forest, which he took to be true.   According to E. A. Temple-Perkins, who studied the irizima in Lake Edward, the monster—especially as it was described by Courtney—may have originated as a local legend intended to explain why waterspouts naturally occur. Given the lack of reliable material from Lake Edward, Bernard Heuvelmans believed that Captain Hichens had accidentally introduced the Lepage-Gapelle fake monster there, leaving Roger Courtney's brief report as the only description of the Lake Edward monster. Karl Shuker, however, asserts that these two contradictory descriptions demonstrate that the term "irizima" is likely used to describe both of the two primary African neodinosaur types found in Lake Edward, the long-necked mokele-mbembe type and the horned emela-ntouka type. Shuker hypothesizes that the irizima, which Hichens described as having numerous horns, may be the same animal as the emela-ntouka and the ngoubou, which resemble Arsinoitherium (a large two horned mammal that went extinct and resemble rhino but the horns being on its brow instead of its snout).   A group of semiaquatic cryptids known as water lions, water leopards, or jungle walruses have been found in rivers and occasionally wetlands throughout tropical Africa, particularly in the Central African Republic. The majority of the time referred to as huge cats , they can be identified by their protruding fangs or tusks and their penchant for hippopotamus slaughter, so they're not all bad. A number of competing theories exist, and some water lions have also been identified or confused with neodinosaurs, water rhinoceroses, and pseudodeinotheria. Ingo Krumbiegel and Bernard Heuvelmans theorized that water lions represent a surviving species of sabre-toothed cat adapted to an amphibious lifestyle and that sounds terrifying. The majority of water lion sighting reports were gathered in the 20th century, however reports of the n'gooli or “water panther”, continue to come from Cameroon.   The Nandi bear, also known as the chemosit (Kalenjin: "devil"), is a cryptid that has been seen in western Kenyan highlands as well as Uganda. It is described as a deadly creature with a matted mane that resembles a bear. Cryptozoologists have determined that the Nandi bear is a fusion of several different cryptids, including maybe two real unknown animals: a huge hyena and a giant baboon, however identities of a living chalicothere (the weird horse/gorilla looking thing) and an unknown bear have also been proposed. Since the 20th century, there have been few or no sightings, and it has been hypothesized that the Nandi bear, if it ever existed, is now extinct. Maybe another version of the sasquatch?    Hope the Cryptids were a little more easy going because now we dive into some… shit.   Sir Edward Frederick William David Walugembe Mutebi Luwangula Mutesa, often known as Kabaka Mutesa II, led a fascinating life. He ruled as Buganda's 36th kabaka (king) from 1939 until his passing on November 21, 1969. In addition, he served as Uganda's first president from 1963 until 1966, when he was ousted and taken into exile by Prime Minister Milton Obote.   Following the passing of his father, King Daudi Cwa II, he succeeded to the throne of Buganda in 1939. He was overthrown twice: once by the colonial governor-general Sir Andrew Cohen in 1953 so that he could be replaced by his half-brother, whom Cohen believed he could better control; and once more in 1966 when Prime Minister Obote forced him to leave for Britain, where he died in exile. Following his first exile of two years, Mutesa II was permitted to reclaim the throne as part of a negotiated agreement that established him as a constitutional monarch and granted the Baganda the opportunity to choose delegates for the kingdom's parliament, the Lukiiko. He had thirteen wives and eleven children by marriage and six through other means.   Initially joining forces to demand self-rule, Sir Edward Mutesa II, KBE and Prime Minister Milton Obote went on to win the 1962 election. Mutesa II was named non-executive president, primarily serving in a ceremonial capacity, but after independence, their relationship started to sour. Obote allegedly instructed Idi Amin-led soldiers to raid his stronghold in 1966. Mutesa II had to escape to the UK once more. Obote declared himself president and assumed total control while he was overseas.   The largest of Uganda's several ethnic groups, the Baganda, were led by Mutesa II as monarch. Despite taking advantage of it, Obote used his position of power to get rid of both the traditional kingships and the independence of the province administrations because Buganda had only agreed to join the state if it had a high degree of autonomy. In 1993, Mutesa's son was elected as the 37th kabaka under a revised constitution. Within Uganda, Buganda is currently a constitutional monarchy. In Uganda, Mutesa II attended King's College, Budo. As a student at Magdalene College in Cambridge, England, he enlisted in an officer training corps and received a captain's commission in the Grenadier Guards. Buganda was then a part of Uganda's British rule. Many of the traditional leaders or kings served as the British's representatives in Uganda. The late fourteenth century is when the Buganda kingly line began. Oddly enough, Obote was deposed in a coup in 1971 by none other than Amin, the head of his own army and closest supporter.   At the age of 45, Mutesa II passed away from alcohol poisoning at his London apartment in 1969. The British authorities determined that he committed suicide, despite his followers' claims that Obote regime assassins were responsible. In 2009, four decades after Mutesa II's passing, a family friend and fellow Ugandan exile living in London told the BBC, "We got warning, people used to write and say somebody has been sent, be aware, take care."   According to JM Kavuma-Kaggwa, an elder from Kyaggwe, Mukono District: “There were rumours that Obote was spending Shs 250,000 per week (a lot of money then) to track down the Kabaka. Their mission had completely failed until luck struck when the late Oscar Kambona of Tanzania who fell out with President Nyerere and fled into exile in London, organised a birthday party in November 1969 in Sir Edward Mutesa's honour.”   “Also in attendance was a beautiful Muganda girl who had reportedly been recruited by the GSU to go to London, befriend Sir Edward, be close to him and poison him. She came close to the Kabaka during the party. It was reported that the Kabaka invited the girl to this birthday party and that was the time she managed to poison him because she was the one in charge of the Kabaka's drinks that evening.”   After Obote was overthrown in 1971, Mutesa II's remains were brought back to Uganda and given a formal funeral by the new president, Idi Amin, who had led the attack on Mutesa's palace in 1966 as the army commander. Definitely an interesting story to say the least. This next event is a little more… unsettling.   On the last night of her life, Rose Nakimuli shut down her little hair salon in rural Uganda at around nine o'clock. The 27-year-old made her way back down to the neighborhood bar for a late-night beverage after walking home to change and turning on her porch light for the evening. Later, while she was strolling along a country road next to a two-lane highway on her way home, a friend leaned out of his small bar to greet her. The following morning, a neighbor discovered her dead; slouched behind banana trees in front of her house. Nakimuli was stripped and forced to kneel on her knees. Her vagina had been penetrated with a cassava stick. Her spouse recognized her by the maroon sweater that was hanging from a tree close by. Considering the porch light was still on suggests that she never actually made it home.   Nakimuli is one of 23 women who have died mysteriously and horribly on the outskirts of Kampala, the expanding metropolis of quickly urbanizing Uganda, from May to November of 2017. The murders have caused fear in the neighborhood, sparked doubts about the nation's dedication to protecting women, and increased scrutiny of the police force, a potent institution criticized for acting with impunity and serving as an extension of the government's ruling political party, the National Resistance Movement.   All of the victims were female, ranging in age from 19 to 38. Four of the individuals have been recognized as sex workers, along with a number of traders and a high school student. Many of the victims had no nearby family and lived alone. Three of the women, at least, are yet unidentified. Many of the murders, according to the police, were committed by witchcraft practitioners who sought financial gain through human sacrifice. Others, according to them, are the result of spousal abuse, drug use among unemployed youth, land disputes, and lone women who fail to take the necessary safeguards.    Twelve or more suspects have been taken into custody. Some have apparently been tortured into confessing. However, not much evidence connecting the suspects to the crimes has been made public.   Locals and activist organizations charge the police with being overburdened and conflicted over the murders of over twenty women.   “What makes me to feel that there is an element of injustice is that it took Rose to die in order for somebody to move,” said Nakimuli's husband, Anatoli Ndyabagyera.   Community watch groups have been established, a curfew has been implemented to prevent women from travelling alone at night, and the local informal economy has collapsed in the interim. Some of the safety measures have not been applied since Idi Amin's regime and the civil conflict that ensued after his overthrow in 1979.   Interior Minister Jeje Odongo blamed a couple of businesspeople at the head of a vast criminal network connected to "the Illuminati" in September 2017 for most of the killings. According to Odongo, the guys, Ivan Katongole and Phillip Tumuhimbise, performed rituals using the victims' blood and body parts in order to increase their wealth.   In Uganda, magic and mysticism still have great power. The rituals that these beliefs usually take the form of can occasionally become more evil. In the past, killings for ceremonial purposes have often involved children in particular.   Jordan Anderson, a researcher who has studied magic in East and Central Africa, claims that the latest killings of women, however, have little in common with conventional ritual homicides. One reason is that it's unusual to preserve a sacrificial body.   “You are killing the person because, in the first sense, you want to use that body part in the ‘medicine' or the potion that you are going to put together,” he said. “It's the particular part of the person you want, not the death per se."   Black magic can also be useful cover for a murderer trying to hide their tracks or an easy scapegoat for incompetent security forces.   “If you have this motif in the media, people can pick it up and copycat it,” Anderson said. “If there's insecurity in this area, if there are murders taking place, this is a great excuse for the politicians, the police and, above all, the people doing the murders.”   In an interview at one of the clubs where she was last seen alive, her husband noted that Nakimuli was regarded as being "extremely sweet." She was unable to stand by as a child sobbed. He couldn't bring himself to clean up her house for two months following her passing.   In small communities like the one where Nakimuli passed away, rumors are easily disseminated, and Ndyabagyera is still dubious of the police's version of what happened to his wife. He thinks Nakimuli's cousin may have set her up as part of a long-standing vendetta.   The small village of Katabi, where Nakimuli and 11 other women were murdered, is located along the main road from Kampala to Entebbe, which is home to the president of Uganda's palace and the country's primary airport on Lake Victoria. Museveni frequently travels this route on his way from his residence to the capital. He didn't go to the town, however, to pay his respects to the deceased until late September.   Museveni interviewed the victims' friends and neighbors during the unexpected visit while keeping a clipboard in his hand and taking careful notes.   The majority of the twelve slain women in the Katabi area were brutalized in ways akin to Nakimuli. Many had been assaulted with cassava sticks, stripped naked, and strangled.   On the opposite side of Kampala, 20 miles north, the bodies of an additional 11 women were found during the same time frame. There, victims were allegedly sexually assaulted and strangled, yet there were no sticks in their genitalia.   An individual named Ibrahim Kaweesa, a chicken dealer who had previously served ten years in prison for robbery, has been connected to those killings. Which seems like a huge escalation. The interior minister claimed that Tumuhimbise, a teenage shopkeeper, employed Kaweesa to murder a dozen women "for ritual performance to protect or improve his wealth."   As part of a loose network supporting law enforcement, 40-year-old Charles Waswa assisted in the arrest of Kaweesa and claimed, "They removed the blood."   Kaweesa resided two-thirds of the way down a short row of apartments, surrounded by women cooking outside and shrieking children. He was labeled by his neighbors as an arrogant and dangerous womanizer.   Kaweesa's neighbor Annette Namkose, 29, stepped in to prevent them from dating. She alleged through a translator that in response, he threatened to kill her, saying, "I'll kill you like I did the ones in Entebbe."   She declared, "He's not a neighbor you want to be with.   Police said that after being detained, Kaweesa swiftly confessed to the crimes. He allegedly led detectives around a number of the crime scenes without being asked.   “I don't believe we have arrested each and every person who knew about this matter,” said Kasingye, the police spokesman.   “I cannot say 100% there isn't going to be any (more) crime because it has never happened anywhere in the world. But at least it (the arrests) shows us we can stop criminals. We can arrest them, we can prosecute them and we can do this throughout the whole country.”   Unfortunately cases like these happen too much in many places around the world. Uganda seems to be trying to get ahead of the curve with the installment of the Anti-Human Sacrifice and Trafficking Task Force following the Anti-Trafficking Act in 2009.    Although reports have shown that the task force has been severely underfunded for a while, we do hope that things start to turn around.   Speaking of human sacrifices, this is a report from only a few weeks ago:   Human sacrifices continue unabated in the remote and rural areas of the landlocked East African country of Uganda despite authorities enacting tough laws and threatening death sentences.   According to officials, 132 incidents of human sacrifices have been recorded in the last three years. The numbers have spiked from 22 sacrifices in 2019, 45 in 2020 and 65 in 2021.   Most victims of such “ritual sacrifices” are children, apparently because they are easier to abduct and seen as “pure” and so of "higher ritual value".   Anadolu Agency quoted authorities as saying on Sunday that the sacrifices are being carried out by witch doctors or local traditional healers, dotting rural areas.   Admitting that human sacrifice is a big problem, Lucas Oweyesigire, the police spokesman for the Kampala region, said most such practices take place in rural areas.   The so-called leader of traditional healing and witch doctors, Mama Fina, has also condemned human sacrifice and described those recommending the sacrifice of human beings as “fake”.   Taking advice from witch doctors   Police spokesman Fred Enanga said only last month they "arrested a man identified as Musilimu Mbwire on suspicion of killing his two sons in human sacrifice.”   According to preliminary investigations, a rich man had paid Mbwire money and convinced him to sacrifice his two sons at the instructions of a witch doctor.   Superstitions lead people in rural areas to seek help from witch doctors, who in turn offer weird prescriptions, including human sacrifices to turn around their luck.   A more worrisome part of the superstition is to undertake human sacrifice to put the body at the foundation of a building to bring good luck.   Timothy Mukasa, a local leader in Kampala's suburb of Kireka, said many multi-storey buildings in the town have been built on a human body.   “The witch doctors tell owners to put a human body at the foundation of the construction of the buildings,” he said.   In 2014, authorities apprehended and later sentenced a tycoon Kato Kajubi for sacrificing a child and then putting his body in the foundation of a building that he was about to construct.   David Musenze, a journalist who studied psychology, said there are not many qualified counsellors to attend to psychological and mental issues of people, which makes them take advice from witch doctors.   "People go to witch doctors to help them get jobs, be promoted at jobs, or kill their enemies, along with many other problems," he said.   So, what about hauntings, you might be thinking to yourself. Well, we found a story from someone living in Uganda from the “your ghost stories” website. I had always thought this sort of nightmare was happening to me alone until I have come across this site. I always took my suffering silently especially the unexplained sickness which always followed devil attacks.   It all started on 28th November 2004 one hour to midnight. Whilst walking home after branching off from the main road. I heard footsteps of someone walking behind me and whoever it was seemed to have been in a hurry, I glanced back and stepped aside to see who it was and let him/her pass as I was in a narrow path.   I saw a hazy form I can't clearly explain here, my hair stood on my head like when you encounter something fearful. A cold shiver enveloped me and a gust of chilly wind wrapped my entire body, like I was putting on a cloak. I let out a silent incoherent scream and ran towards home which was just nearby. That occurrence signalled the beginning of my suffering to date.   Since then, whenever I sleep I am woken up by something touching my foot or a feeling of a being lying beside me, in the morning I find scratches on my body and at first I thought it was me scratching myself during asleep so I used to trim my nails, but the scratches continued.   During the attacks, I fall in a sort of hypotonizing stance. I neither can move nor make any sound except my feet which I use to struggle and try to shrug of the being.   In the past two years the demon has turned sexual, it would turn in a woman form, hugging me in bed trying to initiate sexual intimacy, when I wake up my reproductive organ feels so cold and shrunk. There's pain also in the pelvic area for most of the day.   I have tried all sorts of remedies e.g. Blessed water, salt, prayers etc. But none seems to work, Any suggestions on how to get rid of this demon is welcome.   And lastly, the Haunted Palace of Kabaka Kabak's Palace, also known as Idi Amin's Torture Chambers or Haunted Mansion or Lubiri Palace is located in Lubiri area of Kampala on Mengo Hill Road. It was the home of the Bugandan kings but these days it largely remains unoccupied due to the horrific events that took place under the rule of Idi Amin and President Milton Obote. President Idi Amin built his torture chamber here where hundreds of people were reportedly tortured to death. 1. Another big market drop to start the day. It's a broad based decline as leading sectors such as energy (XLE, XOP) and financials (JPM, GS, KBE) are trading lower. Technology is the big loser again with the NASDAQ Composite down 1.2%. Tech should be avoided now as it looks as if it is in full correction mode and it looks much weaker than everything else. 2. Today's big winner is crypto. Bitcoin and Etherium are both very strong today and has surged above the September 7th pivot top. That is a solid move, now the weekly close will tell me more when it comes to the popular crypto markets. Could this be the beginning of a new crypto rally? 3. Gold/Silver Gold was down earlier but has bounced back and is in the green. Silver down.

Goodwill-towards-man is one thing, but there are limits, and this sudden crush of humanity has made it entirely impossible for me to record the next episode in the dubious saga of VC Adrian Ward, KBE, Alds, &c. Music has always been the epicenter of Geldof's life, and it was also discussed during the interview how Alan felt Sir Bob could've found a life as a writer. Other subjects discussed are relevant bands, social media and how easy it is to be famous with little or no effort. Is there a perfect time for (another) social revolution? Instead, let's discuss the conversations between himself and Prince Charles. It's well known that Bob Geldof was frontman for the late '70s to mid '80s Irish punk rock band the Boomtown Rats. He also conceived and co-wrote Band Aid's tune “Do They Know It's Christmas?” with his friend, musician Midge Ure of Ultravox, which became the 1984 Christmas number one in the UK and the best-selling British pop single until that time. Wikipedia reported that Geldof was granted an honorary knighthood (KBE) by Elizabeth II in 1986 for his charity work in Africa; although it is an honorary award as Geldof is an Irish citizen he is often referred to as ‘Sir Bob'. He is a recipient of the Man of Peace title which recognises individuals who have made “an outstanding contribution to international social justice and peace”, among numerous other awards

Canaccord Genuity Chief Market Strategist Tony Dwyer for a second time joins hosts Aaron Task and Stephen Alpher on the Alpha Trader podcast. "There is no precedent to how high valuations can go," said Dwyer, making some news on Monday by pulling his 3,300 target for the S&P 500 (SP500). I have no idea what multiple you pay for an "unlimited" Fed, says Dwyer. 20x? 24x? 30x? 100x? Dwyer is bullish, but not just because the Fed's new framework and massive U.S. debt loads likely mean 0% policy rates for many more years. There also appears to be a synchronized global recovery at hand. Things won't go up in a straight line - in fact, the current level of frothiness suggests a correction could soon hit - but Dwyer thinks investors ought to be ready to buy any dips. Dwyer reminds that the "market" of late is essentially a small handful of names that seemingly go higher each day. For much of the rest of the equity universe, it's been a bit of a struggle. He expects leadership might soon change though, with cyclical plays like energy (XLE) and the banks ([[KRE]], [[KBE]]) primed to benefit from the global recovery. There's plenty more, including why the current period is way more like 2009 (start of major bull market) vs. 1999 (end of major bull market), the outlook for another fiscal stimulus package, and why zero rates forever might not necessarily drive gold higher and the dollar lower.

1. There was a reshuffling announced in the Dow Jones Industrial Average last night. It looks as if Salesforce.com (CRM) will replace Exxon Mobil (XOM), Amgen (AMGN) will replace Pfizer (PFE), and Honeywell (HON) will replace Raytheon Technologies (RTX) in the DJIA 30. This news is giving these news additions to the DJIA a lift this morning. The official date for the change will be on Monday, August 31, 2020.Major indexes are lower on very light volume. Easy to see them go higher later in the date, but otherwise it’s a backing and filling day. 2. Best Buy (BBY) is selling off after reporting earnings this morning. The earnings were solid, but the chart was already parabolic. This tells us that the stock was priced for perfection and this is a simple sell the news event. This is a perfect reason why you should not trade in front of earnings especially when the stock is parabolic.  3. Yesterday, the financial stocks caught a bid. The $KBE, $JPM, $BAC, $C and other closed nicely higher. For the first time in a long time. Today, they are starting out strong, but it will be important to see if they can hold into the close. Remember, the financials have been laggards for a long time. We shall see if this can start to show some strength and possible leadership. 4. We’ve missed the boat on housing. Still very strong. Waiting for a consolidation. The bus left the station without Nick. Very overbought right now. But financials aren’t and gold and gold miners working off overbought condition. Same with biotechs. Healthcare names pulling back. Look for things on sale.

It was led by a generation of black activists who had participated the Civil Rights movement. By the mid-1960s, a split developed among those activists and many no longer saw nonviolent protests as a viable way of combatting racism. Major turning point: The Watts riots, a 5 day "War" in 1965, that took place in the Watts neighborhood of Los Angeles. Dead‎: ‎34 / Injured‎: ‎1,032 New organizations (the Black Panther Party, the Black Women’s United Front, the Nation of Islam, and others) developed new cultural, political, and economic programs. Desegregation was insufficient. Deconstruction of the white power structures was the new focus. Because they wanted space for black political voices, collective black power, and social autonomy, the movement was often viewed as violent, anti-white, and anti-law enforcement. Key events: the 1965 assassination of Malcom X, and the 1968 assassination of MLK. Major accomplishment: raising the collective level of consciousness, pride, and interest in education. By the mid-1970s, the movement was in decline due to government repression, intragroup squabbles, and further assassinations. Also, police raids, arrests, and harassment. By 1973 African-American activists had begun to concentrate on getting blacks and progressive whites elected to public office. By 1976, the traditional movement was effectually dead. Legacy: Some have compared the modern movement Black Lives Matter to the Black Power movement noting a January 2015 community panel, hosted by the Garfield H.S. – B.S.U. (Seattle) titled: Black Power to Black Lives Matter, connecting the current youth-led struggle of BLACK LIVES MATTER to the Black Power movement of the 1960s-1970s.   Part 2:  Conclusion The Black Power movement not only represented a change in tactical strategy, but also a change in the mind-set of African-Americans. For instance, the black music industry, with its roots in gospel and rhythm and blues became more nationalist. Songs like the Impressions’ “We’re a Winner” (1967), James Brown’s “Say It Loud, I’m Black and I’m Proud” (1968), and the Temptations’ “Message to a Black Man” (1969) helped establish a distinctive sound for a generation of politically conscious young black Americans. Some blacks chose to don African garb and adopt African names. The slogans “Power to the People” and “Black is Beautiful” became very popular. The movement’s style and fashion (military berets, leather gloves and hats, bright powder-blue shirts, and Afro hairstyles were also symbolically important. The independent Tax Review Committee (TRC) established by the Government of PNG recently completed a comprehensive review of the country’s tax system involving extensive and transparent stakeholder consultation over the course of two years. This culminated in two volumes of reports (inclusive of significant economic and tax reform recommendations) which were formally presented to the PNG Government in November, 2015. The final report highlighted the high risks of PNG’s overdependence on the extractive sector and its vulnerability to the cyclical vagaries of global commodities pricing and recommended a ‘paradigm shift’ in the governance, management and stimulation of sustainable economic development. PNG’s vulnerability has become pronounced in recent months with the sudden drop in the oil price resulting in fiscal distress, lack of foreign exchange, and economic slowdown. PNG is at a precarious crossroads and tax reform provides an opportunity for a strategic transformation. The report provides some realistic and practical policy, legal, institutional, and structural reforms to mitigate economic risk and vulnerability by diversifying the economic base to more sustainable sectors like agriculture, forestry, fisheries and tourism. Sir Nagora Bogan, KBE, is Chancellor of the University of Technology, Chairman of the Papua New Guinea Tax panel of experts. He began his career with the PNG Taxation Office and during the 1980s and 1990s took a leading role in modernising tax administration in PNG. From 1991-1995, he was non-salaried Chairman of the London-based Commonwealth Association of Tax Administrators and from 1996 to 2002 Papua New Guinea’s Ambassador to the United States of America with concurrent accreditation as Ambassador to Mexico and High Commissioner to Canada. He served as Chairman of the Nambawan Super Fund Limited for more than a decade and is a director or board member of a number of PNG and regional corporations. The seminar was presented as part of the Development Policy Centre’s PNG Project, which receives funding from the Australian Aid Program through the Pacific Governance and Leadership Precinct.

Cronobacter spp. sind opportunistische pathogene Erreger, die insbesondere nach der Aufnahme kontaminierter Lebensmittel schwere Infektionen mit hohen Letalitätsraten bei Neugeborenen und immungeschwächten Erwachsenen hervorrufen können. Um spezifische immunchemische Nachweisverfahren für diese Keimgruppe zu etablieren, wurden in der vorliegenden Arbeit monoklonale Antikörper (mAK) zum Nachweis von Cronobacter spp. generiert und umfassend charakterisiert. Zur Präparation der Immunogene wurden Cronobacter-Keime mit Polymyxin B behandelt und anschließend wurden Mäuse entweder mit dem durch Zentrifugation erhaltenem Zellpellet (Ghosts) oder mit dem zellfreien Überstand (Lysat) dieser Präparationen immunisiert. Beide Präparationen erwiesen sich als hoch immunogen, die nachweisbaren Titer lagen üblicherweise bei > 1:10.000. Insgesamt konnten 14 stabile Hybridomzelllinien (sieben je Ansatz) etabliert werden. Die Intra- bzw. Inter-Genus-Spezifität und Affinität der entsprechenden mAK wurde umfassend unter Verwendung von indirekten EIA-Verfahren überprüft. Für Studien zur Epitopspezifität der generierten mAK wurden Immunoblots und Immunfluoreszenz-Analysen eingesetzt. Alle mAK, die aus der Immunisierung mit Cronobacter-Ghosts resultierten, zeichneten sich durch ein sehr breites Reaktionsspektrum aus, Kreuzreaktionen wurden vorzugsweise mit Vertretern aus der Familie der Enterobacteriaceae aber auch mit anderen gramnegativen Keimen beobachtet. Für alle mAK konnten Proteine als antigene Determinanten identifiziert werden, die relativen Molekulargewichte reaktiver Proteinbanden lagen üblicherweise im Bereich von > 40 kDa. Demgegenüber zeigten sechs der sieben mAK, die aus der Immunisierung von Mäusen mit Polymyxin B generierten Lysat-Präparationen resultierten, eine hohe Affinität für die O-spezifische Seitenkette der Cronobacter-typischen Lipopolysaccharide (LPS): mAK 2G4 αL reagierte hochspezifisch mit dem C. turicensis-Stamm (MHI 21026; Serotyp O1). Im indirekten EIA war dieser Erreger bei Keimzahlen von ca. 104 KbE/ml noch nachweisbar. Für die weiteren fünf mAK, die alle spezifisch mit C. sakazakii des Serotyps O1 reagierten, wurden im indirekten EIA Nachweisgrenzen im Bereich von 105-107 KbE/ml ermittelt. Alle mAK gegen LPS gehören zum IgG-Subtyp und reagierten in der Immunfluoreszenz mit lebenden Cronobacter-Keimen.

At the same time the problems and failures in Iraq and Afghanistan have strengthened the arguments against such intervention. Both sides have seen in the case of Libya, while Syria has left everyone cautious but frustrated. Where does the debate now stand, and in what circumstances can intervention be both in accordance with international law, and successful?Sir John Holmes GCVO, KBE, CMG is the Director of the Ditchley Foundation and co-chair of the International Rescue Committee UK. He is the former UN Under-Secretary-General for Humanitarian Affairs and Emergency Relief Coordinator. A career diplomat, he has served the British government for more than thirty years and has extensive experience on conflict. Having worked on the Lebanon and Middle East peace processes, he was awarded a knighthood in 1998 primarily for his role in the Northern Ireland Peace Process and the Good Friday Agreement. He served as Private Secretary (Overseas Affairs) to John Major before becoming Principal Private Secretary to Tony Blair in 1997.

Ferner sollten sie die Möglichkeit haben, ihren Kopf unter Wasser zu tauchen. Im Rahmen dieser Studie wurde untersucht, inwieweit die Tränke AquaDuc T® (Firma Big Dutchman GmbH, Vechta) unter Praxisbedingungen die Tierhygiene und verschiedene Gesundheitsparameter von Cherry-Valley Pekingmastenten (Wichmann Geflügelproduktionsgesellschaft mbh, Wachenroth) beeinflusst. Diese Studie ergänzt die Arbeiten von HEUBACH (2007), KÜSTER (2007), KOPP (2005), MANZ (2005), NUSSER (2008) und REMY (2005), in welchen alternative Wasserversorgungsangebote für Pekingmastenten in Kleingruppen erforscht wurden. Die Untersuchungen wurden in drei Entenmastbetrieben durchgeführt, die über Mastkapazitäten zwischen 7.500 und 13.500 Tieren verfügten. Je nach Betrieb entsprach dies einer Besatzdichte von 19,9 - 20,5 kg/m2 (6,6 - 6,8 Tiere/m2). Bei allen Betrieben handelte es sich um Fensterstallungen, die das Umtriebsverfahren betrieben und Bodenhaltung auf Stroheinstreu praktizierten. Die Tränke AquaDuc T® wurde in jeder Stallung grundsätzlich auf der Gefällseite installiert, um einen bestmöglichen Wasserabfluss zu gewährleisten. In Betrieb 1 und Betrieb 3 wurden jeweils acht sich abwechselnde Kontroll- und Versuchsdurchgänge, in der Folge als Besuchsart bezeichnet, durchgeführt. In Betrieb 2 waren es aus betriebsinternen Gründen fünf. Während in den Kontrolldurchgängen die Tiere nur über Nippeltränken mit Tränkwasser versorgt wurden, standen ihnen während der Versuchsdurchgänge auch Rundtränken (ab dem 25. LT täglich für sechs Stunden mit vierstündigem Wasserzulauf) zur Verfügung. Betrieb 1 hatte im Vergleich zu den beiden anderen zusätzlich Auffangschalen unter den Nippeltränken installiert. Die Datenerhebung fand an jeweils zwei Besuchen pro Betrieb und Mastdurchgang statt, jeweils in den Zeitfenstern 28. - 32. und 35. - 39. Lebenstag. Pro Besuch wurden grundsätzlich die Staub- und Ammoniakwerte nach einem bestimmten Schema gemessen, das Wasser der verschiedener Tränkevarianten beprobt (Auffangschalen, Nippeltränken, Rundtränken) und 100 Enten, 50 auf der rundtränkenabgewandten Stallseite und 50 Enten auf der Rundtränkenseite, bonitiert. Parallel wurden Videoaufzeichnungen angefertigt und ethologisch ausgewertet (siehe HARNISCH (2012)). Die Bonitur, die im Rahmen dieser Studie ausgewertet wurde, umfasste die Parameter Paddelhyperkeratose, Paddelnekrose, Verschmutzung Augenumgebung, Augenentzündung und Ulcus corneae. Zusätzlich wurden in jedem Betrieb einmalig Probeschlachtungen für einen Versuchs- und einen Kontrollbesuch an 20 männlichen und 20 weiblichen Enten durchgeführt. Neben den Schlachtparametern wurden die Blutparameter Hämatokrit, Hämoglobin und IgY sowie die Bruchfestigkeit der Ober- und Unterschenkel ermittelt. Die Bayrische Landesanstalt für Landwirtschaft, Lehr-, Versuchs- und Fachzentrum für Geflügelhaltung Kitzingen (LfL) erhob zu jedem Mastdurchgang Produktionsparameter sowie die Lufttemperatur und -feuchte. Außerdem wurden Mistprofile erstellt. Bei der quantitativen Untersuchung der durchschnittlichen Gesamtkeimzahl und Enterobacteriaceae-Anzahl schnitt die Nippeltränke mit einer Gesamtkeimzahl von 10.950 ± 1.583 KbE/ ml (n = 226) und einer Enterobacteriaceae-Anzahl von 113 ± 30 KbE/ml (n = 187) am besten ab. Bei den Rundtränken wurden eine Gesamtkeimzahl von 3.955.864 ± 877.640 KbE/ml (n = 40) und eine Enterobacteriaceae-Anzahl von 14.763 ± 2.459 KbE/ml (n = 33) festgestellt. Das ungünstigste Ergebnis erzielten die Auffangschalen, weil sie häufig mit Futterresten, Federn und Staubpartikeln verschmutzt waren. Hier zeigten sich ein Gesamtkeimzahl von 5.174.412 ± 564.137 KbE/ml (n = 62) und Enterobacteriaceae-Anzahl von 47.301 ± 11.057 KbE/ml (n = 44). Ein signifikanter Einfluss des Zeitfensters auf die gefundene Keimzahl war nicht feststellbar. Hinsichtlich der qualitativen Untersuchung der Proben auf Salmonellen konnten aus einer Nippeltränkenprobe (n = 226), fünf Rundtränkenproben (n = 184) und neun Auffangschalenproben (n = 62) Salmonellen isoliert werden. Am häufigsten war das Serovar S. choleraesuis (zehnmal) zu finden, gefolgt von S. arizonae (dreimal) und S. kottbus (zweimal). Bei der Untersuchung von Rundtränken-Doppelproben (es wurden Proben um 10:00 Uhr während der Rundtränkenbefüllung mit frischem Wasser und regulär um 12:00 Uhr gezogen) fiel auf, dass die Wasserproben um 10:00 Uhr in der Regel niedrigere Gesamtkeimzahlen und Enterobacteriaceae-Gehalte aufwiesen als die um 12:00 Uhr gezogenen Rundtränkenproben. Die Mittelwerte (± SEM) der gemessenen Staubkonzentrationen (n= 5 Durchgänge (Betrieb 2) bzw. 8 Durchgänge (Betrieb 1 und 3)) bewegten sich betriebsunabhängig zwischen 0,53 ± 0,01 mg/m3 (Betrieb 2, Versuch, 1. Zeitfenster) und 1,08 ± 0,21 mg/m3 (Betrieb 1, Kontrolle, 1. Zeitfenster). In keinem der Betriebe konnte binnen eines Zeitfensters eine signifikante Beeinflussung der Staubwerte durch die Besuchsart festgestellt werden. Auch ein signifikanter Einfluss des Zeitfensters innerhalb der Besuchsart Kontrolle oder Versuch war nicht zu ermitteln. Die von PETERMANN (2006) genannten Staubwerte in Geflügelställen und die von ZUCKER et al. (2005) erhobenen Staubwerte in Entenställen wurden im Rahmen dieser Arbeit weder in den Kontroll- noch in den Versuchsbesuchen erreicht. Die gemessenen Ammoniakschadgaskonzentrationen (n= 5 Durchgänge (Betrieb 2) bzw. 8 Durchgänge (Betrieb 1 und 3)) betrugen betriebsunabhängig zwischen 4,33 ± 1,21 ppm (Betrieb 2, Kontrolle, 1. Zeitfenster) und 8,76 ± 0,24 ppm (Betrieb 2, Versuch, 2. Zeitfenster). Wie bei den Staubwerten konnte in keinem der Betriebe innerhalb eines Zeitfensters ein signifikanter Einfluss der Besuchsart auf die Ammoniakwerte festgestellt werden. Eine Beeinflussung der Ammoniakkonzentration der Stallluft durch den Zeitpunkt des Besuchs war ebenfalls nicht erkennbar. In den Vereinbarungen verschiedener Bundesländer über die Haltung von Pekingenten sowie in den Empfehlungen der DEUTSCHEN LANDWIRTSCHAFTS-GESELLSCHAFT E.V. heißt es, dass der Ammoniakgehalt im Tierbereich in der Stallluft unter 10 ppm liegen sollte und dauerhaft 20 ppm nicht überschreiten darf. In Bezug auf diese Forderungen gab es in keinem Betrieb Überschreitungen, weder während der Besuchsart Kontrolle noch während der Besuchsart Versuch. In den selbst durchgeführten Probeschlachtungen schnitten die Versuchstiere von Betrieb 1 und Betrieb 3 in Bezug auf ihr Lebendgewicht signifikant besser ab als die Kontrolltiere. Dieses Ergebnis wurde allerdings durch das LfL, welches die Schlachtergebnisse aller am Feldversuch beteiligten Daten auswertete, widerlegt. Das Lebendgewicht wurde durch die Tränkeform, insgesamt gesehen, nicht signifikant beeinflusst (siehe Dissertation HARNISCH (2012)). Der durchschnittliche Hämoglobingehalt der Enten dieser Untersuchungen schwankte zwischen 6,44 ± 0,09 mmol/l (Betrieb 2, Versuch) und 7,15 ± 0,07 mmol/l (Betrieb 1, Kontrolle), der Hämatokritgehalt zwischen 32,75 ± 0,40 % (Betrieb 3, Kontrolle) und 38,75 ± 0,48 % (Betrieb 2, Kontrolle). In Betrieb 1 und Betrieb 2 konnte sowohl in Bezug auf den Hämoglobin- als auch den Hämatokritgehalt ein signifikanter Unterschied, abgeleitet aus den geschätzten Randmitteln der Wechselwirkung Betrieb * Besuchsart des Regressionsmodells Hämoglobin und Hämatokrit, hinsichtlich der Kontroll- und Versuchsschlachtung festgestellt werden. In beiden Betrieben war das Ergebnis der Versuchsschlachtung signifikant geringer. Diese Werte decken sich mit den Ergebnissen von HATIPOGLU und BAGCI (1996). Von einer Hyperhydratation der Tiere kann daher nicht gesprochen werden. Die ermittelten IgY-Durchschnittswerte dieser Arbeit liegen zwischen 7,77 ± 0,74mg/ml (Betrieb 3, Kontrolle) und 12,63 ± 0,76mg/ml (Betrieb 1, Kontrolle). Hinsichtlich des IgY- Mittelwerts unterschieden sich weder die Kontrolltiere signifikant von den Versuchstieren. Es kann, wie auch in den Arbeiten von MANZ (2005) und HEUBACH (2007) festgestellt, kein nachteiliger Effekt der Rundtränken auf die IgY-Konzentrationen der Enten nachgewiesen werden. Die durchschnittliche Femurknochenlänge bewegte sich zwischen 67,48 ± 0,43 mm (Betrieb 2, Kontrolle) und 69,38 ± 0,46 mm (Betrieb 3, Versuch), die Breite zwischen 6,63 ± 0,06 mm (Betrieb 1, Versuch) und 6,99 ± 0,07 mm (Betrieb 2, Kontrolle) und die Höhe zwischen 7,98 ± 0,06 mm (Betrieb 2, Kontrolle) und 8,48 ± 0,06 mm (Betrieb 3, Versuch). Der durchschnittliche Tibiotarsus maß hingegen in der Länge zwischen 110,56 ± 0,55 mm (Betrieb 1, Kontrolle) und 112,30 ± 0,63 mm (Betrieb 1, Versuch), in der Breite zwischen 7,04 ± 0,06 mm (Betrieb 1, Kontrolle) und 7,62 ± 0,08 mm (Betrieb 2, Versuch) und in der Höhe zwischen 6,32 ± 0,05 mm (Betrieb 2, Kontrolle) und 6,73 ± 0,10 mm (Betrieb 3, Kontrolle). Bei den Größenparametern konnten in Bezug auf die Besuchsart und das Geschlecht der Tiere diverse signifikante Unterschiede festgestellt werden. Ein einheitliches Muster, aus dem allgemein gültige Schlüsse gezogen werden könnten, war nicht abzuleiten. Die gemittelte Bruchfestigkeit (± SEM) der Femura lag zwischen 230,07 ± 4,18 N (Betrieb 1, Versuch) und 235,66 ± 3,71 N (Betrieb 3, Versuch), die der Tibiotarsi zwischen 172,23 ± 4,64 N (Betrieb 1, Versuch) und 195,15 ± 4,64 N (Betrieb 2, Versuch). Die Dehnung der Femura bewegte sich zwischen 2,02 ± 0,04 mm (Betrieb 1, Kontrolle) und 2,18 ± 0,05 mm (Betrieb 3, Kontrolle), die der Tibiotarsi zwischen 3,80 ± 0,10 mm (Betrieb 2, Versuch) und 4,36 ± 0,12 mm (Betrieb 3, Versuch). In keinem der Betriebe zeigte sich hinsichtlich der Knochenbruchfestigkeit ein signifikanter Einfluss der Wechselwirkung Betrieb * Besuchsart oder Betrieb * Besuchsart * Geschlecht innerhalb des dazugehörigen Regressionsmodells in einer Konstellation, welche im Rahmen dieser Arbeit interessant wäre. Eine kontinuierliche Beeinflussung der Dehnung durch die Besuchsart oder das Geschlecht zeigte sich nicht, auch wenn in Betrieb 3 der Dehnungswert der Femura der Kontrolltiere signifikant über dem der Versuchstiere lag und in Betrieb 1 ein Einfluss des Geschlechts erkennbar war. Bei der Bonitur der Paddel konnte in jedem Betrieb – unabhängig von der Besuchsart – eine Hyperkeratose-Rate von über 80% festgestellt werden. Insgesamt war eine signifikant höhere oder niedrigere Hyperkeratose-Rate während der Versuchsbesuche nicht zu verzeichnen. Das Boniturmerkmal "Nekrose der Paddel" ist dagegen von der Besuchsart abhängig. Die Chance des Boniturmerkmals "keine Nekrose der Paddel" verringert sich in allen drei Betrieben während der Besuchsart Versuch. In Betrieb 1 und Betrieb 2 reduziert sich die Chance signifikant (Betrieb 1: p = 0,012; Betrieb 2: p < 0,001). Nach MAYNE (2005) sind die Gründe für das Auftreten von Fußballendermatitis komplex. Zu den zwei wahrscheinlichsten Ursachen zählen feuchte Einstreu und Biotinmangel. Daher ist bei der Installation der Rundtränke unbedingt darauf zu achten, dass ein guter Wasserablauf im Stall gewährleistet ist. Hier besteht weiterer Forschungsbedarf. Die Auswertung der Bonitur der Augen erfolgt zum Großteil in der Dissertation HARNISCH (2012). Bei insgesamt 8.300 Enten wurde eine betriebsunabhängige 3,9%ige Ulcus corneae-Rate festgestellt. Die Besuchsart hatte in Betrieb 2 einen signifikanten Einfluss (p = 0, 001), nicht aber in Betrieb 1 und Betrieb 3. Eventuell kann dies durch betriebsabhängige Faktoren erklärt werden und muss nicht zwingend mit der Wasserversorgung über die Rundtränke in Verbindung gebracht werden. Die deutschen Entenmäster bieten Pekingenten nur zögernd Wasservorrichtungen, wie sie in den EMPFEHLUNGEN IN BEZUG AUF PEKINGENTEN (ANAS PLATYRHYNCHOS) DES STÄNDIGEN AUSSCHUSSES DES EUROPÄISCHEN ÜBEREINKOMMENS ZUM SCHUTZ VON TIEREN IN LANDWIRTSCHAFTLICHER TIERHALTUNG (1999) gefordert werden, an. Sie nennen hierfür zum einen hygienische und zum anderen wirtschaftliche Gründe. Im Verlauf dieser Studie verbesserten sich die Hygiene- und Gesundheitsparameter oder blieben unverändert. Ausnahmen hierzu waren lediglich die signifikant höheren Keimzahlen in den Rundtränken im Vergleich zu den Nippeltränken und der Anstieg der Nekrose-Rate während der Besuchsart Versuch. Letztere kann wahrscheinlich durch ausreichende Entwässerung verbessert werden. Im Hinblick auf die Keimzahlen ist festzustellen, dass die gefundenen Gesamtkeimzahlen in Auffangschalen, welche in der Entenmast durchaus noch vorhanden sind, signifikant höher waren als in Rundtränken. Das pauschale Argument, dass der Einsatz von Rundtränken Tierhygiene und Tiergesundheit negativ beeinflusst, kann aus Sicht dieser Studie nicht bestätigt werden. Die Rundtränke AquaDuc T®, die den Enten Komfortverhalten ermöglicht, erfüllt aus Sicht der vorliegenden Ergebnisse die Voraussetzungen einer tiergerechten Wasserversorgung in der Praxis, und bietet die Möglichkeit die Empfehlungen des Ständigen Ausschusses des Europäischen Übereinkommens in Bezug auf Pekingenten hinsichtlich der Wasserversorgung umzusetzen.

Vorkommen und Expression von ß-Laktamase A und B bei Yersinia enterocolitica 4/O:3 Stämmen aus Bayern mittels Polymerase-Ketten-Reaktion und Agardiffusionstest

Die vermehrte Anwendung antimikrobieller Wirkstoffe in der Human- und Veterinärmedizin hat zu einer Zunahme und Selektion antibakteriell resistenter Zoonoseerreger, wie Yersinia enterocolitica geführt. Resistenzmechanismen dieser Bakterien beruhen auf der Bildung chromosomal-kodierter ß-Laktamasen (A und B). Aufgrund unterschiedlichen Vorkommens der ß-Laktamase-Gene und der Expression dieser Enzyme innerhalb der einzelnen Y. enterocolitica Biotypen weisen diese eine heterogene Antibiotikaempfindlichkeit auf. In Deutschland stehen besonders Infektionen mit Y. enterocolitica Bioserotyp 4/O:3 an erster Stelle. Obwohl dieser Bioserotyp beide chromosomalen ß-Laktamasen bildet, wurden bei Y. enterocolitica Stämmen aus Bayern eine Amoxicillin/Clavulansäure-Antibiotika-Empfindlichkeit belegt. Ziel dieser Studie war das Vorkommen der Gene und die Expression für ß-Laktamase A und B bei 200 aus Bayern isolierten Y. enterocolitica 4/O:3 Stämmen mittels PCR und Agardiffusionstest zu untersuchen. Von den untersuchten Stämmen waren 60 humanen und 140 porcinen Ursprungs. Für den Nachweis der ß-Laktamase-Expression wurden im Doppelplättchenagardiffusionstest pro Stamm je zwei Müller-Hinton-Agarplatten mit einer Bakteriendichte von 107 KbE/ml beimpft und zusammen mit den zwei Antibiotikatestplättchen Ticarcillin (75 μg) und Amoxicillin/Clavulansäure (2/1 μg) bebrütet. Anschließend wurde die Expression der ß-Laktamase A und B anhand der Hemmhofgrößen um Ticarcillin und Amoxicillin/Clavulansäure bestimmt. Zur Überprüfung einer temperaturabhängigen ß-Laktamase-Expression wurde der Doppelplättchenagardiffusionstest bei den Bebrütungstemperaturen 30°C und 25°C durchgeführt. Im Anschluss wurden dieselben Stämme mittels konventioneller PCRMethode auf das Vorliegen der ß-Laktamasegene blaA und blaB überprüft. Die Auswertung der PCR Ergebnisse zeigte, dass bei 199 (99,5 %) der 200 untersuchten Y. enterocolitica 4/O:3 Stämme das blaA-Gen vorlag. Von diesen 199 Stämmen zeigten 198 Stämme auch eine BlaA-Expression in der Agardiffusion. Nur ein aus Schweine-Tonsillen isolierter Stamm zeigte trotz blaA-Gen keine BlaA-Expression. Bei einem der 200 untersuchten Stämme konnte mit Hilfe der PCR kein blaA-Gen nachgewiesen werden. Trotzdem zeigte dieser aus Hackfleisch isolierte Stamm im Agardiffusionstest eine BlaA-Expression. Das blaB-Gen wurde bei allen untersuchten Y. enterocolitica 4/O:3 Stämmen nachgewiesen. Im Agardiffusionstest exprimierten 3 humane und 1 porciner Stamm keine BlaB. Der Vergleich der Bebrütungs-temperaturen 30°C und 25°C zeigte einen Unterschied in der HHR-Größe um Ticarcillin. Während bei 30°C die meisten Stämme einen HHR von 3 mm aufwiesen, hatten die meisten Stämme bei 25°C einen HHR von 2 mm. Die durchgeführte Studie zeigt, dass nahezu alle (199/200) der aus Bayern isolierten und untersuchten Y. enterocolitica 4/O:3 Stämme die chromosomalen Gene für blaA und blaB besitzen. Zusätzlich konnte gezeigt werden, dass fast alle dieser Stämme beide ß-Laktamasen auch exprimieren. Die in einer vorangegangenen Studie ermittelte Empfindlichkeit dieser Stämme gegen Amoxicillin/Clavulansäure (20/10 μg) ist einerseits auf den 10fach höher konzentrierten Wirkstoff, andererseits auf eine geringere Expressionsmenge der ß-Laktamase B zurückzuführen. Die Bebrütungstemperatur zeigte keinen Einfluss auf die eigentliche ß-Laktamase-Expression, lässt aber einen Einfluss auf die Expressionsmenge vermuten.

Untersuchungen zum Vorkommen und zur Toxizität von Bacillus cereus in Gewürzen

B. cereus zählt zu den wichtigsten Verursachern von Qualitätsminderung und Verderb bei Lebensmitteln. Daneben wächst die Bedeutung Toxin-bildender B. cereus Stämme als Auslöser Lebensmittel-bedingter Erkrankungen, die zwei Formen einer gastrointestentinalen Erkrankung hervorrufen können: das diarrhoeische Syndrom, welches durch verschiedene Enterotoxinkomplexe (HBL, Nhe) induziert wird, und das emetische Syndrom, welches durch ein Dodekadepsipetid (Cereulid) ausgelöst wird. In komplexen Lebensmitteln werden vielfach Gewürze als Vektor für B. cereus - Kontaminationen angesehen. Jedoch sind kaum Studien über Gewürze als mögliche Eintragsquelle für B. cereus in Lebensmittel publiziert. Auch liegen nur wenig aktuelle Daten aus dem europäischen Raum über die tatsächliche Belastung von Gewürzen mit diesem Erreger vor. Ziel dieser Arbeit war es, das Vorkommen und die Toxizität von B. cereus in Gewürzen analysieren, um eine aktuelle Übersicht über die Kontamination mit diesem Erreger für eine Bewertung der mikrobiologischen Sicherheit von Gewürzen zu gewinnen. Hierfür wurden insgesamt 60 Gewürzproben zwölf verschiedener Gewürzsorten untersucht. Zunächst wurde mittels kultureller Verfahren die aerobe mesophile Gesamtkeimzahl sowie die Belastung mit Enterobacteriaceae und präsumtiven B. cereus bestimmt. Um auch kleinste Mengen des Erregers sicher nachweisen zu können, wurde zudem ein Real-Time PCR Assay als Alternative zur zeitaufwendigen und diagnostisch ungenauen konventionellen MPN- Methode erarbeitet. Im weiteren Verlauf der Arbeit wurden 151 präsumtive Kolonien, die aus den untersuchten Gewürzproben (n=60) isoliert wurden, mittels biochemischer und molekularbiologischer Analyse als B. cereus bestätigt. Anschließend wurde die Toxigenität und Toxizität der Isolate mittels PCR, immunchemischer Nachweisverfahren (ELISA, RPLA) sowie im Zellkulturtest charakterisiert. Darüberhinaus wurde im Challenge-Test mit marinierten Fleischerzeugnissen untersucht, inwieweit eine Kontamination von Lebensmitteln mit B. cereus tatsächlich aus der Verwendung kontaminierter Gewürze resultiert. Nach mikrobiologischer Analyse der Gewürzprodukte (n=60), waren nahezu alle der Proben als mikrobiologisch unbedenklich einzustufen. Kein Produkt hat den von der DGHM und der Europäischen Kommission empfohlenen Richtwert für B. cereus in Gewürzen von 103 KbE/g überschritten. Mit dem Real-Time PCR-Assay wurde zudem eine zuverlässige Alternative zum konventionellen MPN - Verfahren erarbeitet, der es ermöglichte auch kleinste B. cereus Mengen nach selektiver Anreicherung in den Gewürzproben zu identifizieren. Dies ist insbesondere im Hinblick auf das gesundheitsgefährdende Potential pathogener B. cereus von Bedeutung. So wurden 81% der Isolate (n=151), die aus den Gewürzen gewonnen wurden als diarrhöische und/oder emetische Toxinbildner identifiziert. Zudem wurde gezeigt, dass eine B. cereus Kontamination von Lebensmitteln durchaus aus der Verwendung damit hergestellter, belasteter Gewürze resultieren kann. Jedoch ist das Risiko für den Verbraucher hinsichtlich einer Lebensmittelvergiftung durch kontaminierte Gewürze als gering einzustufen. Zwar wurde B. cereus in 70 % der insgesamt 60 untersuchten Proben detektiert, allerdings in Keimzahlen, die nach derzeitigem Stand der Wissenschaft keine Lebensmittelvergiftung auslösen können. Dennoch ist das Gesundheitsrisiko, das von toxinbildenden B. cereus ausgeht nicht zu unterschätzen, da sie als Sporenbildner Erhitzungsprozesse von Lebensmitteln überleben, wieder auskeimen und sich vermehren können. Das Vorkommen von B. cereus in Gewürzen ist somit unter den Gesichtspunkten eines vorbeugenden Verbraucherschutzes und der Qualitätssicherung zu betrachten, nicht zuletzt da es auch angesichts eines stetig wachsenden Welthandels im Zuge der Globalisierung immer dringlicher wird eine mikrobiologisch unbedenkliche Ware zu gewährleisten. Letztendlich sind weitere Untersuchungen mit größeren Probenzahlen für eine besserere Risikoabschätzung unumgänglich. Mit dem Real-Time PCR Assay steht hierfür eine schnelle und zuverlässige Methode zur Verfügung, die einen Erregernachweis direkt aus Gewürzproben erlaubt.

Zusammenfassung In der vorliegenden Arbeit sollte die Quelle der Kontamination von Würsten mit Lactobacillaceae im Produktionsverlauf herausgefunden werden. Der Betrieb hatte Probleme mit erhöhten Lactobacillaceaezahlen, so dass einzelne Verpackungen vor dem Mindesthaltbarkeitsdatum verdorben waren. In der vorliegenden Untersuchung wurden Würste nach den Arbeitsschritten „Brühen“, „Abkühlen“, „Würsteltrenner“ und „Verpacken“ entnommen und die Gesamtkeimzahl, die Enterobacteriaceae- und die Lactobacillaceaezahl bestimmt. Im Weiteren sind Proben mittels Abklatschplatten von den Euro-Kisten, den Verpackungsmaschinen und vom Würsteltrenner extrahiert worden. An Hand von Tupferproben wurden die Gestelle und die Messer des Würsteltrenners auf die oben genannten Keime untersucht. Außerdem sind Proben von der Kistenwaschanlage und dem Kühlraum auf L. monocytogenes überprüft und weitere Material- und Umgebungsproben auf Campylobacter spp. kontrolliert worden. Als Eingang der Kontamination wurde der Würsteltrenner ausfindig gemacht. Hier zeigten 28,9 % der Wurstproben einen Gehalt an Lactobacillaceae zwischen 101 und 105 KbE/g. Im Vergleich dazu waren es nach dem Brühen nur 2,2 % und nach dem Abkühlen 6,5 % der Proben. Auch an Hand der Umgebungsproben dieses Gerätes wurde eine deutliche Kontamination festgestellt. Durch die Beobachtungen im Betrieb während der Probennahme fiel auf, dass besonders das Reinigungsmanagement nicht optimal verlief. So waren beispielsweise vor Produktionsbeginn, also an frisch gereinigten Geräten, noch deutliche Wurstreste unter den Messerschienen des Würsteltrenners erkennbar. Daher sollte besonders das Hygiene- und Reinigungsmanagement in Bezug auf die Häufigkeit und Gründlichkeit hin verändert werden. Der Schwerpunkt sollte hierbei auf der Schulung der Mitarbeiter beruhen.

In der vorliegenden Arbeit wurde basierend auf der genotypischen Charakterisierung von Stämmen des Mycobacterium tuberculosis - Komplexes anhand des gyrA-Gens mit Hilfe einer auf dem LightCycler® System basierenden gyrA Real-time-PCR ein neuartiges Testverfahren zur Bestimmung von Chinolonresistenzen konzipiert und etabliert. Bei der Evaluierung des Testverfahrens mit 13 phänotypisch resistenten Stämmen mit einer MHK ≥ 4 µg/ml für Ciprofloxacin zeigten 9 Stämme bei der Schmelzpunktanalyse einen eine Mutation anzeigenden Shift des Tm’s, 2 weitere wiesen die typische Schmelzkurve einer Heteroresistenz auf. Die Sequenzierung ergab, dass 5 dieser Stämme eine Mutation des Codon 90 (Ala→Val, GCG→GTG) und 6 Stämme eine Mutation des Codons 94 (Asp→Gly, GAC→GGC) aufwiesen. Die beiden verbliebenen Stämme hatten den Schmelzpunkt des Wildtyps, der durch die Sequenzierung bestätigt werden konnte. Die molekularepidemiologische Unabhängigkeit der resistenten Stämme wurde durch das IS6110 Fingerprinting bestätigt. 44 phänotypisch sensible Stämme wiesen ebenfalls den Tm des Wildtyps auf. Damit besitzt das Testverfahren für den Nachweis von Chinolonresistenzen eine Sensitivität von 85% und eine Spezifität von 100%. Die Speziesspezifität wurde anhand von 7 typischen und 16 atypischen Mykobakterien sowie humanen DNA-Präparationen und Sputumproben überprüft, wobei sich eine Amplifikation nur bei Mitgliedern des M.tb. - Komplexes fand. Das Testverfahren ist somit hochspezifisch für den Mycobacterium tuberculosis - Komplex. Der Test wurde für die Bestimmung von der Primärkultur und für den Direktnachweis aus dem Sputum evaluiert. Der Nachweis von Mykobakterien direkt aus dem Sputum mit dem Testverfahren ist mit 103 KBE/ml eine Größenordnung sensitiver als die Mikroskopie. Das Testergebnis liegt in der Regel innerhalb eines Tages vor. Für die Testdurchführung und die Testauswertung wurden Standardprotokolle erstellt. Anhand einer kritischen Evaluation der Primärliteratur konnte gezeigt werden, dass eine hohe Konkordanz von über 90% zwischen höhergradigen Chinolonresistenzen (MHK ≥ 4 µg/ml) und Mutationen des gyrA-Gens besteht. Die Informationen aus der phänotypischen und der genotypischen Resistenztestung sollte genutzt werden, um die beiden Verfahren gegenseitig zu evaluieren. So sprechen die bisherigen molekularbiologischen Daten für den von der WHO vorgeschlagenen Grenzwert von >2µg/ml bei der phänotypischen Resistenzbestimmung. Anhand von klinischen Studien sollte die Grenzwertsetzung überprüft und vor allem der Nutzen der Techniken für den Patienten evaluiert werden. Chinolone haben eine stark zunehmende Bedeutung bei der Behandlung der Tuberkulose und werden bisher vor allem bei der Behandlung der MDR-Tuberkulose eingesetzt. In mehreren klinischen Studien wird derzeit auch der Einsatz von Gyrasehemmern der neusten Generation als Standardmedikament untersucht. Hiervon verspricht man sich insbesondere eine Verkürzung und eine bessere Verträglichkeit der Therapie. Sollten sich die Chinolone als First Line Medikament zur Behandlung der Tuberkulose durchsetzten, könnte die im Rahmen dieser Arbeit entwickelte auf dem LightCycler® System basierende gyrA Real-time-PCR aufgrund der schnellen Resistenzbestimmung innerhalb eines Tages dem Kliniker entscheidende Hinweise für die primäre Einleitung einer effektiven Therapie geben.

Im Rahmen dieser Arbeit wurden Real-Time PCR-Verfahren auf Basis des TaqMan-Systems für die Detektion von Listeria spp. und von Listeria monocytogenes entwickelt und hinsichtlich der Anwendbarkeit für den Nachweis von Listerien in Milcherzeugnissen und Umfeldproben überprüft. Der genusspezifische Nachweis für Listeria spp. erfasst mit Listeria monocytogenes, Listeria innocua und Listeria seeligeri die wichtigsten und am häufigsten in Milchprodukten nachgewiesenen Listerienspezies. Wenn im gleichen Nachweissystem anstelle der fluoreszenzmarkierten TaqMan-Sonde der interkalierende Fluoreszenzfarbstoff SybrGreen verwendet wird, kann das gesamte Genus Listeria, mit Ausnahme von Listeria grayi ssp. grayi, nachgewiesen werden. Der speziesspezifische Nachweis für Listeria monocytogenes erfasst alle getesteten Stämme dieser Spezies (n = 34) und reagierte mit keiner anderen Spezies falsch positiv. Bei der Untersuchung von Reinkulturen waren für beide Nachweissysteme Keimzahlen von 104-105 KbE/ml für einen sicheren Nachweis erforderlich. Nach der erfolgreichen Etablierung der Nachweise für Reinkulturen wurden künstlich kontaminierte Milchprodukte untersucht. Es wurden sowohl unproblematische Matrizes wie pasteurisierte Vollmilch als auch Produkte mit einem hohen Gehalt an Begleitflora wie Weichkäse untersucht. Hierbei waren für einen zuverlässigen Nachweis niedriger Listerienzahlen von 1-10 KbE/g, aufgrund inhibitorischer Einflüsse einiger Probenmatrizes sowie der Begleitflora auf das Listerienwachstum, Anreicherungszeiten in Halb-Fraser von bis zu 48 h erforderlich. Eine negative Beeinflussung der PCR-Nachweise durch Probenbestandteile wurde nicht beobachtet, wenn die Proben mit dem PrepMan Ultra™ Reagenz aufbereitet wurden. Eine deutliche Verkürzung der Nachweiszeit in problematischen Probenmatrizes konnte durch Separation der Listerien aus der selektiven Anreicherungskultur mittels paramagnetischer Partikel, die mit listerienbindenden Proteinen aus Listeria-spezifischen Phagen beschichtet sind (CBD-MS; KRETZER, 2006), erreicht werden. Die separierten Listerien wurden nach einem kurzen nicht selektiven Anreicherungsschritt (3 h in Trypton-Soja- Bouillon mit Hefeextrakt) mit dem PrepMan Ultra Reagenz aufbereitet und in der PCR eingesetzt. Mit dieser Methode konnte die selektive Anreicherung auf 20 h und die Gesamtnachweiszeit auf 29 h reduziert werden.

In dieser Studie wurden Untersuchungen zu einer tierfreundlichen Wasserversorgung von Pekingmastenten unter hygienischen Aspekten durchgeführt. Insgesamt fanden fünf Versuchsdurchgänge statt. In den Versuchsdurchgängen I, II und V waren die Cherry-Valley-Pekingenten in sechs Abteilen zu je 192 Tieren untergebracht; in Versuchsdurchgang III und IV waren es 226 Tiere pro Abteil. In den Versuchsdurchgängen II, III und IV konnten die Tiere zusätzlich zum Stall einen überdachten Außenbereich nutzen. Da sich durch den Außenbereich die Gesamtnutzungsfläche vergrößerte, wurden in Versuchsdurchgang III und IV mehr Tiere (226) pro Abteil eingestallt. Die Mastdauer betrug 44 bis 47 Tage. In diesen fünf Versuchsdurchgängen stand vor allem die Erprobung von offenen Tränken in Form von Rundtränken, sowie der Einbau von Duschen als Badeersatzmöglichkeit, im Vordergrund. Die Duschen kamen nur in den Versuchsdurchgängen I und IV zum Einsatz. In allen Versuchsdurchgängen wurden Nippeltränken und Rundtränken als Tränkevarianten verwendet. Um bei den Rundtränken den Wasserverbrauch und die Tränkewasserhygiene zu optimieren, wurde die Anzahl der Rundtränken ebenso variiert wie auch ihre zeitliche Verfügbarkeit. Es wurden in jedem Versuchsdurchgang zu Mastbeginn (28.- 30. Masttag) und zu Mastende (41.- 45. Masttag) aus den angebotenen Tränkevarianten Wasserproben entnommen und diese mikrobiologisch auf Gesamtkeimzahl, Enterobacteriaceae-Gehalte, sowie zusätzlich noch qualitativ auf Salmonellen untersucht. Außerdem erfolgten Blutentnahmen zur Bestimmung des IgY-Gehaltes im Plasma, sowie Messungen zur Bestimmung des Ammoniakgehaltes im Stall. Es wurden bei der quantitativen Bestimmung der durchschnittlichen Gesamtkeimzahlen sowie der durchschnittlichen Enterobacteriaceae-Gehalte für die jeweiligen Tränkewasser-Proben zu Mastbeginn höhere Werte gemessen als zu Mastende. Betrachtet man die Medianwerte der Gesamtkeimgehalte, so lagen diese an den Nippeltränken zwischen 165.000 KbE/ml und 980.000 KbE/ml zu Mastanfang und zwischen 260.000 KbE/ml und 705.000 KbE/ml zu Mastende. An den Rundtränken hingegen bewegten sich die medianen Werte zu Beginn der Mast zwischen 415.000 KbE/ml und 16.800.000 KbE/ml und am Ende der Mast zwischen 72.500 KbE/ml und 3.075.000 KbE/ml. Viele der untersuchten Tränkewasser-Proben waren frei von Enterobacteriaceae, die medianen Werte derjenigen Proben, in denen Enterobacteriaceae nachgewiesen werden konnten, lagen zwischen 25.000 KbE/ml und 75.000 KbE/ml. Bei der qualitativen Untersuchung des Tränkewassers auf Salmonellen konnten sowohl aus den Tränkewasserproben der Nippeltränken, als auch aus den Proben der Rundtränken, Salmonellen isoliert werden. Hierbei stellte S.Indiana das am häufigsten isolierte Serovar dar. Die gemessenen durchschnittlichen Schadgaskonzentrationen lagen zu Mastbeginn an den Nippeltränken bei 10,10 ppm, an den Duschen bei 9,40 ppm und an den Rundtränken bei 6,82 ppm. Gegen Mastende erhöhten sich die Werte an den Nippeltränken auf 10,99 ppm, an den Duschen auf 10,90 ppm und an den Rundtränken auf 7,14 ppm. Die durchschnittlichen IgY-Gehalte im Blutplasma betrugen zu Mastbeginn zwischen 5,66 mg/ml und 12,45 mg/ml, zu Mastende lagen sie durchschnittlich zwischen 9,92 mg/ml und 18,11 mg/ml. Tendenziell zeigte sich im Blutplasma mit zunehmenden Alter der Tiere eine Erhöhung der IgY-Werte. Die Mortalität lag in allen fünf Versuchsdurchgängen unter 5 %. Aus hygienischer Sicht kann festgestellt werden, dass offene Tränken in Form von Rundtränken, die zeitlich begrenzt angeboten und regelmäßig gereinigt werden, sich als Alternative zu Nippeltränken anbieten, da sich die Keimbelastung der Tiere bei unterschiedlichen Tränkevarianten nicht wesentlich unterschied und keine negativen Einflüsse der Rundtränken auf die Tiergesundheit festgestellt werden konnten.

Yersinia (Y.) enterocolitica ist ein bedeutender Lebensmittelkeim, der vorwiegend in Schweinefleisch vorkommt. Kulturell ist er sehr schwer nachzuweisen, da er auf Selektivagar sehr schnell von anderen Keimen überwuchert wird. Zudem sind die kulturellen Isolierungsmethoden sehr zeitaufwendig und eine Aussage über pathogene oder apathogene Stämme kann aufgrund der Ergebnisse ohne weitere Diagnostik nicht getroffen werden. Aus diesen Gründen wird in der Diagnostik von Y. enterocolitica aus Lebensmitteln vermehrt der PCR-Nachweis eingesetzt, welcher innerhalb von Stunden, bei vorheriger Anreicherung nach einem Tag, ein zuverlässiges Ergebnis liefert. Abhängig von der Wahl des Zielgenes werden nur pathogene Keime detektiert. Die Sensitivität einer PCR ist in großem Maße von der Aufreinigung abhängig, deshalb sollte ein PCR-Protokoll immer einschließlich Probenvorbereitung etabliert werden. In der vorliegenden Arbeit wurde ein Real-Time PCR-basiertes Nachweisverfahren, zusammen mit der Probenvorbereitung, für Y. enterocolitica in Lebensmitteln entwickelt. Nachgewiesen wurde das chromosomale ail-Gen, sowie das auf dem Plasmid lokalisierte virF-Gen. In Vorversuchen erfolgte die Optimierung von verschiedenen Bedingungen für eine PCR, wie die Mastermix-Konzentration, Primer-Konzentration und Annealing-Temperatur. Anschließend erfolgte die Untersuchung von gezielt kontaminierten Rinderhackfleischproben. Diese Proben wurden nach Anreicherung mit sechs verschiedenen Extraktionsverfahren aufgereinigt und anschließend mit drei PCR-Methoden, davon eine Real-Time PCR mit SYBR Green und eine TaqMan Real-Time PCR sowie eine konventionelle Multiplex-PCR mit Gelelektrophorese, auf Y. enterocolitica untersucht. Parallel dazu wurde der klassische kulturelle Nachweis mit Anreicherung in TSB und ITC und Ausstrich auf CIN-Agar durchgeführt. Abschließend erfolgte die Untersuchung von 150 Schweinefleischproben aus dem Handel. Diese wurden ebenfalls mittels der drei PCR-Methoden und kulturell untersucht. Die Aufreinigung der Proben erfolgte nach Anreicherung mit drei Extraktionsmethoden, welche in den Versuchen mit den beimpften Proben die besten Ergebnisse lieferten. Mit allen drei PCR-Methoden konnten ail-positive Y. enterocolitica ab einer Impfmenge von 10 KbE/10 g Hackfleisch und nachfolgende Anreicherung nachgewiesen werden. Der Nachweis des virF-Genes gelang nur mit der konventionellen PCR, bei der SYBR Green Real-Time PCR konnte das virF-Gen nur selten und nur bei sehr hohen Keimzahlen detektiert werden. Bei der SYBR Green Real-Time PCR (ail) lagen die Ct-Werte bei der niedrigsten Impfmenge von 10 KbE/10 g, je nach Aufreinigungsmethode, im Bereich von 28,5 bis 34,8, bei der höchsten Impfmenge von 105 KbE/10 g konnten Werte ab 19,6 Zyklen erzielt werden. Die besten Ergebnisse konnten mit der Aufreinigung der InstaGene™ Matrix (Biorad), dem Biorad Genomic Tissue Kit und dem Qiagen DNeasy Tissue Kit erzielt werden. Mit diesen Methoden erfolgt auch die Aufreinigung der Schweinefleischproben aus dam Handel. Insgesamt konnte in 43 Proben (29 %) ail-positive Y. enterocolitica detektiert werden. Die höchste Nachweisrate mit 42 positiven Proben (28 %) wurde mit der SYBR Green Real-Time PCR erzielt, wogegen mit der TaqMan Real-Time PCR in 23 Proben (15 %) und mit der konventionellen PCR nur in 20 Proben (13 %) pathogene Y. enterocolitica detektiert werden konnten. Um die Veränderung und Entwicklung der objektiven Rindfleischfarbe während der Lagerung zu untersuchen, wurden vakuumierte Fleischscheiben aus dem M. longissimus dorsi von 30 Rindern über 8 Wochen gelagert. In jeder Lagerwoche wurden die Farbwerte L*a*b* der bestehenden Fleischoberfläche und eines frischen Anschnittes mit dem Minolta Chroma-Meter CR-400 gemessen. Als weitere Qualitätsparameter wurden der mikrobiologische Status (Gesamtkeimzahl, Laktobazillen, Milchsäurebakterien), die sensorischen Eigenschaften und der pH-Wert bestimmt. Ziel der Untersuchungen war, Referenzwerte für die Entwicklung der Rindfleischfarbe L*a*b* während der Reifung und Lagerung zu erarbeiten. Weiterhin sollte in der vorliegenden Studie die Farbmessung an Praxis- und Routinebedingungen in der Fleischwirtschaft angepasst werden, um ihren Einsatz in der Praxis zu verstärken. Drittens dienten die Versuche dazu, Zusammenhänge und Einflussnahme zwischen der Fleischfarbe L*a*b* und anderen Qualitätsparametern von Rindfleisch aufzuzeigen. In Voruntersuchungen wurde die Fleischfarbe von 255 Roastbeefs (15 bis 20 h p. m.) unmittelbar nach der Zerlegung ermittelt. Bei einer verkürzten Farbaufhellung (10 min, + 12°C) unter Praxisbedingungen zeigte der M. longissimus dorsi folgende, durchschnittliche Farbwerte: L* 33,12; a* 18,45; b* 6,62. Die Farbmessung unmittelbar nach der Zerlegung, on-line, eignet sich für einen Vergleich der Fleischfarbe zwischen verschiedenen Teilstücken und Betrieben. In den Vorversuchen konnte zudem nachgewiesen werden, dass signifikante Unterschiede in der Farbe des Roastbeefs zwischen den einzelnen Schlachttierkörpern bestehen. Innerhalb des M. longissimus dorsi eines Tieres erwies sich die Farbverteilung in der vorliegenden Untersuchung jedoch als gleichmäßig. Eine Farbmessung kann damit an jeder Stelle des Roastbeefs vorgenommen werden und ist repräsentativ für den gesamten Muskel. Im Hauptversuch wurde die Fleischfarbe L*a*b* über einen achtwöchigen Untersuchungszeitraum sowohl an der bestehenden Fleischoberfläche als auch an einem frischen Anschnitt gemessen. Dabei zeigte sich, dass zwischen der Farbe von Oberfläche und Anschnitt des M. longissimus dorsi ein starker, statistisch gesicherter Zusammenhang besteht. Für die Farbmessung unter Laborbedingungen wird dennoch weiterhin die Messung an einem frischen Anschnitt nach einstündigem „blooming“ empfohlen. Mit vergleichbar hoher Sicherheit kann jedoch unter Praxisbedingungen im Betrieb die Fleischfarbe durch Messung an der bestehenden Fleischoberfläche bestimmt werden. Die Entwicklung der objektiven Rindfleischfarbe über einen Zeitraum von acht Wochen ist erstmals dokumentiert worden. Die gewonnenen Daten dienen als Richt- und Vergleichswerte für zukünftige Lagerversuche. Am Tag der Zerlegung (15 bis 20 h p. m.) zeigten sich für einen frischen Anschnitt folgende Farbwerte: L* 36,5, a* 21,6, b* 10,5. Die Farbwerte der bestehenden Fleischoberfläche lagen geringfügig höher: L* 37,2, a* 22,9, b* 11,1. In der dritten Lagerwoche, nach Abschluss der Rindfleischreifung, wurden für die Anschnittsfarbe folgende Werte gemessen: L* 39,5, a* 25,5, b* 13,3. Dieser Anstieg der Farbwerte während der Reifung ist auch für die Oberflächenfarbe (L* 40,3, a* 25,9, b* 13,5) festgestellt worden. Die Farbwerte L*a*b* zeigten ab der 4. bzw. 5. Lagerwoche einen nochmaligen Anstieg. In der fünften Lagerwoche lagen die Farbwerte von Anschnitt (L* 39,8, a* 25,7, b* 13,2) und Oberfläche (L* 40,4, a* 25,7, b* 13,2) sehr eng zusammen. Der Anstieg fällt mit dem Ende der Mindesthaltbarkeit (35 Tage) zusammen und deutet auf Veränderungen in der Güte der untersuchten Roastbeefs hin. In den eigenen Untersuchungen zeigte sich eine negative Korrelation der objektiven Fleischfarbe mit dem pH-Wert. Der pH-Wert beeinflusste die Ausprägung der Farbwerte L*a*b* massiv. Er wies ebenfalls einen starken Einfluss auf die sensorische Beurteilung der Fleischfarbe auf. Zur korrekten Beurteilung der gemessenen Farbwerte sowie der visuell ermittelten Farbe muss daher auch immer eine Bestimmung des pH-Wertes erfolgen. Die gemessenen Farbwerte L*a*b* des Roastbeefs wurden nur geringgradig durch den mikrobiologischen Status, vor allem durch die Anzahl der Laktobazillen, beeinflusst. Aus der objektiven Fleischfarbe kann demnach nicht auf die mikrobielle Belastung des Rindfleisches geschlossen werden. Zwischen der objektiven Farbe und den sensorischen Parametern bestanden hoch- bis höchstsignifikante Zusammenhänge. Die Unterschiede der Farbwerte L*a*b* zwischen einzelnen abstufenden Bewertungen der sensorischen Kriterien waren jedoch zu gering, um sichere Richtwerte für wünschenswerte Eigenschaften bzw. Verderbserscheinungen zu ermitteln. Anhand der gemessenen Farbwerte kann daher keine Aussage über die sensorische Beschaffenheit des Rindfleisches getroffen werden. In den vorliegenden Untersuchungen wurde zudem die Entwicklung des mikrobiologischen Status von vakuumiertem Rindfleisch über eine achtwöchige Lagerung nachvollzogen. Erstmals wurden die Gesamtkeimzahl sowie die Anzahl an Laktobazillen und Milchsäurebakterien kontinuierlich in ihrem Verlauf dokumentiert. Die ermittelten Keimzahlen dienen als Referenzwerte für die bakterielle Belastung von hygienisch gewonnenem Rindfleisch im Verlauf der Lagerung. Sie sollen fleischerzeugenden und -vermarktenden Betrieben Vergleichswerte für die Einschätzung des mikrobiologischen Status des eigenen Rindfleisches während der Lagerung an die Hand geben. Sensorische Abweichungen konnten in dieser Studie ab einem Oberflächenkeimgehalt von 106 KbE/cm2 festgestellt werden. Für rohes Rindfleisch wurde in der vorliegenden Arbeit eine beschreibende Sensorik, bei der Oberflächen- und Anschnittsfarbe, Geruch, Konsistenz, Fleischsaftmenge und -beschaffenheit sowie Textur beurteilt wurden, durchgeführt. Die Korrelationen zwischen den einzelnen sensorischen Parametern wurden für rohes Fleisch erstmals ermittelt. Weiterhin wurden die Korrelationen zwischen mikrobiologischen Kriterien und sensorischen Parametern errechnet. Der Zusammenhang zwischen mikrobiologischen und sensorischen Parametern war jedoch unpräzise, so dass zur Überprüfung von Mindesthaltbarkeit und Genusstauglichkeit von Rindfleisch immer sowohl mikrobiologische als auch sensorische Untersuchungen durchgeführt werden sollten. Auf Grundlage der vorliegenden Arbeit ergeben sich neue Anwendungsbereiche für die Farbmessung. Die erarbeiteten Referenzwerte für die Fleischfarbe L*a*b* können im Rahmen des Verbraucherschutzes zur Qualitätsprüfung an Rindfleisch eingesetzt werden. Aber auch die fleischerzeugenden und -vermarktenden Betriebe können aufgrund der erarbeiteten Referenzwerte für die Fleischfarbe in der Zerlegung sowie für die Entwicklung der Farbwerte während der Reifung und Lagerung die Farbmessung im Rahmen ihrer Qualitätssicherung und Eigenkontrollen einsetzen. Des Weiteren dienen die Untersuchungen über die Entwicklung des mikrobiologischen Status und über die sensorische Beurteilung von rohem Fleisch der Verbesserung der Qualitätsbeurteilung an Rindfleisch.

Es wurde eine tiergerechte Wasserversorgung von Pekingenten unter hygienischen Aspekten untersucht. Insgesamt fanden fünf Versuchsdurchgänge statt, wobei nur die Ver¬suchsdurchgänge (VG) III bis V in dieser Studie berücksichtigt wurden. In jedem Versuchs¬durchgang, mit einer Mastdauer von 47 bis 49 Tagen, wurden je 1.152 Cherry-Valley-Pekingenten gehalten, die auf sechs Stallabteile aufgeteilt waren. In diesen drei Versuchsdurchgängen (III bis V) stand die Erprobung verschiedener Tränkevarianten (Nippel-, Rinnen-, und modifizierte Rundtränke) im Vordergrund. Diese Tränkevarianten wurden in Versuchsdurchgang III als Kombinationen – Nippel/Nippel, Nippel/Rinne, Nippel/Rund 24h – rund um die Uhr den Enten angeboten. In den Versuchsdurchgängen IV und V kam nur noch die modifizierte Rundtränke als offene Tränkevariante mit teilweise zeitlich begrenztem Zugang zum Einsatz. So wurde in Versuchsdurchgang IV die modifizierte Rundtränke mit 24h-,8h- und 4h Zugang zusätzlich zu Nippeltränken angeboten, in Versuchsdurchgang V erneut mit 4h- und 2h Zugang sowie einer Kontrollgruppe, denen nur Nippeltränken zur Verfügung standen. Es wurden, jeweils zu Mastbeginn (21.-28. Tag) und zu Mastende (46.-47. Tag) in den Versuchsdurchgängen III-V Wasserproben aus jeder Tränkevariante ge¬zogen und mikrobiologisch auf Enterobacteriaceae-Gehalt, Ge¬samtkeimzahl, sowie qualitativ auf Salmonella-Serovare untersucht. Des Weiteren erfolgten Blut¬entnahmen zur Bestimmung des IgY-Gehaltes im Plasma (mittels neu entwickeltem Sandwich-ELISA) sowie die Bestimmung der Ammoniakkonzentration in der Stallluft. Bei der qualitativen Untersuchung des Tränkewassers auf Salmonella-Serovare konnte aus jeder Tränkevariante Salmonellen isoliert werden. Alle isolierten Serovare konnten als humanpathogene Keime identifiziert werden. S. Saintpaul stellte das häufigste gefundene Serovar dar. Dage¬gen wurde das für den Menschen bedeutsa¬mere Serovar S. Typhimurium nur einmal isoliert. Es wurden bei der quantitativen Bestimmung auf Enterobacteriaceae-Gehalt und Ge¬samtkeimzahl immer zu Mastbeginn höhere Werte gemessen als zu Mastende. Die Messungen in Versuchsdurchgang III (VG III) zu Mastbeginn ergaben durchschnittlich 0 KbE/ml an Enterobacteriaceae in Nippeltränken (vgl. VG IV: 100.000 KbE/ml; bzw. VG V: 0 KbE/ml), bei Rinnentränken lag der Enterobacteriaceae-Gehalt in VG III bei 20.400.000 KbE/ml; an den 24h Rundtränken konnte zu Mastbeginn ein Enterobacteriaceae-Gehalt (VG III) von 4.350.000 KbE/ml (vgl. VG IV: 5.425.000 KbE/ml) festgestellt werden. Des Weiteren konnte zu Mastbeginn an der 8h zugänglichen Rundtränke ein Enterobacteriaceae-Gehalt von 1.335.000 KbE/ml gemessen werden (VG IV), wogegen an der 4h zugänglichen Rundtränke in VG IV 8.875.000 KbE/ml (vgl VG V: 125.000 KbE/ml) festgestellt wurden. An den 2h zugänglichen, modifizierten Rundtränken lag der durchschnittliche Enterobacteriaceae-Gehalt bei 600.000 KbE/ml (VG V). An allen Tränkevarianten konnte über alle drei Versuchsdurchgängen hinweg zum Mastende hin eine Abnahme an Enterobacteriaceae festgestellt werden. Bei Mastbeginn wurde als durchschnittliche Gesamtkeimzahl in VG III 0 KbE/ml (vgl. VG IV: 5.510.000 KbE/ml bzw. VG V: 605.000 KbE/ml) an Nippeltränken ermittelt, an Rinnentränken 83.075.000 KbE/ml (VG III) und an den 24h Rundtränken in VG III 74.500.000 KbE/ml (vgl. VG IV: 73.500.000 KbE/ml). Die Gesamtkeimzahlen konnten in Rundtränken mit einer zeitlichen Zugangsbegrenzung von 8h, 4h und 2h nicht reduziert werden. Die Gesamtkeimzahlen sanken in den Tränken, wie schon bei den Enterobacteriaceae, zu Mastende ab. Die gemessenen Schadgaskonzentrationen, bewegten sich zu Mastbeginn zwischen minimal 3,00 ppm an einer Nippeltränke und maximal 14,00 ppm jeweils an einer Rinnen- und einer 24h Rundtränke. Diese Werte stiegen zu Mastende deutlich an. So ergaben sich zu Mastende Ammoniakwerte von minimal 8,00 ppm an einer Nip¬peltränke und maxi¬mal 32,00 ppm an einer Rinnentränke. Die mittleren IgY-Gehalte im Blutplasma betrugen zu Mastbeginn minimal 3,66 mg/ml an der Tränkekombination Nippel/Rinne in Versuchsdurchgang III und maximal 9,72 mg/ml an der Kombination Nippel/Rund 2h in Versuchsdurchgang V. Die mittleren IgY-Konzentrationen beliefen sich zu Mastende auf minimal 15,26 mg/ml an der Tränkekombination Nip¬pel/Nippel in Versuchsdurchgang III und maximal 26,05 mg/ml an der Kombination Nippel/24h Rundtränke in Versuchsdurchgang IV. Zu Mastbeginn und zu Mastende war keine Korrelation zwischen dem IgY-Gehalt im Plasma der Tiere und dem Gehalt an Enterobacteriaceae sowie dem Gesamtkeimgehalt in offenen Tränken, festzustellen. Es bestand eine signifikante Korrelation zwischen den IgY–Gehalten und den gemes¬senen Ammoniakwerten zu Mastbeginn. Zu Mastende korrelier¬ten diese Parameter nicht signifikant miteinander. Aus hygienischer Sicht kann festgestellt werden, dass sich keine erheblichen Differenzen zwischen den einzelnen offenen Tränken in Bezug auf die Gesamtkeimzahl oder An¬zahl an Enterobacteriaceae ergab. Alle Werte lagen weit über den Richtwerten der Trinkwasserverordnung. Doch diese hohen gemessenen Keimzah¬len in den Tränkevarian¬ten nehmen scheinbar keinen Einfluss auf die IgY–Gehalte im Plasma der Enten.

Für die Mast von Pekingenten wird eine Haltung mit der Möglichkeit gefordert, das arteigene Badeverhalten ausüben zu können. Dies setzt ein Angebot von Tränkesystemen voraus, die es den Tieren ermöglichen, den Ablauf der dafür notwendigen Handlungskette durchzuführen. Ziel der vorliegenden Studie ist es zu untersuchen, inwieweit verschiedene Tränkesysteme in der Lage sind, unter den Gegebenheiten und Managementbedingungen eines Großbetriebes mit insgesamt 161000 Enten Mastkapazität pro Stallkomplex, die Ansprüche der Tiere hinsichtlich der Ausübung ihres Badeverhaltens zu realisieren. Zu diesem Zwecke wurden über den Zeitraum Mai 2003 – Juni 2004 (=12 Mastdurchgänge) in drei Stallabteilen mit einem Besatz von je 3600 Tieren neben einem konventionellen Nippelstrang den Tieren entweder Plassontränken, Sparkcuptränken oder Wasservernebelung mittels Düse in Kombination mit Nippeltränken jeweils als Alternative angeboten. Als Kontrolle diente ein identisches Stallabteil mit gleichem Tierbesatz und zwei Nippelsträngen. Als Beurteilungsparameter für den Systemvergleich diente die Feststellung der Frequentierung der einzelnen Tränkesysteme mittels 2 x 12 Std. Videoaufzeichnung pro Mastdurchgang, die Beurteilung des Befiederungszustandes, der Konjunktiven und Nasenöffnungen am Ausstallungstag sowie die Erhebung von Produktionsparametern wie Verlustrate, Gewichtszuwachs, Stroh- und Wasserverbrauch. Um einen möglichen Einfluss der verschiedenen Tränkesysteme auf die Stallumwelt zu erfassen, wurde neben dem Stallklima und NH3 Gehalt der Stallluft die Einstreuqualität bonitiert. Ferner wurde der mikrobiologische Zustand des Tränkwassers in den Systemen Plasson- und Sparkcuptränken sowie der Einstreu aller Stallabteile kontrolliert. Folgende Ergebnisse konnten statistisch (p < 0,05) gesichert werden: · Plassontränken wurden von den Tieren gegenüber Nippeltränken bevorzugtaufgesucht. · Sparkcuptränken wurden signifikant weniger häufig genutzt als Nippeltränken. · wurden die Nippeltränken mit intermittierender Wasservernebelung durch Düsen kombiniert, führte das zu einer signifikanten Bevorzugung gegenüber der nicht bedüsten Nippeltränkereihe durch die Tiere. · die Einstreuqualität im mit Plassontränken ausgestatten Stall war signifikant schlechter als in den übrigen Ställen. · lagen die Verluste im mit Sparkcuptränke ausgestatteten Stall mit 2,16% über der Kontrolle mit 1,55%. Tendenziell · lagen die Verluste im mit Plassontränken ausgestatteten Stall mit 2,11% über der Kontrolle mit 1,55% (P = 0,056). · lag die Häufigkeit von Tieren ohne den Befund verklebter Nasenöffnungen in der Sparkcupvariante mit 51% über der Plassonvariante mit 32% (P=0,058). Umgekehrt lag die Befundhäufigkeit von zwei verklebten Nasenöffnungen in der Plassonvariant mit 37% über der des Kontrollstalles mit 18% (P=0,058). Kein gesicherter Zusammenhang bestand in den vorliegenden Untersuchungen zwischen den einzelnen Tränkevarianten und den Produktionsparametern Gewichtzuwachs (3,2kg/Durchgang), Strohverbrauch (57 g/Tier/Tag), Befiederungszustand nach Mastende, NH3 Gehalt der Stallluft (9,1 ppm), Enterobakteriengehalt der Einstreu ( 2,58 x 108 KbE/g Einstreu) und dem Auftreten von Konjunktivitis(61,5%). Der Gehalt an Enterobakterien im Tränkewasser von Plasson- und Sparkcuptränken lag bei 1,85 x 106 (KBE/ml). In 2 von 15 Durchgängen konnten aus dem Wasser dieser Systeme Salmonellen isoliert werden. In der Einstreu konnten bei allen Probenahmen und in allen Ställen Salmonellen verschiedener Serovaren (S. Saintpaul,S.Typhimurium, S.Kottbus, S.Indiana, S.Mbandaka) nachgewiesen werden. Stallspezifische Unterschiede bestanden dabei nicht. Insgesamt gesehen wurden von den in der vorliegenden Studie untersuchten alternativen Tränksystemen Plassontränken und Nippeltränken in Kombination mit Wasservernebelung von den Tieren gesichert gegenüber Nippeltränken bevorzugt. Bei den Plassontränken ging dies jedoch mit einer Verschmutzung des Tränkwassers und einer Durchnässung der Einstreu einher. Auf alle geprüften ökonomischen bzw. gesundheitlich relevanten Parameter ergab sich jedoch kein positiver - hinsichtlich hygienischer Aspekte eher ein negativer - Einfluss der Alternativsysteme gegenüber dem konventionellen Nippeltränkesystem.

Der Einfluß der Kryokonservierung auf die mikrobielle Kontamination von Stammzellapheresaten

In einer kontrollierten prospektiven klinisch-experimentellen Untersuchung wurden 24 MNC-Apheresen an 24 freiwilligen gesunden Spendern durchgeführt, das gewonnene Zellkonzentrat portioniert und gezielt mit jeweils einem von fünf häufigen Kontaminationskeimen von Blutprodukten kontaminiert: Escherichia coli, Candida albicans, Staphylokokkus aureus, Koagulase-negative Staphylokokken und Pseudomonas aeruginosa. Dabei wurden zur Inokulation erregerabhängig sepsisähnliche Konzentrationen zwischen zehn hoch minus zwei und zehn hoch drei KBE/ml verwendet. Ein Teil der Proben wurde unter Zusatz von 10 % DMSO kryokonserviert und in der flüssigen Phase von flüssigem Stickstoff bei – 196 ° C gelagert. Von nicht kontaminierten unbehandelten Kontrollen, inokulierten Apheresatproben und gezielt kontaminierten und anschließend kryokonservierten und wiederaufgetauten Proben wurden Blutkulturen in BACTECTMPLUS Aerobic/F* und PLUS Anaerobic/F* Kulturfläschchen angefertigt und zur Untersuchung im BACTEC 9240 Gerät von BECTON DICKINSON in die Mikrobiologie eingesandt. Von positiven Blutkulturen wurden zur Bestätigung des Ergebnisses und zur Isolierung und Identifizierung des Keimes Subkulturen auf Blutagar angefertigt. Die Kryokonservierung zeigte keinen signifikanten Einfluss auf die Viabilität von Mikroorganismen im Apheresat: alle verwertbaren, gezielt kontaminierten Versuchsansätze wiesen nach Kryokonservierung weiterhin Keimwachstum auf. Lediglich bei Escherichia coli konnte, bei Verwendung einer sehr kleinen Inokulationsmenge (zehn hoch minus zwei KBE/ml) zur Kontamination, in drei kryokonservierten und wieder aufgetauten Apheresaten kein Erreger mehr isoliert werden. Da aber bei Verwendung dieser niedrigen Verdünnungsstufe bereits in fünf von zwölf Positivkontrollen kein Wachstum von E. coli nachzuweisen war, dürfen diese Proben nicht in die Auswertung eingehen. Es gibt zu viele Einflussfaktoren auf den Keimnachweis in Blutkulturen, als dass hier von einer Abtötung von E. coli durch den Prozess der Kryokonservierung und des Wiederauftauens ausgegangen werden darf. Als klinisch zuverlässige Maßnahme zur nachträglichen Erzielung von Keimfreiheit bei vorliegender Kontamination eines Aphereseproduktes eignet sich die Kryokonservierung deshalb nicht.
