A recent study in mice explored how a combination of two supplements, lactoferrin and creatine, might help with age-related muscle loss, a condition called sarcopenia. Scientists induced sarcopenia in mice, causing their muscle mass and function to decline. For 8 weeks, these mice were given lactoferrin, creatine, or a mixture of both. The results were promising: the group of mice receiving both lactoferrin and creatine showed the most significant improvements. For example, the lean body content in the sarcopenia model group dropped to around 47-48%, while the group receiving the combination treatment saw their lean body content increase to about 51-52%, bringing them above the levels of the healthy control group (around 50%). In terms of muscle function, the combination group's endurance on a treadmill test was approximately 1200 seconds, a significant improvement compared to the sarcopenia model group (around 600 seconds) and even surpassing the healthy control group (around 900 seconds). Their grip strength also increased to roughly 180g with the combination, compared to about 130g in the model group and 150g in controls. These findings suggest that combining lactoferrin and creatine could be a beneficial nutritional strategy to combat muscle decay associated with aging, potentially by improving how muscles use energy and build protein. 3. Your body makes hydrogen peroxide that is used by the immune system to kill microbes. When hydrogen peroxide combines with iron, it can cause serious cellular damage. People with iron-deficiency anemia often have dysfunctional iron, which means it's unavailable to the red blood cells. This could be why we are not seeing a decrease in anemia even though we've increased iron in the food supply. Iron overload may increase your risk for Alzheimer's, Parkinson's, cancer, heart disease, and more. Excess iron causes oxidative stress and increases inflammation. As we age, we naturally accumulate more iron. Around 70% of our iron is stored in protein in the blood that helps carry oxygen. Ferritin should be inside your tissues to protect you from free iron and control when iron is released. This very efficient system recycles iron in the body. Certain pathogens, microbes, and infections use iron as fuel. This is why the body protects free iron. Some people feel better after donating blood, indicating they have too much iron. Too much iron without enough copper is a big problem. If you think you have this problem, limit iron exposure before taking a copper supplement. Iron is found in enriched flours, grains, and refined carbohydrate products. Shellfish, organ meats, sunflower seeds, and dark chocolate are good sources of copper. To combat oxidative damage caused by iron and other factors like alcohol, smoking, pollution, and junk foods, you'll want to build up your antioxidant reserve. Glutamine found in red meat is vital, but protein is the most important! It's especially important to consume enough protein as you get older. Zinc, manganese, magnesium, and selenium can increase antioxidant networks to help you regulate excess iron. Quercetin, curcumin, black tea, and increasing melatonin are also very beneficial. Lactoferrin is the most powerful chelator for iron. It's found naturally in dairy and can be taken as a supplement.

As a result, the milk quality always suffers, the mozzarella couldn't stretch, it's not enough calcium, not enough nutrients in the milk. So, I gave up that whole idea and was still working for Google."   "So, we bootstrapped initially, brought in some scientists, did a lot of research, and filed our first patent. The technology requires getting memory cells and culturing the cells to express full-function milk, all 2,000 different ingredients in milk. We had our early success and quickly brought this technology to some of the biggest dairy companies in the world, folks like Fonterra and Abbott, and told them, "Hey, we can make milk. What do you think?" "Very quickly they came back with feedback saying, "Well, great that you can make milk, but milk is $2 a gallon. I don't think you can get to the price point anytime soon. You should really be focused on high-value ingredients that are found in milk." So they gave us a list of half a dozen proteins and ingredients that we should look at, and at the top of the list is lactoferrin because of all these great benefits and the high price point it is today. Fengru Lin, Co-Founder and CEO of TurtleTree, is applying precision fermentation technology to produce high-value ingredients for food and beverages without the need for animals. This biotech company's first goal is to expand the production of lactoferrin, a protein found in small quantities in milk that has been shown to promote gut health and immunity. Finding a scalable way to produce lactoferrin will lead to adding this iron-binding element to plant-based milks and energy drinks.   Fengru explains, "A couple of years ago, I was learning how to make cheese as a hobby. A2 Milk's sole infant formula supplier, Synlait Milk, said its half-year profit slumped by 76 per cent to $6.4m, driven by Covid-19 disruption, and said it expected to be "broadly breakeven" in the full year.The full-year outlook has worsened since last December when the company said it expected the 2021 profit to be half that of the previous year's $75.2m.Early this month Synlait withdrew that December guidance due to uncertainty and volatility within its business.In today's result, Synlait's revenue gained 19 per cent to $664.2m while EBITDA fell 29 per cent to $47.7m.Chair Graeme Milne said it was a challenging first half, "and we continue to find ourselves in a period of significant uncertainty and volatility as Synlait faces into several headwinds".A2 Milk - Synlait's biggest customer - has been hit hard as Covid-19 has severely curtailed the important "daigou" unofficial trade channels into China.Synlait said uncertainty was affecting its short-term operations and would impact on the full year's financial result.Chief executive Leon Clement said the company's focus was on mitigating the impact Covid-19 has had its customers."We will need time to get through this, but we remain confident about our future," he said.Synlait has been investing heavily in a bid to diversify its interests away from a2 Milk."Our investment phase is complete. We have the capacity, capability, and customer base to generate significant value," Clement said."Covid-19 hit us late, but we will emerge from the pandemic a stronger, more sustainable Synlait," he said.Synlait signalled earlier this month that it was continuing to experience significant uncertainty and volatility within its business due to ongoing uncertainty in a2 Milk's expected demand for the remainder of 2021 and 2022."Synlait does not currently have sufficient confidence to forecast when this recovery will occur," Clement said."The resulting impact of this on Synlait's business is two-fold: demand for consumer-packaged infant formula remains uncertain, which in turn impacts forward infant base powder production and asset use."Commenting on the ingredients business, Clement said the sudden drop in consumer-packaged infant formula demand, combined with rapidly rising Global Dairy Trade prices, foreign exchange, and a changing product mix, created volatility that limited returns.Furthermore, Synlait said its expectation was that global shipping delays would continue and further impact the 2021.Board and management have considered the above factors and how they will impact Synlait's 2021 profitability."There is still a range of scenarios contributing to the company's profitability, and our current outlook suggests a broadly breakeven FY21 net NPAT result," the company said.While all banking covenant ratios were met during 2021, Synlait has "proactively engaged" with its banking syndicate to increase its leverage ratios to manage any risk at the end of 2021.The 2021 business plan is fully funded by its current banking syndicate.In its result, Synlait said consumer-packaged infant formula sales down 16 per cent to 18,085 tonnes.Lactoferrin production gained up 16 per cent to 13.6 tonnes.Dairyworks, which Synlait bought in 2019 for $112m, reported revenue of $112.6m.Shares in Synlait Milk last traded at $3.41, down 14c or 3.9 per cent from Friday's close.text by Jamie Gray, NZ Herald

Iron levels and biochemistry have impacted my personal performance in a dramatic way. Recently I've been racing cyclocross, something for which I lack talent. For those of you who don't know cyclocross, it's a lot like short track mountain bike racing but most of the bikes look a road bike with drop handlebars. The start is really important, and the course has hundreds of corners and so the sport doesn't suit slow-twitch athletes like me. This year I’ve been enjoying some dramatic gains in my performance. These gains are nothing to do with training, in fact, I've been training less than ever before. The key to performance is health, and that's why I've got Dr. Tommy Wood on again today to talk about some of the biochemical tweaks I've been making and also the serious issue of iron overload (haemochromatosis). First let’s talk about anaemia. Some people think of anaemia as synonymous with iron deficiency, but iron deficiency is just one of many things that can cause anaemia. Diagnosing anaemic tendencies requires the consideration of the production, destruction and loss of red blood cells. Other nutrients such as zinc, B6, B12 and folate are as important as iron for the production of red blood cells. Sometimes we see results that show both iron overload and anaemia. Why is iron overload a problem? Most micronutrients cause little harm when consumed in excess. Vitamin C is a good example--consume too much and you'll simply pee it out. Excess iron is different in that there's no easy way to excrete it. The main way we lose iron is through bleeding, and for men and postmenopausal women this doesn't happen very often. Excess iron becomes bound to proteins and acts as a pro-oxidant, literally causing your tissues to rust. If left untreated, the excess iron can cause diabetes, cirrhosis of the liver and a host of other chronic diseases. The day after we recorded this interview four of my six follow-up appointments were with masters athletes with iron overload. Please don't guess, do the test! How should your diet change if you are diagnosed with iron overload? Research shows diet can have an impact of the absorption of iron. Things that block iron absorption: Calcium rich foods like dairy, leafy vegetables and sardines (whole with bone). Phytates from leafy vegetables and nuts. Tannins in red wine and coffee. Anthocyanins in any colour berry (the darker the colour of the berry the better). Things that increase iron absorption: Vitamin C. Added fructose, table sugar. In this study the effects of the prebiotic acting substance lactoferrin on blood parameters and the specific immune reaction were investigated in the model of orphan-reared neonatal beagle-puppies. Lactoferrin is an iron-binding glycoprotein that can be detected in milk and other external secretions of various mammals. In addition to many other functions it has shown stimulating effects on metabolism and immune system in previous trials. For this 14-week trial 32 beagle-puppies were split in four groups (n = 8) with different lactoferrin inclusion levels (0, 30, 60, 120 mg Lactoferrin/kg milk replacer or dog food). The dogs were fed self-made, balanced milk replacer until weaning during week 8, followed by a commercial dry dog food. Application of Lactoferrin to the puppies had no influence on the increase of body weight. Looking at blood parameters some tendencies of the lactoferrin-supplemented groups within the white blood cell parameters could be detected. Treatment groups showed a decrease of relative lymphocyte numbers compared to those of the control group, what could be due to an anti-proliferating activity of lactoferrin on lymphocytes. Proliferation of neutrophil granulocytes was stimulated by lactoferrin, shown by the slightly increased levels of neutrophils in the treatment groups. No statistical significance could be detected. The typical physiological pattern of the development of red blood cell parameters during the first weeks of newborn puppies could be seen, while oral lactoferrin admission did not change any of these results. The production of anti human serum albumine antibodies was chosen as a parameter for the investigation of the effect of lactoferrin on the immune status. Lactoferrin did not show any influence on the proliferation of specific antibodies, induced by the subcutaneous application of human serum albumine for two times during week 8 and 12. Therefore, no direct association between lactoferrin-supplementation and immune response could be observed. It could be shown, that the application of lactoferrin did not increase the immune response in healthy beagle-puppies, but it can be discussed wether it induces an antiproliferative effect on the production of lymphocytes.

In der vorliegenden Arbeit wurde erstmals der Einfluss von Vitamin C (in Form von Natrium Ascorbyl Monophosphat) und Epigallocatechin Gallat (EGCG) in Kombination mit Lactoferrin (LF) auf die Zahngesundheit der Katze untersucht. Dazu erhielten in einem verblindeten Fütterungsversuch jeweils 9 Katzen in der ersten Versuchsphase von 4 Wochen ein mit Vitamin C (50 mg/kg Körpermasse) bzw. in einer zweiten vierwöchigen Versuchsphase ein mit EGCG in Kombination mit Lactoferrin (je 6,6 mg/kg Körpermasse) versetztes Futter. Zum Vergleich wurde jeweils 9 Katzen parallel ein Kontrollfutter verabreicht. Um die Wirkung des Futters auf die Zahngesundheit beurteilen zu können, wurde die Gingiva, die Tiefe des gingivalen Sulcus, die Plaque- und Zahnsteinbildung sowie Mobilität und Furkation an Zähnen und Parodont mittels Indizes beurteilt. Weiter wurde die Halitosis gemessen und exemplarisch die bakterielle Flora der Maulhöhle von je 2 Katzen aus Kontroll- und Wirkstoffgruppe beurteilt. Zusätzlich wurde die antibakterielle Wirkung der Substanzen in vitro getestet und alle 4 Wochen der Einfluss von Vitamin C und EGCG in Kombination mit Lactoferrin auf den antioxidativen Status der Tiere überprüft. Durch die Fütterung von Vitamin C konnte im Durchschnitt keine Beeinflussung des Geruchs der Atemluft, des Gingivitisindex sowie des Mobilitäts- und Furkationsindex festgestellt werden. Sondierungstiefe und Zahnsteinindex verbesserten sich nur gerringgradig. Für den Plaqueindex ergab sich allerdings durch die Fütterung von Vitamin C eine signifikante Verbesserung im Vergleich zur Kontrollgruppe, die sich in der deutlich geringeren Plaqueakkkumulation bei der Vitamin C-Gruppe zeigte. EGCG in Kombination mit Lactoferrin bewirkte keine Veränderung des Geruchs der Atemluft sowie des Furkations- und Mobilitätsindex. Plaque- und Zahnsteinindex waren nur geringgradig niedriger im Vergleich zur Kontrollgruppe. Signifikante Unterschiede ergaben sich beim Gingivitisindex und der Sondierungstiefe. Der Gingivitisindex verringerte sich innerhalb von 4 Wochen deutlich bei der EGCG/LF-Gruppe und die Sondierungstiefe war im Vergleich zur Kontrollgruppe am Ende der Versuchsphase des EGCG/LF-Versuchs bei der Wirkstoffgruppe deutlich niedriger. In der Zusammensetzung der oralen Bakterienflora ergaben sich innerhalb der 4 Wochen bei keiner der Katzen wesentliche Veränderungen. Die in vitro-Versuche bestätigten bei allen Substanzen deutlich ihre antibakterielle Aktivität. Vitamin C sowie EGCG in Kombination mit Lactoferrin beeinflussten den antioxidativen Status nicht. In der vorliegenden Studie wurde somit gezeigt, dass Vitamin C und EGCG in Kombination mit Lactoferrin einen positiven Effekt auf die Zahngesundheit der Katze ausübten. Dieser Effekt machte sich bei Vitamin C hauptsächlich durch die deutlich verminderte Plaqueakkumulation und bei EGCG in Kombination mit Lactoferrin vor allem durch den verbesserten Zustand der Gingiva bemerkbar.

Parodontopathien sind die am häufigsten vorkommenden Erkrankungen von Hund und Katze. Vor allem bei Katzen beschränken sich Prophylaxemaßnahmen häufig auf eine wiederholte Plaque- und Zahnsteinentfernung in Narkose. Um die Intervalle zwischen diesen Zahnreinigungen zu verlängern, wird versucht, über speziell dafür hergestellte Futtermittel die Neubildung von Plaque und Zahnstein zu reduzieren. In der vorliegenden Arbeit wurden zu diesem Zweck Katzen zwei antibakteriell wirkende Substanzen über das Futter verabreicht und ihre Auswirkung auf die Maulhöhlengesundheit der Katzen sowie ihre Wirksamkeit gegen die aus der Maulhöhle isolierten Keime in vitro untersucht. Als Nebenzielgröße wurde die Auswirkung der Substanzen auf den Antioxidativen Status der Katzen beurteilt. Die Untersuchungen erfolgten in Form von zwei Fütterungsversuchen. In Versuch I wurde der Einfluss von Lactoferrin untersucht. Lactoferrin ist ein eisenbindendes Glykoprotein, das in der Milch verschiedener Säugetierspezies und in diversen Körperflüssigkeiten vorkommt. Lactoferrin hat, neben verschiedenen anderen Wirkungen, antibakterielle und antioxidative Eigenschaften. In Versuch II wurde der Einfluss von Epigallocatechingallat untersucht. Bei Epigallocatechingallat handelt es sich um ein Catechin, das aus den unfermentierten Blättern des Teestrauches Camellia sinensis gewonnen wird (Grüntee-Extrakt). Epigallocatechingallat hat, wie Lactoferrin, unter anderem antibakterielle und antioxidative Eigenschaften. Die zu untersuchenden Substanzen wurden auf ein handelsübliches Trockenfutter für Katzen aufgesprüht, so dass jedes Kilogramm wirkstoffhaltiges Futter für den ersten Versuch 517 mg Lactoferrin und für den zweiten Versuch 227 mg Epigallocatechingallat enthielt. Für jeden der beiden Fütterungsversuche standen 18 Katzen zur Verfügung, die in zwei Gruppen (Wirkstoff- und Kontrollgruppe) mit je 9 Katzen eingeteilt wurden. Zu Beginn der jeweils 28 Tage dauernden Versuche wurde bei allen Tieren der Gesundheitszustand der Maulhöhle anhand verschiedener Indices und Untersuchungen beurteilt sowie Proben für die bakteriologische Untersuchung aus der Maulhöhle entnommen. Zusätzlich wurde Blut zur Bestimmung des Antioxidativen Status abgenommen. Anschließend wurde eine gründliche Reinigung der Zähne mit vollständiger Entfernung von Plaque und Zahnstein durchgeführt („Clean-Tooth“-Modell). In den folgenden 28 Tagen erhielt eine Gruppe wirkstoffhaltiges Futter (in Versuch I Lactoferrin, in Versuch II Epigallocatechingallat), die andere Gruppe jeweils wirkstofffreies Kontrollfutter. Nach 28 Tagen wurden alle Katzen erneut untersucht und die zu Beginn des Versuches durchgeführten Untersuchungen wiederholt. Die an Tag 0 und Tag 28 des jeweiligen Versuches ermittelten Werte wurden innerhalb und zwischen der Wirkstoff- bzw. Kontrollgruppe verglichen, um Auswirkungen von Lactoferrin und Epigallocatechingallat auf die untersuchten Parameter überprüfen zu können. Die Versuche I und II wurden dabei isoliert betrachtet. Weder Lactoferrin noch Epigallocatechingallat konnten in der verwendeten Dosierung und Applikationsart die Maulhöhlengesundheit der Katzen statistisch signifikant verbessern. Beim Einsatz beider Substanzen waren jedoch Tendenzen zur Verbesserung der Maulhöhlengesundheit durch die Wirkstoffe erkennbar. Diese Tendenzen waren bei Verabreichung von Epigallocatechingallat deutlicher als bei Lactoferrin. Die Ergebnisse der In-vitro-Untersuchung der antibakteriellen Aktivität der Substanzen waren vielversprechend. Sowohl Lactoferrin als auch Epigallocatechingallat konnten das Wachstum aus der Maulhöhle der Katzen entnommener Bakterien in vitro verhindern. Aus diesem Grund erscheinen weitere Untersuchungen zum Einsatz von Lactoferrin und Epigallocatechingallat zur Prophylaxe von Parodontopathien bei Katzen sinnvoll. Um bessere In-vivo-Ergebnisse zu erzielen, sollte eine Veränderung der Dosis und unter Umständen auch eine Veränderung der Applikationsform erwogen werden. Die Verabreichung sollte höhere Konzentrationen bzw. eine längere Kontaktzeit der Substanzen in der Maulhöhle ermöglichen. Der als Nebenzielgröße untersuchte Antioxidative Status konnte durch beide Substanzen beeinflusst werden. Einige Parameter des Antioxidativen Status zeigten eine tendenzielle, andere sogar signifikante Verbesserungen gegenüber der Kontrollgruppe, so dass davon auszugehen ist, dass sowohl Lactoferrin als auch Epigallocatechingallat den Antioxidativen Status in der eingesetzten Dosierung positiv beeinflussen.

In der vorliegenden Arbeit wurde erstmals der antioxidative Status von neugeborenen Hundewelpen bestimmt. Zusätzlich wurde untersucht, ob die Zufütterung von Lactoferrin bzw. Enterococcus faecium einen Einfluss auf den antioxidativen Status der Tiere ausübt. Dazu wurden 42 neonatale Beagle-Welpen am 3. Tag nach der Geburt von den Muttertieren getrennt und in zwei unabhängigen Fütterungsversuchen von Hand aufgezogen (Lactoferrin-Versuch und Ec. faecium-Versuch). Der antioxidative Status wurde als TEAC an Tag 3, 7, 14, 28, 56, 70, 84 und 98 im Blut der Welpen gemessen. Zusätzlich wurden die Plasmakonzentrationen des antioxidativ wirksamen Vitamin E bestimmt. Als Vergleich zur mutterlosen Aufzucht dienten 11 neugeborene Beagle-Welpen, die bei ihren Mutterhündinnen aufwuchsen. An Tag 1, 3, 7 und 14 wurden bei ihnen neben der TEAC die Vitamine E und C im Blut gemessen. Außerdem wurde freies Eisen, die latente Eisenbindungskapazität sowie Gesamteisen bestimmt und die totale Eisenbindungskapazität und Transferrinsättigung berechnet. Nach der Geburt kam es bei den neugeborenen Hundewelpen durch erhöhten oxidativen Stress zu einer deutlichen Verminderung des antioxidativen Status im Blut. Dabei konnte kein Einfluss der Lactoferrin- bzw. Ec. faecium-Supplementierung festgestellt werden. Es wurde kein Zusammenhang zwischen dem erniedrigten antioxidativen Status und einem vermehrten Auftreten von Neugeborenenerkrankungen oder einer erhöhten Welpensterblichkeit offensichtlich.

Die Wirkung der prebiotisch wirksamen Substanz Lactoferrin wurde an neonatalen Hundewelpen getestet. Diese wurden am 3. Lebenstag von der Mutter getrennt, in eine Kontroll- bzw. 4 Lactoferringruppen mit unterschiedlichen Dosierungen eingeteilt und mutterlos mittels Milchaustauscher aufgezogen. Es wurde die Gewichtsentwicklung und Futterverwertung, Mikrobiologie des Kotes, Lactat- und Ammoniakgehalt im Kot, sowie der Kot-pH und Blutwerte untersucht. In den Lactoferringruppen wurde eine Erhöhung der Lactatgehalte und erniedrigte Ammoniakwerte im Kot festgestellt. Die aeroben Gesamtkeimzahlen, sowie die Enterococcengehalte wurden durch Lactoferrin verringert, die milchsäurebildenden Keime gefördert. In den Lactoferringruppen waren die Lymphozytenzahlen ab der 10. Lebenswoche um durchschnittlich 35 % im Vergleich zur Kontrolle reduziert.

This study aimed at establishing a method for checking the efficacy of probiotic substances in beagle puppies, in accordance with the requirements of the Guidelines for the assessment of micro-organisms in the animal categories dogs, cats and horses of the European Commission of October 2000. The orphan rearing of neonatal puppies was chosen as trial method, the only method to ensure avoidance of interindividual differences such as quality and quantity of mother milk. Another advantage is exact standardisation by exact prerequisites (time of weaning, type and frequency of feeding, keeping facilities, sample collection, choice of parameter, methods of analysis). In two different feeding trials with beagle puppies (duration 14 weeks) the efficacy of the prebiotic substance Lactoferrin (n=23) and of the probiotic bacteria Enterococcus faecium (n=12) on neonatal organisms was analysed. It has become clear that the general status of the puppies could be sufficiently assessed by general clinical parameters (clinical examination, food intake, weight development, quantity and consistency of faeces, blood parameter). As far as these parameters could be determined based on scoring systems, they contributed to the assessment of the substances as so-called “supporting data“. The specific parameters directly related to the probiotic action that could be established in this model are, among others, ph analysis, ammonia and lactate determination as well as microbiological analyses of faeces. Furthermore gut wall samples of duodenum and colon could be taken for immunohistochemical analysis. Antioxidative and immunmodulatory effects were assessed by means of determining trolox equivalent antioxidative capacity, vitamine E and C, uric acid, bilirubin, free ferrum, lactate, CK and GOT, the overall content of IgG and IgA, and the specific content of antibodies against humane serumalbumine in plasma. The two feeding experiments show what principles can be applied for examining the effect of prebiotically or probiotically effective substances in small animals by means of scientific standards for carrying out experiments, documenting and analysing them. Thus these experiments provide a basis for appropriate application of minimum quality standards in examining these substances, according to the requirements of the European Commission in October 2000.

Hemodialysis membrane-related neutrophil dysfunctions and pentoxifylline - a pilot study

Hemodialysis treatment is associated with activation of neutrophil granulocytes. Pentoxifylline has been shown to inhibit neutrophil activation in vitro and in vivo. We investigated the effect of pentoxifylline on leukocyte and platelet counts and on plasma levels of extracellularly released neutrophil elastase and lactoferrin during a four-hour hemodialysis treatment. Eight patients received 400 mg of pentoxifylline or placebo orally twice a day over 14 days and an additional dose of 400 mg of pentoxifylline intravenously during hemodialysis. Each subject served as his own control in a randomized, double-blind, cross-over study. Combined oral and intravenous treatment with pentoxifylline prevented neither leukopenia nor neutrophil degranulation during the time interval studied. Elastase plasma levels paralleled the drop in leucocyte counts and thereafter increased similarly in both groups. Lactoferrin plasma levels exhibited less increase in the treated group; however, this effect was not statistically significant. This may be due to the small number of cases studied and to difficulties in reaching effective plasma levels without side effects.
